Inulin dietary supplement is conventionally beneficial to gut health and can potentially prevent food allergy (FA). This study aimed to determine how dietary inulin interventions at different doses affect the OVA-induced FA in a BALB/c mouse model. Although the middle dose of inulin (50 mg per mouse) showed the best therapeutic effect on FA, high-inulin supplementation (80 mg per mouse) provoked severe allergic and intestinal inflammatory responses, which were characterized by elevated serum allergic inflammation-related factor levels, dysfunctional gut barrier, unbalanced luminal pH value, decrease in intestinal antioxidant capacity, and disordered gut microecology. Moreover, profiling of SCFAs indicated that the high-inulin-induced excess accumulation of SCFAs in the colon was responsible for the gut immune disorders. Spearman correlation analysis unraveled that the featured bacterial taxa in the high-inulin-treated mice were Ruminococcaceae and Bifidobacterium , of which the relative abundance was negatively correlated with expression of tight junction proteins and improvement of T cell homeostasis, and positively correlated with levels of allergic inflammation-related indexes. Our work suggested that high-inulin dietary supplementation can be detrimental to allergic individuals and highlighted the importance for personalized use of inulin-type dietary supplements to safely improve human health.

This study aimed to investigate the oxidation stability, 3D printability, and β-carotene delivery performance of high internal phase Pickering emulsions (HIPPEs) stabilized by quinoa protein-inulin (QP-Inu) conjugates and chlorogenic acid (CA) ternary complex. Compared with QP-Inu, the ternary complex (QP-Inu/CA non-covalent complex and QP-Inu-CA covalent complex) showed a rougher surface, a higher three-phase angle close to 90°, lower interfacial tension, and an increased absorption rate at the oil-water interface. From particle size and confocal laser scanning microscopy (CLSM) analysis, HIPPEs stabilized by the ternary complex exhibited a compact gel network structure with more tightly packed smaller droplets. This microstructure contributed to their improved oxidation stability from generation of lower levels of primary and secondary lipid oxidation products, enhanced 3D printing capabilities from higher viscosity and elasticity, superior thixotropic recovery rates, and increased printability with improved accuracy and self-supporting ability, as well as enhanced β-carotene delivery from higher encapsulation capacity and bioavailability. These findings suggested that covalent interactions enhanced the interfacial nanoparticle layer, while non-covalent interactions endow greater antioxidant activity to the QP-Inu, providing insights for the designing of multifunctional HIPPEs with broad application potential.

Inflammatory bowel disease (IBD) management remains challenging due to the inadequate efficacy and systemic toxicity of conventional therapies. Herein, we developed an inulin hydrogel loaded with self-assembled nanoparticles of curcumin and glycyrrhizic acid (CURG@IN) for IBD treatment. CURG@IN exhibited remarkable gel characteristics and enzyme-responsive drug release behavior. In vitro, CURG@IN demonstrated potent antioxidant capabilities, effectively protecting cells from oxidative injury, facilitating wound healing of epithelial cells, and regulating macrophage polarization. Additionally, it exerted inhibitory effects on the NF-κB pathway, thereby reducing the production of inflammatory cytokines. In DSS-induced colitis mice, oral CURG@IN significantly ameliorated disease activity index and restored colonic histological architecture. Gut barrier integrity was reinforced through upregulation of tight junction proteins, while microbiota analysis revealed significant restoration of microbial homeostasis. Notably, no significant toxicity was observed during treatment. Collectively, this oral therapeutic strategy provides a multimechanistic approach combining anti-inflammatory, antioxidative, and microbiota-modulating effects for safe IBD therapy, with the potential for clinical application.

Over the last years, inulin, a fructan mixture consisting of oligosaccharides and polysaccharides, has attracted more and more attention from both food industry and researchers, due to its unique functional properties as a natural resource. Therefore, there is an increased interest in the extraction and quantification of inulin for its valorization from inulin rich plants, wastes and by-products. In this work, ultrasonic treatment was applied for inulin extraction, observing a great impact of extraction temperature and ultrasonic power on the inulin content in the obtained extracts. A combined process including trifluoroacetic acid (TFA)-assisted hydrolysis and analysis with high performance liquid chromatography equipped with evaporative light scattering detector (HPLC-ELSD) was developed to quantify inulin content. The effect of hydrolysis parameters was investigated, obtaining the optimal conditions after using TFA at a concentration of 1 mg/mL, hydrolysis temperature of 90 °C, and hydrolysis duration of 60 min. The good linearity (>0.995), precision, recovery (100.27%), and stability obtained during the validation process showed that this developed method allows the quantification of total inulin content in the samples analyzed. This combined method may also contribute to the investigation of the functional properties of inulin (e.g., as prebiotic).

The prebiotics inulin (INU) and isomalto-oligosaccharide (IMO) influence intestinal health and immunity, but their effects on constipation are not clearly established. We evaluated the effects of INU and IMO in a rat model of diphenoxylate-induced constipation. Twenty-four male rats were divided into four groups: basal diet (Con), 40 mg kg−1 diphenoxylate (PCon), 20 g kg−1 INU and treated with 40 mg kg−1 diphenoxylate, and 20 g kg−1 IMO and treated with 40 mg kg−1 diphenoxylate. INU and IMO increased the number, weight, and water content of fecal pellets, and decreased the time to the first black stool in rats with constipation. Serum levels of the gastrointestinal motility-related hormones adrenocorticotropic hormone (ACTH), motilin (MTL), and Substance P (SP) were higher and corticosterone (CORT), vasoactive intestinal peptide (VIP), and calcitonin gene-related peptide (CGRP) were lower in rats treated with prebiotics than in untreated rats. Colon tissue levels of MTL and SP were increased, and VIP and CGRP were decreased by prebiotics. Furthermore, in rats with constipation, INU and IMO increased the colonic contents of short-chain fatty acids. The relative abundance of Bacteroidetes was lower in the prebiotics groups than in the Con and PCon groups. Lactobacillus was more abundant in the INU and IMO groups than in PCon rats. Lactobacillus reuteri and Lactobacillus intestinalis were more abundant in the IMO group than in the PCon group (P < 0.01), and L. intestinalis was more abundant in the INU group than in the PCon group (P < 0.01). In summary, INU and IMO improved constipation and altered the intestinal microbiota in a rat model of constipation.

Autism Spectrum Disorder (ASD), a complex neurodevelopmental disorder marked by social communication deficits and repetitive behaviors, may see symptom amelioration through gut microbiota modulation. This study investigates the effects of a synbiotic – specifically a probiotic amplified by prebiotic supplementation – on ASD-like mouse model's social deficiencies. This model was established via valproic acid injection into pregnant females. Post-weaning, male progeny received daily synbiotic treatment, a combination of Lactobacillus reuteri (L. reuteri) and inulin, for four weeks. Results indicated that the synbiotic rectified social impairments and attenuated inflammatory cytokine expressions in the brain. Moreover, synbiotic intervention protected gut barrier integrity and altered the gut microbiota composition, enhancing the butyrate-producing Bifidobacterium abundance. The synbiotic elevated metabolites such as butyrate and 3-hydroxybutyric acid (3-HB), alongside upregulated genes associated with 3-HB synthesis in the colon and liver, and brain receptors. Conclusively, the synbiotic combination of L. reuteri and inulin mitigated ASD-related social impairments, partially via their regulatory effect on the gut–brain axis.

In this study, we used silver carp (Hypophthalmichthys molitrix) and sea cucumber (Apostichopus japonicus) as the primary protein sources and supplemented them with konjac glucomannan (KGM) or inulin (INU) at concentrations of 0%, 0.5%, 1.0%, and 1.5% to evaluate the gelling properties, textural characteristics, rheological behaviors, and in vitro digestibility of the resulting composite gels. The gastrointestinal digestion behavior, structural changes, and functional characteristics of these gels under simulated adult and elderly physiological conditions were also systematically investigated to clarify how different physiological stages influence their digestive fate. The results showed that incorporating KGM or INU modified hydrogen bonding and protein–polysaccharide interactions within the gel network, thereby regulating the structural integrity and rheological properties of the composite gels. According to the International Dysphagia Diet Standardization Initiative (IDDSI) framework, the KGM/INU-modified gels successfully passed the prescribed tests and were classified as level 4 (pureed/extremely thick) and level 6 (soft and bite-sized) foods, confirming their suitability for dysphagia management. Compared with the control group, the 0.5% KGM group showed the most desirable texture profile, with significantly lower hardness, chewiness, and adhesiveness, and higher cohesiveness (p< 0.05), while the 1.0% INU group exhibited the greatest enhancement in gel strength and viscoelasticity, as evidenced by the highest storage modulus (G′) and loss modulus (G″) (p< 0.05). Moreover, both adult and elderly in vitro digestion models showed extensive proteolysis of the composite gels, together with reduced protein aggregation, a looser microstructure in the digesta, and enhanced antioxidant capacity. Notably, the elderly model generally showed higher digestibility than the adult model, accompanied by greater release of free amino acids and antioxidant constituents (p< 0.05). Overall, these findings indicate that KGM/INU-modified composite gels possess promising potential as texture-modified foods, while the two in vitro physiological models provide a valuable platform for evaluating their digestion-related properties. KGM/INU-modified composite gels thus provide a promising platform for developing texture-modified, nutritionally fortified foods suitable for elderly individuals with swallowing impairments and adults who require safe, readily digestible dietary options.

Anthocyanin (ACN) is a natural pigment with various biological activities, but their stability is compromised by external environmental factors, which limits their practical application in food processing. To enhance the stability of anthocyanin, double-layer-modified anthocyanin nanoliposomes (ACN-NLs) were prepared in this study using pea protein isolate (PPI) and inulin (IN) through layer-by-layer assembly in this study. The preparation conditions of unmodified, single-modified, and double-layer-modified nanoliposomes (ACN-NLs, PPI-ACN-NLs, and IN-PPI-ACN-NLs) were optimized via analysis of their average particle size, zeta potential, and encapsulation efficiency (EE). In addition, the structure of the nanoliposomes was characterized via transmission electron microscopy (TEM) and a Fourier transform infrared (FTIR) spectrometer. Furthermore, the thermal stability of nanoliposomes in hot cocoa and their release behavior during in vitro simulated digestion were evaluated. The results indicated that the optimal formulation for IN-PPI-ACN-NLs was 6% PPI and 2% IN. Under these conditions, the IN-PPI-ACN-NLs had a particle size of 270.2 ± 0.66 nm, a zeta potential of −15.76 ± 0.81 mV, and a high EE of 88.6 ± 0.71%. TEM analysis revealed that IN-PPI-ACN-NLs exhibited a spherical core–shell structure, while FTIR confirmed the interaction between ACNs and the encapsulating materials (PPI and IN). Compared with unmodified or monolayer-modified nanoliposomes, IN-PPI-ACN-NLs exhibited thermal stability in beverage systems and enhanced DPPH radical scavenging activity. During in vitro digestion, IN-PPI-ACN-NLs demonstrated a sustained-release effect and improved the digestive stability of ACN. These properties make it a promising functional additive for applications in the food and pharmaceutical industry.

Whey protein isolate (WPI) is particularly constrained in infant formula owing to the widespread food allergy to milk proteins . Thus, inhibiting the allergens in WPI is crucial. This research aimed to explore the influence of the interaction between glycosylated protein [WPI glycosylated with short-chain inulin (SCI)] and cyanidin-3-glucoside (C3G) on antioxidant activity , thermostability, digestibility, and allergenicity of the proteins. The complex prepared by glycosylated WPI and C3G (2:1, w/w, pH 7.4) exhibited the best stability based on particle size and zeta potential . FTIR results showed that the major forces between glycosylated WPI and C3G were hydrogen bonding force, C–N stretching, and N–H bending. The antioxidant activity and thermal stability were as follows: WPI-SCI/C3G > WPI/C3G > WPI-SCI > WPI, indicating that the introduction of C3G and the interaction between glycosylated WPI and C3G can improve the above mentioned properties of the protein. Furthermore, WPI/C3G and WPI-SCI/C3G had higher resistance to pepsin and trypsin, which slowed down the digestion process and effectively delivered bioactive proteins to the distal end of the gastrointestinal tract. Western blot analysis and enzyme-linked immunosorbent assay showed that the IgE contents of WPI, WPI/C3G, WPI-SCI, and WPI-SCI/C3G were 26.99 ± 0.3, 23.65 ± 0.58, 23.07 ± 0.21, and 10.41 ± 0.14 ng/mL respectively, suggesting that the glycosylated modification and the introduction of C3G can significantly reduce the allergenicity of β-lactoglobulin in WPI. This study contributes to the development of a new promising method for reducing the allergenicity of WPI and its application in food industry.

White hyacinth bean polysaccharide (WHBP) was demonstrated to be effective in weight control and hypolipidemic regulation. Oral Superabsorbent Hydrogels (OSH), a class of dietary hydrogels capable of absorbing large amounts of water in the gastrointestinal tract, have emerged as an innovative carrier system for treating obesity. Here, combination of sodium alginate and inulin was employed to prepare OSH containing WHBP (named WHBP-OSH). Inulin was selected to improve characteristics of WHBP-OSH, and 5 concentrations of inulin (0–20 %) were tested in this work. Results showed that 10 % inulin was optimal for achieving superior encapsulation efficiency, mechanical properties and swelling capacity. Moreover, digestive behaviors suggested that WHBP-OSH presented significant water absorption and swelling, in addition to stability in both stomach and small intestine regions, thereby achieving colon-targeted delivery of WHBP. Subsequently, a 17-week high-fat diet-induced obesity mouse model was used to address roles of WHBP-OSH in obesity, and results showed that WHBP-OSH resulted in higher levels on weight reduction, Lees index, Body Mass Index (BMI) and adipose index than did OSH alone in obese mice, indicating that WHBP enhanced the anti-obesity effects of OSH. Mechanically, anti-obesity effects of WHBP-OSH were associated with decreasing eating behavior by suppressing the appetite reward and motivation aspects of obese mice. Together, WHBP-OSH containing 10 % inulin could develop a promising colon-targeted OSH, and showed significant potential as a weight management strategy via reducing eating behavior.

Algal oil is an aquatic plant source of polyunsaturated fatty acids, especially docosahexaenoic acid (DHA), which may exhibit health benefits when consumed in sufficiently high quantities. But its application as a healthy lipid in functional foods is often limited by its tendency to rapidly oxidize and produce rancid off-flavors. In this study, algal oil-in-water emulsions were prepared using pea protein isolate (PPI)-inulin conjugates as emulsifiers. These conjugates were prepared via the Maillard reaction using a combination of ultrasound and pH-shift treatments. The algal oil phase was mixed with lemon oil (0 to 1.2 wt%) prior to homogenization to improve its sensory characteristics. The addition of 0.6 wt% of lemon oil did not significantly affect the formation and physical stability of the emulsions but it did mask the unpleasant odor of algae oil. The conjugate-stabilized emulsions exhibited shear thinning behavior and had a higher viscosity than PPI-stabilized emulsions. They also had a higher surface load and formed thicker interfacial layers. The conjugates, especially the conjugates prepared by the combined treatments, also significantly improved the physical and oxidative stability of the emulsions. These effects were attributed to an increase in the steric repulsion between the oil droplets due to the hydrophilic inulin chains, as well as to an increase in the antioxidant activity of the interfacial layers due to the presence of more antioxidant proteins. Overall, our results show that the quality attributes and shelf life of DHA-fortified algae oil emulsions can be significantly improved using a plant-based protein-inulin conjugate.
