Self-Assembled Cationic Nanoparticles Combined with Curcumin against Multidrug-Resistant Bacteria

Jian Bin Zhen,Jiajia Yi,Huan Huan Ding,Ke-Wu Yang

The overuse of antibiotics exacerbates the development of antibiotic-resistant bacteria, threatening global public health, while most traditional antibiotics act on specific targets and sterilize through chemical modes. Therefore, it is a desperate need to design novel therapeutics or extraordinary strategies to overcome resistant bacteria. Herein, we report a positively charged nanocomposite PNs-Cur with a hydrodynamic diameter of 289.6 nm, which was fabricated by ring-opening polymerization of ε-caprolactone and Z-Lys-N-carboxyanhydrides (NCAs), and then natural curcumin was loaded onto the PCL core of PNs with a nanostructure through self-assembly, identified through UV–vis, and characterized by scanning electron microscopy (SEM) and dynamic light scattering (DLS). Especially, the self-assembly dynamics of PNs was simulated through molecular modeling to confirm the formation of a core–shell nanostructure. Biological assays revealed that PNs-Cur possessed broad-spectrum and efficient antibacterial activities against both Gram-positive and Gram-negative bacteria, including drug-resistant clinical bacteria and fungus, with MIC values in the range of 8–32 μg/mL. Also, in vivo evaluation showed that PNs-Cur exhibited strong antibacterial activities in infected mice. Importantly, the nanocomposite did not indeed induce the emergence of drug-resistant bacterial strains even after 21 passages, especially showing low toxicity regardless of in vivo or in vitro. The study of the antibacterial mechanism indicated that PNs-Cur could indeed destruct membrane potential, change the membrane potential, and cause the leakage of the cytoplasm. Concurrently, the released curcumin further plays a bactericidal role, eventually leading to bacterial irreversible apoptosis. This unique bacterial mode that PNs-Cur possesses may be the reason why it is not easy to make the bacteria susceptible to easily produce drug resistance. Overall, the constructed PNs-Cur is a promising antibacterial material, which provides a novel strategy to develop efficient antibacterial materials and combat increasingly prevalent bacterial infections.

Solvation effect on photophysical and photochemical properties of mono-biotinylated curcumin

Chen-Chen Zhao,Yang-Lin Jiang,Rong-Yao Gao,Hai-Dan Yao,Peng Wang,Jian-Ping Zhang

Solvent effects on photophysical and photochemical properties of mono-biotinylated curcumin ( mbt -Cur) in comparison with curcumin (Cur) were studied. Same with Cur, mbt -Cur dominantly adopts ‘enol-keto’ configuration in all solvents stabilized by intramolecular H-bond. In non-protic solvents, the photophysical properties of mbt -Cur depend on the solvent polarity parameter Δ f . In H-bond solvent methanol, dramatic deviation of all photophysical properties from the correlation established in non-H-bond solvent occurred. H-bond capability of solvent was found to determine the excitation dynamic mechanism of mbt -Cur at a microsecond timescale. This research laid the foundation for further application of mbt -Cur in biological systems.

Protective effects of curcumin against rat intestinal inflammation‑related motility disorders

Yang Yao,Ranyuan Luo,Shu Xiong,Chang Zhang,Yukun Zhang

Intestinal inflammation frequently occurs alongside dysmotility, which is characterized by altered myosin light chain phosphorylation levels. Curcumin, an active component from the ginger family, is reported to confer anti‑inflammatory effects. However, the effects of curcumin on both diarrhea and constipation associated inflammation remains to be elucidated. The present study was designed to investigate the effects of curcumin on diarrhea and constipation and to determine the related mechanisms. Sprague‑Dawley rats were used to establish diarrhea and constipation models via intracolonic acetic acid (4%) instillation or cold water gavage for 2 weeks, respectively. Blood samples were collected to measure the serum levels of the cytokines TNF‑α and IL‑1β using ELISA kits. Western blotting was performed to measure NF‑κB, RhoA, Rho‑related kinase 2, phosphorylated MLC20, phosphorylated myosin phosphorylated target subunit 1, 130k Da‑MLC kinase (MLCK), c‑kit tyrosine kinase protein expression, and reverse transcription‑quantitative PCR was conducted to measure MLCK expression levels. The results indicated that curcumin reversed the elevations in the pro‑inflammatory cytokines IL‑1β and TNF‑α by inhibiting the NF‑κB pathway in rats with diarrhea and constipation. The results also indicated that myosin light chain (MLC) phosphorylation in intestinal smooth muscle was positively and negatively associated with the motility of inflammation‑related diarrhea and constipation in rats, respectively. Curcumin significantly reversed the increased MLC phosphorylation in the jejunum of the rats with diarrhea, significantly enhanced the reductions in inflammatory mediators, including TNF‑α and IL‑1β, of rats with constipation and significantly ameliorated the related hyper‑motility and hypo‑motility in rats with both diarrhea and constipation. In conclusion, the potential roles of the MLC kinase, c‑kit tyrosine and Rho A/Rho‑associated kinase 2 pathways, which are involved in curcumin‑induced amelioration of inflammation‑related diarrhea and constipation, were explored in the present study. Results from the present study suggested that curcumin has potential therapeutic value for treating intestinal inflammation and inflammation‑related motility disorders.

Curcumin nanoparticles incorporated in PVA/collagen composite films promote wound healing

QingQing Leng,Yue Li,XianLun Pang,BiQiong Wang,ZhouXue Wu,Yun Lu,Kang Xiong,Ling Zhao,Ping Zhou,ShaoZhi Fu

Skin repair remains a common problem in plastic surgery. Wound dressing plays an important role in promoting local skin healing and has been widely studied. This study aimed to manufacture a composite film (CPCF) containing curcumin nanoparticles, collagen, and polyvinyl alcohol (PVA) to effectively promote the healing of skin wounds. Sustained drug release from the composite film provides long-term protection and treatment for skin wounds. Both antibacterial property and good histocompatibility of the CPCF were examined by analyzing antibacterial activity and cytotoxicity to validate its applicability for wound management. Moreover, in vivo studies proved that the CPCF had a rapid healing rate of 98.03%±0.79% and mature epithelialization on day 15 after surgery. Obvious hair follicles and earlier re-epithelialization was also noticed in the CPCF group using H&E staining. The result of Masson’s trichrome staining confirmed that CPCF could promote the formation of collagen fibers. In summary, CPCF may be promising as a wound dressing agent in wound management owing to its rapid wound-healing effects.

Spectral identification and detection of curcumin based on lanthanide upconversion nanoparticles

Litao Liu,Ruinian Hua,Xuan Zhang,Xiang Li,Wei Zhang,Xiaohui Qi,Tieying Zhang,Siwei Yang,Heqiu Zhang,Hongwei Liang

Curcumin is a natural organic substance that has attracted much attention because of its medicinal properties. Because of its immunomodulatory, anti-inflammatory, analgesic and anticarcinogenic effects, medicines containing curcumin are used to treat fever, jaundice, and skin diseases, especially cancers. The luminescence resonance energy transfer (LRET) technique is a powerful and widely used analytical method to verify analytes. In this work, we propose core/shell-structured upconversion nanoparticles (UCNPs) modified with amino groups as a luminescence probe for curcumin detection via LRET; the method is highly sensitive, rapid, easy to operate, and simple and has a low detection limit. In the detection process, curcumin can be brought into close proximity to the core/shell-structured UCNPs@NH 2 via electrostatic attraction and absorb the luminescence of the UCNPs. The results show that the detection limit of curcumin is 0.625 ng/mL. This study indicates that core/shell-structured UCNPs@NH 2 could be used as a valuable probe to detect curcumin.

A Novel Curcumin-Loaded Nanoplatform Alleviates Osteoarthritis by Inhibiting Chondrocyte Ferroptosis

Weibei Sheng,Aikang Li,Yaohang Yue,Qichang Wang,Fei Yu,Jian Weng,Jianjing Lin,Yingqi Chen,Hui Zeng,Deli Wang,Jun Yang,Peng Liu

Osteoarthritis (OA) is a chronic degenerative joint disease characterized by the degradation of articular cartilage. Recent studies have demonstrated that chondrocyte ferroptosis plays a crucial role in the progression of OA. Consequently, developing nanomedicines that suppress chondrocyte ferroptosis is a promising strategy for OA treatment. However, there are few reports on nanomedicines specifically targeting chondrocyte ferroptosis for OA therapy. In this study, Curcumin-loaded nanoparticles (Cur-NPs) are fabricated to suppress chondrocyte ferroptosis by regulating reactive oxygen species (ROS), ferrous ion (Fe 2 ⁺), and Acyl-CoA Synthetase Long-Chain Family Member 4 (ACSL4) levels of chondrocyte. This is achieved by combining the functions of curcumin and an amphiphilic block copolymer with ROS scavenging and iron-chelating properties. The in vitro anti-ferroptotic effects of Cur-NPs are thoroughly investigated. The findings indicate that Cur-NPs decrease the expression of ferroptosis markers such as ROS, Fe 2 ⁺, and ACSL4, while protecting the mitochondrial membrane potential of chondrocytes. Additionally, Cur-NPs attenuated lipid peroxidation in chondrocytes. Furthermore, Cur-NPs significantly reduced the expression of the catabolic factor Matrix Metallopeptidase 13 (MMP13) and increased the expression of the anabolic factor Collagen type II (Col II) in vitro. This study demonstrates that Cur-NPs exhibit enhanced chondroprotective effects through anti-ferroptotic actions, presenting a promising approach for inhibiting chondrocyte ferroptosis using bioactive nanomaterials in OA treatment.

The Stability, Sustained Release and Cellular Antioxidant Activity of Curcumin Nanoliposomes

Xing Chen,Li-Qiang Zou,Jing Niu,Wei Liu,Sheng-Feng Peng,Cheng-Mei Liu

Curcumin is a multifunctional and natural agent considered to be pharmacologically safe. However, its application in the food and medical industry is greatly limited by its poor water solubility, physicochemical instability and inadequate bioavailability. Nanoliposome encapsulation could significantly enhance the solubility and stability of curcumin. Curcumin nanoliposomes exhibited good physicochemical properties (entrapment efficiency = 57.1, particle size = 68.1 nm, polydispersity index = 0.246, and zeta potential = −3.16 mV). Compared with free curcumin, curcumin nanoliposomes exhibited good stability against alkaline pH and metal ions as well as good storage stability at 4 °C. Curcumin nanoliposomes also showed good sustained release properties. Compared with free curcumin, curcumin nanoliposomes presented an equal cellular antioxidant activity, which is mainly attributed to its lower cellular uptake as detected by fluorescence microscopy and flow cytometry. This study provide theoretical and practical guides for the further application of curcumin nanoliposomes. Keywords:curcumin nanoliposomes;stability;sustained release;cellular antioxidant activity;cellular uptake

Sensitizing nanoparticle based platinum(IV) drugs by curcumin for better chemotherapy

Xiang Kang,Chen Zhao,Lesan Yan,Ruogu Qi,Xiabin Jing,Zehua Wang

A polymer-cisplatin(IV) conjugate was prepared by attaching Pt(IV)-COOH to a biodegradable amphiphilic block copolymer containing pendant OH groups. The conjugate can assemble into micelles (M(Pt)) with a mean diameter of ca. 169   nm. Further, curcumin (CM) was used to sensitize platinum drug based nanoparticles to overcome cisplatin resistance and enhance antitumor efficacy. In vitro studies showed that M(Pt)/CM combinations had great synergistic effect both on cisplatin sensitive and cisplatin resistant cell lines (A2780 and A2780DDP). In vivo studies showed that M(Pt)/CM had a much lower systemic toxicity and an enhanced antitumor efficacy compared to cisplatin alone or the corresponding cisplatin/CM combinations. Therefore, polymer-cisplatin(IV) conjugate with small molecules that serve as a non-cytotoxic or minimally cytotoxic sensitizer or enhancer provide a promising strategy, which may have potential clinical implications in the near future.

Anti-inflammatory effect of combined tetramethylpyrazine, resveratrol and curcumin in vivo

Chen Long,Liu Tianjun,Wang Qiangsong,Liu Juan

Background Resveratrol and curcumin, as natural flavones products, have good therapeutic effect in acute and chronic inflammation; on the other hand, tetramethylpyrazine (TMP) has angiogenesis and vessel protection effect as well as anti-inflammatory function. In this paper, the anti-inflammatory effect of the tetramethylpyrazine, resveratrol and curcumin (TRC) combination in acute and chronic inflammation was reported in vivo. Methods The dose of the combined three natural products was optimized based on the acute paw swelling mouse model with a Uniform Design methodology. The therapeutic effect of TRC combination on chronic inflammation was investigated by using the collagen-induced arthritis (CIA) rat model based upon the following indexes: the volume of paw swelling, arthritis score, serum mediators and histological examination as well as immunohistochemical staining. The levels of alanine aminotransferase (ALT) and aspartate transaminase (AST) in serum were measured and the pathological sections of liver and kidney were analysed. LD50 was measured based on the acute oral toxicity (AOT) standard method. Results The best formulation was the three components combined at the same mass proportion revealed by the Uniform Design methodology. This combination could significantly reduce the paw swelling in acute paw swelling mouse model, could reduce paw swelling and alleviate the damage in joint structural of ankle, cartilages and fibrous tissue in CIA rat model. The dose relationship was clear in both cases. Immunohistochemical staining of ankle tissue revealed that TRC combination was able to inhibit the expression of NF-κB p65 and TNF-α which were closely related to the inflammatory process. Analysis of serum mediators revealed TRC combination could inhibit the production of TNF-α, IL-1β, and IL-6 in the serum. Toxic study revealed this formulation was low toxic, LD50 was larger than 5 g/kg, both the level of ALT and AST and histopathology in the liver and kidney exhibited no distinctions between the TRC combination and the blank group, no mortality occurred at the administered doses of 5 g/kg.Conclusion sThe results showed this formulation could provide a novel potent treatment for acute and chronic inflammation (RA) without side effect like gastric injury occurring in NSAIDs.

Magnetic molecularly imprinted polymers for spectrophotometric quantification of curcumin in food

Xiuying Liu,Lijie Zhu,Xue Gao,Yuxin Wang,Haixia Lu,Yiwei Tang,Jianrong Li

Herein, we present a simple and rapid method for monitoring curcumin in food samples using a magnetic molecularly imprinted technique combined with ultraviolet–visible (UV–Vis) spectrophotometry. Magnetic molecularly imprinted polymers (mag-MIPs) were first synthesized by fabricating MIPs on to the surface of Fe 3 O 4 nanobeads using curcumin as a template and methacrylic acid as a functional monomer. The obtained mag-MIPs were evaluated in detail with different techniques (such as binding isotherm, Scatchard analysis, and selectivity) and various adsorption experiments. Finally, mag-MIPs were constructed and UV–Vis spectrophotometry was used to quantify curcumin under optimized conditions. Good recoveries between 79.37% and 88.89% were obtained with the limits of detection and quantification of 1.31 and 4.38   μg/mL, respectively.

Photoenhanced gene transfection by a curcumin loaded CS-g-PZLL micelle

Jian-Tao Lin,Wen-Jia Pan,Jun-Ai Zhang,Wei Wang,Jia Zhong,Jia-Min Su,Tong Li,Ying Zou,Guan-Hai Wang

The codelivery of drug and gene is a promising method for cancer treatment. In our previous works, we prepared a cationic micelles based on chitosan and poly-( N -3-carbobenzyloxylysine) (CS-g-PZLL), but transfection ratio of CS-g-PZLL to Hela cell was low. Herein, to improve the transfection efficiency of CS-g-PZLL, curcumin was loaded in the CS-g-PZLL micelle. After irradiation, the obtained curcumin loaded micelle showed a better transfection, and the p53 protein expression in Hela cells was higher. The apoptosis assay showed that the complex could induce a more significant apoptosis to Hela cells than that of curcumin or p53 used alone, and the curcumin loaded micelle inducing apoptosis was best after irradiation. Therefore, CS-g-PZLL is a safe and effective carrier for the codelivery of drug/gene, and curcumin could be used as a photosensitizer to induce a photoenhanced gene transfection, which should be encouraged in improving transfection and tumor therapy.

Curcumin- and quercetin-functionalized polypropylene membranes as active food packaging material

Zeynep Karahaliloğlu,Baki Hazer

A wide range of active agents, synthetic and natural agents such as essential oils, chitosan and polyphneols consisting of curcumin, gallic acid, anthocyanins, and catechins have been used in order to develop antimicrobial packaging systems, and among them, natural polyphenolic compounds, specially curcumin (Cur) has great potential due to effective biological activities in developing food packaging material. Quercetin (Quer) is also the mostly studied flavonol as a color-changing indicator in the food industry and has been already developed as a realistic alternative for smart and active food packaging. The reason for choosing these two polyphenolic compounds is that they simultaneously possess many beneficial properties such as antioxidant, antibacterial, antiviral, antitumoral, and anti-inflammatory effects. Additionally, the main objective of the study is to combine polypropylene (PP), which is the most preferred and cost-effective polymer in the packaging industry, with these active ingredients, rather than using more expensive polymer types. In this context, PP-Quer or PP-Cur membranes, which are new experiences based on these literatures were chemically characterized by Fourier transform infrared spectroscopy, and the surface morphology of these composite membranes was characterized by scanning electron microscopy. The antibacterial response against gram-positive ( Staphylococcus aureus ) and gram-negative ( Escherichia coli ) bacteria species was investigated. Furthermore, the reactive oxygen species generation and anticancer activity of these composite membranes using human colorectal adenocarcinoma (HT-29) were observed. We proposed that PP-Quer or PP-Cur composite membranes can be a potential candidate as active packaging material in the food industry.
