Fabrication, characterization, and bioactivity of self-assembled carrier-free colloidal dispersions from Citrus × Limon ‘Rosso’ essential oil and tea polyphenols

Fangying Cheng,Mianhong Chen,Zhihao Duan,Ying Zou,Yunxia He,Fanke Zeng,Yuan Yuan,Tiaokun Fu,Hao Tu,Ruyi Li,Jihua Li,Wei Zhou

Carrier-free nanodelivery systems are fully self-assembled from active ingredients through interactions, offering the advantages of green, safe, and large-scale manufacturing. To improve the dispersion of Citrus × limon ‘Rosso’ peel essential oil (CEO) in water and boost the biological activity of CEO and tea polyphenols (TP), self-assembled CEO-TP colloidal dispersions (CEO-TP Colloids) were fabricated through sonication without surfactants or carriers. The optimal CEO and TP concentrations in the CEO-TP Colloids were determined to be 10.0 and 20.0 mg/mL by particle size and stability analyzer, respectively. The CEO self-assembled with TP to form spherical nanoparticles through hydrophobic and hydrogen-bonding interactions, whereas the CEO in CEO-TP Colloids weakened TP intramolecular aggregation. Meanwhile, the CEO-TP Colloids showed synergistic effects with better antibacterial, cellular antioxidant, and anti-inflammatory activities than single components. This study opens up the possibility of carrier-free co-delivery of hydrophobic and hydrophilic active components developed into food-grade formulations with multiple bioactivities.

Flavonoid-rich sesame leaf extract-mediated synthesis of nanozymes: Extraction optimization, chemical composition identification and bioactivity evaluation

Ruiying Zhang,Yufei Liu,Yiqiao Gao,Dengfeng Peng,Qian Luan,Ziliang Li,Xiaoyang Xia,Xia Xiang

Sesame leaves contain rich phenolic acids and flavonoids. However, their potential in nanozyme synthesis has not been investigated yet. Herein, we report the preparation of flavonoid-rich sesame leaf extract (SLE), composition identification, and its use in the construction of iron (Fe)-based nanozymes (Fe-SLE CPNs). SLE was obtained with an extraction yield of ∼14.5% with a total flavonoid content (TFC) of ∼850.85 mg RE/g. There were 83 flavonoid compounds in SLE, primarily including scutellarin, apigenin-7-glucuronid, narcissin, and hyperoside. Fe-SLE CPNs exhibited nanodot morphology with a hydrodynamic size of 79.34 nm and good stability in various physiological solutions, pH levels, and temperatures. The Fe-SLE CPNs were more efficient in the scavenging ability of reactive oxygen species (ROS) than SLE alone. Furthermore, a stronger anti-inflammatory effect of the Fe-SLE CPNs was shown by modulating the MyD88-NF-κB-MAPK signaling pathways. These findings imply that SLE-based nanozymes hold great potential for diverse applications.

Flavor Characteristics, Antioxidant Activity and In Vitro Digestion Properties of Bread with Large-Leaf Yellow Tea Powder

Gexing Zhang,Yang Zhong,Xinzhen Zhang,Yuqi Wang,Yue Sun,Xueling Li,Zhengquan Liu,Jin Liang

Foods containing tea could be widely utilized due to the addition of good tea ingredients, especially large-leaf yellow tea, which is rich with a good flavor. Applying this change to bread containing tea would improve its product quality. In this research, large-leaf yellow tea bread (LYB), possessing a special flavor, was developed using ultrafine large-leaf yellow tea powder and flour as the main raw materials. The amount of ultrafine large-leaf yellow tea powder added to bread was optimized using texture, sensation, and specific volume as comprehensive evaluation indicators. At the optimal dosage, the free amino acids, volatile flavor compounds, antioxidant activity, and in vitro starch digestibility of LYB were measured. Response surface optimization experimental results showed that the comprehensive score of bread was highest when the added amount of ultrafine large-leaf yellow tea powder was 3%. In particular, compared to blank bread (BB), adding ultrafine large-leaf yellow tea powder into bread could effectively increase its amino acid composition, enhance its volatile flavor compounds, improve the antioxidant capacity, and reduce the digestibility of starch.

Formulation, Characterization, Antibacterial Activity, Antioxidant Activity, and Safety Evaluation of Camphora longepaniculata Essential Oil Nanoemulsions Through High-Pressure Homogenization

Yue Yan,Changhe Wei,Xin Liu,Xin Zhao,Shanmei Zhao,Shuai Tong,Guoyou Ren,Qin Wei

The volatility, instability, and water insolubility ofCamphora longepaniculataessential oil (CLEO) limit its practical applications in the food, pharmaceutical, and cosmetics industries. CLEO nanoemulsions (CLNEs) were formulated and characterized to overcome the aforementioned issues. The volatile compounds of CLEO were identified by gas chromatography–mass spectrometry. CLNEs were prepared using EL-40 (5%,w/w) as the surfactant via the high-pressure homogenization method. The formation of nanoemulsions was verified by Fourier transform infrared spectroscopy and transmission electron microscopy. Homogenized nanoemulsions had smaller particle sizes of 39.99 ± 0.47 nm and exhibited enhanced stability. The nanostructured CLEO showed an antibacterial effect againstEscherichia coliandStaphylococcus aureus. The antibacterial mechanism was explored through bacterial morphology and intracellular lysate leakage. CLNEs disrupted the structure of bacterial cells and impaired the permeability of cell membranes, resulting in the leakage of bacterial intracellular contents. The nanoemulsions exhibited superior radical scavenging ability compared to the pure oil. Furthermore, evaluations of the cellular safety of the CLNEs demonstrated that the survival rate of exposed HOECs was greater than 90%, with an apoptosis rate of less than 10% in a concentration range. The results demonstrated that nanoemulsification improved the stability, solubility in aqueous media, and bioavailability of CLEO, thereby broadening its potential industrial applications as a natural antibacterial and antioxidant agent.

Fractional extraction of lignin from coffee beans with low cytotoxicity, excellent anticancer and antioxidant activities

Shiwen Liu,Shan Xiao,Bo Wang,Yanxue Cai,Ruihong Xie,Xing Wang,Jihui Wang

Lignin, a biopolymer generated from renewable resources, is widely present in terrestrial plants and possesses notable biosafety characteristics. The objective of this work was to assess the edible safety, in vitro antioxidant, and anti-cancer properties of various lignin fractions isolated from commercially available coffee beans often used for coffee preparation. The findings suggest that the phenolic hydroxyl content increased from 3.26 mmol/g (ED70L) to 5.81 mmol/g (ED0L) with decreasing molecular weight, which resulted in more significant antioxidant properties of the low molecular weight lignin fraction. The findings of the study indicate that the viability of RAW 264.7 and HaCaT cells decreased as the quantity of lignin fractions increased. It was observed that concentrations below 200 μg/mL did not exhibit any harmful effects on normal cells. The results of the study demonstrated a significant reduction of cancer cell growth (specifically A375 cells) at a concentration of 800 μg/mL for all lignin fractions, with an observed inhibition rate of 95 %. The results of this study indicate that the lignin extracts derived from coffee beans exhibit significant potential in mitigating diseases resulting from excessive radical production. Furthermore, these extracts show promise as natural antioxidants and anti-cancer agents.

Ginkgo biloba leaf polysaccharide-stabled selenium nanozyme as an efficient glutathione peroxidase mimic for the preservation of bananas and cherry tomatoes

Jiahui Zou,Huimin Zhong,Cong Jiang,Guancheng Zhu,Xueer Lin,Yanyan Huang

To develop functional, sustainable and eco-friendly active packaging materials as alternatives to plastic films, we successfully prepared Ginkgo biloba leaf polysaccharide-stabilized selenium nanomaterials (Se-GBLP). Se-GBLP with glutathione peroxidase-like activity could efficiently remove harmful reactive oxygen species. As a functional additive, Se-GBLP was incorporated into degradable chitosan (CS) to fabricate CS/Se-GBLP films. The addition of Se-GBLP improved the mechanical properties, UV–visible light barrier performance, water vapor permeability, and antioxidant activity of the films. Preservation experiments demonstrated CS/Se-GBLP film could maintain quality and prolong the storage time of bananas and cherry tomatoes . It was the first time to use selenium-based nanozyme for fruit preservation. This work offered a cost-effective solution to reduce post-harvest losses, increasing sustainability and profitability. Future research should focus on more factors affecting freshness such as variety, maturity, harvest and storage conditions to improve preservation, as well as on the material's safety concern and environmental impact.

Green Extraction of Antioxidant-Rich Flavonoids from Fagonia cretica Using Deep Eutectic Solvents

Jafar Khan,Sajjad Asaf,Lubna,Ashraf M. M. Abdelbacki,Rahmatullah Jan,Kyung-Min Kim

This study optimized the extraction of flavonoids fromFagonia creticausing deep eutectic solvents (DESs), focusing on key factors such as the type of DES used, molar ratio, water content, solid/liquid ratio, extraction temperature, and time. Among six DESs tested, the betaine–acetic acid combination exhibited the highest extraction efficiency, attributed to its low viscosity (4.98 mPa·s). Optimal extraction conditions were determined to be a 1:4 molar ratio of betaine to acetic acid, a 25% water content, a solid/liquid ratio of 1:60 g/mL, an extraction temperature of 50 °C, and an extraction time of 30 min. Under these conditions, the flavonoid yield was maximized while preserving bioactivity. Antioxidant assays revealed that flavonoids extracted with DESs exhibited superior scavenging activity against DPPH and hydroxyl radical compared to ethanol-extracted flavonoids, highlighting DESs’ potential to enhance antioxidant properties. The recyclability of DESs was demonstrated using ultracapacitor porous activated carbon, achieving an 89.78% recovery efficiency. The reused DES maintained a high flavonoid extraction yield, retaining 92% efficiency after six cycles, emphasizing its sustainability and cost-effectiveness. This study establishes DES-based extraction as an environmentally friendly and efficient approach for isolating flavonoids with strong antioxidant properties, offering significant advantages in green chemistry and bioactive compound recovery.

Green preparation of licorice flavonoids/ZnO/attapulgite nanocomposites for multifunctional chitosan-based food packaging films

Wenting Gao,Bin Mu,Fangfang Yang,Yalong Li,Aiqin Wang

Multifunctional biodegradable packaging films with antibacterial and antioxidant properties are highly desirable for food preservation. In this study, multifunctional bioactive chitosan-based composite films were fabricated for loquat preservation after incorporation of licorice flavonoids/ZnO/attapulgite (LF/ZnO/APT) nanocomposites, which were prepared by anchoring ZnO nanoparticles on the surface of attapulgite followed by mechanochemical-assisted loading of licorice flavonoids. The structure and physicochemical properties of composite films were characterized using Fourier transform infrared spectroscopy, transmission electron microscopy, X-ray powder diffraction, etc., while the effects of the added amount of LF/ZnO/APT nanocomposites on the mechanical, antibacterial and antioxidant performances of composite films were investigated. The results indicated that introduction of LF/ZnO/APT nanocomposites obviously enhanced the mechanical properties and antibacterial ratios against Escherichia coli (97.04 ± 2.08%) and Staphylococcus aureus (98.68 ± 1.10%) of composite films. Moreover, the composite films also exhibited excellent scavenging capacities against 2,2-diphenyl-1-picrylhydrazyl and hydroxyl radicals. Therefore, the as-prepared composite films significantly extended the shelf life of loquats, and it is feasible to be served as promising active food packaging materials based on component synergistic effect of composite films.

Green Starch-Based Hydrogels with Excellent Injectability, Self-Healing, Adhesion, Photothermal Effect, and Antibacterial Activity for Promoting Wound Healing

Ke Xu,Xinyu Sun,Chuanyu Chong,Li Ren,Lili Tan,Haonan Sun,Xin Wang,Lihua Li,Junfang Xia,Ruoxi Zhang,Li Wang

Hydrogel materials have proven valuable in wound healing, but improving the safety of these hydrogels is still challenging. Therefore, designing multifunctional natural polymeric-based hydrogels with excellent mechanical properties to replace toxic or potentially risky, refractory chemical polymer-based hydrogels such as polyacrylamide and polyethylene glycol is of particular significance. Here, a green starch-based hydrogel (Starch@Ca/CGC hydrogel) with injectability, self-healing, and instant adhesion was constructed by coordination interaction, electrostatic interaction, and intramolecular and intermolecular hydrogen bonds. Therein, natural bioactive small molecules gallic acid (GA) and carvacrol (CA) were coordinated with metal ions by the ultrasonic-triggered self-assembly and ionic cross-linking codriven strategy to prepare Cu-gallic acid-carvacrol nanospheres (CGC NPs), which conferred the hydrogel with near-infrared light (NIR)-controlled CA release and photothermal synergistic sterilization properties, as well as antioxidant and anti-infection capabilities. More importantly, the multifunctional hydrogel platforms could completely cover an irregular wound shape to prevent secondary injury and significantly accelerate wound healing under NIR with more skin appendages like hair follicles and blood vessels appearing. Therefore, it is expected that this starch-based hydrogel could serve as a competitive multifunctional dressing in the biomedical field, including bacteria-derived wound infection and other tissue repair.

Green tea polyphenol alleviates silica particle-induced lung injury by suppressing IL-17/NF-κB p65 signaling-driven inflammation

Yunyi Xu,Qionghua Ding,Yuhuan Xie,Qingqing Zhang,Yangyu Zhou,Hairui Sun,Rui Qian,Liqun Wang,Xuxi Chen,Ying Gao,Yuqin Yao,Yue Xiao

Background Silicosis, an interstitial lung disease caused by inhalation of silica particles, poses a significant health concern globally. Green tea polyphenol (TP) stands out as a promising therapeutic candidate, yet its specific protective effects and in-depth mechanisms against silicosis have not been thoroughly investigated. Purpose This study aimed to systematically assess the protective potential of TP against silicosis and to elucidate the underlying mechanisms of its action. Methods A combination of physiological, transcriptomic, molecular, and computational techniques was employed. HPLC was used to identify the components of TP, and its antioxidant properties were tested with DPPH and ABTS assays. The effects of TP on lung injury were assessed in silicosis mice using histopathology, qRT-PCR, and western blot. Transcriptomic analysis was applied to explore the differentially expressed genes and pathways in response to TP intervention. In vitro studies with mouse alveolar macrophages (MH-S) examined TP's effects on cell viability, proliferation, apoptosis, and inflammation responses. Integrated qRT-PCR, western blot, immunohistochemistry, and molecular docking were performed to confirm the molecular mechanism underlying the protective effects of TP against silicosis. Results TP effectively attenuated pulmonary inflammation and fibrosis in silicosis mice, as evidenced by significant reductions in inflammation and fibrotic markers. Moreover, TP's therapeutic benefits were linked to its cytoprotective effects on alveolar macrophages, notably its ability to protect MH-S cells from silica particle-induced apoptosis, inhibition of proliferation, and inflammatory response, underscoring its targeted protective effects at the cellular level. Mechanistically, TP exerted its anti-silicosis activity by targeting key pathways implicated in inflammatory responses, notably through the inhibition of the IL-17/NF-κB p65 signaling cascade. Molecular docking simulations corroborated these findings, demonstrating favorable binding affinities between TP's bioactive components (EGC, ECG, and EGCG) and crucial proteins (IL-17A, IL-17F, p65, TNF-α, IL-6, and IL-1β) involved in the IL-17/NF-κB p65 signaling pathway. This pathway inhibition led to a significant decrease in the production of pro-inflammatory cytokines, such as TNF-α, IL-6, and IL-1β, thus mitigated silicosis. Conclusion TP demonstrates efficacy in alleviating silica particle-induced lung injury by suppressing inflammation through the IL-17/NF-κB p65 signaling pathway, underscoring its potential as a valuable natural compound for silicosis management.

Kang Hu,Zhuo Huang,Qinying Tang,Danyang Chen,Lianxu Chen,Lu Chen,Guohua Jiang,Qianfei Huang,Langjie Chai,Hang Chen,Liang Guo,Bin Li

Currently, searching for safer and more effective approaches to promote skin wound healing and tissue regeneration is a significant research focus in the field of public health. Eliminating excessive reactive oxygen species (ROS) and using electrically active dressings to accelerate wound healing has gained significant attention. Herein, green tea-based carbon dots (GCDs) are synthesized from leaves of green tea and proven to possess strong antioxidant, antibacterial, and anti-inflammatory properties. Then, GCDs, polyvinyl alcohol (PVA), and poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS) are successfully constructed into a multifunctional conductive PVA-PEDOT:PSS/GCDs hydrogel (PPPCD) using a repeated freeze-thaw method for promoting skin wound healing. Highly electrical conductivity is imparted by introducing PEDOT:PSS, while antioxidant and antibacterial properties are conferred by GCDs. In vitro assays demonstrated the excellent biocompatibility of the hydrogel, as well as its ability to scavenge excessive ROS and promote cell migration. Furthermore, the hydrogel not only accelerates wound healing by promoting blood vessel formation and epidermal regeneration but also alleviating excessive inflammation in vivo. Overall, the developed hydrogel dressing demonstrates promising prospects for skin wound healing and offers a new perspective for tissue engineering.

Hybrid nano-microstructured and bioinspired conductive hydrogels with tunable multifunctionality

Manting Wang,Jiaqi Zhang,Yaoyi Guo,Xiaoyong Zhou,Jie-Xin Wang,Yuan Le

Conductive hydrogels with tunable multifunctionality show great potential in wearable smart devices. In this work, inspired by the nano-structured architecture strategy and natural mussel adhesive mechanism, a conductive hydrogel (AgPMP hydrogel) was fabricated through facile direct in situ polymerization. Silver nanowires (Ag NWs) and polydopamine (PDA) chains were integrated into the poly(N-isopropylacrylamide-co-acrylamide) (P(NIPAM-co-AM)) gel matrix, with activated PNIPAM microgels (MGs) serving as nano-crosslinkers. The overall internal structure of the hydrogel presented a heterogeneous cross-linked mesh-like structure with numerous irregular wrinkles. The AgPMP hydrogel demonstrated significantly enhanced mechanical strength, achieving a remarkable stretchability of 1140% attributed to the nano-microstructured architecture built using AM-based networks, nano-crosslinkers and Ag NWs. Moreover, due to the synergistic effect of PDA and Ag NWs, the hydrogel exhibited impressive photothermal antibacterial activity. Meanwhile, in the presence of abundant active catechol groups, this hydrogel possessed adequate tissue-adhesive and self-healing abilities as well as good antioxidant properties. Notably, the AgPMP hydrogel exhibited outstanding strain-sensitive conductivity. The resultant strain sensor showed high sensitivity with a gauge factor (GF) of up to 8.35 at a strain of 250%. The stable response signals during continuous and gradual cycling proved its electrical durability and stability. Combined with the great merits of the above performance, this sensor holds great promise for detecting human movements. The relative resistance change ranged from 16% to 64% as the finger bending angle changed from 30° to 90°. Thanks to its simple fabrication process, this multifunctional conductive hydrogel is expected to have widespread potential applications in tissue engineering, soft electronic skin and human-health monitoring.
