We investigated the regulatory effects of citrus pectin oligosaccharides (POS) from an innovative, chemically controllable degradation process on cholesterol metabolism and the gut microbial composition. The modulatory role of the intestinal flora was explored. Four-week-old male C57BL/6 mice were fed either a standard diet; a high-fat (HF) diet; or a HF diet with 0.15, 0.45, and 0.9 g/kg body weight POS for 30 days. POS reduced serum total cholesterol (TC) and low-density lipoprotein-cholesterol (LDL-C) in a dose-dependent manner. The relative abundances of specific bacterial groups in the feces and the concentrations of their metabolites were higher in the POS groups. There were significant correlations among Bifidobacterium, Lactobacillus, and Bacteroides and short-chain fatty acids, as well as among serum TC, LDL-C, fecal bile acids, and liver cholesterol 7-α-hydroxylase and 3-hydroxy-3-methylglutaryl-coenzyme A reductase. These findings indicate that the prepared POS exhibited hypocholesterolemic effects and that the potential regulatory mechanisms of citrus POS on cholesterol metabolism are modulated by specific bacterial groups together with their metabolites.

Paeoniflorin-6'-O-benzene sulfonate (CP-25), is a novel derivative of paeoniflorin (Pae) with improved anti-arthritic effects. The aim of this study was to evaluate the therapeutic effects and pharmacokinetics of CP-25 when co-administered with MTX in adjuvant-induced arthritis (AA) rats. AA rats were randomly divided into 6 groups and treated as follows: TGP (50 mg/kg/day, ig), CP-25 (50 mg/kg/day, ig), MTX (0.5 mg/kg, 3 times/week, ig), TGP (50 mg/kg/day, ig) + MTX (0.5 mg/kg, 3 times/week, ig) and CP-25 (50 mg/kg/day, ig) + MTX (0.5 mg/kg, 3 times/week, ig) from days 14 - 35 after induction. Clinical, biochemical, and histological scores were used to assess the severity of arthritis while novel ultra-performance liquid chromatography mass spectrometry (UPLC-MS/MS) was used to detect plasma concentration of CP-25. Co-administration of CP-25 and MTX significantly reduced clinical signs of inflammation, suppressed the serum production of IL-17 compared to TGP (50 mg/kg/day, ig) and MTX (0.5 mg/kg, 3 times/week, ig). Pharmacokinetics studies showed increased AUC 0-t , C max , and t 1/2 of CP-25 while V/F and CL/F decreased when co-administered with MTX. We observed an increase concentration and longer exposure with decreased clearance of CP-25 when combined with MTX, that should be further explored for clinical RA therapy.

A Beta-KIT-6 (BK) micro-mesoporous composite material was assembled using zeolite Beta seed solution as raw material. BK composite contained both zeolite Beta units and three-dimensional cubic Ia3d mesostructure of KIT-6, which served as a catalyst support for hydroisomerization of n-heptane. The composite was characterized by a variety of techniques. Results show that catalyst Pt/HBK using the composite as the support exhibited the highest isoheptanes yield among all catalysts, which is attributed to both appropriate surface acidity and excellent mass transfer properties of Beta-KIT-6 composite. Experimental results verified that the indexed isomerization factor as a general performance descriptor could accurately predict the hydroisomerization performance of support materials.

The synergetic effect of a reactive compatibilizer and organo-montmorillonite (OMMT) on phase morphology and properties of polyamide 6 (PA6)/polydimethylsilicone (PDMS) blend with a high viscosity ratio of dispersed phase and matrix was studied. A small amount of an amine-terminated copolymer of polyether amine (PEA) and PDMS (PDMS-b-PEA) as a reactive compatibilizer can significantly decrease the dispersed phase domain size. However, the dispersed phase domain size has still been above 2 μm even when the PDMS-b-PEA concentration reaches 5 wt %. OMMT as the second additive can further dramatically decrease the dispersed phase domain size due to the barrier effect for effectively preventing the coalescence of the dispersed phase domain. Moreover, the presence of PDMS-b-PEA can promote the uniform dispersion of OMMT. The synergetic effect of PDMS-b-PEA and OMMT can also improve tensile properties and decrease water absorption.

Two novel ruthenium(II) complexes coordinated with 6-chloro-5-hydroxylpyrido[3,2-a]phenazine (CQM), [Ru(L)2(CQM)]ClO4 [L = 1,10-phenanthroline, (1) and 2,2′-bipyridine, (2)], were investigated as potential fluorescent probes to track dynamic changes in the nuclei of living cells. Confocal laser technology was used to observe their co-location inside the cells. The results showed that both complexes were taken up by HepG2 cells, especially 1, which was localized in the cell nuclei, whereas 2 was distributed in the cell nuclei and mitochondria. Further studies by real-time fluorescence observations revealed that 1 rapidly entered the living cells, namely, HepG2, HeLa and MCF-7 cells, imaged the dynamic changes of the nuclei of living tumor cells, and exhibited low toxicity toward cells. The results demonstrated that 1 may be developed into a novel fluorescent probe for living cell nuclei. This study facilitates the development of fluorescent chemosensors with metal complexes.

Context: Flavonoid-enriched extract from Hippophae rhamnoides L. (Elaeagnaceae) seed (FSH) has shown beneficial effects in anti-hypertension and lowering cholesterol level. However, evidence for its efficacy in treating obesity is limited. Objective: We sought to determine if FSH can reduce body weight and regulate lipid metabolism disorder in high fat diet (HFD)-induced obese mouse model, and to investigate potential molecular targets involved.Materials and methods: C57BL/6 mice were fed with HFD for 8 weeks to induce obesity. The modeled mice were divided into four groups and treated with vehicle, rosiglitazone (2 mg/kg), low (100 mg/kg) and high (300 mg/kg) dose of FSH, respectively. Normal control was also used. The treatments were administered orally for 9 weeks. We measured the effect of FSH on regulating body weight, various liver and serum parameters, and molecular targets that are key to lipid metabolism. Results: FSH administration at 100 and 300 mg/kg significantly reduced body weight gain by 33.06 and 43.51%, respectively. Additionally, triglyceride concentration in serum and liver were decreased by 15.67 and 49.56%, individually, after FSH (300 mg/kg) treatment. Upon FSH (100 and 300 mg/kg) treatment, PPARα mRNA expression was upregulated in liver (1.24- and 1.42-fold) and in adipose tissue (1.66- and 1.72-fold). Furthermore, FSH downregulated PPARγ protein level both in liver and adipose tissue. Moreover, FSH inhibited macrophage infiltration into adipose tissues, and downregulated TNFα mRNA expression in adipose tissue (38.01–47.70%).Conclusion: This effect was mediated via regulation of PPARγ and PPARα gene expression, and suppression of adipose tissue inflammation.

Enteromorpha prolifera polysaccharide (EP2) protection against acute alcoholic liver injury (AALI) in mice was investigated. By integration of physiological indicators, gut microbiota, and short-chain fatty acids (SCFAs), the mechanism of EP2 in alleviating AALI was disclosed. The results showed that EP2 significantly ameliorated alcohol-induced abnormal transaminase activities, liver and intestinal systemic inflammation, and intestinal environmental disorders. EP2 significantly reduces liver and serum LPS contents by 1.69-fold and 1.54-fold. Furthermore, inhibition of the NF-κB signaling pathway by EP2 reduced the production of proinflammatory cytokines such as TNF-α (1.83-fold), IL-6 (11.09-fold), and IL-1β (1.99-fold). EP2 restored SCFAs to normal levels by upregulating the abundance of beneficial bacteria (Colidextribacter, Ruminococcus, unclassified_Lachnospiraceae, and Akkermansia). The alleviation of AALI by EP2 occurs through protection of the intestinal mucosal barrier and reduction of LPS permeating in serum. The decrease in LPS inactivates the NF-κB signaling pathway and prevents inflammation. In short, EP2 regulates the gut–liver axis and inflammation, alleviating effects in AALI mice.

Interleukin-6 (IL-6) is a common proinflammatory cytokine and can be adopted as an important biomarker for several diseases including nasopharynx cancer, pancreatitis, infections, and cardiovascular. It is thus necessary to design an effective method for monitoring IL-6. Herein, by preparing gold nanoparticles/graphdiyne (Au/GRD) nanohybrid with simple electroless deposition as the sensing substrate and UiO-66 metal–organic frameworks/ferrocene (UiO/Fc) as the signaling-amplifier, a highly efficient sandwich-type electrochemical immunosensing (SEI) platform was proposed for IL-6 determination. The superior electrical properties, easy preparation, high surface area, and stability of Au/GRD make it a desirable substrate to immobilize the primary antibody of IL-6. Meanwhile, the unique structure of UiO-66 is beneficial to immobilizing the Fc probe and the secondary antibody. By optimizing experimental conditions, the Fc current obtained at the as-designed SEI platform increases along with IL-6 increase, and the related linearity is 0.5 pg ml−1–1200.0 pg ml−1 coupled with a low detection limit of 0.2 pg ml−1. It is expected the as-designed SEI platform based on Au/GRD and UiO/Fc shows potential applications for IL-6 analysis.

Reuteran is a functional homopolysaccharide comprising major (α1 → 4) and interspersed (α1 → 6) linkages whose properties and functions are directly associated with the structure. In order to enrich the structural variety and application prospects of reuteran, a GtfB-type 4,6-α-glucanotransferase from Limosilactobacillus mucosae L24-B (LmGtfB), which can synthesize reuteran with high-content linear (α1 → 6) linkages, was screened. LmGtfB exhibited 4,6-α-glucanotransferase activity against starch or dextrin and can synthesize both linear and branching (α1 → 6) linkages, which offered advantages in terms of source and cost. The as-produced reuteran had the highest content of linear (α1 → 6) linkages (19% in the methylation result) among the reuterans synthesized by other GtfB-type 4,6-α-glucanotransferases. A purification method was established and applied to the extraction of the core structure of the LmGtfB products. 1H NMR and enzymatic hydrolysis revealed that the core structure contains 46.24% (α1 → 6) linkages and alternating (α1 → 4)/(α1 → 6) linkages, which was formed by enzymatic transfer of maltose.

Isobaric vapour–liquid equilibrium (VLE) data for binary systems containing o-cresol (OC) + m-cresol (MC), OC + 2, 6-dimethylphenol (DMP), and MC + DMP at 20.0 kPa were measured by using a modified Othmer still. Its reliability was confirmed by measuring the saturated vapour pressure data for the components of MC and DMP. Herington area test was used to confirm the thermodynamic consistency for these binary systems, and the results showed good thermodynamic consistency. The binary VLE experimental data were correlated and predicted by Wilson, NRTL, and UNIFAC models. The results correlated by Wilson and NRTL models agree well with the experimental values. The results predicted by UNIFAC are in accordance with experimental data with rational deviations for systems OC + MC and OC + DMP but show relatively large deviations between the predicted values and experimental data for MC + DMP system. By defining the group =COH in DMP and MC as two different groups, the experimental data were correlated, and new groups with binary interaction parameters were obtained. The results calculated by UNIFAC with new defining groups are in accordance with the experimental data.

The mechanical properties of carbon fiber (CF) reinforced thermoplastic polymer composites are primarily governed by the interphase between CFs and matrix. However, the inherent inertness of CF surfaces combined with the high viscosity and processing temperatures of thermoplastic resin often result in relatively weak interfacial bonding. This study aims to enhance the interfacial adhesion of carbon fiber reinforced polyamide 6 composites to improve their mechanical properties. CFs were de-sized and oxidized, followed by re-sizing with silanized carbon nanotubes. Fracture morphology and composition analysis of the fibers were conducted, and the fibers were subsequently incorporated into composites for mechanical testing. Results revealed a 20.0% increase in tensile strength, a 25.11% increase in flexural strength, and a 24.88% increase in interlaminar shear strength for the resized-carbon fiber reinforced polyamide 6 composites compared to the pristine-carbon fiber reinforced polyamide composites. The cross-sectional morphology of the modified composites exhibited a zig-zag fracture pattern. Dynamic mechanical analysis indicated that the modified fibers required higher activation energy for the free movement of the polyamide 6 molecular chain. These findings suggest that surface treatment enhances the interfacial adhesive between CF and resin, thereby significantly improving the mechanical properties of carbon fiber reinforced polyamide 6 composites. Highlights An efficient and reliable carbon fiber surface treatment method is proposed. Surface modification improves surface chemical activity of carbon fibers. Composites show substantial improvements in mechanical properties. Interfacial performance enhancement mechanism of composite was revealed.

Perfluorinated compounds are of extensive concern due to their prevalent use, persistence, and toxicities. There is a demand for extraction adsorbents that are efficient, easy to synthesize, and cost-effective. Herein, a magnetic nylon 6 nanocomposite (M-Ny6) was synthesized through a simple process and utilized as an adsorbent in magnetic solid-phase extraction (MSPE) of perfluorooctanoic acid (PFOA). The MSPE condition was optimized and the adsorption characteristics for PFOA was investigated. Based on the interaction between the amide groups on M-Ny6 and PFOA, the MSPE method was coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) for analyzing trace levels of PFOA in environmental water and bottled beverages. The established method demonstrated a detection limit of 1.2 ng L −1 with linear range across 50–800 ng L −1 and satisfactory recoveries (82.3–113.0 %) in spiked samples. The experimental results indicated that M-Ny6 is a promising and cost-effective MSPE adsorbent offering potential for large-scale applications in food analysis.
