Abstract Maize ear rot and stalk rot are mainly caused by Fusarium graminearum, which severely reduces maize yield. This pathogen produces DON mycotoxin to contaminate maize and threaten food safety. Fludioxonil exhibits high control efficacy against this fungus, while its regulatory mechanism remains poorly understood. This study elucidates the molecular mechanism of transcription factor FgHOY1 in response to fludioxonil. FgHOY1 regulates hyphal growth, conidiation, pathogenicity and DON biosynthesis, and ΔFgHOY1 strain shows markedly enhanced fludioxonil resistance. Fludioxonil treatment significantly downregulates the expression of FgHOY1. Meanwhile, FgHOY1 directly binds to the OS1 promoter to modulate transcription and regulates Hog1 phosphorylation. This study clarifies the FgHOY1-mediated regulatory pathway of fludioxonil resistance, providing a theoretical basis for exploring fungicide-resistance networks, disease green prevention and rational fungicide application.

For optimal proteolytic function, the proteasome core (CP or 20S) must associate with activators. The cAMP-PKA pathway is reported to affect the activity of the proteasome in humans. However, the relationship between the proteasome and PKA is not well characterized. Our results showed that the major catalytic subunit Cpk1 was degraded without the protection of Pkr. Eleven (out of 67)pkrsuppressors had FgBlm10 C-terminal truncation, one suppressor had an amino acid change mutation in thePRE6 ortholog (FGRRES_07282), and one in thePRE5 ortholog (FGRRES_05222). These mutations rescued the defects in growth and conidial morphology, Cpk1 stability, and PKA activities in thepkrmutant. The interaction of FgBlm10 with FgPre5 and FgPre6 were detected by co-immunoprecipitation, and the essential elements for their interaction were characterized, including the FgBlm10 C-terminus, amino acid D82 of FgPre6 and K62 of FgPre5. Additional FgBlm10-interacting proteins were identified in the wild type andpkrmutant, suggesting that PKA regulates the preference of FgBlm10-mediated proteasome assembly. In addition, PKA indirectly affected the phosphorylation of FgBlm10, and its localization in the nucleus. The truncation of the FgBlm10 C terminus also enhanced nuclear import and bleomycin resistance, suggesting its role in proteasome assembly at DNA damage sites. Collectively, our data demonstrated that regulation between PKA and proteasome degradation is critical for the vegetative growth ofF. graminearum.Keywords:FgBlm10;26S proteasome;PKA;degradation;Fusarium graminearum

The calcium-calcineurin and high-osmolarity glycerol (HOG) pathways play crucial roles in fungal development , pathogenicity , and in responses to various environmental stresses. However, interaction of these pathways in regulating fungicide sensitivity remains largely unknown in phytopathogenic fungi. In this study, we investigated the function of the calcium-calcineurin signalling pathway in Fusarium graminearum , the causal agent of Fusarium head blight. Inhibitors of Ca 2+ and calcineurin enhanced antifungal activity of tebuconazole (an azole fungicide) against F. graminearum . Deletion of the putative downstream transcription factor FgCrz1 resulted in significantly increased sensitivity of F. graminearum to tebuconazole. FgCrz1-GFP was translocated to the nucleus upon tebuconazole treatment in a calcineurin-dependent manner. In addition, deletion of FgCrz1 increased the phosphorylation of FgHog1 in response to tebuconazole. Moreover, the calcium-calcineurin and HOG signalling pathways exhibited synergistic effect in regulating pathogenicity and sensitivity of F. graminearum to tebuconazole and multiple other stresses. RNA-seq data revealed that FgCrz1 regulated expression of a set of non-CYP51 genes that are associated with tebuconazole sensitivity, including multidrug transporters, membrane lipid biosynthesis and metabolism, and cell wall organization. Our findings demonstrate that the calcium-calcineurin and HOG pathways act coordinately to orchestrate tebuconazole sensitivity and pathogenicity in F. graminearum , which may provide novel insights in management of Fusarium disease.

BACKGROUND Kiwifruit rot is an important disease caused by different fungal pathogens, which can lead to huge economic loss in the kiwifruit industry. The aims of this study were to discover an effective botanical compound that significantly inhibits the pathogens causing kiwifruit rot, evaluate its control efficacy against this disease, and reveal the underlying mechanisms. RESULTS A strain of Fusarium tricinctum (GF-1), isolated from diseased kiwifruit, could cause fruit rot in both Actinidia chinensis var. chinensis and Actinidia chinensis var. deliciosa . Different botanical chemicals were used for antifungal activity test against GF-1 and thymol was the most effective one with a 50% effective concentration (EC 50 ) of 30.98 mg L −1 . The minimal inhibitory concentration (MIC) of thymol against GF-1 was 90 mg L −1 . Control efficacy of thymol against kiwifruit rot was evaluated and the results indicated that thymol could effectively decrease the occurrence and spread of kiwifruit rot. The mechanisms underlying the antifungal activity of thymol against F. tricinctum were investigated, and it showed that thymol could significantly damage the ultrastructure, destroy the plasma membrane integrity, and instantaneously increase energy metabolisms of F. tricinctum . Further investigations indicated that thymol could extend shelf life of kiwifruit by increasing their storability. CONCLUSION Thymol can effectively inhibit F. tricinctum that is one of the causal agents of kiwifruit rot. Multiple modes of action are involved in the antifungal activity. The results of this study indicate that thymol can be a promising botanical fungicide to control kiwifruit rot and provide useful references for thymol application in agriculture system. © 2023 Society of Chemical Industry.

Fungi are among the main producers of diverse bioactive secondary metabolites. This study aimed to characterize the chemical constituents and evaluate the antimicrobial and antidiabetic activities of metabolites produced by the fungal Fusarium lichenicola isolated from the cicada Cryptotympana mandarina . Gas chromatography–mass spectrometry (GC-MS) analysis revealed that the fungal ethyl acetate extract contained 23 metabolites, including hydrocarbons, esters, terpenoids, alcohols and ketones, fatty acids, aldehydes, phenolics, and sulfur-containing compounds. In vitro assays indicated the fungal extract exhibited weak antimicrobial activity against Gram-positive and Gram-negative bacteria, as well as pathogenic fungi, with minimum inhibitory concentrations (MICs) ranging from 128 to 512 μg/ml. Antidiabetic potential of the fungal extract was demonstrated by inhibition of α-amylase and α-glucosidase enzymes, with IC 50 values of 1202 ± 35 to 1347 ± 40 μg/ml, whereas representative metabolites demonstrated activities in the range of 285 ± 32 to 892 ± 85 μM. Molecular docking simulation identified the potential antidiabetic metabolites from the fungal extract. Further analyses revealed moderate correlation between docking scores and experimental activities, with regression R 2 values of 0.58-0.62 and Pearson correlation coefficients of 0.76-0.79. These results highlight the predictive value of docking for identifying active metabolites, while emphasizing the necessity of experimental confirmation. Overall, these findings suggest that F. lichenicola derived from a cicada host represents a potential source of antidiabetic agents.

Deoxynivalenol (DON) is a mycotoxin mainly produced by theFusarium graminearumcomplex, which are important phytopathogens that can infect crops and lead to a serious disease calledFusariumhead blight (FHB). As the most common B type trichothecene mycotoxin, DON has toxic effects on animals and humans, which poses a risk to food security. Thus, efforts have been devoted to control DON contamination in different ways. Management of DON production byTrichodermastrains as a biological control-based strategy has drawn great attention recently. In our study, eight selectedTrichodermastrains were evaluated for their antagonistic activities onF. graminearumby dual culture on potato dextrose agar (PDA) medium. As potential antagonists,Trichodermastrains showed prominent inhibitory effects on mycelial growth and mycotoxin production ofF. graminearum. In addition, the modified mycotoxin deoxynivalenol-3-glucoside (D3G), which was once regarded as a detoxification product of DON in plant defense, was detected whenTrichodermawere confronted withF. graminearum. The occurrence of D3G inF. graminearumandTrichodermainteraction was reported for the first time, and these findings provide evidence thatTrichodermastrains possess a self-protection mechanism as plants to detoxify DON into D3G when competing withF. graminearum. Keywords:mycotoxin;toxigenicFusarium;biological control;Trichoderma;modified mycotoxin

Soil-borne diseases severely threaten global crop production and urgently require pesticide formulations with targeted effects to improve control efficiency. In this study, a one-pot synthesis method combined with solvent ratio screening was used to successfully prepare Pyr@ZIF-NH₂ loaded with pyraclostrobin (Pyr) achieving a high encapsulation efficiency of 97.42 % and a pesticide loading rate of 20.59 %. The Pyr@ZIF-NH 2 was modified with pectin to obtain Pyr@ZIF@P (119.4 nm), and its successful synthesis was confirmed through characterization. Pyr@ZIF@P enhanced the UV photostability of Pyr and effectively inhibited the growth of Fusarium graminearum , resulting in mycelial malformation. In vitro release studies showed significantly higher Pyr release under acidic, pectinase-rich conditions (mimicking infection sites) with cumulative releases rates of 83.1 % and 74.11 % within 72 h. Foliar application revealed that Pyr@ZIF@P improved foliar retention and promoted downward translocation compared to the commercial suspension, thereby enhancing the control of soil-borne diseases. Moreover, root deaching of Pyr@ZIF@P resulted in more substantial accumulation of active Pyr in the stem, thereby enhancing the control of Fusarium graminearum. Additionally, the zinc-based framework of Pyr@ZIF@P not only facilitates pesticide delivery but also contributes to enhanced root development in wheat seedlings. Taken together, this study presents a novel approach for controlling soil-borne diseases and supporting sustainable agricultural production.

Fusarium graminearum is a preeminent fungal pathogen exacting a heavy toll on global crop production. Fludioxonil, a highly effective broad-spectrum fungicide, has been extensively deployed to mitigate the devastation wrought by F. graminearum. Nevertheless, the molecular mechanisms of fludioxonil resistance in this pathogen remain largely enigmatic. In this study, we identify a novel point mutation (L837F) in FgOs1 (osmosensor-like histidine kinase), which is located in the HIS_KIN (histidine kinase) domain. Biological assays revealed that the FgOs1L837F mutation reduced the ability of F. graminearum to adapt to low-temperature stress, osmotic stress, and metal ion stress and also decreased its pathogenicity. Additionally, this mutation did not confer cross-resistance to the fungicides phenamacril, tebuconazole, and carbendazim. To facilitate field monitoring, we employed allele-specific PCR (AS-PCR) to detect FgOs1L837F point mutation in F. graminearum. This study offers crucial support for fungal disease control and crop protection.

The soil-born filamentous fungal pathogen Fusarium oxysporum f. sp. cubense (FOC), which causes vascular wilt disease in banana plants, is one of the most economically important Fusarium species. Biocontrol using endophytic microorganisms is among the most effective methods for controlling banana Fusarium wilt. In this study, volatile organic compounds (VOCs) showed strong antifungal activity against FOC. Seventeen compounds were identified from the VOCs produced by endophytic fungi Sarocladium brachiariae HND5, and three (2-methoxy-4-vinylphenol, 3,4-dimethoxystyrol and caryophyllene) showed antifungal activity against FOC with 50% effective concentrations of 36, 60 and 2900 μL/L headspace, respectively. Transmission electron microscopy (TEM) and double fluorescence staining revealed that 2-methoxy-4-vinylphenol and 3,4-dimethoxystyrol damaged the plasma membranes, resulting in cell death. 3,4-dimethoxystyrol also could induce expression of chitin synthases genes and altered the cell walls of FOC hyphae. Dichloro-dihydro-fluorescein diacetate staining indicated the caryophyllene induced accumulation of reactive oxygen species (ROS) in FOC hyphae. FOC secondary metabolism also responded to active VOC challenge by producing less fusaric acid and expressions of genes related to fusaric acid production were interrupted at sublethal concentrations. These findings indicate the potential of S . brachiariae HND5 as a biocontrol agent against FOC and the antifungal VOCs as fumigants.

Film mulching is one of the most important methods to control soil-borne diseases. However, the traditional mulch may cause microplastic pollution and soil ecological damage. Herein, a biodegradable film was developed using oxidized starch and carboxymethyl chitosan and incorporated ZIF-8 carrying fludioxonil to sustainably control soil-borne disease. The microstructure, mechanical properties, optical properties, and water barrier properties of the composite films (Flu@ZIF-8-OS/CMCS) were investigated. The results show that Flu@ZIF-8-OS/CMCS had a smooth and uniform surface and excellent light transmittance. The excellent mechanical properties of the films were verified by tensile strength , elongation at break and Young's modulus . Higher contact angle and lower water vapor permeability indicate water retention capacity of the soil was improved through using Flu@ZIF-8-OS/CMCS. Furthermore, the release properties, biological activity, degradability and safety to soil organisms of Flu@ZIF-8-OS/CMCS was determined. The addition of ZIF-8 significantly improved the film's ability to retard the release of Flu, while the Flu@ZIF-8-OS/CMCS has good soil degradability. In vitro antifungal assays and pot experiments demonstrated excellent inhibitory activity against Fusarium oxysporum f. sp. Lycopersici . Flu@ZIF-8-OS/CMCS caused only 13.33 % mortality of earthworms within 7 d. This research provides a new approach to reducing microplastic pollution and effectively managing soil-borne diseases.

Background: Fusarium species are opportunistic causative agents of superficial and disseminated human infections. Fast and accurate identification and targeted antifungal therapy give help to improve the patients’ prognosis. Objectives: This study aimed to evaluate the effectiveness of matrix-assisted laser desorption ionisation time of flight mass spectrometry (MALDI-TOF MS) for Fusarium identification, and investigate the epidemiology and antifungal susceptibility profiles of clinical Fusarium isolates in Southern China. Methods: There were 95 clinical Fusarium isolates identified by DNA sequencing of translation elongation factor 1-alpha (TEF1α) and MALDI-TOF MS, respectively. Antifungal susceptibility testing of isolates was performed by broth microdilution according to the CLSI approved standard M38-A3 document. Results: Seven species complexes (SC) with 17 Fusarium species were identified. The most prevalent SC was the F. solani SC (70.5%, 67/95), followed by the F. fujikuroi SC (16.8%, 16/95). F. keratoplasticum within the F. solani SC was the most prevalent species (32.6%, 31/95). There were 91.6% (87/95) of isolates identified by MALDI-TOF MS at the SC level. In most of species, amphotericin B and voriconazole showed lower MICs compared to itraconazole and terbinafine. The F. solani SC showed higher MICs to these antifungal agents compared to the other SCs. There were 10.5% (10/95) of strains with high MICs for amphotericin B (≥8 μg/ml), terbinafine (≥32 μg/ml) and itraconazole (≥32 μg/ml) simultaneously, mostly focusing on F. keratoplasticum (9/10).Conclusion: MALDI-TOF MS exhibited good performance on the identification of Fusarium strains at the SC level. The F. solani SC was the most prevalent clinical SC in Southern China. The MICs varied significantly among different species or SCs to different antifungal agents.

Fusarium poses a significant threat to the global crop productivity and food security. This study evaluated the mechanisms of nanoscale boron nitride (nano-BN, 0–500 mg/kg) against Fusarium oxysporum in cucumber. Fusarium infection severely impaired plant growth with biomass declining by 60%. However, 50 mg/kg nano-BN treatment significantly increased shoot biomass by 64.9% compared to disease controls, restoring photosynthetic parameters to near-healthy levels. Nano-BN inhibited Fusarium proliferation by disrupting hyphal and spore structures and reduced mycotoxin production (beauvericin and enniatin) by over 63%. Metabolomic analysis demonstrates that nano-BN mitigated oxidative stress by enhancing glutathione metabolism, with significant increases in glutathione and ascorbic acid content by 166.67% and 478.78%, respectively. Importantly, the protein–protein interaction network shows that nano-BN counteracted Fusarium-induced suppression of ribosomal proteins and endoplasmic reticulum stress-related proteins, promoting protein synthesis and folding. The coexpression network identified sucrose and geshoidin as key metabolites linked to ribosomal and mitochondrial proteins, bridging metabolic resilience with enhanced disease resistance. Multiomics analysis suggests that nano-BN alleviated the Fusarium stress by regulating the phenylpropanoid biosynthesis and restoring the expression levels of key enzymes in carbohydrate metabolism. Overall, nano-BN effectively mitigates Fusarium stress by enhancing plant growth and modulating metabolic processes, offering a promising strategy for plant protection.
