Simple Method for Directly Synthesizing Ag Nanoparticles with Silver Ammonia and Polydopamine in a Microreactor toward the Conversion of 4-NP to 4-AP

Kun Wang,Rong Chen,Xun Zhu,Qiang Liao,Dingding Ye,Gang Chen,Ming Liu

In this study, a simple method for directly synthesizing Ag nanoparticles on the inner wall of a microreactor was proposed to catalytically convert 4-NP to 4-AP. The proposed method employed silver ammonia as the precursor and polydopamine (PDA) to modify the surface of the microreactor. Because PDA contains numerous reducing agents of phenolic hydroxyl groups, the silver ions can be effectively reduced to Ag nanoparticles. Moreover, due to the zwitterionic nature of PDA, the use of silver ammonia as a precursor, which intrinsically has a high pH value, can allow for more silver ions to be adsorbed on the surface than the conventionally used AgNO3 as a precursor. Accordingly, the precursor utilization could be greatly improved, leading to the increase of the catalyst loading and thereby boosting the catalytic conversion and durability. It was also found that an increase in the reduction time required for the catalyst preparation could lead to the increase of the catalyst loading and thereby the catalytic performance but a too long reduction time did not continue to make more contribution. Experimental results demonstrate that the microreactor prepared by this simple method is able to exhibit good catalytic activity and durability, providing a new approach for developing high-performance microreactors.

Ultrasensitive Determination of Thiabendazole in Fruit Juice by Surface-Enhanced Raman Scattering (SERS) Using a Covalent Organic Framework (COF) Gold Nanoparticle (Au NP) Substrate

Xinxing Jiang,Jihong Fu,Dandan Li,Wenxia Xue,Yaxue Zhang

An effective and sensitive SERS substrate (TPDH-COF/Au NPs) on a covalent organic framework (TPDH-COF) decorated with gold nanoparticles (Au NPs) was fabricated via electrostatic self-assembly and successfully applied for the determination of thiabendazole (TBZ) in fruit juice. The “raspberry-like” structure SERS substrate combined the properties of the abundant “hot spots” between the Au NPs and the adsorption performance of TPDH-COFs for trace analytes. Excellent SERS performance was observed, which achieved a high enhancement factor value of 2.01 × 108 for rhodamine 6 G, with considerable reproducibility and excellent long-term stability of at least 28 d. The limit of detection was estimated to be 5 ppb for TBZ in fruit juice. The result demonstrated that the TPDH-COF/Au NPs have great potential for SERS analyses and determination of pesticide residues in foods.

The Three-Pronged Strategy: A Bilayer Hydrogel Treats Diabetic Chronic Wound through Microalgae Oxygen Therapy, Ag─FA NP Antibacterial, and Synergistic Scavenging of ROS

Ruru Xiong,Min Zhou,Huishuang Li,Lijuan Wang,Guixia Ling,Peng Zhang

Due to hypoxia, excessive reactive oxygen species (ROS), dense bacterial biofilms, and complex inflammatory microenvironment, chronic diabetic wound healing presents a huge challenge. Currently, sustainable treatments for this multifaceted problem have not been found. Here, microalgae and silver (Ag)─fulvic acid (FA) NPs are used to develop a multinetwork hydrogel (C@Ag─KGCM) based on oxidized konjac glucomannan (OKG) for the treatment of diabetic chronic wound (DCW). The lower hydrogel carries microrobots of Chlamydomonas rheinhardtii (C. rheinhardtii) , which continuously release bio-oxygen under light irradiation to alleviate hypoxia in deep wound tissues. The upper layer carries Ag─FA NPs formed by Ag + and FA self-assembly chelation complex, which can effectively inhibit bacterial growth and remove bacterial biofilms through photothermal-therapy-based Near - Infrared (NIR) 808 nm. C. reinhardtii and Ag─FA NPs synergistically scavenge excessive ROS. Therefore, a new concept of “three-pronged strategies” that addresses multiple problems in the wound microenvironment of diabetic patients has been proposed. Histological analysis of wound tissues shows that the C@Ag─KGCM bilayer hydrogel can promote chronic diabetic wound healing and tissue regeneration, indicating promising potential for its treatment of DCW healing.

Efficient photocatalytic degradation of perfluorooctanoic acid by bismuth nanoparticle modified titanium dioxide

Zhi Wang,Mingyang Li,Wei Cao,Zhenhua Liu,Deyang Kong,Wei Jiang

Perfluorooctanoic acid (PFOA) is potentially toxic and exceptionally stable attributed to its robust C F bond, which is hard to be removed by UV/TiO 2 systems. In this research, bismuth nanoparticle (Bi NP) modified titanium oxides (Bi/TiO 2 ) were synthesized by a simple photochemical deposition-calcination method and were applied as photocatalysts for the first time to degrade PFOA. The removal rate of 50 mg/L PFOA reached 99.3 % with 58.6 % defluorination rate after 30 min of irradiation via a mercury lamp. Bi/TiO 2 exhibited superior performance in PFOA degradation compared to commercial photocatalysts (TiO 2 , Ga 2 O 3 , Bi 2 O 3 and In 2 O 3 ). In addition, Bi/TiO 2 showed high degradation activity under actual sunlight, achieved 100 % removal rate and 59.3 % defluorination rate within 2 h. Bi NPs increase the light trapping ability of Bi/TiO 2 and promote the separation of photogenerated electron-hole pairs via local surface plasmon resonance (LSPR) effect, which results in more photogenerated holes (h + ) and hydroxyl radicals ( OH). Combined with DFT calculations and intermediate detections, the degradation reaction is initiated from the oxidation of the PFOA carboxyl group via h + , followed by the loss of the CF 2 unit step by step with the participation of OH. This work presents a novel approach for the practical implementation of TiO 2 -based photocatalysts to achieve highly efficient photocatalytic degradation of perfluorocarboxylic acids (PFCAs).

Lactiplantibacillus plantarum P101 Ameliorates TiO2 NP-Induced Bone Injury in Young Rats by Remodeling the Gut Microbiota and Inhibiting the Production of Pro-Inflammatory Cytokines

Lixin Xie,Lihua Feng,Xiaomin Tang,Yunping Xu,Hengyi Xu,Yang Liu

Purpose To evaluate the therapeutic effect of oral administration of Lactiplantibacillus plantarum P101 (P101) on skeletal injury in young rats exposed to titanium dioxide nanoparticles (TiO2 NPs), and explore the potential mechanism. Methods Four-week-old male rats were orally administration to TiO2 NPs and supplemented with P101 2 hours later for 4 weeks. The growth and development, food intake, bone metabolism and serum inflammatory markers of the rats were evaluated. Their tibias were observed and evaluated using microcomputed tomography (micro-CT), tartrate-resistant acid phosphatase (TRAP) staining, immunohistochemistry (IHC) and real-time quantitative PCR (RT-qPCR). We observed the tibia growth plate using safranin and fast green staining. 16S rDNA sequence analysis of fecal samples was performed to observe changes in the gut microbiota. Results Our results showed that TiO2 NPs can lead to bone growth inhibition and osteoporosis, induce intestinal flora imbalance, and induce inflammation in young rats. Further mechanistic studies suggested that TiO2 NPs disrupts intestinal flora and increases serum IL-1β levels, which increased the expression of RANKL in bone, thereby enhancing osteoclast differentiation and function, leading to bone loss. Through a P101 supplementation experiment, we found that P101 ameliorated the inflammation and osteoporosis on bone caused by TiO2 NPs.Conclusion This study showed that the mechanism by which P101 alleviates bone damage caused by TiO2 NPs may be through restoring intestinal microbial homeostasis and inhibiting inflammatory response.

High-capacity NCNT-encapsulated metal NP catalysts on carbonised loofah with dual-reaction centres over C–M bond bridges for Fenton-like degradation of antibiotics

Xiao Zhang,Zhongping Yao,Jiankang Wang,Wanqian Guo,Xiaohong Wu,Zhaohua Jiang

The intensification of electron transport between the dual-reaction centres in Fenton-like catalyst is demonstrated through the incorporation of carbon-metal (C–M) bond bridges. Herein, nitrogen-doped carbon nanotube-encapsulated Fe 3 C (Fe 3 C@NCNT) with dual-reaction centres over short C–Fe bond was constructed through DFT calculation, and accordingly a series of NCNTs-encapsulated Fe/Fe 3 C, Co, and Mo 2 C nanoparticles on carbonised loofah were developed by dynamic thermal impregnation and subsequent pyrolysis process . The resultant catalysts displayed satisfactory degradation properties of antibiotics, in which Fe/Fe 3 C@NCNTs-CL-800 achieved norfloxacin (NOR) removal efficiency of 93.8% in 90 min under neutral pH, and presented excellent cycle stability in NOR removal as well as universality in various antibiotic degradation systems. The remarkable degradation performance of heterogeneous Fenton-like catalysts was attributed to enhancing electron transport to electron-rich M centres over short C–M bond bridges. This study will open a new perspective on the rational design of catalysts for Fenton-like degradation.

Dual-aptamer-based colorimetric assay for the accurate identification of circulating tumor cells via Fe3O4@Pt NP nanozymes and G-quadruplex/hemin for signal amplification

Ye He,Panlin Wang,Zhuzheng Wu,Yating Chen,Xiaohao Gan,Fangjie Li,Wenxiang Wang

The precise identification and detection of circulating tumor cells (CTCs) is significant for the early diagnosis and prognostic treatment of cancer. However, it remains a challenge to detect CTCs with high sensitivity and specificity. In this study, a colorimetric sensor for the analysis of CTCs was constructed based on dual-aptamer recognition and core (Fe3O4)–shell (Pt) nanoparticle (Fe3O4@Pt NP) nanozymes coupled with a G-quadruplex as a signal transducer/amplifier. The results show that the linear range of the designed colorimetric sensor is 50–4000 cells and the detection limit is 4 cells. The constructed aptasensor could be extended to identify various CTCs by changing the aptamers, and holds great potential in precise tumor diagnosis.

Gold Nanoparticle (Au NP)-Decorated Ionic Liquid (IL) Based Liposome: A Stable, Biocompatible, and Conductive Biomimetic Platform for the Fabrication of an Enzymatic Electrochemical Glucose Biosensor

Hongzhuang Chen,Jialin Fan,Xue Chen,Zhenkuan Ma,Ling Zhang,Xuwei Chen

A novel liposomal nanocomposite with biocompatibility and conductivity, Au@ILs-polysome, was fabricated and first utilized as the electrode material to immobilize the redox enzyme glucose oxidase (GOD) for the sensitive determination of glucose. Ionic liquids (ILs)-based polysome (ILs-polysome) was prepared via the radical polymerization of ILs based liposome (ILs-liposome) and improved by ion exchange with chloroauric acid to prepare Au nanoparticles (Au NPs) modified ILs based polysome (Au@ILs-polysome). The morphology, surface properties, and structure of Au@ILs-polysome were characterized; the results suggest that the nanocomposite exhibited excellent stability on the surface of solid electrodes without fusion. Electrochemical impedance spectroscopy (EIS) demonstrated that the nanocomposite had enhanced conductivity. Cyclic voltammetry (CV) shows that the direct electron transfer of GOD was achieved on the GOD/Au@ILs-polysome/GCE electrode since the Au@ILs-polysome provided a biocompatible microenvironment that made the immobilized enzyme stay natural and active. Differential pulse voltammetry (DPV) demonstrates that the GOD/Au@ILs-polysome/GCE electrode exhibited improved electrocatalytic activity compared with the GOD/ILs-polysome/GCE electrode. The electrochemical sensor exhibited a low detection limit of 0.02 mM, high sensitivity of 32.52 µA mM−1 cm−2, high selectivity toward glucose, and good stability. The practical application of the sensor was verified by the determination of glucose in human serum. The multi-component liposomal nanocomposites possess universal significance in protein-based direct electrochemical devices while offering a facile route to fabricating a highly sensitive glucose sensor for practical clinical diagnosis.

A novel spherical GelMA-HAMA hydrogel encapsulating APET×2 polypeptide and CFIm25-targeting sgRNA for immune microenvironment modulation and nucleus pulposus regeneration in intervertebral discs

Yu Xiao-Jun,Zhao Yuan-Ting,Abudouaini Haimiti,Zou Peng,Li Tian-Qi,Bai Xiao-Fan,Wang Shan-Xi,Guan Jian-Bin,Li Meng-wei,Wang Xiao-dong,Wang Ying-guang,Hao Ding-Jun

Methods Single-cell transcriptomics and high-throughput transcriptomics were used to screen factors significantly correlated with intervertebral disc degeneration (IDD). Expression changes of CFIm25 were determined via RT-qPCR and Western blot. NP cells were isolated from mouse intervertebral discs and induced to degrade with TNF-α and IL-1β. CFIm25 was knocked out using CRISPR-Cas9, and CFIm25 knockout and overexpressing nucleus pulposus (NP) cell lines were generated through lentiviral transfection. Proteoglycan expression, protein expression, inflammatory factor expression, cell viability, proliferation, migration, gene expression, and protein expression were analyzed using various assays (alcian blue staining, immunofluorescence, ELISA, CCK-8, EDU labeling, transwell migration, scratch assay, RT-qPCR, Western blot). The GelMA-HAMA hydrogel loaded with APET×2 polypeptide and sgRNA was designed, and its effects on NP regeneration were assessed through in vitro and mouse model experiments. The progression of IDD in mice was evaluated using X-ray, H&E staining, and Safranin O-Fast Green staining. Immunohistochemistry was performed to determine protein expression in NP tissue. Proteomic analysis combined with in vitro and in vivo experiments was conducted to elucidate the mechanisms of hydrogel action. Results CFIm25 was upregulated in IDD NP tissue and significantly correlated with disease progression. Inhibition of CFIm25 improved NP cell degeneration, enhanced cell proliferation, and migration. The hydrogel effectively knocked down CFIm25 expression, improved NP cell degeneration, promoted cell proliferation and migration, and mitigated IDD progression in a mouse model. The hydrogel inhibited inflammatory factor expression (IL-6, iNOS, IL-1β, TNF-α) by targeting the p38/NF-κB signaling pathway, increased collagen COLII and proteoglycan Aggrecan expression, and suppressed NP degeneration-related factors (COX-2, MMP-3). Conclusion The study highlighted the crucial role of CFIm25 in IDD and introduced a promising therapeutic strategy using a porous spherical GelMA-HAMA hydrogel loaded with APET×2 polypeptide and sgRNA. This innovative approach offers new possibilities for treating degenerated intervertebral discs. A generally minimalist strategy of constructing biomineralized high-efficiency personalized nanovaccine combined with immune checkpoint blockade for cancer immunotherapy

Sijia Zhang,Yuanji Feng,Meng Meng,Zhen Li,Huixin Li,Lin Lin,Caina Xu,Jie Chen,Kai Hao,Zhaohui Tang,Huayu Tian,Xuesi Chen

As a representative of tumor immunotherapy, tumor vaccine can inhibit tumor growth by activating tumor-specific immune response, which has the advantages of relatively low toxicity and high efficiency, and has attracted much attention in recent years. However, there are still difficulties in how to effectively deliver tumor vaccines in vivo and make them work efficiently. It is a relatively mature method to load tumor specific antigens with suitable carriers to produce tumor vaccines. Here, a generally minimalist construction method of tumor nanovaccine was developed. A high-efficiency tumor nanovaccine (NV) was prepared in one step by a biomineralization-like method, which contained ovalbumin (OVA, model antigen), unmethylated cytosine-phosphate-guanine (CpG, adjuvant) and Mn-NP (carrier and adjuvant). NV not only showed good tumor preventive effect, but also could successfully inhibited tumor development and metastasis when combined with anti-PD-L1, and induced long-term immune memory effect. However, the method of screening tumor specific antigen to construct nanovaccine is cumbersome and tumors are heterogeneous. Therefore, surgically resected tumor tissue is the best source of antigens for preparing tumor vaccines. Next, based on the strong loading ability of the carrier, we designed a personalized tumor nanovaccine (PNV) using the supernatant of tumor abrasive fluid (STAF) as antigen based on the generally minimalist tumor nanovaccine construction strategy. PNV combined with anti-PD-L1 could successfully inhibit post-surgical tumor recurrence and induce strong and durable immune memory effects. This study presents a novel, general, and minimalist strategy to construct high-efficiency personalized nanovaccine, which has a wide range of potential applications in the field of tumor treatment.

MicroSphere 3D Structures Delay Tissue Senescence through Mechanotransduction

Ziang Li,Jincheng Tang,Liang Zhou,Jiannan Mao,Wei Wang,Ziyan Huang,Lichen Zhang,Jie Wu,Xinzhao Jiang,Zhouye Ding,Kun Xi,Feng Cai,Yong Gu,Liang Chen

The extracellular matrix (ECM) stores signaling molecules and facilitates mechanical and biochemical signaling in cells. However, the influence of biomimetic “rejuvenation” ECM structures on aging- and degeneration-related cellular activities and tissue repair is not well understood. We combined physical extrusion and precise “on–off” alternating cross-linking methods to create anisotropic biomaterial microgels (MicroRod and MicroSphere) and explored how they regulate the cell activities of the nucleus pulposus (NP) and their potential antidegenerative effects on intervertebral discs. NP cells exhibited aligned growth along the surface of the MicroRod, enhanced proliferation, and reduced apoptosis. This suggests an adaptive cellular response involving adhesion and mechanosensing, which causes cytoskeletal extension via environmental cues. NP cells maintain nuclear membrane integrity through the YAP/TAZ pathway, which activates the cGAS-STING pathway to rectify the aging mechanisms. In vivo, MicroRod carries NP cells and reduces inflammatory factor and protease secretion in degenerated intervertebral discs, inhibiting degeneration and promoting NP tissue regeneration. Our findings highlight the role of mechanical stress in maintaining cellular activity and antiaging effects in harsh environments, providing a foundation for further research and development of antidegenerative biomaterials.

CARMA3 Drives NF-κB Activation and Promotes Intervertebral Disc Degeneration: Involvement of CARMA3-BCL10-MALT1 Signalosome

Liu Yadong,Zhang Guiqi,Wu Jiani,Meng Yi,Hu Jianyu,Fu Hao,Yang Dongfang

Intervertebral disc degeneration (IDD) diseases are common and frequent diseases in orthopedics. The caspase recruitment domain (CARD) and membrane-associated guanylate kinase-like protein 3 (CARMA3) is crucial in the activation of the NF-κB pathway. However, the biological function of CARMA3 in IDD remains unknown. Here, CARMA3 expression was elevated in nucleus pulposus (NP) tissues of IDD rats and nutrient deprivation (ND)-induced NP cells. The main pathological manifestations observed in IDD rats were shrinkage of the NP, reduction of NP cells, fibrosis of NP tissues, and massive reduction of proteoglycans. These changes were accompanied by a decrease in the expression of collagen II and aggrecan, an increase in the expression of the extracellular matrix (ECM) catabolic proteases MMP-3, MMP-13, and metalloprotease with ADAMTS-5, and an increase in the activity of the pro-apoptotic protease caspase-3. The expression of p-IκBα Ser32/36 and p-p65 Ser536 was also upregulated. However, these effects were reversed with the knockdown of CARMA3. Mechanistically, CARMA3 bound to BCL10 and MALT1 to form a signalosome. Knockdown of CARMA3 reduced the CARMA3-BCL10-MALT1 signalosome-mediated NF-κB activation. CARMA3 activated the NF-κB signaling pathway in a manner that bound to BCL10 and MALT1 to form a signalosome, which affects NP cell damage and is involved in the development of IDD. This supports CARMA3-BCL10-MALT1-NF-κB as a promising targeting axis for the treatment of IDD.
