Large-area three-dimensional (3D) well-ordered Co(OH)2 nanoflower array films decorated with Ag nanoparticles (Ag NPs) with a novel structure were designed as sensitive SERS substrates for trace detection of pesticides. Co(OH)2 nanoflowers were electrodeposited on the surface of molded polyacrylonitrile (PAN) films with ordered nanopillar arrays and then Ag NPs were sputtered onto of the surface of each Co(OH)2 nanoflower. The SERS performance of the as-prepared films can be optimized by tuning the Ag-sputtering duration and the morphology of the Co(OH)2 nanoflower arrays. Owing to the formation of high-density SERS “hotspots” created in the nanogaps between the adjacent Ag NPs located on the same and neighbouring Co(OH)2 nanosheets, the optimized substrates show SERS activity for probe molecules. As a result, 10−11 M rhodamine 6G and 10−9 M 4-aminothiophenol can be SERS recognized, respectively. To verify their practical application, the optimized SERS substrates were employed for trace detection of thiram (a type of pesticide), and 10−9 M thiram can be recognized, showing great potential in pesticide detection.

Developing a rapid, low cost and sensitive sensing strategy for undifferentiated detection and fast killing of bacterial pathogens are critical to alleviating bacteria infections. Here, we propose a direct photoreduction method to synthesize the SERS tag by integrating poly(4-cyanostyrene) nanoparticles (NPs) and silver ions , which are applied as bio-sensing system for bacteria sensing and fast killing. Under a focused laser spot, silver ions on the surface of the poly(4-cyanostyrene) NPs could be photoreduced into Ag NPs, thereby causing the Raman signal amplification of poly(4-cyanostyrene) NPs up to 40 times, and there is a good linear correlation between the Raman intensity of poly(4-cyanostyrene) NPs and different concentrations of Ag + . Moreover, 4-mercaptophenylboronic acid, performing the same recognition function for both the Gram-positive and Gram-negative bacteria, is used as bridge between the bacteria and Ag + by the inherent chemical bonding. Based on further constructed bio-sensing system, we achieved the quick count and killing of both Gram-positive bacteria, e.g. , Staphylococcus aureus ( S. aureus ), and Gram-negative bacteria, e.g. , Escherichia coli ( E. coli ). Notably, the sensing strategy can detect at least ∼100 cells of E. coli , ∼10 cells of S. aureus and ∼10 cells of their mixture in less than 40 min. The detection accuracy for actual samples can also reach over 80% and the bacteria were entirely killed by Ag + after the detection, avoiding bacterial contamination in the environment. This novel method is anticipated to perform as a simple yet effective tool for fast and sensitive bacteria counting and killing.

Some noble metal colloids with negative charges were mostly used as surface-enhanced Raman scattering (SERS) substrates, that the Raman signals of negatively charged molecules are hard to be detected because of the electrostatic repulsion among them and the negatively charged metal nanoparticles . Herein, Ag nanoparticles (Ag NPs) with positive charges and its Ag nanofilm were prepared by poly dimethyl diallyl ammonium chloride (PDDA) to realize the SERS detection of negatively charged molecules directly. The zeta potential and its particle sizes were characterized to select the optimum condition of PDDA. In addition, the morphology of positive Ag NPs and Ag nanofilm was characterized by UV–Vis spectroscopy and transmission electron microscopy . The SERS effect of negatively charged thiamphenicol on positively charged Ag nanofilm was significantly enhanced at the optimum condition and its sensitivity was significantly improved compared with normal Ag nanofilm. The proposed novel positively charged Ag nanofilm can be further applied to detect other negatively charged molecules by SERS.

Circulating tumor cells (CTCs) can be the seeds of tumor metastasis and are closely linked to cancer-related death. Fast and effective detection of CTCs is important for the early diagnosis of cancer and the evaluation of micrometastasis. However, the extreme rarity and heterogeneity of CTCs in peripheral blood make sensitive detection of CTCs a big challenge. In this paper, a TiO2-based surface-enhanced Raman scattering (SERS) bioprobe is reported for the first time with outstanding ultrasensitive specificity, excellent stability of the signal, and good biocompatibility for the detection of CTCs. The TiO2 NPs were encoded with alizarin red (AR) and functionalized with reduced bovine serum protein (rBSA) and folic acid (FA). The limit of detection (LOD) for 4-mercaptobenzoic acid (4-MBA) and AR molecules adsorbed on the TiO2 SERS substrate is 5 × 10−7 M. The designed TiO2-based SERS bioprobe can be effectively utilized in detecting four diverse types of cancer cells in rabbit blood, which shows good sensitivity of the SERS detection technology. Finally, precise targeting of CTCs based on the SERS bioprobe with the function of fluorescence imaging is also confirmed by the fluorescence colouration test. This work offers a novel strategy for CTC detection and the development of non-noble metal semiconductor-based SERS platforms for tumor diagnosis.

The integration of surface-enhanced Raman scattering (SERS) spectroscopy with magnetic fluid provides significant utility in point-of-care (POC) testing applications. Bifunctional magnetic–plasmonic composites have been widely employed as SERS substrates. In this study, a simple and cost-effective approach was developed to synthesize magnetic–plasmonic SERS substrates by decorating silver nanoparticles onto magnetic Fe3O4nanoparticles (AgMNPs), which function both as SERS-active substrates and magnetic fluid particles. The strong magnetic responsivity from AgMNPs can isolate, concentrate, and detect target analytes from the irregular surface of fish skin rapidly. We fabricate a microfluid chip with three sample reservoirs that confine AgMNPs into ever smaller volumes under an applied magnetic field, which enhances the SERS signal and improves the detection limit by two orders of magnitude. The magnetic fluid POC sensor successfully detected malachite green from fish with excellent selectivity and high sensitivity down to the picomolar level. This work achieves a label-free, non-destructive optical sensing approach with promising potential for the detection of various harmful ingredients in food or the environment. Keywords:magnetic fluid;surface-enhanced Raman scattering;POC sensor;AgMNPs;pesticides

The spread of COVID-19 over the past three years is largely due to the continuous mutation of the virus, which has significantly impeded global efforts to prevent and control this epidemic. Specifically, mutations in the amino acid sequence of the surface spike (S) protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have directly impacted its biological functions, leading to enhanced transmission and triggering an immune escape effect. Therefore, prompt identification of these mutations is crucial for formulating targeted treatment plans and implementing precise prevention and control measures. In this study, the label-free surface-enhanced Raman scattering (SERS) technology combined with machine learning (ML) algorithms provide a potential solution for accurate identification of SARS-CoV-2 variants. We establish a SERS spectral database of SARS-CoV-2 variants and demonstrate that a diagnostic classifier using a logistic regression (LR) algorithm can provide accurate results within 10 min. Our classifier achieves 100% accuracy for Beta (B.1.351/501Y.V2), Delta (B.1.617), Wuhan (COVID-19) and Omicron (BA.1) variants. In addition, our method achieves 100% accuracy in blind tests of positive and negative human nasal swabs based on the LR model. This method enables detection and classification of variants in complex biological samples. Therefore, ML-based SERS technology is expected to accurately discriminate various SARS-CoV-2 variants and may be used for rapid diagnosis and therapeutic decision-making.

Heavy metal ions, which are over-emitted from industrial production, pose a major threat to the ecological environment and human beings. Among the present detection technologies, achieving rapid and on-site detection of contaminants remains a challenge. Herein, capillaries with three-dimensional (3D) hot spot constructures are fabricated to achieve repaid and ultrasensitive mercury ion (Hg2+) detection in water based on surface-enhanced Raman scattering (SERS). The 4-mercapto pyridine (4-Mpy) serves as the Raman reporter with high selectivity, enabling the detection of Hg2+ by changes in adsorption configuration at the trace level. Under optimized conditions, the SERS response of 4-Mpy for Hg2+ exhibits good linearity, ranging from 1 pM to 0.1 μM in a few minutes, and the detection limit of 0.2 pM is much lower than the maximum Hg2+ concentration of 10 nM allowed in drinking water, as defined by the US Environmental Protection Agency (EPA). Simultaneously, combined with the theoretical simulation and experimental results, the above results indicate that the SERS substrates possess outstanding performances in specificity, recovery rate and stability, which may hold great potential for achieving rapid and on-site environmental pollutant detection using a portable Raman spectrometer.

Accurate detection of natural dyes in archaeological textiles is challenging due to limited sample availability. In situ detection of fibers by surface-enhanced Raman scattering (SERS) can overcome this challenge. In this study, we present two methods for the preparation of SERS substrates based on the laser photoreduction reaction and the galvanic displacement reaction, respectively. Our results show that the substrates prepared by these methods, including 450 nm-AgNPs, Cu-based Ag, and Zn-based Ag, offer several advantages such as rapid preparation, easy handling, low cost, high sensitivity and excellent reproducibility. Using these substrates, we successfully detected anthraquinones (alizarin, purpurin), indigoids (indigotin, indirubin), and protoberberine-type dyes in archaeological textiles from the Wei and Jin Dynasties. Notably, we were able to detect both indigo and protoberberine-type dyes in one archaeological fiber. The minimally invasive, in situ analytical approach developed provides a valuable tool for analytical applications where sampling is limited to microscopic amounts.

To achieve high sensitivity and uniformity simultaneously in a surface-enhanced Raman scattering (SERS) substrate, this paper presents the preparation of a flexible and transparent three-dimensional (3D) ordered hemispherical array polydimethylsiloxane (PDMS) film. This is achieved by self-assembling a single-layer polystyrene (PS) microsphere array on a silicon substrate. The liquid-liquid interface method is then used to transfer Ag nanoparticles onto the PDMS film, which includes open nanocavity arrays created by etching the PS microsphere array. An open nanocavity assistant soft SERS sample, “Ag@PDMS,” is then prepared. For electromagnetic simulation of our sample, we utilized Comsol software. It has been experimentally confirmed that the Ag@PDMS substrate with silver particles of 50 nm in size is capable of achieving the largest localized electromagnetic hot spots in space. The optimal sample, Ag@PDMS, exhibits ultra-high sensitivity towards Rhodamine 6 G (R6G) probe molecules, with a limit of detection (LOD) of 10−15 mol/L, and an enhancement factor (EF) of ∼1012. Additionally, the substrate exhibits a highly uniform signal intensity for probe molecules, with a relative standard deviation (RSD) of approximately 6.86%. Moreover, it is capable of detecting multiple molecules and can perform real detection on non-flat surfaces.

We present here a novel strategy for textile sample treatment, using galvanic displacement-based substrates, to identify anthraquinone dyes by surface enhanced Raman spectroscopy (SERS). This global procedure of SERS analysis, using several silver microparticles , in particularly those of dendritic shape, formed by reducing silver nitrate with zinc or copper, can be accomplished within 20 min. Compared to the synthesis of standard Lee-Meisel colloids, the preparation of silver dendrites provides a simple, fast and cost-effective route to produce reliable SERS substrates. The most fascinating feature is that the textile fibers analyzed are almost simultaneously covered with the substrates once the galvanic displacement reaction is triggered. This method was successfully applied to discriminating between alizarin and purpurin in a 6th-9th century samite with bird roundels, and carminic acid and laccaic acid in a 1740s chinoiserie brocade. However, as is the case with other silver substrates, the Raman signals of the insect dyes normally overlapped those of the plant dyes in the SERS profiles.

In this paper, a simple and cost-efficient strategy was used to construct a uniform Au NPs distribution on the surface of flexible filter paper for the detection of benzidine. Taking full advantage of the adsorption properties of filter paper, small gold nanoparticles were adsorbed onto its surface as gold seeds, and subsequently grown by electroless plating to form a highly uniform distribution of Au NPs substrates. By changing the electroless plating time, an optimal substrate was obtained. The as-prepared substrate exhibited satisfactory sensitivity with a low detection limit of 10−13 M for 4-ATP, and good reproducibility and homogeneity. Furthermore, the as-prepared substrates were successfully used for the detection of benzidine in environmental water, with a minimum detection concentration as low as 0.1 ppm and recoveries in the range of 92.4 to ∼108.5%. This study indicated that filter paper-based SERS substrates have great potential value in the detection of environmental organic pollutants.

Effective charge separation is essential in plasmon-mediated photochemistry and is usually achieved by constructing plasmon–semiconductor interfaces, which is usually challenging. In this work, by monitoring the plasmon-mediated silver oxidation with in situ Raman spectroscopy, we demonstrate that the adsorbed thiophenol molecules could modulate the rate of photochemical reactions by tuning the charge separation at the plasmon–molecule interfaces. It is found that the thiophenol molecules with strong electron-withdrawing or donating functional groups could accelerate or decelerate the rate of plasmon-mediated silver oxidation, respectively. Owing to the easy tuning of the electronic structures of organic molecules via substitution, our method provides a versatile and convenient approach for the fine modulation of plasmon-mediated photochemical reactions.
