Surface modification of porous PLGA scaffolds with plasma for preventing dimensional shrinkage and promoting scaffold–cell/tissue interactions

Peiming Liu,Lian Sun,Pingying Liu,Wenqian Yu,Qianhui Zhang,Weibing Zhang,Jing Ma,Pingsheng Liu,Jian Shen

Due to numerous merits, polyesters have been widely used for the preparation of porous scaffolds for biomedical applications. However, insufficient dimensional stability and weak cell interaction are two critical challenges to highly porous polyester scaffolds fulfilling their roles during applications. Here, we report the surface modification of PLGA scaffolds with air plasma for simultaneously tackling the dimensional shrinkage of PLGA scaffolds and improving scaffold–tissue integration. Highly porous PLGA scaffolds with interconnected pore structures and well orientated microtubules were prepared through lyophilization, and an air plasma process with varied duration was applied to the porous scaffolds. Water contact angle measurements indicated that moderate treatment (120 seconds) can significantly increase the wettability without distorting the morphology of and damaging the PLGA scaffolds. Further characterization (SEM, XPS, μ-CT, and theoretical computations) revealed that the plasma treatment enabled the generation of carboxyl groups on the surface of the PLGA scaffold, and the hydrophilic PLGA chain tends to extend under aqueous conditions in comparison to the hydrophobic pristine PLGA chain. Consequently, this could eliminate the hydrophobic-scaffold/water interface and weaken the driving force behind the distortion of the scaffolds in aqueous conditions, and finally inhibit overall shrinkage and maintain the microstructures of the porous scaffolds. Similar results were observed with porous PLA scaffolds, indicating the generality of air plasma treatment in inhibiting the shrinkage of polyester scaffolds. In the meantime, in vitro cell attachment experiments and in vivo subcutaneous implantation results consistently indicated that better maintaining the microstructure could significantly enhance the short-term cell attachment/proliferation on the scaffold and the long-term scaffold–tissue integration. These findings may inspire many creative and efficient strategies for the surface modification of polyester scaffolds to improve their performance for biomedical applications.

Ultrasound-Modulated Shape Memory and Payload Release Effects in a Biodegradable Cylindrical Rod Made of Chitosan-Functionalized PLGA Microspheres

Min Bao,Qihui Zhou,Wen Dong,Xiangxin Lou,Yanzhong Zhang

Minimally invasive implants and/or scaffolds integrated with multiple functionalities are of interest in the clinical settings. In this paper, chitosan (CTS) functionalized poly(lactic-co-glycolic acid) (PLGA) microspheres containing a model payload, lysozyme (Lyz), were prepared by a water-in-oil-in-water emulsion method, from which cylindrical shaped rod (5 mm in diameter) was fabricated by sintering the composite microspheres in a mold. High-intensity focused ultrasound (HIFU) was then employed as a unique technique to enable shape memory and payload release effects of the three-dimensional (3-D) structure. It was found that incorporation of CTS into PLGA microspheres could regulate the transition temperature Ttrans of the microsphere from 45 to 50 °C and affect shape memory ratio of the fabricated cylindrical rod to some extent. Shape memory test and drug release assay proved that HIFU could modulate the shape recovery process and synchronize the release kinetics of the encapsulated Lyz in the rod in a switchable manner. Moreover, the two processes could be manipulated by varying the acoustic power and insonation duration. Mechanical tests of the microspheres-based rod before and after ultrasound irradiation revealed its compressive properties in the range of trabecular bone. Examination of the degradation behavior indicated that the introduction of CTS into the PLGA microspheres also alleviated acidic degradation characteristic of the PLGA-dominant cylindrical rod. With HIFU, this study thus demonstrated the desired capabilities of shape recovery and payload release effects integrated in one microspheres-based biodegradable cylindrical structure.

PLGA-PEG-fucoxanthin nanoparticles protect against ischemic stroke in vivo

Qiyao Wang,Difan Zhang,Jiani Lu,Jiaying Zhang,Zhenquan Xuan,Luyun Gong,Mengxiang Yang,Lingli Jin,Jingyang Le,An Zhu,Hongze Liang,C. Benjamin Naman,Jinrong Zhang,Li Zhao,Shan He,Qinwen Wang,Hao Liu,Xiaojun Yan,Lingling Zhao,Wei Cui

Fucoxanthin is the most abundant marine carotenoid derived from edible brown seaweeds and has been used as a functional ingredient for the treatment of obesity. However, poor brain bioavailability of fucoxanthin has limited its application in combating neurological diseases. In this study, we demonstrated that 40 % fucoxanthin was released from polylactic- co -glycolic acid-polyethylene glycol (PLGA-PEG) nanoparticles at one day after incubation. Fucoxanthin nanoparticles (20–40 mg/kg, i.v. ) significantly prevented neurological and behavioral deficits in middle cerebral artery occlusion/reperfusion (MCAO)-treated rats via the reduction of M1 microglial activation, decrease of pro-inflammatory factor production, inactivation of the nuclear factor-κB (NF-κB) pathway, and elevation of the nuclear factor-E2-related factor 2 (Nrf2) pathway in ischemic penumbra. Moreover, fucoxanthin nanoparticles (5–15 μg/mL) inhibited hypoxia-stimulated neurotoxicity in a mouse hippocampal neuron cell line (HT22 cells). These results suggest that fucoxanthin nanoparticles enhanced brain bioavailability and produced neuroprotection against ischemic stroke in vivo with much lower effective concentrations than fucoxanthin itself, providing further support for the development of fucoxanthin as a functional ingredient for the prevention of ischemic stroke with proper nanoparticle formulations.

Preparation of porous PLGA/Ti biphasic scaffold and osteochondral defect repair

Chaoyong Zhao,Hongfang Zhang,Bing Cai,Guanglin Wang,Hongsong Fan,Xingdong Zhang

In this study, a porous PLGA/Ti biphasic scaffold was prepared and its potential to repair osteochondral defects was evaluated. The microstructure and mechanical properties of porous PLGA and porous Ti and their interface were characterized. The as-prepared biphasic scaffolds were implanted into rabbit knees for 1 and 3 months (PLGA/Ti group), and the porous PLGA (PLGA group) or empty defect (Untreated group) was also used. The results showed that both porous PLGA and porous Ti had an interconnected porous structure. The mechanical strength of porous PLGA and porous Ti was similar to that of cartilage and subchondral bone, respectively. The interface analysis revealed that the as-prepared biphasic scaffold had good overall integrity and interface stability. The gross observation and histological evaluation of specimens showed that hyaline-like cartilage filled the defects to a certain extent at 3 months for both PLGA/Ti and PLGA groups while the defects remained in an untreated group. In the PLGA/Ti group, better cartilage and subchondral bone repair was observed, and quantitative macroscopic and histological score evaluations confirmed this result, indicating that the porous biphasic PLGA/Ti scaffold had good biocompatibility and the best osteochondral defect repair ability in this study. The as-prepared porous biphasic scaffold showed the potential to be used in osteochondral tissue engineering for osteochondral defect repair.

Immobilizing natural macromolecule on PLGA electrospun nanofiber with surface entrapment and entrapment-graft techniques

Z.X. Meng,Q.T. Zeng,Z.Z. Sun,X.X. Xu,Y.S. Wang,W. Zheng,Y.F. Zheng

Surface entrapment is a convenient method to immobilize the natural macromolecules on the surface of synthetic polymers. In this study, the gelatin modified and sodium alginate/gelatin modified PLGA nanofibrous membranes were fabricated via surface entrapment and entrapment-graft techniques. The surface morphology of the each single modified PLGA nanofiber was as smooth as that of untreated PLGA nanofibers. The results of water angle contact measurements and tensile tests showed that the surface entrapment cannot only improve the hydrophilicity but also enhance mechanical properties of the modified nanofibrous membranes. In addition, the sodium alginate/gelatin modified electrospun PLGA nanofibrous membrane exhibited higher hydrophilicity and better biocompatibility than the simply gelatin modified PLGA nanofibrous membrane, which suggested the surface entrapment is a facile and efficient approach to surface modification for electrospun nanofibours membranes.

Curcumin encapsulated PLGA-chitosan nanocarrier: Fabrication, characterization, and in vitro differentiation evaluation on PC12 cells

Renzhang Liang,Cuilian Yu,Dong Ma,Xiaohan Cao,Liying Bai,Yulan Li,Jingxian Zhang,Jiayue Liu,Xia Qiao,Bingren Tian

Curcumin, a polyphenolic compound, demonstrates neuroprotective potential through its antioxidant, anti-inflammatory, and anti-amyloid activities. To enhance its delivery, we fabricated poly(lactic-co-glycolic acid) (PLGA) nanoparticles encapsulating curcumin and stabilized them with chitosan. Successful formulation was confirmed by FTIR, XRD, and SEM analyses. Zeta potential measurement confirmed a surface charge reversal from -28.1 mV (uncoated PLGA) to + 14.7 mV after chitosan coating. The nanoparticles exhibited favorable drug loading (4.09 ± 0.02 % to 5.67 ± 0.01 %) and encapsulation efficiency (14.33 ± 0.08 % to 19.85 ± 0.04 %), and provided a sustained release profile (>24 h) enhanced by chitosan. The curcumin encapsulated PLGA-chitosan nanocarriers demonstrated potent antioxidant activity (>80 % DPPH scavenging at 0.75 mg/mL) and no significant cytotoxicity against PC12 cells at concentrations up to 80 μg/mL. In HCT116 cells, the chitosan coating enhanced cellular uptake and cytoplasmic distribution. Crucially, the nanocarriers promoted PC12 cells differentiation, resulting in a high proportion of neurite-bearing cells. These findings indicate that the designed chitosan-coated PLGA nanoparticles are a promising delivery system for promoting neuronal differentiation.

Controllable Subtractive Nanoimprint Lithography for Precisely Fabricating Paclitaxel-Loaded PLGA Nanocylinders to Enhance Anticancer Efficacy

Xiang Zhang,Yurui Xu,Weiwei Zhang,Xinxin Fu,Zongbin Hao,Mengjia He,Denis Trefilov,Xinghai Ning,Haixiong Ge,Yanfeng Chen

Nanoimprint lithography presents a new strategy for preparing uniform nanostructures with predefined sizes and shapes and has the potential for developing nanosized drug delivery systems. However, the current nanoimprint lithography is a type of an additive nanofabrication method that has limited potential due to its restricted template-dependent innate character. Herein, we have developed a novel subtractive UV-nanoimprint lithography (sUNL) for the scalable fabrication of PLGA nanostructures with variable sizes for the first time. sUNL can not only fabricate a variety of predefined nanostructures by simply utilizing different nanoimprint molds but also precisely prepare scalable nanocylinders with different length to diameter ratios. Particularly, sUNL can fabricate paclitaxel-loaded PLGA nanocylinders (PTX-PLGA NCs) with high drug-loading rate of 40% and long storage stability over a year. We demonstrate that PTX-PLGA NCs target clathrin- and caveolae-mediated cell transport pathways and display increased cellular uptake, in comparison to traditional PTX-loaded PLGA nanoparticles (PTX-PLGA NPs), leading to enhanced anticancer effects. Therefore, sUNL represents a promising nanofabrication method for efficiently developing predefined drug delivery systems.

An ultrasound-driven PLGA/Zn-KNN hybrid piezoelectric scaffold with direct and immunoregulatory antibacterial activity for bone infection

Yuhao Zheng,Shu Wang,Wenhe Jin,Zhuoxuan Li,Guoju Yang,Xiaoxu Li,Ning Li,Yue Wang,Fan Sheng,Zhiming Song

Antibacterial piezoelectric materials have broad application prospects in the medical field because of their broad-spectrum antibacterial properties and no bacterial drug resistance. At present, one of the main problems in the application of piezoelectric materials is the low electrocatalytic efficiency, which limits its application in antibacterial field. In this study, a piezoelectric antibacterial (PLGA/Zn-KNN) scaffold was fabricated by incorporating zinc oxide (ZnO) into potassium-sodium niobate (KNN) and composited with a poly (lactic-co-glycolic acid) (PLGA) to achieve multicombination antibacterial for bone infection. The physicochemical properties of piezoelectric antibacterial scaffolds were analyzed. Bacterial, cell, and animal experiments were performed to characterize the antibacterial and infection treatment capabilities of piezoelectric scaffolds. The piezoelectric properties of the PLGA/Zn-KNN scaffold were enhanced by embedding ZnO particles into the KNN solid solution matrix. Furthermore, the piezoelectric scaffold released zinc ions, and electrical stimulation driven by ultrasound resulted in significant antibacterial effects through direct and immunoregulatory antibacterial pathways. Mechanistic investigation suggested that extracellular matrix ligands and complement and coagulation cascades may have a moderate effect on macrophage phagocytosis. This work highlights potential application methods for fabricating novel antibacterial hybrid piezoelectric scaffolds and engineering macrophages with immunoregulatory antibacterial activity.

Primary M1 macrophages as multifunctional carrier combined with PLGA nanoparticle delivering anticancer drug for efficient glioma therapy

Liang Pang,Ying Zhu,Jing Qin,Wenjie Zhao,Jianxin Wang

Glioma remains difficult to treat because of the infiltrative growth of tumor cells and their resistance to standard therapy. Despite rapid development of targeted drug delivery system, the current therapeutic efficacy is still challenging. Based on our previous studies, macrophages have been proved to be promising drug carrier for active glioma delivery. To make full use of macrophage carrier, primary M1 macrophages were proposed to replace regular macrophage to deliver nanodrugs into glioma, because M1 macrophages not only have the natural ability to home into tumor tissues, but they also have stronger phagocytic capability than other types of macrophage, which can enable them to uptake enough drug-loaded nanoparticles for therapy. In addition, M1 macrophages are not easily affected by harsh tumor microenvironment and inhibit tumor growth themselves. In this study, M1 macrophage-loaded nanoparticles (M1-NPs) were prepared by incubating poly(lactide-co-glycolide) (PLGA) nanoparticles with primary M1 macrophages. In vitro cell assays demonstrated M1 macrophage still maintained good tumor tropism capability after particle loading, and could efficiently carry particles across endothelial barrier into tumor tissues. In vivo imaging verified that M1-NPs exhibited higher brain tumor distribution than free nanoparticles. DOX@M1-NPs (doxorubicin-loaded M1-NPs) presented significantly enhanced anti-glioma effect with prolonged survival median and increased cell apoptosis. In conclusion, the results provided a new strategy exploiting M1 macrophage as carrier for drug delivery, which improved targeting efficiency and therapeutic efficacy of chemodrugs for glioma therapy.

An HPLC method for the pharmacokinetic study of vincristine sulfate-loaded PLGA–PEG nanoparticle formulations after injection to rats

Jianian Chen,Huijuan He,Shaoshun Li,Qi Shen

The aim of this study is to develop a simple and applicable HPLC method for the detection of vincristine in rat plasma after administration of poly(lactic- co -glycolic acid)–poly(ethylene glycol) (PLGA–PEG) nanoparticles loaded with vincristine sulfate (VCR). Vincristine was extracted from rat plasma and vinblastine sulfate was chosen as the internal standard (IS). Chromatographic separation of VCR and IS was achieved by a Dikma Dimonsil C 18 column (200   mm   ×   4.6   mm) with the mobile phase consisting of 0.02   M sodium dihydrogen phosphate–methanol (36:64, v/v, pH   =   4.7) at a flow rate of 1.0   mL/min. The ultraviolet detection wavelength was set at 276   nm. The calibration curve was linear over a concentration range of 0.05–5.0   μg/mL. The intra-day and inter-day accuracy for three quality controls (QC) samples was 93.48–107.74% and 92.61–96.58%, respectively; the precision was less than 9%. The average method recoveries for vincristine from spiked plasma at all QC levels were over 83%; and extraction recoveries were between 66 and 70%. Vincristine was stable in rat plasma for one month at −80   °C, for 8   h at room temperature , as well as during three freeze–thaw cycles. This HPLC method was applied successfully to the pharmacokinetic study of vincristine in rats after a single intravenous injection of VCR in physiological saline (F-VCR) solution, VCR-loaded PLGA–mPEG nanoparticles with (NP1) and PLGA–PEG–folate nanoparticles (NP2) suspension, respectively. There were significant differences in main pharmacokinetic parameters between F-VCR and the nanoparticles. Both kinds of VCR-loaded nanoparticles displayed improved pharmacokinetic profiles.

Development of composite PLGA microspheres containing exenatide-encapsulated lecithin nanoparticles for sustained drug release

Ni Dong,Chune Zhu,Junhuang Jiang,Di Huang,Xing Li,Guilan Quan,Yang Liu,Wen Tan,Xin Pan,Chuanbin Wu

This study aimed to prepare poly (D, L -lactic-co-glycolic acid) microspheres (PLGA-Ms) by a modified solid-in-oil-in-water (S/O/W) multi-emulsion technique in order to achieve sustained release with reduced initial burst and maintain efficient drug concentration for a prolonged period of time. Composite PLGA microspheres containing exenatide-encapsulated lecithin nanoparticles (Ex-NPs-PLGA-Ms) were obtained by initial fabrication of exenatide-loaded lecithin nanoparticles (Ex-NPs) via the alcohol injection method, followed by encapsulation of Ex-NPs into PLGA microspheres. Compared to Ms prepared by the conventional water-in-oil-in-water (W/O/W) technique (Ex-PLGA-Ms), Ex-NPs-PLGA-Ms showed a more uniform particle size distribution, reduced initial burst release, and sustained release for over 60 d in vitro . Cytotoxicity studies showed that Ms prepared by both techniques had superior biocompatibility without causing any detectable cytotoxicity. In pharmacokinetic studies, the effective drug concentration was maintained for over 30 d following a single subcutaneous injection of two types of Ms formulation in rats, potentially prolonging the therapeutic action of Ex. In addition, administration of Ex-NPs-PLGA-Ms resulted in a more smooth plasma concentration-time profile with a higher area under the curve (AUC) compared to that of Ex-PLGA-Ms. Overall, Ex-NPs-PLGA-Ms prepared by the novel S/O/W method could be a promising sustained drug release system with reduced initial burst release and prolonged therapeutic efficacy.

Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.
