Composition and Characteristics of Yam Juice Fermented by Lactobacillus plantarum and Streptococcus thermophilus

Wenxue Chen,Jianzhong Zhu,Hui Niu,Yizhe Song,Weimin Zhang,Haiming Chen,Weijun Chen

In this study, Lactobacillus plantarum (LP), alone or in combination with Streptococcus thermophilus , was used to ferment yam juice. Changes in the composition (phenols, organic acids, reducing sugars and volatile substances) and functional characteristics (antioxidative activity and ability to regulate the intestinal flora) of yam juice during fermentation were investigated. The results showed that the total phenolic (TP) content increased from 201.27 to 281.27 and 285.77 μg/mL for LP- and L. plantarum and S. thermophilus (LPST)-fermented yam juice, respectively. The antioxidative activity of yam juice improved significantly after fermentation, highly correlating with its TP content. In addition, LP- or LPST-fermented yam juice had positive effects on members of the human intestinal flora, improving the activity of Bifidobacterium and inhibiting the growth of Escherichia coli . Sensory analysis showed that LPST-fermented yam juice had a highest score. The results of this study showed that fermented yam juice can serve as a healthy beverage for consumers with low immunity or an imbalance of the intestinal flora.

Lactobacillus reuteri Biofilms Inhibit Pathogens and Regulate Microbiota in In Vitro Fecal Fermentation

Meng-Xin Hu,Fei He,Ya-Xin Guo,Li-Zhen Mo,Xuan Zhu

Bacteria colonizing the gastrointestinal tract generally grow well in biofilms. In recent years, probiotic biofilms have been considered the most promising fourth-generation probiotics. However, the research into the functions of probiotic biofilms is just starting. In this study, Lactobacillus reuteri DSM 17938 biofilms formed on electrospun cellulose acetate nanofibrous scaffolds were contrasted with planktonic cells. Pathogen inhibition analysis of Escherichia coli, Staphylococcus aureus, and Listeria monocytogenes suggested a significant distinction between the planktonic and biofilm groups. In human fecal fermentation, L. reuteri remodeled the microbiota by decreasing the relative abundances of Proteobacteria, Escherichia–Shigella, and Desulfovibrio and increasing the relative abundances of Phascolarctobacterium, Bacteroides, and Lactobacillus. Moreover, L. reuteri biofilms played more positive roles in microbiota modulation and short-chain fatty acid production than planktonic L. reuteri. These findings provide an understanding of the beneficial effects of probiotic biofilms, laying a foundation for the application of probiotic biofilms as a health promoter.

Lactobacillus plantarum modulate gut microbiota and intestinal immunity in cyclophosphamide-treated mice model

Zhibo Zeng,Zonghao Huang,Wen Yue,Shah Nawaz,Xinzhu Chen,Jing Liu

Gut microbiota (GM) contributes to the production of immune-regulatory molecules and cytokines. However, our understanding regarding intricate relationship between Lactobacillus plantarum and GM on regulation of immune function remained limited. To investigate the effect of Lactobacillus plantarum on an immunosuppressed mouse model, we employed cyclophosphamide treatment and conducted various analysis including H&E (hematoxylin-eosin staining), immunohistochemistry, 16S rRNA gene sequencing, and RT-PCR. Our results demonstrated that the administration of Lactobacillus plantarum had significant immunoenhancing effects in the immune-suppressed mice, as evidenced by the restoration of functional expression of specific immune markers in the spleen and an increase in the number of goblet cells in intestine (P < 0.05). Microbial taxonomic analysis revealed alterations in the gut microbiota composition, characterized by a decrease in the richness of Firmicutes and an increase in the proportion of Verrucomicrobia and Actinobacteria following cyclophosphamide treatment. Furthermore, cyclophosphamide treatment significantly suppressed the mRNA expression of inflammatory cytokines (P < 0.05), which were subsequently restored after administration of Lactobacillus plantarum . These observations provide valuable insights into the complex interplay between probiotics, gut microbiota, and immune system functioning.

Quercetin-enriched Lactobacillus aviarius alleviates hyperuricemia by hydrolase-mediated degradation of purine nucleosides

Depeng Li,Meihong Zhang,A.La Teng Zhu La,Zhengtian Lyu,Xin Li,Yuqing Feng,Dan Liu,Yuming Guo,Yongfei Hu

The development of hyperuricemia (HUA) and gout is associated with dysbiosis of the gut microbiota. Quercetin can reduce serum uric acid levels and thus alleviate HUA by modulating the gut microbiota. However, the detailed mechanisms involved in this process are not fully understood. Here, we showed that quercetin significantly reduced the serum uric acid level in a chicken HUA model by altering the chicken cecal microbiota structure and function and increasing the abundance of Lactobacillus aviarius . An L. aviarius strain, CML180, was isolated from the quercetin-treated chicken gut microbiota. Strain characterization indicated that quercetin promoted the growth of L. aviarius CML180 and increased its adhesion, hydrophobicity, and co-aggregation abilities. Gavage of live L. aviarius CML180 to a mouse model of HUA—established by adenosine and potassium oxonate—reduced the serum uric acid level and alleviated HUA. The ability of L. aviarius CML180 to decrease the level of uric acid was due to its degradation of purine nucleosides, which are the precursors for uric acid production. A nucleoside hydrolase gene, nhy69 , was identified from the genome of L. aviarius CML180, and the resulting protein, Nhy69, exhibited strong purine nucleoside-hydrolyzing activity at mesophilic temperature and neutral pH conditions. These findings provide mechanistic insights into the potential of quercetin to treat HUA or gout diseases via a specific gut microbe.

A rational designed synthetic three-species alliance system for synergetic improvement on power generation from microbial fuel cell

Jun-Ying Han,Han-Lei Zhang,Hui Guo,An-Qi Liu,Said Nawab,Na Liu,Ming Hui,Dan-Dan Zhai,Yang-Chun Yong

Microbial fuel cells with mixed bacteria always show better power output capacity than those with single bacteria due to synergetic effects between species. Here, a rational-designed synthetic three-species alliance system based on Shewanella oneidensis MR-1 (the model exoelectrogen), Pseudomonas aeruginosa, and Lactobacillus plantarum was constructed upon process optimization, the three-species alliance system in MFC delivered a maximum power density of ∼ 207 mW/m 2 by using the glucose as the substrate, which was 2 ∼ 58 times higher than that of MFC inoculated with single species. In addition, the synthetic alliance system could generate electricity from broad substrate spectrum (lactose, saccharose, maltose, arabinose, cellobiose) with enhanced performance. Electrochemical and metabolism analysis indicated inter-species synergetic enhancement on charge transfer, substrate utilization, and biofilm formation was the underlying mechanism for performance improvement in MFC by this synthetic alliance system. In this system, L. plantarum uses glucose to produce lactic acid as the electrogenic substrate and electron donor of S. oneidensis MR-1, and P. aeruginosa uses glucose to produce another electron shuttle phenazine, which can also promote the growth of S. oneidensis MR-1 biofilm and help the alliance system to transfer electrons better. This work demonstrated a new strategy to enhance the MFC performance with a rational designed synthetic bacterial community, which would further renew the toolbox for MFC optimization.

Taking SCFAs produced by Lactobacillus reuteri orally reshapes gut microbiota and elicits antitumor responses

Li Nannan,Niu Lili,Liu Yao,Wang Yang,Su Xiaomin,Xu Ce,Sun Zanya,Guo Huishu,Gong Jingru,Shen Shun

Background Colorectal cancer (CRC) incidence is increasing in recent years due to intestinal flora imbalance, making oral probiotics a hotspot for research. However, numerous studies related to intestinal flora regulation ignore its internal mechanisms without in-depth research. Results Here, we developed a probiotic microgel delivery system (L.r@(SA-CS)2) through the layer-by-layer encapsulation technology of alginate (SA) and chitosan (CS) to improve gut microbiota dysbiosis and enhance anti-tumor therapeutic effect. Short chain fatty acids (SCFAs) produced by L.r have direct anti-tumor effects. Additionally, it reduces harmful bacteria such as Proteobacteria and Fusobacteriota, and through bacteria mutualophy increases beneficial bacteria such as Bacteroidota and Firmicutes which produce butyric acid. By binding to the G protein-coupled receptor 109A (GPR109A) on the surface of colonic epithelial cells, butyric acid can induce apoptosis in abnormal cells. Due to the low expression of GPR109A in colon cancer cells, MK-6892 (MK) can be used to stimulate GPR109A. With increased production of butyrate, activated GPR109A is able to bind more butyrate, which further promotes apoptosis of cancer cells and triggers an antitumor response. Conclusion It appears that the oral administration of L.r@(SA-CS)2 microgels may provide a treatment option for CRC by modifying the gut microbiota.

Directional Mining and Thermostability Modification of Novel Acid-Resistant Maltogenic Amylase from Lactobacillus paralimentarius

Qian Hou,Zongxiao Huang,Huicong Chen,Weining Huang,Ning Li,Xiaoqing Mu

Due to the low pH and high temperature in dough rising and baking processes of starchy foods, high thermostability and acidic resistance have become the most important characteristics for commercial maltogenic amylase as a baking antiaging additive. Therefore, a novel maltogenic amylase from Lactobacillus paralimentarius, exhibiting an activity of 274.79 U·mg–1 under optimal conditions at 50 °C and pH 5.0, was screened through EnzymeMiner-assisted preliminary screening, microbial sources-assisted reselection, and structure analysis-assisted final selection, which was based on the bioinformatics. Data-driven rational design was carried out to construct a mutation library for the improvement of thermostability; the optimal reaction temperature and activity of mutant A375P/S257P were increased to 65 °C and 430.15 U·mg–1, respectively, while the half-life at 60 °C increased by 7.09-fold. The structural analysis indicated that the formation of new hydrogen bonds and the increase in rigidity reduce the free energy of mutant A375P/S257P, thereby further enhancing its thermostability.

Glycine betaine transport conditions of Lactobacillus delbrueckii subsp. bulgaricus in salt induced hyperosmotic stress

Xue Han,Huiying Wu,Peng Yu,Lijuan Zhang,Sainan Zhao,Lanwei Zhang

Glycine betaine (GB) is a naturally occurring osmolyte that helps cells adapt to osmotic stress. In this study, the osmoprotective role of GB in Lactobacillus delbrueckii subsp. bulgaricus 34.5 under salt stress was investigated. A concentration of 0.2  m NaCl provided sufficient osmotic constraint while allowing the cells to survive. Under conditions of salt stress, Lactobacillus bulgaricus 34.5 continued to proliferate in the presence of GB, and the optimum growth was seen at 1.2 m m  GB. The optimum culture conditions for L. bulgaricus 34.5 were 37 °C and pH 6.0 for 24 h. In addition, intracellular GB content increased in response to salt stress, and the extent of GB transport in L. bulgaricus 34.5 under high salt concentrations depended on the culture conditions. In conclusion, GB played a protective role in alleviating the detrimental effect of salt on L. bulgaricus 34.5.

Postbiotics From Lactobacillus Johnsonii Activates Gut Innate Immunity to Mitigate Alcohol-Associated Liver Disease

Ruopeng Yin,Tao Wang,Jingzu Sun,Huanqin Dai,Yuting Zhang,Ningning Liu,Hongwei Liu

Prolonged alcohol consumption disrupts the gut microbiota and the immune system, contributing to the pathogenesis of alcohol-associated liver disease (ALD). Probiotic-postbiotic intervention strategies can effectively relieve ALD by maintaining gut homeostasis. Herein, the efficacy of heat-killed Lactobacillus johnsonii (HKLJ) in mitigating alcoholic liver damage is demonstrated in mouse models of ALD. The gut-liver axis is identified as a pivotal pathway for the protective effects of L. johnsonii against ALD. Specifically, HKLJ is found to upregulate the expression of intestinal lysozymes, thereby enhancing the production of immunoregulatory substances from gut bacteria, which subsequently activated the Nucleotide-binding oligomerization domain 2 (NOD2)-interleukin (IL-23)-IL-22 innate immune axis. The elevated IL-22 upregulated the antimicrobial peptide synthesis to maintain intestinal homeostasis and moreover activated the Signal transducer and activator of Transcription3 (STAT3) pathway in the liver to facilitate the repair of hepatic injuries. The heat-killed L. johnsonii provoked immunity helps correct the gut microbiota dysbiosis, specifically by reversing the reduction of butyrate-producing bacteria (such as Faecalibaculum rodentium ) and the expansion of opportunistic pathogens (such as Helicobacter sp. and Pichia kudriavzevii ) induced by ethanol. The findings provide novel insights into the gut microbiota-liver axis that may be leveraged to enhance the treatment of ALD.

Inhibition of Helicobacter pylori by Lactobacillus rhamnosus L08 in combination with Curcuma longa extracts

Qing Liu,Yan Song,Ruonan Wang,Jinwei Sun,Jian He,Qiming Li,Guofang Zhang,Xinkai Ma,Chun Li,Libo Liu

Helicobacter pylori ( H. pylori ) resistance due to antibiotics is becoming increasingly prevalent, and the search for healthy alternatives to drugs has become a popular research topic. Curcuma longa and Lactobacillus rhamnosus are not only inhibitory to H. pylori but also have good compatibility. It was found that acid-tolerant L. rhamnosus had a significant coaggregation effect on H. pylori , but the inhibitory effect was weak. The polyphenol or flavonoid content of Curcuma extract was significantly correlated with the antimicrobial effect, but the inhibition time would be shortened due to its decomposition or destabilization in the digestive system. Both were able to synergistically inhibit H. pylori adhesion and expression of certain genes ( SabA , UreE , UreI , and UreG ), with no significant synergistic effect in inhibiting urease. The combination of Curcuma extract with L. rhamnosus enhanced the inhibitory effect, solved the problem of drug resistance, and improved the short duration of inhibition of Curcuma extract. This study aimed to combine the antimicrobial effect of Curcuma extract with the function of L. rhamnosus in regulating the gastrointestinal flora and to investigate the synergistic inhibitory effect of the two on H. pylori . The findings provide a theoretical basis for subsequent cellular, animal, or clinical experiments.

In vivo evaluations of Lactobacillus-fermented Eucheuma spinosum polysaccharides on alleviating food allergy activity

Jun Zhang,Qingmei Liu,Fudie Gu,Yan Li,Huiying Chen,Meng Liu,Yu Zhou,Hong Liu,Xiaofeng Wei,Guangming Liu

In order to explore the in vivo anti-food allergy activity of Lactobacillus sakei subsp. sakei-fermented Eucheuma spinosum polysaccharides F1-ESP-3, an ovalbumin (OVA)-induced food allergy mouse model was established by ascites immunization and gavage. The weight, temperature, incidence of diarrhea, levels of allergic mediators and inflammatory factors in the serum of mice were analyzed. We analyzed the differentiation of mouse spleen lymphocytes and the proportion of sensitized mast cells by flow cytometry. The intestinal barrier status of mice was analyzed by intestinal pathological tissue sections and microbiota sequencing. The results showed that F1-ESP-3 could alleviate the food allergy symptoms of mice, such as hypothermia and loose stool; levels of OVA-specific immunoglobulin E, mast cell protease and histamine in the serum of sensitized mice and the proportion of dendritic cells and mast cells in mouse spleen were significantly reduced; in addition, F1-ESP-3 may protect the intestinal barrier and further improve the intestinal microenvironment of food-allergic mice by regulating the abundance of Bacteroidetes and Firmicutes. F1-ESP-3 can further improve the intestinal microenvironment of food-allergic mice by upregulating the levels of Lachnospiraceae, and may affect the signal pathways such as NOD-like receptor, MAPK, I kappa B and antigen processing and presentation.

Improving the survival of Lactobacillus plantarum FZU3013 by phase separated caseinate/alginate gel beads

Ruiying Hao,Zhiyang Chen,Ya Wu,Dongdong Li,Binxi Qi,Chenxin Lin,Lan Zhao,Tingting Xiao,Kunfeng Zhang,Jia Wu

The phase separation behavior of mixed solution of caseinate (Cas) and alginate (Alg) was investigated. Lactobacillus plantarum FZU3013 was encapsulated using 4 % Cas/1 % Alg gel beads with a phase-separated structure. The bacteria were predominantly distributed in the Alg-rich continuous phase. The use of 4 % Cas/1 % Alg beads resulted in higher encapsulation efficiency for L. plantarum FZU3013 compared to 1 % Alg beads. After 5 weeks of storage at 4 °C, the viable count in 4 % Cas/1 % Alg beads was 8.3 log CFU/g, which was 1.1 log CFU/g higher than that of the 1 % Alg beads. When 1 % Alg beads of the smallest size were subjected to in vitro digestion, no viable bacteria could be detected at the end of the digestion, whereas the 4 % Cas/1 % Alg beads of the smallest size had a viable count of 3.9 log CFU/g. When the size of the 4 % Cas/1 % Alg beads was increased to 1000 μm, the viable count was 7.0 log CFU/g after digestion. The results of infrared spectroscopy and zeta potential indicated that hydrogen bonding and electrostatic interactions between caseinate and alginate reinforced the structure of the gel beads and improved the protection for L. plantarum FZU 3013.
