Glyphosate exposure impairs visual function in zebrafish larvae through retinoic acid-dependent regulation of six3b expression

Mian Zhang,Nani Xu,Xueru Zhang,Yu Li,Wei Jiang,Baihuan Feng,Wenjun Sun

Glyphosate (GLY) is a widely used, broad-spectrum organophosphorus herbicide. Epidemiological studies have raised concerns about its potential risks due to environmental residues and occupational exposure, yet the molecular mechanisms underlying its toxicity, particularly in the visual system, remain poorly understood. This study investigates the toxic effects and mechanisms of GLY on the visual system using zebrafish. Wild-type and six homeobox 3b ( six3b )-overexpression zebrafish embryos at 5 h post-fertilization (hpf) were exposed to environmentally relevant concentrations of GLY (0.14, 0.7, 1.4, and 2.8 mg/L) until 144 hpf. Results indicated that exposure to varying concentrations of GLY did not significantly affect basic developmental parameters, such as hatchability, mortality, malformation rates, or larval eye diameter. However, it markedly impaired visual motor response and optomotor response in zebrafish larvae. At 1.4 mg/L, GLY exposure reduced optic nerve size, suppressed six3b gene transcription, decreased Six3 protein expression, and lowered retinoic acid (RA) concentration in larvae. Both six3b overexpression and RA supplementation rescued GLY-induced optomotor response deficits. Moreover, RA supplementation restored six3b expression and normalized optic nerve morphology in GLY-exposed larvae. Taken together, these findings demonstrated that exposure to environmentally relevant concentrations of GLY impairs visual behavior in zebrafish larvae through optic nerve hypoplasia, mediated by RA-dependent downregulation of six3b expression.

Visual detection of glyphosate by Al3+-regulated carbon dots/CdTe quantum dots ratiometric fluorescent sensing platform

Ziyi Qin,Jinju Xu,Yuanxin Cao,Chunhui Liao,Tian Lan,Shuyun Shi

Highly sensitive monitoring of pesticides has garnered attention for food safety. Here, a ratiometric fluorescent probe (AG-CDs-Al 3+ /CdTe QDs) was fabricated for specific and sensitive sensing of glyphosate. Surface modifications revealed the chelation-enhanced fluorescence process between the surface -OH of AG-CDs and Al 3+ . Positively charged green-emission AG-CDs-Al 3+ assembled with negatively charged red-emission CdTe QDs, and photoinduced electron transfer (PET) occurred from CdTe QDs to AG CDs-Al 3+ . Glyphosate specifically coordinated with Al 3+ , causing remarkable variations in F 500 /F 630 (linear range, 0.2–4.0 μM; detection limit, 5.7 nM) and a distinguishable green-yellow-red color change on the paper platform (linear range, 0.5–8.0 μM; detection limit, 26 nM). Furthermore, for real sample analysis, the sensor has the advantages of fast response (5 min), excellent accuracy (spiked recoveries, 95.0–105.0 %), high precision (relative standard deviation, < 3.33 %), and superior specificity, which endowed the designed sensing platform with great potential practicality in the field of food analysis.

In situ biosynthetic-driven fluorescence sensor combined with a smartphone platform for portable detection of glyphosate

Yeru Liu,Mengxue Zhang,Xiaolei Zhao,Yifan Zhou,Hongna Yin,Li Zhou,Zhicheng Liu,Qiyong Cai

Glyphosate, a broad-spectrum herbicide extensively applied in agriculture, may bioaccumulate in humans via the food chain and pose toxicological risks. In this work, an integrated biosensing strategy for glyphosate is developed, integrating a self-developed Android application (APP) with alkaline phosphatase (ALP)-triggered silicon nanodot (Si NDs) fluorescence biosensing to achieve field-deployable quantification. In the sensing system, ascorbic acid (AA), produced by ALP-catalyzed substrate 2-phospho- l -ascorbic acid trisodium salt (AA2P), can reduce the silane precursor 3-aminopropyltrimethoxysilane (APTMS) at room temperature to nucleate and grow green fluorescent Si NDs with an emission wavelength of 505 nm. The fluorescence intensity is dynamically modulated by the inhibitory effect of glyphosate on ALP activity, enabling a quantitative relationship between pesticide concentration and signal output. The sensor displayed a robust linear response toward glyphosate across 0.005–100 μM, with detection limits of 2.4 nM (fluorimeter) and 18 nM (APP). Importantly, the custom-designed Android application integrates image acquisition, RGB signal extraction, and calibration modeling, transforming smartphone into portable analytical devices. This synergy between nanomaterial innovation and digital technology not only delivered high recovery rates (80–109.5%) in spiked real samples but also demonstrated excellent reproducibility (RSD < 2.9%) and selectivity, underscoring its promise as a next-generation strategy for on-site glyphosate monitoring.

Chronic Co-exposure to Environmentally Relevant Concentrations of Cadmium and Glyphosate Exerts Aggravating Nephrotoxicity in Mice

Zhong Zhao-Jin,Chen Kai,Qiu Bo,Chen Pan,Lian Cai-Yu,Wang Lin

Cadmium (Cd) and glyphosate (GLY) are two prevalent environmental contaminants. Their widespread use in agricultural activities has caused frequent co-contamination incidents within ecosystems. The kidney is a primary target organ and the major excretory organ for these pollutants. However, the nephrotoxic effects of their co-exposure on mammals remain unclear. Sixty healthy mice housed in a specific pathogen-free facility were randomly divided into 4 groups ( n  = 15) and exposed to GLY (8.75 mg GLY/kg feed) and/or Cd (1.45 mg Cd/kg feed) via diet for 24 weeks; subsequently, the urine, serum, and kidney samples were collected for respective analyses. Data showed that serum levels of renal injury biomarkers in the combined group were significantly higher than those in the alone group, consistent with the renal pathological findings. Urinary levels of proteins and enzymes in the combined group were also markedly higher than those in the alone group, respectively, consistent with the ultrastructural changes of proximal tubular microvilli. Moreover, mitochondria and nuclei in the renal cortex of combined group exhibited more severe ultrastructural damage than those in the alone group, indicating the characteristic of apoptosis. Data from apoptotic assays further suggested that the combined group exhibited more obvious apoptotic damage than those in the alone group. Collectively, these findings demonstrated that GLY itself exhibits low nephrotoxicity, but combined exposure to GLY and Cd significantly enhances Cd’s nephrotoxic effects, characterized by exacerbated apoptotic damage to proximal tubule cells.

Co-exposure to polystyrene microplastics and glyphosate induces gut microbiota dysbiosis and cognitive impairment in honeybees

Yiqing Wu,Lina Qin,Yimeng Zhang,Yifan Jia,Yaoyang Lü,Nianlong Wang,Hao Zheng,Liang Li,Zijing Zhang

Microplastics, as emerging persistent environmental contaminants, can act as vectors for the absorption of other pollutants in ecosystems, such as pesticide residues. However, the combined toxicological effects of microplastics and pesticides on pollinators remain poorly understood. Here, we evaluated the toxicity of polystyrene microplastics (PS) and glyphosate (GLY), both individually and in combination, in honeybees ( Apis mellifera ). While no significant changes in body weight gain or food consumption were observed after 20 days of exposure, co-exposure to PS and GLY significantly increased bee mortality and induced midgut damage and microbial dysbiosis. In the gut, pathways associated with cell proliferation and differentiation, along with genes related to oxidative stress, detoxification, and immunity, were significantly downregulated. Furthermore, the combination of PS and GLY impaired sucrose responsiveness, a key cognitive behavior in bees. Brain transcriptomics indicated a downregulation of serotonergic synapse-related genes, which were correlated with shifts in core gut bacteria such as Snodgrassella and Lactobacillus . Our findings demonstrate that co-exposure to PS and GLY exacerbates gut homeostasis disruption and impairs cognitive behavior, suggesting a potential role of the gut–brain axis. This study extends our understanding of the combined ecological risks posed by multiple environmental contaminants to bees as insect pollinators and emphasizes the need for comprehensive hazard assessments in insect conservation.

Synergistic dual-recognition enabled by single-atom nanozyme and aptamer for ultrasensitive colorimetric detection of glyphosate

Lijun Wang,Kunrong Zhao,Minghui Luo,Yi Li,Xiao Yang,Xianggui Chen

A dual-recognition strategy was developed to detect glyphosate. • The dual-recognition system incorporated Fe/Cu-N-C SAzymes and aptamers. • The LOD was 0.0581 μg/mL. • The method had high specificity and excellent tolerance to matrix effects. • Recovery rates of the proposed method ranged from 90.00% to 96.67%. While nanozyme-based inhibition strategies are attractive for on-site detection of pesticides, their sensitivity is primarily hampered by insufficient interactions at the nanozyme-pesticide interface. To address this, we developed a dual-recognition system that integrates a Fe/Cu-N-C single-atom nanozyme (SAzyme) with an aptamer for sensitive detection of glyphosate. The Fe/Cu-N-C SAzyme overcomes the low atomic exposure of conventional nanozymes by offering abundant metal atomic sites for glyphosate binding. Meanwhile, the surface-grafted aptamers on the SAzyme facilitate glyphosate enrichment on the SAzyme surface, thereby increasing the inhibition efficiency from 14.4% to 26.1%. Under optimized conditions, this enhancement translated into a detection limit of 0.0581 μg/mL, along with high specificity and excellent tolerance to matrix effects. Validation experiments with three spiked samples yielded recoveries ranging from 90.00% to 96.67%, comparable to the results of liquid chromatography-tandem mass spectrometry, demonstrating the reliability of this approach for glyphosate detection in foods. Smartphone-Integrated Ratiometric Fluorescent Sensing Platform Based on Carbon Dots for One-Step Detection of Glyphosate

Tingting Lei,Yating Xu,Siyi Tan,Jiafu Xiao,Shaojing Zhao,Minhuan Lan

An ability to effectively detect glyphosate residues in food and the environment on-site remains a challenge. Herein, we developed a ratiometric fluorescent sensing platform based on blue fluorescent carbon dots (B-CDs) and red carbon dots (R-CDs) for rapid, visual, and quantitative detection of glyphosate. Although B-CDs, which were functionalized with abundant amino and hydroxyl groups, caused glyphosate-mediated aggregation-caused quenching (ACQ) via hydrogen bonding effect, R-CDs acted as a stable internal reference. The system could detect glyphosate in one step within 20 s and had a low detection limit of 2.87 μM. During the detection process, the fluorescent color changes from blue to red. On-site detection can be achieved by using the RGB analysis integrated in the smartphone. The sensor had high selectivity to glyphosate, despite the presence of metal ions, biomolecules, and various pesticides. It was successfully applied to detect actual water samples with recovery rates of 81.8%–96.6% (RSD ≤ 11.1%). With its rapid response, smartphone compatibility, and robustness in detecting complex matrices, the proposed sensor has promising potential for the monitoring of glyphosate in agricultural and environmental samples.

Thermodynamics of the Protonation of an Analogue of Glyphosate, N-(phosphonomethyl)-l-proline, in Aqueous Solutions

Li Qiang,Liu Bijun,Tian Yin,Yu Qianhong,Mu Wanjun,Wei Hongyuan,Wang Dongqi,Li Xingliang,Luo Shunzhong

N -(phosphonomethyl)- l -proline is an analogue of glyphosate. The protonation for N -(phosphonomethyl)- l -proline was studied by potentiometry, calorimetry, 31 P NMR spectroscopy and quantum chemical calculations to further understand the protonation process of glyphosate. The results confirmed that the order of successive protonation sites of totally deprotonated N -(phosphonomethyl)- l -proline are a phosphonate oxygen, amino nitrogen, and finally the carboxylate oxygen. The results can improve the understanding of the biological activity of these types of molecules in solution.

Gold Nanoclusters and Silica-Coated Carbon Dots-Assisted Ratiometric Fluorescent Nanosensors for Ultrasensitive Detection of Glyphosate

Runran Ma,Lizhu Fu,Nan Long,Haipeng Guo,Yujiao Hou,Ying Li,Peng Li,Jiabo Wang,Lidong Zhou,Weijun Kong

In this work, a novel ratiometric fluorescent nanosensor by coupling mesoporous silica spheres-coated nitrogen-doped carbon dots (N-CDs@SiO2) and bovine serum albumin-stabilized gold nanoclusters (BSA-AuNCs) was developed for sensitive detection of glyphosate. BSA-AuNCs nanocomposites were covalently connected onto the surface of amino-functionalized N-CDs@SiO2 nanospheres, causing the core-satellite probes with two fluorescence emissions at 436 and 651 nm, respectively, under a single excitation of 360 nm. The N-CDs@SiO2@BSA-AuNCs “signal on–off–on” ratiometric fluorescent probes could realize highly selective differentiation and accurate detection of glyphosate based on the fluorescence quenching of Cu2+ on BSA-AuNCs and glyphosate-induced fluorescence recovery due to the strong complexation of glyphosate toward Cu2+. The developed ratiometric fluorescent nanosensor exhibited excellent selectivity to glyphosate, wide detectability in a concentration range of 5–100 ng/mL, high sensitivity with a low detection limit of 3.4 ng/mL, as well as fascinating reliability and practicability in real malt samples with recoveries of 94.81–101.61%. This study provides a universal platform for versatile sensing of more trace analytes in complex matrices and can be extended for wide application in the environment and food safety fields.

Residue and dietary risk assessment of glyphosate, glufosinate-ammonium, and their metabolites in maize and soybean

Kuan Wang,Bin Jiao,Haixiang Gao,Xinglu Pan,Xiaohu Wu,Jun Xu,Fengshou Dong,Yongquan Zheng

Persistent weed growing in herbicide-resistant maize and soybean fields is controlled increasingly by glyphosate (GLY) and glufosinate-ammonium (GLU). There is increasing concern that consumers may be exposed to herbicide residues . A Total of 224 samples involved supervised trials and market sampling were carried out. And the levels of GLY and GLU were determined in maize and soybean samples by UPLC−MS/MS method. The residues of GLY and GLU were at a low level in all samples and ranged from <0.04 to 0.29 mg/kg and <0.04 to 0.91 mg/kg, respectively. Deterministic and probabilistic models were used to assess the dietary risk, and the highest risk quotient (RQ) values of GLY and GLU were 2.24 × 10 −4 and 3.27 × 10 −2 , respectively. The RQs of GLU are significantly higher than that of GLY. Risk assessment results showed that GLY and GLU residues should not pose serious health risks to Chinese consumers.

BSA-carbon dots a promising “off–on” fluorescence probe for detecting glyphosate residues in agricultural products

Wang Qian,Wang Miao,Zheng Lufei,She Yongxin,Wang Jing,Jia Minghong,Abd El-Aty A. M.

The widespread and extensive use of glyphosate in agriculture has raised concerns about its potential impact on the quality and safety of agricultural products. Conventional detection methods require long analysis times, making them impractical for the rapid detection of large quantities of samples. Therefore, developing a fast and simple detection system for glyphosate pesticide residues is urgent. In this study, the development of a facile fluorescence probe synthesized using a simple one-pot hydrothermal method for the determination of glyphosate is an important step toward addressing the need for a fast and simple detection system. The present sensor was created using bovine serum albumin (BSA) as a precursor, and the sensor operates by producing an “off–on” fluorescent signal. The bovine albumin-derived BSA-CDs emitted light yellow fluorescence, but this fluorescence was quenched (or suppressed) by the presence of Cu 2+ ions. However, the fluorescence can be restored by the presence of glyphosate, which interacts with the Cu 2+ ions to form a complex and release the BSA-CDs from suppression. The functional groups in glyphosate can capture Cu 2+ and break the BSA-CDs/Cu 2+ combinatorial system. The BSA-CDs/Cu 2+ fluorescence quenching system had good selectivity for glyphosate. The detection limit of the BSA-CD/Cu 2+ fluorescence sensor was 0.05 µg/mL. This developed method was utilized to successfully detect glyphosate in Chinese wheat. The average recoveries ranged from 98.9 to 100.7%, with a relative standard deviation < 3.0%, showing good prospects for practical applicability.

Highly selective and sensitive colorimetric detection for glyphosate based on β-CD@DNA-CuNCs enzyme mimics

Shengmei Tai,Zhijuan Qian,Hongxin Ren,Alberta Osei Barimah,Chifang Peng,Xinlin Wei

A colorimetric assay based on an enzyme-inhibition strategy is promising for the on-site detection of pesticide residues . However, very few works of pesticide detection were reported based on the inhibition toward nanozymes although nanozymes have demonstrated many advantages in sensing various targets. Herein, a facile colorimetric detection for Glyp was developed based on β [email protected] enzyme mimics. The β [email protected] with high peroxidase-like activity was synthesized using random DNA double strands as template and β -CD as surface ligand. β [email protected] could catalyze the H 2 O 2 -3,3′,5,5′-tetramethylbenzidine (TMB) system. The oxidation product OxTMB with a blue color and presented a large absorption signal at 652 nm. However, Glyp could destroy the synergic effect between redox doublet (Cu 2+ /Cu + ) on the β [email protected] A-CuNCs surface, resulting in the inhibition of the peroxidase-like activity. Colorimetric detection for Glyp could be established by detecting the changes of absorption signal at 652 nm. The linear range was 0.02–2  μ g/mL and the detection limit was 0.85 ng/mL (3δ/s). The method was applied in measuring Glyp spiked in lake water and various food samples. This method had rapidness, high sensitivity, and selectivity advantages, indicating the high application potential in monitoring Glyp residue in food.
