PM 2 . 5 translocated across the air-blood barrier (ABB) can accumulate in the lungs and extrapulmonary organs, leading to both pulmonary and systemic pathophysiological effects. However, the mechanisms governing the translocation of micron-sized PM 2 . 5 (MPs) remain unclear. Here, a physiologically relevant in vitro ABB model is developed, comprising a triple co-culture system under respiratory-like cyclic strain. Using this platform, it is demonstrated that cyclic stretching enhances dTHP-1 phagocytosis and transmigration, synergistically promoting macrophage-mediated MPs translocation across the ABB in combination with strain-induced disruption of epithelial tight junctions. This cooperative mechanism governs MPs translocation across the ABB and markedly increases the transport of 2 µm particles compared to static conditions. By using scavenger receptor A (SR-A) inhibitors, this process are confirmed to be predominantly driven by macrophage phagocytosis. Furthermore, this study explores the potential fate of particles post-translocation, proposing their subsequent release from macrophages via active lysosomal exocytosis and passive release mediated by cell death. These findings provide new insights into mitigating PM 2 . 5 -induced health risks and inform macrophage-assisted pulmonary drug delivery strategies.

Exacerbated by human activities and natural events, air pollution poses severe health risks, requiring effective control measures to ensure healthy living environments. Traditional filtration systems that employ high-efficiency particulate air (HEPA) filters are capable of effectively removing particulate matter (PM) in indoor environments . However, these systems often work without considering the fluctuations in air pollution levels, leading to high energy consumption. This study proposed a novel PM 2.5 pollution-level adaptive air filtration system that combined elastic thermoplastic polyurethane (TPU) filters and an Internet of Things (IoT) system. The developed system can effectively adjust its filtration performance (i.e., pressure drop and PM 2.5 filtration efficiency) in response to real-time air quality conditions by mechanically altering the structures of TPU filters. Furthermore, while operating in varied pollution conditions, the proposed system demonstrated remarkable reductions in pressure drop without notably compromising the pollution control capability. Finally, the energy consumption of the pollution-level adaptive air filtration system was estimated when applied in mechanical ventilation systems in different cities (Hong Kong, Beijing, and Xi’an) with various pollution conditions. The results revealed that, compared to a traditional fixed system, the annual energy consumption could be reduced by up to ∼26.4 % in Hong Kong.

Objectives: Fine particulate matter (PM2.5), a small molecule particulate pollutant, can reach the lungs via respiration and cause lung damage. Currently, effective strategies and measures are lacking to prevent and treat the pulmonary toxicity of PM2.5. Astaxanthin (ASX), a natural xanthophyll carotenoid, has attracted attention due to its unique biological activity. Our research aims to probe into the prevention and treatment of ASX on PM2.5-induced lung injury and clarify its potential mechanism. Methods: Sprague–Dawley (SD) rats were given olive oil and different concentrations of ASX orally daily for 21 days. PM2.5 suspension was instilled into the trachea of rats every two days for one week to successfully develop the PM2.5 exposure model in the PM2.5-exposed and ASX-treated groups of rats. The bronchoalveolar lavage fluid (BALF) was collected, and the content of lung injury-related markers was detected. Histomorphological changes and expression of markers associated with oxidative stress, inflammation, iron death, and apoptosis were detected in lung tissue. Results: PM2.5 exposure can cause changes in lung histochemistry and increase the expression levels of TP, AKP, ALB, and LDH in the BALF. Simultaneously, inflammatory responses and oxidative stress were promoted in rat lung tissue after exposure to particulate matter. Additionally, ASX preconditioning can alleviate histomorphological changes, oxidative stress, and inflammation caused by PM2.5 and reduce PM2.5-related ferroptosis and apoptosis. Conclusion: ASX preconditioning can alleviate lung injury after PM2.5 exposure by inhibiting ferroptosis and apoptosis.

In this paper, a high-performance composite nanofiber membrane with excellent air filtration and dye degradation properties was prepared by incorporating MIL-53(Fe) nanoparticles into the electrospinning solution. The results showed that the filtration efficiency for PM2.5 of the PP/PVDF-HFP/MIL-53(Fe) composite nanofiber membrane reached up to 99.3% with the addition of 2 wt% MIL-53(Fe). Moreover, the composite nanofiber membrane exhibited steady filtration efficiency across different parameters, including air velocity, PM2.5 concentration, and extended testing time. Furthermore, the incorporation of MIL-53(Fe) significantly enhanced the removal efficiency of the composite nanofiber membrane toward Rh B, resulting in a remarkable 91% removal rate. The polymer/MOF composite nanofiber membranes prepared by simple and cheap electrospinning technology have a wide range of practical applications in the field of environmental protection. Highlights PPM composite nanofiber membrane was prepared via electrospinning method. MIL-53(Fe) nanoparticles were introduced to PPM composite nanofiber membrane. PPM composite nanofiber membrane showed excellent removal efficiency for PM2.5. PPM composite nanofiber showed a good removal performance for RhB in solution.

The growing threat of airborne particulate matter and pathogenic microorganisms to public health necessitates advanced air purification technologies capable of simultaneously removing particles and inactivating pathogens. In this study, we present a composite air filter based on a copper nanowire-modified phenolic resin meltblown nonwoven mat (CuNWs-PRMBM), which integrates physical filtration with Joule heating-induced sterilization. The PRMBM substrate, fabricated via melt-blowing, exhibits excellent thermal stability (decomposition temperature >450 °C) and low thermal conductivity (0.0369 W/m·K), offering a robust platform for Joule heating applications. Surface-deposited copper nanowires establish an efficient conductive network, enabling rapid (≤10 s) and uniform heating up to 124.6 °C under a low voltage of 3.0 V, with exceptional cycling stability. The composite displays superior filtration efficiency, achieving 99.6 % PM 2 . 5 removal at a low pressure drop of 87 Pa under 32 L·min −1 airflow, and maintains 94.9 % efficiency even at 85 L·min −1 . Furthermore, at 2.5 V (102.6 °C), the filter achieves complete inactivation of both S. aureus and E. coli within 15 min. Long-term tests confirm consistent performance over 120 h with sustained removal efficiencies exceeding 99.7 %. This work demonstrates a practical strategy for developing energy-efficient and safe air purification systems suitable for air purifiers and heating, ventilation, and air conditioning applications.

Lung damage can be caused by fine particulate matter (PM2.5). Thus, effective prevention strategies for PM2.5-induced lung injury are urgently required. Sesamin (Ses) is a natural polyphenolic compound that has attracted considerable attention of researchers because of its wide range of pharmacological activities. The present study aims to elucidate whether Ses pretreatment could alleviate PM2.5-induced lung damage and identify its possible mechanisms. Sprague–Dawley rats were orally dosed with 0.5% carboxymethylcellulose (CMC) and different concentrations of Ses once a day for 21 days. Then, the rats of the PM2.5 exposure group and Ses-treated group were exposed to PM2.5 by intratracheal instillation every 2 days for 1 week. Biomarkers associated with lung injury were detected in bronchoalveolar lavage fluid (BALF). Lung tissue was collected for histology, inflammation, oxidative stress, immunohistochemistry, and Western blot. Our results showed that PM2.5 exposure could cause pathological changes in lung tissue and increase levels of TP, AKP, and ALB in BALF. Meanwhile, exposure to PM2.5 can cause oxidative stress and inflammation in the lungs. In addition, Ses pretreatment could ameliorate histopathological injury, oxidative stress, and inflammation caused by PM2.5 exposure. It could also inhibit PM2.5-induced apoptosis and upregulation of autophagy-associated proteins. Collectively, our study indicated that Ses pretreatment could ameliorate PM2.5-induced lung damage via inhibiting apoptosis and autophagy in rats.

Background Abnormal lipid accumulation in macrophages may lead to macrophages foaming, which is the most important pathological process of atherosclerosis. Atmospheric PM 2.5 could enter the blood circulation and further affect the lipid metabolism of macrophages. But the underlying mechanism is not unclear. This study was undertaken to clarify the effect of PM 2.5 on lipid metabolism in macrophages, and to explore the role of inflammatory reaction and JAK2/STAT3 signaling pathway in this process. Method Macrophages derived from THP-1 cells were exposed to PM 2.5 (0,100,200,400 μg/mL) for 6 h and 12 h. STAT3 agonist ColivelinTFA is used to specifically excite STAT3. The survival rate of macrophages was detected by CCK-8. The lipid levels in macrophages were detected by colorimetry. The levels of inflammatory factors secreted by macrophages were detected by ELISA. Q-PCR was used to detect the mRNA expression levels, and Western Blot was used to detect the protein expression levels of JAK2/STAT3 pathway genes. Result The survival rate of macrophages was reduced by PM 2.5 , and the levels of TG, T-CHO and LDL-C of macrophages exposed to PM 2.5 were increased. PM 2.5 led to the increasing level of IL-6 and the decreasing level of IL-4, and the JAK2/STAT3 signaling pathway was inhibited by PM 2.5 . Colivelin TFA significantly decreased the increasing levels of TG, T-CHO and LDL-C levels, and increased the decreasing mRNA levels of IL-4, and LPL induced by PM 2.5 ( p < 0.05). Discussion PM 2.5 could cause the lipid accumulation of macrophages by inhibiting the JAK2/STAT3 signaling pathway, and inflammatory responses may be involved in this process.

Antimony (Sb), a carcinogenic pollutant, poses significant health risks via particulate matter with aerodynamic diameter <2.5 μm bound Sb (SbPM2.5). However, their sources and transboundary dynamics in ecologically fragile regions such as the Himalayas remain poorly resolved. Here, we combined antimony stable isotopes (ε123Sb), air-mass trajectory analysis, and multivariate mixing modeling to elucidate the sources and seasonal transport mechanisms of SbPM2.5 at Mount Qomolangma Station (QOMS) on the Tibetan Plateau, based on year-round samples collected during the premonsoon, monsoon, postmonsoon, and winter seasons. The annual mean Sb concentration (0.88 ± 0.64 ng m–3) was comparable to values reported for European urban sites, indicating a substantial transboundary influence. Sb isotopic signatures (ε123Sb: −1.92 to +1.61 ε) exhibited clear seasonality: lighter values prevailed in winter and premonsoon samples and were associated with South Asian coal combustion (28 ± 10%) and biomass burning (29 ± 10%), whereas heavier values in the postmonsoon period reflected an increased contribution from desert dust (41 ± 11%). Monsoon-driven precipitation modulated the observed isotopic composition with wet scavenging preferentially removing lighter isotopes. A multivariate mixing model that integrates Sb isotopes and Rb/Sr ratios resolved three primary sources, desert dust (23–64%), coal combustion (13–46%), and biomass burning (14–45%), whose influences are linked to westerly circulation and glacial-valley winds. This study represents the first application of Sb isotopes to trace atmospheric Sb at a high-altitude site and demonstrates monsoon-regulated transport and a dominant anthropogenic signal from South Asia. These findings provide critical insights into trans-boundary heavy-metal pollution in high-altitude ecosystems and inform strategies for mitigating environmental risks in the Himalayas and comparable regions worldwide.

Air particulate pollution from ever-increasing industrialization poses an enormous threat to public health. Thus, the development of a green air filter with high efficiency and performance is of urgent necessity. In this study, we introduce a new effective air filtration membrane that can be used for outdoor protection. The air filter’s composite nanofibre materials were prepared from polyvinyl alcohol (PVA)-sodium lignosulfonate (LS) via a “green” electrospinning method and thermal crosslinking. The addition of LS helped increase the PM 2.5 removal efficiency compared to that of a pure PVA nanofibre membrane. The pressure drops of the electrospun PVA-LS membranes exceeded those of the pristine PVA air filter. The remarkable air filtration performance was maintained even after 10 cycles of circulation filtration. In addition, the PVA-LS composite nanofibre membrane exhibited excellent mechanical properties and transparency due to the introduction of LS. This study provides new insight into the design and development of high-performance and high-visibility green filter media, which include personal protection and building screens.

Introduction Fine particulate matter (PM2.5) is a major airborne pollutant in low-temperature livestock housing. Individual environmental factors have been widely studied. However, the actual growth environments of livestock and poultry usually involve the combined effects of multiple environmental factors. Objectives This study aims to investigate the fundamental mechanisms underlying energy metabolism disorders in the lung-intestine axis of finishing pigs under the combined effects of low temperature and PM2.5. Methods This study characterized the physical, chemical, and microbial composition of the PM2.5 collected from pig houses. Inflammatory responses and energy metabolism were assessed at the tissue level. Key signaling pathways were functionally validated using in vitro cell models. Analysis of the lung-gut transcriptome elucidated the specific mechanisms by which the lung-gut axis responds to adverse environmental injuries. Results This study found that PM2.5 affects the energy metabolism process of lung macrophages, and low temperatures exacerbate the effects of PM2.5. Environmental factors trigger lactate production and mitochondrial dysfunction in macrophages through the HIF-PDK pathway. This dysfunction results in the release of pro-inflammatory factors that mediate systemic inflammation. Elevated levels of intestinal injury markers detected in serum indicate intestinal damage. Lung-gut transcriptomic analysis revealed impairments in immune and metabolic pathways. These findings were validated in intestinal tissues, supporting the activation of the lung-gut axis, which mediates systemic inflammation and intestinal damage following respiratory exposure. Conclusion In summary, these findings elucidate the specific mechanisms through which combined cold exposure and PM2.5 exposure disrupt pulmonary and intestinal immune-metabolic homeostasis. This underscores the necessity of implementing interventions in intensive livestock production systems.

PM 2.5 is the main particulate air pollutant whose aerodynamic diameter is less than 2.5 micron. The inflammation of various respiratory diseases are associated with PM 2.5 inhalation. Pro-inflammatory cytokine IL-1β generated from effected cells usually plays a crucial role in many kinds of lung inflammatory reactions. The exacerbation of Th immune responses are identified in some PM 2.5 related diseases. To elucidate the underlying mechanism of PM 2.5 -induced acute lung inflammation, we exposed Balb/c mice to PM 2.5 intratracheally and established a mice model. Acute lung inflammation and increased IL-1β expression was observed after PM 2.5 instillation. Regulatory factors of IL-1β (TLR4/MyD88 signaling pathway and NLRP3 inflammasome) participated in this lung inflammatory response as well. Treatment with compound essential oils (CEOs) substantially attenuated PM 2.5 -induced acute lung inflammation. The decreased IL-1β and Th immune responses after CEOs treatment were significant. PM 2.5 may increase the secretion of IL-1β through TLR4/MyD88 and NLRP3 pathway resulting in murine airway inflammation. CEOs could attenuate the lung inflammation by reducing IL-1β and Th immune responses in this model. This study describes a potentially important mechanism of PM 2.5 -induced acute lung inflammation and that may bring about novel therapies for the inflammatory diseases associated with PM 2.5 inhalation.
