Synergistic U(VI) immobilization by a ternary Shewanella putrefaciens–chlorite–humic acid system: coupled adsorption and bio-reduction

Wang Xinyu,Qin Yilin,Gong Junyuan,Zhang Guohao,Peng Yinyin,Sun Gen,Ji Yanhao,Huang Yaling,Li Yahui,Nie Xiaoqin

This study aimed to enhance U(VI) removal from water by constructing a ternary system of Shewanella putrefaciens ( S. putrefaciens ), chlorite, and humic acid (HA). The ternary system achieved significantly higher U(VI) immobilization than single or binary systems, owing to synergistic effects of adsorption enrichment, electron shuttling, and bioreduction. Under optimal conditions, the U(VI) removal efficiency reached 91.6%. Importantly, pre-forming the ternary composite before U(VI) addition avoided false removal caused by HA-U(VI) complexation, ensuring accurate assessment of reductive immobilization. This work provides a reliable strategy for uranium-contaminated water remediation. Graphical abstract

Common Divalent Cations Elicit Sublethal Effects of Negatively Charged Nanoplastics on Shewanella oneidensis by Compromising Membrane Protein Functionality

Siyu Zhang,Boyu Jia,Weilin Huang,Changdong Ke,Yuekang Chen,Yanping Deng,Guining Lu,Zhi Dang,Chuling Guo

The toxicity of negatively charged nanoplastics (NNP) to bacteria is generally subtler than that of positively charged counterparts, owing to limited NNP-cell interaction. This study hypothesized that common environmental cations (Na+, Mg2+, Ca2+) could enhance interaction between NNP and Shewanella oneidensis, thereby inducing biological effects. Settling experiments and dynamic light scattering analyses showed that NNP-cell interaction increased in the order of Ca2+ ≈ Mg2+ > Na+, which can be attributed to the decreased electrostatic repulsion, as confirmed by extended Derjaguin–Landau–Verwey–Overbeek theory calculations. Although coexposure to NNP and cations did not result in significant lethality, extracellular electron transfer (EET) to insoluble electron acceptors was significantly inhibited by coexposing to NNP and Ca2+ (NNP+Ca2+, by 37%) or Mg2+ (NNP+Mg2+, by 20%), but not by NNP alone or NNP and Na+ treatments. Two-dimensional correlation spectroscopy indicated that membrane proteins predominantly mediate bacterial interactions with NNP. Physical membrane damage and structural alterations of membrane proteins were observed following coexposure to NNP+Ca2+ and NNP+Mg2+, impairing the direct EET pathways. Transcriptomic and physiological analyses further revealed that NNP+Ca2+ upregulated persister marker genes (spoT, ppx, relA) and induced ATP depletion, triggering cellular dormancy and suppressing membrane protein-mediated processes. By contrast, NNP+Mg2+ exposure activated protective responses, including two-component systems and flagellar assembly, consistent with the milder impairment of EET. Notably, these effects were absent in treatments with either cations alone or NNP alone. These findings reveal an overlooked ecological impact of NNP and underscore the potential for distinct bacterial responses to NNP in varying aquatic environments.

Decolorization and detoxification of water-insoluble Sudan dye by Shewanella putrefaciens CN32 co-cultured with Bacillus circulans BWL1061

Weijie Liu,Yanting You,Di Sun,Shiwei Wang,Jingrong Zhu,Cong Liu

Effluents loaded with various synthetic dyes are considered as a huge burden to the surrounding ecosystems. Sudan dyes are relatively difficult to decolorize due to its water-insolubility. In the present study, the strain Shewanella putrefaciens CN32 was firstly applied to decolorize Sudan dyes under the anaerobic condition , and the physicochemical parameters on the decolorization were optimized. The results demonstrated that the suitable decolorization condition was temperature 26 °C, initial pH 7.0–8.0 and NaCl concentrations 0–20 g/L. Electron competitive acceptors including nitrite, nitrate, dimethyl sulphoxide and oxygen could cause the significant inhibition to the decolorization of Sudan dyes. Biosurfactant rhamnolipid played a positive role in enhancing the decolorization of Sudan I. The co-culture of S. putrefaciens CN32 and Bacillus circulans BWL1061 is reported for the first time to accelerate the decolorization through improving the synergistic effect of enzymatic degradation and biological reductive effect. The highest decolorization of 90.23% to Sudan I was achieved within 108 h, suggesting that co-culture technique has a good potential in the treatment of dyeing wastewater. Furthermore, the microbial toxicity tests indicated that the toxicity of Sudan I to Escherichia coli BL21 and Bacillus subtilis 168 was obviously decreased after the decolorization.

Effect of Plasma-Activated Water on Shewanella putrefaciens Population Growth and Quality of Yellow River Carp (Cyprinus carpio) Fillets

Xiao Liu,Mingli Zhang,Xi Meng,Yanhong Bai,Xiuping Dong

ABSTRACT Plasma-activated water (PAW) is a new sanitizer that has received considerable attention for application in food industries. This research aimed to evaluate the effect of PAW on the inactivation of Shewanella putrefaciens and quality attributes of Yellow River carp ( Cyprinus carpio ) fillets. The carp fillet samples were immersed in sterile deionized water (SDW) or SDW activated by plasma discharge for 120 s (PAW120) for 1.5, 3.0, 4.5, or 6.0 min. After being treated by PAW120 for 6 min, the population of S. putrefaciens on carp fillets was significantly ( P < 0.05) decreased by 1.03 log CFU/g. Compared with SDW-treated samples, the L* value of PAW120–treated carp fillets was increased and the a* value was decreased after PAW120 treatment, whereas there was no significant ( P > 0.05) difference in the b* value. Compared with SDW, PAW120 caused no significant ( P > 0.05) changes in sensory properties and texture attributes of carp fillets including hardness, springiness, gumminess, and cohesiveness. However, 6-min PAW120 treatment caused a significant increase in the lipid oxidation level and a decrease in the pH value of the carp fillets. This work provides a basis for the potential application of PAW in the preservation of aquatic products.

Nitrate and iron salts enhanced Shewanella MR-1 bioreduction on Cr(VI) by stimulating cytochrome c electron transmission system

Chuanlong Huang,Mengqi Hao,Shurun Yao,Lirong Wang,Gang Liu,Zhuo Wei

Bioreduction of Cr(VI) by Shewanella through extracellular electron transmission (EET) attracts increasing interest due to cytochrome c (cyts c ) system especially conductive flagella (“nanowire”) structure. The responses of bioreduction efficiency, “nanowire” growth, cyts c synthesis, and e (electron) transmission intensity through CymA→MtrA→MtrB→MtrC→OmcA (Route 1) and CymA→NapA→NapB (Route 2) to iron ion (Fe 3 + ) and nitrate (NO 3 - ) were evaluated. Results showed that Fe 3+ and NO 3 - of 1–1.5 mM significantly ( p  < 0.01) enhanced bioreduction of Cr(VI) from 67% to 82% and 79%, respectively with promoted cyts c -mediated c onductivity (C p ). Both Fe 3+ and NO 3 - stimulated the growth of “nanowire” and synthesis of cyts c participating in EET with Fe 3+ showing stronger stimulation on Routes 1 while NO 3 - on Routes 2. The genes expression of cyts c on inner membrane (CymA), periplasmic space (NapA/NapB), and “nanowire” (MtrA/MtrB/MtrC, OmcA) show synchronous responses to Fe 3+ and NO 3 - . However, contents of mobile cyts c in extracellular polymeric substances (EPS) and structural cyts c in cell membrane showed inconsistent responses to Fe 3+ and NO 3 - . Overall, this study illustrated the regulation mechanism of e acceptors on cyts c- electron transmission system of Shewanella , which help improving the electrochemical efficiency on Shewanella and provide novel idea in chromium remediation strategy .

Enhanced Cd(II) Removal by Nano-Fe3O4/Alginate-Immobilized Shewanella-Serratia Consortium: Adsorption Performance, Reusability, and Preliminary Mechanistic Interpretation

Yan Shiying,Zhang Jian,Liu Yimeng,Chen Yiping,Jiang Tianlan,Gao Yu,Zhang Jie,Gao Jingqi,Li Fosheng,Zhao Jian,Feng Su

This study presents a composite biosorbent, Nano-Fe—SA—XB, synthesized by immobilizing a consortium of Shewanella putrefaciens and Serratia sp. B16 (XB) within a sodium alginate (SA) matrix reinforced with Nano-Fe 3 O 4 (Nano-Fe). A preliminary orthogonal formulation screening identified 4% Nano-Fe, 4% SA, 30 g/L XB biomass, and 4 h crosslinking as the selected preparation condition. At pH 7, using a wet dosage of 0.6 g (equivalent to a calculated dry mass of ~ 0.03 g), Nano-Fe—SA—XB achieved an 80.51% removal efficiency at a 20 mg/L initial Cd(II) concentration. Normalized to dry weight, the experimental maximum adsorption capacity was 72.5 mg/g at 150 mg/L, while the Langmuir-estimated capacity reached 87.8 mg/g. Kinetic and isotherm fitting suggested that Cd(II) uptake was consistent with pseudo-second-order kinetics and Sips/Langmuir-type adsorption behavior. SEM–EDS and FTIR analyses suggested that Cd(II) removal was associated with surface complexation and ion-exchange interactions involving hydroxyl, amide/carboxyl, phosphate, and calcium-containing sites. XRD showed no detectable new crystalline Cd-containing precipitates after adsorption. The material also showed NaCl-regeneration potential and 30-day Cd(II) retention/release stability. Overall, Nano-Fe—SA—XB shows potential as a reusable biosorbent for Cd(II)-contaminated water, while further mechanical, thermodynamic, surface-charge, pore-structure, magnetic, and chemical-state characterization is still needed. Exploring the interactions between Shewanella putrefaciens and Pseudomonas fluorescen on protein degradation of refrigerated large yellow croaker (Pseudosciaena crocea)

Jingxin Ye,Weiqing Lan,Jing Xie

Exploring the roles of dominant bacteria to spoilage is important in inhibiting spoilage. This study investigated related quality, protein degradation, and muscle microstructure in refrigerated large yellow croaker inoculated with Shewanella putrefaciens (S) and Pseudomonas fluorescens (P) as mono- or co-culture (SP). P. fluorescens had the most robust spoilage ability. P. fluorescens and the co-culture degraded major myofibrillar proteins and the links to disrupt muscle fibers, induced significant free water loss in muscle and formed amounts of trichloroacetic acid (TCA)-soluble peptides and total volatile basic nitrogen (TVB-N). S. putrefaciens caused severe mutual separation in myofibers. The competitive interactions between the two spoilage bacteria inhibited spoilage in the co-culture, while the numbers of differentially abundant peptides in the co-culture were enriched. Therefore, this study revealed competitive interactions of the co-culture of S. putrefaciens and P. fluorescens , which offered a thorough understanding of bacterial spoilage behavior in large yellow croaker.

Riboflavin-mediated reactivation of aged nano zero-valent iron by Shewanella oneidensis MR-1 for sustained Cr(VI) detoxification

Chengyu Yang,Jie Wei,Yixuan Li,Yuanhao Peng,Xuan Wu,Xianrui Zhang,Yingxin Ding,Shumi Lu,Chongling Feng,Chao Huang

Aging-induced surface passivation of nano zero-valent iron (nZVI) significantly diminishes its reactivity for Cr(VI) reduction, thereby impeding its practical application in environmental remediation. Herein, we propose a novel system integrating aged nZVI (AnZVI), Shewanella oneidensis MR-1, and the endogenous electron shuttle riboflavin (RF) to overcome the passivation limitations and enhance Cr(VI) removal. Remarkably, the AnZVI/MR-1/RF system achieved complete removal of 20 mg L − 1 Cr(VI) within 3 h, outperforming AnZVI/MR-1 and AnZVI alone by 23.8 % and 36.6 %, respectively. Mechanistic investigations revealed that RF mediated direct interfacial contact between AnZVI and MR-1, promoting microbial reduction of the AnZVI passivation layer. Electrochemical analyses confirmed RF’s critical role as an electron mediator in accelerating charge transfer between AnZVI and MR-1. Three-dimensional fluorescence mapping (3D-EEM) analysis revealed microbial extracellular polymeric substances (EPS) contributions to chromium immobilization processes. The immobilization of chromium is primarily attributed to the reduction of Cr(VI) to insoluble Cr(III) species, in conjunction with adsorption and surface complexation mechanisms. TEM validated a significant reduction in the thickness of the iron passivation layer, resulting in the exposure of the underlying Fe⁰ core and a resultant enhancement in reductive capacity. This work provides a robust strategy to overcome AnZVI’s limitations by coupling microbial activity with electron shuttling, offering new insights into sustainable remediation via biotic-abiotic synergies.

Unveiling the Biosynthesis Pathways of PdAu Nanoparticles Mediated by Cellular Oxidoreductases and Extracellular Polymeric Substances

Jin-Feng Ma,Feng-Ai Yang,Haonan Guo,Ya-Nan Hou,Zhihua Liu,Nanqi Ren,Ai-Jie Wang,Cong Huang

Biogenic PdAu nanoparticles (bio-PdAu NPs) synthesized by microorganisms exhibit remarkable catalytic activity relevant to environmental remediation. This study investigates the biosynthetic pathway of bio-PdAu NPs synthesized by Shewanella oneidensis MR-1, revealing that the bioreduction of Pd(II) and Au(III) follows first-order kinetics. Inhibition of NADH dehydrogenase (rotenone, Rot-PdAu), c-Cyts (diuron, Diu-PdAu), and hydrogenase (Cu2+, Cu-PdAu) decreased total metal-ion reduction efficiencies and Pd/Au composition, in which hydrogenases contributed most, followed by NADH dehydrogenase and c-Cyts. Notably, the presence of inhibitors affected the Au(III) reduction efficiency, altering particle size and composition. Statistical analysis indicated average particle sizes of 45.45, 50.15, and 13.16 nm for Rot-PdAu, Diu-PdAu, and Cu-PdAu, respectively, with a positive correlation to kinetic constants (R2 = 0.82). During biosynthesis, intracellular and extracellular pathways contributed 76.55 and 23.44%, respectively. For electrocatalytic hydrogen evolution reaction, the maximum current density (jmax) of bio-PdAu NPs exhibits substantially superior electrocatalytic performance compared to monometallic counterparts, with current densities approximately 1.8–3.2 times higher than individual bio-Pd or bio-Au systems. These findings deepen the understanding of the controlled synthesis pathways of bio-PdAu NPs and offer insights for developing biobimetallic NPs for environmental applications.

Effect of Mo2C-functionalized electrode interface on enhancing microbial cathode electrocatalysis: Beyond electrochemical hydrogen evolution

Qi Zhu,Jiaxin Peng,Yunhong Huang,Haiyan Ni,Zhong-er Long,Long Zou

Mo 2 C has recently been introduced into microbial cathode electrocatalysis due to its well-known catalytic activity for hydrogen evolution reaction (HER), but a low electrode potential (i.e., a high energy input) is required usually. However, in the range of potential where HER cannot occur, its effect on microorganism/cathode interfacial kinetics remains unknown. Herein, an active Mo 2 C interface is constructed on conventional carbon cloth electrode to investigate its impact on electric-driven fumarate reduction by a typical electroactive microorganism ( Shewanella oneidensis MR-1) at a potential of −0.36 V (vs . standard hydrogen electrode) that cannot meet the HER requirement. The Mo 2 C-functionalized electrode achieves a greatly increased current consumption density of ∼0.041 mA cm −2 , about three times that of the control, indicating a faster fumarate reduction rate. This substantially proves that the enhancement of microbial electrocatalytic kinetics at Mo 2 C-functionalized interface is not confined to the induced hydrogen evolution. By analyzing its interaction with extracellular electron transfer mediators (e.g., c -type cytochromes and riboflavin), a robust inward extracellular electron transfer process based on the rapid electrochemical reaction of riboflavin at the Mo 2 C-functionalized interface is elaborated. This work provides a new understanding of the mechanism by which the nanostructured Mo 2 C interface enhances microbial cathode electrocatalysis.

Polydopamine as electron shuttle and dispersant: Unlocking nZVI reactivity coupled with Shewanella oneidensis MR-1 for efficient Cr(VI) removal

Liwen Zhang,Shaohua Chen,Shanyi Tan,Huizi Wen,Shuyu He,Fen Zhang,Xiangru Zuo,Ling Xiong

The inherent limitations of nano zero-valent iron (nZVI) severely limit its widespread application in heavy metal pollution remediation. To address this, Shewanella oneidenis MR-1 (MR-1) was utilized to counteract the iron oxide passivation layer on nZVI surface, however, this approach suffered from low electron transfer rate between MR-1 and the passivation layer. In this research, a novel PDA-modified nZVI composite (PDA-nZVI) was synthesized. Compared to unmodified nZVI, the proportions of Fe 0 and Fe(II) on the PDA-nZVI surface increased 2.06 % and 21.63 %, respectively. The electrochemical tests (e.g., a more negative corrosion potential, a lower charge-transfer resistance, a higher current density, and a 3.4-times larger electrochemical active surface area for PDA-nZVI compared to nZVI) confirm that PDA enhances electron transfer from iron core to the outside, thereby promoting Fe 0 availability and Fe(II) generation on the PDA-nZVI surface. Meanwhile, PDA modification successfully enhanced the material's dispersibility and hydrophilicity in aqueous solutions. Results demonstrate that the Cr(VI) elimination capacity of the PDA-nZVI and MR-1 combined system (PDA-nZVI/MR-1) was markedly superior to that of the nZVI/MR-1 system. Mechanism analyses demonstrate that PDA can act as an electron shuttle to accelerate electron transfer between MR-1 and the passivation layer, thereby promoting more Fe(III) transformation to Fe(II). This process enables more Cr(VI) to be reduced to Cr(III), which is subsequently adsorbed or complexed on the material surface as Cr 2 O 3 /FeCr 2 O 4 and Cr x Fe 1-x (OH) 3 , ultimately being removed from the solution via co-precipitation. The PDA-nZVI/MR-1 system successfully overcomes the inherent limitations and exhibits an exceptional Cr(VI) elimination capability

Dual-photosensitizers photodynamic system for non-thermal inactivation of Shewanella putrefaciens and inhibition of microbial growth of refrigerated Litopenaeus vannamei

Dehua Wang,Yanzhe Lou,Xiaoming Jiang,Biqian Wei,Yong Xue,Zhaojie Li,Qingjuan Tang

Aquatic products are highly susceptible to microbial contamination and spoilage during storage, while traditional thermal sterilization often compromises their nutritional and sensory quality. In this study, a non-thermal dual-photosensitizer photodynamic treatment system composed of curcumin and octyl gallate was developed and activated under 420 nm light. The synergistic action of the two photosensitizers significantly enhanced the generation of reactive oxygen species (ROS), resulting in a 3.30 log reduction in Shewanella putrefaciens counts via membrane disruption, intracellular leakage, and genomic DNA degradation. Furthermore, the curcumin/octyl gallate-photodynamic treatment system was incorporated into a chitosan-based coating and applied to Litopenaeus vannamei . This treatment significantly inhibited increases in total viable counts, TVB-N, TBARS, and pH, thereby effectively extending the refrigerated shelf life. Microbial community analysis revealed marked shifts in bacterial composition, with suppression of dominant spoilage genera such as Psychrobacter, Pseudomonas , and Shewanella , confirming the broad-spectrum antibacterial efficacy of the treatment. Overall, curcumin/octyl gallate-photodynamic treatment represents an innovative, low-dose, and high-efficiency non-thermal sterilization approach for seafood preservation, offering a feasible alternative to traditional methods while maintaining microbial safety and product quality.
