Hyphenated method of dispersive solid phase microextraction and surface-enhanced Raman spectroscopy for rapid analysis of PAHs

Meihui Gu,Jianing Wang,Shihao Ma,Kang Cheng,Xi Bian,Hao Gao,Shaokai Jian,Yongchao Lai

Thioctic acid-modified Ag NPs have been utilized as a pH-responsive platform. • A hyphenated method of DSPME and SERS is established for the analysis of PAHs. • Extraction, enrichment, and detection are integrated into one step. • This method demonstrates high sensitivity and good reproducibility. Polycyclic aromatic hydrocarbons (PAHs) pose a significant threat to human health owing to their carcinogenic, teratogenic, and mutagenic properties, underscoring the urgent need for the expeditious development of efficient detection methodologies. The prevailing analytical methodologies for PAHs are characterized by two primary drawbacks: the necessity of extensive sample pretreatment and the use of high-cost instrumentation. The present study proposes a hyphenated method of dispersive solid phase microextraction (DSPME) and surface-enhanced Raman spectroscopy (SERS) for rapid analysis of PAHs. Thioctic acid-modified silver nanoparticles (Ag NPs) can precisely regulate the dispersion and aggregation states by varying the pH value, thereby facilitating the DSPME enrichment and SERS analysis of PAHs. This method demonstrates high sensitivity (limits of detection: 8.2 ppb for fluoranthene, 0.75 ppb for benzo[ a ]pyrene), good reproducibility (batch-to-batch variation <5%), and excellent selectivity. Furthermore, the method reveals satisfactory recovery rates and robust anti-interference ability in real environmental water samples, offering considerable promise for expeditious, dependable, on-site PAH detection with minimal pretreatment and portable equipment. Persulfate advanced oxidation based on a novel CeO2-Fe3O4 heterojunction catalyst for enhanced degradation of soil PAHs

Zhuoqun Deng,Nianxi Wang,Yang Wang,Xujing Li,Haixiang Li,Dexun Zou,Yanping Liu

Due to the persistence and toxicity of PAHs, the accumulation of PAHs in soil causes environmental challenges. Utilizing nanoparticles to activate persulfate (PS) is an eco-friendly and cost-effective method for degrading PAHs. Herein, an advanced oxidation system with Fe 3 O 4 and CeO 2 loaded on BC as PS activators (Fe 3 O 4 -Ce@BC/PS) has been constructed, achieving a total removal of 89.34 % of PAHs from soil at 48 h of reaction at room temperature. Notably, High Resolution Transmission Electron Microscope analysis confirmed the presence of a CeO 2 -Fe 3 O 4 heterojunction, and Density functional theory calculations further revealed the key function of this structure. It reduced the adsorption energy of PS (−4.03 eV), thereby improving its utilization efficiency. In addition, during the reaction process, OH and SO 4 − as the main active species collaborated with 1 O 2 , O 2 − and electron transfer to degrade PAHs. In a word, the Fe 3 O 4 -Ce@BC/PS system developed in this study provided an effective solution to the problem of PAHs-contaminated soil.

Mechanistic insights into microbial and metabolomic responses in PAHs-contaminated soil derived from a coal gasification site

Ben Zhang,Jianli Jia,Jiaqi Zhang,Yichi Ma,Bing Xiao,Haokun Yuan,Yuxin Han,Yequan Wang,Ravi Naidu

Microbial remediation offers a promising in situ solution for polycyclic aromatic hydrocarbons (PAHs)-contaminated soils at coal gasification sites, where these persistent pollutants cause long-term ecological risks. However, the mechanistic links between microbial metabolic processes, community dynamics, and degradation efficiency under realistic remediation conditions remain insufficiently understood. In this study, a large-scale soil column experiment evaluated an integrated bioremediation approach combining bacterial consortium inoculation, nutrient supplementation, and intermittent bio-ventilation (BA treatment). The BA treatment significantly accelerated the degradation of PAHs in soil, particularly high-concentration PAHs such as naphthalene (NAP), phenanthrene (PHE), pyrene (PYR) and benzo[a]pyrene (BaP), and resulted in pronounced changes in differential metabolites (DEMs), with notable enrichment of key transformation intermediates such as pyrene-1,6-dione and phenanthrene-1,2-oxide. KEGG pathway analysis indicated significant enrichment in xenobiotic biodegradation, amino acid metabolism, and lipid metabolism pathways, reflecting microbial functional expression associated with pollutant breakdown and cellular metabolic activity. Simultaneously, BA treatment enhanced dehydrogenase and fluorescein diacetate hydrolase activities while also increasing available phosphorus, which were strongly correlated with PAHs removal and metabolite profiles. Mantel tests and co-occurrence network analyses revealed strong linkages among microbial genera, PAHs, DEMs and soil biochemical properties, highlighting synergistic interactions underlying degradation. Specific bacterial genera exhibited close associations with PAH-related metabolites, suggesting functional contributions to pollutant transformation. Overall, the enhanced microbial treatment not only promoted PAHs removal but also altered soil metabolic profiles and biochemical activity, providing evidence of synergistic microbe–metabolite interactions that contribute to efficient bioremediation.

Damage of anodic biofilms by high salinity deteriorates PAHs degradation in single-chamber microbial electrolysis cell reactor

Peng Ding,Ping Wu,Zhang Jie,Min-Hua Cui,He Liu

The anaerobic biodegradation of polycyclic aromatic hydrocarbons (PAHs) in high salinity wastewater is rather hard due to the inhibition of microorganisms by complex and high dosage of salts. Microbial electrolysis cell (MEC), with its excellent characteristic of anodic biofilms, can be an effective way to enhance the PAHs biodegradation. This work evaluated the impact of NaCl concentrations (0 g/L, 10 g/L, 30 g/L, and 60 g/L) on naphthalene biodegradation and analyzed the damage protection mechanism of anodic biofilms in batching MECs. Compared with the open circuit, the degradation efficiency of naphthalene under the closed circuit with 10 g/L NaCl concentration reached the maximum of 95.17% within 5 days. Even when NaCl concentration reached 60 g/L, the degradation efficiency only decreased by 10.02%, compared with the MEC without additional NaCl. Confocal scanning laser microscope (CSLM) proved the superiority of the biofilm states of MEC anode under high salinity in terms of thicker biofilms and higher proportion of live/dead bacteria cells. The highest dehydrogenase activity (DHA) was found in the MEC with 10 g/L NaCl concentration. Moreover, microbial diversity analysis demonstrated the classical electroactive microorganisms Geobacter and Pseudomonas were found on the anodic biofilms of MECs, which have both PAHs degradability and the electrochemical activity. Therefore, this study proved that high salinity had adverse effects on the anodic biofilms, but MEC alleviated the damage caused by high salinity.

Contamination of polycyclic aromatic hydrocarbons (PAHs) in surface sediments and plants of mangrove swamps in Shenzhen, China

Fenglan Li,Xiaokang Zeng,Junda Yang,Kai Zhou,Qijie Zan,Anping Lei,Nora F.Y. Tam

The concentrations of 16 individual and total polycyclic aromatic hydrocarbons (∑PAHs) in sediments, roots and leaves of three mangrove swamps in Shenzhen, China, namely Futian, Baguang and Waterlands, were determined. The mean concentration of ∑PAHs in Futian (4480   ng   g −1 ) was significantly higher than that in Baguang (1262   ng   g −1 ) and Waterlands (2711   ng   g −1 ). Among the 16 PAHs, the concentration of naphthalene was the highest. Based on the ratios of phenanthrene/anthracene and fluoranthene/pyrene, PAHs in Futian and Waterlands came from petrogenic and pyrolytic sources, while Baguang was mainly from pyrolytic. More PAHs were accumulated in leaves, as reflected by its higher mean concentration of ∑PAHs (3697   ng   g −1 ) and bioconcentration factor of PAHs (BCF) (>1.5) than that in roots. The BCF values in plants collected from Futian were significantly higher than that from Waterlands. These results indicated that more attention should be paid to the PAH contamination in Futian.

Mechanisms of biostimulant-enhanced biodegradation of PAHs and BTEX mixed contaminants in soil by native microbial consortium

Mukhtiar Ali,Xin Song,Qing Wang,Zhuanxia Zhang,Jilu Che,Xing Chen,Zhiwen Tang,Xin Liu

Despite the co-occurrence of polycyclic aromatic hydrocarbons (PAHs) and benzene, toluene, ethylbenzene, and xylene (BTEX) in the field, to date, knowledge on the bioremediation of benzene and benzo[a]pyrene (BaP) mixed contaminants is limited. In this study, the mechanisms underlying the biodegradation of benzene and BaP under individual and co-contaminated conditions followed by the enhanced biodegradation using methanol, ethanol, and vegetable oil as biostimulants were investigated. The results demonstrated that the benzene biodegradation was highly reduced under the co-contaminated condition compared to the individual benzene contamination, whereas the BaP biodegradation was slightly enhanced with the co-contamination of benzene. Moreover, biostimulation significantly improved the biodegradation of both contaminants under co-contaminated conditions. A trend of significant reduction in the bioavailable BaP contents was observed in all biostimulant-enhanced groups, implying that the bioavailable BaP was the preferred biodegradable BaP fraction. Furthermore, the enzymatic activity analysis revealed a significant increase in lipase and dehydrogenase (DHA) activities, as well as a reduction in the catalase and polyphenol oxidase , suggesting that the increased hydrolysis of fats and proton transfer, as well as the reduced oxidative stress , contributed to the enhanced benzene and BaP biodegradation in the vegetable oil treatment. In addition, the microbial composition analysis results demonstrated that the enriched functional genera contributed to the increased biodegradation efficiency, and the functional genera in the microbial consortium responded differently to different biostimulants, and competitive growth was observed in the biostimulant-enhanced treatments. In addition, the enrichment of Pseudomonas and Rhodococcus species was noticed during the biostimulation of benzene and BaP co-contamination soil, and was positively correlated with the DHA enzyme activities , indicating that these species encode DHA genes which contributed to the higher biodegradation. In conclusion, multiple lines of evidence were provided to shed light on the mechanisms of biostimulant-enhanced biodegradation of PAHs and BTEX co-contamination with native microbial consortiums.

Effects of bioaugmentation by isolated Achromobacter xylosoxidans BP1 on PAHs degradation and microbial community in contaminated soil

Ben Zhang,Wei Xu,Yichi Ma,Xiaolong Gao,Huyang Ming,Jianli Jia

Polycyclic aromatic hydrocarbons (PAHs) are a group of organic pollutants ubiquitous and persistent in soil. In order to provide a viable solution for bioremediation of PAHs-contaminated soil, a strain of Achromobacter xylosoxidans BP1 with superior PAHs degradation ability was isolated from contaminated soil at a coal chemical site in northern China. The degradation of phenanthrene (PHE) and benzo[a]pyrene (BaP) by strain BP1 was investigated in three different liquid phase cultures, and the removal rates of PHE and BaP by strain BP1 were 98.47% and 29.86% after 7 days under the conditions of PHE and BaP as the only carbon source, respectively. In the medium with the coexistence of PHE and BaP, the removal rates of BP1 were 89.44% and 9.42% after 7 days, respectively. Then, strain BP1 was investigated for its feasibility in remediating PAH-contaminated soil. Among the four PAHs-contaminated soils treated differently, the treatment inoculated with BP1 exhibited higher removal rates of PHE and BaP (p < 0.05), especially the CS-BP1 treatment (inoculation of BP1 into unsterilized PAHs-contaminated soil) showed 67.72%, 13.48% removal of PHE and BaP, respectively, over 49 days of incubation. Bioaugmentation also significantly increased the activity of dehydrogenase and catalase in the soil (p<0.05). Furthermore, the effect of bioaugmentation on the removal of PAHs was investigated by measuring the activity of dehydrogenase (DH) and catalase (CAT) during incubation. Among them, the DH and CAT activities of CS-BP1 and SCS-BP1 (inoculation of BP1 into sterilized PAHs-contaminated soil) treatments inoculated with strain BP1 were significantly higher than those of treatments without BP1 addition during incubation (p < 0.01). The structure of the microbial community varied among treatments, but the Proteobacteria phylum showed the highest relative abundance in all treatments of the bioremediation process, and most of the bacteria with higher relative abundance at the genus level also belonged to the Proteobacteria phylum. Prediction of microbial functions in soil by FAPROTAX analysis showed that bioaugmentation enhanced microbial functions associated with the degradation of PAHs. These results demonstrate the effectiveness of Achromobacter xylosoxidans BP1 as a PAH-contaminated soil degrader for the risk control of PAHs contamination.

Relationship between PAH4 formation and thermal reaction products in model lipids and possible pathways of PAHs formation

Xiangxin Xu,Xiaofang Liu,Shuai Wang,Yongping Zou,Jixian Zhang,Li Liang,Chaoting Wen,Youdong Li,Xiaowei Xu,Xudong He,Guoyan Liu,Xin Xu

Lipids are closely related to the generation of PAHs during food thermal processing. During heating, lipids mainly triglycerides undergo hydrolysis, oxidation and decomposition. The relationship between the various products and the formation of PAHs is still unclear. This paper investigated the effect of different lipid standards on PAH4 production, and explored their thermal stability and reaction products to delve into nature of the differences in PAH4 production. Fatty acids were more prone to generate PAH4 than glycerides. The higher the degree of esterification of glycerides, the higher its thermal stability and the lower the content of PAH4 generated, implying that hydrolysis of glycerides promoted the generation of PAH4. In addition, there was a positive correlation between unsaturation in lipids and the PAH4 production. After heat treatment, hydroperoxides, unsaturated fatty alcohols and aldehydes, alkenes and aromatic substances were abundant in oleic acid and linoleic acid which produced the most PAH4. Thermal decomposition of lipid hydroperoxides was the pathway for the generation of conjugated hydrocarbon radicals, alcohols, aldehydes, and alkenes . The intramolecular cyclization and Diels-Alder reaction acted as ring-forming reactions, with consequent dehydrogenation , decarboxylation , side-chain breaks and radical reorganization, ultimately facilitating the amplification of the aromatic rings and the formation of PAHs.

In situ growth of IRMOF-3 combined with ionic liquids to prepare solid-phase microextraction fibers

Juan Zheng,Siyan Li,Ying Wang,Lei Li,Chengyong Su,Hong Liu,Fang Zhu,Ruifen Jiang,Gangfeng Ouyang

A superior solid-phase microextraction (SPME) fiber-coating material, IRMOF-3@ILs/PDMS, was prepared by the in situ growth of IRMOF-3 onto stainless-steel wires and protection with ionic liquids (ILs) and polydimethylsiloxane (PDMS). The ILs can efficiently prevent the substantial cracking of IRMOF-3 caused by moisture, and a thin PDMS film can protect the IRMOF-3@ILs material to achieve a much better extraction efficiency as well as excellent resistance to high temperature and high humidity. This IRMOF-3@ILs/PDMS coating possessed a porous structure, a rough surface and an increased lifespan (by at least 100 times) compared with that of IRMOF-3. The coating was evaluated by analyzing four polycyclic aromatic hydrocarbons (PAHs) in water, and good precision (<7.7%), low detection limits (12.0–15.4   ng   L −1 ), and wide linearity (50–20,000   ng   L −1 ) were achieved under the optimized conditions. The fiber was successfully applied to the sensitive analysis of PAHs in rainwater by coupling it with gas chromatography–mass spectrometry (GC–MS).

Moisture stable Ni-Zn MOF/g-C3N4 nanoflowers: A highly efficient adsorbent for solid-phase microextraction of PAHs

Ning Zhang,Chuanhui Huang,Ping Tong,Zunmei Feng,Xiaoping Wu,Lan Zhang

Volatile polycyclic aromatic hydrocarbons (PAHs) in water and soil are associated with status in the human body. Development of simple, efficient detection method is challenging due to the coating could be attacked by the abundance of water in the direct-immersion solid-phase microextraction. The stability of coating is essential to the analysis results. In this paper, a stable Ni-Zn MOF/g-C 3 N 4 (MG NFs) nanoflowers with cavity traps structure was firstly reported and acted as solid-phase microextraction (SPME) adsorbent for PAHs. Markedly enhanced moisture and acid stability of the MG NFs was obtained through the doping the hydrophobic graphitic carbon nitride (g-C 3 N 4 ) and metal ions into metal organic frameworks (MOFs). The aperture environment and ambient environment of MG NFs were changed by the doping of the Ni and the g-C 3 N 4, respectively. The moisture and acid stability of MG NFs were prominently increased under the dual protection. Compared to commonly used commercial coatings, the MG NFs own large surface area, unique nanoflowers structure and numerous open metal sites on the nanosheets, which demonstrated significant extraction superiority for PAHs. The MG NFs coated fiber was used for the SPME preconcentration of PAHs and couped with GC–MS for detecting PAHs. It presented low detection limits (0.1–3.0 ng L −1 ), wide linearity (0.3–5000.0 ng L −1 ) and good linearity (the correlation coefficient >0.9951). The inter-day and intra-day relative standard deviation (RSD) (n = 3) for three replicate extractions using one fiber was 3.8%–9.1%, and 3.5%–9.2%, respectively. The fiber-to-fiber reproducibility (n = 3) was 4.2–11.8%. The coupling method was successfully applied in the analysis of real water and soil samples with satisfactory recoveries of 82.9–109.2%, 84.2–106.4%, and the corresponding RSDs were 2.4–11.3%, 3.6–10.8%, respectively. The results indicated the effectiveness of NG NFs coated fiber in further practical application.

Surface modification of BiOBr/TiO2 by reduced AgBr for solar-driven PAHs degradation: Mechanism insight and application assessment

Xianbo Sun,Weiyu He,Xiaodi Hao,Haodong Ji,Wen Liu,Zhengqing Cai

A novel solar active AgBr/BiOBr/TiO 2 catalyst was synthesized by a facile coprecipitation method for solar-driven water remediation . The synthesized material composed of flower-like TiO 2 nanoparticles loaded on BiOBr nanosheets and with homogeneous surface distributed Ag/AgBr nanoparticles. The internal electric field between BiOBr/TiO 2 heterojunction greatly facilitated the charge carrier migration; the introduction of narrow band gap semiconductors (AgBr and BiOBr) promoted the visible light adsorption; and the Ag/AgBr nanoparticles acted as photosensitizer to further improve the light utilization. The new material showed 7.6- and 4.0-times activity of pure TiO 2 and BiOBr under solar light, and the contribution of reactive species on anthracene degradation followed the order of h + > O 2 • − > •OH. The degradation mechanism and pathway were proposed based on intermediates analysis and DFT calculation. The QSAR analysis revealed that the environmental risks of contaminants were greatly reduced during the photocatalysis process but some intermediates were still toxic. The high photocatalytic activity, stability and adaptability all indicated that this new material owns great application potential for cost-effective photocatalytic remediation of persistent organic contaminants under solar light.

Impact of arsenic and PAHs compound contamination on microorganisms in coking sites: From a community to individual perspective

Meng Zhou,Zishu Liu,Baolan Hu

Arsenic (As) and polycyclic aromatic hydrocarbons (PAHs) are highly toxic, carcinogenic and teratogenic, and are commonly found in soils of industrial sites such as coking plants. They exert environmental stresses on soil microorganisms, but their compounding effects have not been systematically studied. Exploring the effects of compound contamination on microbial communities, species and genes is important for revealing the ecological damage caused by compound contamination and offering scientific insights into soil remediation strategies. In this study, we selected soil samples from 0 to 100 cm depth of a coking site with As, PAHs and compound contamination. We investigated the compound effects of As and PAHs on microbial communities by combining high-throughput sequencing, metagenomic sequencing and genome assembly. Compared with single contamination, compound contamination reduced the microbial community diversity by 10.68%–12.07% and reduced the community richness by 8.39%–18.61%. The compound contamination decreased 32.41%–46.02% of microbial PAHs metabolic gene abundance, 11.36%–19.25% of cell membrane transport gene abundance and 12.62%–57.77% of cell motility gene abundance. Xanthobacteraceae , the biomarker for compound contaminated soils, harbors arsenic reduction genes and PAHs degradation pathways of naphthalene, benzo [a]pyrene, fluorene, anthracene, and phenanthrene. Its broad metabolic capabilities, encompassing sulfur metabolism and quorum sensing, facilitate the acquisition of energy and nutrients, thereby conferring ecological niche advantages in compound contaminated environments. This study underscores the significant impacts of As and PAHs on the composition and function of microbial communities, thereby enriching our understanding of their combined effects and providing insights for the remediation of compound contaminated sites.
