As an emerging molecular imaging modality, Photoacoustic Tomography (PAT) is capable of mapping tissue physiological metabolism and exogenous contrast agent information with high specificity. Due to its ultrasonic detection mechanism, the precise localization of targeted lesions has long been a challenge for PAT imaging. The poor soft-tissue contrast of the PAT image makes this process difficult and inaccurate. To meet this challenge, in this study, we first make use of the rich and clear structural information brought about by another advanced imaging modality, Magnetic Resonance Imaging (MRI), to assist organ segmentation and correct for the light fluence attenuation of PAT. We demonstrate improved feature visibility and enhanced localization of endogenous and exogenous agents in the fluence corrected PAT images. Compared with PAT-based methods, the contrast-to-noise ratio (CNR) of our MRI-assisted method increases by 29.1% in live animal experiments. Furthermore, we show that the co-registered MRI image can also be incorporated into PAT image restoration, and achieves improved anatomical landscape and soft-tissue contrast (CNR increased by 25.36%) while preserving similar spatial resolution. This PAT-MRI combination provides excellent structural, functional and molecular images of the subject, and may enable more comprehensive analysis of various preclinical research applications.

This study aims to design and evaluate a two-dimensional Ti 3 C 2 -MXene contrast agent, codoped with iron and manganese, with the aim of achieving T 1 /T 2 dual-mode magnetic resonance imaging contrast, and to investigate its effect on Chemical Exchange Saturation Transfer (CEST) contrast agents. Single-layer Ti 3 C 2 was synthesized via the HCL/LiF etching technique, with the flake size subsequently tailored and the hydroxylated surface modified through a hydrothermal process. Fe 3 O 4 nanoparticles and MnO 2 nanosheets were integrated onto the two-dimensional Ti 3 C 2 -MXene surface using an in situ doping approach, and the final modification was achieved with carboxymethyl chitosan. Various characterization techniques were employed to assess the physical and chemical properties of the probe, which were then combined with a CEST contrast agent to investigate the mechanisms of multimodal magnetic resonance imaging and in vitro photothermal effects. Upon integrating the multi-modal molecular probe with Ioversol for imaging purposes, at a magnetic field strength of B 1 = 1.25 μT and with a molecular probe concentration of 0.5 mg/mL, the CEST signal from Ioversol is almost entirely suppressed. This allows for the successful identification of a distinct concentration range where both contrast agents can coexist. In vitro photothermal studies on MXene codoped with iron and manganese have shown that the composite molecular probe exhibits excellent photothermal efficiency. The method combining complex nanoprobes with CEST contrast agents successfully achieved simultaneous imaging of both contrast agents. In vitro MRI experiments successfully obtained distinguishable concentration ranges for both, with changes in CEST signals reflecting the concentration variations of the complex molecular probes, which also exhibit good photothermal therapy (PTT) capabilities.

Contrast-enhanced magnetic resonance imaging (CE-MRI) is a promising approach for the diagnosis of kidney diseases. However, safety concerns, including nephrogenic systemic fibrosis, limit the administration of gadolinium (Gd)-based contrast agents (GBCAs) in patients who suffer from renal impairment. Meanwhile, nanomaterials meet biosafety concerns because of their long-term retention in the body. Herein, we propose a small-molecule manganese-based imaging probe Mn-PhDTA as an alternative to GBCAs to assess renal insufficiency for the first time. Mn-PhDTA was synthesized via a simple three-step reaction with a total yield of up to 33.6%, and a gram-scale synthesis can be realized. Mn-PhDTA has an r1 relaxivity of 2.72 mM–1 s–1 at 3.0 T and superior kinetic inertness over Gd-DTPA and Mn-EDTA with a dissociation time of 60 min in the presence of excess Zn2+. In vivo and in vitro experiments demonstrate their good stability and biocompatibility. In the unilateral ureteral obstruction rats, Mn-PhDTA provided significant MR signal enhancement, enabled distinguishing structure changes between the normal and damaged kidneys, and evaluated the renal function at different injured stages. Mn-PhDTA could act as a potential MRI contrast agent candidate for the replacement of GBCAs in the early detection of kidney dysfunction and analysis of kidney disease progression.

Recent studies highlight piezocatalytic therapy (PCT) as a superior curative strategy owing to its stable dynamic control compared to sonoluminescence-activated sonodynamic therapy (SDT). However, most nano-piezoelectric catalysts' catalytic efficiency in biosafety and biocompatibility is unsatisfactory. Herein, this study modifies the typical piezoelectric material tetragonal barium titanate (T-BTO). It introduces novel manganese (Mn) -coated T-BTO nanoparticles (MCTB) that possess magnetic resonance imaging (MRI) ability and enhanced PCT effects. The encapsulation of Mn masks the piezoelectric effect of the nanoparticles, and the acidic tumor microenvironment (TME) triggers the stripping of Mn and restoration of the piezoelectric effect of T-BTO, which produces ROS under ultrasonic irradiation. The presence of Mn ions also enhances the MRI property of the tumor site as a contrast agent. Additionally, ultrasonic irradiation promotes the conversion of low-valence Mn into a high-valence state, further stimulating ROS production. The biosafety, therapeutic effects, T1-weighted MRI, and piezoresponse of MCTB are investigated in vitro. Animal studies also indicate that the nanoparticles display good treatment performance when activated by ultrasound and guided by MRI. Therefore, this study develops an MCTB particle that boosts ROS generation, enables tumor-specific ROS release, and enhances T1-weighted MRI, offering a safe PCT approach for tumors.

The morbidity and mortality rates of hepatocellular carcinoma (HCC) are high and continue to increase. The antitumor effects of single therapies are limited because of tumor heterogeneity and drug resistance, and the lack of real-time monitoring of tumor progression during the treatment process leads to poor therapeutic outcomes. Therefore, novel nanodelivery platforms combining tumor therapy and diagnosis have garnered extensive attention. In this study, we developed a multifunctional nanodelivery vector, LPSD-DOX/siRNA, which was loaded with oleic acid-modified superparamagnetic iron oxide nanoparticles (OA-SPION) and the antitumor drug doxorubicin (DOX), further modified by DOTAP to carry small interfering RNA targeting phosphatidylinositol proteoglycan-3 (Glypican-3, GPC3) (siRNA-GPC3). These components were utilized for the combined treatment of HCC and tumor monitoring with magnetic resonance imaging. LPSD-DOX/siRNA exhibited high drug loading, high gene transfection efficiency, and low toxicity. Pharmacokinetic and in vivo distribution experiments showed that LPSD-DOX/siRNA significantly prolonged the circulation time of DOX and enhanced drug accumulation at the tumor site. Magnetic resonance imaging demonstrated that LPSD-DOX/siRNA can serve as a T2 imaging contrast agent to enhance the imaging contrast between the tumor site and other tissues and facilitate the imaging monitoring of tumor tissues. Antitumor experiments revealed that the effects of DOX were promoted by inhibiting the expression of GPC3 protein in HepG2 cell-transplanted tumors, with increased tumor apoptosis. In conclusion, LPSD-DOX/siRNA serves as a promising strategy for combination therapy and monitoring of HCC, with significant potential in antitumor therapy.

ABSTRACT
The development of nanomaterials, especially the development of multi-modality imaging nanoprobe technology has become a reality for tumor imaging diagnosis. In this study, the gadolinium ion chelating agent (2,2′,2′′-(10-(2-(2, 5-dioxopyrrolidin-1-oxyl)-2
oxoethyl)-1, 4, 7, 10-tetraazacyclododecane-1, 4, 7-tri)triacetic acid) (DOTA-NHS) and polyethylene glycol (PEG)-modified targeting ligand PEG-folic acid (FA) were grafted onto the PEG-modified fifth-generation polyamide-amine dendrimer (P5-NH2), which were undertaken as standards
to wrap gold nanoparticles. Then, remaining amino group was acetylated to acquire Gd–Au DENPS-FA nanoparticles. Biocompatibility and cytotoxicity of nanoparticles were analyzed while characterizing them. The cases with unidentified tumors (mass diameter >4.5 cm) of abdominal or pelvic
origin were selected and used for clinical study of computed tomography (CT)/magnetic resonance imaging (MRI) dual-modality abdominal tumors. In the test, average particle size of Gd-Au DENPs-FA nanoparticles was 4.2 nm, and they had good water solubility and stability in aqueous solution.
When concentration of Au+ nanoparticles increased, the cell morphology remained normal, only a small number of round cells appeared, and the cell activity remained above 80%. GD-Au DENPs-FA nanoparticles had a good targeting specificity for KB cells with high folate receptor expression.
During 6–24 hours of injection, the signal at the tumor site of the patient was enhanced markedly, namely the targeted gold nanoparticles were bound to the tumor tissue and the metabolic rate was slow. Based on this material, CT/MRI imaging could be performed in the patient’s body
and further used for the early diagnosis of abdominal tumors.

Background Medical imaging modalities, such as magnetic resonance imaging (MRI), ultrasound, and fluorescence imaging, have gained widespread acceptance in clinical practice for tumor diagnosis. Each imaging modality has its own unique principles, advantages, and limitations, thus necessitating a multimodal approach for a comprehensive disease understanding of the disease process. To enhance diagnostic precision, physicians frequently integrate data from multiple imaging modalities, driving research advancements in multimodal imaging technology research. Methods In this study, hematoporphyrin-poly (lactic acid) (HP-PLLA) polymer was prepared via ring-opening polymerization and thoroughly characterized using FT-IR, 1H-NMR, XRD, and TGA. HP-PLLA based nanoparticles encapsulating perfluoropentane (PFP) and salicylic acid were prepared via emulsion-solvent evaporation. Zeta potential and mean diameter were assessed using DLS and TEM. Biocompatibility was evaluated via cell migration, hemolysis, and cytotoxicity assays. Ultrasonic imaging was performed with a dedicated apparatus, while CEST MRI was conducted using a 7.0 T animal scanner. Results We designed and prepared a novel dual-mode nanoimaging probe SA/PFP@HP-PLLA NPs. PFP enhanced US imaging, while salicylic acid bolstered CEST imaging. With an average size of 74.43 ± 1.12 nm, a polydispersity index of 0.175 ± 0.015, and a surface zeta potential of −64.1 ± 2.11 mV. These NPs exhibit excellent biocompatibility and stability. Both in vitro and in vivo experiments confirmed the SA/PFP@HP-PLLA NP’s ability to improve tumor characterization and diagnostic precision. Conclusion The SA/PFP@HP-PLLA NPs demonstrate promising dual-modality imaging capabilities, indicating their potential for preclinical and clinical use as a contrast agent.

Developing ultrasensitive contrast agents for the early detection of malignant tumors in liver is highly demanded. Constructing hepatic tumors specific targeting probes could provide more sensitive imaging information but still faces great challenges. Here we report a novel approach for the synthesis of ultra-small Fe 3 O 4 nanoparticles conjugated with c(RGDyK) and their applications as active-target T 1 -weighted magnetic resonance imaging (MRI) contrast agent (T 1 -Fe 3 O 4 ) for imaging tiny hepatic tumors in vivo . RGD-modified T 1 -Fe 3 O 4 nanoprobes exhibited high r 1 of 7.74 mM -1 s -1 and ultralow r 2 /r 1 of 2.8 at 3 T, reflecting their excellent T 1 contrast effect at clinically relevant magnetic field. High targeting specificity together with favorable biocompatibility and strong ability to resist against non-specific uptake were evaluated through in vitro studies. Owing to the outstanding properties of tumor angiogenesis targeting with little phagocytosis in liver parenchyma, hepatic tumor as small as 2.2 mm was successfully detected via the T 1 contrast enhancement of RGD-modified T 1 -Fe 3 O 4 . It is emphasized that this is the first report on active-target T 1 imaging of hepatic tumors, which could not only significantly improve diagnostic sensitivity, but also provide post therapeutic assessments for patients with liver cancer.

High-performance magnetic resonance (MR) imaging contrast agents are the key to reliable and accurate diagnosis of diseases using the MR imaging technique. In particular, dual-modal MR imaging nanoprobes that integrated the advantages of a high contrast effect and impressive sensitivity of T1- and T2-weighted MR imaging, respectively, have attracted increasing attention in recent years. However, the current methods for the fabrication of the dual-modal MR imaging nanoprobes suffer from a tedious synthesis procedure, harsh synthesis condition and ambiguous toxicity, strongly hindering their clinic transformation. Herein, we proposed a biomineralization approach for the fabrication of bovine serum albumin (BSA)–Fe3O4 nanoparticles with undoubted biosafety and robust T1 and T2 MR imaging ability in a facile way under mild conditions. BSA not only served as a template to control the growth of nanocrystal, but also enabled the favorable water solubility of the nanoprobe. The outstanding biocompatibility of BSA–Fe3O4 nanoparticles was proven by systematic in vitro and in vivo toxicity assessments, and dual-modal T1 and T2 MR imaging in vivo was achieved successfully. In addition, the nanoprobe was successfully applied for in vivo MR imaging of liver with a broad time window, and the metabolic investigation of BSA–Fe3O4 nanoparticles revealed that they could be eliminated from the body, ensuring a negligible adverse effect. The proposed biomineralization approach opens a new way for the fabrication of dual-modal MR contrast imaging nanoprobe with undoubted biosafety, and other functional proteins also exhibit great potential for the fabrication of dual-modal MR contrast imaging nanoprobe with specific functions with respect to biomineralization.

An ultra-sensitive T2-weighted MR imaging contrast agent was prepared based on Fe3O4 nanoparticles and Gd3+ ions (Fe3O4@Gd). Amino modified Fe3O4 nanoparticles were conjugated to diethylenetriamine pentaacetic acid, and finally coordinated with Gd3+ ions. The nanoparticles had a uniform morphology with a size of 100 nm and a Gd/Fe mass ratio of 1/110. The r2 (transverse relaxivity) of the Fe3O4 nanoparticles increased from 131.89 mM−1 s−1 to 202.06 mM−1 s−1 after coordination with Gd3+ ions. MR measurements showed that the aqueous dispersion of Fe3O4@Gd nanoparticles had an obvious concentration-dependent negative contrast enhancement. Hepatoma cells were selected to test the cytotoxicity and MR imaging effect. The application of Fe3O4@Gd nanoparticles as contrast agents was also exploited in vivo for T2-weighted MR imaging of rat livers. All the results showed the effectiveness of the nanoparticles in MR diagnosis.

Multifunctional polymer nanoparticles with great promise for biomedical applications have attracted many attentions. Using an aggregation-enhanced emission (AEE) molecule DPPBPA and magnetic Fe3O4 as the core, and biocompatible polymer Pluronic F-127 as the encapsulation matrix, multifunctional polymer NPs Fe3O4/DPPBPA@F-127 were fabricated by self-assembly procedures. These NPs have bright near-infrared fluorescence λex at 654 nm with a high fluorescence quantum yield of 18.3%, and strong magnetism, and superparamagnetism. With good monodispersity and biocompatibility, the NPs not only can show effective MRI ability, but also can stain in cytoplasm with a strong near-infrared fluorescent signal, as well as little toxicity to living cells, which show a very good prospect in the field of biological applications.

Effective vascular and hepatic enhancement and better safety are the key drivers for exploring gadolinium-free hepatobiliary contrast agents. Herein, a facile strategy proposes that the high lipophilicity may be favorable to enhancing sequentially vascular and hepatobiliary signal intensity based on the structure–activity relationship that both hepatic uptake and interaction with serum albumins partly depend on lipophilicity. Therefore, 11 newly synthesized derivatives of manganese o-phenylenediamine-N,N,N′,N′-tetraacetic acid (MnLs) were evaluated as vascular and hepatobiliary agents. The maximum signal intensities of the heart, liver, and kidneys were strongly correlated with log P, a key indicator of lipophilicity. The most lipophilic agent, MnL6, showed favorable relaxivity when binding with serum albumin, good vascular enhancement, rapid excretion, and reliable hepatobiliary phases comparable to a classic hepatobiliary agent, gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid (Gd-EOB-DTPA) for in vivo liver tumor imaging. Inhibition experiments confirmed the hepatic targeting of MnL6 is mediated by organic anion-transporting polypeptides.
