Whole Genome Sequencing and RNA-seq-Driven Discovery of New Targets That Affect Carotenoid Synthesis in Phaffia rhodozyma

Shi Zhihui,He Xiaoxian,Zhang Hailiang,Guo Xuena,Cheng Yanfei,Liu Xuelian,Wang Zhaoyue,He Xiuping

Carotenoids are unsaturated compounds with terpene groups. Among them, astaxanthin has strong antioxidant properties. It is widely used in aquaculture, food, medicine, and cosmetics with a broad market prospect. Phaffia rhodozyma is an important microorganism that synthesizes astaxanthin, but its wild strains have low pigment content, long growth cycle, and low fermentation temperature. Therefore, it is important to research the genetic improvement of the physiological and biochemical properties of P. rhodozyma. In this study, the atmospheric and room temperature plasma mutagenesis technology was adopted, through the functional evolution of the carotenoid production performance; then, through the comparative analysis of the genomics and transcriptomics of the wild strain and evolved strain, the key factor GST1 gene that affects carotenoid synthesis was discovered.

Chitin-rich shrimp shells-based nitrogen-doped green fluorescence carbon dots: New insights for multi-orbital applications

Mingying Chen,Danni Wu,Xiaoye Deng,Yingjie Li,Xiaoqin Sun,Junjie Ma,Congjin Chen,Xiufen Liao,Xijun Liu

Carbon dots (CDs), revolutionary carbon-based nanomaterial, have demonstrated tremendous potential for applications across multiple areas. Chitin-rich shrimp shells are a sustainable and renewable resource, and their utilization can turn waste into treasure. This work is dedicated to efficiently preparing nitrogen-doped green fluorescent CDs (N-CDs) for multi-modal applications from waste shrimp shells by hydrothermal methods. The detailed spectroscopic characterization reveals that the N-CDs possess excellent optical properties. The quantum yield is as high as 33.2%. Specifically, the N-CDs show excellent selectivity and sensitivity in the quantitative detection of Fe 3+ . Additionally, in response to pH changes, the surface functional groups of the N-CDs undergo protonation and deprotonation, leading to corresponding alterations in fluorescence colors. Capitalizing on this characteristic, a visual portable pH-measuring platform is developed. This platform ingeniously integrates nanoscale optical sensors with smartphone image processing technology, enabling real-time and highly accurate monitoring of environmental pH levels. Furthermore, they are ideal materials for fluorescence anti-counterfeiting detection due to their unique optical properties. This work simultaneously accomplishes the reuse of waste biomass and provides novel insights for multi-orbital applications, including metal ion detection, pH monitoring, and the anti-counterfeiting field.

Microencapsulation of norfloxacin in chitosan/chitosan oligosaccharides and its application in shrimp culture

Ziru Lian,Rong Pan,Jiangtao Wang

Norfloxacin chitosan/chitosan oligosaccharide microcapsules (NCCM) were prepared by emulsion-chemical crosslinking method. The characteristics of obtained microcapsules were evaluated by scanning electron microscopy, Fourier transform infrared spectroscopy and release experiments. Cumulative release profile of norfloxacin from the chitosan microcapsules in natural seawater was measured and the controlled release of drugs was at a uniform rate in 48   h. The chitosan microcapsules were applied onto the antibacterial study of the shrimp culture in natural seawater. It is observed that the seawater in the NCCM added groups was relatively clear and the biomass of Vibrio increased slowly in contrast to the control and norfloxacin groups. The inhibition rate of Vibrio in norfloxacin groups obvioursly decreased after the 5 th day, whereas, it remained high and stable during experiment period in NCCM groups. The results showed that the chitosan microcapsules as release materials have excellent antibacterial effects on Vibrio in the farming of Penaeus vannamei Boone. The controlled release could obviously reduce dosage of antibiotics and delivery times, and effectively improve the utilization rate of norfloxacin drugs for shrimps

Preparation of Antarctic Krill Oil Emulsion and Its Stability under Catalase Treatment

Zhenxiao Zheng,Kai Zhu,Zhiyuan Dai

Making Antarctic krill oil into emulsion is a good way to utilize Antarctic krill, but proliferation of microorganisms cannot be ignored. H2O2is widely used in the sterilization of liquid food since its decomposition products are environmentally friendly, although residual H2O2should be removed for food safety. Adding catalase (CAT) is an effective means to do this. However, the enzyme activity center of CAT is the iron porphyrin group, which has the risk of accelerating lipid oxidation in the oil emulsion. Therefore, we hypothesized that CAT might not be suitable for the removal of H2O2in Antarctic krill oil emulsion. In this paper, Antarctic krill oil emulsion was prepared, and then the effect of CAT on the emulsion was studied through visual observation, microscopic morphology observation, turbidity and stability, particle size, and ζ-potential; finally, the mechanism of CAT destroying the emulsion was explored from the perspective of lipid oxidation. The results showed that a stable Antarctic krill emulsion was prepared using Tween-80 as the emulsifier, with the oil concentration of 1% (v/v) and the ratio of surfactant to oil phase of 1:5 (v/v). The emulsion treated with CAT had undergone demulsification, stratification, and coagulation after 2 days of incubation, while the emulsion treated with superoxide dismutase (SOD) and bovine serum albumin (BSA) changed little. In addition, the thiobarbituric acid reactive substances (TBARS) value and the content of hydroxyl radicals in the CAT group increased significantly. The preliminary research results indicated that the effect of CAT on the emulsion related to the lipid oxidation caused by the iron porphyrin group at the center of the enzyme activity. All these results indicated that CAT was not suitable for the removal of residual H2O2in Antarctic krill oil emulsion. Moreover, it is helpful to avoid the contact of Antarctic krill oil emulsion and CAT during the processing of the krill. Keywords:Antarctic krill emulsion;catalase;iron porphyrin;lipid oxidation

Adsorption of Tetracycline by Shrimp Shell Waste from Aqueous Solutions: Adsorption Isotherm, Kinetics Modeling, and Mechanism

Jing Chang,Zhen Shen,Xiude Hu,Emily Schulman,Chunyue Cui,Qingjie Guo,Hongjing Tian

The highly efficient removal of tetracycline (TC) from an aqueous solution was accomplished by using the raw shrimp shell waste (SSW) as an environmentally friendly adsorbent. The SSW without any treatment removed TC more efficiently than the SSW after being treated with HCl and NaOH solutions. The SSW was characterized using nitrogen adsorption–desorption isotherms, scanning electron microscopy alongside energy-dispersive X-ray spectroscopy, Fourier transform infrared spectroscopy, a thermogravimetric-derivative thermogravimetry analyzer, and a ζ-potential analyzer. The maximum adsorption capacity of 400 mg/L SSW was 229.98 mg/g for 36 h at 55 °C. Both the Langmuir isotherm model and the pseudo-second-order kinetic model well described the experimental data. According to the values of the Gibbs free energy and enthalpy changes, the TC adsorption by SSW proved to be spontaneous and endothermic. The TC adsorption process was controlled by intraparticle diffusion and liquid film diffusion.

Effects of nonylphenol exposure on histological changes, apoptosis and time-course transcriptome in gills of white shrimp Litopenaeus vannamei

Hui Guo,Zhi Liang,Peihua Zheng,Ling Li,Jian'an Xian,Xiaowen Zhu

Nonylphenol (NP) is considered as one of the persistent organic pollutants (POPs) in the environment. Pacific white shrimp Litopenaeus vannamei is the predominant species in China, which is frequently affected by environmental pollutants. However, potential toxicity mechanism of NP in shrimp has not been comprehensively studied. To explore the physiological changes and molecular mechanism involved in NP exposure of shrimp, we analyzed histological alterations, apoptosis and transcriptional responses of L. vannamei subjected to NP. Results indicated that significant changes in the histoarchitecture of the gills were observed after NP exposure for 3, 12 and 48 h. Apoptosis was also detected in a time-dependent manner. Numerous differentially expressed genes (DEGs) were obtained at 3 h, 12 h and 48 h after exposure. On the basis of the expression patterns over the time course, these DEGs were classified into 12 clusters. GO and KEGG enrichment analysis of these DEGs was carried out and a dynamic and global view was obtained in shrimp after NP exposure on a transcriptome level. In addition, 15 DEGs involved in immune response, apoptosis, DNA repair, osmoregulation etc. were selected for qRT-PCR validation. The expression patterns of these DEGs kept a well consistent with the high-throughput data at different timepoints, which confirmed the accuracy and reliability of the transcriptome data. All the results demonstrated that NP exposure might lead to impairments of biological functions in gills, alter immune and antioxidant response, compromise DNA repair and anti-apoptosis abilities of shrimp, cause severe histopathological changes and eventually trigger apoptosis. The present study enriched the information on the toxicity mechanism of crustaceans in response to NP exposure.

Preparation of carrier-free astaxanthin nanoparticles with improved antioxidant capacity

Yu Fei,Chen Jiaxin,Wei Zizhan,Zhu Pingchuan,Qing Qing,Li Bangda,Chen Huimin,Lin Weiying,Yang Hua,Qi Zhongquan,Hong Xuehui,Chen Xiao Dong

Astaxanthin (AST), a red pigment of the carotenoids, has various advantageous biological activities. Nevertheless, the wide application of AST is restricted due to its poor water solubility and highly unsaturated structure. To overcome these limitations, carrier-free astaxanthin nanoparticles (AST-NPs) were fabricated through the anti-solvent precipitation method. The AST-NPs had a small particle size, negative zeta potential and high loading capacity. Analysis of DSC and XRD demonstrated that amorphous AST existed in AST-NPs. In comparison with free AST, AST-NPs displayed enhanced stability during storage. Besides, it also showed outstanding stability when exposed to UV light. Furthermore, the antioxidant capacity of AST-NPs was significantly increased. In vitro release study showed that AST-NPs significantly delayed the release of AST in the releasing medium. These findings indicated that AST-NPs would be an ideal formulation for AST, which could contribute to the development of novel functional foods.

Development of intelligent/active food packaging film based on TEMPO-oxidized bacterial cellulose containing thymol and anthocyanin-rich purple potato extract for shelf life extension of shrimp

Yanyi Wen,Jia Liu,Lei Jiang,Zhongjie Zhu,Suirui He,Shu He,Wei Shao

An intelligent and active film was developed based on 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO)-oxidized bacterial cellulose (TOBC) containing thymol (THY) and anthocyanin-rich purple potato extract (ANT). The microstructure, thermal stability, water vapor permeability, mechanical property and UV protection performance of TOBC-based films were evaluated. Scanning Electron Microscopy (SEM) and Atomic Force Microscope (AFM) results show the additions of THY and ANT bring about the uneven distribution of TOBC fibers with slightly increased surface roughness . Meanwhile, the introduction of THY and ANT could increase the thermal stability, UV protection and water vapor barrier property of TOBC with slightly decreased tensile strength . The prepared ANT exhibits a pH-sensitive activity, and it endows the fabricated TOBC/THY/ANT film with colorimetric response ability to volatile ammonia. TOBC/THY/ANT film also exhibits good antibacterial and antioxidant activities . Particular finding is that TOBC/THY/ANT film possesses simultaneously freshness real-time monitoring and good shrimp preservation abilities. The antioxidant, antibacterial and color response performances of TOBC/THY/ANT film were measured for three cycles in order to prove its recycling ability. These unique features of TOBC/THY/ANT film demonstrate the potential in commercial shrimp packaging with freshness real-time monitoring.

A novel curcumin-based bilayer indicator film for freshness evaluation of shrimp with excellent mechanical properties and moisture resistance

Qi Zhang,Jian Zhang,Qingwei Ping,Zhihui Sui,Xiangyu Cao,Tianyuan Xiao

Curcumin-based smart films have gained widespread attention in the packaging field due to their convenience, ease of use, and environmental sustainability. However, its poor pH response, unsatisfactory mechanical properties , and limited moisture resistance severely restrict its applications. In this study, we prepared a curcumin-based bilayer indicator film with a protective layer consisting of gelatin, carboxymethyl cellulose , and hexadecyl trimethoxy silane to provide mechanical protection and moisture resistance, and an indicator layer consisting of gelatin, curcumin, and malvidin to improve the pH sensitivity of the curcumin-based film and broaden the range of color indication. Owing to the hydrophobic effect of the protective layer and cross-linking between the bilayers, the curcumin-based bilayer film exhibited excellent mechanical properties with a stress of 15.3 MPa, a strain of 101.6 %, and a Young's modulus of 38.9 MPa. The moisture resistance of the film was also improved, with a water vapor permeability as low as 1.11 ± 0.15 × 10 −10  g m m −2  Pa −1  s −1 . In addition, with the addition of malvidin, the curcumin-based bilayer film showed greater sensitivity to pH and more pronounced color changes compared to the gelatin/curcumin film for practical applications in monitoring shrimp freshness. Overall, this study demonstrated the applicability of the prepared curcumin-based bilayer indicator film for monitoring shrimp freshness and also broadened the prospects of curcumin-based films in the field of smart packaging.

Selectively extraction of astaxanthin from Haematococcus pluvialis by aqueous biphasic systems composed of ionic liquids and deep eutectic solutions

Lili Zhang,Yan Li,Jing Gao

Haematococcus pluvialis is rich in astaxanthin ester (Ast 1 ) and free astaxanthin (Ast 2 ). The astaxanthin is natural antioxidant, and shows higher biodegradability and eco-friendly characteristics than commercial astaxanthin. However, the existing extraction technology is difficult to achieve selectively separation of Ast 1 and Ast 2 . Novel aqueous biphasic systems (ABSs) composed of ionic liquids (ILs) and deep eutectic solvents (DESs) were developed to selectively extract Ast 1 and Ast 2 . The results showed that the ABS composed of tributyloctylphosphine chloride ([P 4448 ]Cl) and choline chloride: d -fructose ([ChCl][Fru]) performed best. The yield and partition coefficient of astaxanthin was 2.35 mg·g −1 and 103.59, respectively. Moreover, Ast 1 and Ast 2 could be selectively separately with the highest coefficient of specificity of 48.31. Furthermore, the ABS maintained an excellent extraction capacity after three cycles of recycling. As compared with organic solvent extraction and prior methods, the IL-DES ABS is more efficient, recyclable, and environmentally friendly for selectively extraction of astaxanthin.

Effects of Low Molecular Weight Peptides from Red Shrimp (Solenocera crassicornis) Head on Immune Response in Immunosuppressed Mice

Rui Zhao,Shuoqi Jiang,Yunping Tang,Guofang Ding

This study aimed to investigate the immunoenhancement effects of low molecular weight peptides (SCHPs-F1) from red shrimp (Solenocera crassicornis) head against cyclophosphamide (CTX)-induced immunosuppressed mice. ICR mice were intraperitoneally injected with 80 mg/kg CTX for 5 consecutive days to establish the immunosuppressive model and then intragastrically administered with SCHPs-F1 (100 mg/kg, 200 mg/kg, and 400 mg/kg) to investigate its improving effect on immunosuppressed mice and explore its potential mechanism using Western blot. SCHPs-F1 could effectively improve the spleen and thymus index, promoting serum cytokines and immunoglobulins production and upregulating the proliferative activity of splenic lymphocytes and peritoneal macrophages of the CTX-treated mice. Moreover, SCHPs-F1 could significantly promote the expression levels of related proteins in the NF-κB and MAPK pathways in the spleen tissues. Overall, the results suggested that SCHPs-F1 could effectively ameliorate the immune deficiency caused by CTX and had the potential to explore as an immunomodulator in functional foods or dietary supplements. Keywords:low molecular weight peptides;immunoenhancement;cyclophosphamide;Solenocera crassicornis;signal pathways

Preparation of porous chitin beads from waste crayfish shell and application in the co-immobilization of PLP and its dependent enzyme

Guoguang Wei,Chi Zhang,Ning Zhou,Bin Wu,Hui Li,Alei Zhang,Pingkai Ouyang,Kequan Chen

In this study, co-immobilization of PLP and its dependent enzyme were investigated using a novel type of porous chitin bead (PCB). Crayfish shell was used to prepare PCB via dissolution of it to form beads, followed by the removal of CaCO 3 and protein in-situ. Scanning electron microscopy, Fourier transform infrared spectroscopy , and Brunauer-Emmett-Teller method showed that the PCB had abundant porous structures with deacetylation degree of 33 % and the specific surface area of 35.87 m 2 /g. Then, the beads are used to co-immobilize pyridoxal 5-phosphate (PLP) and l -lysine decarboxylase fused with chitin-binding protein ( Sp LDC-ChBD). Laser scanning confocal microscopy revealed that the beads could co-immobilize PLP and Sp LDC-ChBD successfully. In addition, a packed bed was also constructed using the PCB containing co-immobilized Sp LDC-ChBD and PLP. The substrate conversion remained at 91.09 % after 48 h with 50 g/L  l -lysine, which showed good continuous catalysis ability. This study provides a novel method for co-immobilization of enzyme and PLP, as well as develops a new application of waste crustacean shells.
