Zein-fucoidan nanoparticles (ZFNPs) as an excellent colloidal delivery system for bioactive compounds, such as curcumin (CUR), show a great application potential in the food industry. Herein, various amount of ZFNPs, ZFNPs loaded with CUR (CZFNPs) and free CUR were added into golden threadfin bream ( Nemipterus virgatus ) surimi to explore their impacts on the gel properties. Dynamic rheological tests showed that storage modulus (G') and loss modulus (G") of surimi gels with the additive amount of ZFNPs and CZFNPs under 0.8 % significantly increased and their loss tangent (tan δ) slightly reduced during thermal gelation, indicating stronger networks and better structural configuration. Puncture test and textural profile analysis confirmed the improvements in breaking force, hardness, chewiness and gumminess of surimi gels. Furthermore, 0.8 % of ZFNPs and CZFNPs induced conformational changes of surimi proteins from α-helix to β-sheet and random coil, and then facilitating the cross-link between proteins. Low-field nuclear magnetic resonance revealed that ZFNPs and CZFNPs promoted the redistribution of water in surimi gel networks, converting more free water to tightly bounded water, and thus increasing the water holding capacity. However, the addition of equivalent amount of CUR caused detrimental to surimi gel properties. In conclusion, ZFNPs and CZFNPs conferred better gel properties and nutritional characteristics to surimi gels, and this study provided a theoretical basis for the development of high-quality surimi products.

Fucoidan is widely applied in food and pharmaceutical industry for the promising bioactivities. Low-molecular weight hydrolyzed fucoidan has gained attention for its beneficial health effects. Here, the modulation on microbiome and metabolome features of fucoidan and its acidolyzed derivatives (HMAF, 1.5–20 kDa; LMAF, <1.5 kDa) were investigated through human fecal cultures. Fucose is the main monosaccharide component in fucoidan and LMAF, while HMAF contains abundant glucuronic acid. LMAF fermentation resulted in the highest production of short-chain fatty acids, with acetate and propionate reaching maximum levels of 13.46 mmol/L and 11.57 mmol/L, respectively. Conversely, HMAF exhibited a maximum butyrate production of 9.28 mmol/L. Both fucoidan and acidolyzed derivatives decreased the abundance of Escherichia-Shigella and Klebsiella in human fecal cultures. Fucoidan and HMAF prefer to improve the abundance of Bacteroides . However, LMAF showed positive influence on Bifidobacterium , Lactobacillus , and Megamonas . Untargeted metabolome indicated that fucoidan and its derivatives mainly altered the metabolic level of lipids, indole, and their derivatives, with fucoidan and HMAF promoting higher level of indole-3-propionic acid and indole-3-carboxaldehyde compared to LMAF. Considering the chemical structural differences, this study suggested that hydrolyzed fucoidan can provide potential therapeutic applications for targeted regulation of microbial communities.

Hydrogen bonding and hydrophobic interactions were the major forces involved in the OVA-FUC complex. • The formation of the OVA-FUC complex resulted in enhanced emulsifying properties compared to individual OVA or FUC. • The galangin-loaded emulsion based on the OVA-FUC complex possessed potent antioxidant activity. • Galangin-loaded emulsion potentiates the oxidative stress resistance of Caenorhabditis . The effective delivery of hydrophobic bioactive compounds remains a major challenge in functional food and nutraceutical applications because of their poor aqueous solubility, limited physicochemical stability, and insufficient bioavailability. In this study, ovalbumin-fucoidan (OVA-FUC) complexes were constructed as food-grade stabilizers to develop galangin (GAL)-loaded emulsions with improved stability and antioxidant efficacy. The interaction mechanism, emulsion stability, and antioxidant activity were evaluated using structural characterization, interfacial analysis, storage tests, chemical antioxidant assays, and an ethanol-induced oxidative stress model in Caenorhabditis elegans . Results showed that OVA and FUC formed complexes mainly through hydrogen bonding and hydrophobic interactions. The 1-1 OVA-FUC complex exhibited the most favorable interfacial behavior and produced emulsions with superior storage stability. The GAL-loaded emulsion showed strong ABTS radical-scavenging activity (74.33 ± 7.60%) and effectively reduced ROS, lipofuscin, and MDA levels while increasing glutathione content and improving locomotor activity in C. elegans. These findings reveal that controlled OVA-FUC interactions can optimize interfacial assembly and emulsion stability, offering a practical strategy to improve the functional delivery of hydrophobic antioxidants in food systems. Fucoxanthin (FX), a marine-origin carotenoid, possesses various physiological activities. However, FX has instability and low water solubility. Encapsulation using nanoparticles effectively addresses these challenges. Nanoparticles loaded with FX were fabricated using a pH-driven method, with pea protein isolate (PPI) and fucoidan (FUC) serving as the raw materials. The optimal nanoparticles were prepared at pH = 7.0 with a PPI:FUC = 1:3, yielding a particle size of 166.60 ± 0.55 nm and a zeta potential of −40.88 ± 0.68 mV. The formation of FX@PPI/FUC nanoparticles were primarily driven by hydrogen bonding and hydrophobic interactions. Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and fluorescence spectroscopy were used to research structure of nanoparticle and interaction during the formation. The FX@PPI/FUC nanoparticles demonstrated excellent thermal and pH stability in neutral and alkaline environments, effectively released FX and showcased antioxidant properties. Additionally, a W/O/W FX@PPI-FUC Pickering emulsion was formulated, containing 65 % of the oil phase, which exhibited a favorable particle size of 26.5 ± 0.28 μm and a zeta potential of −67.2 ± 0.94 mV. Furthermore, the FX@PPI-FUC Pickering emulsion demonstrated outstanding thermal and storage stability, indicating its potential for application in functional food.

Acute kidney injury (AKI) is a common and serious complication in clinical practice, especially when using chemotherapy drug cisplatin, which severely limits its anti-cancer efficacy. This study developed a novel nano delivery system (Fu-4-PBA/Po NPs) that combines fucoidan (Fu), endoplasmic reticulum stress (ERS) inhibitor 4-phenylbutyric acid (4-PBA), and natural antioxidant Polydatin (Po) to simultaneously alleviate cisplatin induced oxidative stress and ERS. The optimized Fu-4-PBA/Po NPs showed an average size of 102 ± 3.46 nm, a zeta potential of −16.5 ± 0.49 mV, and a high drug loading capacity of 10.34 ± 0.6 %. They exhibited excellent stability and a sustained release profile in vitro . In vitro results showed that treatment with Fu-4-PBA/Po NPs led to a marked increase in cell survival by 34.73 ± 3.54 percentage points compared to the model group, which had a survival rate of 45.97 ± 2.76 %. Furthermore, Fu-4-PBA/Po NPs effectively inhibited the accumulation of reactive oxygen species (ROS), mitochondrial membrane potential decline, and calcium ion release by targeting P-selectin and endoplasmic reticulum (ER) at the site of inflammation, and significantly alleviate cisplatin induced cell apoptosis. Mechanism studies showed that nanocomposites alleviate ERS by inhibiting the PERK-eIF2α-ATF4-CHOP pathway, while blocking the activation of the cGAS-STING pathway, thereby reducing DNA damage and inflammatory response. Pharmacokinetic studies showed that the peak concentration (Cmax) of the nanoparticles reached 2.2 times that of free Po, and the area under the curve (AUC) increased by 2 times. In animal studies, administration of Fu-4-PBA/Po NPs at a dose of 100 mg/kg notably ameliorated renal function, as indicated by reductions in serum creatinine (SCr) and blood urea nitrogen (BUN) levels by 84.3 % and 62.4 %, respectively, and also improved kidney histopathology. This study provided a dual targeted nano-delivery strategy for cisplatin induced AKI therapy, which has important clinical application potential.

Type 2 diabetes mellitus (T2DM) is frequently associated with liver injury. This study examines the therapeutic potential of Fucus vesiculosus -derived low-molecular-weight fucoidan (Fuc-S) in mitigating T2DM-related hepatic damage. In STZ/HFD-induced diabetic mice, Fuc-S treatment (100 or 200 mg/kg, 5 weeks) significantly improved glucose tolerance, lipid metabolism, and liver function, while reducing hepatic steatosis and serum ALT/AST levels. Fuc-S enhanced hepatic antioxidant defenses, increasing SOD, CAT, and GSH-Px activity while decreasing MDA levels. Gut microbiota analysis showed that Fuc-S promoted the growth of beneficial bacteria ( Bacteroides acidifaciens ) and elevated fecal short-chain fatty acids (SCFAs), such as acetate, propionate, and butyrate. Furthermore, Fuc-S reinforced intestinal barrier integrity by upregulating tight junction proteins (ZO-1 and occludin). These results indicate that Fuc-S alleviates T2DM-induced liver injury by modulating the gut microbiota-SCFA-liver axis, thereby reducing oxidative stress and inflammation. The study suggests Fuc-S as a promising dietary intervention for T2DM, acting through multi-target microbiota-metabolite interactions.

Curcumin (Cur) features versatile bioactivities but suffers from poor stability and high vulnerability to gastrointestinal degradation. To address these bottlenecks, an effective and facile strategy for constructing a sustainable hydrogel delivery system with pH-responsive function and controlled gastrointestinal release properties is reported. The hydrogel beads were developed through physical crosslinking between fucoidan (Fuc), carboxymethyl chitosan (CMCS), and the encapsulated bioactive compound Cur. Structural characterization confirmed that intermolecular hydrogen bonding between Fuc and CMCS played a pivotal role in the hydrogel network formation. Cur was efficiently encapsulated within the hydrogel beads, functioning as an additional crosslinking agent that increased the crosslinking density and reduced the equilibrium water content of the system. Notably, the hydrogel beads protected Cur via non-covalent interactions and physical barriers, enhancing its thermal and UV stabilities. Furthermore, the hydrogel beads exhibited pronounced pH-responsive swelling and release behaviors, showing minimal swelling and limited release under acidic gastric conditions, in contrast to sharp swelling and near-complete release in alkaline intestinal environments, attributed to the pH-dependent protonation states of polysaccharides' functional groups altering intermolecular interactions. Importantly, the encapsulated Cur retained more than 90% antioxidant capacity following simulated gastrointestinal digestion. This crosslinker-free and food-grade system provides a safe delivery platform for labile polyphenols, showing considerable potential in functional foods and dietary supplements.

Enterococcus faecalis ( E. faecalis ) biofilm-associated persistent endodontic infections (PEIs) are one of the most common tooth lesions, causing chronic periapical periodontitis, root resorption, and even tooth loss. Clinical root canal disinfectants have the risk of damaging soft tissues (e.g., mucosa and tongue) and teeth in the oral cavity, unsatisfactory to the therapy of PEIs. Nanomaterials with remarkable antibacterial properties and good biocompatibility have been developed as a promising strategy for removing pathogenic bacteria and related biofilm. Herein, carbon dots (CDs) derived from fucoidan (FD) are prepared through a one-pot hydrothermal method for the treatment of PEIs. The prepared FDCDs (7.15 nm) with sulfate groups and fluorescence property are well dispersed and stable in water. Further, it is found that in vitro FDCDs display excellent inhibiting effects on E. faecalis and its biofilm by inducing the formation of intracellular and extracellular reactive oxygen species and altering bacterial permeability. Importantly, the FDCDs penetrated the root canals and dentinal tubules, removing located E. faecalis biofilm. Moreover, the cellular assays show that the developed FDCDs have satisfactory cytocompatibility and promote macrophage recruitment. Thus, the developed FDCDs hold great potential for the management of PEIs. Graphical Abstract

Previous studies have indicated that fucoidan could resist high-fat-diet (HFD)-induced obesity by modulating the composition of gut microbiota. However, the outcome of dietary intervention may differ between individuals due to large inter-individual variability in gut microbiota. Thus, the present study aimed to investigate the possible discrepancy of the anti-obesity effects of fucoidan supplementation in HFD-fed mice models with different gut microbiota communities. In the present study, the anti-obesity effects of fucoidan isolated from Laminaria japonica (FucLj) on normal mice and microbiota-altered mice treated with penicillin or metronidazole were compared and investigated. The 16S rRNA sequencing revealed the differences of gut microbiota among penicillin-treated, metronidazole-treated and normal groups, and mice treated with penicillin were characterized by greater relative abundance of the phylum Bacteroidetes and the families Muribaculaceae and Bacteroidaceae. Furthermore, FucLj ameliorated HFD-induced body weight gain, fat accumulation, serum lipid profiles, insulin resistance, hepatic steatosis and adipocyte hypertrophy in penicillin-treated and untreated mice, while no effects were observed in metronidazole-treated mice. Overall, mice with different initial gut microbiota responded differently to FucLj supplementation on a high-fat diet, and metronidazole-sensitive gut bacteria negatively correlated with obesity symptoms and were required for the anti-obesity effects of FucLj. Moreover, the anti-obesity effects were not dependent on the utilization of FucLj by gut microbiota to produce SCFAs. These findings indicate that evaluation of the gut microbiota structure before dietary interventions is helpful for enhancing the beneficial outcomes of dietary fiber supplementation and provide a rationale for the further application of dietary fucoidan in a personalized way.

Polyvinyl alcohol (PVA)/fucoidan (FUC) blend nanofibers were systematically fabricated to co-encapsulate probiotic Lactiplantibacillus plantarum 69–2 (LP69–2) and four kinds of polyphenols by electrospinning for the first time. Scanning electron microscopy showed that some areas of PVA/FUC nanofibers encapsulated with LP69–2 were locally broadened. Attenuated total reflectance-Fourier transform infrared spectroscopy and X-ray diffraction suggested that LP69–2 and polyphenol were successfully encapsulated in PVA/FUC electrospun nanofibers. Thermogravimetric analysis revealed that the addition of LP69–2 and polyphenol enhanced the thermal stability of nanofibers. Moreover, the incorporation of FUC and polyphenol significantly increased the ABTS + and DPPH radical scavenging ability of PVA nanofibers ( P < 0.05). Notably, PVA/FUC/LP69–2/DMY nanofibers displayed the highest DPPH radical scavenging ability. After 21 d, these nanofibers loaded with polyphenols could maintain viability of LP69–2 over 7 lg CFU/g at 4 °C and the viability of LP69–2 in PVA/FUC/DMY nanofibers was the highest. Overall, the co -encapsulation of probiotic and polyphenol within PVA/FUC electrospun nanofibers increased the viability of probiotics and enhanced antioxidant activity of nanofibers. This study provided unique insights for protecting probiotics and developing novel functional foods with higher probiotics.

Using of low-fat emulsion gels stabilized by quinoa protein (QP) for 3D food printing was limited by their defective rheological properties. The study was to explore the feasibility of using fucoidan (FU) to improve the printability and curcumin encapsulation stability of QP emulsion gel. The gels with 0.5–0.75 % FU incorporated were proved to be the most promising ink. Rheological analysis confirmed their great printing potential from the three-stages simulated 3D printing process. The incorporation of FU into emulsions induced more QP molecules adsorbed at the oil-water interface and smaller droplets size. When gel was induced by gluconate δ-lactone, these electrostatic interactions transformed into electrostatic attraction, resulting in a denser gel network. Adding FU to the gel improved its textural properties. Moreover, the gels with FU incorporated exerted better curcumin storage stability. This study provides a simple way to develop and improve the edible 3D printing inks for future food manufacturing.

Introduction Quercetin (Que), a physiologically versatile flavonoid, faces application limitations in food and pharmaceuticals due to poor aqueous solubility and stability. Method To address this, we developed quercetin-loaded zein-sodium caseinate-fucoidan (Que-ZE-SC-FD) ternary nanoparticles using a green, pH-driven approach. Results and discussion The Que-ZE-SC-FD nanoparticle exhibited a spherical morphology stabilized by hydrogen bonding, electrostatic, and hydrophobic interactions, with a mean diameter of 137.8 ± 11.6 nm, PDI of 0.38 ± 0.04, z-potential of 34.9 ± 0.6mV, and high quercetin loading efficiency (92.8% ± 1.1%). Crucially, SC and FD demonstrated synergistic stabilization effects. The Que-ZE-SC-FD nanoparticle exhibited a mean particle size of 150.8 ± 0.6 nm at a pH of 8.0, and solution remained clear and transparent with no observable sediment. Under a NaCl concentration of 3.0 mol/L, the particle size decreased to 127.8 ± 4.5 nm. Upon heating at 80°C for 2 h, the particle size further reduced to 121.3 ± 1.2 nm, with a PDI of 0.34 ± 0.02. After 28 days of storage, the particle size decreased to 125.1 ± 1.9 nm, while the PDI decreased slightly to 0.32 ± 0.01 and the zeta potential increased to 31.6 ± 1.5mV, collectively indicating excellent stability. Under simulated gastrointestinal conditions, the Que release from ZE-SC-FD nanoparticles was only 22.2 ± 0.5% in gastric fluid; however, a significantly higher release rate of 75.0 ± 0.5% was achieved in intestinal fluid. These results demonstrate that ZE-SC-FD nanoparticles serve as a robust nanocarrier system for encapsulating, protecting, and delivering quercetin.
