Fluorinated Hypoxia-Responsive Aza-BODIPY for NIR-II FL/19F MR/PA Imaging and Phototherapy of Lung Cancer

Anfeng Li,Fang Wang,Mou Jiang,Yu Li,Xin Zhou,Zhong-Xing Jiang

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.

NIR/TME-responsive release of zoledronic acid-loaded LDHs on ZE21C magnesium alloy with multi-therapy for osteosarcoma

Dong Li,Tong Zhang,Shaosheng Jia,Jianan Yu,Rongbang Sun,Bin Yin,Liguo Wang,Lingchuang Bai,Lan Chen,Shaokang Guan

Osteosarcoma (OS) poses a severe threat to human health, and the standard clinical treatment involves prosthetic replacement after surgical removal combined with radiotherapy and chemotherapy. However, the recurrence of residual tumors and extensive bone defects leads to a poor prognosis for patients. Zoledronic acid (ZA) is clinically used for tumor treatment and osteoclast inhibition. Nevertheless, intravenous infusion of ZA has poor targeting and significant toxicity to normal tissues. Biodegradable Mg alloys have excellent mechanical and biological compatibility, making them ideal for orthopedic implants. However, their application is limited due to poor corrosion resistance. This work applied a multi-composite layered double hydroxide (LDH)-ZA/PDA coating to the surface of ZE21C alloy. The composite coating adsorbed ZA in weakly alkaline or neutral conditions and released ZA under near-infrared (NIR) irradiation in the tumor microenvironment (TME). The stimuli-responsive release of ZA was confirmed to be related to the NIR-TME-triggered change in LDH interlayer spacing. In vitro and in vivo experiments showed that this multi-composite coating improved corrosion resistance, responded to TME and NIR stimuli to release ZA and reactive oxygen species, effectively targeting and killing tumor cells and osteoclasts through the AKT/GSK-3β signaling pathway. It also transformed OS from “cold tumors” into “hot tumors”, reactivated macrophage-mediated tumor immunity, and promoted osteogenesis. The chemo-thermal-therapeutic approach of the composite coating achieves a “three-birds-with-one-stone” effect. This study offers new insights into applying Mg alloys in prosthetic replacements after OS surgery.

Engineering of 2D Ultrathin CuS-PMA Heterostructures for Enhanced NIR-II Photothermal Therapy Facilitating Oxidative Stress and Immunotherapy

Dong Wang,Feng Yuan,Xuliang Deng,Haoyang Li,Siyang Nie,Qingda Liu,Xun Wang

Therapy based on heat and reactive oxygen species (ROS) generation have attracted increasing attention, while the weak photothermal ability and low ROS production have severely limited the efficiency of the tumor treatment. Here, 2D ultrathin CuS-phosphomolybdic acid (PMA) nanosheets are synthesized at sub-nanoscale. Typically, the improved second near-infrared light (NIR-II) light absorption, small bandgap, fast hot carrier response, abundant oxygen vacancies, and the electron delocalization between closely connected PMA and CuS units in sub-nanoscale result in the notably enhanced NIR-II photothermal therapy (PTT) performance of CuS-PMA. The reversible redox of Mo active sites in PMA provides basis for glutathione (GSH) depletion while triggering chemodynamic therapy (CDT) in the tumor microenvironment (TME), and the heat produced by the PTT greatly promotes the CDT and GSH depletion, where CuS-PMA presents sevenfold more ROS generation and fourfold more GSH depletion under PTT. In addition, the ROS generation/GSH depletion can downregulate heat shock proteins, weakening the heat resistance of tumor cells and thereby amplifying the killing effect of PTT. Moreover, the combination of antiprogrammed death-ligand 1 (αPD-L1) with the treatment of CuS-PMA promote the immunotherapy, resulting in the suppression of untreated abscopal tumors.

Au nanostars-assembled PVA/PEI nanofibers as a photothermal catalyst for enhanced NIR-induced reduction of 4-nitrophenol

Yujiao Zou,Bo Ge,Yanhui Hu,Yulu Yang,Ni Li,Yili Zhao

The high catalytic performance induced by the photothermal conversion effect holds immense potential in environmental purification, attracting widespread attention. Herein, the synthesis of PVA/PEI/Au nanofibers was successfully achieved, and they were applied as plasmonic photothermal catalysts to facilitate the reduction of 4-nitrophenol (4-NP). The photothermal performance of nanofibers loaded with different concentrations of Au nanostars (NSs) was compared under near-infrared (NIR) laser irradiation. Remarkably, at low laser power densities under NIR irradiation, the PVA/PEI/Au nanofibers catalyzed the reduction of 4-NP, resulting in a 1.5-fold increase in reaction rate and achieving a 98% conversion efficiency within 8 min. Additionally, the nanofibers demonstrated excellent recyclability, maintaining a conversion efficiency exceeding 97% over six consecutive cycles. The PVA/PEI/Au nanofibers hold significant promise for the practical application of electrospun nanofibers in the photothermal degradation of environmental pollutants.

Construction of Chitosan-Coated TiO2/In2S3 Heterojunction on Ti surfaces for NIR Triggered Photothermal/photodynamic Synergistic Enhanced Antibacterial

Peixin Dong,Yangkai Liu,Fulin Deng,Anning Zhao,Min Ren,Linjie Song,Yuanzhen Zhou,Yumeng Dong,Yunjie Xiang,Ugo D‘ Amora,Liqun Xu,Xi Rao

In 2 S 3 forms a Z-scheme heterojunction with TiO 2 , broadening the light response range, improving ROS generation. • Photothermal-photodynamic synergy reduces the required photothermal temperature. • The TiO 2 / In 2 S 3 /Chitosan composite coating exhibits enhanced antibacterial activity. • The composite coating exhibits excellent biocompatibility. Implant-associated bacterial infections remain a major challenge in orthopedic and dental applications, often compromising implant integration and longevity. For this reason, the synergistic antibacterial strategy of photothermal and photocatalysis has become one of the important methods in the field of antibacterial. To synthesize high-efficiency near-infrared (NIR) light-responsive catalysts, a titanium oxide/indium sulfide (TiO 2 /In 2 S 3 ) heterojunction antibacterial coating was prepared on the titanium-based surface using anodic oxidation and wet chemical deposition methods. Compared with titanium alloy, the TiO 2 /In 2 S 3 heterojunction showed a suitable energy band structure, improved light absorption, and enhanced carrier separation capability, thus enhancing photothermal catalytic activity and promoting the production of bacterial reactive oxygen species under NIR light. Indeed, the TiO 2 /In 2 S 3 heterojunction narrowed the bandgap (~2.5 eV) and efficiently generated reactive oxygen species (•O₂⁻ and •OH) under NIR irradiation. Furthermore, the successful deposition of chitosan (CS) endowed the TiO 2 /In 2 S 3 heterojunction with osteogenic activity, promoting the proliferation and differentiation of osteoblasts. In vitro , results demonstrated effective antibacterial performance against Escherichia coli and S taphylococcus. aureus , inhibited biofilm formation, and supported collagen secretion and mineralization. In vivo , NIR irradiation raised the implant site temperature to below 50   °C, enabling safe and efficient sterilization, and showing no organ toxicity. These results highlight CS-coated TiO 2 /In 2 S 3 heterojunction as a promising multifunctional coating that combines infection control with osteogenic support, providing a potential strategy for improving the safety and longevity of titanium-based implants. Pixel-Registered Trimodal Visible/NIR/X-Ray Imaging Enabled by a Single-Material Rare-Earth-Free Double Perovskite

Qiming Huang,Jialiang Jiang,Hui Fu,Xi Chen,Hui Peng,Kanybek Osmonaliev,Samat Toktogonov,Jinju Zheng,Jialong Zhao,Weiyou Yang

Multimodal imaging that integrates x-ray, white light, and near-infrared (NIR) information offers enhanced diagnostic accuracy. However, the absence of material that can efficiently respond to all three types of signals requires the use of separate hardware for each modality, inevitably causing pixel misalignment and complicating system integration. To address this, we develop a Te 4+ /Mo 4+ co-doped Cs 2 HfCl 6 perovskite microcrystals (MCs) platform. Rational dopant engineering yields an optimized composition (3%Te 4+ , 5%Mo 4+ ) that simultaneously emits in the visible (570 nm, PLQY  =  17.6%, from Te 4+ self-trapped excitons) and NIR (910 nm, PLQY  =  41.7%, from Mo 4+ d-d transitions) regions. The high-Z host also ensures strong x-ray absorption, leading to exceptional x-ray scintillation performance with a light yield of 35 073 photons/MeV, a detection limit of 81.43 nGy air /s, and a spatial resolution of 13.6 lp/mm. The resulting flexible PDMS composite films function concurrently as white-light and NIR-emitting diodes, as well as high-resolution x-ray screens. Using a single detector with switchable excitation (450 nm LED or x-rays), we demonstrated pixel-registered trimodal imaging of a screw-ink-capsule phantom without the need for post-processing alignment. This rare-earth-free, all-in-one material strategy provides a compact and scalable route toward next-generation multimodal medical diagnostics.

A Photosensitizer-Engineered Nanoplatform Exposed to NIR Laser for Mitochondria Targeting and Chemo/Photodynamic Synergistic Therapy against Glioma

Fengtian Zhang,Weihong Luo,Zhongjie Min,Darong Lu,Weiliang Chen,Lijuan Wen

Targeted mitochondrial-based therapeutic strategies for tumors have emerged as a significant research focus in the field of cancer treatment in recent years. Mitochondria, often referred to as the “energy factory” of cells, play a crucial role in the metabolic reprogramming of tumor cells, as well as in oxidative stress and immune evasion. In this study, we utilized a mitochondria-targeted photosensitizer, IR780, to engineer Celastrol (Cela)-loaded lipid nanoparticles, designed as Cela@Lip-IRH, for the synergistic therapy of chemotherapy and photodynamic therapy (PDT) against glioma. Cela@Lip-IRH could respond to the weakly alkaline environment of tumor cell mitochondria for rapid Cela release. Leveraging the mitochondrial targeting capacity and PDT property of IR780, Cela@Lip-IRH nanoparticles showed enhanced cellular uptake and mitochondria targeting distribution in vitro, as well as obvious tumor accumulation in vivo. Cela@Lip-IRH nanoparticles exhibited prominent intracellular reactive oxygen species (ROS) generation upon NIR laser irradiation, which was accompanied by pronounced photocytotoxicity and cell apoptosis, finally producing remarkable tumor growth suppression efficacy in vivo. We further verified that the potential antitumor molecular mechanism was triggered by the release of Cytochrome C, leading to an activated mitochondrial apoptosis pathway, alongside an immune response characterized by enhanced recruitment of CD4+ T cells and CD8+ T cells in tumor tissues. Currently, this mitochondria-targeted nanoplatform presents an ideal method to enhance the synergistic performance of chemotherapy and PDT in glioma, and it is anticipated that such a strategy will provide a promising alternative to optimize the tumor-targeted therapies.

Efficient broadband near-infrared luminescence of Cr3+ doped fluoride K2NaInF6 and its NIR-LED application toward veins imaging

Zixuan Wu,Xinxin Han,Yayun Zhou,Ke Xing,Sheng Cao,Li Chen,Ruosheng Zeng,Jialong Zhao,Bingsuo Zou

High performance broadband Near-infrared (NIR) phosphors with high efficiency and excellent thermal stability are highly demanded for constructing the next generation of NIR radiation sources. Herein, a broadband NIR emitting fluoride phosphor K 2 NaInF 6 :Cr 3+ peaking at 774 nm with a Full width at half maximum (FWHM) of 116 nm (1906 cm −1 ) was demonstrated. A high Internal quantum efficiency (IQE) value of 70.2% can be obtained in the as-synthesized K 2 NaInF 6 :Cr 3+ phosphor. Additionally, benefiting from the relatively weak electron phonon coupling effect, the phosphor also exhibits excellent anti-thermal quenching behavior with the emission intensity at 150 °C retaining 78.3% of that at room temperature. A high power NIR pc-LED with NIR radiant flux of 492.85 mW@1050 mA is obtained by combing the as-synthesized K 2 NaInF 6 :Cr 3+ phosphor and a blue LED chip. Utilizing this high power NIR pc-LED as NIR radiation source, high-resolution human hand veins imaging is realized with the assistance of a NIR camera, which indicates that K 2 NaInF 6 :Cr 3+ phosphor has high potential in biological applications.

3D printed core-shell hydrogel fiber scaffolds with NIR-triggered drug release for localized therapy of breast cancer

Xiaoyue Wei,Chunyang Liu,Zhiyong Wang,Yongxiang Luo

Localized therapy using hydrogels-based drug delivery system (DDS) is a promising strategy for the treatment of diseases such as cancer in superficial tissues. In this study, we presented a facile method to prepare core-shell hydrogel fibers/scaffolds with controlled drug delivery and designed structures for the treatment of the residual breast cancer and prevention of local recurrence after surgery. Mixtures of polydopamine (PDA) and concentrated alginate inks (15.3 wt%) as the shell layer, and drug-loaded temperature-sensitive hydrogels as the core part were co-injected and coaxial 3D printed into core-shell hydrogel fibers and scaffolds. Under near infrared (NIR) irradiation, PDA with excellent photothermal effect could raise the temperature of core-shell fibers, which induced the gel-sol transition of the core gels, and subsequently resulted in the drug release from the loosened hydrogel network. The photothermal effect and the released drugs could eliminate cancer effectively. Thus, our prepared core/shell fibers and scaffolds with NIR-triggered on-demand drug release would be promising candidates to fill the cavity of breast tissues after surgical resection of cancer achieving a therapeutic effect for the residual and recurred cancer.

Double pH-sensitive nanotheranostics of polypeptide nanoparticle encapsulated BODIPY with both NIR activated fluorescence and enhanced photodynamic therapy

Huiping Dang,Quan Cheng,Youliang Tian,Changchang Teng,Kai Xie,Lifeng Yan

To achieve accurate fluorescence imaging-guided cancer therapy, intelligent systems with specific responsiveness to the tumor microenvironment need to be designed. Here, we have achieved both enhanced NIR fluorescence and photodynamic therapy by introducing a dimethylamino functional group in BODIPY dyes, which can be used as a pH sensor under acidic conditions by coordinating with the proton. At pH 7.4, the fluorescence is quenched due to the photo-induced electron transfer (PET) process. After the photosensitizer is protonated in tumor cell lysosomes (pH 4.0–5.5), the PET process is inhibited and the fluorophore emission capacity is restored (fluorescence enhancement up to 10-fold), resulting in near-infrared fluorescence with the OFF/ON transition inside the tumor and enhanced singlet oxygen production for lysosome targeting capability. Due to the substitution of heavy atom iodine, the compound has a high singlet oxygen quantum yield of 81.8% in dichloromethane. In addition, using a pH-sensitive amphiphilic polypeptide (POEGMA23-PE9) as a carrier to wrap the photosensitizer BDPI can release enough drug in the acidic environment (pH 5.5–6.5) of intracellular endosomes/lysosomes, which is conducive to more adequate interactions of the photosensitizer with H+ and more effective enhancement of fluorescence emission and 1O2 production, achieving precise fluorescence imaging capability and extremely low background toxicity.

Fabrication of Dissolving Microneedles with Thermal-Responsive Coating for NIR-Triggered Transdermal Delivery of Metformin on Diabetic Rats

Depeng Liu,Yang Zhang,Guohua Jiang,Weijiang Yu,Bin Xu,Jiangyin Zhu

This study described near-infrared (NIR) light-responsive polymer-nanodot composite microneedles (MNs) used for on-demand transdermal drug delivery. Bismuth (Bi) nanodots stabilized by poly(vinylpyrrolidone) (PVP) as a photothermal conversion agent and metformin as an antidiabetic drug were introduced into the dissolving MNs coated with lauric acid (LA). When the MNs were irradiated with NIR light, light-to-heat transduction induced by the Bi nanodots caused the LA to melt. As a result, the polymer matrix was dissolved after absorbing the interstitial fluid, enabling the encapsulated metformin release from the MNs into skin tissue. Compared with subcutaneous injection of metformin, the administration using the Bi nanodots-induced NIR responsive MNs developed in the current research exhibited a remarkable hypoglycemic effect in vivo. This work indicates that the as-fabricated Bi nanodot-induced NIR responsive and LA-coated MNs have potential applications in diabetes treatment. Additionally, these artificial MNs also present a promising platform for delivering other therapeutic drugs.

Microenvironment-programmed self-activating cascaded chemodynamic antibacterial packaging film with NIR-regulated catalytic activity for prolonged food preservation

Yiya Ping,Liang Zhang,Mengyi Wang,Bingzhi Li,Linpin Luo,Wenze Wang,Yuechun Li,Wentao Zhang,Mingqiang Zhu,Jianlong Wang

In this study, a nano-platform driven by the microenvironment and photo-controlled catalytic activity was meticulously designed, and a CaO 2 @CAFe functional filler was constructed by using chlorogenic acid as a phenolic ligand and ferric ion to co-encapsulate calcium peroxide, creating an antibacterial packaging system (CCF-CS) with in situ self-activation cascade catalytic activity and intelligent enhancement effect of near-infrared light response. In this system, the acidic microenvironment provided by the chitosan (CS) film matrix triggers the CaO 2 @CAFe particles to continuously release H 2 O 2 , the catalytic substrate in which peroxidase plays a role, and "substrate self-supply" is achieved through a cascade reaction. Moreover, the catalytic efficiency of the nanozymes can be positively modulated by the "smart switch" of near-infrared light, enabling a dual-mode antibacterial behavior of "on-demand burst and routine protection". The CCF 0.20 -CS film was 99.9 % effective in killing both bacteria and fungi, and significantly extended the shelf life of cherry tomatoes by 10 days. Furthermore, the film exhibits enhanced UV-shielding capability and improved mechanical strength, alongside validated biosafety. These optimal properties underscore the innovative potential and practical superiority of the CCF 0.20 -CS packaging system in addressing microbial contamination challenges in food preservation.
