The protein corona on nano drug carriers is an important well-known biological issue that often induce biological incompatibility and screens the targeting molecules on the surfaces of carriers, therefore, the design of NPs with good protein corona-free property is highly desired and challenged. The natural polysaccharide has been demonstrated as one types of stealth materials after the functional group modification process, but the types and structures of their chains has never been considered. Here, we have designed five types of core-shell starch-coated poly (methyl methacrylate) nanoparticles and we found the starch coated NPs with low amylose content (<15%) could exhibit the excellent protein corona-free property without any modification and the starch with high amylose content coated NPs can also exhibit protein corona-free property after etherifying the surface of NPs to positive surface charge. Therefore, the combined impact of both low amylose content and positive surface charges by etherification modification of the starch can provide the excellent protein corona-free property for starch coated polymer NPs, that is very promising for highly efficient nano drug carries and marine coatings.

Pancreatic cancer is one of the most lethal malignancies in the world and remains one of the leading causes of cancer related death. For its treatment, a lot of investigations have dealt not only with individual chemotherapy by using polymeric carriers to deliver anticancer drugs, but also with individual gene therapy by using polymeric carriers to deliver nucleic acids such as small interfering RNA (siRNA) and plasmid DNA . However, relatively few studies have been focused on the co -delivery of gene and anticancer drug by multifunctional polymeric carriers for its synergistic therapy. In this work, a DPLL-functionalized amylose (ADP) was prepared by the click reaction between azidized amylose and propargyl focal point poly( l -lysine) dendrons, and then used to co-deliver plasmid pIRES2-EGFP-TNFα and curcumin for pancreatic cancer treatment. Due to the internal hydrophobic cavity of amylose component, ADP could load efficiently curcumin with anticancer activity and showed a sustained release behavior. Moreover, the curcumin-loaded ADP could form colloidally stable nanocomplexes with plasmid DNA in aqueous system due to the existence of cationic poly( l -lysine) dendrons and exhibited high gene transfection efficiency. The in vitro and in vivo tests confirmed the effectiveness of using ADP to co-deliver plasmid pIRES2-EGFP-TNFα and curcumin for synergistic therapy of pancreatic cancer.

Three-dimensional (3D)-printed intestinal stents based on thermoplastic starch (TPS)/polylactic acid (PLA) are a promising biodegradable alternative to conventional metal stents. However, the influence of the structure and conformation of starch on the properties of 3D-printed TPS/PLA intestinal stents requires clarification. This study evaluated the effects of the amylose/amylopectin ratio of starch on the molecular orientation, molecular interactions, crystallization behavior, microstructure, and thermodynamic, mechanical and hydrolysis performance of 3D-printed TPS/PLA intestinal stents. The molecular orientation of starch was determined during the preparation of the filaments for 3D printing. The 3D printing process promoted intermolecular interactions by further improving the molecular orientation, and enhanced the short-range order and crystallinity of the starch molecules. Decreasing the amylose/amylopectin ratio enhanced the short-range order of the starch molecules by increasing molecular orientation, which improved the circumferential compression performance of the 3D-printed intestinal stents but not the axial compression performance. However, the reduction in plasticized starch particles and improved PLA continuity caused by decreasing the amylose/amylopectin ratio contributed to improving the circumferential and axial compression performance as well as the anti-hydrolysis performance. These findings highlight the potential of starch to replace PLA as an inexpensive raw material for the development of high-performance biodegradable 3D-printed intestinal stents.

BACKGROUND The formation of starch–lipid complexes is of interest to food processing and human nutrition. Fatty acid (FA) structure is important for the formation and structure of starch–FA complexes. However, there is limited research regarding the complexing behavior between amylose and different kinds of FAs, as well as the relationship between fine structures and digestibility of the formed complexes. This study aimed to investigate the behavior, fine structure, and digestibility of complexes formed between high amylose maize starch (HMS) and FA having various chain lengths and unsaturation degrees. RESULTS Complexes containing different FA structures showed V 6III -type crystals. Complexes containing 18-carbon unsaturated FAs displayed significantly higher complexing index ( P < 0.05) than other complexes. Complexes containing 12-carbon FAs and 18-carbon FAs with one unsaturation degree showed a higher degree of structural order and resistant starch (RS) content than other complexes. The 12-carbon FAs exhibited a higher binding degree with helical cavity of amylose than other FAs. Additionally, 10-carbon and 18-carbon saturated FAs tended to combine with HMS outside amylose helices more than other FAs. Laser confocal micro-Raman imaging revealed that the physically embedded 10-carbon and 18-carbon saturated FAs showed heterogeneous distribution in complexes, and that the complexed 18-carbon FAs with one unsaturation degree exhibited homogeneous distribution. CONCLUSION The behavior, structural order and digestibility of complexes could be regulated by FA structure. The 12-carbon FAs and 18-carbon FAs with one unsaturation degree were more suitable for the production of HMS–FA complexes with higher structural order and RS content than other FAs. © 2022 Society of Chemical Industry.

O/W emulsions stabilized by polyphenol/amylose (AM) complexes with several polyphenol/AM mass ratios and different polyphenols (gallic acid (GA), epigallocatechin gallate (EGCG) and tannic acid (TA)) were prepared by a high-intensity ultrasound emulsification technique. The effect of the pyrogallol group number of polyphenols and the mass ratio of polyphenols/AM on polyphenol/AM complexes and emulsions was studied. The soluble and/or insoluble complexes gradually formed upon adding polyphenols into the AM system. However, insoluble complexes were not formed in the GA/AM systems because GA has only one pyrogallol group. In addition, the hydrophobicity of AM could also be improved by forming polyphenol/AM complexes. The emulsion size decreased with increasing pyrogallol group number on the polyphenol molecules at a fixed ratio, and the size could also be controlled by the polyphenol/AM ratio. Moreover, all emulsions presented various degrees of creaming, which was restrained by decreasing emulsion size or the formation of a thick complex network. The complex network was enhanced by increasing the ratio or pyrogallol group number on the polyphenol molecules, which was because the increasing number of complexes was adsorbed onto the interface. Altogether, compared to GA/AM and EGCG/AM, the TA/AM complex emulsifier had the best hydrophobicity and emulsifying properties, and the TA/AM emulsion had the best emulsion stability.

Our aim was to investigate the preparation of self-assembled garlic essential oil-amylose inclusion complexes (SGAs) using garlic essential oil (GEO) and corn starch (CS), and evaluated their release properties. SGAs were fabricated by pre-gelatinization coupling with high-speed shear at different GEO-CS mass ratios. When the mass ratio of GEO to pre-gelatinized corn starch was set at 15 % (SGA-15 %), with a fixed shear rate of 9000 rpm and a shear time of 30 min, the allicin content was 0.573 ± 0.023 mg/g. X-ray diffraction (XRD) results revealed a starch V-type crystalline structure in SGAs with peaks at 13.0°, 18.0°, and 20.0° (2θ). Fourier Transform Infrared (FTIR) spectra of SGAs displayed a shift in the characteristic peak of diallyl trisulfide from 987.51 cm −1 to 991.45 cm −1 . Scanning electron microscope (SEM) images revealed that SGAs exhibited lamellar structures covered with small granules. SGAs exhibited higher residual mass (approximately 12 %) than other samples. The resistant starch content of SGAs increased from 10.1 % to 18.4 % as GEO contents varied from 5 % to 15 %. In vitro digestion tests showed that about 53.21 % of allicin remained in SGA-15 % after 8 h. Therefore, this dual treatment can be a new method for fabricating controlled-release inclusion complexes of guest molecules.

Nuciferine exhibits properties such as reducing blood sugar and fat, however, it is hindered by its poor water solubility and low bioavailability. Pickering emulsions can efficiently encapsulate, protect and deliver active ingredients. In recent years, the use of biologically derived natural materials as emulsifiers to construct Pickering emulsions has become a research hotspot. This research utilized an enzymatic hydrolysis technique to produce short amylose. Subsequently, a ternary composite of short amylose (DBS), zein, and pectin (PEC) was formulated to stabilize Pickering emulsion, with the incorporation of nuciferine aiming to enhance the performance of lotus leaves in terms of both stability and bioavailability. The study revealed that varying amounts of DBS addition had a significant impact on the micromorphological structure and functional properties of DBS-Zein-PEC ternary composite particles. Specifically, the addition of 0.4 g of DBS led to a notable reduction in particle size to 735.2 nm and Zeta potential to −29.6 mV, creating a three-dimensional network with a closely packed lamellar structure. Optimal process conditions for preparing Pickering emulsion included a 3-minute homogenization time, rotation speed of 15000 rpm, and 5 % ternary composite particle addition. Under these conditions, O/W Pickering emulsion was successfully prepared, achieving a 90.5 % encapsulation rate for nuciferine. The resulting emulsion exhibited a minimum particle size of 4.09 μm, displayed good storage stability, resistance to salt ions and pH variations, viscous fluid characteristics, tolerance to oral and gastric environments, and slow release of nuciferine in the small intestine, thereby enhancing its bioavailability. These findings offer insights into the loading and delivery of nuciferine and serve as a technical guide for developing highly stable emulsion gel systems.

Curcumin-loaded DES was successfully encapsulated in hydrogel beads. • Water distribution and microstructure varied with HAMS content. • Higher-HAMS formulations showed slower intestinal-stage curcumin release. • DES-hydrogel integration provides a simple strategy for hydrophobic delivery. Curcumin is a hydrophobic bioactive compound with limited oral applicability due to its poor water solubility and low stability. In this study, high-amylose maize starch (HAMS)-containing hydrogel beads were developed for the encapsulation of curcumin-loaded natural deep eutectic solvent (CLDES). Sodium alginate (SA) formed an ionically crosslinked hydrogel matrix through Ca 2+ -induced gelation, while HAMS contributed to additional physical associations within the hydrogel system. The hydrogel beads showed HAMS-content-dependent differences in their structural properties. The average bead diameter increased from 2.41 ± 0.02 mm (H0) to 3.14 ± 0.02 mm (H25). SEM showed a progressively denser internal structure with increasing HAMS content. LF-NMR analysis revealed differences in water mobility and distribution among formulations with different HAMS contents. In addition, the maximum thermal degradation temperature increased from 248.3 °C (H0) to 313.7 °C (H35), indicating formulation-dependent differences in thermal degradation behavior at higher HAMS levels. The swelling ratio and water holding capacity decreased with increasing HAMS content, while the loading capacity gradually stabilized at higher HAMS levels. Under simplified simulated gastrointestinal conditions, all hydrogel beads exhibited limited curcumin release in simulated gastric fluid (below 3% within 2 h), followed by markedly increased release in simulated intestinal fluid. Release kinetic analysis suggested that the release behavior was associated with combined effects of network relaxation, swelling, and structural disintegration rather than simple Fickian diffusion. Overall, the HAMS-containing CLDES hydrogel beads developed in this study provide a facile strategy for the encapsulation and controlled release of hydrophobic bioactive compounds. The stability of hydrophobic bioactive compound indole-3-carbinol (I3C) is a challenge for application. In this work, Pickering emulsions were prepared to encapsulate I3C. As the emulsifier, high amylose corn starch was pretreated by acid hydrolysis, afterwards modified by different concentrations of octenyl succinic anhydride (OSA), and their emulsions were evaluated. The XRD, SEM and FTIR results indicated the successful modification. ζ-potential, mean droplet size and emulsification index (EI) of the emulsions confirmed that modified starch with a higher degree of substitution (DS) was more effective for enhancing the storage stability. The results of encapsulation efficiency (EE) and retention degree of I3C after 14 d also proved the assumption. Moreover, the Pickering emulsions protected I3C against ultraviolet light and achieved controlled release in vitro . The food-grade Pickering emulsion loading I3C is promising to be used as a nutrient or dietary supplement for food applications.

Granule-associated surface lipids (GASLs) and internal lipids showed different lipid–amylose relationships, contents, and distributions, suggesting their differing biological origins and functions, among waxy, normal, and high-amylose rice starch. The GASL content mainly depended on the pore size, while internal lipids regulated starch biosynthesis, as indicated by correlations of internal lipids with the chain length distribution of amylopectin and amylose content. Of the 1346 lipids detected, 628, 562, and 408 differentially expressed lipids were observed between normal–waxy, high-amylose–waxy, and normal–high-amylose starch, respectively. After the removal of GASLs, the higher lysophospholipid content induced greater decreases in the peak and breakdown viscosity and swelling power, while the highest digestibility increase was found with the highest triacylglycerol content. Thus, different GASL compositions led to different digestibility, swelling, and pasting outcomes. This study sheds new light on the mechanism of the role of GASLs in the structure and properties of starch, as well as in potential modifications and amyloplast membrane development.

In this study, active packaging films were developed by incorporating nano TiO 2 and pomegranate peel extract (PPE) into high-amylose corn starch (HCS)/konjac glucomannan (KG) matrix for the first time. The aim of present work was to investigate the influence of nano TiO 2 and PPE on physicochemical and functional properties of HCS/KG-based films. The films were characterized by XRD, FT-IR and SEM, and the tensile, antibacterial and antioxidant properties were evaluated. The results showed that the crystallinity of the composite films was increased and the microstructure was more uniform and dense after adding TiO 2 and PPE, and the intermolecular hydrogen bonds were formed between TiO 2 , PPE, and film matrix. Incorporation of TiO 2 and PPE significantly improved tensile properties and decreased water solubility and water vapor permeability of the composite films (p < 0.05). HCS/KG films incorporated with TiO 2 and PPE presented remarkable antibacterial activity against Escherichia coli and Staphylococcus aureus , and exhibited strong antioxidant activity due to the polyphenol compounds in PPE. All films properties not only changed with the content of TiO 2 and PPE, but also improved synergistically when they were added together. The developed composite films were used as coating for the preservation of Agaricus bisporus , and the weight, firmness and total soluble solids of A. bisporus were significantly maintained and browning was effectively inhibited during storage. Therefore, HCS/KG composite film/coating containing TiO 2 and PPE has great potential as an attractive commercialization technology to ensure the quality and extend the shelf life of foods.

Starch is an important raw material in the food industry, and the ratios of amylose to amylopectin within it is closely related to the quality of starch products. Therefore, the development of rapid detection methods for the amylose/amylopectin ratios in starch is a practical demand to meet industrial development. Consequently, the development of a rapid, sensitive, and portable detection method is of great significance. In this study, an electrochemical detection method for the amylose-to-amylopectin ratios was established by utilizing alcohol dehydrogenase (ADH) and pullulanase to fabricate an ADH@pullulanase-AuNPs biosensor. Specifically, protein-metal nanoflowers were synthesized by regulating the interactions of Cu 2+ and PO 4 3− with the enzymes to achieve the spatially localized immobilization of ADH and pullulanase; meanwhile, AuNPs were modified to enhance electrical conductivity. Additionally, the applicability of this method to rice products was investigated. The experimental results showed that the ADH@pullulanase cluster exhibited a porous flower-like structure, good enzymatic activity, and excellent electrochemical performance. Under optimal conditions, this method exhibited a wide linear range (1 × 10 −6 –1 × 10 −4 mol/L) for the amylose/amylopectin ratios, a low limit of detection (0.5 × 10 −6 mol/L), and a short detection time (< 10 min). When applied to the detection of rice products, the average relative deviation between it and the iodine colorimetric method was 6.9%. The successful development of this method provides a technical reference and theoretical basis for the on-site rapid and sensitive detection of the amylose/amylopectin ratios.
