Photoreduced Ag+ surrounding single poly(4-cyanostyrene) nanoparticles for undifferentiated SERS sensing and killing of bacteria

Ya-Qin Liu,Wei Zhu,Quan Yuan,Ji-Ming Hu,Xin Zhang,Ai-Guo Shen

Developing a rapid, low cost and sensitive sensing strategy for undifferentiated detection and fast killing of bacterial pathogens are critical to alleviating bacteria infections. Here, we propose a direct photoreduction method to synthesize the SERS tag by integrating poly(4-cyanostyrene) nanoparticles (NPs) and silver ions , which are applied as bio-sensing system for bacteria sensing and fast killing. Under a focused laser spot, silver ions on the surface of the poly(4-cyanostyrene) NPs could be photoreduced into Ag NPs, thereby causing the Raman signal amplification of poly(4-cyanostyrene) NPs up to 40 times, and there is a good linear correlation between the Raman intensity of poly(4-cyanostyrene) NPs and different concentrations of Ag + . Moreover, 4-mercaptophenylboronic acid, performing the same recognition function for both the Gram-positive and Gram-negative bacteria, is used as bridge between the bacteria and Ag + by the inherent chemical bonding. Based on further constructed bio-sensing system, we achieved the quick count and killing of both Gram-positive bacteria, e.g. , Staphylococcus aureus ( S. aureus ), and Gram-negative bacteria, e.g. , Escherichia coli ( E. coli ). Notably, the sensing strategy can detect at least ∼100 cells of E. coli , ∼10 cells of S. aureus and ∼10 cells of their mixture in less than 40 min. The detection accuracy for actual samples can also reach over 80% and the bacteria were entirely killed by Ag + after the detection, avoiding bacterial contamination in the environment. This novel method is anticipated to perform as a simple yet effective tool for fast and sensitive bacteria counting and killing.

A TiO2-based bioprobe enabling excellent SERS activity in the detection of diverse circulating tumor cells

Yanping Xu,Jie Lin,Xiaoxia Wu,Xiawei Xu,Dinghu Zhang,Yujiao Xie,Ting Pan,Yiwei He,Aiguo Wu,Guoliang Shao

Circulating tumor cells (CTCs) can be the seeds of tumor metastasis and are closely linked to cancer-related death. Fast and effective detection of CTCs is important for the early diagnosis of cancer and the evaluation of micrometastasis. However, the extreme rarity and heterogeneity of CTCs in peripheral blood make sensitive detection of CTCs a big challenge. In this paper, a TiO2-based surface-enhanced Raman scattering (SERS) bioprobe is reported for the first time with outstanding ultrasensitive specificity, excellent stability of the signal, and good biocompatibility for the detection of CTCs. The TiO2 NPs were encoded with alizarin red (AR) and functionalized with reduced bovine serum protein (rBSA) and folic acid (FA). The limit of detection (LOD) for 4-mercaptobenzoic acid (4-MBA) and AR molecules adsorbed on the TiO2 SERS substrate is 5 × 10−7 M. The designed TiO2-based SERS bioprobe can be effectively utilized in detecting four diverse types of cancer cells in rabbit blood, which shows good sensitivity of the SERS detection technology. Finally, precise targeting of CTCs based on the SERS bioprobe with the function of fluorescence imaging is also confirmed by the fluorescence colouration test. This work offers a novel strategy for CTC detection and the development of non-noble metal semiconductor-based SERS platforms for tumor diagnosis.

Ultra-Sensitive, Rapid and On-Site Sensing Harmful Ingredients Used in Aquaculture with Magnetic Fluid SERS

Meizhen Zhang,Jingru Liao,Xianming Kong,Qian Yu,Miao Zhang,Alan X. Wang

The integration of surface-enhanced Raman scattering (SERS) spectroscopy with magnetic fluid provides significant utility in point-of-care (POC) testing applications. Bifunctional magnetic–plasmonic composites have been widely employed as SERS substrates. In this study, a simple and cost-effective approach was developed to synthesize magnetic–plasmonic SERS substrates by decorating silver nanoparticles onto magnetic Fe3O4nanoparticles (AgMNPs), which function both as SERS-active substrates and magnetic fluid particles. The strong magnetic responsivity from AgMNPs can isolate, concentrate, and detect target analytes from the irregular surface of fish skin rapidly. We fabricate a microfluid chip with three sample reservoirs that confine AgMNPs into ever smaller volumes under an applied magnetic field, which enhances the SERS signal and improves the detection limit by two orders of magnitude. The magnetic fluid POC sensor successfully detected malachite green from fish with excellent selectivity and high sensitivity down to the picomolar level. This work achieves a label-free, non-destructive optical sensing approach with promising potential for the detection of various harmful ingredients in food or the environment. Keywords:magnetic fluid;surface-enhanced Raman scattering;POC sensor;AgMNPs;pesticides

Preparation of SERS active filter paper for filtration and detection of pesticides residue from complex sample

Min Hou,Ning Li,Xiaoran Tian,Qian Yu,Juan-P. Hinestroza,Xianming Kong

The selectivity is needed mostly in SERS sensing because analytes of interest are commonly present in a complex mixture containing particles and impurities, which hinder the interactions between the laser and analyte being detected. In this manuscript, we describe our efforts developing a simple and instant. method to prepare a filter paper SERS sensor. Colloidal Ag nanoparticles were immobilized on one side of filter paper via an in-situ growth method. The fabrication process of the sensor could be finished in several minutes, and no special facility needed. The filter paper SERS sensor demonstrated a spectra uniformity with a 7.0 % point-by-point signal deviation. And the filter function of the sensor could effectively filter out interferences from samples in 1 min, that allowing the direct detection of thiram in ketchup by SERS with detection limit of 93 ppb. Furthermore, we used a Quick Easy Cheap Effective Rugged and Safe (QuEChERS) sample preparation method to detect malachite green (MG) in soil with a sensitivity as low as 0.01 ppm without any sample pre-treatment or purification. A SERS filter paper may open a new avenue for rapid testing of food quality during manufacturing as well as fast detection of potential contaminants in a myriad of substrates.

Synthesis of an organic phosphoric acid-based multilayered SERS imprinted sensor for selective detection of dichlorophenol

Hongji Li,Hongda Xu,Jinyue Zhang,Yi Li,Haochen Yu,Yibo Zhao,Dandan Wang,Yunhui Li,Jianwei Zhu

A novel surface enhanced Raman scattering (SERS) substrate was synthesized by wrapping Ag nanoparticles on the surface of the MIL-101 (Fe) shell under the assistance of IP6, and combining with molecular imprinted polymers (MIPs) for the selective detection of 2,6-DCP. In this study, as a stabilizer and linking agent, IP6 can form a chemical bond with MIL-101 (Fe) to decrease the distance between two Ag nanoparticles and generate more “hot spots”. On the other hand, the inositol hexaphosphate (IP6) and MIL-101 (Fe) shell acted as a protective shell layer, preventing Ag nanoparticles from being polluted by the external environment and reduced SERS activity. In addition, the SERS-MIPs can selectively detect the target molecule, avoiding the influence of other interfering substances on the detection effect. The prepared sensor (Ag/IP6@MIL-101 (Fe)@MIPs, AIM@MIPs) presented sensitive SERS properties, and the detection of limit can reach 10−9 M. The AIM@MIPs also presented strong selective detection to the target molecules, and can be applied for practical sample detection. This test provides a potential application in the detection of environmental pollutants.

Large-area Co(OH)2 nanoflower array films decorated with Ag nanoparticles as sensitive SERS substrates

Tingting Fan,Yan Ke,Lijun Zhang,Li Cai,Zhongbo Li

Large-area three-dimensional (3D) well-ordered Co(OH)2 nanoflower array films decorated with Ag nanoparticles (Ag NPs) with a novel structure were designed as sensitive SERS substrates for trace detection of pesticides. Co(OH)2 nanoflowers were electrodeposited on the surface of molded polyacrylonitrile (PAN) films with ordered nanopillar arrays and then Ag NPs were sputtered onto of the surface of each Co(OH)2 nanoflower. The SERS performance of the as-prepared films can be optimized by tuning the Ag-sputtering duration and the morphology of the Co(OH)2 nanoflower arrays. Owing to the formation of high-density SERS “hotspots” created in the nanogaps between the adjacent Ag NPs located on the same and neighbouring Co(OH)2 nanosheets, the optimized substrates show SERS activity for probe molecules. As a result, 10−11 M rhodamine 6G and 10−9 M 4-aminothiophenol can be SERS recognized, respectively. To verify their practical application, the optimized SERS substrates were employed for trace detection of thiram (a type of pesticide), and 10−9 M thiram can be recognized, showing great potential in pesticide detection.

Dynamic monitoring of bacteriostatic process by SERS analysis based on a simple but effective detection strategy

Songtao Xiang,Yuxiang Lan,Zhiliang Mai,Feng Tian,Hua Mao

Investigating antibacterial process at a molecular level is helpful to fully understand the mechanism of bacteriostasis and develop new antimicrobial agents. Herein, a simple but effective sensor strategy of antibacterial nanocomposite combined with surface-enhanced Raman scattering (SERS) substrate was applied for the robust detection of bacteriostatic process. The synergistic SERS effect of nanocomposite and Ag nanoparticles (NPs) substrate was confirmed by finite difference time domain (FDTD) solutions. A curcumin [email protected] NPs nanocomposite was designed and prepared as a kind of bacteriostatic agent and SERS material as well. By means of electrostatic attraction between the nanocomposite and bacteria (methicillin resistant staphylococcus aureus, MRSA), specific detection of MRSA and monitoring of the molecular structure changes after bacteriostatic   effect were realized by SERS. Important intermediates produced in the bacteriostatic process were also measured at the same time. The relationship between the relative peak intensities and the structure of MRSA were thus established. The results were verified by high performance liquid chromatography-mass spectrometry (HPLC-MS), reactive oxygen species (ROS) kit, and flow cytometry. The detection strategy we proposed could not only be used for real-time detection of bacteriostatic processes with a high efficiency, but also a powerful tool for analyzing the mechanism in biochemical processes.

Preparation of a superhydrophobic AgNP/GF substrate and its SERS application in a complex detection environment

Chundong Liu,Fengcai Lei,Yisheng Wei,Zhen Li,Chao Zhang,Qianqian Peng,Baoyuan Man,Baoyuan Man,Jing Yu,Jing Yu

Surface-enhanced Raman scattering (SERS) is widely considered to be a fingerprint spectrum that can realize molecular identification, and it continues to receive a lot of attention due to its high sensitivity and powerful qualitative analysis capabilities. In recent years, there has been a lot of work and reports on super-sensitive SERS substrates, but often the enhanced ability of the substrate is also effective for impurities and irrelevant molecules. Therefore, a problem that still remains to be solved is how to perform effective trace detection of specific substances in a complex detection environment. Herein, a superhydrophobic Ag nanoparticle/glass microfibre filter (AgNP/GF) substrate was designed to realize the Raman detection of complex multiphase solutions. The hydrophobic three-dimensional net-like structure provides efficient Raman enhancement, making the substrate have extremely high detection limits for dye molecules and even achieving specific detection of the hexane phase component (thiram molecule) in a multiphase solution.

Metal–organic framework modified by silver nanoparticles for SERS-based determination of sildenafil and pioglitazone hydrochloride

Ding Yanru,Cheng Yuqi,Hao Baoqin,Zhu Lin,Zhang Nan,Zhao Bing,Tian Yuan

A versatile surface-enhanced Raman scattering (SERS) assay has been established that can realize rapid and sensitive determination of sildenafil (SIL) and pioglitazone hydrochloride (PIO) adulteration in healthcare products. Metal–organic frameworks-silver nanoparticles (MOFs-AgNPs) with SERS activity were successfully prepared via in situ synthesis AgNPs on the MOFs surface. By virtue of the adsorptivity of MOFs, the MOFs-AgNPs could effectively concentrate the drug molecules on the electromagnetic enhancement areas of AgNPs. Moreover, the MOFs-AgNPs substrate exhibited more sensitive SERS activity than classical AgNPs with linear range of 1.0 × 10 –7 –1.0 × 10 –5  mol L −1 for SIL and 8.0 × 10 –7 –3.0 × 10 –5  mol L −1 for PIO and limit of detection (LOD) of 4.8 × 10 –8  mol L −1 for SIL and 1.4 × 10 –7  mol L −1 for PIO. The designed method realized the determination of SIL and PIO in commercial tablets and healthcare products with recoveries of 93.8–108.0% and 93.0–104.0%, respectively, with relative standard deviation (RSD) of 2.7–4.1% and 2.2–4.2%, respectively. The present system displayed little interference effect on determination. This work provides a multifunctional route for the determination of other drugs via the SERS technology. Graphical abstract

Modulating the plasmon-mediated silver oxidation using thiophenol molecules as monitored by in situ SERS spectroscopy

Guilin Wang,Kai Wang,Chengyu Zhang,Yameng Zhu,Xueyan Jiang,Zhuoyao Li,Chengrong Yin,Huili Ma,Juqing Liu,Xiao Huang,Gang Lu

Effective charge separation is essential in plasmon-mediated photochemistry and is usually achieved by constructing plasmon–semiconductor interfaces, which is usually challenging. In this work, by monitoring the plasmon-mediated silver oxidation with in situ Raman spectroscopy, we demonstrate that the adsorbed thiophenol molecules could modulate the rate of photochemical reactions by tuning the charge separation at the plasmon–molecule interfaces. It is found that the thiophenol molecules with strong electron-withdrawing or donating functional groups could accelerate or decelerate the rate of plasmon-mediated silver oxidation, respectively. Owing to the easy tuning of the electronic structures of organic molecules via substitution, our method provides a versatile and convenient approach for the fine modulation of plasmon-mediated photochemical reactions.

Controllable synthesis of the homogeneous 3D Ag nanoflowers on FTO substrate for ultra-sensitive SERS detection

Qianqian Shang,Xin Tan,Mingcan Chen,Songyue Han,Tao Yu

High-efficient SERS substrates containing dense hot spots were fabricated by assembling 3D Ag nanoflowers (AgNFs)@Au-FTO substrate by using citrate ions as the shape-guiding agent. The vertical growth of 3D AgNFs could be attributed to the synergistic effects of the concentration gradient layer of Ag + and the density gradient of the charge carrier. Combined with the simulation result of FDTD method, the enhanced SERS sensitivity and repeatability of AgNFs@Au-FTO could be attributed to the enlarged specific surface area, and the improved density of hot spots contributed by the overlap between the vertically stacked AgNSs with a high population of sharp edges.

Development of Au NPs-decorated filter paper as a SERS platform for the detection of benzidine

Rong Wang,Hongyan cao

In this paper, a simple and cost-efficient strategy was used to construct a uniform Au NPs distribution on the surface of flexible filter paper for the detection of benzidine. Taking full advantage of the adsorption properties of filter paper, small gold nanoparticles were adsorbed onto its surface as gold seeds, and subsequently grown by electroless plating to form a highly uniform distribution of Au NPs substrates. By changing the electroless plating time, an optimal substrate was obtained. The as-prepared substrate exhibited satisfactory sensitivity with a low detection limit of 10−13 M for 4-ATP, and good reproducibility and homogeneity. Furthermore, the as-prepared substrates were successfully used for the detection of benzidine in environmental water, with a minimum detection concentration as low as 0.1 ppm and recoveries in the range of 92.4 to ∼108.5%. This study indicated that filter paper-based SERS substrates have great potential value in the detection of environmental organic pollutants.
