Ultrafast Mapping of Subcellular Domains via Nanopipette-Based Electroosmotically Modulated Delivery into a Single Living Cell

Ruo-Can Qian,Jian Lv,Yi-Tao Long

Recently, a variety of strategies have been developed for single-cell detection. However, the precise probing of the given area at single-cell level is still a challenge. Here, we put forward a rapid and targeted imaging approach for the mapping of subcelluar domains, which realizes the precise injection of multifluorescence into a single living cell via an ultrasmall quartz capillary nanopipette (∼100 nm) and can successfully transport different fluorescent probe molecules to the pointing subcellullar area around the tip in the cytoplasm within 20 s. This method is also applied for monitoring the loss of intracellular mitochondrial membrane potential under the treatment of metformin in a single MCF-7 breast cancer cell. The major driven force in the nanopipette, electroosmotic flow, is evaluated by a theory calculation method and finite element simulations, and the solution indicates a confined solute distribution profile around the tip within the working range. Overall, the nanopipette approach realizes the precise and simultaneous delivery of multiple probe molecules into the single living cell through the electroosmotically modulated, nondestructive, and one-step injection, which is especially powerful and convenient for multichannel single-cell imaging and monitoring, indicating favorable potential for understanding, monitoring, and controlling the biological processes from the single cell to subcellular organelles.

Integrating mass spectrometry imaging and data-driven segmentation for spatial metabolic mapping of diabetic eye disease

Shuohan Cheng,Shuo Wang,Tianfang Lan,Hongtao Jin,Zhou zhi,Zhonghua Wang,Zeper Abliz

Diabetic eye disease (DED) is a leading cause of vision impairment worldwide, yet the molecular mechanisms underlying its progression remain incompletely understood. In this study, we applied a dual-platform spatial metabolomics strategy integrating air flow-assisted desorption electrospray ionization mass spectrometry imaging (AFADESI-MSI) and matrix-assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI) to characterize spatial metabolic alterations in the eyes of diabetic rats. Data-driven segmentation of retinal micro-regions using SCiLS Lab software enabled fine-scale mapping of metabolic heterogeneity. Physiological, biochemical, and histopathological analyses were combined with spatial metabolite mapping to construct a metabolic atlas and evaluate the regulatory effects of ferulic acid. We established a comprehensive spatial metabolome atlas of the rat eye, identifying 135 annotated metabolites and revealing significant region-specific metabolic heterogeneity. Unsupervised k-means clustering was further applied to the high-resolution MALDI-MSI data, successfully delineating distinct functional micro-regions of the retina solely based on endogenous metabolic profiles, demonstrating the power of data-driven tissue segmentation. In diabetic eyes, 39 metabolites were significantly dysregulated, involving amino acid, glucose, lipid, and redox metabolism. Notably, lysine, arginine, carnitine, and GSH were depleted, while glucose-6-phosphate (G6P), glycerol-3-phosphate (G3P), and pro-inflammatory lipids were elevated, highlighting profound metabolic reprogramming across ocular compartments. Ferulic acid treatment restored nine key metabolites, alleviated oxidative stress, normalized lipid and glucose metabolism, and improved retinal structural integrity in a dose-dependent manner. This study shows that integrating mass spectrometry imaging with data-driven tissue segmentation reveals spatial metabolic reprogramming in DED and highlights ferulic acid as a promising therapeutic candidate.

Mapping CSC-Mediated Ovarian Cancer Chemoresistance via CXCR4-PET to Guide Precision Cisplatin Re-Sensitization Therapy

Lixia Feng,Simei Zhao,Zheng Wei,Wenwen Wang,Feiquan Ying,Lin Huang,Mengna Zhu,Mengqing Chen,Qiang Yang,Si Sun,Dawei Jiang,Lingling Gao,Jing Cai

Therapy targeting cancer stem cells (CSCs) has been proposed as a promising strategy to reduce chemoresistance and relapse risks in ovarian cancer (OC) patients. However, the lack of targetable markers impedes research progress. Here, we demonstrate that CXC motif chemokine receptor 4 (CXCR4) may be a targetable functional marker of ovarian CSCs and propose a new translational model incorporating targeted imaging and CXCR4 blockade in CXCR4 + tumors. Expression profile analysis of chemoresistant CSC-like ovarian cancer cells highlighted that CXCR4 functions as a potential stemness marker. CXCR4 + ovarian cancer cells exhibited high self-renewal capacity in vitro and in vivo, and an association with chemoresistance. CXCR4 inhibitor AMD3100 significantly impaired the self-renewal ability of CSC-like ovarian cancer cells and enhanced their sensitivity to cisplatin. CXCR4-targeted [ 68 Ga]Ga-Pentixafor was highly specific in delineating CXCR4-high cell line-derived xenografts and patient-derived xenografts (PDXs) via positron emission tomography (PET) imaging, with precise tumor-targeting and persistent retention. A combination of AMD3100 and CDDP exerted an excellent antitumor effect in CXCR4-high PDXs, but not in CXCR4-low PDXs. These results suggest that CXCR4 may represent a functional CSC marker associated with chemoresistance. Moreover, [ 68 Ga]Ga-Pentixafor PET imaging can guide decision-making for AMD3100 therapy, paving the way for further clinical translation.

3D stress mapping reveals the origin of lithium-deposition heterogeneity in solid-state lithium-metal batteries

Jinhua Hu,Zhetao Sun,Yirong Gao,Ping Li,Yifan Wu,Shiwei Chen,Ruibin Wang,Nana Li,Wenge Yang,Yongxing Shen,Shou-Hang Bo

Solid-state lithium-metal batteries possess intrinsic advantages in terms of both safety and energy density. However, the fundamental origin of electrochemical lithium deposition heterogeneity in solid-state batteries is much less understood than that in lithium-metal batteries using a liquid electrolyte, partly due to the difficulties of directly mapping lithium-deposition reaction fronts and the associated changes in local stress of the solid-state electrolyte. Here, we trace the evolution of three-dimensional microscopic stress and demonstrate that the stress distribution is rather broad in a garnet solid-state electrolyte during processing and battery cycling using confocal Raman spectroscopy. We further discuss the effect of local stress variations on the overpotential of lithium deposition as the most likely origin of lithium-deposition heterogeneity in garnet systems. The ex situ stress-mapping tool developed in this work provides a strong basis for understanding the electromechanical effects, a prerequisite to fully unlock the potential of solid-state lithium-metal batteries.

Atomic-Scale Mapping of Atomically Dispersed Light-Element Species in Oxide Nanomaterials by Solid-State NMR

Xiaoli Xia,Fang Wang,Changju Yang,Yujie Wen,Xiang Ning,Zhengting Zhang,Juan Wen,Zhonghan Feng,Xiaokang Ke,Zhiwu Yu,Hanxi Guan,Wenhua Hou,Xuefeng Guo,Weiping Ding,Di Wu,Yadong Li,Luming Peng

Light-element isolated species represent an emerging class of atomically dispersed nanomaterials, yet their precise local and higher-shell coordination environments remain largely inaccessible due to the intrinsic limitations of conventional nanoscale probes. Here, we establish a solid-state NMR method, combining 23Na and 17O spectroscopy with first-principles calculations, to directly resolve the atomic-scale structure of light-element isolated species beyond the first coordination shell. Using Na-modified CeO2 as a representative system, isolated and aggregated sodium species are distinguished by characteristic 23Na NMR signatures, while 17O NMR further reveals how light element dopants perturb oxygen coordination across the first to third atomic shells. This approach enables clear differentiation of Na–O–Ce linkages from more remote Na–O–Ce–O environments that are inaccessible to electron microscopy and X-ray-based techniques. As a demonstration of atomic-scale structure-adsorption correlation, CO2 is shown to preferentially adsorb at Na–O–Ce sites rather than at Na–O–H groups.

Self-Assembled Microsphere-Integrated SERS Microneedles for the 3D Mapping of Agrochemicals Penetration in Fresh Produce

Qiong Wang,Feifei Yang,Xinru Chen,Yan Lu,Siqi Zhou,Yaxian Wang,Yuxin Yang,Tingting Wang,Hui Liu,Hui Song,Mengke Su,Honglin Liu

Spatially resolved quantification of agrochemical deposition, penetration, and absorption on and within fresh produce is critical for optimizing efficacy and managing residual risks. This study presents a hydrogel microneedle (MN)-based sensing platform that integrates surface-enhanced Raman scattering (SERS) with silver nanowire (AgNW) arrays. The platform overcomes the inherent limitation of conventional self-assembly methods, which rely on unstable biphasic interfaces, by elucidating a curing mechanism for liquid–liquid interface self-assembled nanostructures. This process enables a controllable transformation of dynamic interfacial assemblies into stable, solid plasmonic microspheres (PMSs). These PMSs function as high-performance SERS units, exhibiting uniform dispersion, long-term stability, and a reproducible electromagnetic hotspot distribution, thereby addressing the signal irreproducibility associated with random nanoparticle encapsulation in MNs. Subsequent integration of the PMSs into a polyethylene glycol diacrylate (PEGDA) hydrogel yields mechanically robust MN arrays with tunable swelling properties. This design confers an integrated pierce–extract–detect functionality, enabling rapid agrochemical screening. For the common pesticide molecules thiram and thiabendazole (TBZ), in the theoretical system, the limits of detection (LODs) are 3.47 × 10–10 M and 8.81 × 10–8 M, respectively. When coupled with SERS mapping technology, the platform facilitates two-dimensional and three-dimensional visualization of pesticide distribution on and within fresh produce, revealing spatial patterns governed by multifactor synergy. This work underscores the significant potential of the developed SERS platform in agrochemical residue detection and distribution analysis, opening new avenues for applications in food safety monitoring and sustainable agrochemical development.

Temperature-driven antisolvent crystallization of ellagic acid with choline chloride: structure–function mapping and antioxidant performance

Anshu Sharma,Bong-Seop Lee,Byung Heung Park

Ellagic acid (EA), a bioactive polyphenol, exhibits low aqueous solubility, limiting its pharmaceutical and nutraceutical applications. In this study, EA was recrystallized from ethanol with choline chloride (ChCl) at three controlled temperatures (313.15 K, 333.15 K, and 353.15 K) to enhance its physicochemical properties. The resulting recrystallized ellagic acid (REA) samples were systematically characterized using FTIR, XRD, SEM, DSC, and NMR spectroscopy. XRD analysis revealed improved crystallinity with distinct peak sharpening at 2θ = 28.4° and 29.3° for the 333.15 K REA, while SEM imaging confirmed more defined crystal morphology. Importantly, 1 H NMR spectroscopy revealed chemical shift variations: the aromatic proton signal at 7.47 ppm in native EA exhibited downfield shifts and broadening in the 313.15 K and 353.15 K REA, indicating altered hydrogen bonding and π-π stacking. The 333.15 K REA retained spectral fidelity to native EA, suggesting minimal perturbation in molecular symmetry. These temperature-induced structural modifications significantly enhanced the antioxidant activity, with the 333.15 K REA demonstrating a 1.6-fold increase in DPPH radical scavenging efficiency in comparison with other REA at other temperatures. This study establishes a simple recrystallization approach to modulate EA’s solid-state properties and improve its functional performance. The primary objective is to perform a single-factor evaluation to quantify EA recovery after interaction with ChCl under defined conditions.

Interface-engineered calcium phosphate-hydroxyapatite heterostructure with enhanced full-spectrum mechanoluminescence for bridge stress distribution mapping

Renjie Xu,Shuangqiang Fang,Mingsheng Cai,Haoliang Cheng,Jiaqi Ou,Jiali Yu,Jing Ma,Tiancheng Zhang,Juan Kang,Le Wang

Mechanoluminescent (ML) materials, with their self-powered and real-time response characteristics, have demonstrated significant potential in structural health monitoring. However, limitations such as narrow spectral width and insufficient quantitative analysis capability still restrict their practical applications. This work presents an Eu-doped Ca 3 (PO 4 ) 2 /Ca 5 (PO 4 ) 3 OH (CPO/CPOH) heterostructured ML material. By synergistically optimizing interfacial electron behavior and phase composition, this heterostructure design increases ML intensity by 230 % compared to single-phase materials. Notably, an interface-induced Eu 3 + →Eu 2+ self-reduction phenomenon is first observed . Introducing a reducing atmosphere , ML intensity is further amplified by 10.7 times, and full-spectrum ML emission with a full width at half maximum (FWHM) of 170 nm is successfully realized. Density functional theory (DFT) calculations confirm that the oxygen-bridge-connected interface structure exhibits thermodynamic stability, and directional electron transfer from CPO to CPOH reduces the system band gap (2.73 eV), providing an efficient pathway for carrier migration. Combined with thermoluminescence analysis, cycling stability, and interfacial electric field correlation, the ML mechanism is clarified as an interfacial electric field-driven carrier detrapping process. Furthermore, by integrating digital image correlation (DIC) technology with Lagrange transformation, this material successfully visualizes and enables quasi-quantitative analysis of stress concentration regions (0.05 - 1.16 MPa) in bridge structures, validating compressive performance differences among T-beam bridges, cylindrical bridges, and arch bridges. This study not only provides a novel heterostructure ML system and regulating spectral properties but also establishes a reliable visual quasi - quantitative analysis platform for real-time health monitoring of large-scale structures.

SMILE: Extraction-free submicron-resolution mapping of lipid chain length and unsaturation by stimulated Raman imaging

Yihui Zhou,Xiangjie Huang,Xingxin Liu,Ling Zhang,Jie Huang,Jing Zhao,Yongqing Zhang,Jin Zhang,Yanqing Gong,Delong Zhang,Hyeon Jeong Lee

Lipid spatial heterogeneity is essential for linking metabolic regulation to cellular behavior. Yet, subcellular lipid profiling remains a challenge, as exogenous labels cannot detect lipid speciation and species diversity. Here, we developed submicron-resolution mapping of intracellular lipid elements (SMILE), a vibrational spectrometry platform based on hyperspectral stimulated Raman scattering imaging with a spectral analysis pipeline for pixel-resolved lipid profiling. SMILE enables quantitative mapping of lipid chain length and unsaturation at 300 nm resolution without the need for lipid extraction or labeling. Using SMILE, we revealed the spatial heterogeneity of lipid species and identified a distinct subset of highly aggregated lipid droplets (LDs) enriched in long-chain polyunsaturated lipids in invasive cancer cells. Lipid elongation was found to promote LD aggregation and cancer migration/invasion, thereby defining this as a unique spatial lipid-metabolic signature associated with cancer aggressiveness. Mechanistically, we identified linolenic acid as a key mediator driving aggressive lipid signatures and cancer invasion. Collectively, these findings demonstrate SMILE as an enabling spatial-omics platform that expands the chemical resolution of vibrational spectroscopic imaging for fundamental and translational research.

Bimodal Thermoelectric/AIE E-Skin Decouples Contact-Area Ambiguity for Concurrent Pain Perception and Injury Mapping

Bingchen Huo,Fengxia Kuang,Chunyu Du,Chengzhi Chen,Qi Sun,Wanqiang Li,Lirong Liang,Hanfu Wang,Cun-Yue Guo,Guangming Chen

Thermoreceptive electronic skins face fundamental limitations due to signal ambiguity caused by the contact-area effect in thermoelectric devices. Since electrical output depends on total heat flux, localized high-temperature stimuli generate indistinguishable signals from widespread low-temperature stimuli, leading to unreliable thermal hazard assessment. To address this challenge, we developed a dual-modality bilayer e-skin integrating a single-walled carbon nanotube-based thermoelectric layer and an aggregation-induced emission luminogen-based photoluminescent layer. The bottom thermoelectric layer functions as a fast-response nociceptor, converting temperature gradients into voltage-encoded “pain” signals. The top AIE layer provides contact-area-independent optical mapping of thermal fields through photoluminescence quenching, enabling direct visual decoupling of temperature from contact area without computational processing. This integrated platform achieves real-time injury visualization, accurate temperature recognition (>97% accuracy), and reliable nociceptive-like sensing. Validated by a biomimetic robotic reflex system, the e-skin offers a robust solution for intelligent safety protection and enhanced human-machine interaction in dynamic thermal environments.

Automated Modeling of Road Networks for High-Definition Maps in OpenDRIVE Format Using Mobile Mapping Measurements

Kai-Wei Chiang,Hao-Yu Pai,Jhih-Cing Zeng,Meng-Lun Tsai,Naser El-Sheimy

With growing attention being devoted to autonomous vehicle (AV) safety, people have recently attached importance to high-definition (HD) maps. HD maps are not limited by environmental factors and can limit AVs driving in certain lanes. HD maps provide accurate auxiliary information on factors such as road geometry, traffic sign placement, and traffic topology. Nowadays, most HD maps are made from point clouds data, and this data contains accurate 3D position information. However, the production costs associated with HD maps are substantial. This article proposes an algorithm that reduces a great amount of time and human resource. The algorithm is divided into three phases, lane lines’ extraction from point clouds, modelling lane lines with attributes, and building OpenDRIVE file. The algorithm extracts lane lines resting on intensity value within the range of roads. Next, it models lane lines by cubic spline interpolation with the result of first phase, and build the OpenDRIVE file following the announcement of OpenDRIVE. The final result is compared with the verified HD map from the mapping company to analyze the accuracy. The root mean square (RMSE) obtained were 0.069 and 0.079 m for 2D and 3D maps, respectively.
