4-Methylbenzylidene camphor (4-MBC), a typical organic UV filter (OUVF) in personal care products, is considered to be a potential endocrine disruptor due to its estrogenic activity and bioaccumulation. Although 4-MBC residues have been extensively identified in aquatic waters, little is known about their occurrence, levels, and potential risk in coastal waters. This study developed a reliable sampling approach, based on diffusive gradients in thin films (DGT) with XAD-2 as the binding agent, for monitoring 4-MBC in coastal waters. The diffusion coefficients of 4-MBC in freshwater and artificial seawater were 3.65 × 10 −6 cm 2 /s and 3.83 × 10 −6 cm 2 /s, respectively. XAD-2 binding gel showed rapid adsorption to 4-MBC. The accumulated masses of 4-MBC in XAD-2 DGT increased linearly with deployed time for 7 days in freshwater and seawater, which agreed well with theoretical predictions. The sampling performance was independent of ionic strength (0.0001–0.5 M), pH (4.0–8.5), and dissolved organic matter (0–20 mg/L). Field deployment in the river estuary and bathing beach showed that DGT-measured 4-MBC concentrations were consistent in comparison with grab sampling. Environmental risk assessment showed that 4-MBC may pose a medium risk to aquatic organisms based on computed risk quotient (RQ) values. Sewage discharge is the main source of 4-MBC risk, while the residue in recreation beaches contributes more significantly in summer. The established DGT sampling is suitable for seasonal monitoring, source identification, and risk assessment of 4-MBC in coastal waters. Graphical abstract

Simultaneously reducing and immobilizing Cr(VI) anions in groundwater are crucial to minimize their hazardous effects. Herein, benefiting from the feasible chemical-/photo-reduction capability of greigite, together with the ion-exchange functionality of zeolites, the integrated three-in-one function in greigite/Re-ANA composites (GRAs) material has been employed for removing and immobilizing Cr(VI) in groundwater remediation. GRA-3 achieved a 99.4% Cr(VI) removal efficiency in a Cr(VI) solution with 1 mg/L. Cr(VI) residue concentration is 0.006 mg/L, complying with the control guideline in drinking water according to WHO regulations. A comprehensive mechanism study reveals that the chemical reduction and photoreduction processes are involved within greigite, which drives the process of reducing Cr(VI) to Cr(III). Subsequently, the as-reduced Cr(III) is either adsorbed on a greigite surface or ion-exchanged with zeolite microporous channels, thus getting immobilized. Furthermore, environmentally relevant Cr(VI) contaminated groundwater sampling from the chromate plant has been employed for the fixed-bed column study, which achieved the discharge standard for practical Cr(VI) polluted groundwater. This work reported a composite material matrix with great potential and significance for Cr(VI) remediation, which could be employed in permeable reactive barriers for Cr(VI)-polluted groundwater remediation in the future.

Zwitterionic microspheres of poly2-(dimethylamino) ethyl methacrylate (PDMAEMA) block polymethacrylic acid (MAA) were prepared by RAFT dispersion polymerization in alcohols. 4-Cyano-4-(phenylcarbonothioylthio) pentanoic acid (CPDB) was used as the RAFT agent. 2,2′-Azobis(2,4-dimethyl) valeronitrile (V65) was used as the initiator, and PDMAEMA-CPDB was used as the macro-chain transfer agent (CTA). 1 HNMR was applied to determine the polymerization degrees of both PDMAEMA-CTA and PDMAEMA-b-PMAA. SEM, dynamic light scattering (DLS), and small-angle X-ray diffraction (SAX) were employed to characterize the microspheres. It was observed that the polarity of alcohols impacted the partition of MAA and V65 between the mini-droplets of PDMAEMA-CTA/MAA complexes and the continuous phase, thereby affected on the diameters of microspheres, the total conversion of MAA, and the polymerization degree of PMAA in the copolymers. For example, in methanol, the smallest diameter and polymerization degree of PMAA in the block copolymers were obtained, whereas the total conversion of MAA of the RAFT polymerization system was the highest. Most of RAFT polymerization of MAA occurred nearby PDMAEMA-CTA chains; thus, PDMAEMA-CTA acted as a template to regulate the length of PMAA in the copolymers. pH-sensitive characteristics of microspheres were dependent to the extra length of PDMAEMA-CTA, i.e., the difference of lengths between two blocks. When the length of PMAA was too short, e.g., PDMAEMA 600 -b-PMAA 87 , or the length of PMAA was closed to that of PDMAEMA, e.g., PDMAEMA 79 -b-PMAA 93 and PDMAEMA 102 -b-PMAA 131 , the amphoteric characteristics of microspheres disappeared. The self-assembly behavior of diblock copolymers in KCl alcohol-water solution was also determined by the extra length of PDMAEMA. Microspheres turned into nanofibers or nano-ribbons, but for PDMAEMA 600 -b-PMAA 87 , the microspheres maintained their original spheric morphology. These results indicated that the charge-screening effects of salts played the key roles in the self-assembly.

Olmesartan medoxomil (OM) has been used in treatment of hypertension disease, as a prodrug of olmesartan. However, its clinical application is limited by the low oral bioavailability (~28%). Ternary solid dispersion of OM was prepared with hydroxypropyl- β -cyclodextrin and N -methyl- d -glucamine to improve its oral bioavailability using roll mill ML 007. Its physicochemical characterization was measured by scanning electron microscope, differential scanning calorimetry , powder X-ray diffraction and transform infrared spectroscopy . The solutions properties of OM in ternary solid dispersion were investigated involving the solubility, dissolution profiles, 1 H NMR analysis, phase solubility study and PAMPA study. Instrument analysis results revealed an amorphous solid dispersion had been synthesized without any side-products. It also showed enhanced solubility and dissolution rate in vitro . Pharmacokinetic study demonstrated an improved oral bioavailability in vivo . The rapid storage assay showed that the chemical stability of inclusion complexes had no change. The synthesized OM ternary solid dispersion is promising to be a better alternative to its pure formation in the market with improved oral bioavailability.

Aberrant activation of the Ras–ERK signaling pathway drives many important cancer phenotypes, and several inhibitors targeting such pathways are under investigation and/or approved by the FDA as single- or multi-agent therapy for patients with melanoma and non–small-cell lung cancer (NSCLC). Here, we show that betulinic acid (BA), a natural pentacyclic triterpenoid, inhibits cell proliferation, and induces apoptosis and protective autophagy in NSCLC cells. Thus, the cancer cell killing activity of BA is enhanced by autophagy inhibition. Mitogen-activated protein kinases, and especially ERK that facilitates cancer cell survival, are also activated by BA treatment. As such, in the presence of ERK inhibitors (ERKi), lung cancer cells are much more sensitive to BA. However, the dual treatment of BA and ERKi results in increased protective autophagy and AKT phosphorylation. Accordingly, inhibition of AKT has a highly synergistic anticancer effect with co-treatment of BA and ERKi. Notably, autophagy inhibition by hydroxychloroquine (HCQ) increases the response of lung cancer cells to BA in combination with ERKi. In vivo, the three-drug combination (BA, ERKi, and HCQ), resulted in superior therapeutic efficacy than single or dual treatments in the xenograft mouse model. Thus, our study provides a combined therapy strategy that is a highly effective treatment for patients with NSCLC.

In situ monitoring of Sb speciation improves the understanding of Sb biogeochemistry and toxicity in ecosystems. Precise measurement of Sb is a challenge due to its instability of oxidation and ultratrace concentration. The development of simple and reliable methods specific to SbIII measurement is not only appealing but essential for implementing regulations. Here, we present an in situ speciation analysis method for SbIII, using the diffusive gradients in thin films (DGT) technique, combined with mercapto-functionalized SBA-15 mesoporous silica nanoparticles (MSBA). Laboratory performance tests confirmed MSBA-DGT uptake was independent of pH (4–9) and ionic strength (0.1–200 mmol L–1). DGT devices equipped with MSBA-based binding gels showed a theoretically linear accumulation of SbIII and exhibited a high capacity for SbIII at 65 μg/gel disc, with negligible accumulation of SbV over a 72 h deployment. Compared with commercial 3-mercaptopropyl-functionalized silica (MFS), the nanosized MSBA facilitate its even distribution in the binding gels. Furthermore, the good selectivity and high homogeneity of the MSBA gel enabled it to be applied in a rice rhizosphere in conjunction with AgI gel to investigate the effects of sulfur application on the SbIII solubility. In summary, the newly developed MSBA-DGT provides a selective measurement of SbIII, showing potential for environmental monitoring and further application in understanding the biogeochemical process of Sb.

The C-X-C motif chemokine 12/C-X-C chemokine receptor type 4 (CXCL12/ CXCR4) biological axis plays an important role in the pathogenesis of liver fibrosis . Curcumin is known to have an anti-fibrosis effect, but the specific mechanism needs to be elucidated. There is currently no evidence illustrating a connection between curcumin and the CXCL12/CXCR4 axis in liver fibrosis . Here, we investigated the contribution of curcumin on CXCL12/ CXCR4 biological axis in liver fibrosis. Our results showed that curcumin remarkably improved hepatic function and liver fibrosis, and the effects are similar as silymarin . The alleviation of liver fibrosis with curcumin treatment was associated with a reduction of CXCL12, CXCR4, α-SMA and RhoA. In addition, curcumin markedly inhibited the proliferation and migration of HSC-T6 cells. This study indicates that curcumin could protect against hepatic stellate cells activation and migration by inhibiting the CXCL12/CXCR4 biological axis in liver fibrosis.

Summary This study aims to identify novel antioxidant peptides within black soldier fly larvae protein (BSFLP) using simulated gastrointestinal processes, LC–MS/MS analysis, and virtual screening. Two peptides, PFCPK and ADFW, exhibited significant antioxidant properties in both in vitro radical scavenging experiments and in vivo evaluations using zebrafish embryos induced with 2,2′-azobis(2-methylpropionamidine) dihydrochloride. Treatment with 25 μg mL −1 of PFCPK or ADFW significantly ( P < 0.05) enhanced zebrafish embryo survival, reduced abnormal heartbeats and lowered ROS generation rate by 62.98% and 83.44%, lipid peroxidation by 52.54% and 68.35%, and cell death rate by 86.92% and 97.54%, respectively. Moreover, they restored antioxidant enzyme activities and MDA levels to near-normal values, demonstrating significant therapeutic effects. The observed antioxidant effects may potentially be linked to the modulation of the Keap1/Nrf2 pathway. This study provides valuable insights for developing novel natural antioxidants and maximising the high-value applications of BSFLP.

“One stone–three birds”: we developed a smart portable electrochemical sensor based on electrodeposited ferrocene-functionalized multiwalled carbon nanotubes (FC-MWCNTs) for the in vitro and in vivo detection of nicotine (NIC) in tobacco samples. The “one stone” was the establishment of the standard curve for the concentrationof NIC based on the oxidation of NIC itself as a single signal on the FC-MWCNT modified electrode or to use the FC-MWCNTs as a ratiometric probe to establish the standard curve for the NIC concentration on the modified electrode. The “three birds” were: (1) to broaden the application of the Nernst equation (the correlation between the potential and the concentration); (2) to use the function of materials modified by electrodeposition from multiple perspectives; and (3) to use a portable electrochemical workstation with a smartphone as an intelligent electrochemical sensing platform for NIC detection. The Tyndall effect, scanning electron microscopy, X-ray diffraction, UV-vis spectrophotometry, Fourier transform infrared spectrophotometry, Raman spectroscopy, cyclic voltammetry and electrochemical impedance spectrometry were used to characterize the properties of the sensor materials. The effects of the electrochemical properties, such as the electrochemical active area, the charge transfer resistance and the adsorbed charge were investigated. The sensor was then studied by differential pulse voltammetry. Under the optimized conditions, the detection range of NIC was 60–1000 μmol L−1 and the detection limit was 4.25 μmol L−1 in the single-signal mode and 0.44 μmol L−1 in the ratiometric signal mode. The sensor can be used for the rapid and accurate detection of NIC in tobacco samples, both in vitro and in vivo.

Limited studies have demonstrated that inorganic arsenic exposure is positively associated with serum vitamin D levels, although the correlation between urinary arsenic species and serum vitamin D has not been investigated in areas of water-borne arsenicosis. A cross-sectional study of 762 participants was conducted in Wenshui Country, Shanxi Province, a water-borne arsenicosis area. The results showed a positive relationship between urinary arsenic species (inorganic arsenic (iAs), methylarsonic acid (MMAV), dimethylarsinic acid (DMAV) and serum 25(OH)D. Log-binomial regression analysis indicated a 0.4% increase in the risk of vitamin D excess for every 1-unit increment in the Box–Cox transformed urinary DMAVafter adjustment for covariates. After stratifying populations by inorganic arsenic methylation metabolic capacity, serum 25(OH)D levels in the populations with iAs% above the median and primary methylation index (PMI) below the median increased by 0.064 ng/mL (95% CI: 0.032 to 0.096) for every one-unit increase in the Box–Cox transformed total arsenic (tAs) levels. Serum 25(OH)D levels increased by 0.592 ng/mL (95% CI: 0.041 to 1.143) for every one-unit rise in the Box–Cox transformed iAs levels in people with skin hyperkeratosis. Overall, our findings support a positive relationship between urinary arsenic species and serum 25(OH)D. It was recommended that those residing in regions with water-borne arsenicosis should take moderate vitamin D supplements to avoid vitamin D poisoning.

Background Intracerebral hemorrhage (ICH) is one deadly subtype of stroke with high morbidity and mortality. Oxidative stress and inflammation are major factors contributing to blood-brain barrier (BBB) dysfunction, which induces perihematoma edema and exacerbates ICH-induced secondary brain injury. SYN1 is a crucial neuronal phosphoprotein that plays a pivotal role in neuronal development. Our study was devised to clarify the function of SYN1 in hemin-induced cell injury in PC12 cells and brain injury in ICH rat models. Methods For in vitro assay, PC12 cells were first stimulated by 150 µM hemin for 4 h and then transfected with si-SYN1 and pcDNA3.1-SP1. Cell viability was tested through lactate dehydrogenase (LDH) release and CCK-8 assays. Cell apoptosis was observed through TUNEL staining. Inflammatory factors (IL-1β, IL-6, and TNF-α) were evaluated through western blotting. Oxidative markers (GSH, MDA, and SOD) were detected by adopting commercial kits. The binding sites of SP1 on the SYN1 promoter were predicted by using the JASPAR database and were validated through performing CHIP and luciferase reporter assays. SP1 and SYN1 expression in PC12 cells was investigated by RT-qPCR. For in vivo assay, rats were administrated intracerebroventricularly with si-SYN1 at 72 h before ICH induction. SYN1 and SP1 expression in ICH rats was assessed by RT-qPCR. Neurological deficits, brain edema, and BBB disruption at 24 h following ICH were assessed by conducting neurobehavioral tests, brain water content examination, and Evans blue extravasation assay. Results In vitro, hemin stimulation elevated SYN1 expression, attenuated cell viability, enhanced apoptosis, and induced inflammation and oxidative stress in PC12 cells, which were offset by SYN1 knockdown. SP1 bound to the SYN1 promoter region and positively regulated SYN1 expression. Overexpressing SP1 antagonized the impacts of SYN1 downregulation on hemin-induced damage in PC12 cells. In vivo , SYN1 and SP1 expression was upregulated in ICH rat models. Administration of si-SYN1 effectually improved neurological function, attenuated brain edema, and ameliorated BBB disruption following ICH. Conclusion SP1-mediated SYN1 probably participates in the process of neuronal damage, BBB disruption, and brain injury following ICH.

Root exudates play an important role in the phytoremediation of soils contaminated by organic pollutants, but how root exudate components affect the remediation process is not well understood. In this study, we explored the effects and mechanisms of the major root exudates, including glucose, organic acids, and serine, in the rhizoremediation of pyrene-contaminated soil. The results showed that glucose increased the degradation of pyrene (54.3 ± 1.7%) most significantly compared to the organic acids (45.5 ± 2.5%) and serine (43.2 ± 0.1%). Glucose could significantly facilitate the removal of pyrene in soil through promoting dehydrogenase activity indicated by a positive correlation between the removal efficiency of pyrene and the soil dehydrogenase activity ( p < 0.01). Furthermore, root exudates were able to change soil microbial community, particularly the bacterial taxonomic composition, thereby affecting the biodegradation of pyrene. Glucose could alter soil microbial community and enhance the amount of Mycobacterium markedly, which is dominant in the degradation of pyrene. These findings provide insights into the mechanisms by which root exudates enhance the degradation of organic contaminants and advance our understanding of the micro-processes involved in rhizoremediation.
