Ros-sensitive hydrogel system incorporating mitochondria-targeted liposomes for periodontitis treatment

Qing Liu,Rui Cong,Jinping Yu,Xiaoxin Zhang,Jiasheng Tu,Yingchun Dong,Lei Jiang,Bin Chen

Periodontitis, the sixth most prevalent human disease, involves excessive ROS and mitochondrial dysfunction as key pathological drivers. However, therapeutic strategies simultaneously targeting both mechanisms for optimal regeneration remain scarce. To address this, we developed a ROS-responsive hydrogel (Gel) integrated with mitochondria-targeted liposomes encapsulating Coenzyme Q10 (CoQ10-MLip/Gel). The CoQ10-MLip combines CoQ10 (mitochondrial electron carrier) with arginine residues (mitochondrial-targeting motifs). The phenylboronic ester-based Gel degrades under high ROS, enabling controlled CoQ10-MLip release. Its self-healing properties and injectability ensure prolonged retention in periodontal pockets. This system effectively eliminated ROS, restored mitochondrial integrity, and significantly mitigated periodontal inflammation in vivo. The CoQ10-MLip/Gel platform thus offers a promising strategy for periodontitis treatment and potentially other chronic inflammatory diseases.

ROS-responsive dimeric prodrug-based nanomedicine targeted therapy for gastric cancer

Jiachi Ma,Yuzhong Chen,Wanqing Liang,Lei Li,Jun Du,Chengwu Pan,Chensong Zhang

Gastric cancer (GC) remains a major public health problem. Ursolic acid (UA) is reported to be effective in inhibiting GC; however, its low solubility and poor biocompatibility have greatly hindered its clinical application. Herein, an innovative reactive oxygen species (ROS)-sensitive UA dimeric prodrug is developed by coupling two UA molecules via a ROS-cleavable linkage, which can self-assemble into stable nanoparticles in the presence of surfactant. This new UA-based delivery system comprises the following major components: (I) dimeric prodrug inner core that can achieve high drug-loading (55%, w/w) and undergo rapid and selective conversion into intact drug molecules in response to ROS; (II) a polyethylene glycol (PEG) shell to improve colloid stability and extend blood circulation, and (III) surface-modified internalizing RGD (iRGD) to increase tumor targeting. Enhancement of the antitumor effect of this delivery system was demonstrated against GC tumors in vitro and in vivo. This novel approach offers the potential for clinical applications of UA.

ROS-responsive drug-releasing injectable microgels for ameliorating myocardial infarction

Shuqin Wang,Yuejun Yao,Liang Song,Zihe Zhai,Shifen Li,Zhaolong Wang,Liyin Shen,Yang Zhu,Wenxing Liu,Changyou Gao

Despite of the recent advances in regulatory T cell (Treg) therapy, a limited number of available cells and specificity at the desired tissue site have severely compromised their efficacy. Herein, an injectable drug-releasing (MTK-TK-drug) microgel system in response to in situ stimulation by reactive oxygen species (ROS) was constructed with a coaxial capillary microfluidic system and UV curing. The spherical microgels with a size of 150 μm were obtained. The MTK-TK-drug microgels efficiently converted the pro-inflammatory Th17 cells into anti-inflammatory regulatory T cells (Treg) cells in vitro , and the ROS-scavenging materials synergistically enhanced the effect by modulating the inflammation microenvironment. Thus, the microgels significantly reduced cardiomyocyte apoptosis and decreased the inflammatory response in the early stages of post-myocardial infarction (MI) in vivo , thereby reducing fibrosis, promoting vascularization, and preserving cardiac function. Overall, our results indicate that the MTK-TK-drug microgels can attenuate the inflammatory response and improve MI therapeutic effects in vivo .

Ginsenoside Rk1 inhibits postharvest fruit anthracnose by disrupting reactive oxygen species (ROS) homeostasis and activating the autophagic mechanism in Colletotrichum gloeosporioides

Xin-Yv Li,Yu-Jie Liu,Xian-Hua Meng,Chu Gong,Jun-Li Yang,Yin Qiang,Jun Wang,Xiu-Dan Hou

Anthracnose is a highly contagious and destructive plant disease caused by Colletotrichum gloeosporioides . The objective of this study was to investigate the antifungal activity and mechanism of ginsenoside Rk1 ( GRk1 ) against C. gloeosporioides. In vitro experiments demonstrated that GRk1 exhibited substantial antifungal activity, with EC 50 values of 64.58 mg L −1 , surpassing the plant-derived pesticide d -limonene . Additionally, GRk1 could effectively suppress the dissemination of anthracnose spots in postharvest fruit. By observing the microstructure of C. gloeosporioides , it was observed that the mycelium structure underwent notable changes, and autophagosomes engulfed the organelles in the experimental groups. Fluorescent staining revealed specific signals in mycelial autophagosomes, and Western Blot analysis confirmed the upregulation of the autophagy-related protein Atg8. These findings indicate that GRk1 may activate the autophagy process by promoting Atg8 expression. Proteomic analysis revealed that the expression of Sec20 and Mpv17 proteins was significantly downregulated, whose dysregulated expression is associated with the crosstalk between SNAREs and peroxisomes during vesicle trafficking and metabolic processes. Assessment of catalase (CAT) activity, hydrogen peroxide (H₂O₂) content, and superoxide anion (O₂ − ) levels further demonstrated that GRk1 could disrupt intracellular reactive oxygen species (ROS) homeostasis in mycelia. Moreover, the introduction of the ROS scavenger N -acetylcysteine (NAC) markedly inhibited autophagy, indicating that GRk1 promotes autophagy by inducing oxidative stress. Collectively, GRk1 inhibits pathogens via the ROS-autophagy axis. These findings suggested that GRk1 holds potential as a promising alternative to fungicides for the biological control of anthracnose.

Engineering ROS-responsive double network hydrogel as bioactive barrier for postoperative abdominal adhesions prevention

Lingling Ren,Yong Luo,Yanjuan Huang,Xianmin Shi,Huanxin Lin,Tao Zhang,Yujun Gong,Yao Liu,Dong Zheng,Wanzhen Li,Danni Xiao,Shengzhi Wang,Chunshun Zhao

Postoperative adhesions are common and severe complications, which affect up to 90 % of patients undergoing abdominal surgery. Despite the application of various strategies to minimize adhesions, the clinical outcomes remain far from satisfactory. Herein, we engineered a ROS-responsive and scavenging double-network hydrogel (PD-OHN) with multiple biofunctions and good mechanical properties for effective PAA prevention. First, a novel ROS-cleavable dithiothreitol (DTT) crosslinking monomer (DPBA) was synthesized. Subsequently, PD-OHN hydrogel was fabricated within 5 s by forming phenylborate ester bond networks between DPBA and polyvinyl alcohol (PVA), and acylhydrazone bond networks between oxidized hyaluronic acid (OHA) and adipic acid dihydrazide-modified hyaluronic acid (HA-ADH). After spraying, it can form a uniform and stretchable hydrogel film. Results showed that PD-OHN had good mechanical properties with a storage modulus about 20 kPa, satisfactory tissue adhesion strength of approximately 8 kPa, and an appropriate in vivo cecum retention time of about 21 days with good biosafety. More importantly, DPBA in PD-OHN hydrogel scavenged ROS via phenylboronate bond cleavage and the subsequent release of DTT, which intelligently alleviated oxidative stress according to the ROS levels in wound sites and induced pro-inflammatory M1 macrophages to polarize into anti-inflammatory M2 phenotype to alleviate inflammation. Further, the fibrinolytic system balance was recovered and fibrosis was reduced. Consequently, PD-OHN hydrogel effectively prevented adhesion formation in a cecum-sidewall abrasion rat model, and provided a promising whole course care anti-adhesion barrier for effective PAA prevention.

Preparation of meso-Ag/Al2O3 and synergistic water disinfection of metallic silver and ROS under visible light

Huajun Chen,Wenjie Tian,Wuxiu Ding

Metallic sliver-doped mesoporous Al 2 O 3 fibers (meso-Ag/Al 2 O 3 ) that can harvest visible light to speed up solar water purification, were synthesized by one-pot hydrothermal method. Insightful analyses of XRD, XRF and XPS of meso-Ag/ Al 2 O 3 verified that metallic silver occupied interstitial positions of Al 2 O 3 lattice forming interstitial solid solution. N 2 adsorption/desorption illustrated that meso-Ag/Al 2 O 3 exhibited significantly increases in the specific surface area, pore volume and average pore diameter after doping with silver. The UV–Vis diffuse reflectance spectra and disinfection performance indicated that meso-Ag/Al 2 O 3 can harvest visible light to promote the conversion of dioxygen to ROS for water disinfection compared with the widely used TiO 2 . With our approach, we achieved water disinfection of >99.999% inactivation of bacteria in 120 min with a small amount of material (8 mg/L) under simulated visible light.

N-acetylcysteine regulates dental follicle stem cell osteogenesis and alveolar bone repair via ROS scavenging

Meng Zhaosong,Liu Jiacheng,Feng Zhipeng,Guo Shuling,Wang Mingzhe,Wang Zheng,Li Zhe,Li Hongjie,Sui Lei

Background Dental follicle stem cells (DFSCs) show mesenchymal stem cell properties with the potential for alveolar bone regeneration. Stem cell properties can be impaired by reactive oxygen species (ROS), prompting us to examine the importance of scavenging ROS for stem cell-based tissue regeneration. This study aimed to investigate the effect and mechanism of N-acetylcysteine (NAC), a promising antioxidant, on the properties of DFSCs and DFSC-based alveolar bone regeneration. Methods DFSCs were cultured in media supplemented with different concentrations of NAC (0–10 mM). Cytologic experiments, RNA-sequencing and antioxidant assays were performed in vitro in human DFSCs (hDFSCs). Rat maxillary first molar extraction models were constructed, histological and radiological examinations were performed at day 7 post-surgery to investigate alveolar bone regeneration in tooth extraction sockets after local transplantation of NAC, rat DFSCs (rDFSCs) or NAC-treated rDFSCs.Results 5 mM NAC-treated hDFSCs exhibited better proliferation, less senescent rate, higher stem cell-specific marker and immune-related factor expression with the strongest osteogenic differentiation; other concentrations were also beneficial for maintaining stem cell properties. RNA-sequencing identified 803 differentially expressed genes between hDFSCs with and without 5 mM NAC. “Developmental process (GO:0032502)” was prominent, bioinformatic analysis of 394 involved genes revealed functional and pathway enrichment of ossification and PI3K/AKT pathway, respectively. Furthermore, after NAC treatment, the reduction of ROS levels (ROS, superoxide, hydrogen peroxide), the induction of antioxidant levels (glutathione, catalase, superoxide dismutase), the upregulation of PI3K/AKT signaling (PI3K-p110, PI3K-p85, AKT, phosphorylated-PI3K-p85, phosphorylated-AKT) and the rebound of ROS level upon PI3K/AKT inhibition were showed. Local transplantation of NAC, rDFSCs or NAC-treated rDFSCs was safe and promoted oral socket bone formation after tooth extraction, with application of NAC-treated rDFSCs possessing the best effect. Conclusion sThe proper concentration of NAC enhances DFSC properties, especially osteogenesis, via PI3K/AKT/ROS signaling, and offers clinical potential for stem cell-based alveolar bone regeneration.

A NIR-II Photoactivatable “ROS Bomb” with High-Density Cu2O-Supported MoS2 Nanoflowers for Anticancer Therapy

Jia Huang,Guiming Deng,Shuya Wang,Tianjiao Zhao,Qiaohui Chen,Yuqi Yang,Yongqi Yang,Jinping Zhang,Yayun Nan,Zhaoqian Liu,Ke Cao,Qiong Huang,Kelong Ai

The fast conversion of hydrogen peroxide (H 2 O 2 ) into reactive oxygen species (ROS) at tumor sites is a promising anticancer strategy by manipulating nanomedicines with near-infrared light in the second region (NIR-II). However, this strategy is greatly compromised by the powerful antioxidant capacity of tumors and the limited ROS generation rate of nanomedicines. This dilemma mainly stems from the lack of an effective synthesis method to support high-density copper-based nanocatalysts on the surface of photothermal nanomaterials. Herein, a multifunctional nanoplatform (MCPQZ) with high–density cuprous (Cu 2 O) supported molybdenum disulfide (MoS 2 ) nanoflowers (MC NFs) is developed for the efficient killing of tumors via a potent ROS storm by an innovative method. Under NIR-II light irradiation, the ROS intensity and maximum reaction velocity ( V max ) produced by MC NFs are 21.6 and 33.8 times that of the non–irradiation group in vitro, which is much higher than most current nanomedicines. Moreover, the strong ROS storm in cancer cells is efficiently formed by MCPQZ (increased by 27.8 times compared to the control), thanks to the fact that MCPQZ effectively pre–weakens the multiple antioxidant systems of cancer cells. This work provides a novel insight to solve the bottleneck of ROS-based cancer therapy.

Small Ceria Nanoclusters with High ROS Scavenging Activity and Favorable Pharmacokinetic Parameters for the Amelioration of Chronic Kidney Disease

Qianqian Wu,Lu Yang,Ling Zou,Wang Yang,Qingshan Liu,Anwei Zhang,Jiang Cao,Guangyou Shi,Jian He,Xiaochao Yang

The over production of reactive oxygen species (ROS) plays a critical role in the progression of chronic kidney disease (CKD). Organic ROS scavengers currently used for CKD treatment do not satisfy low dosage and high efficiency requirements. Ceria nanomaterials featured with renewable ROS scavenging activity are potential candidates for CKD treatment. Herein, a method for the synthesis of ceria nanoclusters (NCs) featured with small size of ≈1.2 nm is reported. The synthesized NCs are modified by three hydrophilic ligands with different molecular weights, including succinic acid (SA), polyethylene glycol diacid 600 (PEG600), and polyethylene glycol diacid 2000 (PEG2000). The surface modified NCs exhibit excellent ROS scavenging activity due to the high Ce 3+ /Ce 4+ ratio in their crystal structures. Compared with bigger-sized ceria nanoparticles (NPs) (≈45 nm), NCs demonstrate smoother blood concentration-time curve, lower organ accumulation, and faster metabolic rate superiorities. The administration of NCs to CKD mice, especially PEG600 and PEG2000 modified NCs, can effectively inhibit oxidative stress, inflammation, renal fibrosis, and apoptosis in their kidneys. Due to these benefits, the constructed NCs can ameliorate the progression of CKD. These findings suggest that NCs is a potential redox nanomedicine for future clinical treatment of CKD.

The role of GLS1-mediated glutaminolysis/2-HG/H3K4me3 and GSH/ROS signals in Th17 responses counteracted by PPARγ agonists

Yumeng Miao,Yun Zheng,Yanzhi Geng,Ling Yang,Na Cao,Yue Dai,Zhifeng Wei

Background: Peroxisome proliferator-activated receptor gamma (PPARγ) has the ability to counter Th17 responses, but the full mechanisms remain elusive. Herein, we aimed to elucidate this process in view of cellular metabolism, especially glutaminolysis. Methods: MTT, CCK-8, Annexin V-FITC/PI staining or trypan blue exclusion assays were used to analyze cytotoxicity. Flow cytometry and Q-PCR assays were applied to determine Th17 responses. The detection of metabolite levels using commercial kits and rate-limiting enzyme expression using western blotting assays was performed to illustrate the metabolic activity. ChIP assays were used to examine H3K4me3 modifications. Mouse models of dextran sulfate sodium (DSS)-induced colitis and house dust mite (HDM)/lipopolysaccharide (LPS)-induced asthma were established to confirm the mechanisms studied in vitro . Results: The PPARγ agonists rosiglitazone and pioglitazone blocked glutaminolysis but not glycolysis under Th17-skewing conditions, as indicated by the detection of intracellular lactate and α-KG and the fluorescence ratios of BCECF-AM. The PPARγ agonists prevented the utilization of glutamine and thus directly limited Th17 responses even when Foxp3 was deficient. The mechanisms were ascribed to restricted conversion of glutamine to glutamate by reducing the expression of the rate-limiting enzyme GLS1, which was confirmed by GLS1 overexpression. Replenishment of α-KG and 2-HG but not succinate weakened the effects of PPARγ agonists, and α-KG-promoted Th17 responses were dampened by siIDH1/2. Inhibition of KDM5 but not KDM4/6 restrained the inhibitory effect of PPARγ agonists on IL-17A expression, and the H3K4me3 level in the promoter and CNS2 region of the il-17 gene locus down-regulated by PPARγ agonists was rescued by 2-HG and GLS1 overexpression. However, the limitation of PPARγ agonists on the mRNA expression of RORγt was unable to be stopped by 2-HG but was attributed to GSH/ROS signals subsequent to GLS1. The exact role of PPARγ was proved by GW9662 or PPARγ knockout, and the mechanisms for PPARγ-inhibited Th17 responses were further confirmed by GLS1 overexpression in vivo . Conclusion: PPARγ agonists repressed Th17 responses by counteracting GLS1-mediated glutaminolysis/2-HG/H3K4me3 and GSH/ROS signals, which is beneficial for Th17 cell-related immune dysregulation.

A ROS-responsive multifunctional targeted prodrug micelle for atherosclerosis treatment

Zishuo Zhou,Yaxue Liu,Pei Xie,Zongning Yin

Atherosclerosis is a chronic multifactorial cardiovascular disease. To combat atherosclerosis effectively, it is necessary to develop precision and targeted therapy in the early stages of plaque formation. In this study, a simvastatin (SV)-containing prodrug micelle SPCPV was developed by incorporating a peroxalate ester bond (PO). SPCPV could specifically target VCAM-1 overexpressed at atherosclerotic lesions. SPCPV contains a carrier (CP) composed of cyclodextrin (CD) and polyethylene glycol (PEG). At the lesions, CP and SV exerted multifaceted anti-atherosclerotic effects. In vitro studies demonstrated that intracellular reactive oxygen species (ROS) could induce the release of SV from SPCPV. The uptake of SPCPV was higher in inflammatory cells than in normal cells. Furthermore, in vitro experiments showed that SPCPV effectively reduced ROS levels, possessed anti-inflammatory properties, inhibited foam cell formation, and promoted cholesterol efflux. In vivo studies using atherosclerotic rats showed that SPCPV reduced the thickness of the vascular wall and low-density lipoprotein (LDL). This study developed a drug delivery strategy that could target atherosclerotic plaques and treat atherosclerosis by integrating the carrier with SV. The findings demonstrated that SPCPV possessed high stability and safety and had great therapeutic potential for treating early-stage atherosclerosis.

Zuojin Pill inhibits the proliferation of esophageal cancer cells by inducing ROS production and apoptosis via regulation of PI3K/Akt signalling

Lingyu Wang,Ruolan Li,Meiyan Li,Jiayi Sun,Jia Liu,Min Xie,Yunhui Chen,Songqi Tang

This study aimed to investigate the inhibitory effect of Zuojin Pill (ZJP) on KYSE 150 cells and xenograft tumours of the digestive tract cancer – esophageal cancer (EC) and its underlying mechanism. Initially, in vivo research demonstrated that oral administration of ZJP effectively inhibited the growth of EC tumours, and in vitro studies revealed that ZJP suppressed the proliferation, migration, and invasion of cells. Then, network pharmacology prediction identified the core target, active ingredient, and involvement of the PI3K-Akt signalling pathway in ZJP treatment of EC. Cell studies validated that ZJP affected the expression of core targets, and its active ingredients inhibited the proliferation of cells. Additionally, ZJP was shown to induce cell apoptosis, decrease mitochondrial membrane potential, and increase the level of intracellular oxidative stress. Further experimental verifications suggested that the inhibitory effects of ZJP on EC might be attributed to the modulation of PI3 K/Akt signalling pathways.
