Design and construction of metal sulfide/phosphide heterostructures with optimized d-band center and boosted electrocatalytic oxygen evolution

Xianyu Chu,Yanan Wang,Li Jing,Wei Jiang,Yuanyuan Wu,Ming Lu,Bo Liu,Chunbo Liu,Yantao Sun,Guangbo Che

Constructing heterostructures to regulate the electronic structure is an effective strategy for enhancing the oxygen evolution reaction (OER) electrocatalytic activity. Herein, we prepared heterostructured Co 4 S 3 /CoP 3 (CoPS/NFF) electrocatalysts through a one-step phosphorization and sulfuration process. The synthesized electrode drives an overpotential of 190, 272, and 331 mV at 20, 50, and 100 mA cm −2 for OER in 1 M KOH alkaline media, respectively. These values outperformed those of monophase Co 4 S 3 and CoP 3 as well as the majority of transition metal-based catalysts previously reported. Furthermore, the Density Functional Theory (DFT) calculation results show that charge redistribution occurs at CoPS/NFF heterogeneous interfaces, which facilitates charge transfer and improves catalytic activity. The CoPS/NFF with heterostructure optimizes the adsorption strength of oxygen-containing intermediates (*O, *OH, and *OOH) by appropriately adjusting the d-band center energy level, thereby reducing the energy barrier of OER. This work provides a new perspective on rational design strategies for efficient transition metal-based electrocatalysts by inducing d-band center regulation through the construction of heterostructures.

A simple strategy for d/l-carnitine analysis in food samples using ion mobility spectrometry and theoretical calculations

Cong Liu,Ying Zou,Manli Zhang,Chaoxian Chi,Di Zhang,Fangling Wu,Chuan-Fan Ding

This work presents a straightforward approach to the separation d / l -carnitine ( d / l -Carn) using ion mobility–mass spectrometry (IM-MS) and theoretical calculations. Natamycin (Nat) was used as separation reagent to interact with the Carn, metal ions (G) were employed as ligand, the resultant ternary complexes [ d / l -Carn + Nat + G] + were observed experimentally. IM-MS results revealed that d / l -Carn could be baseline separated via complex formation using Li + , Na + , K + , Rb + , and Cs + , with a maximum peak separation resolution (R p-p ) of 2.91; Theoretical calculations were performed to determine the optimal conformations of [ d / l -Carn + Nat + Li/K] + , and the predicted collisional cross section values were consistent with the experimental values. Conformational analysis was used to elucidate the enantiomeric separation of d / l -Carn at the molecular level via the formation of ternary complexes. Furthermore, quantitative analyses for the determination of the enantiomers were established with effective linearity and acceptable sensitivity. Finally, the proposed method was successfully applied in the determination of d / l -Carn in food samples.

Bird's Nest-Shaped Sb2WO6/D-Fru Composite for Multi-Stage Evaporator and Tandem Solar Light-Heat-Electricity Generators

Yanfen Wan,Yingfei Hu,Hongyu Tu,Wenbo Zhuang,Xuemin Geng,Tian Zhang,Jinghao Zhang,Ming Wen,Peng Yang

Herein, an integrated solar-thermal-power protocol is presented at a micro-nanoscopic level to maximize the energy utilization efficiency involving utilization period and utilization patterns, and the nexus of freshwater production and nanogeneration is realized. This sophisticated vaporization device is constructed with the merits of thermally confined evaporation space in favor of recycling latent heat of condensation and optimizing light absorption based on the local sunlight angle of incidence. Inspired by a bird's nest, Sb 2 WO 6 /D-Fructose composites are prepared as photothermal absorbers to achieve a superior water evaporation rate of 2.78 kg m −2 h −1 in the Multi-stage evaporator. In addition, a synergistic tandem photo thermal-electric device with a combination of solar-driven water evaporation and further waterflow-driven hydrovoltaic generation, which can output a stable voltage of up to 360.8 mV with effective utilization of steam energy and a limited water source, is exploited. Such integrated configurations pave a pathway for clean water production and renewable power generation simultaneously toward energy issues.

Chang-Kang-Fang alleviates diarrhea predominant irritable bowel syndrome (IBS-D) through inhibiting TLR4/NF-κB/NLRP3 pathway

Sihao Zhang,Danmei Tian,Zixuan Xia,Fengge Yang,Yanhui Chen,Zhihong Yao,Yi He,Xinglong Miao,Guirong Zhou,Xinsheng Yao,Jinshan Tang

Ethnopharmacological relevance Chang-Kang-Fang (CKF), originated from traditional Chinese medicine (TCM) formulas, has been utilized to treat diarrhea predominant irritable bowel syndrome (IBS-D) based on clinical experience. However, the underlying mechanism of CKF for treating IBS-D remains unclear and need further clarification. Aim of the study The objective of this present investigation was to validate the efficacy of CKF on IBS-D model rats and to uncover its potential mechanism for the treatment of IBS-D. Materials and Methods We first established the IBS-D rat model through neonatal maternal separation (NMS) in combination with restraint stress (RS) and the administration of senna decoction via gavage. To confirm the therapeutic effect of CKF on treating IBS-D, abdominal withdrawal reflex (AWR) scores, the quantity of fecal pellets, and the fecal water content (FWC) were measured to evaluate the influence of CKF on visceral hypersensitivity and the severity of diarrhea symptom after the intragastric administration of CKF for 14 days. Subsequently, enzyme linked immunosorbent assay (ELISA) was applied to assess the effect of CKF on neuropeptides substance P (SP) and 5-hydroxytryptamine (5-HT), as well as inflammatory cytokines in serum and in intestinal tissues. Further, colonic pathological changes, the amount of colonic mast cells, and the expression level of occludin in rat colon tissues, were investigated by hematoxylin-eosin (HE) staining, toluidine blue staining, and immunohistochemistry, respectively. To explore the underlying mechanisms, alterations in colonic RNA transcriptomics for the normal, model, and CKF treatment groups were assessed using RNA sequencing (RNA-Seq). Subsequently, quantitative real-time polymerase chain reaction (qRT-PCR), western blot (WB), and immunofluorescence (IF) assays were applied to validate the effect of CKF on predicted pathways in vivo and in vitro . In addition, to elucidate the potential active compounds in CKF, 11 representative components found in CKF were selected, and their anti-inflammation potentials were evaluated using LPS-treated RAW264.7 cell models. Results CKF treatment significantly reduced the number of fecal pellets, attenuated visceral hypersensitivity, and decreased 5-HT and SP concentrations in serum and colon tissues, along with a reduction in colonic mast cell counts, correlating with improved symptoms in IBS-D rats. Meanwhile, CKF treatment reduced the colonic inflammatory cell infiltration, lowered the levels of IL-6, TNF- α , and IL-1 β in serum and colon tissues, and increased the occludin protein expression in colon tissues to improve inflammatory response and colonic barrier function. RNA-Seq, in conjugation with our previous network pharmacology analysis, indicated that CKF might mitigate the symptoms of IBS-D rats by inhibiting the Toll like receptor 4/Nuclear factor kappa-B/NLR family pyrin domain-containing protein 3 (TLR4/NF- κ B/NLRP3) pathway, which was confirmed by WB, IF, and qRT-PCR experiments in vivo and in vitro . Furthermore, coptisine, berberine, hyperoside, epicatechin, and gallic acid present in CKF emerged as potential active components for treating IBS-D, as they demonstrated in vitro anti-inflammatory effects. Conclusion Our findings demonstrate that CKF effectively improves the symptoms of IBS-D rats, potentially through the inhibition of the TLR4/NF- κ B/NLRP3 pathway. Moreover, this study unveils the potential bioactive components in CKF that could be applied in the treatment of IBS-D.

Platycodin D Ameliorates Cognitive Impairment in Type 2 Diabetes Mellitus Mice via Regulating PI3K/Akt/GSK3β Signaling Pathway

Ya-wei Lu,Li-ya Xie,Meng-han Qi,Shen Ren,Yue-qi Wang,Jun-nan Hu,Zi Wang,Shan Tang,Jing-tian Zhang,Wei Li

Objectives: The aim of this study was to investigate the ameliorative effect of platycodin D (PD) on cognitive dysfunction in type 2 diabetes mellitus (T2DM) and its potential molecular mechanisms of action in vivo and in vitro. Materials and methods: An animal model of cognitive impairment in T2DM was established using a single intraperitoneal injection of streptozotocin (100 mg/kg) after 8 weeks of feeding a high-fat diet to C57BL/6 mice. In vitro, immunofluorescence staining and Western blot were employed to analyze the effects of PD on glucose-induced neurotoxicity in mouse hippocampal neuronal cells (HT22). Results: PD (2.5 mg/kg) treatment for 4 weeks significantly suppressed the rise in fasting blood glucose in T2DM mice, improved insulin secretion deficiency, and reversed abnormalities in serum triglyceride, cholesterol, low-density lipoprotein, and high-density lipoprotein levels. Meanwhile, PD ameliorated choline dysfunction in T2DM mice and inhibited the production of oxidative stress and apoptosis-related proteins of the caspase family. Notably, PD dose-dependently prevents the loss of mitochondrial membrane potential, promotes phosphorylation of phosphatidylinositol 3 kinase and protein kinase B (Akt) in vitro, activates glycogen synthase kinase 3β (GSK3β) expression at the Ser9 site, and inhibits Tau protein hyperphosphorylation. Conclusions: These findings clearly indicated that PD could alleviate the neurological damage caused by T2DM, and the phosphorylation of Akt at Ser473 may be the key to its effect.

Manipulating the d- and p-Band centers of amorphous alloys by variable composition for robust oxygen evolution reaction

Yuci Xin,Yong Wu,Xingan Dong,Yuhan Li,Zhenxiang Cheng,Jianli Wang,Xiaolong Guo,Peng Yu

Amorphous electrocatalysts display several unique advantages in electricity-driven water splitting compared to their crystalline analogs, but understanding their structure–activity relationships remains a major challenge. Herein, we show that the d- and p-electronic states of amorphous Ni-Fe-B can be subtly manipulated by varying the Ni and Fe contents. The optimal Ni-Fe-B alloy exhibits a high performance in the oxygen evolution reaction (OER), as supported by its impressive stability (no clear degradation after 100 h) and considerably lower overpotential compared to those of its crystalline analogs. Based on theoretical calculations, different Ni and Fe contents can cause significant shifts in the d-band levels of Ni and Fe and the p-band level of B, thus altering the OER activity. Additionally, the energy difference between the d- and p-band centers (ΔE ad-p ) may be an effective index for use in reflecting the structure–activity relationship of an amorphous Ni-Fe-B alloy in the OER. An amorphous Ni-Fe-B alloy with a smaller ΔE ad-p displays a higher intrinsic activity. This study supplies a unique direction for use in constructing the structure–activity relationships of amorphous electrocatalysts by revealing the role of ΔE ad-p , which promotes fundamental research and the practical application of amorphous electrocatalysts.

Gray mold , caused by Botrytis cinerea is an intractable fungal disease that causes extensive damage to agricultural products. In the search for novel antifungal active ingredients, we discovered a linear pyranocoumarin Pd-D-V was effective against B. cinerea in both in vitro and in vivo assays. Furthermore, this study investigated the effects of Ca 2+ and the Ca 2+ -calcineurin signaling pathway on its antifungal activity against B. cinerea . The results indicated that Pd-D-V reduced the concentration of Ca 2+ in the mycelia of B. cinerea ; CaCl 2 , the Ca 2+ channel blocker verapamil , or the calcineurin inhibitor cyclosporin A could affect the sensitivity of Pd-D-V against B. cinerea ; the expression of genes ( Bccch1, Bcmid1, BccnA, Bccnb1, Bcpmc1, and Bcpmr1 ) of the Ca 2+ -calcineurin signaling pathway decreased after Pd-D-V treatment. In summary, Pd-D-V is compound for developing fungicides against B. cinerea . Pd-D-V can reduce intracellular Ca 2+ concentration and disturb Ca 2+ homeostasis . The Ca 2+ -calcineurin signaling pathway is important in the antifungal activity of Pd-D-V against B. cinerea .

Axial pyridine coordination-induced Co d-orbital rearrangement boosts peroxymonosulfate activation for singlet oxygen evolution in water remediation

Huan Chen,Yan Zhao,Zhe Zhang,Bo Niu,Yayun Zhang,Donghui Long

In this work, a cobalt phthalocyanine catalyst with axial pyridine coordination anchored on carbon nanotubes (CoPc-Py-CNT) was developed for enhanced peroxymonosulfate (PMS) activation toward singlet oxygen ( 1 O 2 ) evolution. Axial pyridine coordination induces Co 3 d -N 2 p rehybridization, elevating the Co d z 2 orbital energy level to strengthens d-p orbital hybridization with the oxygen atom in PMS, thereby lowering the activation energy barrier for O-O bond cleavage. The CoPc-Py-CNT/PMS system exhibits an 8.3-fold increase in ciprofloxacin degradation rate and nearly quantitative selectivity in aromatic alcohol oxidation to corresponding aldehydes compared to axially unmodified CoPc-CNT. CoPc-Py-CNT maintains excellent performance in both continuous-flow and 50 L pilot-scale water treatment systems, with minimal cobalt leaching (<20 μg L −1 ) and negligible ecotoxicity. This work proposes a tailored symmetry-breaking strategy via axial coordination to modulate the electronic structure of SACs, offering a robust platform for 1 O 2 -mediated water remediation and selective oxidation reactions in green chemistry applications.

Saikosaponin D Mitigates Radioresistance in Triple-Negative Breast Cancer by Inducing MRE11 De-Lactylation via HIF1α/HDAC5 Pathway

Jingyi Li,Liang Feng,Lei Zhang,Weiguo Zhang,Yanting Zhang,Xiaoqin Liu,Qianqian Cai,Weiming Zhao,Gang Huang,Changlian Lu

Background: Triple-negative breast cancer (TNBC), the most aggressive breast cancer subtype, exhibits poor prognosis due to radiotherapy resistance. However, the underlying mechanisms and effective therapeutic agents remain elusive. Methods: We employed lactate/oxamate to assess DNA damage/repair in irradiated TNBC cell lines. Lentiviral vectors for MRE11/HDAC5 constructs and shRNA were used to explore lactylation via Western blot/Co-IP. TCGA data mining, tissue microarrays, proteomics-MS, and gene expression profiling were used to dissect Saikosaponin D (SSD)'s radiosensitizing mechanisms. Promoter luciferase assays and ChIP-qPCR were performed to map SSD-induced HIF1α binding sites on the HDAC5 promoter. Results: Elevated endogenous lactate in radioresistant TNBC cells promoted DNA repair via MRE11 Lys 673 lactylation, a critical modification conferring radioresistance. HDAC5 was identified as the key delactylase for MRE11 Lys 673, validated by HADDOCK docking (hydrogen bond between MRE11 Lys 673 and HDAC5 Ser 18 ) and Co-IP (HDAC5 overexpression reduced K 673 lactylation). TCGA and clinical tissue microarrays confirmed HDAC5 downregulation in TNBC. SSD inhibits the malignant phenotype of TNBC and enhances radiotherapy efficacy by inhibition on MRE11 lactylation via upregulating HDAC5. Mechanistically, SSD upregulated HIF1α to bind the HDAC5 promoter (-342bp to -20bp region) to activate its expression. Conclusion: Lactate-driven MRE11 Lys 673 lactylation mediates radioresistance, while SSD reverses this via HIF1α/HDAC5 axis activation. Our findings identify SSD as a radiosensitizer and HDAC5/MRE11 as potential therapeutic targets for TNBC.

Augmented recruitment of host Grb2 by Helicobacter pylori tyrosine phosphorylated CagA EPIYA-D motif promotes cellular oncogenic transformation

Huilin Zhao,Shuzhen Liu,Jiangfan Shan,Juan Zhang,Qianwen Wu,Jianhui Zhang,Zekun Sun,Xinying Cao,Shu Chen,Mingzhu Feng,Yinuo Zhang,Xiang Cheng,Boqing Li,Xiaofei Ji

CagA, one of the key virulence factors of Helicobacter pylori , plays a significant role in H. pylori -associated gastric cancer by actively participating in neoplastic transformation. Among CagA variants, East Asian type CagA (CagA E ) bearing the EPIYA-D motif exhibits a higher risk than the Western-type (CagA W ) with EPIYA-C motifs. In this study, we investigated the interactions of CagA E and CagA W and host intracellular Grb2 and found a pronouncedly greater recruitment of Grb2 by CagA E compared to CagA W . Our findings revealed the phosphorylated tyrosine in the EPIYA motif is very important for the binding of CagA to Grb2. Phe at Y + 5 position in EPIYA-D, which interacts with Trp 121 in SH2 domain of Grb2, established the higher affinity than the interaction of EPIYA-C and Grb2-SH2. The substitute of Phe to Asp/Ala in CagA E EPIYA-D reduced the recruitment of Grb2 by CagA E , and neutralized the stronger induced malignant characteristics of the recipient cells. These findings provide additional insights into the distinct regulatory mechanisms employed by CagA E and CagA W , contributing to a better understanding of the higher oncogenic risk associated with H. pylori CagA E .

Redistribution of local electronic structure in RuO2 via empty d-orbital yttrium doping for robust acidic water oxidation

Moxing Cheng,Haoyun Sheng,Yin’an Zhu,Ziqi Tian,Yichao Lin

The development of highly active and durable catalysts for the acidic oxygen evolution reaction (OER) is urgently needed to accelerate reaction kinetics and reduce energy barriers. Herein, we propose to tune the local electronic structure in RuO 2 via yttrium ion (Y 3+ ) doping to facilitate the OER process following oxide path mechanism (OPM). The electron-deficient of Y 3+ redistributes local charge density, strengthens metal–oxygen bonds to suppress lattice degradation. Moreover, the incorporation of larger Y 3+ radius reduce the Ru–Ru distance, and thereby benefits O–O coupling. The optimized Y 0.11 Ru 0.89 O 2−δ catalyst exhibits an exceptionally low overpotential of 195 mV at 10 mA cm −2 for acidic OER and operates stably for 250 h at 100 mA cm −2 in a PEM electrolyzer. Theoretical investigations reveal that Y–O–Ru structure effectively modifies the chemical environment and optimizes the atomic configuration of active sites. This study establishes electron-deficient, large-radius dopants as an effective strategy for designing efficient and durable acidic OER catalysts, enhancing RuO 2 performance.

Identification of a novel ipp gene cluster responsible for 2-isopropylphenol degradation in strain Rhodococcus sp. D-6

Qian Zhu,Kangning Wei,Kaihua Pan,Gang Hu,Weihao Zhu,Yanni Huang,Changchang Wang,Qian Li,Mingliang Zhang,Jiguo Qiu,Qing Hong

Isoprocarb (IPC), a widely used carbamate insecticide, causes environmental contamination and poses risks to humans and ecosystems. Rhodococcus sp. D-6, capable of utilizing IPC as the sole growth substrate, was isolated by our lab. Strain D-6 initiates IPC degradation through ester bond hydrolysis by the hydrolase IpcH, yielding 2-isopropylphenol (IPP). In this study, the catabolic pathway of IPP in strain D-6 was elucidated, and the ipp gene cluster responsible for its degradation was predicted by transcriptomic analysis. Key genes, including a novel two-component IPP monooxygenase gene ippA1A2 and a 2-isopropylhydroquinone dioxygenase gene ippB, are responsible for the successive conversion of IPP, resulting in the ring cleavage of IPP. IppA1, a group D flavin-dependent monooxygenase, along with its reductase component IppA2, hydroxylates IPP to 2-isopropylhydroquinone using reduced nicotinamide adenine dinucleotide (NADH) and flavin adenine dinucleotide (FAD) as cofactors. The optimal molar ratio of IppA1 to IppA2 was 3:1. The apparent Km and kcat values of IppA1 for IPP were 60.52 ± 1.27 µM and 70.07 ± 0.85 s−1, respectively. IppB, an extradiol dioxygenase, catalyzes 2-isopropylhydroquinone ring cleavage to generate 2-isopropyl-4-hydroxymuconic semialdehyde. The apparent Km and kcat values of IppB for 2-isopropylhydroquinone were 29.07 ± 1.54 μM and 147.65 ± 1.92 s−1, respectively. This study provides critical insights into the molecular mechanisms of IPP degradation, contributing to a comprehensive understanding of IPC degradation.
