Ginger ( Zingiber officinale Rosc.) is a common dietary adjunct that contributes to the taste and flavor of foods, and is also an important Traditional Chinese medicine (TCM). Different processing methods can produce different processed gingers with dissimilar chemical constituents and pharmacological activities. In this study, an ultra-performance liquid chromatography/quadrupole-time-of-flight mass spectrometry (UPLC/QTOF-MS) was applied to identify the complicated components from fresh, dried, stir-frying and carbonized ginger extracts. All of the 27 compounds were identified from four kinds of ginger samples (fresh, dried, stir-frying and carbonized ginger). Five main constituents (zingerone, 6-gingerol, 8-gingerol, 6-shogaol and 10-gingerol) in these four kinds of ginger sample extracts were simultaneously determined by UPLC-PDA. Meanwhile, the antioxidant effect of fresh, dried, stir-frying and carbonized gingers were evaluated by three assays (2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azinobis(3-ethylbenzthiazolinesulfonic acid) diammonium salt (ABTS), and ferric reducing antioxidant power (FRAP)). The results demonstrated that antioxidant activity of dried ginger was the highest, for its phenolic contents are 5.2-, 1.1- and 2.4-fold higher than that of fresh, stir-frying and carbonized ginger, respectively, the antioxidant activities’ results indicated a similar tendency with phenolic contents: dried ginger > stir-frying ginger > fresh ginger > carbonized ginger. The processing contributed to the decreased concentration of gingerols and the increased levels of shogaols, which reducing the antioxidant effects in pace with processing. This study elucidated the relationship of the heating process with the constituents and antioxidant activity, and provided a guide for choosing different kinds of ginger samples on clinical application.

Zizyphus jujuba has been used as a traditional Chinese medicinal herb since ancient time. Polysaccharides have been found to be important bioactive compounds in the jujube. This work was designed to investigate the physicochemical characteristics and bioactivities of polysaccharides purified from Zizyphus jujuba cv. Muzao . Water-soluble polysaccharides were extracted using an ultrasonically assisted extraction method. The purified polysaccharides (HJP) were obtained by deproteinization and decoloration. Three main fractions (HJP1, HJP2 and HJP3) were isolated using DEAE-Sepharose fast flow ion-exchange chromatography. The purified polysaccharides were found to consist of mannose, rhamnose, galactose, galacturonic acid, glucose and arabinose at various levels for different fractions. The HJP and its three main fractions displayed DPPH radical scavenging activities as well as relatively strong reducing power and HJP had stronger activities than homogeneous compositions. Moreover, the results from in vitro immunological activities studies indicated that HJP could improve the phagocytosis activity of THP-l cells and had effect on the expression of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6). In conclusion, the polysaccharides from Zizyphus jujuba cv. Muzao were discovered to have antioxidative and immunological activities.

Pectin, a natural polysaccharide found in the cell wall of most higher plant such as citrus, has drawn much attention due to its potential beneficial role in facilitating the treatment of many diseases like cancer, hyper cholesterol and diabetes. However, the broad application of pectin faces great limitations as the large molecular size of pectin severely prevents its bioavailability in vivo . In this study, we report an effective and highly convenient approach to degrade natural pectin into lower molecular pectin. By combining ultrasound with Fenton system (US-Fenton), we show that ultrasound synergistically enhances the efficiency of Fenton reaction to degrade pectin into 5.5 kDa within only 35 minutes. Importantly, RG-I domain, the most effective portion of natural pectin, was well preserved and highly enriched. In addition, the antioxidant activities of US-Fenton-treated pectin was significantly elevated. The mechanism of this novel observation was further investigated through the multiple structural analyses including HPLC, IR and NMR. Taken together, we present a novel and convenient approach to generate ultra-low molecular weight pectin with high efficiency and higher bioactivity. We expect our approach will have broader applications in improving the bioavailability and bioactivity of other polysaccharide-based natural compounds.

Blends of phthalonitrile ( CN ) and alicyclic imide compounds synthesized from tetrahydrophthalic anhydride ( CC ), methyl tetrahydrophthalic anhydride ( MCC ) and hexahydrophthalic anhydride ( HCC ), respectively, were prepared and characterized. Results from rheological studies demonstrated the scope of a novel Thermal Synergistic Polymerization (TSP) effect between phthalonitrile and the alicyclic imide compounds. FTIR data showed a unique phenomenon, which is, the C N of the phthalonitrile compound could be completely and rapidly consumed without generating any triazine ring. Further works on cured products of CC / CN system showed comparable or even higher thermal oxidation stability, low boiling water absorbance and moderate dielectric constant ( ε′ in the range of 4.8∼4.5) with almost no voids compared with that of the phthalonitrile resins promoted by traditional curing system of active hydrogen. Model system was designed to study the mechanism of TSP effect of alicyclic imide moiety/phthalonitrile. The systematic characterizations by FTIR, UV–Vis, MALDI-TOF MS, free radical scavenger (DPPH), EPR , etc. showed that the TSP reaction undergoes both copolymerization and homopolymerization with no triazine formation, involving a free radical process. Structural analysis of the oligomers suggested that phthalocyanine rings were a minor component of the thermoset networks, while polyisoindolines were the primary structural motif.

In order to explore the potential of different subtropical fruit leaves as natural sources of bioactive compounds, free, esterified and insoluble-bound forms of total phenolic contents (TPC) and total flavonoid contents (TFC) were determined. The antioxidant activities of fruit leaves was determined by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (DPPH), ferric reducing antioxidant power (FRAP), and trolox equivalent antioxidant capacity (TEAC) assays. Furthermore, several phenolic compounds were detected using ultra performance liquid chromatography (UPLC) technique. Rutin, (+)-catechin, isoquercitrin, quercitrin and quercetin were widely found in these fruit leaves. The highest TPC, TFC and antioxidant capacities were found in Averrhoa carambola , Dimocarpus longgana Lour, Litchi chinensis Sonn , Mangifera indica and Psidium guajava Linn . The results implied that these leaves are important natural sources of antioxidants for preparing functional food ingredients and preventing oxidative stress diseases.

In the current study the influence of different lipid-based formulations (Pickering and nanoemulsions) and their droplet size on curcumin encapsulation and bioaccessibility, as well as on its anti-oxidant activity was investigated. Oil-in-water Pickering emulsion stabilized by chitosan-tripolyphosphate (CS-TPP) nanoparticles and nanoemulsions containing an organic phase (Span80:Tween80), were prepared with either medium chain triglyceride (MCT) or corn oil as long chain triglyceride (LCT). An in vitro gastrointestinal (GIT) model consisting of mouth, gastric and intestinal phases was used to characterize the rate and extent of lipid phase digestion of the ingested samples. A centrifugation method determined fraction of curcumin released into mixed micelles after digestion (bioaccessibility). These findings showed that after subjecting to simulated GIT model, all the emulsion systems experienced a progressive increase in mean particle size, due to droplet flocculation and coalescence after digestion. Electrical charge (ζ) of particles was observed to become highly negative as they passed through GIT due to accumulation of anionic bile salts, phospholipids and free fatty acids at their interfaces. The rate and extent of lipid digestion and bioaccessibility of curcumin increased with decreasing mean droplet diameter (NMCT > NCO > PMCT > PCO). Finally, we showed that as compared to free curcumin, the encapsulated curcumin showed higher radical scavenging activity (RSA), which confirmed the protective effect of the emulsion systems on the antioxidant activity of curcumin.

BACKGROUND Due to the disadvantages of traditional enzymolysis, pretreatments are crucial to enhance protein enzymolysis. Enzymolysis kinetics and thermodynamics, amino acids composition, molecular weight distribution, fluorescence spectroscopy and antioxidant activity of thermal (HT) and single frequency counter-current ultrasound (SCFU) pretreated sodium caseinate (NaCas) were studied. RESULTS Enzymolysis of untreated NaCas (control) improved significantly ( P < 0.05) by SFCU and followed by HT. Values of the Michaelis–Menten constant ( K M ) of SFCU and HT were 0.0212 and 0.0250, respectively. HT and SFCU increased ( P < 0.05) the reaction rate constant ( k ) by 38.64 and 90.91%, respectively at 298 K. k values decreased with increasing temperature. The initial activation energy (46.39 kJ mol −1 ) reduced ( P < 0.05) by HT (39.66 kJ mol −1 ) and further by SFCU (33.42 kJ mol −1 ). SFCU-pretreated NaCas hydrolysates had the highest contents of hydrophobic, aromatic, positively and negatively charged amino acids. Medium-sized peptides (5000–1000 Da) are higher in SFCU (78.11%) than HT and the control. SFCU induced molecular unfolding of NaCas proteins. Accordingly, SFCU-pretreated NaCas hydrolysate exhibited the highest scavenging activity on DPPH and hydroxyl radicals, reducing power, and iron chelating ability. CONCLUSION SFCU pretreatment would be a useful tool for production of bioactive peptides from NaCas hydrolysate. © 2016 Society of Chemical Industry

Six lactic acid bacterial (LAB) strains were isolated from traditionally fermented Xinjiang cheese and evaluated for functional and probiotic properties and potentials as starter cultures. The isolated six LAB strains comprised Lactobacillus rhamnosus (one strain), Lactobacillus helveticus (one strain), and Enterococcus hirae (four strains). All of the six strains were tolerant to acidic and bile salt conditions. Among which, the L. rhamnosus R4 strain showed more desirable antimicrobial, auto-aggregation, and hydrophobic activity. In addition, the strain L. rhamnosus R4 exhibited the highest level of free radical scavenging activity (53.78% of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals and 45.79% of hydroxyl radicals). L. rhamnosus R4 also demonstrated cholesterol and triglyceride degradation by 50.97% and 28.92%, respectively. To further examine the health-promoting effects of these LAB strains on host lifespan, Caenorhabditis elegans was used as an in vivo model. Worms fed LAB as a food source had significant differences in lifespan compared to those fed Escherichia coli OP50 (as a negative control). Feeding of L. rhamnosus R4 extended the mean lifespan of C. elegans by up to 36.1% compared to that of the control. The results suggest that the strains isolated from Xinjiang fermented dairy products have high potential as starter cultures in the cheese industry.

Background
Dracocephalum heterophyllum was a traditional Tibetan medicine possesses various pharmacological effects involved in anti-inflammatory, antibacterial activities. However, its anti-hepatitis, antioxidant activity and bioactive compounds have not been reported, the objective of this research work was to investigate the pharmacological activity and bioactive compounds of D. heterophyllum extracts. Results In the present study, the anti-hepatics and antioxidant activities of four D. heterophyllum extracts (i.e. petroleum ether extracts, ethyl acetate extracts, n -BuOH extracts, and water extracts) were conducted. The main chemical constituent of petroleum ether and ethyl acetate extracts were also isolated using chromatographic techniques and identified by NMR spectroscopic methods. The anti-hepatitis assay showed that the petroleum ether and ethyl acetate extracts of D. heterophyllum significantly prolonged the mean survival times and reduced the mortality of mouse hepatitis model induced by concanavalin A (ConA). The levels of alanine transaminase, aspartate transaminase in blood serum could be decreased obviously by ethyl acetate extracts compared with ConA group (P < 0.01). The histological analysis demonstrated that the ethyl acetate extracts could inhibit apoptosis and necrosis caused by ConA. In addition, the antioxidant activities of the four extracts of D. heterophyllum were measured by DPPH assay, ABTS assay, anti-lipidperoxidation assay, ferric reducing antioxidant power assay, ferrous metal ions chelating assay and determination of total phenolic contents. The results showed that the ethyl acetate extract had the highest antioxidant activities, followed by petroleum ether extract. Finally, nine mainly compounds were isolated from the Petroleum ether and ethyl acetate extracts, including four triterpenes: oleanolic acid ( 1 ), ursolic acid ( 2 ), pomolic acid ( 3 ), 2α- hydroxyl ursolic acid ( 4 ), three flavonoids: apigenin-7- O -rutinoside ( 5 ), luteolin ( 8 ), diosmetin ( 9 ) and two phenolic acids: rosmarinic acid ( 6 ), methyl rosmarinate ( 7 ). Conclusion The Ethyl acetate extract of D. heterophyllum had the highest anti-hepatitis and antioxidants activities, followed by petroleum ether extract. The bioactive substances may be triterpenes, flavonoids and phenolic acids, the ethyl acetate extracts of D. heterophyllum may be possible candidates in developing anti-hepatitis medicine.

The fruit (aril) of the endangered genus Taxus plants is an abandoned herbal resource. Traditionally, people enthusiastically focus on its bark, its renewable, tremendous arils fall into the soil with seeds after they are mature. The present research investigated the fruit of two species from the genus Taxus , Taxus chinensis var. mairei , and Taxus media , with regards to their antioxidant and antihyperglycaemic activities, safety, and bioactive constituents. Results showed that T . chinensis var. mairei and T . media both had certain biological activities with T . chinensis var. mairei better in antioxidant activity and T . media better in antihyperglycaemic activity. Correlation analysis revealed that the differences in bioactivities depended on content of their mainly chemical components. The mice acute oral toxicity test indicated that the methanol extracts of the two biotypes of Taxus were safe. And nineteen compounds were tentatively assigned from the two varieties, via tandem mass spectrometry using a LC-ESI-Q-TOF-MS instrument, which included phenols, flavonoids, and terpenes. These results indicate a possible application of Taxus fruit extracts in various fields like in food industry, however, this still needs further investigations. Graphical

ICAM-1 overexpression and subsequent adhesion of leukocytes to endothelial cells play critical roles in the early stage of atherosclerosis. Danshenol A (DA) is an abietane-type diterpenoid isolated from traditional Chinese herb Salvia miltiorrhiza Bunge. The mechanisms under its regulation of adhesion of molecular expression are explored. Here, the effect of DA on TNF-α-induced ICAM-1 expression was investigated in endothelial cells. TNF-α-induced ICAM-1 expression and subsequent adhesion of monocytes, as well as elevated reactive oxygen species (ROS) generation and NOX4 expression were all significantly reversed by DA, siNOX4 and NOX4 inhibitor GKT137831. Furthermore, TNF-α-induced ICAM-1 expression, which was increased via IKKβ/IκBα-mediated activation of NF-κB p65, was also inhibited by DA. Interestingly, NOX4 overexpression suppressed the ICAM-1 expression, and this finding may be ascribed to the activation of Nrf-2. Additionally, NF-κB inhibitor PDTC, siNOX4, or DA can decrease the TNF-α-induced ICAM-1 expression and suppress the adhesion of monocytes. In all, DA inhibited TNF-α-induced ICAM-1 expression and subsequent monocyte adhesion to endothelial cells through the NOX4-dependent IKKβ/NF-κB pathway. Besides, NOX4 played dual role in regulating ICAM-1 expression via diverse signal pathway. This novel bioactivity will make DA a good candidate to be further explored for therapeutic or preventive application for atherosclerosis.
