Preparation of PEG/ZIF-8@HF drug delivery system for melanoma treatment via oral administration

Luxi Peng,Jiajun Qiu,Lidan Liu,Xiaoyu Li,Xuanyong Liu,Yongjun Zhang

Melanoma is one of the highly malignant tumors whose incidence and fatality rates have been increased year by year. However, in addition to early surgical resection, there still lacks specific targeted drugs and treatment strategies. In this study, it was discovered that hinokiflavone (HF) encapsulated in zeolitic imidazolate framework-8 (ZIF-8) exhibited a superior anti-melanoma effect in vitro and in vivo. HF was encapsulated in ZIF-8 through a one-step synthesis method, and polyethylene glycol (PEG-2000) was used to optimize the size and dispersion of the drug-loaded complex (PEG/ZIF-8@HF). The results show that the prepared PEG/ZIF-8@HF has a high encapsulation efficiency (92.12%) and can achieve selective drug release in an acidic microenvironment. The results of in vitro anti-melanoma experiments indicate that PEG/ZIF-8@HF shows up-regulation of reactive oxygen species (ROS) levels and can restrain the migration and invasion of B16F10 cells. Moreover, in vivo animal experiments further confirm that PEG/ZIF-8@HF shows anti-tumor effect by up-regulating the pro-apoptotic proteins caspase-3 and caspase-8, and down-regulating the migration-promoting invasion protein MMP-9. This study developed a safe and effective oral administration of HF based on the high-efficiency delivery ZIF-8 system, which provides an effective treatment strategy for melanoma.

Study on the Strengthening Mechanism of a MIBC–PEG Mixed Surfactant on Foam Stability

Mengdi Xu,Fangyu Guo,Xicheng Bao,Xiahui Gui,Yaowen Xing,Yijun Cao

In the flotation process, the frother, which is typically a surfactant, can be added to the pulp to reduce the surface tension and create stable foam. Currently, the nonionic mixed surfactant is widely employed as the frother for fine coal flotation. In this study, we focused on examining the foam properties of a mixed surfactant comprising short-chain methyl isobutyl carbinol (MIBC) and long-chain polyethylene glycol-1000 (PEG). Analytical techniques such as surface tension measurement, dynamic foam stability measurement, bubble morphology observation, and foam film drainage measurement were used to investigate the foam properties in single and mixed surfactant solution from a macroscopic scale to a microscopic scale. The surface tension results indicated that PEG exhibited higher surface activity than MIBC, and the addition of PEG to MIBC resulted in a significant reduction in solution surface tension. The dynamic foam stability analysis revealed that the incorporation of a small amount of PEG into MIBC solution notably improved foam stability. Furthermore, the addition of PEG to the MIBC solution led to a shift in the bubble size distribution curve from a “double peak” to a “single peak” shape. This shift indicated a substantial reduction in bubble size, indicating an enhanced inhibition of bubble coalescence. Additionally, the liquid film drainage rate was significantly slowed down, and the stability of the liquid film was improved upon the addition of PEG to MIBC. This improvement can be attributed to the synergistic effect of MIBC and PEG molecules adsorbed at the gas–liquid interface. The synergistic effect of mixed MIBC–PEG was due to the additional surface tension gradient created by the difference in surface activity between PEG and MIBC. This surface tension gradient enhances the Marangoni flow of surfactant molecules, thereby improving the self-healing ability of the liquid film and increasing its stability.

CuS@mSiO2-PEG core–shell nanoparticles as a NIR light responsive drug delivery nanoplatform for efficient chemo-photothermal therapy

Xijian Liu,Qilong Ren,Fanfan Fu,Rujia Zou,Qian Wang,Guobing Xin,Zhiyin Xiao,Xiaojuan Huang,Qian Liu,Junqing Hu

We report a facile and low-cost approach to design a difunctional nanoplatform (CuS@mSiO2-PEG) as a near-infrared (NIR) light responsive drug delivery system for efficient chemo-photothermal therapy. The nanoplatform demonstrated good biocompatibility and colloidal stability, as well as high loading capacity for the anticancer drug (26.5 wt% for doxorubicin (DOX)). The CuS nanocrystals (core) within these CuS@mSiO2-PEG core–shell nanoparticles can effectively absorb and convert NIR light to fatal heat under NIR light irradiation for photothermal therapy, and the release of DOX from the mesoporous silica (shell) can be triggered by pH and NIR light for chemotherapy. When the CuS@mSiO2-PEG/DOX nanocomposites were irradiated by 980 nm light, both chemotherapy and photothermal therapy were simultaneously driven, resulting in a synergistic effect for killing cancer cells. Importantly, compared with chemotherapy or photothermal treatment alone, the combined therapy significantly improved the therapeutic efficacy.

Preparation of acrylamide-based poly-HIPEs with enhanced mechanical strength using PVDBM-b-PEG-emulsified CO2-in-water emulsions

Liwen Wang,Yongjia Liu,Lei Bao,Dongdong Hu,Yuan Zong,Gangsheng Tong,Ling Zhao,Tao Liu

Poly(vinyl acetate- alt -dibutyl maleate)- block -poly(ethylene glycol) (PVDBM- b -PEG) copolymers were synthesized via reversible addition–fragmentation chain transfer radical polymerization and used as emulsifiers to form stable CO 2 -in-water high internal phase emulsions (C/W HIPEs). Then, highly interconnected cellular polyacrylamide (PAM) and poly(acrylamide- co - N -hydroxymethyl acrylamide) [P(AM- co -HMAM)] poly-HIPEs with enhanced mechanical strength were prepared based on the stable C/W HIPEs. The porous structures of the PAM poly-HIPEs, as well as morphology and compressive modulus, could be influenced by the surfactant concentration and the length of the CO 2 -philic tails of the surfactants. PAM poly-HIPEs with the smallest average pore diameter (11.12 ± 0.62 μm) and the highest compressive modulus (22.65 ± 0.10 MPa) could be obtained by using the short CO 2 -philic chains of the PVDBM- b -PEG surfactant at a high concentration (1.0 wt %). Moreover, with the copolymerization of N -hydroxymethyl acrylamide (HMAM) comonomers with acrylamide, the compressive modulus of the obtained P(AM- co -HMAM) poly-HIPEs was three times higher than that of PAM poly-HIPEs. Both PAM and P(AM- co -HMAM) poly-HIPEs were employed as scaffolds to guide H9c2 cardiac muscle cellular growth. Fluorescence images showed that a smaller average pore size and a narrower pore-size distribution were helpful for cell growth and proliferation on these materials. © 2018 Wiley Periodicals, Inc. J. High Drug Payload 10-Hydroxycamptothecin Nanosuspensions Stabilized by Cholesterol-PEG: In Vitro and In Vivo Investigation

Yang Linjie,Jiang Jizong,Hong Jingyi,Di Jing,Liao Yonghong,Kuang Haixue,Wang Xiangtao

The objective of this study was to evaluate the feasibility of producing 10-hydroxycamptothecin nanosuspensions with high drug payload and to then determine the in vitro and in vivo characteristics of these nanosuspensions. Using cholesterol-PEG600 as a stabilizer, 10-hydroxycamptothecin nanosuspensions were successfully prepared using a precipitation-combined high-pressure homogenization method. A satisfactory drug payload of 90.39% (w/w) was achieved. The obtained nanosuspensions were spherical, with a mean particle size of 115.0 ± 0.4 nm, and they were monodisperse with a polydispersity index of 0.134 ± 0.001. The 10-hydroxycamptothecin remained in the same crystalline form in both the nanosuspensions and the bulk powder. In vitro, the 10-hydroxycamptothecin nanosuspensions released the encapsulated drug with nearly zero-order kinetics, and the accumulative release reached 90% within 72 hours. In vitro cytotoxicity assay showed that the 10-hydroxycamptothecin nanosuspensions had significantly enhanced cytotoxicity against HepG2 cells compared to the commercially available 10-hydroxycamptothecin injections. The in vivo study with H22 tumor-bearing mice and intravenous injection of the drug showed that in contrast to the 10-hydroxycamptothecin injections, the 10-hydroxycamptothecin nanosuspensions exhibited significantly enhanced biodistribution, particularly in the lung (393.40-fold AUC0–24 h, liver (192.35-fold AUC0–24 h, spleen (141.67-fold AUC0–24 h and tumor (64.21-fold AUC0–24 h. The 10-hydroxycamptothecin nanosuspensions also showed improved antitumor therapeutic efficacy over the injections (89.83% vs. 30.56%). This suggests that cholesterol-PEG600 may be an effective stabilizer for the preparation of hydrophobic drug nanosuspensions and that 10-hydroxycamptothecin nanosuspensions are a promising drug delivery system for tumor treatment.

Effects of PEG-TMS on the stability and antifouling performances of hydrocarbon-modified amphiphilic xerogel coatings

Dan Shang,Xiaoying Sun,Xiaoyao Shen,Jianzhong Hang,Lujiang Jin,Liyi Shi

Amphiphilic xerogel films with slight swelling were prepared by the acid-catalysed sol-gel method starting from tetraethoxysilane (TEOS), triethoxyoctylsilane (C8Si), n -octadecyl trimethoxysilane (C18Si) and poly(ethylene glycol) siloxane (PEG-TMS). In these amphiphilic xerogel films, the presence of hydrophobic chains, especially C18Si facilitated the emigration of PEG chain to the coating/air interface during the film formation/curing process, and then the PEG chains enriched at the surface of coatings further slightly emigrated to form hydration layer underwater confirmed by energy dispersive X-ray spectroscopic (EDS) analysis, captive air-bubble contact angle (CA) and CA hysteresis. Furthermore, the amphiphilic xerogel films showed the optimal water absorption ratio lower than 1.0% due to the restraint of higher crosslinked density and little network aperture of the films on PEG chains. The antifouling performances of amphiphilic xerogel films included the protein resistance for FITC-BSA, antibacterial property for Escherichia coli and the resistance for diatom Navicula , were better than those of the elastic silicone-based coatings and xerogel film without hydrophilic PEG chains. In general, the amphiphilic xerogel films containing both PEG and proper levels of C8Si and C18Si possessed excellent fouling release properties and slight swelling due to the synergistic effects.

Self-repairing nonfouling polyurethane coatings via 3D-grafting of PEG-b-PHEMA-b-PMPC copolymer

Kunlin Chen,Shuxue Zhou,Limin Wu

Durability of nonfouling coatings is a critical problem to be solved for their practical application. In this paper, self-repairing nonfouling polyurethane (PU) coating was fabricated by spraying of a triblock copolymer of polyethylene glycol (PEG), 2-hydroxyethyl methacrylate (HEMA) and 2-methacryloyloxyethyl phosphorylcholine (MPC) on pre-cured acrylic-based PU coatings. The triblock copolymer was synthesized through living radical polymerization mediated by an atom transfer radical polymerization. Anchoring of the copolymer took place both on the surface and in the interior (namely, 3D-grafting) of PU coatings through the diffusion of the copolymer and subsequently the chemical reaction between the pendent hydroxyl group of the copolymer and the remaining isocyanate groups in the pre-cured PU coatings. This 3D grafting procedure is rather simple and favorable for large-area application. The 3D-grafted PU coating has higher hydrophilicity (water contact angle: 33°). In contrast, if the triblock copolymer was used as an additive in PU coatings, the hydrophilicity of the PU coating did not obviously change. The 3D-grafted PU coating effectively inhibited the adhesion of protein and human platelet cells due to the synergistic effect of PEG and MPC. Moreover, after the coatings were detached and mechanically damaged, the surfaces can restore their hydrophilicity and possess better long-term anti-fouling ability than the control surface-grafted coatings underwater.

Fluorene methoxycarbonyl-PEG-deferoxamine conjugates “hitchhike” with albumin in situ for iron overload therapy

Linyi Xu,Rou Guan,Bohong Yu,Yicheng Li,Hongzhuo Liu,Yiguo Jiang

Although deferoxamine (DFO) has been approved for the treatment the iron overloaded diseases, its clinical application is impeded by very short circulation time and its relating toxicity. In this work, the fluorene methoxycarbonyl (FMOC) for “albumin hitchhiking” was used to prolong the plasma circulation time of DFO and reduce toxicity. The designed FMOC-PEG-DFO conjugates were found to reversible bind to albumin and gradually release DFO in vivo . Herein, the FMOC-PEG 1000 -DFO conjugates could increase 30 times the blood circulation time of DFO with the improvement of the iron elimination efficacy. Meanwhile, the conjugates markedly reduced the cytotoxicity of DFO. Taken together, the result demonstrated the FMOC-PEG 1000 -DFO conjugates could be a potential therapeutic choice for iron-overload-related diseases.

Controlled release of brefeldin A from electrospun PEG–PLLA nanofibers and their in vitro antitumor activity against HepG2 cells

Wanyun Liu,Junchao Wei,Ping Huo,Yunhua Lu,Yiwang Chen,Yen Wei

In this work, PEG–PLLA electrospun fibers were developed as a new controlled release system for macrolide antibiotic drug brefeldin A (BFA). SEM and XRD analyses of the BFA-loaded PEG–PLLA fibers revealed that the average diameter of fibers was below 950   nm with smooth surfaces, and the drug was well incorporated into the fibers in amorphous form. The release profiles of BFA in PBS were measured by HPLC, demonstrating that the controlled release of BFA could be gained for long time. The in vitro antitumor activity against human liver carcinoma HepG2 cells of the fibers containing 3%, 6%, 9%, 12% and 15% BFA were examined by MTT method, and the results showed that cell growth inhibition rates at 72   h were 64%, 77%, 80%, 81% and 85%, respectively. These results strongly suggested that the BFA/PEG–PLLA fibers had an effect of controlled release of BFA and were suitable for postoperative chemotherapy of cancers.

PEGylated molybdenum dichalcogenide (PEG-MoS2) nanosheets with enhanced peroxidase-like activity for the colorimetric detection of H2O2

Huimin Zhao,Yaxi Li,Bing Tan,Yaobin Zhang,Xiaochi Chen,Xie Quan

To improve the peroxidase-like activity of molybdenum dichalcogenides (MoS2), surface modification, such as combing with monometallic or bimetallic nanoparticles, is of great and widespread interest. However, only a few studies have been reported on the modification of MoS2 with non-metallic materials. Herein, we report a study on the peroxidase-like activity of novel PEGylated MoS2 (PEG-MoS2) nanosheets prepared using a simple liquid-phase exfoliation and pegylation process. Compared to the undecorated MoS2 nanosheets, PEG-MoS2 nanosheets have improved water dispersiblity and stability, which are beneficial for improving the peroxidase-like activity, as well as the catalysis velocity and affinity for TMB or H2O2. Based on PEGylated MoS2, we successfully established a new platform for the colorimetric detection of H2O2 with a limit of detection (LOD) of 1.18 μM, which is comparable to that of the majority of the reported enzyme mimics of inorganic graphene analogues. More importantly, the as-prepared PEG-MoS2 nanosheets are environmentally friendly, water dispersible, low cytotoxic, and easily prepared, which indicate that PEG-MoS2 is a type of promising enzyme mimics and extremely likely to be applied in cell-based assay systems without any adverse reactions.

PLGA-PEG-fucoxanthin nanoparticles protect against ischemic stroke in vivo

Qiyao Wang,Difan Zhang,Jiani Lu,Jiaying Zhang,Zhenquan Xuan,Luyun Gong,Mengxiang Yang,Lingli Jin,Jingyang Le,An Zhu,Hongze Liang,C. Benjamin Naman,Jinrong Zhang,Li Zhao,Shan He,Qinwen Wang,Hao Liu,Xiaojun Yan,Lingling Zhao,Wei Cui

Fucoxanthin is the most abundant marine carotenoid derived from edible brown seaweeds and has been used as a functional ingredient for the treatment of obesity. However, poor brain bioavailability of fucoxanthin has limited its application in combating neurological diseases. In this study, we demonstrated that 40 % fucoxanthin was released from polylactic- co -glycolic acid-polyethylene glycol (PLGA-PEG) nanoparticles at one day after incubation. Fucoxanthin nanoparticles (20–40 mg/kg, i.v. ) significantly prevented neurological and behavioral deficits in middle cerebral artery occlusion/reperfusion (MCAO)-treated rats via the reduction of M1 microglial activation, decrease of pro-inflammatory factor production, inactivation of the nuclear factor-κB (NF-κB) pathway, and elevation of the nuclear factor-E2-related factor 2 (Nrf2) pathway in ischemic penumbra. Moreover, fucoxanthin nanoparticles (5–15 μg/mL) inhibited hypoxia-stimulated neurotoxicity in a mouse hippocampal neuron cell line (HT22 cells). These results suggest that fucoxanthin nanoparticles enhanced brain bioavailability and produced neuroprotection against ischemic stroke in vivo with much lower effective concentrations than fucoxanthin itself, providing further support for the development of fucoxanthin as a functional ingredient for the prevention of ischemic stroke with proper nanoparticle formulations.

Engineering M2 macrophage-derived exosome-loaded PEG hydrogels for preventing pancreatic leakage and facilitating anastomotic healing

Chunyou Lai,Yunchao Yang,Pengfei Wei,Guangming Xiang,Guan Wang,Shasha Liu,Li Zeng,Guanghong Du,Ke Yang,Maozhu Yang,Xueying Zhang,Yunhuan Li,Xueqiao Yu,Bo Zhao,Yutong Yao

Pancreaticojejunostomy presents significant challenges due to the high risk of complications such as pancreatic leakage and poor tissue regeneration. However, current approaches to enhance surgical outcomes are often limited by issues related to biocompatibility, tissue repair efficacy, and inflammation modulation. Here, we report the potential of an adhesive polyethylene glycol (PEG) hydrogel incorporating M2 macrophage-derived exosomes (PEG/Exo M2 ) for improving tissue repair in pancreaticojejunostomy. M2 macrophage-derived exosomes exhibit potent anti-inflammatory properties and pro-angiogenic effects, making them ideal candidates for tissue regeneration. The PEG/Exo M2 hydrogel demonstrates excellent mechanical strength and strong antibacterial activity. In vitro assays reveal that the PEG/Exo M2 hydrogel promotes angiogenesis and cell migration while modulating inflammatory responses. In animal models, the PEG/Exo M2 hydrogel effectively reduces fibrosis and inflammation in a subcutaneous implantation model, and significantly improves tissue regeneration. In a pancreaticojejunostomy model, the PEG/Exo M2 hydrogel facilitates wound sealing, lowers the risk of pancreatic leakage, and enhances anastomotic repair, outperforming conventional sutures. These results highlight the PEG/Exo M2 hydrogel as a promising biomaterial for tissue repair and regeneration, with strong potential for clinical applications in surgical wound healing.
