Cervical cancer (CC) is one of the most prevalent cancers among women globally. The primary cause of CC is persistent infection with high-risk types of human papillomavirus (HPV), particularly HPV16, whose E7 oncoprotein plays a pivotal role in carcinogenesis and the maintenance of stem cell-like characteristics. RTKN2 participates in the progression of various cancers. However, the precise functions of RTKN2 in regulating CC remain unclear. The effects of HPV16 E7 in CC cells were evaluated using MTT, western blotting, Transwell, and sphere formation assays. Transcriptome sequencing and bioinformatics analyses were used to identify the targets of HPV16 E7. The expression levels of the target (RTKN2) in clinical samples were assessed using immunohistochemistry (IHC). The function and mechanism of RTKN2 in CC cells were investigated by the knockdown and overexpression approaches, as well as dual-luciferase reporter assay. HPV16 E7 exhibited a positive correlation on the malignant phenotype and stemness of CC cells. RTKN2 was identified as a target of HPV16 E7, and a reduction in its expression levels was caused by knockdown of HPV16 E7. The high expression of RTKN2 was associated with a poor prognosis in CC. HPV16 E7 may regulate RTKN2 expression by modulating the binding activity of E2F1 to the RTKN2 promoter. Upregulated RTKN2 activates the NF-κB signaling pathway, enhances the stemness of CC cells, and ultimately promotes malignant progression.

Huperzine A (Hup A), a reversible acetylcholinesterase inhibitor, shows protective potential against neurotoxic poisoning. Current formulations (oral tablets/capsules and injectables) face limitations: oral administration suffers from first-pass metabolism and fluctuating plasma concentrations due to frequent dosing, while injectables require sterile administration, reducing long-term compliance. The aim of this study was to prepare dissolving microneedles (MNs) patches containing Hup A for pretreatment against soman (GD) poisoning. Hyaluronic acid (HA), polyvinylpyrrolidone K30 (PVP K30) and sorbitol were selected as matrix materials in specific proportions and concentration ranges based on single-factor screening experiments and orthogonal design optimization. In order to further evaluate the potential of the Hup A MNs, optimized formulations were selected for subjected to characterization. This evaluation encompassed morphological observation, mechanical properties, skin penetration efficacy, recovery time, in vitro drug release study, permeability assays, pharmacokinetic study, and pharmacodynamic study. Statistical analyses were performed using Student's t-tests for pairwise comparisons and one-way ANOVA for multiple comparisons. The results demonstrated that prepared Hup A MNs had a conical shape with a smooth surface and sharp tips. Hup A MNs had sufficient mechanical strength (122.3 ± 15.3 N) to penetrate through the skin and could be quickly skin of recovery 6 min. In permeability assays, the cumulative Hup A release was about 35.4 μg at 2 h and 58 μg at 10 h. Pharmacodynamic results demonstrated that compared to oral administration, Hup A MNs reduced the onset time (30 min vs . 60 min) and extended the effective prevention duration against GD poisoning (6 h vs . 2 h). Pharmacokinetic analysis showed that MNS administration can maintain a more lasting and stable blood concentration as compared to oral administration. MNs are a valuable drug delivery system, benefiting the patients with minimal skin invasion. Taken together, these results demonstrated that Hup A-loaded MNs are acquiring a new alternative for pretreatment of OPNAs. Graphical

Textiles are prone to bacterial proliferation, which may lead to unpleasant odors, infection, and inflammation. To prepare antibacterial textiles, post-finishing is a convenient and scalable method. However, conventional post-finishing coatings often exhibit poor adhesion to polyethylene terephthalate (PET) textiles, limiting their loading efficiency. This study developed a quaternized chitosan/tannic acid (QACS/TA) bicomponent finishing agent to address this challenge. When the finishing agent contained 2 wt% QACS and 1 wt% TA, the post-finishing coating demonstrated optimized adhesion to PET, resulting in a weight gain of more than 15% for the treated PET textiles. The pyrogallol groups on TA could form abundant hydrogen bonds with ester groups of PET and the amino and hydroxyl groups of QACS, acting as binders between QACS and PET. The treated textiles showed an antibacterial rate of 99.2% against E.coli and almost 100% against S.aureus . This study presents a convenient and effective approach to preparing antibacterial PET textiles with improved coating adhesion.

Nasopharyngeal carcinoma (NPC) is one of the most prevalent cancers in the head and neck region. The development of innovative therapies that are effective at eliminating tumors while minimizing damage to surrounding normal tissues due to postoperative complications is crucial. To solve this problem, a gelatin-sodium alginate (Gel-SA) hydrogel containing bismuth selenide nanosheets (Bi 2 Se 3 NSs) has been developed, which introduces a “switch” of near-infrared (NIR) light for photodynamic therapy (PDT) and photothermal therapy (PTT) to significantly increase the level of oxidative stress while inducing ferroptosis and apoptosis to synergistically treat tumors. Once the treatment of the tumor has been completed, NSs remove a large number of free radicals left by NIR irradiation because of their excellent anti-inflammatory performance and combine with the rich nutrients in Gel-SA to protect and repair the tissues around the tumor. Combining Bi 2 Se 3 NSs and Gel-SA is an effective solution for treating NPC, as it ablates tumors while protecting surrounding tissues. Graphical abstract

Advanced Glycation End Products (AGEs) are the molecular mediators that contribute to the progression of diabetic complications. However, there is a paucity of research on effective AGEs inhibitors and strategies for blocking the AGEs-RAGE pathway. To solve the problem, we synthesized polymetformin (PL), which for the first time proved the ability to inhibit AGEs formation and block the AGEs-RAGE pathway to protect the vascular from being damaged. Based on gene ontology (GO) analysis and Kegg enrichment, we found that PL could antagonize AGEs. Molecular docking and dynamics analyses showed that PL formed stable structures with AGEs through electrostatic interactions and hydrogen bonding. PL treatment altered AGEs structures have been proven in FT-IR results. The study discovered that PL interacted with AGEs via both non-covalent and covalent modifications, altering AGEs’ binding sites and antagonizing the AGEs-RAGE pathway. Immunofluorescence assays indicated lower levels of RAGE, IL-1β, and TNFα, while ROS assays demonstrated lower ROS levels, highlighting PL’s inhibitory effects and biocompatibility. Our work underscores PL’s potential to treat diabetic complications by elucidating its mechanism of action against the AGEs-RAGE pathway and inflammatory factors for the first time. This research provides insights for managing chronic illnesses linked to the AGEs-RAGE pathway beyond diabetes complications.

Kidney stones are among the most common renal diseases with high global incidence and recurrence rates, with calcium oxalate (CaOx) stones constituting 65.9% to 80% of all cases. Minimally invasive surgery remains the primary approach for kidney stones, while limited progress has been made in the drug therapy for CaOx kidney stones over the recent years. The limitations of traditional drug therapy, such as renal toxicity and poor targeting, along with an elusive pathogenesis of CaOx kidney stones, have prevented its widespread clinical application. Renal tubular epithelial cells injury has been reported to play a crucial role in the occurrence and development of CaOx kidney stones. Nanozymes with potent antioxidant and anti-inflammatory properties have the potential to treat CaOx crystal-induced kidney injury. Moreover, as a key stone inhibitor, potassium citrate is widely used to inhibit stone formation due to its ability to modify urinary chemistry. Herein, we designed citrate-coated Prussian blue nanozyme hitchhiking on the neutrophils (NM@CPBzyme) with injured kidney targeting capability and good biosafety. The results showed that NM@CPBzyme alleviated CaOx crystal-induced kidney injury and CaOx crystals deposition. On the one hand, NM@CPBzyme has been demonstrated to not only suppress oxidative stress but also chelate calcium ions, thereby facilitating crystal dissolution. On the other hand, NM@CPBzyme could mitigate neutrophil infiltration, NETosis and inhibit pyroptosis in vitro and in vivo. In addition, RNA sequencing and bioinformatic analysis further showed that NM@CPBzyme ameliorates CaOx crystal-induced kidney injury via oxidative stress and neutrophil mediated inflammatory response. In summary, our results revealed that NM@CPBzyme is a novel strategy for protecting against CaOx crystal-induced kidney injury.

Spinal cord injury (SCI) initiates secondary injury cascades, including ferroptosis and neuroinflammation, which contribute to progressive neuronal and myelin loss. Single-cell RNA sequencing defines a therapeutically actionable window for selenium (Se) replenishment: neuronal and oligodendrocyte selenoproteins—especially Gpx4—show a transient rise at 1-day post-injury followed by sustained suppression with induction of ferroptosis drivers, indicating Se-limited antioxidant collapse. In this study, we extracted a novel Polygonatum -derived fructan and, for the first time, used it to coat selenium nanoparticles, synthesizing PRP@SeNPs via a green, ascorbate-mediated reduction. The PRP coating yields smaller hydrodynamic size, a more negative zeta potential, and a front-loaded yet sustained Se-release profile that aligns with the scRNA-seq–identified supplementation window. In vitro, PRP@SeNPs restore Gpx4 expression, reduce lipid peroxidation, scavenge ROS, and promote M2 microglial polarization. In situ administration in a T-cut SCI mouse model suppresses ferroptosis and glial activation, preserves neuronal and myelin integrity, enhances axonal regeneration, and improves motor function (Basso Mouse Scale, gait analysis, electrophysiology). PRP@SeNPs thus provide a drug-free, biocompatible nanotherapeutic strategy to replenish Se, mitigate secondary injury mechanisms, and promote neuroprotection and remyelination for advanced functional recovery after SCI. Yes-associated protein 1 ( YAP1 ), a core effector of the Hippo signaling pathway, has garnered increasing attention for its role in ferroptosis signaling in breast cancer. This study investigates the antitumor properties of Lycium barbarum polysaccharides (LBPs) and develops LBPs-loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles (LBPs NPs), further exploring the mechanisms of free LBPs and LBPs NPs in normal and malignant breast cells. Results demonstrated that YAP1 overexpression drives malignant transformation in normal mammary cells, including aberrant proliferation, apoptosis resistance, cytoskeletal remodeling, and enhanced invasiveness, alongside mitochondrial hyperfunction and altered energy metabolism. However, LBPs effectively suppressed these malignant phenotypes and counteracted the associated mitochondrial and metabolic enhancements by inhibiting YAP1 upregulation. In breast cancer cells, YAP1 overexpression similarly promoted proliferation, invasion, cytoskeletal reorganization, and apoptosis resistance, accompanied by enhanced mitochondrial dysfunction and energy, and acquired ferroptosis resistance. Both LBPs and LBPs NPs significantly inhibited cell proliferation, invasion, and cytoskeletal reorganization, disrupted energy metabolism, and ultimately induced ferroptosis via downregulation of YAP1 expression. Notably, both free LBPs and LBPs NPs effectively suppressed YAP1 expression, with LBPs NPs exhibiting significantly superior efficacy compared to free LBPs. This enhancement is attributed to improved drug delivery efficiency facilitated by the PLGA-based nanocarrier system. Mechanistically, LBPs NPs target the Hippo-YAP1 signaling axis, leading to altered phosphorylation status of YAP1 (p-YAP1), and synergistically induce changes in energy metabolism and ferroptosis to inhibit breast cancer metastasis. These findings provide a proof-of-concept at the in vitro level and a potential nanoparticle-based formulation platform for the treatment of breast cancer.

Objectives To synthesize a novel chlorinated catechol monomer, N-(2-chloro-4,5-dihydroxyphenethyl) methacrylamide (Cl-DMA), and evaluate its efficacy as a primer in enhancing dentin bond durability and imparting antibacterial properties. Methods Cl-DMA at varying concentrations (0, 1, 3, and 5 wt%) dissolved in 50% DMSO/H₂O were applied as primer agent to dentin surfaces for 60 s. The interaction between Cl-DMA with type I collagen was assessed by ATR-FTIR and molecular docking. The effects of Cl-DMA primers on dentin bond strength and durability were comprehensively evaluated using degree of conversion (DC), micro-tensile bond strength (µTBS) test, fracture pattern observation, nanoleakage evaluation, and in situ zymography analysis. The antibacterial properties of Cl-DMA were assessed through colony forming unit (CFU) counting, CCK-8 assay, crystal violet staining, scanning electron microscopy (SEM) observation, and live/dead bacterial fluorescence staining. The assessment of Cl-DMA-induced cytotoxicity was performed by measuring cell viability with a CCK-8 kit and fluorescence staining. Results Cl-DMA potentially interacted with type I collagen via hydrogen bond. Pretreatment with Cl-DMA solutions effectively maintained resin-dentin bond strength, preserved stability of the bonded interface, and inhibited endogenous protease activity. Antibacterial effects against Streptococcus mutans biofilm formation and bactericidal capacity improved gradually with increasing Cl-DMA concentration. Cl-DMA was well-tolerated by the cells compared with Bis-GMA, with no observed cytotoxicity at doses up to 50 µg/mL. Conclusions Pretreating dentin surface with Cl-DMA primer for 60 s can effectively stabilize the bonded interface and exhibit significant antibacterial efficacy.

Conventional hydrogel dressings often fail to simultaneously achieve adhesivity, rapid self-healing, and structural adaptability required for dynamic and irregular wound environments. Here, we report the fabrication of a dual-crosslinked nanocellulose hydrogel via a gradient temperature-controlled strategy, which confers tissue adhesion, tunable porosity, and rapid self-repair. In this process, cellulose nanofibers (CNF) and polyvinyl alcohol (PVA) were dissolved at 90 °C, gelatin was introduced at 70 °C, and reversible borate ester crosslinking was induced at 37 °C, enabling controlled assembly and uniform network formation. The resulting hydrogel exhibited exceptional mechanical performance, with stretchability exceeding 1000% strain and ultrafast self-healing within 8 s at 37 °C. It also showed intrinsic antibacterial activity, achieving an 84.3% inhibition rate against Staphylococcus aureus . Mechanistic studies revealed that the stepwise assembly stabilized CNF/PVA hydrogen-bonded frameworks and facilitated energy dissipation via dynamic borate bond reconfiguration, thereby underpinning the multifunctional properties. In vivo, the hydrogel accelerated wound closure (95.8% at day 14 vs. 84.4% for gauze) and enhanced collagen deposition. Furthermore, it conformed seamlessly to irregular anatomical sites such as joints, minimized secondary tissue damage, and maintained reliable adhesion strength (4.38 kPa). This design thus establishes a rational paradigm for developing next-generation, multifunctional hydrogel dressings for precision wound care.

Phenolic resin waste (PRW) is a carbon-rich industrial byproduct, and its improper disposal leads to environmental pollution and resource loss. In this study, a porous carbon material (PRWPC) with a well-developed porous structure and a large specific surface area (1760.6107 m 2 g −1 ) was prepared from PRW via microwave-assisted alkaline activation and applied for methylene blue (MeB) removal from aqueous solution. Under the optimized conditions with an initial MeB concentration of 100 mg L −1 , an adsorbent dosage of 10 mg, a contact time of 40 min, a temperature of 328 K, and pH = 11, PRWPC exhibits high adsorption performance, achieving a maximum adsorption capacity of 1482.35 mg g −1 with a removal efficiency of 98.8%. Kinetic analysis indicates that the adsorption process follows a pseudo-second-order model, while equilibrium data are well described by the Langmuir isotherm, suggesting monolayer adsorption dominated by micropore filling. Thermodynamic analysis reveals that the adsorption process is spontaneous and endothermic. Overall, this study demonstrates that microwave-assisted conversion of phenolic resin waste provides a feasible, low-cost, and sustainable strategy for the efficient removal of cationic dyes from wastewater.

With the rise in public health awareness and the increasing demand for environmental protection, the biotoxicity assessment of chemical substances has become a multidisciplinary research hotspot and has necessitated the development of multiresponsive, user-friendly sensing materials and methods for the identification of various analytes. This study formulated a novel sensing strategy based on the Hofmeister effect, which modulated the network structure and multimodal physicochemical properties of cellulose nanofiber/gelatin hydrogels through ion-specific interactions. A multiparameter responsive sensitivity coefficient (CPR) analysis model was also developed. This model enabled the qualitative and quantitative analysis of various anions and cations by evaluating the electrical, mechanical, and physical responses of the hydrogel, thereby overcoming the limitations of single-parameter sensing. Experimental results demonstrated that the compressive strength of the hydrogel increased 3.92-fold through the salting-out effect of Na 2 SO 4 , yielding an electrical conductivity of up to 2.89 S/m in a composite ion environment. More importantly, by combining the CPR model with linear discriminant analysis, high-precision identification of nine single ions and six composite ions was successfully achieved. In simulated sweat detection, the sensor achieved a 100% identification accuracy for three physiological states: healthy, dehydrated, and postexercise. This technology is applicable to various fields, including environmental monitoring, medical and health services, and sports management optimization.
