In this work, the supersensitive and selective determination of lincomycin (Lin) was achieved using a novel electroluminescent (ECL) aptasensor based on the synergistic integration of gold functionalized upconversion nanoparticles (UCNPs) and thiolated 3,4,9,10-perylene tetracarboxylic acid (PTCA). The integration of two luminophores of UCNPs and PTCA combined the merits of the cathodoluminescence stability of UCNPs and the high quantum yield of PTCA, which significantly promoted the ECL signal and analytical performance of the proposed sensor. The introduction of gold nanoparticles in UCNPs can not only improve the conductivity and ECL performance of UCNPs but also cause them to easily integrate with thiolated PTCA (t-PTCA) via an Au-S bond. The ECL signal of UCNPs@Au/t-PTCA/GCE was almost twice as strong as that of t-PTCA/GCE and tenfold higher than that of UCNPs@Au/GCE. Because of the non-conductive protein of the Lin aptamer, the ECL intensity of apt/UCNPs@Au/t-PTCA/GCE noticeably decreased. In the presence of Lin, the aptamer was pulled down from the sensing interface, resulting in the recovery of the ECL intensity of the sensor. Under optimal conditions, our proposed sensor can quantify the concentration of Lin in the range from 1.0 × 10−15 to 1.0 × 10−7 M with a low detection limit of 2.4 × 10−16 M (S/N = 3), exhibiting high sensitivity and specificity for the determination of Lin.

An ultrasensitive sandwich-type electrochemiluminescence (ECL) immunosensor was developed for detection of prostate specific antigen (PSA) using an amplification strategy based on ZnO nanorods- l -cysteine-luminol nanocomposites and the biotin-streptavidin system. The biotin-streptavidin system served as a capture probe to increase the number of antibodies. ZnO nanorods not only acted as a nanocarrier that increased the number of luminol molecules and secondary antibodies, but also enhanced the ECL signal of luminol-H 2 O 2 system by promoting H 2 O 2 decomposition, which can further increase ECL intensity. Under optimized conditions, the proposed immunosensor demonstrated excellent analytical performance with a wide linear detection range of 0.03 pg mL −1 to 30 ng mL −1 and a detection limit of 0.01 pg mL −1 (the detection limit in real samples was 0.021 pg mL −1 ). This method exhibited excellent stability, reproducibility, and selectivity. In addition, the results of PSA determinations in human serum samples obtained using the proposed immunosensor were consistent with data collected using the commercial chemiluminescence immunoassay analyzer.

Recent years have witnessed the great development of electrochemiluminescence (ECL) as a platform for the analytical processes in many fields, which includes pharmaceutical analysis, immunoassay, DNA analysis as well as clinical analysis. Herein, a novel electrochemiluminescence (ECL) sensor was fabricated for ultrasensitive detection of atropine based on Ru(bpy) 3 2+ /carbon nanospheres/Nafion composite film. The carbon nanospheres (CNPs) in composite film were obtained through a hydrothermal method. Then, electrochemical and ECL behaviors of the proposed sensor were investigated. Under the optimal conditions, the constructed atropine sensor showed a wide linear range from 0.5 to 250 μM with a detection limit of 0.17 μM (at signal-to-noise ratio of 3). Moreover, the results indicated that the CNPs-based ECL sensor exhibited excellent sensitivity and favorable stability, which might be ascribed to the high surface area and the good electrical conductivity of CNPs. Furthermore, the ECL sensor was applied for the determination of atropine in atropine sulfate injection samples and displayed a desirable result. The proposed ECL sensor could be easily obtained and be promising used for drug assay.

In this work, we designed an ECL ratiometric biosensor with a three-stranded Y-type DNA (Y-DNA) probe and induced a hybridization chain reaction (HCR) for the highly sensitive detection of SARS-CoV-2 nucleic acid. The important component of this system is the self-assembled Y-Shaped probe based on three nucleic acids. Y1, Y2, and Y3 can be linked by complementary base pairing to Hairpin1 (H1), Hairpin2 (H2), and Ru modified DNA (Ru1), respectively. H1 and H2 can trigger the HCR reaction when activated by the SARS-CoV-2 RdRp gene and the 5′ end of Ru1. The 5′ end of Ru1 is modified with the Ru complex, which can produce a strong electrochemiluminescence luminescence signal at 620 nm under an applied voltage. Through the amplification of Y-DNA-induced HCR reaction, Ru1 on the electrode surface gradually increased, the ECL signal at 460 nm was gradually quenched, and the signal at 620 nm was steadily generated. The SARS-CoV-2 RdRp gene can be quantified according to the degree of decrease of ECL signal at 460 nm and the increase of ECL signal at 620 nm. Combining the two signal amplification strategies, this ratiometric ECL biosensor can accurately and efficiently detect the target gene with a detection limit of 59 aM.

We propose a novel washing-free electrochemiluminescence (ECL) biosensor for the simultaneous detection of two types of N6 methyladenosines-RNAs (m6A-RNAs), which are potential cancer biomarkers, on the basis of binding-induced DNA strand displacement (BINSD). The biosensor integrated a tri-double resolution strategy that combined spatial and potential resolution, hybridization and antibody recognition, and ECL luminescence and quenching. The biosensor was fabricated by separately immobilizing two ECL reagents (gold nanoparticles/g-C3N4 nanosheets and ruthenium bipyridine derivative/gold nanoparticles/Nafion) and the capture DNA probe on the two sections of glassy carbon electrode. As a proof of concept, m6A-Let-7a-5p and m6A-miR-17-5p were chosen as model analytes, while m6A antibody-DNA3/ferrocene-DNA4/ferrocene-DNA5 was designed as an m6A-binding probe and DNA6/DNA7 was designed as a hybridization probe with DNA3 to release the quenching probes ferrocene-DNA4/ferrocene-DNA5. The recognition process led to the quenching of the ECL signals from both probes via BINSD. The proposed biosensor has the advantage of being washing-free. The ECL methods using the fabricated ECL biosensor with the designed probes exhibited a low detection limit of 0.03 pM for two m6A-RNAs and high selectivity. This work reveals that this strategy is promising for developing an ECL method for the simultaneous detection of two m6A-RNAs. The proposed strategy could be expanded to develop the analytical methods for the simultaneous detection of other RNA modifications by changing the antibody and hybridization probe sequences.

Graphdiyne (GDY) is considered as another promising carbon material in biosensing field. However, the application of GDY in ECL sensing is in its infancy stage, and different roles of GDY are urgent to be discovered. In this work, strong anodic Ru(bpy) 3 2+ electrochemiluminescence (ECL) was obtained at a cucurbit[8]uril (Q[8]) modified electrode. Although GDY exhibited electrocatalytic effect on the oxidation of Ru(bpy) 3 2+ , an apparent inhibiting effect of GDY on the ECL signal was observed due to the energy transfer. An “on-off-on” mode ECL sensor was fabricated to detect pancuronium (PMD) based on the competitive host–guest interaction between ferrocene (Fc), PMD and Q[8]. Fc modified single-stranded DNA was immobilized onto the surface of GDY, which could be attached onto the electrode through the interaction between Fc and Q[8]. The quenching effect of GDY could apparently decrease ECL signal. PMD exhibited stronger combining ability with Q[8] than Fc, and could replace Fc from the cavity of Q[8], leading to the recovery of ECL signal. The variation of ECL intensity changed linearly with the PMD concentration in the range of 10 nM to 50 μM with a detection limit of 5 nM (3σ). The proposed sensor has high sensitivity, outstanding specificity and accuracy, indicating that GDY has promising potential application in ECL sensing field based on its inhibiting effect.

The successful application of electrochemiluminescence (ECL) in various fields required continuous exploration of novel ECL signal emitters. In this work, we have proposed a pristine ECL luminophor named NiRu MOFs, which owned extremely high and stable ECL transmission efficiency and was synthesized via a straightforward two-step hydrothermal pathway. The foundation framework of pure Ni-MOFs with the initial structure was layered-pillared constructed by the coordinated octahedrally divalent between nickel and terephthalic acid (BDC). The terephthalates were coordinated and pillared directly to the nickel hydroxide layers and the three-dimensional framework was formed, which had a weak ECL response strength. Then, the ruthenium pyridine complex was recombined with pure Ni-MOFs to produce NiRu MOFs and part of the introduced ruthenium was atomically dispersed in the layered-pillared structure through an ion-exchange method, which led to the ECL luminous efficiency being significantly boosted more than pure Ni-MOFs. In order to verify the superiority of this newly synthesized illuminant, an ECL immunoassay model has been designed, and the results demonstrated that it had extremely strong and steady signal output in practical application. This study realized an efficient platform in ECL immunoassay application with the limit of detection of 0.32 pg mL–1 for neuron-specific enolase (NSE). Therefore, the approach which combined the pristine pure Ni-MOFs and the star-illuminant ruthenium pyridine complex would provide a convenient and meaningful solution for exploring the next-generation ECL emitters.

A novel electrochemiluminescence biosensor was presented for the assay of hepatitis B virus (HBV) DNA. Coralloid poly(aniline-luminol)-MWCNTs was prepared on an electrode surface, which was followed by the attachment of capture DNA via amide bond formation. The HBV-DNA and ferrocene labelled DNA hybridized with capture DNA to form a sandwich structure on electrode surface. Ferrocene can effectively increase the signal output of the ECL sensor due to the catalysis of ferrocene on electrochemiluminescence of luminol. The biosensor rendered a linear dependence over the range from 1.0×10 −16 to 1.0×10 −7 M with a low detection limit of 2.4×10 −17 M.

Exploring novel and sensitive analysis methods for monitoring lincomycin (Lin) residues is of great significance since overuse of it would cause a serious threat to public health. Herein, a Zr-porphyrin metal-organic frameworks (Zr-TCPP) with covalently modified polyamidoamine (PAMAM) dendrimers was synthesized as a novel intramolecular self-enhanced ECL reagent, which exhibited greatly improved ECL response due to the promotion of SO 4 •– generation and the shortening of the electron transfer distance. Graphene oxide modified with gold nanoparticles (Au@GO) was synthesized as the quencher for the ECL sensor construction based on the quenching strategy. The present aptasensor achieved a wide linear range of 1.0 × 10 −14 − 5.0 × 10 −9 g/mL and a low detection limit of 1.7 fg/mL, which was applied for the determination of Lin in different real samples with satisfactory results.

A bipolar electrode electrochemiluminescence (BPE-ECL) with β-cyclodextrin (β-CD) host–guest recognition was constructed for the rapid detection of Salmonella typhimurium ( S. typhimurium). Firstly, the sulfhydryl β-CD (SH-β-CD, with inner diameter size: 6.0–6.5 Å) was assembled on the gold-plated cathode of BPE. Ferrocene (Fc, length: 3.5 Å; width: 4.4 Å) labelled signal probe and parts Fc-aptamer complement each other to form a double-stranded DNA (dsDNA). Due to its large size, the dsDNA with a double Fc (width > 8.8 Å) could not be embedded into the cavities of β-CD. In the presence of S. typhimurium, Fc-aptamer bond specifically to S. typhimurium, and the signal probe was shed from the dsDNA. Then, the signal probe (guest) was captured by the SH-β-CD (host) on the surface of the BPE. Due to the principle of spatial matching between Fc and SH-β-CD, a stable inclusion complex could be formed, which made Fc close to the cathode surface of the BPE. When a certain driving voltage was applied, Fc was oxidized to Fc + . The cathode surface of Fc + would promote electron transfer, causing the [Ru(bpy) 3 ] 2+ at the anode emit light, and effectively convert the electrochemical signal into an ECL signal. 10 1 -10 5 CFU mL −1 S. typhimurium was positively correlated with Fc + and ECL intensity, with a detection limit of 10 1 CFU mL −1 . Therefore, S. typhimurium can be quantitatively detected by the electron transfer rate of the BPE.

Background Epithelial cell adhesion molecules (EpCAM) protein is one of the most frequently and intensely studied tumor-associated antigens because of its overexpression in most cancer cells. Thus, it is regarded as a tumor biomarker associated with a wide variety of carcinomas and reflects the existence and growth of tumors in the human body. It is highly required to detect low concentrations of EpCAM in early stage for ensuring human health. Results In this study, a novel sandwich electrochemiluminescence (ECL) aptasensor was developed for EpCAM detection. Wherein oxalate-based Zn-MOF supported tris(2,2′-bipyridyl)ruthenium (II) (Ru(bpy) 3 2+ @Zn-MOF) was used as luminophore and gold nanoparticle modified o-phenylenediamine (PoPD@Au NPs) as quencher. The 3D Ru(bpy) 3 2+ @Zn-MOF generated self-enhanced ECL without additional co-reactants. Furthermore, it remained stable and had short electron transfer distance, ensuring high and stable ECL signal. The ECL signal could be effectively quenched by PoPD@Au NPs through resonance energy transfer (RET) and electron transfer (ET), due to its absorption spectrum overlapping with the ECL emission spectrum of Ru(bpy) 3 2+ @Zn-MOF. In the presence of EpCAM, the aptamer acted as a linker between the luminophore and quencher, forming a sandwich structure that had reduced ECL intensity. Under optimal conditions, EpCAM could be sensitively detected over the range of 1 pg mL -1 -100 ng mL -1 , with a detection limit of 0.34 pg mL -1 . Significance This work developed a ECL aptasensor with self-enhanced Ru(bpy) 3 2+ @Zn-MOF luminophore for EpCAM detection. The ECL system was simple, but ultrasensitive for EpCAM monitoring, thanks to the ECL-RET and ET between Ru(bpy) 3 2+ @Zn-MOF and PoPD@Au NPs. It also could be applied to detect other biomolecules by replacing the aptamer, offering significant potential in disease diagnosis.

A MIP-ECL sensor was developed for the dual-potential resolved simultaneous detection of RUX and LEN. • The combining dummy template imprinting with probes click-labeling enables simultaneous detection of signal-free analytes. • This strategy provides a new detection platform for therapeutic drug monitoring in combination therapy. Combination therapy is widely used in clinics to improve efficacy and reduce side effects by administering two or more drugs for personalized precise dosing. For the combination therapy of ruxolitinib (RUX) and lenalidomide (LEN) in primary myelofibrosis treatment, therapeutic drug monitoring (TDM) of both drugs in blood is required. This paper presents the development of a molecularly imprinted (MIP) electrochemiluminescence (ECL) sensor for the simultaneous detection of RUX and LEN. To address the challenge that neither RUX nor LEN generates ECL signals on electrodes, a dummy template molecule with terminal allyl groups, structurally similar to RUX, was designed to copolymerize with LEN for the preparation of the MIP film. A novel strategy based on dummy template competitive adsorption and photo-initiated click reactions was developed for simultaneous determination of RUX and LEN. It relies on the competitive adsorption between RUX and its dummy template on MIP, followed by click labeling of two ECL probes onto the dummy template and LEN, with detection of RUX and LEN achieved by recording the cathodic and anodic ECL signals, respectively. The MIP-ECL sensor exhibits a detection range of 5 to 5000 ng/mL for both RUX and LEN, with detection limits of 0.11 ng/mL and 0.49 ng/mL, respectively. The sensor was successfully applied to the analysis of human serum samples. It not only provides a novel approach for the detection of multiple targets without responding signals using MIP, but also offers a more sensitive and convenient technical platform for TDM. An MIP-ECL sensor combining dummy-template imprinting with probe click-labeling was developed for the potential-resolved simultaneous detection of ruxolitinib and lenalidomide. This strategy overcomes the challenge of simultaneously detecting signal-free analytes and amplifies detection signals, improving sensitivity with LODs of 0.11 ng/mL for ruxolitinib and 0.49 ng/mL for lenalidomide.
