NIR-Triggered Metal-Polyphenol Nanoparticles Enhance HPV-Driven Cancer Immunotherapy via Immunogenic Cell Death and STING Sequential Activation

Maoyu Liu,Jindong Zhang,Shuning Chen,Jiao Zheng,Linlin Xiao,Xiaoli Liu,Yang Cao,Shenyin Zhu,Shufang Chang

HPV-associated malignancies consistently express E6/E7 oncoproteins, making these viral antigens prime targets for therapeutic vaccination. However, insufficient antigen exposure and presentation remain major obstacles for potent immunotherapy. Here, a novel metal-polyphenol network-coated human serum albumin nanoplatform (IMT@H) is engineered to co-deliver IR780 and manganese ions (Mn 2+ ) to achieve enhanced immunogenic cell death (ICD) and cGAS-STING-dependent antigen presentation. The metal-polyphenol nanostructure facilitates the pH-responsive release of Mn 2+ , which subsequently initiates Fenton-like reactions to generate hydroxyl radicals (·OH). Meanwhile, under near-infrared (NIR) light irradiation, IR780 induces mitochondrial-targeted phototherapy and concurrently produces reactive oxygen species (ROS). These processes act synergistically to amplify the oxidative damage and ICD in TC-1 tumors, leading to the release of damage-associated molecular patterns (DAMPs). These ICD-derived DAMPs cooperate with Mn 2+ to sustain activation of the cGAS-STING pathway in dendritic cells. This combinatorial strategy successfully transforms tumor antigens into endogenous vaccines, eventually inhibiting the growth of primary tumors and producing strong abscopal effects. Notably, mice primed with nanovaccines exhibit strong anti-HPV16 E7-specific immune responses and tumor resistance. With its dual therapeutic and preventive functionality, IMT@H represents a paradigm-shifting strategy for virus-driven malignancies and offers a blueprint for engineering self-adjuvanting nanovaccines against viral oncogenesis.

Self-healing Ppy-hydrogel promotes diabetic skin wound healing through enhanced sterilization and macrophage orchestration triggered by NIR

Zhuangzhuang Chu,Xingdan Liu,Tong Zhao,Dongya Jiang,Jing Zhao,Xiaohua Dong,Kelvin W.K. Yeung,Xuanyong Liu,Yun Liao,Liping Ouyang

Non-healing diabetic foot ulcers are the knotty public health issue due to the uncontrolled bacterial infection, prolonged inflammation, and inferior vessel remodeling. In this work, polypyrrole (Ppy) was added into the hybrid hydrogel containing polyvinyl alcohol (PVA), polyethylene glycol (PEG), and hyaluronan (HA) to acquire superior mechanism and photothermal ability. The Ppy composited hybrid hydrogel could effectively kill bacteria through accumulating heat on the hydrogel surface. RNA-Seq analysis shows that the heat accumulation could enhance phagosome of macrophage and M1 activation, which further accelerate bacteria clearance. Benefitting from the bacteria clearance, macrophage could transform its phenotype to M2 in Ppy composited hybrid hydrogel group with near infrared light (NIR) stimulation. The related genes expression in keratinization, keratinocyte differentiation, and establishment of the skin barrier in the skin were up-regulated and collagen and vascular endothelial growth factor (VEGF) expression level are also enhanced. In summary, Ppy composited hybrid hydrogel could effectively solve the issues of infection and poor wound healing in diabetic foot ulcers, making it an ideal candidate dressing for the treatment of chronic wounds.

Enhanced near-infrared II and III emission in Ga2O3: Ni2+ phosphor via charge compensation for NIR spectroscopy application

Yi Zhang,Zhexuan Gao,Yinyan Li,Huanping Wang,Shilong Zhao,Yang Shen,Degang Deng,Shiqing Xu

A novel near-infrared emitting Ga 2 O 3 : Ni 2+ phosphor was prepared, and its emission characteristics were improved through melting aid and charge balance. Under 410 nm excitation, the Ga 2 O 3 : Ni 2+ phosphor exhibits broadband near-infrared emission in the second region, with a peak at 1455 nm and a half maximum width of up to 300 nm. At the same time, the crystallinity of the sample was improved and the charge imbalance during substitution between Ga 3+ and Ni 2+ was eliminated through melting assistance and charge compensation. The results show that the emission intensity of Ga 1.97 O 3 : 0.015Ni 2+ , 0.015Ge 4+ phosphor (with added flux H 3 BO 3 ) is 10 times higher than that of Ga 1.985 O 3 : 0.015Ni 2+ sample. Its quantum yield is as high as 31.2 %, significantly higher than the reported Ni 2+ ion doped phosphors. According to the distinct absorptions of C–H, O–H as well as N–H bonds in the shortwave near-infrared range, samples of ethanol, n-hexanol, water and ammonia demonstrated different absorption peaks under the penetration of the constructed phosphor conversion light-emitting diode, which proved the potential application of Ga 2 O 3 : Ni 2+ , Ge 4+ -H 3 BO 3 phosphor in composition analysis and detection.

Investigating the sensitization of the ytterbium(III) NIR emission by non-sandwich type Yb(III)-porphyrin coordination compounds

Yu-Li Dang,Li-Xia Xie,Na-Na Fei,Ru Qiao,Zhan-Qi Cao,Li-Jie Liu,Guo-Xing Liu,Xin Zheng,Yun-Lai Ren,Xia Sheng,Sheng-Qiang Guo,Cao-Yuan Niu

Two porphyrin ligands with peripheral chelate groups linked at meso and β positions and their two new non-sandwich-type ytterbium(III)-porphyrin coordination compounds have been synthesized. New porphyrin compounds and the two coordination compounds were characterized by Nuclear Magnetic Resonance, High Resolution Mass Spectrum, Ultraviolet and visible, and fluorescence in the visible and Near Infrared (NIR) regions. The two Yb(III) complexes afforded the photoluminescence quantum yields of 2.0% and 1.6% for Yb(III) characteristic NIR emission in DMSO. Finally and importantly, the impact factors on the sensitization of Yb(III) NIR emission by porphyrin ligands via non-sandwich-type linkage were investigated by theoretical calculations and further measurements on fluorescent properties. It is proposed that the sensitization efficiency for the distance between the sensitizer and Yb(III) NIR emitter is better than that for conjugated degree of the peripheral porphyrin linkage.

An NIR light-responsive “on-off-on” photoelectrochemical aptasensor for carcinoembryonic antigen assay based on Y-shaped DNA

Bo Liu,Yonghao Ge,Yahui Lu,Yibo Huang,Xiaoru Zhang,Xunyi Yuan

This work develops a novel photoelectrochemical sensor for the detection of carcinoembryonic antigen (CEA) based on the composite of UCNPs with semiconductors and conformational changes in the DNA structure . Firstly, SnS 2 , ZnIn 2 S 4 and UCNPs were assembled on the surface of the ITO electrode. Then Au NPs were dropped, which could facilitate the coupling of CdSe NPs modified DNA1 via Au–S bond, giving an ITO/SnS 2 /ZnIn 2 S 4 /UCNPs/CdSe heterojunction structure. When irradiated with 980 nm near-infrared (NIR) light, the UV–visible light emitted by the UCNPs could excite the nanocomposite , producing an enhanced photoelectric reaction. Subsequently, CEA aptamer and DNA2-modified SiO 2 were added to form a Y-shaped DNA structure. At this time, the photocurrent was significantly reduced by the combination of the light-blocking effect of SiO 2 and the departure of CdSe NPs from the electrode surface . When the target CEA was added, the recognition between CEA and the aptamer led to the collapse of the Y-shaped DNA structure, the restoration of hairpin DNA and the proximity of CdSe to the electrode. Accordingly, the photocurrent signals enhanced again. Under optimal experimental conditions, the detection limit as low as 0.3 pg mL −1 was obtained with good selectivity , achieving a sensitive “on-off-on” photoelectrochemical sensor for CEA detection.

NIR-triggered tea polyphenol-modified gold nanoparticles-loaded hydrogel treats periodontitis by inhibiting bacteria and inducing bone regeneration

Zhiqiang Dong,Yiyi Lin,Shibo Xu,Linna Chang,Xingjun Zhao,Xifan Mei,Xiuqiu Gao

Periodontitis caused by bacteria is the most common oral disease. In the face of plaque formation, traditional scaling and antibiotic treatment can lead to gum recession, drug resistance and other side effects. The nanoplatform of multifunctional complex treatment is attractive for the treatment of periodontitis. In this study, epigallocatechin gallate (EGCG) was loaded into gold nanoparticle-modified hydrogels ( [email protected] ) to achieve the synergistic functions of near-infrared (NIR) photosensitization, bactericidal and periodontal tissue regeneration. [email protected] rapidly heats up to 50.7 °C within 5 min under NIR light irradiation, exerting a good photothermal effect, while the NIR spectrum can effectively control the release of tea polyphenols, enhance the antibacterial effect, promote angiogenesis and improve bone regeneration. In vitro studies showed that NIR-irradiated composites inhibited S. aureus , E. coli and S. aureus biofilms by 94 %, 92 % and 74 %, increased alkaline phosphatase activity 5-fold after 7 days and increased extracellular matrix mineralization rate 3-fold after 21 days. The rat periodontitis model successfully demonstrated that [email protected] with NIR light irradiation inhibited dental plaque biofilm by 87 % and promoted alveolar bone regeneration by 38 %. The experimental results suggest that this hydrogel with a NIR-sensitive composite nanoplatform offers new possibilities for the treatment of periodontal disease.

NIR Activated Multimodal Therapeutics Based on Metal–Phenolic Networks-Functionalized Nanoplatform for Combating against Multidrug Resistance and Metastasis

Chen Cheng,Weixi Jiang,Yuanli Luo,Li Wan,Xun Guo,Zhuoyan Xie,Rui Tang,Tong Huang,Jingxue Wang,Chier Du,Zhigang Wang,Haitao Ran,Pan Li,Zhiyi Zhou,Jianli Ren

Multidrug resistance (MDR) and metastasis in cancer have become increasingly serious problems since antitumor efficiency is greatly restricted by a single therapeutic modality and the insensitive tumor microenvironment (TME). Herein, metal–phenolic network-functionalized nanoparticles (t-P@TFP NPs) are designed to realize multiple therapeutic modalities and reshape the TME from insensitive to sensitive under multimodal imaging monitoring. After a single irradiation, a near-infrared laser-activated multistage reaction occurs. t-P@TFP NPs trigger the phase transition of perfluoropentane (PFP) to release tannic acid (TA)/ferric ion (Fe 3+ )-coated paclitaxel (PTX) and cause hyperthermia in the tumor region to efficiently kill cancer cells. Additionally, PTX is released after the disassembly of the TA-Fe 3+ film by the abundant adenosine triphosphate (ATP) in the malignant tumor, which concurrently inhibits ATP-dependent drug efflux to improve sensitivity to chemotherapeutic agents. Furthermore, hyperthermia-induced immunogenic cell death (ICD) transforms “cold” tumors into “hot” tumors with the assistance of PD-1/PD-L1 blockade to evoke antitumor immunogenicity. This work carefully reveals the mechanisms underlying the abilities of these multifunctional NPs, providing new insights into combating the proliferation and metastasis of multidrug-resistant tumors.

NIR-triggered Synergetic Photothermal and Chemotherapy Cancer Treatment Based on SiO2@Au@SiO2@QDs-DOX Composite Structural Particles

Xinbing Jiang,Lu Qiao,Huan Yang,Ben Q Li,Shujiang Ding

Graphical SiO 2 @Au@SiO 2 @QDs nanocomposite with dual-functionality of simultaneous heating and temperature sensing were prepared. The heating is activated by a NIR laser through SPR effect while thermal sensing is accomplished by QDs PL thermometry. These nanocomposites are used as a drug carrier for in vitro chemotherapy-photothermal co-treatment for malignant cells. This paper reports the synthesis of multi-layer SiO 2 @Au@SiO 2 @QDs nanoparticles with a dual-functionality of simultaneous heating and temperature sensing and their applications to in vitro chemo-thermal therapy. The heating is activated by a NIR-light through resonance excitation of surface plasma while in-situ thermal sensing is accomplished by the QDs photoluminescent (PL) effect. These dual function nanoparticles are used as a drug carrier for in vitro chemotherapy-photothermal co-treatment for malignant cells. A comparative study of drug release profiles was performed with and without 808 nm laser irradiation. The drug release rate was accelerated by a rise of temperature, which is induced by plasmonic heat generation associated with Au nanoshells; while the temperature change is monitored by the QD nanoparticles at the same time. The results showed that SiO 2 @Au@SiO 2 @QDs-DOX platform enabled the combination of local specific chemotherapy with external near-infrared (NIR) photothermal therapy and significantly improved the efficacy of cancer treatment. This combined treatment demonstrated synergistic chemotherapeutic-thermal effects compared to chemotherapy or photothermal therapy alone, resulting in higher efficacy.

Polyoxometalate-based heterojunction with NIR light-facilitated photocatalytic W6+/W5+ redox cycling for enhanced bacteria-infected wound healing

Mingzhu Lv,Zhou Xu,Yuan Yong,Huan Wang,Chenghui Liu,Qiqi Xu,Guobo Du,Jiani Xie,Yong You,Jiangwei Xiao,Guohui Jiang

Metal-based nanomaterials with shifting or mixed redox states have been widely applied for combating against bacteria-infected diseases by virtue of their peroxidase (POD)-like catalytic activity. Herein, a polyoxometalate-based heterojunction (GdP 5 W 30 @WS 2 ) with shifting or mixed redox states (W 6+ /W 5+ ) is devised to achieve boosted reactive oxygen species (ROS) generation by near-infrared (NIR) laser-facilitated W 6+ /W 5+ redox cycling for accelerated bacteria-infected wound healing. On the one hand, under the exposure to NIR laser, WS 2 as electron donor could be stimulated to transfer electrons to GdP 5 W 30 , which promotes the reduction from W 6+ to W 5+ active sites on the surface of the GdP 5 W 30 . Consequently, the regenerated W 5+ active sites could catalyze endogenous overexpressed H 2 O 2 to persistently generate ROS, which displays enhanced POD-like catalytic activity for chemodynamic therapy (CDT) by NIR-driven photocatalytic W 6+ /W 5+ redox cycling. Moreover, GdP 5 W 30 @WS 2 also displays favorable glutathione (GSH)-activated photothermal performance by W 6+ -triggered GSH exhaustion for sufficient ROS generation, thereby facilitating the synergistic therapeutic efficiency of photothermal therapy (PTT) and CDT. We expect that the present work will promote the development of the heterojunction with shifting or mixed redox states for biofilm elimination based on NIR-facilitated enzyme-like catalytic activity, which may act as a candidate for BME-activated synergistic therapy.

Gold nanobipyramid@copper sulfide nanotheranostics for image-guided NIR-II photo/chemodynamic cancer therapy with enhanced immune response

Yiyu Chen,Ping Liu,Chunze Zhou,Tao Zhang,Tianxing Zhou,Dandan Men,Guihua Jiang,Lifeng Hang

Photothermal therapy (PTT), photodynamic therapy (PDT), and chemodynamic therapy (CDT) can cause cancer cell death through an immunogenic process. However, the study of second near-infrared window (NIR-II)-triggered PTT and PDT combined with CDT to induce an immune response has not been recently reported. Here, we integrated gold nanobipyramids and copper sulfide in a core/shell architecture (AuNBP@CuS). The material displays both photodynamic and photothermal properties under irradiation with a NIR-II laser. The released Cu 2+ from CuS under an acidic tumor microenvironment can be converted to Cu + by glutathione following a Fenton-like reaction with hydrogen peroxide to generate highly toxic hydroxyl radicals in the tumor region. Both in vitro and in vivo results demonstrated that such multifunctional nanoplatforms could achieve enhanced efficiency for image-guided tumor suppression based on the NIR-II photo/chemodynamic therapy. We found that damage-associated molecular pattern molecules such as adenosine triphosphate, pre-apoptotic calreticulin , and high mobility group box-1 in dying cells induced by the NIR-II photo/chemodynamic therapy could simultaneously trigger adaptive immune responses. This is the first report revealing that NIR-II photo/chemodynamic therapy based on AuNBP@CuS had promising performance on tumor suppressor with an effective immunogenic cell death process. Statement of significance 1. AuNBP@CuS displays both NIR-II photodynamic and photothermal properties. 2. Cu + following a Fenton-like reaction to generate highly toxic hydroxyl radicals. 3. The NIR-II photo/chemodynamic therapy can trigger adaptive immune responses. 4. Such multifunctional nanoplatforms could achieve enhanced efficiency for tumor suppression.

NIR-Triggered and ROS-Boosted Nanoplatform for Enhanced Chemo/PDT/PTT Synergistic Therapy of Sorafenib in Hepatocellular Carcinoma

Wang Chonggao,Cheng Xiaolan,Peng Hao,Zhang Yewei

Although being the first-line treatment of advanced hepatocellular carcinoma (HCC), sorafenib (SOR) outcome is limited due to drug resistance and low tumor accumulation. Herein, with MnO 2 as photothermal agent and chlorine6 (Ce6) as photosensitizer, a tumor-targeting and NIR-triggered multifunctional nanoplatform loading sorafenib (MnO 2 -SOR-Ce6@PDA-PEG-FA, MSCPF) was constructed. Owing to oxygen generator MnO 2 , MSCPF could generate excessive ROS, thus can alleviate tumor hypoxia and improve sorafenib accumulation in cancer cells. Besides, ROS production further strengthens Ce6-mediated PDT and PDA-mediated PTT. By exploiting these features, MSCPF exhibited excellent antitumor effects on HCC in the in vitro and in vivo studies, compared to solo sorafenib or PDT/PTT treatment. Further mechanism experiments suggested that MSCPF could inhibit P-gp expression and induce ferroptosis via deactivation of GPX4 and SLC7A11, which ultimately enhanced the antitumor efficacy of SOR. In summary, our work highlights a promising NIR-triggered and ROS-boosted nanoplatform for enhanced chemo/PDT/PTT synergistic therapy of SOR in HCC treatment.

An ultrasensitive NIR-IIa’ fluorescence-based multiplex immunochromatographic strip test platform for antibiotic residues detection in milk samples

Yunyue Zhang,Tao Liao,Guoxin Wang,Juan Xu,Mohan Wang,Fazheng Ren,Hao Zhang

Introduction Widely used in livestock breeding, residues of antibiotic drugs in milk have become a threat to food safety and human health. Current rapid detection technologies using colorimetric immunochromatographic strip tests (IST) lack the necessary sensitivity for on-site trace monitoring. Fluorescence-based detection in the near-infrared IIa’ (NIR-IIa’) region (1000 ∼ 1300 nm) has enormous potential due to greatly minimized auto-fluorescence and light scattering. Objectives The aim of this work is to develop an ultrasensitive IST platform using NIR-IIa’ fluorescent nanoparticles as labels for multiplex antibiotic residues detection in milk. Methods NIR-IIa’ fluorescent nanoparticles were assembled by encapsulating synthesized NIR-IIa’ fluorophores into carboxyl - modified polystyrene nanoparticles. The NIR-IIa’ nanoparticles were subsequently used as labels in an IST platform to detect sulfonamides, quinolones, and lincomycin simultaneously in milk. A portable fluorescent reader was fabricated to provide on-site detection. To further validate the developed IST platform, the detection was compared with LC-MS/MS in 22 real milk samples. Results Fluorescent nanoparticles were synthesized with low energy emission (1030 nm) and large Stokes shift (>250 nm) showing a much higher signal-to-noise ratio compared with fluorophores emitting in the NIR-I region. The developed IST platform yielded a highly sensitive, simultaneous quantification of sulfonamides, quinolones, and lincomycin in milk with detection limits of 46.7, 27.6 and 51.4 pg/mL, respectively, achieving a wide detection range (up to 50 ng/mL). The IST platform showed good accuracy, reproducibility, and specificity with the portable fluorescent reader which could rapidly quantify in 10 s. These results were better than reported immunochromatographic assays using fluorescent labels, and remarkably, showed a higher recognition ability than LC-MS/MS for real samples. Conclusion The utility of NIR-IIa’ fluorescence-based IST platform for the fast, sensitive, and accurate detection of antibiotics in milk was demonstrated, successfully verifying the potential of this platform in detecting trace materials in complex matrices.
