Biomimetic thermal regulation in intelligent fruit packaging: A fungal hypothermia-bioinspired hydrogel coating

Lei Wang,Zhongzhen Li,Xiaochen Zhang,Dong Li,Chenjuan Zeng,Tiefeng Li,Liang Zou,Zisheng Luo,Xuxu Yang,Li Li

Ambient temperature constitutes a critical environmental factor in postharvest fruit logistics and retail ecosystems. Here, we presented an design of intelligent fruit packaging incorporating poly(N-isopropyl acrylamide) (pNIPAM)-based hydrogel coating to enable thermoregulation, inspired from the natural thermoregulatory mechanism of fungal hypothermia. The as-prepared coating exhibited tough mechanical properties (tensile strength of 62.62 ± 2.84 kPa, Young’s modulus of 57.46 ± 1.69 kPa) and durable thermal responsiveness. When ambient temperature surpassed the lower critical solution temperature (LCST) of hydrogel coating, the packaging transited from hydrophilic to hydrophobic, and deswelled water to the surface, promoting evaporative cooling. Simultaneously, the coating underwent an optical transition from transparent to opaque state, reducing the solar irradiance transmission, including visible light, infrared light, and ultraviolet light. Notably, the LCST exhibited precise tunability across a food logistics-relevant temperature range (3–32 °C) as required through NaCl concentration modulation. Here, we prepared intelligent packagings for different fruit with various LCST, which effectively achieved the temperature reduction within the packaging, and maintained the fruit quality. This biomimetic application enables autonomous thermal regulation in agri-food logistics, while providing a way for intelligent food packaging systems.

A Tocopherol-Related Methyltransferase SlVTE3(2) Regulates Tomato Fruit Ripening and Tocopherol Accumulation

Zheng Bian,Jiaxin Xiong,Bowen Li,Cailian Wang,Chunyang Wang,Benzhong Zhu

Abstract Tomato fruit ripening is a critical developmental process that determines the fruit quality. However, the regulatory mechanisms underlying the ripening initiation remain incompletely understood. Here, we characterized SlVTE3(2), a tocopherol-biosynthesis-related methyltransferase, and investigated its role in tomato fruit ripening. CRISPR/Cas9-mediated mutation of SlVTE3(2) accelerated ripening initiation, resulting in earlier ethylene production, faster softening, and an earlier color transition, whereas overexpression delayed these processes. Transcriptome analysis showed that SlVTE3(2) affected multiple ripening-related pathways, including ethylene biosynthesis and signaling, carotenoid metabolism, and cell wall modification. Although SlVTE3(2) significantly altered the tocopherol composition in tomato fruits, exogenous tocopherol treatment did not affect ripening initiation. These findings provide new insights into the regulation of tomato fruit ripening by the metabolic enzymes.

Synergistic 3D nanofibers for enhanced microbial control and moisture regulation in fruit preservation

Lin Wang,Wei Liu,Xianrui Chen,Zhenzhen Wu,Nitong Bu,Huimin Xiao,Chen Ma,Jingcheng Hao

Food spoilage and waste present significant global challenges, highlighting the need for innovative and sustainable packaging solutions. This study introduces a biodegradable Janus nanofiber, designed with dual-sided water impermeability, carefully engineered wettability gradients, and multiscale structural features. The design integrates moisture management with microbial control, creating an optimal microenvironment for the preservation of tropical fruits. The nanofiber retains moisture on its hydrophilic side while preventing external moisture ingress through the hydrophobic side, effectively reducing weight loss and extending shelf life. In 15-day shelf-life tests at room temperature and 4 °C, the nanofiber outperformed commercial packaging options, reducing fruit weight loss by >20%, while maintaining texture, firmness, and vitamin C content, and significantly delaying microbial spoilage. These improved results are supported by multiscale dissipative particle dynamics simulations, which demonstrate the material's ability to control moisture transport and suppress microbial growth. This work offers a scalable and high-performance solution for biodegradable antimicrobial packaging, enhancing sustainable food preservation practices. The novel 3D PHB-AT NPs show strong potential for tropical fruit preservation and can be extended to other sustainable food storage applications, addressing major challenges in the global food supply chain.

Transcriptional analysis reveals that 1-methylcyclopropene treatment promotes anthocyanin synthesis in peach fruit

Xiaoqin Wu,Zhiyi Shan,Lufan Wang,Wenwei Shi,Yanyan Li,Hongfang Cai,Liang Zhou,Chen Huan

Anthocyanins are bioactive substances that are not only responsible for the pigmentation of various fruits, but also hold potential benefits to human health. In this study, postharvest peaches were treated with 10 ppm 1-MCP, and changes of anthocyanin biosynthesis were then revealed at transcriptional level to gain insight into the underlying molecular mechanisms. The functions of key transcription factors (TFs) were systematically analyzed. Results demonstrated that 1-MCP effectively up-regulated expression of structural genes and stimulated anthocyanin accumulation. A novel transcription factor PpbHLH135 can respond positively to 1-MCP treatment and has been noted for its remarkable gene expression and protein abundance. The transcription factor bHLH3 was also investigated in this study due to its widespread interest and attention, serving to compare the regulatory function of bHLH135 in anthocyanin synthesis, although it didn’t show a significant response to 1-MCP treatment in this study. PpbHLH3 TF showed a stronger regulatory effect on PpF3H , while PpbHLH135 was more dominant over the regulation of PpUFGT . Future genetic and molecular experimentation will elucidate whether PpbHLH135 TF, in combination with other TFs, can play a stronger regulating role in fruit pigmentation, or in other metabolic pathways.

Humidity-responsive PCL/ICs/PVA pad with directional water transport for fruit preservation

Xuling Song,Zhengfeng Wang,Minxin Zhou,Yuwei Li,Yujie Wei,Yi Luo,Jianfeng Li,Bei Xu,Baijun Shang,Qingshan Duan

A smart humidity-responsive antimicrobial pad with directional water transport (DWT) was developed to address the rapid spoilage of fruits after harvest due to high humidity and water accumulation. This pad was fabricated from polycaprolactone/cinnamaldehyde β-cyclodextrin inclusion complexes/polyvinyl alcohol (PCL/ICs/PVA) using electrospinning and co-precipitation methods. The inclusion complexes formed by β-cyclodextrin and the guest molecule cinnamaldehyde (CIN) undergo crystalline and conformational changes upon moisture absorption, generating gaps or channels enabling humidity-responsive release of CIN. Meanwhile, capillary action, water uptake by the hydrophilic layer, and reverse water penetration resistance from the hydrophobic layer jointly promote DWT. In the PCL/ICs/PVA pad, the hydrophobic PCL coalesces free water molecules in the air to form tiny droplets to promote DWT, ICs release CIN in response to humidity, and the hydrophilic PVA absorbs droplets, together enhancing the antibacterial performance of the pad under high humidity conditions. The 14 PCL layer (electrospun from 14 wt% PCL solution) with honeycomb-like morphology was beneficial for loading ICs and humidity response. At 98 % RH, the pad with 33.3 % ICs sustained CIN release for 168 h and extended strawberry shelf life. This study holds significant implications for fruit preservation in high-humidity environments.

Preparation of fruit stem preservation column and its application in postharvest grape storage

Youwei Yu,Xinyu Yang,Silong Jia,Qiyuan Feng,Tian Li,Hesheng Wang,Linna Wang,Limin Guo,Baoguo Ji,Shaoying Zhang

Postharvest fruits and vegetables are susceptible to spoilage, which may result in food safety hazards and economic losses. In this study, a stem-insertable preservation column was developed and applied for the preservation of post-harvest grapes. The preservation column incorporates orange essential oil Pickering emulsion as the active component, with natural antimicrobial substances being effectively stabilized through physical emulsification technology. Experimental results showed that the inserted preservation columns enable targeted release of active compounds through the vascular system and achieved significant inhibition against pathogenic microorganisms including Staphylococcus aureus at grape stem junctions. The preservation column treatment reduced grape decay incidence by over 60 % compared to untreated controls. Furthermore, this technology effectively mitigates fruit water loss, decreasing the mass reduction rate by 45 %, while improving nutrient retention - notably reducing vitamin C loss from 70.22 % in controls to 52.20 % in the P-CZ treatment group. These effects collectively maintain optimal sensory characteristics and marketability of grape berries. The developed method exhibits multiple advantages including operational simplicity, absence of chemical residues, and consistent preservation efficacy. These findings confirm that the essential oil-based preservation column technology represents a safe and effective innovation for grape postharvest management.

Harnessing the power of lignin from Litsea cubeba fruit residues: Characterization and valorization

Kaili Zhang,Xiao Jiang,Ran Tao,Jingcong Xie,Jian Zhao,Ning Zhang,Yicun Chen,Yangdong Wang,Kui Wang,Jianchun Jiang

Lignin is an important component of plants, and its utilization is essential for biorefinery. Litsea cubeba fruit residues (LSRs) are abundant renewable lignin-rich bioresource with enormous potential for producing value-added monophenols. Herein, the lignin from LFRs was isolated with organic solvent, which was characterized by two-dimensional HSQC-NMR, FT-IR, Py-GCMS and GPC. The results indicate that the lignin in LFRs is mainly composed of guaiacyl (G) subunits (93 %), with small amounts of syringyl (S)/ p -hydroxyphenyl (H) structural subunits. The reductive catalytic fractionation (RCF) of LFRs was investigated using methanol (MeOH) as the solvent and a commercial Pd/C (5 wt% Pd) catalyst. Two major phenolic compounds, 4- n -propylguaiacol and 4- n -propanolguaiacol derived from β-O-4 structure, were generated with high yield (19.4 wt%). This result was different from G-type pine wood powder, where 4- n -propanolguaiacol was the main product with a selectivity of 93.8 %. Besides, the influences about several important reaction parameters like time, temperature, solvent, H 2 pressure, and catalyst stability were also investigated based on monophenol yields and average molecular weight. Overall, the lignin components of Litsea cubeba fruit residues were identified and transformed into high functional phenolic chemicals in this work.

Efficient pretreatment of passion fruit husks using choline chloride based deep eutectic solvents

Xiao-Ping Zheng,Meng Li,Yu Chai,Ya-Peng Du,Yu-Cang Zhang,Yan-Zhen Zheng

Substituting renewable lignocellulose for fossil fuels is crucial for economic development and environmental protection. Deep eutectic solvents (DESs) demonstrate efficacy in facilitating the fractionation of high-purity lignin and cellulose during lignocellulose pretreatment. In this study, agricultural by-product passion fruit husks underwent pretreatment using DESs comprised of choline chloride (ChCl) and organic acids, aiming to efficiently separate cellulose and lignin. Among the various DESs tested, ChCl combined with lactic acid (LA) exhibited optimal performance in the pretreatment process. This combination demonstrated a remarkable synergistic effect in breaking down the lignin-carbohydrate complex bonds and deconstructing passion fruit husks. Characterizations of the cellulose rich solid residues were performed using FTIR, SEM, and XRD techniques, confirming the effectiveness of lignin removal and cellulose retention through the pretreatment. The regenerated lignin exhibited homogeneous distribution and well-preserved structure, as evidenced by 2D-HSQC analysis. Furthermore, it displayed notable antioxidant activity and antibacterial property. The mechanism underlying the pretreatment was elucidated through density functional theory (DFT) calculations, confirming hydrogen-bond interactions between DESs and lignin, which positively contributed to lignin dissolution. Additionally, hydrogen-bond properties between DESs and lignin were also revealed.

Dual-functional Zn@melanin nanoparticles for enhanced antibacterial activity and prolonged fruit preservation

Yangjing Li,Wei Tian,Tao Chen,Yanfei Li,Runan Zhao,Yuxin Li,Xuesong Chi,Di Wu,Yinan Du,Jiangning Hu

Mitigating food spoilage from microbial infections remains a critical challenge in food preservation. Although Zn 2+ ions are used as chemical bactericides, their use alone often requires high doses. A novel nanomaterial, Zn@MNPs, combining photothermal properties with the controlled release of Zn 2+ , was synthesized through a coordination of Zn 2+ with melanin nanoparticles (MNPs) derived from cuttlefish ink. Zn@MNPs were capable of attaching onto the bacterial surfaces, enabling high-efficiency release of Zn 2+ under mildly acidic conditions typically associated with bacterial infections. This leads to sustained antibacterial activity, causing bacterial membrane rupture and leakage of intracellular components. Incorporating Zn@MNPs into polyvinyl alcohol (PVA)-based films improved their ability to block UV light and reduce oxygen and water vapor permeability. These films effectively reduced dehydration, preserved nutritional content, and extended fruit shelf life. This study highlights the potential of Zn@MNPs-based PVA films as biodegradable, bioactive packaging materials for enhancing food preservation and safety.

Cu-Doped MnO2 Catalysts for Effective Fruit Preservation via Ozone Synergistic Catalytic Oxidation

Jianguo Huang,Rashid Khan,Chunhui Zhai,Xianting Ding,Li-Sha Zhang,Jin-Ming Wu,Zhizhen Ye

Developing and implementing technologies that can significantly reduce food loss during storage and transport are of paramount importance. Ozone synergistic catalytic oxidation (OSCO) technology has been developed, which sterilizes bacteria and viruses on the surface of food and degrades ethylene released during fruit storage through the active oxygen produced by the catalytic decomposition of ozone. Herein, we report the hydrothermal synthesis of MnO2with distinct phase compositions and nanostructures through simply varying the reaction temperatures. Optimized copper-doped α-MnO2nanorods exhibited remarkable efficacy in activating ozone at a concentration of 40 ppb, and this activation resulted in the complete eradication of indicator bacteria on food surfaces within a 24 h period. Moreover, these nanorods demonstrated high effectiveness in decomposing more than 80% of the ethylene molecules emitted by apples and bananas during the preservation period. The high concentration of surface oxygen vacancies is believed to contribute to the enhanced catalytic activity of the Cu-doped α-MnO2catalyst in the OSCO procedure by reducing ethylene production and maintaining the fruit quality during the preservation period.

Enhancing fruit preservation with sodium alginate films incorporating propolis extract and graphene oxide

Junyan Guo,Mohammad Rizwan Khan,Naushad Ahmad,Wanli Zhang

In this work, sodium alginate (SA) composite films containing propolis extract (PRO) and graphene oxide (GO) were developed. Subsequently, the effects of PRO and GO on different properties of SA composite films were studied, and the films were characterized by scanning electron microscopy, fourier transform infrared spectroscopy, X-ray diffraction, and thermogravimetric analysis. The PRO release properties and fruit preservation performance of the developed composite films were also investigated. The results showed that the incorporation of PRO resulted in a 51.16% increase in tensile strength. The simultaneous incorporation of PRO and GO reduced water vapor permeability (WVP) by 22.56% compared to the SA film. The temperatures at which the SA/GO/PRO film lost 5% of its weight were 8.0°C higher than those of the SA film. The incorporation of GO into the SA/PRO composite film also modulates the release of PRO. Furthermore, the incorporation of PRO and GO improved the tensile strength of the SA film, as reflected in the microstructure of the films. The reduced WVP of the SA composite film allowed the packaged blueberries to exhibit less weight loss and shrinkage, thereby prolonging their shelf life.
