Evaluation of photon-counting micro-CT scanning for imaging marine organisms

Mengjia Xue,Tianwu Xie,Qian Liu

Introduction: Anatomical information is crucial in marine biology research. Traditionally, destructive techniques, such as slice imaging of the sample, are used to image the inside of the sample, which are unsuitable for valuable or rare specimens. While energy-integrating detectors computed tomography (CT) can provide non-invasive anatomical information, it suffers from limitations such as low spatial resolution, high noise, and an inability to analyze material composition. Methods: Photon-counting (PC) micro-CT offers a promising alternative, enabling noninvasive imaging of both internal and external structures of marine organisms. Various species, including bivalves, conchs, and fish otoliths, were scanned using this advanced technology. The influence of different tube voltages on image quality is analyzed in this paper. Results: One of the key capabilities of PC micro-CT is the reconstruction of virtual monoenergetic images and material decomposition, which has been tested. To further enhance soft tissue visualization, iodixanol solution was employed for contrast imaging.Discussion: PC micro-CT provides high resolution both in non-contrast images and iodixanol-contrast images which digestive tracts and glands can be seen. Material decomposition of fish otoliths revealed the characteristic wheel-shaped distribution of calcium content. PC micro-CT has the potential ability for imaging marine organisms and promising future in broader utilization in marine biological research.

Bimodal Molecule as NIR-CT Contrast Agent for Hepatoma Specific Imaging

Tianbai Shuai,Yizhou Zhou,Guoqiang Shao,Rui Yang,Letian Wang,Jinglin Wang,Jie Sun,Longfei Ren,Jintao Wang,Yan Liao,Mian Wei,Qingxiang Xu,Yuyan Li,Li Zhao

With currently available molecular imaging techniques, hepatocellular carcinoma (HCC), a liver cancer with high mortality rates and poor treatment responses, is mostly diagnosed at its late stage. This is largely due to the lack of highly sensitive contrast agents with high liver specificity. Herein, we report a novel bimodal contrast agent molecule CNCI-1 for the effective detection of HCC at its early stage both in vitro and in vivo. The agent has high liver specificity with effective X-ray computed tomography (CT)/near-infrared (NIR) imaging functions. It has been successfully applied to in vivo NIR imaging with high sensitivity and high selectivity to the HCC region of the HepG2 tumor-xenografted mice model and LM3 orthotopic hepatoma mice model. Moreover, the agent was found to be noninvasive and hepatocarcinoma cells preferential. Furthermore, it also enhanced the tumor imaging by revealing the blood vessels nearby for the CT image acquisition in the VX2 orthotopic hepatoma rabbit model. Our design strategy provides a new avenue to develop the medical relevant bimodal contrast agents for diagnosis of HCC at its early stage.

PVP-Modified Multifunctional Bi2WO6 Nanosheets for Enhanced CT Imaging and Cancer Radiotherapy

Yifan Hao,Bo Peng,Chao Si,Bo Wang,Chengfeng Luo,Menghao Chen,Cheng Luo,Baijuan Gong,Zhimin Li

Malignant tumors are one of the main causes of human death. The clinical treatment of malignant tumors is usually surgery, chemotherapy, radiotherapy, and so forth. Radiotherapy, as a traditional and effective treatment method for cancer, is widely used in clinical practice, but the radiation resistance of tumor cells and the toxic side effects to normal cells are still the Achilles heel of radiotherapy. Multifunctional inorganic high-atom nanomaterials are expected to enhance the effect of tumor radiotherapy. Tungsten and bismuth, which contain elements with high atomic coefficients, have strong X-ray energy attenuation capability. We synthesized Bi2WO6 nanosheets (NSs) using a hydrothermal synthesis method and modified polyvinylpyrrolidone (PVP) on their surface to make them more stable. PVP–Bi2WO6 NSs have a variety of effects after absorbing X-rays (such as the photoelectric effect and Compton effect) and release a variety of particles such as photoelectrons, Compton electrons, auger electrons, and so forth, which can react with organic molecules or water in cells, generate a large number of free radicals, and promote cell apoptosis, thereby improving the effect of radiotherapy. We show through γ-H2AX and DCFH-DA probe analysis experiments that PVP–Bi2WO6 NSs can effectively increase cell DNA damage and reactive oxygen species formation under X-ray irradiation. Clone formation analysis showed that PVP–Bi2WO6 NSs can effectively suppress cell colony formation under X-ray irradiation. These versatile functions endow PVP–Bi2WO6 NSs with enhanced radiotherapy efficacy in animal models. In addition, PVP–Bi2WO6 NSs can also be used as contrast agents for X-ray computed tomography (CT) imaging with obvious effects. Therefore, PVP–Bi2WO6 NSs can be used as CT imaging contrast agents and tumor radiotherapy sensitizers and have potential medical applications.

Gram-Scale Synthesis of Renal-Clearable Tantalum Nanodots with High Water Solubility for Computed Tomography Imaging In Vivo

Yanzhi Dong,Gang Shu,Yibo Wei,Jinbin Pan,Dong Li,Shao-Kai Sun

Tantalum (Ta) emerges as a promising element for advanced computed tomography (CT) imaging probes owing to its high X-ray attenuation coefficient and excellent biocompatibility. Nevertheless, the synthesis of renally clear Ta-based imaging probes through simple methods remains a significant challenge. Herein, we introduce a simple and gram-scale approach for the synthesis of renal-clearable Ta nanodots with high water solubility for CT imaging in vivo. The Ta nanodots, coordination polymers, are fabricated via coordination reactions involving Ta(OH)5, citric acid (CA), and hydrogen peroxide. The Ta nanodots exhibit an ultrasmall hydrodynamic diameter (2.8 nm), high water solubility (1.88 g/mL, 688 mg Ta/mL), superior X-ray absorption capacity, gram-scale production capability (>10 g in lab synthesis), renal-clearable ability, and good biocompatibility. The Ta nanodots possess superior CT imaging efficacy across diverse tube voltages, enabling highly sensitive gastrointestinal CT imaging, renal CT imaging, and CT angiography (CTA). Moreover, Ta nanodots maintain robust CT imaging capabilities even at high X-ray energies, and Ta nanodots-based spectral CT achieves metallic artifacts-minimized CTA. The proposed Ta nanodots present substantial potential as a potent CT imaging probe for diagnosing various diseases.

Metabolizable dopamine-coated gold nanoparticle aggregates: preparation, characteristics, computed tomography imaging, acute toxicity, and metabolism in vivo

Yao Yu,Youshen Wu,Jiajun Liu,Ke Li,Daocheng Wu

To improve the computed tomography (CT) imaging ability and the toxicity of gold nanoparticles (AuNPs), hydrophilic dopamine (DPA)-coated gold nanoparticle (AuNP) (MSA AuNPs@DPA) aggregates (42 ± 2.65 nm) were obtained by assembling MSA AuNPs@DPA (5–6 nm) with the use of polyethyleneimine (PEI). They rapidly degraded into MSA AuNPs@DPA with PEI in blood. The acute toxicity test showed that the maximum tolerated doses of MSA AuNPs@DPA and MSA AuNPs@DPA aggregates were larger than 4.8 and 4.6 g kg−1, respectively, which are much higher doses than those of the commonly used citric acid-stabilized AuNPs (CA AuNPs) (2.97 g kg−1). The metabolic test in vivo showed that the elimination rates of MSA AuNPs@DPA aggregates and MSA AuNPs@DPA were 0.848 and 0.955, respectively. Most of the aggregates were eliminated by the kidney after 5 days and excreted by urine, whereas CA AuNPs remained in vivo and concentrated in some organs. The t0.5 values of MSA AuNPs@DPA and MSA AuNPs@DPA aggregates were 24.28 and 43.68 h, respectively, meaning that MSA AuNPs@DPA aggregates had a much longer circulation time. Similarly, the CT absorption value of MSA AuNPs@DPA aggregates was much higher than that of the commonly used nonionic iodinated CT contrast agent iohexol and CA AuNPs at the same concentration. Thus, MSA AuNPs@DPA aggregates possess characteristics such as ease of fabrication, long circulation time, hypotoxicity, and excellent CT absorption value, which suggest their great potential applications in vivo.

DiceCT applied to fossilized hard tissues: A preliminary case study using a miocene bird

Alida M. Bailleul,Jing Lu,Zhiheng Li

Iodine-based contrasting agents for computed tomography (CT) have been used for decades in medicine. Agents like Lugol's iodine enhance the contrasts between soft tissues and mineralized (skeletal) tissues. Because a recent study on extant avian heads showed that iodine–ethanol (I 2 E) is a better contrast enhancer overall than the standard Lugol's iodine, here, we tested if I 2 E could also enhance the CT contrasts of two fossilized skeletal tissues: bone and calcified cartilage. For this, we used a partial ankle joint from an extinct pheasant from the Late Miocene of Northwest China (Linxia Basin). The pre-staining CT scans showed no microstructural details of the sample. After being immersed into a solution of 1% I 2 E for 8 days and scanned a second time, the contrasts were drastically enhanced between the mineralized tissues (bony trabeculae and calcified cartilage) and the sediments and minerals inside vascular spaces. After three other staining-scanning cycles in 2%, 3%, and 6% I 2 E solutions, the best contrasts were obtained after immersion in 6% I 2 E for 7 days. Energy Dispersive Spectroscopy showed that iodine was preferentially absorbed by the mineralized tissues and the minerals in the vascular spaces, but not by the sediments. This method not only effectively increased the contrasts of two different fossilized skeletal tissues, it was also non-destructive and reversible because part of the fossil was successfully de-stained after a few days in pure ethanol. These preliminary results indicate that iodine-ethanol has the potential to be used widely in vertebrate paleontology to improve CT imaging of fossilized tissues. Research Highlights We immersed the partial ankle joint of a fossil bird from the Miocene of China into solutions of iodine-ethanol. The CT contrasts of the fossilized bony trabeculae and the calcified cartilage were positively enhanced; in a reversible and non-destructive manner.

Tetramethylpyrazine-caffeic Acid Hybrid CT-011 Exerts Dopaminergic Neuroprotection Through Inhibiting Microglia-mediated Neuroinflammation

Xie Min,Cheng Yu,Yang Lizhen,Lin Zijin,Shen Hao,Zhou Xinhua,Liu Zheng,Xing Jieyu,Zhou Sigui,Chen Haiyun

Parkinson’s disease (PD) is the second most prevalent neurological disorder without a clear etiology and specific cure. Microglia-mediated neuroinflammation plays a pivotal role in the pathogenesis and progression of PD, which is influenced by various factors and involves multiple signaling pathways. CT-011, synthesized by combining nitrone-tetramethyl pyrazine (TMP) and caffeic acid through an ester bond, has shown neuroprotection against ischemic stroke and glutamate-induced neuronal damage in animal models. The purpose of this study is to explore the impact of CT-011 on PD-related neuroinflammation and its potential in treating PD. Results showed that CT-011 significantly inhibited lipopolysaccharide (LPS)-induced release of pro-inflammatory cytokines and mediators in BV2 microglial cells. CT-011 also effectively mitigated LPS-induced mitochondrial membrane potential (MMP) reduction, mitochondrial and intracellular reactive oxygen species (ROS) production. Meanwhile, CT-011's anti-inflammatory effect was related to its inhibition of the TLR4-mediated MyD88/NF-B and PI3K-mediated AKT/GSK3β pathways. More significantly, CT-011 repressed the NLRP3 inflammasome activation. Additionally, CT-011 protected primary neurons against microglial-mediated neurotoxicity in vitro , and ameliorated dopaminergic neuronal damage in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice with a consistent anti-neuroinflammatory effect in vivo. These results demonstrate that CT-011 effectively inhibits microglia-mediated neuroinflammation and exerts neuroprotective effects on dopamine neurons, thus making it a promising therapeutic candidate for PD.

Reconstructing Nanohydroxyapatite Prosthesis Based on CT-Scanning Data and Its Application in Spinal Injury

Wang Jian,Li Ying,Xu Ting,Zhao Jie,Yuan Cuihua,Wen Baojun

This study investigated the nanohydroxyapatite (nHA) prosthesis application effect based on CT-scanning data in spinal injury. This study chose 26 spinal injury patients treated in our hospital from September 2017 to September 2018, who were randomly divided into two groups. nHA prosthesis based on CT-scanning data was implanted in the nHA group, whereas titanium mesh was implanted in the titanium mesh group. Consequently, osteoblasts were cultured to test the biological activity of nHA and titanium alloy. In cell tests, we found osteoblasts could better adhere to nHA, and proliferation and activity were higher when planted on nHA material. After surgical treatment, all patients’ spinal symptoms (VAS score, JOA score, and Cobb angle) had improved and did not cause obvious inflammatory foreign body reactions. During a two-year follow-up, the fusion time and support settlement in the nHA group was lower, and the vertebral fusion rate and ASIA score were higher than those in the titanium mesh group. Thus, CT-scanning data could further improve the vertebral fusion rate in the nHA group. Consequentially, nHA prosthesis based on CT-scanning data is a better choice for spinal injury therapy.

Anti-Scattering Perovskite Scintillator Arrays for High-Resolution Computed Tomography Imaging

Jinmei Song,Yuhong He,Haijing Hu,Mingbian Li,Chenglong Li,Bai Yang,Haotong Wei

Computed tomography (CT) imaging has emerge as an effective medical diagnostic technique due to its rapid and 3D imaging capabilities, often employing indirect imaging methods through scintillator materials. Arraying scintillators that can confine light scattering to enable high-resolution CT imaging remains an area of ongoing exploration for emerging perovskite scintillators. Here an anti-scattering cesium lead bromide (CsPbBr 3 ) scintillator array embedded within a polyurethane acrylate matrix for CT imaging using a cost-effective solution-processed method is reported. Due to the large refractive index contrast between the scintillator and matrix, photon propagation can be well confined within the CsPbBr 3 scintillator array to significantly suppress the light scattering and enhance the light collection efficiency by nearly two times compared to the monolithic film. Furthermore, the scintillator array exhibits low-dosage and high-resolution CT imaging capability by reconstructing a 3D tooth image with a good spatial resolution of 20.1 lp cm −1 at a low effective dose of 0.22 mSv. This work highlights that the CsPbBr 3 scintillator array is a highly promising candidate for CT imaging.

Large-scale synthesis of non-ionic bismuth chelate for computed tomography imaging in vivo

Yuping Zhang,Jianqi Deng,Mingbin Lang,Gang Shu,Jinbin Pan,Cai Zhang,Ran Cheng,Shao-Kai Sun

High atomic number elements-based X-ray computed tomography (CT) contrast agents offer a promising solution to address the inherent deficiencies of FDA-approved iodine contrast agents. However, they face substantial challenges in balancing imaging performance, safety, and large-scale production for clinical translation. Herein, inspired by the history of clinical gadolinium- and iodine-based contrast agents, we report a large-scale approach for synthesizing non-ionic bismuth (Bi) chelate for high-performance CT imaging in vivo . Bi-HPDO3A can be easily obtained from low-cost precursor within 4 steps at 6 g-scale. The non-ionic macrocyclic structure endows it with low osmolality, low viscosity, high stability, good renal clearable capability and biocompatibility. Additionally, Bi-HPDO3A realizes superior imaging performance across various in vivo applications, including gastrointestinal imaging, renal imaging, and computed tomography angiography (CTA). Especially, Bi-HPDO3A exhibits superior spectral imaging capability owing to the high K-edge of element Bi, achieving metal artifact-free CTA in vivo . The proposed Bi-HPDO3A that balances overall performance can serve as a high-performance CT contrast agent with potential for clinical translation.

Morphology-Controlled Synthesis and Multifunctional Luminescence/MRI/CT Imaging Applications of Octahedral BaGdF5:Eu3+ Phosphors

Jingjing Liu,Jing Huang,Meng Lv,Shuguo Wang,Gaoqi chen,Zhihao Zhang,Jiajun Tao,Xu Hou,Xudong Yang,Yu Wang,Shuo Wang,Fei Li,Tong Xu,Yunru Wan,Jiaying Zhao,Xiangyu Gong

RE 3+ -activated BaGdF 5 phosphors have broad applications in LED-based fluorescence displays and biological tracing. In this research, various types (GdF 3 :5%Eu 3+ , BaGdF 5 :5%Eu 3+ ) and different morphologies (peanut, octahedron, microsphere) of gadolinium-based fluoride micro/nano-materials with down-shifting luminescence were synthesized via the hydrothermal method. The coordination environments of Gd 3+ , Eu 3+ , Ba 2+ , and F - ions were tuned by systematically varying the reaction time (6–36 h), the initial pH (3–11), and surfactants/chelating agents (CTAB, EDTA-2Na, Na 3 Cit and PVP). The optimal synthesis conditions (36 h, pH=3) were identified through X-ray diffraction (XRD) and scanning electron microscopy (SEM) analysis. The samples were comprehensively characterized using thermogravimetric analysis (TGA), Brunauer–Emmett–Teller (BET) surface area measurements, Fourier-transform infrared (FT-IR) spectroscopy, and photoluminescence (PL) spectroscopy. The octahedral BaGdF 5 :5%Eu 3+ particles synthesized with CTAB demonstrated good thermal stability and the strongest fluorescence emission intensity at 595 nm, producing an orange-red light with CIE chromaticity coordinates of (0.5141, 0.4845). The luminescence intensity was 2.23 times greater than that of samples prepared with PVP. Moreover, the signal intensity in MRI T 1 -weighted imaging exhibits a positive correlation with Gd 3+ concentration. The composite material containing 2% wt% BaGdF 5 :5% Eu 3+ within the chitin matrix demonstrated optimal performance in CT imaging. Research indicates that the synthesised octahedral BaGdF 5 :5% Eu 3+ compound holds potential applications for down-shifting luminescence and multimodal MRI/CT imaging.

Rod-shape MSN@MoS2 Nanoplatform for FL/MSOT/CT Imaging-Guided Photothermal and Photodynamic Therapy

Lifang Yang,Jinping Wang,Shan Yang,Qianglan Lu,Peishan Li,Nan Li

Rod-shape nanoplatform have received tremendous attention owing to their enhanced ability for cell internalization and high capacity for drug loading. MoS 2 , widely used in electronic devices, electrocatalysis, sensor and energy-storage, has been studied as photothermal agents over the years. However, the efficacy of rod-shape MoS 2 based photothermal agents for photothermal therapy has not been studied before. Here, a near-infrared (NIR) light-absorbing MoS 2 nanosheets coated mesoporous silica nanorods with human serum albumin (HSA) modifying and Ce6 loading (MSNR@MoS 2 -HSA/Ce6) were constructed for combined photothermal and photodynamic therapy. Methods : The near-infrared (NIR) light was used to trigger the synergistic anti-tumor therapy. In addition, breast cancer cell line was applied to evaluate the in vitro anti-tumor activity. The multi-modal imaging capacity and tumor-killing efficiency of the designed nanocomposites in vivo was also demonstrated with the 4T1 tumor-bearing mouse model. Results : These nanocomposites could not only perform NIR light triggered photodynamic therapy (PDT) and photothermal therapy (PTT), but also achieve in vivo fluorescence (FL) /multispectral optical tomography (MSOT)/X-ray computed tomography (CT) triple-model bioimaging. What's more, the rod-shape nanoplatform could be endowed with better anti-tumor ability based on the EPR effect and HSA-mediated active tumor targeting. At the same time, the hyperthermia generated by MoS 2 could synergistically improve the PDT effect with the acceleration of the blood flow, leading to the increase of the oxygen level in tumor tissue. Conclusion : MSNR@MoS 2 -HSA/Ce6 proves to be a promising multi-functional nanoplatform for effective treatment of tumor.
