Exploring urinary modified nucleosides as biomarkers for diabetic retinopathy: Development and validation of a ultra performance liquid chromatography-tandem mass spectrometry method

Chen Yao,Daizhu Lv,Xueqing Zhou,Pengcheng Fu,Wen Sun,Jinlian Chen,Huan Lin

The dynamic modification of RNA plays a crucial role in biological regulation and is strongly linked to human disease development and progression. Notably, modified nucleosides in urine have shown promising potential as early diagnostic biomarkers for various conditions. In this study, we developed and validated a rapid, sensitive, and accurate UPLC-MS/MS method for quantifying eight types of modified nucleosides ( N 1 -methyladenosine (m 1 A), N 6 -methyladenosine (m 6 A), 5-methyluridine (m 5 U), 5-taurinomethyl-2-thiouridine (τm 5 s 2 U), 5-methylcytidine (m 5 C), 2'-O-methylcytidine (Cm), N 1 -methylguanosine (m 1 G), and N 7 -methylguanosine (m 7 G) in human urine. Using the method, we measured the urinary concentrations of m 1 A, m 6 A, m 5 U, τm 5 s 2 U, m 5 C, Cm, m 1 G, and m 7 G in a total of 21 control individuals and 23 patients diagnosed with diabetic retinopathy (DR). Cm levels showed promise as a diagnostic marker for diabetic retinopathy (DR), with a significant value (P < 0.01) and an AUC of 0.735. Other modified nucleosides also exhibited significant differences within specific subpopulations. As non-proliferative diabetic retinopathy (NPDR) signifies the latent early stage of diabetic retinopathy, we developed a multivariate linear model that integrates patients' sex, age, height, and urinary concentration of modified nucleosides which aims to predict and differentiate between healthy individuals, NPDR patients, and proliferative diabetic retinopathy (PDR) patients. Encouragingly, the model achieved satisfactory accuracy rates: healthy (81%), NPDR (75%), and PDR (80%). Our findings provide valuable insights into the development of an early, cost-effective, and noninvasive diagnostic approach for diabetic retinopathy.

Potential interactions between triazole antifungal agents and lorlatinib based on ultra-performance liquid chromatography-tandem mass spectrometry in rat plasma

Ye Zhongjiang,Wang Chenxiang,Li Rongqi,Chen Chuang,Yang Jianhui,Chen Yizhang,Fu Jing,Zhou Tao,Jin Mi,Huang Aifang,Zhang Xiuhua,Zhou Ziye

AimOur study is to investigate the effects of triazole antifungal drugs on the pharmacokinetics of lorlatinib in rats.MethodsThe samples were precipitated with methanol. Chromatographic separation was performed on a ultra-performance liquid chromatography (UPLC) system using a BEH C18 column. The mobile phase consisted of 0.1% formic acid water and methanol. Lorlatinib and crizotinib (internal standard) were detected in multiple reaction monitoring mode. The fragment ions were 407.3–228.07 for lorlatinib and m/z 450.3–260.0 for crizotinib. Lorlatinib and different triazole antifungal drugs were given to Sprague Dawley rats by gavage, and blood was collected from the tail vein at a certain time point. The validated UPLC–MS/MS method was applied to a drug interaction study of ketoconazole, voriconazole, itraconazole, and posaconazole with lorlatinib in rats.ResultsKetoconazole and voriconazole significantly inhibited lorlatinib metabolism. When administration with ketoconazole and voriconazole, the area under the curve from time zero to infinity of lorlatinib increased by 49.0% and 104.3%, respectively; the clearance decreased by 40.0% and 40.0%, respectively. While itraconazole and posaconazole did not affect lorlatinib pharmacokinetics.ConclusionThe UPLC-MS/MS-based assay is helpful to further understand the pharmacokinetics of lorlatinib in rats, and confirmed the findings that the combination of lorlatinib with CYP3A inhibitors should be avoided as predicted by our pre-clinical studies.

Synthesis of multiwalled carbon nanotubes/metal-organic framework composite for the determination of neonicotinoid pesticides in medicine and food homology products

Haizhi Huang,Na Li,Ya Chen,Xuping Shentu,Xiaoping Yu,Zihong Ye

A novel extraction adsorbent composite of MWCNTs/NH2-MIL-101(Fe) was synthesized, and was used to extract 6 kinds of neonicotinoid pesticides in medicine and food homology products. The composite was characterized by scanning electron microscope (SEM), fourier transform-infrared (FT-IR) spectroscopy, and X-ray powder diffraction (XRD). MWCNTs were enveloped around MOFs to provide physical support for the crystal structure. The adsorbent has higher adsorption capacity and reusability than pure NH2-MIL-101(Fe). Combined with UPLC-MS/MS, the method showed the low limit of detection (LOD) and limit of quantitation (LOQ) of 0.01–0.07 μg/kg and 0.04–0.22 µg/kg, respectively. It exhibited high extraction recovery of 77.86–101.10% for neonicotinoid pesticides in spiked samples. Meanwhile, this novel method could be successfully employed for the detection of other medicine and food homology products. Compared with previous reports, this method has advantages in detection limit and extraction recovery, indicating that it can be a preferential choice for the detection of neonicotinoid pesticides.

Comparative serum imidazole propionate profiling in colorectal adenoma and cancer by UPLC-MS/MS

Lv Jie,Chen Qianqian,Yang Lu,Guan Jing,Wang Sheng,Gui Gen,Yang Zhaoyun,Wang Xu,Sun Bin

IntroductionThe incidence of colorectal cancer (CRC) is increasing with colorectal adenomas recognized as key precancerous lesions. Emerging evidence suggests that Imidazole propionate (ImP), a metabolite of gut microbiota, is elevated in patients, indicating a potential role in tumorigenesis of CRC.ObjectivesThis study aimed to validate a novel method for detecting serum ImP using 3-piperazin-1-yl-propionic acid as an internal standard (IS), and to compare ImP levels between patients with colorectal adenomas and colorectal cancer. Methods Serum ImP were measured using Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry (UPLC-MS/MS) with 3-piperazin-1-yl-propionic acid as the IS. Four distinct patient groups were analyzed. Results The use of 3-piperazin-1-yl-propionic acid as an IS was successfully validated for the quantification of ImP. Serum ImP differed significantly among the groups, showing a stepwise increase from healthy controls to patients with colorectal adenomas and CRC. Notably, ImP concentrations were significantly higher in CRC patients than in other groups. Conclusion This study demonstrates the successful application of 3-piperazin-1-yl-propionic acid as an IS provides a viable method for measuring serum ImP concentrations. The results suggest a potential link between ImP and colorectal cancer development.

Rapid determination of amoxicillin in porcine tissues by UPLC-MS/MS with internal standard

Caiyun Zhang,Jiaxiong Zeng,Wenguang Xiong,Zhenling Zeng

A rapid, simple and reliable method using amoxicillin-D4 as internal standard (IS) for determination of amoxicillin (AMO) in porcine tissues was developed. The analytical methodology used a phosphate buffer (0.01 M, pH = 6.3) with trichloroacetic acid (50 mg/mL) for extraction and Oasis® HLB cartridges for clean-up followed by UPLC-MS/MS analysis. The optimized method was validated according to EU guideline 2002/657/EC and FDA guideline. The LOQ in muscle, fat, liver and kidney were set to 10 μg/kg which was below the maximum residue limits, while the LOD were set to 5 μg/kg. Results showed that using AMO-D4 as IS can effectively eliminate relative matrix interference and improve the recovery and precision. A good linearity was observed at the range of 10−500 μg/kg with a correlation coefficients (R 2 ) larger than 0.99. The mean recoveries were higher than 87.0 %, and the RSDs were lower than 11.2 %. This method could be applied to conveniently analysis the residue of AMO in different porcine tissues.

Yuan Shuai,Wang Xin,Wang Roujia,Luo Ruxin,Shi Yan,Shen Baohua,Liu Wei,Yu Zhiguo,Xiang Ping

Assessing collective drug consumption based on the concentrations of illicit drugs and their metabolites in wastewater is a new technology. Currently, this technology is receiving attention in China, and methods for multiple illicit drug detection in wastewater are urgently needed. In our study, a method with a short runtime (7 min), a small solid-phase extraction (SPE) loading volume (50 mL) and high sensitivity (lower limits of quantitation (LLOQs) ranged from 0.2 to 5 ng/L) was developed for the simultaneous determination of amphetamines, ketamine, opiates, cocaine and their metabolites in wastewater. Samples were enriched by SPE on a mixed-mode sorbent (Oasis MCX) and analyzed by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The limits of detection (LODs) ranged from 0.1 to 2 ng/L, and the LLOQs varied between 0.2 and 5 ng/L. Moreover, the method developed was applied to real wastewater samples collected from 15 different wastewater treatment plants (WWTPs). In the results, the most abundant compounds were morphine (1.8–46.6 ng/L) and codeine (3.7–24.9 ng/L), which were detected in 13 WWTPs. After successful optimization of the UPLC-MS/MS conditions and sample loading pH, the method developed is able to meet the needs of common illicit drug monitoring and high-throughput analysis requirements. SPE-UPLC-MS/MS method was developed for the for determination of 11 illicit drugs in wastewater. The method has a smaller SPE loading volume, a shorter UPLC runtime and higher sensitivity than other methods. The method can meet the requirements for current drug abuse monitoring in China.

Jigucao capsules (JGCC) have the effects of soothing the liver and gallbladder and clearing heat and detoxification. It is a good medicine for treating acute and chronic hepatitis cholecystitis with damp heat of the liver and gallbladder. However, the existing quality standard of JGCC does not have content determination items, which is not conducive to quality control. In this study, serum pharmacochemistry technology and UNIFI data processing software were used to identify the blood prototype components and metabolites under the condition of the obvious drug effects of JGCC, and the referenced literature reports and the results from in vitro analysis of JGCC in the early stage revealed a total of 43 prototype blood components and 33 metabolites in JGCC. Quality markers (Q-markers) were discovered, such as abrine, trigonelline, hypaphorine and isoschaftoside. In addition, ultra-high-performance liquid chromatography–triple quadrupole mass spectrometry (UPLC-QQQ-MS) was used to determine the active ingredients in JGCC. The components of quantitative analysis have good correlation in the linear range with R2≥ 0.9993. The recovery rate is 93.15%~108.92% and the relative standard deviation (RSD) is less than 9.48%. The established UPLC-MS/MS quantitative analysis method has high sensitivity and accuracy, and can be used for the quality evaluation of JGCC.Keywords:serum pharmacochemistry;jigucao capsule;quality markers;ultra-high-performance liquid chromatography-mass spectrometry;quality evaluation

Fe3O4@SiO2–BD–DADB-COF is proposed as a novel magnetic covalent organic framework for the determination and extraction of 15 macrolide antibiotics in water and honey

Hao Zhang,Weihao Ma,Chunyu Qiang,Jiayuan Nie,Ling Ma,Yawei Zhang,Ke Wang

In our research, an emerging magnetic covalent organic framework (Fe3O4@SiO2–BD–DADB-COF) was formulated through Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), etc. Several parameters affecting the extraction process were refined. Accordingly, a novel method of determining 15 macrolides (MALs) in honey and water was established through the Fe3O4@SiO2–BD–DADB-COF as a magnetic solid-phase extraction (MSPE) adsorbent and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). In consequence, the standard curves for the 15 MALs exhibited an exceptional linearity from 0.1 to 200 μg L−1, and the correlation coefficients (R2) varied from 0.9990 to 0.9999. The recoveries fell between 70.01% and 115.56%, with the relative standard deviations (RSDs) being below 9.93% (n = 5). The detection limits reached 0.001–0.075 μg L−1 with the quantification limits being 0.004–0.228 μg L−1. Ultimately, the method was excellently applied to the analysis of MALs in honey and water.

Quantitative and Discriminative Evaluation of Contents of Phenolic and Flavonoid and Antioxidant Competence for Chinese Honeys from Different Botanical Origins

Shi Shen,Jingbo Wang,Qin Zhuo,Xi Chen,Tingting Liu,Shuang-Qing Zhang

Phenolics and flavonoids in honey are considered as the main phytonutrients which not only act as natural antioxidants, but can also be used as floral markers for honey identification. In this study, the chemical profiles of phenolics and flavonoids, antioxidant competences including total phenolic content, DPPH and ABTS assays and discrimination using chemometric analysis of various Chinese monofloral honeys from six botanical origins (acacia,Vitex, linden, rapeseed,AstragalusandCodonopsis) were examined. A reproducible and sensitive ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was optimized and validated for the simultaneous determination of 38 phenolics, flavonoids and abscisic acid in honey. Formononetin, ononin, calycosin and calycosin-7-O-β-d-glucoside were identified and quantified in honeys for the first time. Principal component analysis (PCA) showed obvious differences among the honey samples in three-dimensional space accounting for 72.63% of the total variance. Hierarchical cluster analysis (HCA) also revealed that the botanical origins of honey samples correlated with their phenolic and flavonoid contents. Partial least squares-discriminant analysis (PLS-DA) classification was performed to derive a model with high prediction ability. Orthogonal partial least squares-discriminant analysis (OPLS-DA) model was employed to identify markers specific to a particular honey type. The results indicated that Chinese honeys contained various and discriminative phenolics and flavonoids, as well as antioxidant competence from different botanical origins, which was an alternative approach to honey identification and nutritional evaluation.Keywords:UPLC-MS/MS;antioxidant competence;phenolic;flavonoid;chemometrics;monofloral honey

UPLC-MS/MS and network pharmacology-based antidepressant study of Chaihu Jia Longgu Muli decoction

Huang Huang,Xiuhui Guo,Siqi Quan,Xiangli Yan,Yucheng Li,Erping Xu

Background Depression is a mental disorder characterized by persistent low mood and cognitive impairment. Chaihu Jia Longgu Muli Decoction (CLMD) is an effective and relatively safe therapeutic method for depression. However, the chemical composition of CLM and its underlying mechanisms remain unclear. Objective This study aimed to explore the potential mechanisms of CLMD in treating depression by integrating UPLC-MS/MS and network pharmacology. Methods Ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was used to determine the ingredients of CLMD. The targets of these components were predicted and screened using the TCMSP, SuperPred, and SwissTargetPrediction databases. The disease targets related to depression were obtained from the GeneCards, OMIM, and DisGeNET databases. The protein-protein interaction (PPI) networks of CLMD and depression were constructed using the STRING database, and Origin 2022 software was used to identify cross-targets by screening the core-acting targets. A network diagram was constructed using Cytoscape3.7.2 software. The DAVID database was used for GO and KEGG enrichment analysis of the core genes. Then, molecular docking was performed to verify the results using AutoDock 4.2.6, AutoDock Tools 1.5.6, PyMOL2.3.0, and Ligplot V 2.2.4 software. Molecular dynamics simulation was performed by iNODS. Finally, a series of in vitro experiments were performed to examine the function of baicalin in corticosterone-stimulated HT22 cells. Results A total of 1011 components in CLMD were detected, and 778 targets were identified through database screening. Key active ingredients included baicalin, luteolin, rhein, etc . A total of 472 targets were screened based on the degree values, among which there were 3 core target genes: PIK3R1, AKT1, and MAPK1. There were many enriched significant pathways, including PI3K/AKT and MAPK pathways. Baicalin binds well to the three core proteins PIK3R1, AKT1, and MAPK1. Baicalin inhibited corticosterone-induced depression-like neuronal injury through activating PI3K/AKT and MAPK/ERK pathways. Conclusion These results suggested that the antidepressant effects of CLMD could be achieved by its active ingredient baicalin, which targeted PIK3R1, AKT1, and MAPK1 to affect PI3K/AKT and MAPK/ERK pathways. This study provided a basis for further in-depth study on the therapeutic mechanism of CLMD in depression.

Pharmacokinetic profile and bioavailability assessment of rubusoside in mouse plasma utilizing UPLC-MS/MS analysis

Meiling Zhang,Siyu Zhuo,Fangmei Zhang,Jianshe Ma,Lufeng Hu,Xianqin Wang

The ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) technique was utilized to detect rubusoside in mouse plasma and evaluate its pharmacokinetic behavior following oral administration at doses of 10 mg/kg and 20 mg/kg, as well as intravenous administration at 10 mg/kg. Plasma samples were prepared using a protein precipitation method employing acetonitrile as the precipitating agent and isoscoparin as the internal standard (IS). Chromatographic separation was performed on a UPLC high strength silica (HSS) T3 column under gradient elution conditions, using acetonitrile and 0.1 % formic acid in water as the mobile phase system. The calibration curve for rubusoside demonstrated excellent linearity across a concentration range of 3.5 to 1120 ng/mL. Both intra-day and inter-day precision values remained below 7 %, with accuracy ranging from 92.3 % to 108.1 %. Matrix effects were consistently within the range of 107.1 % to 110.0 %, and the recovery rate exceeded 74.2 %. Following oral administration, the absolute bioavailability of rubusoside was determined to be 1.00 % and 0.98 % at doses of 10 mg/kg and 20 mg/kg, respectively. These findings confirm that the developed UPLC-MS/MS method effectively supports the pharmacokinetic evaluation of rubusoside

Network pharmacology and UPLC‐MS/MS‐based study of active ingredients in Jiu Wei decoction

Chen-Chen MAO,Qi HU,Yi-Ming NING,Da-Jin ZHANG,Yu-Kun GAO,Xiang-Yan XU,Yu-Dong WANG,Mei-Yun SHI,Qiang MA,Hong-Yu XUE

Background and objectives Jiu Wei decoction (JWD) is a traditional Chinese medicine (TCM) applied to control rheumatoid arthritis (RA). However, the active ingredients of JWD are still unclear. In this study, bioinformatics and ultra-high-performance liquid chromatography coupled with mass spectrometry (UPLC-MS/MS) were used to uncover the material basis and mechanism of JWD against RA. Materials and methods Ingredients and potential targets of herbs in JWD were collected from online databases. Then, these targets and RA related targets were overlapped, and the overlapped targets were subjected to further PPI (protein-protein interaction, PPI) analysis. Drug-component-target network and molecular docking were used to predict the potential active compounds. UPLC-MS/MS method was applied to detect and quantify these compounds in JWD alcohol extract. Results A total of 90 constituents and 520 targets were collected from JWD and 95 potential targets were overlapped by comparing these targets with RA-related targets. PPI network analysis indicated that TNF-α, IL-6, AKT1 and VEGFA were the main targets of JWD to RA, and the pharmacological mechanism of these targets might be related to anti-inflammation, pro-apoptosis, and inhibition of angiogenesis. Drug-component-target network and molecular docking results showed that quercetin , wogonin , baicalein , ferulic acid , kaempferol and berberine might be the potential compounds of JWD to RA. Through UPLC-MS/MS analysis, four compounds including quercetin, wogonin, baicalein, and berberine were detected and quantified. Moreover, among the four ingredients, quercetin, wogonin, berberine and baicalein accounted for 15.85%, 25.84%, 15.01%, and 43.3%, respectively. Conclusion Collectively, our study proved that bioinformatic analysis combined with UPLC-MS/MS is a promising method to explore the material basis of anti-RA in JWD and the method is also applicable to other TCMs. The effect of the active compounds still needs further experimental verification in the disease cell model.
