Efficient Preparation of High-Purity Sulforaphene: Selective Immobilization of Myrosinase by Designing a Concanavalin A-Loaded Metal–Organic Framework

Mintong Tian,Yuman Guo,Meng Zhou,Purui Yu,Mingming Li,Zehua Li,Qipeng Yuan

Isothiocyanates (ITCs) in cruciferous plants are natural active ingredients with cancer-preventive, anti-inflammatory, and antibacterial functions. ITCs are generated through the hydrolysis of glucosinolates by myrosinase (MYR). However, the presence of specifier proteins, especially epithionitrile-specifier proteins (ESPs), significantly impacts the yield, purification, and application of ITCs. Herein, we designed a metal–organic framework to selectively immobilize MYR by constructing a concanavalin A (ConA)-loaded ZIF-8 core. The 1.55-fold higher sulforaphane (SFE)-generation activity of ConA-MYR@ZIF-8 compared to ConA-free groups demonstrated the selectivity of ConA@ZIF-8 toward MYR. Impurities, including nitrile generated by ESP, were effectively excluded. ConA-MYR@ZIF-8 maintained over 93.9% activity after 1 week of storage at 4 °C and retained 77.9% activity after 20 cycles. SFE with 97.8% purity was prepared in a yield of 14.9 mg/(g·h) through enzymatic reactions coupled with real-time dichloromethane column extraction, achieving a recovery rate of 90.1%. This study provided new insights for the efficient preparation of SFE and other ITCs.

Mercapto-modified layered double hydroxide: high-efficiency emerging amendment for cadmium immobilization in cadmium-contaminated acidic and alkaline soil

Xusheng Gao,Xilin Chen,Yifei Shu,Kexin Chen,Ge Gao,Xiaomin Liu,Qingqing Huang,Lin Wang,Yuebing Sun,Yujie Zhao,Xuefeng Liang

Soil cadmium (Cd) contamination has emerged as a critical environmental challenge in Chinese agricultural systems. The remediation of Cd-contaminated farmland necessitates high-efficient immobilization amendments. The present study successfully synthesized mercapto-modified MgAl- and ZnAl-layered double hydroxides (MgAl/ZnAl-MP-LDHs) as structurally stable and high-performance amendments via a coprecipitation-condensation method. Batch adsorption experiments revealed superior Cd 2+ sorption capacities for MP-LDHs, with the maximum Cd 2+ capacities increased by 1.6–1.8 times compared to the unmodified materials. The XPS analysis further elucidated the dual sorption mechanisms involve in Cd 2+ absorption by MP-LDHs: complexation via mercapto groups and precipitation mediated by hydroxyl groups. Moreover, soil application of MP-LDHs at 0.025% dosage (w/w) demonstrated rapid immobilization efficacy, reducing available Cd content by 38.41%–93.20% within 24   h in alkaline and acidic soil. Subsequent analysis revealed that MP-LDHs exhibited pH-resilient performance in soil Cd immobilization. Potting experiment further validated practical applicability of MP-LDHs, reducing pakchoi shoot Cd concentration by 53.52%–95.24% at 0.025%–0.1% dosage. The findings established MP-LDHs as multifunctional amendments with ultralow application dosage requirements, excellent immobilization efficiency, and universal performance across varying soil pH levels, hence positioning MP-LDHs as a scalable solution for safe utilization of Cd-contaminated soil.

Phase-Specific Immobilization of Phospholipase D as an Efficient Pickering Emulsion Interfacial Catalyst for Converting Antarctic Krill Oil Phospholipid

Lili Yin,Wenjia Feng,Yang Hu,Xiangzhao Mao

Phosphatidylserine (PS), a rare phospholipid in Antarctic krill oil (AKO) critical for brain development, can be produced from the abundant phosphatidylcholine (PC) using phospholipase D (PLD) in Pickering emulsion interfacial catalysis (PIC) systems. However, the exposure of PLD to organic solvent around the emulsion interface diminished PLD activity, limiting the conversion efficiency of PS. In this study, we proposed a strategy to fabricate a PIC system with high efficiency and stability by immobilizing PLD in a specific phase on the emulsion interface, based on investigating the effect of the interfacial microenvironment on PLD activity. Janus-poly(acrylic acid)/polystyrene (JPP) and Janus-polyethylenimine/octadecane (JPO) particles were fabricated as carriers to realize the specific-phase immobilization of PLD. The highest activity was observed when PLD was immobilized on the hydrophilic side of JPP (PLD@JPP(W)), 1.9-fold that of free PLD. The catalytic efficiency of PLD@JPP(W) was 1.7-fold that of free PLD, confirmed by the kcat/Km value enhancement. Immobilization on the hydrophilic side also enhanced the thermal stability of PLD. The half-lives of PLD were extended from 4 to 36 h at 40 °C and from 6 to 28 days at 4 °C. Importantly, PLD@JPP(W) showed excellent catalytic efficiency as a PIC system, achieving a PS productivity of 93% within a short time of 2 h at an enzyme dosage of 0.05 mg. PLD@JPP(W) exhibited a 3.6 times higher yield than free PLD in the production of PS from PC rich in Antarctic krill oil. The strategy in this work could also be applied to other lipases, providing a promising method for the efficient conversion of functional lipids.

Enabling Stable Recycling of L-Arabinose Isomerase Through Whole-Cell Immobilization for Efficient and Cost-Effective D-Tagatose Production

Zepeng Li,Runmin Wang,Xiantai Lai,Wenyi Liao,Runfeng Liao,Zhuohong Wu,Guoyan Zhang,Xianghui Qi

D-tagatose is a functional sweetener with glucose-regulating and prebiotic properties, but its bioproduction from D-galactose faces many limitations, particularly the high production costs. In particular, the current biosynthesis of D-tagatose suffers from thermal instability and the substrate selectivity issues of L-arabinose isomerase (L-AI) required to convert D-galactose into D-tagatose. In this study, recombinantEscherichia coliBW25113/pQE-80L-araAF118M/F279Iexpressing double mutant L-AI was immobilized to enhance its stability and reusability. The optimal conditions for whole-cell catalysis were 60 °C, pH 6.5, 5 mM Mn2+, and 20 h, with a yield of 55.2 g/L of D-tagatose. Immobilization with 3% sodium alginate and 2% CaCl2retained 90% of the production efficiency displayed by free cells. Notably, the immobilized cells exhibited enhanced heat resistance (60–70 °C) and operational stability, retaining 76% activity after five cycles. The D-tagatose production was further increased to 129.43 g/L by increasing the substrate concentration to 250 g/L. Compared to free cells, immobilized cells retained 83.6% of the initial yield up to 10 batches. This study presents a cost-effective and sustainable method for the production of D-tagatose using optimized whole-cell catalysis through immobilization, which paves the way to solve industrial challenges such as thermal instability and low substrate efficiency.

Amphiphilic molecular brush-grafted nanofiltration membranes via stable initiator immobilization: A dual defense strategy against complex fouling in leachate

Chengyu Li,Wentian Zhang,Huilin Li,Lichao Xia,Shangwei Zhang,Fangang Meng,Shanshan Zhao

Conventional nanofiltration (NF) membranes often encounter severe membrane fouling issue in advanced treatment of leachate, primarily due to the presence of complex foulants. Herein, we developed a novel dual-defense functionalized robust antifouling NF membrane through covalent grafting of initiator α -bromoisobutyric acid (Biba) onto a NF membrane surface followed by sequential grafting of zwitterionic polymer (PSBMA) and fluoropolymer (PHFBM) brushes via surface-initiated atom transfer radical polymerization (SI-ATRP). The chemical grafting of novel initiator Biba supplanted the previously reported toxic initiator and unstable polydopamine coating strategy, thereby facilitating robust covalent immobilization of antifouling functional layers. The modified membrane demonstrated exceptional antifouling capabilities during synthetic wastewater treatment, exhibiting a flux decline ratio (FDR) of only 12.4 % and a flux recovery ratio (FRR) of 94.7 %. Furthermore, it also showed remarkable durability during three cycles of filtration using actual leachate biological treatment effluent, consistently maintaining a FRR of 94.2 %. The molecular dynamics simulations, adhesion force tests, and free energy calculations elucidated the fouling resistance and fouling release mechanisms of the PSBMA and PHFBM brushes. This work provides a new initiator immobilization strategy for fabricating a robust fouling resistant NF membrane with a dual-defense functional polymer brushes coating layer. Meanwhile, this robust NF membrane exhibits substantial potential for sustainable leachate treatment applications.

Immobilization of phospholipase D on macroporous SiO2/cationic polymer nano-composited support for the highly efficient synthesis of phosphatidylserine

Jia-Qin Wu,Xiao-Mei Xu,Ding-Lin Wang,Neng-Bing Long,Rui-Feng Zhang

Novel nano-composites were prepared by coating epoxy resin-based cationic polymer in nano-thickness via in-situ curing on the nano-wall of macroporous SiO 2 with pore size of 0.5∼1 μm. By changing the thickness of polymer coating the specific surface area and porosity varied in range of 115∼74 m 2 /g and 90.4∼83.9 %, respectively. Through ion exchange phospholipase D (PLD, from Streptomyces sp ) was efficiently immobilized on the nano-composites as support and the immobilized PLD was applied for the highly efficient synthesis of phosphatidylserine (PS). The loading amount of PLD on the nano-composited support reached to a maximum of 90.2 mg/g support , 4 times as high as that on the pure macroporous silica. The specific activity of the immobilized PLD reached as high as 16,230 U/g protein , while that of free PLD was 18,780 U/g protein . Under a wide range of temperature and pH the stability and activity of the immobilized PLD were greatly improved as compared with the free ones. Under optimized conditions at 45 °C and pH 7.0, the PS yield reached as high as 96.2 % within 40 min. After 28 days storage the immobilized PLD retained 82.2 % of original activity, and after 12 cycling reuses it retained 79.3 % of PS yield, which indicated that the immobilized PLD exhibited good stability.

A novel alginate-assisted immobilization strategy for enhanced biofilm development and nitrogen removal in membrane aerated biofilm reactors (MABRs)

Abdallah Abdelfattah,Reham Eltawab,Ke Li,Zhong-Ting Hu,Liang Cheng

The prolonged startup period caused by slow and unstable biofilm formation remains a major barrier to large-scale application of membrane-aerated biofilm reactors (MABRs). This study developed a novel sodium alginate–assisted immobilization strategy to accelerate biofilm establishment and enhance nitrogen removal. A homogeneous mixture of sodium alginate and activated sludge was spray-coated onto plasma-treated PVDF hollow-fiber membranes, followed by CaCl₂ mist crosslinking to form a thin gel layer immobilizing active biomass. The coated MABR achieved 85 ± 5 % surface coverage within seven days—over twice that of the control—and complete ammonium oxidation (>90 %) in 10 days, reducing startup time by about 50 %. Digital microscopy and SEM revealed rapid transition from the synthetic alginate matrix to a self-sustained EPS network, while 16S rRNA sequencing confirmed enriched Nitrosomonas and Nitrospira with enhanced microbial diversity. The oxygen transfer rate initially decreased by 18 % after coating but recovered to 95 % of the original after partial scaffold dissolution, showing reversible mass-transfer adaptability. Mechanistically, the biodegradable Ca 2+ –alginate scaffold provided an initial attachment platform evolving into a stable stratified biofilm enabling simultaneous nitrification and denitrification. This study establishes a mechanistic and practical basis for biopolymer-assisted biofilm engineering toward rapid-startup, high-efficiency MABR systems.

Interlayer functionalization of silica nanosheets derived from natural clay for stable ionic liquids immobilization and efficient carbon dioxide capture

Xiaoyu Li,Ruihong Li,Haiyang Xue,Chenyu Wang,Kang Peng

Supported ionic liquids (ILs) within porous supports are efficient carbon dioxide (CO 2 ) adsorbents and has become a promising alternative to solid amine adsorbents owing to their negligible volatility, excellent chemical/thermal stability, and non-corrosive nature. In this work, we explore the confinement of ILs within the hierarchical nanochannels of laminated silica nanosheets derived from low-cost vermiculite (AEV) for efficient CO 2 capture. A series of AEV/ILs (ILs confined within AEV) composites were synthesized via scalable wet impregnation approach, and the effects of several key factors, including ILs species, IL loading (10–50 wt%), adsorption temperature (25–65 °C), CO 2 partial pressure, and gas flow rate (50–100 mL/min), were systematically evaluated. Characterization data indicated that the ILs were successfully incorporated within the laminated porous network of AEV, leading to an intercalated confinement structure. Under atmospheric pressure and 35 °C in a 40 % CO 2 /60 % N 2 flow, the AEV/BF 4 –20 adsorbent (20 wt% 1-butyl-3-methylimidazolium tetrafluoroborate ([BMIM][BF 4 ]) loading within AEV) achieved a CO 2 uptake of 1.28 mmol/g, combined with rapid adsorption kinetics and stable recyclability, showing only an average capacity loss of 0.78 % per cycle over long-term testing. The hierarchical porous structure of AEV appears to promote efficient ILs dispersion and enhance CO 2 diffusion, thereby improving CO 2 capture performances. Furthermore, interactions between [BMIM][BF 4 ] and AEV nanosheets, together with multiscale adsorption pathways (surface, interlayer, intraparticle), are considered to synergistically enhance the CO 2 adsorption behavior. These low-cost raw materials, uncomplicated synthesis process and highly adsorption activity make AEV/ILs composites possible for large-scale production, showing great potential for various practical CO 2 capture and separation processes.

Long-term stability of montmorillonite modified with ferrihydrite in freeze-thaw cycles: Enhanced heavy metal immobilization via crystalline transformation

Miao Jiang,Guangzhi Wang,Qingliang Zhao,Kun Wang,Ruiyuan Xue,Liangliang Wei,Junqiu Jiang,Jing Ding

The widespread industrial application of As, Sb, and Pb has resulted in frequent co-contamination of soils, raising significant environmental and public health concerns. Our previous studies demonstrated that amorphous ferrihydrite-modified montmorillonite (amFe@Mont) possesses excellent stabilization capacity; however, its long-term performance under freeze–thaw (F–T) stress has not been investigated. Herein, the stability of amFe@Mont-remediated As/Sb/Pb-contaminated soil was systematically evaluated under 120 repeated F–T cycles using TCLP, SBET, and DTPA extraction methods. Even after 120 cycles, amFe@Mont maintained high immobilization efficiencies of 93.06 % for As, 84.22 % for Sb, and 87.59 % for Pb. Although F–T cycles increased the bioavailability of these metals in untreated soil, amendment with amFe@Mont significantly reduced their extractability. The amendment also modulated key soil properties, including pH, electrical conductivity, redox potential, and organic carbon content. Microbial community analysis revealed adaptive shifts toward heavy metal-resistant and cold-tolerant taxa. Mechanistically, silicon doping from montmorillonite inhibited Fe crystallization, enabling amFe@Mont to retain poorly crystalline Fe oxides that incorporated As and Sb into Fe–O lattices, while Pb immobilization occurred primarily via surface complexation enhanced by Fe oxide transformation. The enrichment of resistant genera such as Ralstonia and cold-tolerant taxa such as Mucilaginibacter further reinforced contaminant stabilization. Collectively, this study provides the first evidence of dual chemical–microbial resilience in Fe–clay composites under F–T stress, offering a promising strategy for the remediation of co-contaminated soils in cold regions.

Bimetallic metal–organic framework for enzyme immobilization by biomimetic mineralization: Constructing a mimic enzyme and simultaneously immobilizing natural enzymes

Junning Wang,Mengyuan Bao,Tianxiang Wei,Zhaoyin Wang,Zhihui Dai

An iron-nickel bimetallic metal−organic framework (FeNi-MOF) with inner peroxidase-like activity as a support for enzyme immobilization was synthesized for the first time. And the GOx-mediated synthesis of mimic multienzyme system (GOx/FeNi-MOF) was obtained by a one-step biomimetic mineralization . FeNi-MOF played a dual role of a peroxidase mimic and a protective coating. Therefore, the obtained multienzyme system showed both peroxidase-like activity of FeNi-MOF and the biological activity of natural enzyme . FeNi-MOF acted as a peroxidase mimic, showing a higher affinity for hydrogen peroxide (H 2 O 2 ). The enhanced catalytic activity of the FeNi-MOF may result from synergistic effect between iron and nickel. In addition, the GOx/FeNi-MOF was selected as a proof of concept and succeeded in a one-step colorimetric detection of glucose by tandem catalysis. A small amount of the product was used for glucose detection, and the detection range was 0.3–35 mM, which indicated that our sensor can meet the clinical needs for diagnose diabetes.

Microbial investigations of new hydrogel-biochar composites as soil amendments for simultaneous nitrogen-use improvement and heavy metal immobilization

Lixun Zhang,Yuntao Guan

Agricultural sustainability is challenging because of increasingly serious and co-existing issues, e.g., poor nitrogen-fertilizer use and heavy metal pollution. Herein, we introduced a new poly(acrylic acid)-grafted chitosan and biochar composite (PAA/CTS/BC) for soil amendment, and provided a first microbial insight into how PAA/CTS/BC amendment simultaneously improved nitrogen cycling and immobilized heavy metals. Our results suggest that the PAA/CTS/BC amendment significantly promoted soil ammonium retention, and reduced nitrate accumulation, nitrous oxide emission and ammonia volatilization during the rice cultivation. The availability of various heavy metals (Fe, Mn, Cu, Zn, Ni, Pb, Cr, and As) markedly decreased in the PAA/CTS/BC amended soil, thereby reducing their accumulation in rice root. The PAA/CTS/BC amendment significantly altered the structure and function of soil microbial communities . Importantly, the co-occurrence networks of microbial communities became more complex and function-specific after PAA/CTS/BC addition. For example, the keystone species related to organic matter degradation, denitrification, and plant resistance to pathogen or stresses were enriched within the network. In addition to direct adsorption, the effects of PAA/CTS/BC on shaping microbial communities played dominant roles in the soil amendment. Our findings provide a promising strategy of simultaneous nitrogen-use improvement and heavy metal immobilization for achieving crop production improvement, pollution control , and climate change mitigation .

Convenient Immobilization of α-L-Rhamnosidase on Cerium-based Metal-Organic Frameworks Nanoparticles for Enhanced Enzymatic Activity and Recyclability

Lingling Peng,Wansen Tan,Yuting Lu,Ayan Yao,Dayuan Zheng,Le Li,Jingran Xiao,Lijun Li,Qingbiao Li,Shu-feng Zhou,Guowu Zhan

Graphical Immobilized Rha on MOFs : Herein, we explored five typical metal-organic frameworks (MOFs) as porous carriers to immobilize Rha, and the obtained Rha@Ce-BTC showed the highest enzymatic activity. Also, Rha@Ce-BTC manifested enhanced enzyme-substrate affinity, great reusability/storage stability, and catalytic efficiency compared to free Rha. Significantly, the as-prepared Rha@Ce-BTC might be assembled as a membrane reactor for efficient biocatalysis reactions. The α-L-Rhamnosidase (Rha) is a useful glycoside hydrolase for selectively hydrolyzing the terminal L-rhamnose residues in flavonoids, being vital to food and pharmaceutical industries. However, Rha suffers from low recyclability and poor stability in harsh environments. Herein, we explored five typical metal-organic frameworks (MOFs) as porous carriers to immobilize Rha, and the activities of the resultant Rha@MOF composites were compared with the free enzyme. The locations of the enzyme in MOFs were proved by a series of characterization techniques. It was found that Rha@Ce-BTC (with enzyme immobilization efficiency of 23 % and enzyme loading content of 8.8 %) showed the highest enzymatic activity. The immobilized Rha@Ce-BTC showed 80 % residual activity after five consecutive cycles, suggesting a limited leaching effect. Also, Rha@Ce-BTC manifested markedly enhanced enzyme-substrate affinity and catalytic efficiency compared to free Rha, as supported by Michaelis-Menten kinetic studies. Accordingly, the Ce-BTC would be an appealing carrier for enzyme immobilization, and the as-designed enzyme/Ce-BTC composites are promising candidates for industrial use with remarkably high activity, recyclability, and storage stability.
