In surface-enhanced Raman scattering (SERS) detection research, the shape, structure, surface modification, and material selection of nanoparticles can significantly impact the SERS intensity. Petal-like gap-enhanced Raman tags (P-GERTs) possess numerous sharp tips and edges, which generate localized electric field enhancements, further amplifying the electric field enhancement effect on neighboring molecules and enhancing the SERS signal. Additionally, the surface of P-GERTs can be modified with functional molecules, enabling their application in the detection of disease biomarkers. Using COVID-19 as an example, the performance of P-GERTs in disease biomarker detection was validated, demonstrating that the signal intensity of this probe can reach 55 times that of regular gold nanoparticles and 36.7 times that of smooth shell gap-enhanced Raman tags (S-GERTs). Furthermore, in combination with magnetically retrievable magnetic bead substrates, the N-protein antigen was specifically detected in a one-step process. N-protein was detected within 15 min using a portable Raman spectrometer. The limit of detection (LOD) for the standard sample was 4.28 pg/mL, and the LOD for the actual throat swab sample system was 25.4 pg/mL. This workflow can be extended to the detection of other biomarkers, making it widely applicable.

Background As COVID-19 continues to spread globally, traditional emergency management measures are facing many practical limitations. The application of big data analysis technology provides an opportunity for local governments to conduct the COVID-19 epidemic emergency management more scientifically. The present study, based on emergency management lifecycle theory, includes a comprehensive analysis of the application framework of China’s SARS epidemic emergency management lacked the support of big data technology in 2003. In contrast, this study first proposes a more agile and efficient application framework, supported by big data technology, for the COVID-19 epidemic emergency management and then analyses the differences between the two frameworks. Methods This study takes Hainan Province, China as its case study by using a file content analysis and semistructured interviews to systematically comprehend the strategy and mechanism of Hainan’s application of big data technology in its COVID-19 epidemic emergency management. Results Hainan Province adopted big data technology during the four stages, i.e., migration, preparedness, response, and recovery, of its COVID-19 epidemic emergency management. Hainan Province developed advanced big data management mechanisms and technologies for practical epidemic emergency management, thereby verifying the feasibility and value of the big data technology application framework we propose. Conclusions This study provides empirical evidence for certain aspects of the theory, mechanism, and technology for local governments in different countries and regions to apply, in a precise, agile, and evidence-based manner, big data technology in their formulations of comprehensive COVID-19 epidemic emergency management strategies.

Background:The COVID-19 pandemic has placed severe restrictions on people’s behavior worldwide with school closures in many countries. These closures have shifted education from the classroom to the home. This change is unprecedented, and home-schooling has placed substantial stress on families across the world. As of 9 April 2020, 1.57 billion children were being educated by families that had little or no experience of protracted home-schooling. An essential but neglected issue related to COVID-19 is the psychological impact of home-schooling on family wellbeing, especially considering the other stressors they are experiencing including social isolation, fears of infection, frustration, boredom, inadequate information, and financial stress. This study explores the impact of home-schooling on family psychological wellbeing during COVID-19. These findings will help develop supports and interventions for this population. Methods:An exploratory qualitative study will be conducted using semi-structured interviews with a convenience sample of families (at least one parent and one child). Participants will be recruited using social media outlets, contacts of academic members and snowball sampling. Interviews will take place using Microsoft Teams and via telephone and recorded for transcription purposes. These transcripts will be analyzed using interpretative phenomenological analysis (IPA). Ethical approval has been granted by the Social Research Ethics Subcommittee (SRESC) Maynooth University (2407411). The Science Foundation Ireland (SFI) has funded the research with the COVID-19 rapid response grant. The dissemination of findings will be through open access journal publications, distribution of lay summaries, press release and policy papers.Discussion:The research findings will discuss the impact that home-schooling has had on family psychological wellbeing. It will examine how parents are managing their children’s education and learning while handling the other stresses associated with COVID-19. A deeper understanding of the impact of school closures and home-schooling on family processes is essential if the psychological wellbeing of families is to be protected and supported during challenging times such as health-related disasters.

With the prevalence of COVID-19, wearing medical surgical masks has become a requisite measure to protect against the invasion of the virus. Therefore, a huge amount of discarded medical surgical masks will be produced, which will become a potential hazard to pollute the environment and endanger the health of organisms without our awareness. Herein, a green and cost-effective way for the reasonable disposal of waste masks becomes necessary. In this work, we realized the transformation from waste medical surgical masks into high-quality carbon–nickel composite nanowires, which not only benefit the protection of the environment and ecosystem but also contribute to the realization of economic value. The obtained composite carbon-based materials demonstrate 70 S m–1 conductivity, 5.2 nm average pore diameters, 234 m2 g–1 surface areas, and proper graphitization degree. As an anode material for lithium-ion batteries, the prepared carbon composite materials demonstrate a specific capacity of 420 mA h g–1 after 800 cycles at a current density of 0.2 A g–1. It also displays good rate performance and decent cycling stability. Therefore, this study provides an approach to converting the discarded medical surgical masks into high-quality carbon nanowire anode materials to turn waste into treasure.

Bitespiramycin, has been shown to have a therapeutic effect against respiratory tract inflammation, including a potential effect against COVID-19. A current clinical trial in China showed that bitespiramycin was an effective treatment for severe pneumonia and intracranial infection. However, there is lack of an analytical method to elucidate the distribution of bitespiramycin. In this study, a highly sensitive, rapid and reliable UPLC–MS/MS method was developed to comprehensively characterize the bitespiramycin distribution in various bio-samples, which is significantly improved upon the published work. A rapid sample preparation method was developed by usingn-butanol as the solvent to extract bitespiramycin from different bio-samples. The extract was then directly analyzed by UPLC–MS/MS coupled with an alkaline-resistant column after centrifugation which avoids the time-consuming concentration process under nitrogen and redissolution. The method was employed to accurately quantify bitespiramycin and its metabolites in rat plasma, tissues, and human cerebrospinal fluid. Notably, the presence of bitespiramycin and its metabolites was identified for the first time in various rat organs including brain, testis, bladder and prostate as well as in human cerebrospinal fluid. This newly developed approach shows great promise for drug distribution assays including other antibiotics and can help elucidate the ADME of bitespiramycin.

In this work, we constructed an exonuclease III cleavage reaction-based isothermal amplification of nucleic acids with CRISPR/Cas12a-mediated pH-induced regenerative Electrochemiluminescence (ECL) biosensor for ultrasensitive and specific detection of SARS-CoV-2 nucleic acids for SARS-CoV-2 diagnosis. The triple-stranded nucleic acid in this biosensor has an extreme dependence on pH, which makes our constructed biosensor reproducible. This is essential for effective large-scale screening of SARS-CoV-2 in areas where resources are currently relatively scarce. Using this pH-induced regenerative biosensor, we detected the SARS-CoV-2 RdRp gene with a detection limit of 43.70 aM. In addition, the detection system has good stability and reproducibility, and we expect that this method may provide a potential platform for the diagnosis of COVID-19.

Herein, a novel and simple electrospray (ES) printing technique was developed for the fabrication of ultrathin graphene layers with precisely controlled nanometer-scale thickness, where graphene oxide (GO) was electrosprayed on wafers and subsequently chemically reduced into reduced GO (rGO). Utilizing that technique, we prepared ultrathin rGO in-plane graphene field-effect transistor (GFET)-based biosensors coupled with a portable prototype measuring system for point-of-care detection of pathogens. We illustrate the use of such prepared GFETs to detect COVID-19, using the SARS-CoV-2 nucleocapsid protein antigen (N-protein) and genomic viral RNA as detection targets. The electrosprayed and chemically reduced rGO films enhance the molecular detection in GFET sensors through significant local gating effects. The device detects the N-protein from the SARS-CoV-2 Omicron variant in a culture medium with an LOD of 1.44 PFU/mL and in clinical oropharyngeal samples with an LOD of 45 genome copies/mL in 5 min. It also successfully detects viral RNA in oropharyngeal swabs within 10 min. The GFET sensor responses were further analyzed using our proprietary wireless, miniaturized, and portable FET analyzer, coupled with a smartphone detecting app. Altogether, we present low-cost and mass-producible GFETs with high-quality graphene channels, enabling a portable, efficient, and accurate solution for point-of-care pathogen detection and in clinical testing. This technology has the potential to become a crucial tool in preventing future global epidemic outbreaks.

Both increasing emission of chlorophenol chemicals and plastic waste pollution are severe global environmental challenges, which is worth being explored to respond with a "waste eliminated by waste" coupling treatment strategy. Herein, we propose a one-step recyclable nano-MgO template strategy to prepare in-situ N, O-doped novel porous carbon coral reefs (PCNCR) directly derived from the mixture of real waste PET and melamine formaldehyde resin. The template precursor can be conveniently recycled by facile acetic acid pickling, vacuum evaporation and drying, and the whole preparing process does not involve any toxic solvents, massive consumption of pore-forming agents, or discharge of salty wastewater. The obtained PCNCRs have hierarchical porous structures and abundant N, O-doped species, and the textural parameters can be modulation linearly by varying the mass ratio of magnesium acetate to mixed waste plastics. The optimized PCNCR-1.25 exhibited remarkable adsorption capacity for chlorophenol contaminants (777.5 mg/g for 4-Chloro-3,5-dimethylphenol, PCMX and 538.74 mg/g for 4-Chloro-3-methylphenol, PCMC) in aqueous solution. Adsorption isotherm fitting studies revealed chlorophenol contaminants tended to be rapidly adsorbed on the PCNCR surface in the form of monolayer. And the system reached adsorption equilibrium at about 15 min, which was described by linear driving force model. Reusability test verified that PCNCR-1.25 had excellent adsorption stability for chlorophenol in continuous adsorption-regeneration cycles. DFT calculation illustrated the enhancement of chlorophenol adsorption by heteroatom species, especially nitrogen dopants. This work highlighted a low-cost and sustainable approach to defuse the dual environmental threat of chlorophenol emissions and plastic waste pollution simultaneously.

The study combined UHPLC-Q-Orbitrap-MS analysis with authentic standards, to create a novel strategy for isomers recognition and putative identification. Through the strategy, anti -Covid-19 Jinhua Qinggan Granule was found to comprise 28 isomers and 45 potential anti -Covid-19 constituents. The detection of three constituents (Danshensu, cryptotanshin, and tanshinone IIA) suggests Danshen as confidential additive. Based on this, 6 constituents are recommended as quality-marker candidates, including chlorogenic acid, acteoside, peimisine, baicalein, licoricesaponin H2, and tanshinone IIA. Obviously, the study can not only help the public to really understand the Granule’s formula and chemistry, but also facilitate its Pharmacopoeia collection in future.

Based on the drug repositioning strategy, niclosamide (NCL) has shown potential applications for treating COVID-19. However, the development of new formulations for effective NCL delivery is still challenging. Herein, NCL-embedded dry powder for inhalation (NeDPI) was fabricated by a novel spray freeze drying technology. The addition of Tween-80 together with 1,2-Distearoyl-sn-glycero-3-phosphocholine showed the synergistic effects on improving both the dispersibility of primary NCL nanocrystals suspended in the feed liquid and the spherical structure integrity of the spray freeze dried (SFD) microparticle. The SFD microparticle size, morphology, crystal properties, flowability and aerosol performance were systematically investigated by regulating the feed liquid composition and freezing temperature. The addition of leucine as the aerosol enhancer promoted the microparticle sphericity with greatly improved flowability. The optimal sample (SF − 80 D-N 20 L 2 D 2 T 1 ) showed the highest fine particle fraction of ∼47.83%, equivalently over 3.8 mg NCL that could reach the deep lung when inhaling 10 mg dry powders.

By exploring the China International Fair for Investment and Trade’s development process, this study analyzes its absolute advantages in future development, gradually lost comparative advantages, and potential crises. Through the data envelopment analysis model, the study analyzes its resource allocation efficiency based on two cooperation modes: traditional “Offline” investment mode and “Online + Offline” investment mode. Then we use vector autoregressive model to comprehensively investigate the impact and causality of the two input factors on output. We find that its comprehensive allocation efficiency presents a “U” shape that reflects the characteristics of its three stages: the first stage, 2001–2005; the second stage, 2006–2012; and the third stage, 2013–2020. The main factors affecting resource allocation efficiency are then deduced from the results: exhibition scale utilization, booth design innovation, project strength of participating enterprises, and the signing rate of overseas customers. The number of industrial and commercial groups (X 4 ) and participating countries and regions (X 6 ) have an important impact on the output indicators: signed contract projects (Y 1 ). The empirical results verify that the “Online + Offline” investment mode is an effective and suitable mechanism to solve the problem of cooperation and investment constrained by the COVID-19 pandemic. Based on the results of empirical analysis, this study posits the path and countermeasures of realizing the transformation to “cloud” exhibition in the post-pandemic era. Especially, we should focus on building new mechanisms of business environment that will promote the active participation of national, regional, and international industrial and commercial groups. The purpose is to continuously strengthen the foundation of cooperative trust and innovate a new trust model of online cooperative trading.

Loratadine (LORA), is a topical antihistamine utilized in the treatment of ocular symptoms of COVID-19. The study aimed to develop a Loratadine Nanostructured Lipid Carriers Ocugel (LORA-NLCs Ocugel), enhance its solubility, trans-corneal penetrability, and bioavailability. full-factorial design was established with 24 trials to investigate the impact of several variables upon NLCs properties. LORA-NLCs were fabricated by using hot melt emulsification combined with high-speed stirring and ultrasonication methods. All obtained formulae were assessed in terms of percent of entrapment efficiency (EE%), size of the particle (PS), zeta potential (ZP), as well as in-vitro release. Via using Design Expert® software the optimum formula was selected, characterized using FTIR, Raman spectroscopy, and stability studies. Gel-based of optimized LORA-NLCs was prepared using 4% HPMC k100m which was further evaluated in terms of physicochemical properties, Ex-vivo, and In-vivo studies. The optimized LORA-NLCs, comprising Compritol 888 ATO®, Labrasol®, and Span® 60 showed EE% of 95.78 ± 0.67%, PS of 156.11 ± 0.54 nm, ZP of −40.10 ± 0.55 Mv, and Qh6% of 99.67 ± 1.09%, respectively. Additionally, it illustrated a spherical morphology and compatibility of LORA with other excipients. Consequently, gel-based on optimized LORA-NLCs showed pH (7.11 ± 0.52), drug content (98.62%± 1.31%), viscosity 2736 cp, and Q12% (90.49 ± 1.32%). LORA-NLCs and LORA-NLCs Ocugel exhibited higher ex-vivo trans-corneal penetrability compared with the aqueous drug dispersion. Confocal laser scanning showed valuable penetration of fluoro-labeled optimized formula and LORA-NLCs Ocugel through corneal. The optimized formula was subjected to an ocular irritation test (Draize Test) that showed the absence of any signs of inflammation in rabbits, and histological analysis showed no effect or damage to rabbit eyeballs. Cmax and the AUC0–24 were higher in LORA-NLCs Ocugel compared with pure Lora dispersion-loaded gel The research findings confirmed that NLCs could enhance solubility, trans-corneal penetrability, and the bioavailability of LORA.
