Recently, 3D printing technology has been widely applied in microfluidics due to its capacity of three-dimensional construction, rapid and automatic fabrication, and less time consumption. However, a droplet-based PCR in 3D-printed microfluidic chips is rarely reported. In this study, a 3D-printed microfluidic chip was designed and fabricated for droplet-based PCR detection of miRNA-21. In this chip, the capacity of monodisperse droplet generation was demonstrated. Moreover, the temperature of the chip was uniform and sufficiently stable, meeting the requirement for the PCR process. In addition, the working conditions of the droplet-based PCR process in the 3D-printed chip were explored. Finally, the droplet-based PCR detection of miRNA-21 levels in the MCF-10A cells and MDA-MB-231 cells was successfully achieved using the 3D-printed chip. We expect that the enormous potential of 3D printing technology in the research area of microfluidics will be explored in the future.

Although various liver chips have been developed using emerging organ-on-a-chip techniques, it remains an enormous challenge to replicate the liver lobules with self-assembled perfusable hepatic sinusoid networks. Herein we develop a lifelike bionic liver lobule chip (LLC), on which the perfusable hepatic sinusoid networks are achieved using a microflow-guided angiogenesis methodology; additionally, during and after self-assembly, oxygen concentration is regulated to mimic physiologically dissolved levels supplied by actual hepatic arterioles and venules. This liver lobule design thereby produces more bionic liver microstructures, higher metabolic abilities, and longer lasting hepatocyte function than other liver-on-a-chip techniques that are able to deliver. We found that the flow through the unique micropillar design in the cell coculture zone guides the radiating assembly of the hepatic sinusoid, the oxygen concentration affects the morphology of the sinusoid by proliferation, and the oxygen gradient plays a key role in prolonging hepatocyte function. The expected breadth of applications our LLC is suited to is demonstrated by means of preliminarily testing chronic and acute hepatotoxicity of drugs and replicating growth of tumors in situ. This work provides new insights into designing more extensive bionic vascularized liver chips, while achieving longer lasting ex-vivo hepatocyte function.

Photocatalytic seawater splitting is an attractive way for producing green hydrogen. Significant progresses have been made recently in catalytic efficiencies, but the activity of catalysts can only maintain stable for about 10 h. Here, we develop a vacancy-engineered Ag 3 PO 4 /CdS porous microreactor chip photocatalyst, operating in seawater with a performance stability exceeding 300 h. This is achieved by the establishment of both catalytic selectivity for impurity ions and tailored interactions between vacancies and sulfur species. Efficient transport of carriers with strong redox ability is ensured by forming a heterojunction within a space charge region, where the visualization of potential distribution confirms the key design concept of our chip. Moreover, the separation of oxidation and reduction reactions in space inhibits the reverse recombination, making the chip capable of working at atmospheric pressure. Consequently, in the presence of Pt co-catalysts, a high solar-to-hydrogen efficiency of 0.81% can be achieved in the whole durability test. When using a fully solar-driven 256 cm 2 hydrogen production prototype, a H 2 evolution rate of 68.01 mmol h −1 m −2 can be achieved under outdoor insolation. Our findings provide a novel approach to achieve high selectivity, and demonstrate an efficient and scalable prototype suitable for practical solar H 2 production.

Immunosensors offer great promise for cancer diagnostics owing to their high sensitivity, specificity, and potential for miniaturization. However, conventional electrochemical immunosensors often rely on single-step enzyme reactions and single-biomarker detection, limiting their signal amplification capacity and diagnostic accuracy for complex diseases such as prostate cancer. Here, we presented an ultrasensitive cascade enzyme immunoassay electrochemical microfluidic chip (CEE-CHIP) for the simultaneous detection of multiple prostate cancer biomarkers. The system employed primary antibody-functionalized magnetic metal-organic frameworks (Fe3O4@MOF), followed by target antigens binding specifically and glucose oxidase-labeled secondary antibodies conjugating to form stable sandwich immunocomplexes. The functionalized Fe3O4@MOF were magnetically positioned within a microfluidic channel and a subsequent cascade reaction involving glucose oxidase generates H2O2. H2O2 was detected by a Co3O4/MXene-modified working electrode, resulting in amplified electrochemical signals. This system allowed for the simultaneous detection of multiple tumor markers, including prostate-specific antigen and Engrailed 2, with detection limits as low as 0.154 and 0.146 pg/mL, respectively. With a broad linear range (0.001−100 ng/mL), excellent sensitivity, and minimal sample consumption, the CEE-CHIP represented a promising non-invasive, point-of-care diagnostic platform for early prostate cancer detection and broader clinical applications.

Abstract Tumor cells (TCs) typically achieve brain metastasis through two distinct post-extravasation routes: invasion into the parenchyma or migration along the abluminal surface of vessels. However, the biomechanical cues dictating this route selection remain poorly understood. Here, we employed a triparallel extravasation-on-chip that enables independent modulation of key mechanical cues within the brain extravasation microenvironment. Traction force microscopy (TFM) and atomic force microscopy (AFM)-based mechanical characterization were used to quantify tumor-cell mechanical responses. We show that, compared with soft extravascular matrix, stiff extravascular matrix markedly suppresses invasion of TCs into the matrix after extravasation. Strikingly, this inhibitory effect can be reversed when vascular wall compactness is increased and fluid shear stress (FSS) is applied, even under stiff-matrix condition. To elucidate the mechanical basis underlying this adaptive invasion, AFM and TFM measurements revealed that tumor-cell stiffening, accompanied by increased F-actin organization, was associated with enhanced traction generation and matrix deformation, consistent with the increased matrix invasion observed under these conditions. These results suggest that tumor-cell stiffening represents a mechanoadaptive state, highlighting tumor-cell mechanical adaptation as an additional regulator of post-extravasation invasion beyond matrix compliance alone. This study provides a controllable platform to dissect TCs-matrix interfacial mechanics and elucidate how physical microenvironmental cues regulate organ-specific metastatic progression.

Tumor-on-a-chip platforms are increasingly recognized as promising preclinical tools for developing personalized therapies. However, effectively recapitulating tumor heterogeneity, three-dimensional architecture, and the dynamic in vivo microenvironment remains a significant challenge. Here, we developed magneto-responsive microcontainers for controllable culture of the patient-derived colorectal tumor cells (pCRCs) spheroids. Using microfluidic electrospray technology, microcontainers with a biocompatible carboxymethyl cellulose (CMC) core and a magnetic Fe 3 O 4 @alginate shell were fabricated for efficient harvest of pCRCs-spheroids. In addition, the microcontainers underwent rotational motion under an external magnetic field, thereby modulating local molecular transport. Moreover, integration of such pCRCs-laden microcontainer into a branched microfluidic chip enabled the generation of drug concentration gradients. Functional on-chip assays revealed heterogeneous drug responses among patient-derived spheroids and enabled preliminary comparison with the available clinical outcomes. Collectively, this magnetic chip-based platform offers a controllable in vitro approach for high-throughput, patient-specific chemotherapy screening in colorectal cancer.

Endothelial glycocalyx dysregulation remains the leading cause of the development and progression of cardiovascular diseases, which can increase the risk of organ failure and even cause death. However, the construction of a reliable in vitro drug screening model system to investigate how to prevent glycocalyx degradation is still challenging. Herein, an advanced and biomimetic vessel-on-a-chip based on the inverse opal hydrogel film has been designed for endothelial glycocalyx research and drug evaluation. The inverse opal film, with ordered and uniform pore structures, is prepared through replicating the closely packed nanoparticle assemblies. The proposed vessel-on-a-chip is generated from the integration of an inverse opal film with a custom-designed microfluidic chip. Benefiting from the three-dimensional (3D) ordered porous structures of inverse opal films and the biomimetic dynamic in vivo environment of the chip, endothelial cells and smooth muscle cells can be co-cultured on the inverse opal film of this microfluidic chip for the endothelial glycocalyx study. Specifically, the results demonstrate that the glycocalyx layer is susceptible to degradation under low shear stress, whereas such dysregulation can be effectively prevented through the administration of dexmedetomidine anesthesia. Furthermore, the specific mechanism underlying this glycocalyx-preserving effect of dexmedetomidine is demonstrated to be associated with mitigation of the low shear stress-induced oxidative stress in endothelial cells, thereby attenuating mitochondrial apoptosis. These features reveal that our vessel-on-a-chip provides a promising platform for preclinical drug screening and evaluation in cardiovascular diseases.

Spectral computing and reconstruction technology does not require the complex optical paths and optical components in traditional spectral detection, and can achieve the integration of the detection system. It has important application prospects in medical imaging, industrial inspection, remote sensing, and environmental monitoring. However, photodetectors (PDs) used for spectral reconstruction are often prepared with filters. Once the photodetectors under this scheme are prepared, they cannot be dynamically adjusted. Meanwhile, the extensive use of filters will also cause mutual constraints between spectral resolution and spatial resolution. Herein, we propose a multilayer organic–inorganic halide perovskite single-crystal heterojunction (MPSCH) for spectral reconstruction via solution-processed epitaxial growth, with a graded-bandgap structure of MAPbCl3/MAPbBr1.5Cl1.5/MAPbBr3/MAPbI1.5Br1.5/MAPbI2Br to achieve wavelength-dependent photon absorption across the vertical stack. Instead of relying on static physical filters, we exploit the nonlinear extraction of photogenerated carriers under varying electric fields. This bias-tunable mechanism allows for the dynamic modulation of spectral response, enabling a number of detection channels that significantly exceed the limit of physical filter arrays, thus facilitating higher spectral resolution without sacrificing spatial resolution. The unknown spectrum is fitted by using the Gaussian function through the least-squares method based on this device. The final structure can achieve spectral calculation and reconstruction within the range of 420 nm–740 nm. Compared with the commonly used 9/16/25 channels in filter array-based devices, our device can achieve more than 60 reconstruction channels. Single-point scanning imaging of the picture can be achieved through this device. The minimum mean square error (MSE) order of magnitude of the reconstructed spectrum calculated by it, compared with commercial spectrometers, can reach 10–4.

Rapid diagnosis of viral diseases in pepper (Capsicum annuum L.) is crucial for preventing pathogen spread, reducing yield losses, and supporting the green and sustainable development of the pepper industry. Here, we present an integrated, lowcost centrifugal microfluidic chip that simultaneously detects nucleic acids from six pepper viruses. All reagents are prestored on-chip as dried powders and are used to perform real-time fluorescence reverse transcription loop-mediated isothermal amplification (RT-LAMP). The developed assay showed strong agreement with conventional diagnostic methods of polymerase chain reaction (PCR). This platform offers a practical approach for rapid nucleic acid-based diagnostics and epidemiological monitoring of pepper viral diseases.

Spherical micro/nanorobots are reported as attractive candidates in nano-surgery, contamination degradation, analytical chemistry, medical engineering, etc. Unlike macroscopic robots, the standard fabrication and inspection strategy to control the high quality of microrobots are rarely explored before. Overcoming the geometrical discrepancy is now essential to keep an identical locomotion behavior for spherical microrobot individuals for complex usages. Herein, acoustic-based microbubble grippers are proposed controlled by a machine vision system to realize agile recognition and sorting of unqualified spherical microrobot models. Theoretical analysis and numerical simulation reveal the basic grabbing mechanisms via acoustic stimuli. The acoustic resonation of microbubbles can generate a locally enhanced acoustic field, attracting microrobots toward the bubble surface by the acoustic forces. The viscosity of microfluid makes it possible to instantaneously grab or release just by controlling the microbubble activation. Further, a machine vision-based monitor and control system is proposed to enable automatic control for the entire process. Based on extracted characteristic parameters, target microrobot models are quickly selected and grabbed out. It is expected that such a combination of microgrippers with an advanced control system will pave the way for high-qualified inspection for microrobots and improve their unique versatility in myriad micromanipulation scenarios.

Pharmacological chemotherapy remains a cornerstone in treating osteosarcoma (OS), where the application of drug combinations not only enhances therapeutic efficacy but also mitigates adverse side effects. However, the absence of an efficient and reliable drug screening platform poses a significant challenge in optimizing these combination therapies. In this study, we introduce a novel OS chip designed to facilitate the high-throughput and precise evaluation of drug combination efficacy, addressing this critical gap in OS treatment research. Leveraging the precise control of microfluidic electrospray technology, we successfully fabricated core–shell microcarriers (CSMs) encapsulating OS cells with uniform size and monodisperse distribution. Through three-dimensional culture, OS cell spheroids were efficiently formed within the CSMs, which were subsequently integrated into a microfluidic chip equipped with a concentration gradient generator and cell culture chambers. This innovative platform enables high-throughput screening of single drugs and diverse drug combinations. Our results demonstrate that this OS-on-a-chip exhibits significant potential for clinical drug screening, offering a robust tool for optimizing therapeutic strategies in OS treatment. Summary Innovative osteosarcoma-on-a-chip for high-throughput drug screening Core–shell microcarriers enable uniform 3D OS spheroid formation Microfluidic electrospray technology for precise drug combination testing Concentration gradient generator enhances multi-drug efficacy evaluation
