Highly sensitive T 1–T 2 dual-mode MRI probe based on ultra-small gadolinium oxide-decorated iron oxide nanocrystals

Yashuang Miao,Peirong Chen,Miao Yan,Jianmin Xiao,Biao Hong,Ke Zhou,Guilong Zhang,Junchao Qian,Zhengyan Wu

Single-mode magnetic resonance imaging (MRI) contrast agents (CAs) in clinical settings are easily disturbed by calcification, bleeding, and adipose signals, which result in inaccurate diagnoses. In this study, we developed a highly efficient T1–T2 dual-mode MRI CA using an ultra-small gadolinium oxide-decorated magnetic iron oxide nanocrystal (GMIO). The gadolinium element could effectively alter the magnetic properties of the GMIO from soft-ferromagnetism to superparamagnetism. In addition, when the Gd/Fe ratio was 15% (designated as GMIO-2), the GMIO-2 possessed the best superparamagnetism and highest magnetism. Subsequently, T1 and T2 values of GMIO-2 were measured through a series of turbo spin-echo images and then multi-spin echo sequence, respectively. Based on this, T1 and T2 relaxivities of GMIO-2 were calculated and were the highest (r1: 1.306 m M−1 s−1 and r2: 234.5 m M−1 s−1) when compared to other groups. The cytotoxicity of GMIO-2 was negligible under a wide range of dosages, thus exhibiting excellent cell biocompatibility. Moreover, GMIO-2 could quickly diffuse into cells, leading to its effective accumulation. The systemic delivery of GMIO-2 resulted in an excellent T1–T2 dual-mode MRI contrast effect in kidneys, which is expected to improve the diagnosis of kidney lesions. Therefore, this work provides a promising candidate for the development of a T1–T2 dual-mode MRI CA.

Manganese-Doped Carbon Dots with Redshifted Orange Emission for Enhanced Fluorescence and Magnetic Resonance Imaging

Sijia Sun,Lining Zhao,Di Wu,Hongxin Zhang,Haichen Lian,Xiaolong Zhao,Aiguo Wu,Leyong Zeng

Metal-doped carbon dots (CDs) exhibited promising application in fluorescence and magnetic resonance (MR) imaging, but developing manganese-doped CDs (Mn-CDs) with long wavelength emission and enhanced MR performance is a challenge. Herein, using a one-step solvothermal method, Mn-CDs with redshifted orange emission and enhanced longitudinal relaxation were synthesized for fluorescence and MR imaging. The results indicated that the prepared Mn-CDs had a uniform size distribution, and the average size was 5 nm. Moreover, Mn-CDs possessed a stronger fluorescence performance than Mn-free CDs, and simultaneously, the emission wavelength can redshift from 542 nm (green emission) to 578 nm (orange emission), owing to the increasing N-doping. Because of the movement limit of Mn2+, Mn-CDs exhibited high T1-weighted MR imaging performance with a longitudinal relaxation rate of 12.69 mM–1·s–1. The in vivo experiments demonstrated their excellent fluorescence and MR imaging with safety and reliability. Therefore, the prepared Mn-CDs with orange emission can be an excellent candidate as a dual-modal nanoprobe for fluorescence and MR imaging.

Development of hollow ferrogadolinium nanonetworks for dual-modal MRI guided cancer chemotherapy

Ting Tang,Xiao Sun,Xuedong Xu,Yifeng Bian,Xiaojie Ma,Ning Chen

The development of hollow ferrogadolinium nanonetworks has not been reported for nanomedicine application until now. In this study, we developed a hollow and porous ferrogadolinium nanonetwork structure using the one-pot solvothermal method. This nanoparticle could be simultaneously used as a T1 and T2 dual-modal magnetic resonance imaging (MRI) contrast agent. In addition, the hollow lumen and abundant pores of the nanonetworks maximized the loading capacity and conferred the nanoplatforms for suitable anticancer drug loading capacity. Using these nanonetworks, MRI and anticancer experiments were conducted in vitro and satisfactory dual-modal MRI and cancer chemotherapy results were obtained. Therefore, the nanonetworks with dual-modal MRI and drug loading abilities effectively complement the ferrogadolinium composites' library and hold great promise in nanomedicine for simultaneous cancer diagnosis and chemotherapy.

Qiang Liu,Jin-Zhu Zhao,Fu-Sheng Zhou,Jiong Lan,Qiang Lu,Hong-Can Ren

GFH009 is a potent, highly selective, small molecule that targets and inhibits the activity of the CDK9/cyclin T1 regulatory complex of P-TEFb. This study aimed to develop and validate a highly selective and sensitive ultrahigh-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method for precise quantification of GFH009 in rat plasma. This method was subsequently employed for conducting toxicokinetic studies of GFH009 in rats. Plasma was prepared using a simple protein precipitation method by acetonitrile. Chromatographic separation of the analytes was achieved on a BEH C 18 analytical column with a rapid 3.0 min run time and a flow rate of 0.5 ml/min. The calibration curves for plasma samples exhibited excellent linearity over a wide concentration range of 1.0–1,000 ng/ml for GFH009. Intra- and inter-day accuracies were within 92.7–105.7%, and precisions were no more than 6.7%. Furthermore, the analyte demonstrated stability under four different storage conditions, with variations of <15.0%. This study pioneers a methodological innovation by introducing a highly reliable, specific and sensitive analytical method for GFH009 in rat plasma. The successful application of this method in toxicokinetic studies further underscores its significance, offering valuable insights for the methodology of clinical pharmacokinetic research.

Biomimetic Microenvironment Nanomodulators for Reactive Oxygen Species-Responsive T1-T2 Dual-Modal Magnetic Resonance Imaging and Visualized Treatment of Atherosclerosis

Xinhua Liu,Sunli Wan,Tingting Yao,Dan Wang,Mingkang Wang,Xinlong Liu,Xiaohui Deng,Runjianya Ling,Wangshu Zhu,Ze-Jian Wang,Yuanyuan Guo,Chuan Zhang,Yuehua Li

Atherosclerosis (AS) is a key pathological basis causing major cardiovascular diseases. Its proinflammatory microenvironment, characterized by aggregated inflammatory macrophages, excessive reactive oxygen species, and deficient phagocytosis, plays a pivotal role in driving AS pathogenesis. However, given the complexity of the microenvironment inside atherosclerotic plaques, developing highly effective and comprehensive nanoplatforms for precise diagnosis, efficient therapy, and reliable prognostic evaluation remains a major challenge. Herein, we construct a biomimetic nanomodulator (FeMn/Cur@CM) with responsive T1-T2 dual-modal magnetic resonance imaging (MRI) and multifaceted regulation functionalities of the plaque microenvironment for visualized treatment of AS. Coated with a macrophage membrane, curcumin-loaded FeMn/Cur@CM nanoparticles achieve prolonged circulation and specific targeting of inflammatory plaques. Diagnostically, FeMn/Cur@CM functions as a microenvironment-responsive dual-modal MRI contrast agent, amplifying T1-T2 signals in plaques. Therapeutically, FeMn/Cur@CM multifacetedly modulates the plaque microenvironment by alleviating oxidative stress, neutralizing protons, promoting macrophage reprogramming, and restoring macrophage efferocytosis. As a result, the integrated theranostic platform demonstrates remarkable plaque regression and simultaneously realizes real-time visualization of lesion progression via dual-modal MRI, providing precise diagnosis of plaque milieu. Through inflammation-targeted intervention coupled with real-time imaging feedback, this biomimetic nanomodulator establishes a promising strategy for the precise treatment of inflammatory vascular diseases, offering translational potential for AS management.

Potent dual-mode hyperthermia and chemodynamic therapy triggered by smart magnetic ring-shaped nanozymes under switchable T1–T2 MRI navigation

Ying Hu,Shuangshuang Guo,Jun Liu,Yuhao Yang,Jing Wang,Miaomiao Chen,Wenqian Zhao,Dandan Fan,Yuchuan Zhuang,Jing Li,Yanbo Dong,Andrey Tulupov,Xiao Wang,Jianfeng Bao

Emerging nanotechnologies in cancer therapy increasingly highlight the importance of integrating therapeutic and diagnostic functions into a single platform for enhanced efficacy and real-time monitoring. Here, we present a vortex‐shaped nanoring Fe 3 O 4 @MnO 2 @PDA (polydopamine) engineered for dual‐mode T 1 – T 2 magnetic resonance imaging (MRI)‐guided, dual‐thermal, and Fenton‐like chemodynamic therapy (CDT). The ring‐shaped nano Fe 3 O 4 core provides magnetothermal hyperthermia, while the MnO 2 layer, acting as a responsive nanozyme, catalyzes endogenous H 2 O 2 to generate oxygen and bolster CDT. Meanwhile, the PDA coating efficiently absorbs near‐infrared light, amplifying photothermal effects for tumor ablation. Under dual‐heating conditions, the temperature rise rate (2.05–2.92 times) and final equilibrium temperatures (1.57–2.27 times) exceed those of single‐mode heating, underscoring a robust synergistic effect. Additionally, due to the shell’s shielding effect weakening T 2 signals and the tumor microenvironment enhancing T 1 relaxation, the T 1 (5.75 Mm −1 s −1 ) to T 2 (91.56 mM −1 s −1 ) ratio is optimized specifically at tumor sites, thus achieving effective dual‐modal MRI contrast. Moreover, the smart MnO 2 design not only furnishes dynamic imaging enhancements but also enables environment‐triggered Mn 2+ release, offering further diagnostic and therapeutic benefits. In vitro and in vivo evaluations confirm significant tumor suppression with minimal systemic toxicity. These findings position nanoring Fe 3 O 4 @MnO 2 @PDA as a promising dual‐mode MRI‐guided, dual‐thermal, and chemodynamic platform for advanced cancer therapy.

Ultrasmall Ferrite Nanoparticles Synthesized via Dynamic Simultaneous Thermal Decomposition for High-Performance and Multifunctional T1 Magnetic Resonance Imaging Contrast Agent

Huan Zhang,Li Li,Xiao Li Liu,Ju Jiao,Cheng-Teng Ng,Jia Bao Yi,Yan E Luo,Boon-Huat Bay,Ling Yun Zhao,Ming Li Peng,Ning Gu,Hai Ming Fan

Large-scale synthesis of monodisperse ultrasmall metal ferrite nanoparticles as well as understanding the correlations between chemical composition and MR signal enhancement is critical for developing next-generation, ultrasensitive T1 magnetic resonance imaging (MRI) nanoprobes. Herein, taking ultrasmall MnFe2O4 nanoparticles (UMFNPs) as a model system, we report a general dynamic simultaneous thermal decomposition (DSTD) strategy for controllable synthesis of monodisperse ultrasmall metal ferrite nanoparticles with sizes smaller than 4 nm. The comparison study revealed that the DSTD using the iron-eruciate paired with a metal-oleate precursor enabled a nucleation-doping process, which is crucial for particle size and distribution control of ultrasmall metal ferrite nanoparticles. The principle of DSTD synthesis has been further confirmed by synthesizing NiFe2O4 and CoFe2O4 nanoparticles with well-controlled sizes of ∼3 nm. More significantly, the success in DSTD synthesis allows us to tune both MR and biochemical properties of magnetic iron oxide nanoprobes by adjusting their chemical composition. Beneficial from the Mn2+ dopant, the synthesized UMFNPs exhibited the highest r1 relaxivity (up to 8.43 mM–1 s–1) among the ferrite nanoparticles with similar sizes reported so far and demonstrated a multifunctional T1 MR nanoprobe for in vivo high-resolution blood pool and liver-specific MRI simultaneously. Our study provides a general strategy to synthesize ultrasmall multicomponent magnetic nanoparticles, which offers possibilities for the chemical design of a highly sensitive ultrasmall magnetic nanoparticle based T1 MRI probe for various clinical diagnosis applications.

Fluorine and Nitrogen Co-Doped Carbon Dot Complexation with Fe(III) as a T1 Contrast Agent for Magnetic Resonance Imaging

Jingmin Wang,Xiaolong Hu,Haizhen Ding,Xin Huang,Mingsheng Xu,Zhenzhen Li,Dong Wang,Xu Yan,Yang Lu,Yunjun Xu,Yan Chen,Paulo Cesar Morais,Yupeng Tian,Ren-Quan Zhang,Hong Bi

Commercial gadolinium-based materials have been widely used as contrast agents for magnetic resonance imaging (MRI), but the high toxicity of leaking free Gd3+ ions still raises biosafety concerns. Here, we develop a novel, safe, and efficient MRI contrast agent based on a stable Fe(III) complex of fluorine and nitrogen co-doped carbon dots (F,N-CDs) that was prepared from glucose and levofloxacin by a simple microwave-assisted thermal decomposition method. The obtained Fe3+@F,N-CD complex exhibits higher longitudinal relaxivity (r1 = 5.79 mM–1·s–1) than that of the control samples of the Fe3+@CD complex (r1 = 4.23 mM–1 s–1) and free Fe3+ (r1 = 1.59 mM–1 s–1) in aqueous solution, as assessed by a 1.5 T NMR analyzer. More importantly, the Fe3+@F,N-CD complex is very stable with a large coordination constant of 1.06 × 107 in aqueous medium. While incubated with HeLa cells, the Fe3+@F,N-CD complex shows clear MR images, demonstrating that it has potential to be an excellent MRI contrast agent. Furthermore, in vivo MRI experiments indicate that the Fe3+@F,N-CD complex provides high-resolution MRI pictures of 4T1 tumor bearing BALB/c mice 15 min after injection and can be completely excreted 2 h after administration. No cytotoxicity was observed with F,N-CDs and Fe concentration up to 0.2 mg/mL and 0.3 mM in 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cell proliferation assay, respectively. The possible mechanism of the enhanced MRI effect of the Fe3+@F,N-CD complex is therefore proposed. The extremely low toxicity, high r1 relaxivity, strong photoluminescence, and low synthetic cost enable the Fe3+@F,N-CD complex to be a safe and promising T1-weighted MRI contrast agent for clinical applications.

Microwave-assisted preparation of paramagnetic zwitterionic amphiphilic copolymer hybrid molybdenum disulfide for T1-weighted magnetic resonance imaging-guided photothermal therapy

Yiming Yu,Bin Chi,Leping Lin,Zhe Yang,Qianyuan He,Zushun Xu,Changfeng Yi,Jing Wang

Magnetic resonance imaging (MRI)-guided photothermal therapy (PTT) has recently attracted tremendous attention. In this study, a paramagnetic zwitterionic amphiphilic copolymer (PZAC) was successfully prepared and utilized as a multifunctional surfactant to form micelles in diethylene glycol to coordinate with a molybdenum disulfide precursor (MoS2). Uniform spherical MoS2 nanohybrids (MoS2@PZAC) were first prepared by a microwave-assisted solvothermal process, which is simpler, easier and more efficient than hydrothermal methods or exfoliation processes. In this nanoplatform, MoS2 serves as a phototherapeutic agent possessing a high photothermal conversion efficiency (33.8%), while PZAC acts as a T1-weighted MRI contrast agent. This nanoplatform has the advantages of ultralow toxicity, prolonged circulation time and bio-imaging guided capability. The cytotoxicity assessment showed the good cytocompatibility of MoS2@PZAC. Furthermore, MoS2@PZAC could effectively kill cancer cells upon 2 W cm−2 808 nm laser irradiation for 10 min in both in vitro and in vivo experiments. This work provides a novel and efficient solution to synthesize a multifunctional and uniform theranostic agent, and the results show that the as-prepared MoS2@PZAC can be used as a promising theranostic agent for T1-weighted MRI-guided PTT of cancer cells.

Integration of PEG-conjugated gadolinium complex and superparamagnetic iron oxide nanoparticles as T1–T2 dual-mode magnetic resonance imaging probes

Yang Li,Fu Shengxiang,Cai Zhongyuan,Liu Li,Xia Chunchao,Gong Qiyong,Song Bin,Ai Hua

The T1−T2 dual-mode probes for magnetic resonance imaging (MRI) can non-invasively acquire comprehensive information of different tissues or generate self-complementary information of the same tissue at the same time, making MRI a more flexible imaging modality for complicated applications. In this work, three Gadolinium-diethylene-triaminepentaaceticacid (Gd-DTPA) complex conjugated superparamagnetic iron oxide (SPIO) nanoparticles with different Gd/Fe molar ratio (0.94, 1.28 and 1.67) were prepared as T1–T2 dual-mode MRI probes, named as SPIO@PEG-GdDTPA0.94, SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67, respectively. All SPIO@PEG-GdDTPA nanocomposites with 8 nm spherical SPIO nanocrystals showed good Gd3+ chelate stability. SPIO@PEG-GdDTPA0.94 nanocomposites with lowest Gd/Fe molar ratio show no cytotoxicity to Raw 264.7 cells as compared to SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67. SPIO@PEG-GdDTPA0.94 nanocomposites with r1 (8.4 mM−1s−1), r2 (83.2 mM−1s−1) and relatively ideal r2/r1 ratio (9.9) were selected for T1–T2 dual-mode MRI of blood vessels and liver tissue in vivo. Good contrast images were obtained for both cardiovascular system and liver in animal studies under a clinical 3 T scanner. Importantly, one can get high-quality contrast-enhanced blood vessel images within the first 2 h after contrast agent administration and acquire liver tissue anatomy information up to 24 h. Overall, the strategy of one shot of the dual mode MRI agent could bring numerous benefits not only for patients but also to the radiologists and clinicians, e.g. saving time, lowering side effects and collecting data of different organs sequentially.

Achieving Ultrasmall Prussian Blue Nanoparticles as High-Performance Biomedical Agents with Multifunctions

Zhiguo Qin,Bo Chen,Yu Mao,Chu Shi,Yan Li,Xiao Huang,Fang Yang,Ning Gu

Prussian blue nanoparticles (PBNPs), which belong to the iron-based metal–organic frameworks, are important biomedical agents. Reducing the size of PBNPs can bring improved functional properties, but unfortunately, has been a long-standing challenge. Herein, sub-5 nm ultrasmall PBNPs (USPBNPs) were successfully synthesized by using ethanol/water mixture as the solvent and polyvinyl pyrrolidone (PVP) as the surface capping agent. Adjusting the ethanol/water ratio is not only able to control the nucleation time and size of PBNPs but also tune the conformation of PVP molecules so as to prevent interparticle attachment and enlargement. At an ethanol/water ratio of 3:1, highly stable USPBNPs with a size of ∼3.4 nm were synthesized. Due to their large specific surface area, they demonstrated high peroxidase-like and catalase-like activities, which outperform PBNPs synthesized by a conventional method. In addition, they also showed a high longitudinal relaxation rate (r1) of 1.3 mM–1 S–1, suggesting their potential to be used as T1 MRI agent.

Albumin-constrained large-scale synthesis of renal clearable ferrous sulfide quantum dots for T1-Weighted MR imaging and phototheranostics of tumors

Weitao Yang,Chenyang Xiang,Yan Xu,Shizhen Chen,Weiwei Zeng,Kai Liu,Xiao Jin,Xin Zhou,Bingbo Zhang

Ultrasmall-sized iron-based nanoparticles are showing increasing potentials to be alternatives as T 1 -weighted magnetic resonance imaging (MRI) contrast agents to the currently-used gadolinium-based compounds. However, their synthesis particularly in a large-scale and green fashion is still a big challenge. Herein, we report an albumin-constrained strategy to synthesize tiny and highly dispersible ferrous sulfide (termed [email protected] ) quantum dots (QDs) at ambient conditions. [email protected] QDs exhibit ultrasmall size of ca . 3.0 nm with an ultralow magnetization, affording them an appealing longitudinal relaxivity for T 1 -weighted MRI. The design principle leverages on albumin-mediated biomimetic synthesis and spatial isolation of the protein interface prevents bulk aggregation of the particles. Albumin was found to play crucial roles in the synthesis process: a constrained-microenvironment reactor for particle growth, a water-soluble layer for colloidal stability and a carrier for multi-functionality. This synthetic strategy was found facile, green and particularly large-scalable to 10 L. Mice experiments show good T 1 -weighted MRI capability of [email protected] QDs, significantly lighting the whole body organs, blood vessels and tumors. And interestingly, these QDs can be further used to conduct phototheranostic of tumor benefited from their intense absorption at near-infrared region. In particular, they can be cleared via glomerular filtration into bladder after treatment. Given this approach is biomimetic, scalable and does not require any complicated chemical synthesis or modifications, the method demonstrated here will find great potentials for clinical translation in T 1 -weighted MRI of diseases and inspire other functional tiny nanoprobes for biomedical applications.
