Glutathione coated gold and silver nanoclusters (GSH-Au/AgNCs) were synthesized by one-pot reduction methods and are found to be viable fluorescent nanoprobes for cysteine (Cys) and arginine (Arg), with good selectivity over other amino acids. The GSH-Au/AgNCs have two emissions at 616 nm and 412 nm when excited at 360 nm. With the increased concentration of Cys, the ratio of the emission intensities (I 616 /I 412 ) linearly decreases with Cys in concentration ranging from 0.05 to 10 μM and from 10 to 50 μM, respectively. With increased concentrations of Arg, the ratio of I 616 /I 412 linearly decreases with Arg concentration ranging from 0 to 50 μM and from 50 to 100 μM, respectively. The probe was applied to the determination of Cys and Arg in spiked samples of serum and urine where it gave good recoveries.

In this work, the rapid unfolding kinetics of pancreas α-amylase (PPA) induced by l -arginine and the interaction mechanism were investigated. The unfolding followed a first-level reaction kinetics equation, without intermediates. l -arginine interacted with PPA though diffusion-controlled process rather than complexion. The interaction between l -arginine and PPA resulted in a pronounced decrease in β-sheet and a significant increase in random coil, and thereby the enzyme activity decreased. However, the unfolding of PPA could be compensated and the second structure change could be recovered to some extent by the macromolecular crowded medium of Pluronics. Further insight into the mechanism disclosed that the broken H-bond network of water may contribute to PPA unfolding. This work provides a new perspective on the interaction of l -arginine with digestive enzyme. The unfolding mechanism of enzymes by may help to understand the effects of other structurally similar drugs, which is of concern in food-drug interactions.

Antibacterial waterborne polyurethane (WPU) is attracting increasing interest in a wide range of industrial applications due to its versatility and eco-friendliness. However, single system antimicrobial WPU often does not meet the requirements of highly antimicrobial coating fields. Therefore, WPU was synergistically modified by L-arginine (L-Arg) and quaternary ammonium salts (QAs) in this study. They were introduced into the molecular structure of WPU by in-situ emulsion polymerization to form the composite antibacterial system (WRPUs). The results showed that the composite systems had excellent antibacterial activity against both Escherichia coli and Staphylococcus aureus compared to single system. When the content of both L-Arg and QAs was 7%, the antibacterial effect of WRPU7 was optimal, from the sum ( E. coli : 8.3 mm + 8.3 mm = 16.6 mm and S. aureus : 9.8 mm + 9.5 mm = 19.3 mm ) of the inhibition circles of L-Arg and QAs to 21.4 and 22.6 mm, respectively, net efficiency increase of 29% and 17%, fully reflecting the synergistic effect of “ 1 + 1 > 2 . ” Taken together, the results presented in this study confirm the significance of L-Arg and QAs as synergistic antimicrobial building blocks. In this way, the antimicrobial performance of WPU can be significantly improved and better biocompatibility can be exhibited, thus broadening the application of WPU in highly antibacterial fields.

Neurogenesis in the developing and adult brain is intimately linked to remodeling of cellular metabolism. However, it is still unclear how distinct metabolic programs and energy sources govern neural stem cell (NSC) behavior and subsequent neuronal differentiation. Here, we found that adult mice lacking the mitochondrial urea metabolism enzyme, Arginase-II (Arg-II), exhibited NSC overactivation, thereby leading to accelerated NSC pool depletion and decreased hippocampal neurogenesis over time. Mechanistically, Arg-II deficiency resulted in elevated L-arginine levels and induction of a metabolic shift from glycolysis to oxidative phosphorylation (OXPHOS) caused by impaired attachment of hexokinase-I to mitochondria. Notably, selective inhibition of OXPHOS ameliorated NSC overactivation and restored abnormal neurogenesis in Arg-II deficient mice. Therefore, Arg-II-mediated intracellular L-arginine homeostasis directly influences the metabolic fitness of neural stem cells that is essential to maintain neurogenesis with age.

Arginine is considered as a biomarker of cystinuria and other diseases, and thus, it is of urgency to develop a simple and rapid method with high sensitivity and selectivity for arginine detection to meet the demand of on-site analysis and bedside diagnosis. In this work, a lanthanide metal–organic framework, La(TATB), was prepared using a triazine-based planar ligand, 4,4′,4″-s-triazine-2,4,6-triyltribenzoate (H3TATB), and lanthanide ion (La3+). La(TATB) can be used as a highly photosensitive agent to activate molecular oxygen to 1O2 to achieve efficient photosensitive oxidation of arginine accompanied by strong blue fluorescence emission under 302 nm UV irradiation. Due to the porous structure and high specific surface area of La(TATB), short-life 1O2 can effectively approach and react with amino acid substrate molecules, thus leading to higher sensitivity than other systems. Therefore, the “turn-on” fluorescence sensing of trace arginine can be realized, with a measured linear response range of 10–20,000 nM and a limit of detection as low as 7 nM. This method can be used for the detection of trace arginine in urine, which is conducive to the bedside diagnosis and rapid screening of cystinuria and other diseases. The proposed method not only expands the application scope of Ln-MOFs but also provides a new construction strategy for “turn-on” luminescence sensors.

In the field of drug delivery, there is growing concern over the self-assembly of fatty acids, since their structures and properties are similar to liposomes, which are characteristically nontoxic for humans, possess good biocompatibility and biodegradation, and are without immunogenicity. Changes in pH, temperature, concentration, molecular structure of the amphiphilic molecule, head group type and hydrocarbon chain length have significant effects on the self-assembled structures; however, the degree of unsaturation can also influence the self-assembled structures of fatty acids. The aggregation behaviors of the mixtures of arginine and three kinds of unsaturated fatty acids (UFAs, oleic acid, linoleic acid and linolenic acid) in aqueous solutions have been investigated. Phase transition from transparent micelles (L1 phase) to birefringent bilayer structures (Lα phase) occurred with increasing amounts of UFAs. However, the Lα phase was different for macro and micro structures. Oleic acid with only one double bond was regarded as less flexible and curved, but with higher stacking, leading to a gradual phase transition process from the vesicle phase to the stacked lamellar phase in the Lα phase region, which could be proved by cryogenic transmission electron microscopy (cryo-TEM) and polarizing microscopy observations combined with 2H-nuclear magnetic resonance (2H NMR) and rheological properties. The turbid vesicle phase with weak viscoelastic properties were only observed in linoleic acid and linolenic acid systems because UFAs with more than one double bond are easier to bend, indicating that highly curved vesicles are likely to form. Through the analysis of the proposed mechanism and FT-IR spectra, the synergistic effects of non-covalent interactions, including hydrogen bonding, electrostatic interaction, and hydrophobicity, were considered to be responsible for the aggregation behaviors. Finally, vesicles displayed their potential in encapsulating water-soluble model drugs such as calcein. The results show that drug-loaded vesicles could play a role in the potential applications in drug sustained delivery systems.

Arginine (Arg) has been recognized as a crucial biomarker for cystinuria, with its concentration levels markedly influencing human health. In this study, Eu-TATB were synthesized via a solvothermal method using 4,4′,4″- s -triazine-2,4,6-triyl-tribenzoic acid (H₃TATB) as the organic ligand and Eu ions. Eu-TATB, benefiting from its intrinsic photosensitive characteristics and the characteristic Eu 3+ emission at 618 nm, was utilized to construct a novel ratiometric fluorescent sensor. By harnessing the photosensitization of Eu-TATB under ultraviolet light irradiation, significant oxidation of arginine was catalyzed, leading to the emergence of a new blue fluorescence peak at 430 nm. The intensity of this blue fluorescence increased proportionally with the arginine concentration, while the red emission at 618 nm gradually decreased. Utilizing the cross response of this dual-emission intensity, Eu-TATB exhibits ratiometric fluorescence sensing property for arginine, with a broad linear range of 0.1–2 mM and a low limit of detection (LOD) of 3.22 μM, and validated the practicality of the proposed sensor through the quantitative detection of arginine in human serum. Notably, the variable luminescence colors that are visible to naked eye endow the feasibility of Eu-TATB for in situ visualization analysis of arginine.

Apoptosis at bone defects drives an anti-osteogenic oxidative stress environment • IL-4 drives a pro-reparative macrophage phenotype and upregulates the Arg1 pathway • Exogenous Arg fuels efferocytosis and secretion of pro-regenerative factors • Co-delivery of IL-4 and Arg via injectable hydrogel accelerates bone regeneration Summary Host bone microenvironment is a critical factor for successful bone regeneration during implantation. In this work, we identified a spatiotemporal mismatch at drilling-induced injury sites where initial apoptosis outpaces macrophage recruitment, leading to apoptotic cell accumulation and oxidative stress that impairs osteogenic differentiation. To mitigate this phenomenon, we developed a synergistic strategy co-delivering interleukin 4 (IL-4) and arginine (Arg) to reprogram the microenvironment. Mechanistically, IL-4 recruits and polarizes macrophages toward a pro-reparative M2 phenotype, while Arg fuels the arginase-1 metabolic pathway to sustain continual efferocytosis and secretion of pro-regenerative cytokines. In a mouse bone injury model, transcriptomic and histological analyses confirmed that this strategy accelerates apoptotic clearance and establishes an M2-dominant environment, significantly enhancing early bone regeneration. Altogether, this work proposes a novel therapeutic paradigm targeting macrophage efferocytosis to resolve early microenvironmental imbalances, providing a promising therapeutic approach to promote bone defect healing and offering novel insights into optimizing host microenvironments for osseointegration following implantation. By self-assembly of MnCl 2 and arginine under alkaline conditions, ultra-small MnArg nanoparticles were successfully constructed as oxidase (OXD) mimics for intelligent detection of the Ginkgo toxin 4′- O -methylpyridoxal (MPN). The obtained MnArg nanozymes possessed excellent OXD-like activity and thermal stability. Based on the inhibitory effect of MPN for the catalytic activity of MnArg, this system was utilized for the colorimetric sensing of MPN with a low limit of detection (LOD) of 2.16 µg mL −1 . The detection system exhibited good selectivity against other potential interferents. FTIR data showed that the presence of MPN binds with MnArg and shields the active sites, thereby interfering with the oxidase-like activity. Combined with a smartphone and the ColorMax software, this nanozyme-based intelligent detection platform could effectively detect MPN with a LOD of 2.1 µg mL −1 . Our MnArg nanozyme–based system was applied to detect real ginkgo nut samples with recoveries of 92.4–108.7%, and the relative standard deviations were less than 0.7%. This work may promote the development of novel nanozymes and expand their applications in the field of food safety detection. Graphical Abstract

Given that excessive CO 2 induces reactive oxygen species (ROS) which inhibit photosynthesis and microalgal survival, this study explores how exogenous arginine enhances stress resilience and biomass yield of Nannochloropsis oceanica under high CO 2 stress. Arginine promoted NO synthesis, enhancing antioxidant enzyme activities to combat ROS damage. Transcriptomic analysis revealed that key enzymes in the Calvin cycle (PRK, GAPDH, FBPase and SBPase) and TCA cycle (CS, IDH, SCS and MDH) were significantly up-regulated, supporting carbon fixation and energy metabolism. Arginine also alleviated cell necrosis by extending the G1 phase, promoting DNA replication and protein synthesis, which increased the proportion of viable cells from 47 % to 84 %. Furthermore, the synthesis of chlorophyll a and carotenoid were enhanced, leading to 46 % increase in biomass yield. The results highlighted arginine as a promising metabolic regulator for enhancing stress tolerance and carbon fixation efficiency of N. oceanica under high CO 2 conditions.

To create fluorescent sensors for Cr (VI) and l-arginine detection, blue-emitting carbon dots (CDs, named as M-CDs) were prepared via a solvothermal synthesis by using m-phenylenediamine. The introduction of sulfuric acid changed the blue-emitting of CDs into green-emitting (named as S-CDs), in which the photoluminescence (PL) peak wavelengths of samples M-CDs and S-CDs were 450 and 540 nm, while their PL efficiencies were 21.7 and 84.2 % respectively. Sulfuric acid resulted in the average fluorescent lifetime of the CDs was prolonged into 8.19 from 4.65 ns. The photo and thermal stable tests demonstrated that theses CDs revealed excellent stability. Meanwhile, CDs flexible films with bright blue- and green-emitting were prepared utilizing the favorable solubility of CDs in polar solvents. Mono-dispersed CDs as luminescent materials demonstrated it promising prospects in luminescent devices field. Furthermore, M-CDs exhibited excellent selectivity towards Cr(VI) and l-arginine. The detected limit (0.86 μM for Cr(VI) and 0.43 μM for l-arginine) was determined by assessing the response of M-CDs to various concentrations of Cr(VI) ion and l-arginine solutions. These bright CDs revealed great potential for accurate detection of Cr(VI) and l-arginine.

A novel smartphone-assisted fluorescent sensor based on europium/zirconium metal-organic framework (Eu 0.5 /Zr 0.5 -MOF) was developed for the fast and sensitive determination of doxycycline (DOX) and L-arginine (Arg). After the addition of DOX, the fluorescence of Eu 0.5 /Zr 0.5 -MOF was quenched owing to the inner filter effect (IFE). When Arg was introduced into the Eu 0.5 /Zr 0.5 -MOF@DOX complex system, the fluorescence was recovered because the interaction between Arg and Eu 0.5 /Zr 0.5 -MOF@DOX weakened the IFE. Moreover, the Eu 0.5 /Zr 0.5 -MOF produced continuous fluorescence color changes for the visual measurement of DOX and Arg. The fluorescent probe for DOX and Arg offered broad linear ranges of 0.05–80 and 0.1–60 μg/mL, respectively, with detection limits as low as 2.07 and 67.5 ng/mL. The proposed method was successfully applied to monitor DOX in eggs and Arg in human serum. This work provides a powerful platform for the real-time and visual analysis of DOX and Arg in food and biological samples.
