Patulin disrupts SLC7A11-cystine-cysteine-GSH antioxidant system and promotes renal cell ferroptosis both in vitro and in vivo

Hui Chen,Lixing Cao,Kai Han,Han Zhang,Jinling Cui,Xuan Ma,Shuang Zhao,Chong Zhao,Shutao Yin,Lihong Fan,Hongbo Hu

Patulin (PAT) is a common food-borne mycotoxin with diverse toxic effects including nephrotoxicity . The induction of oxidative stress is suggested to be a key mechanism contributed to toxicities of PAT. Reduced glutathione (GSH), a sulfhydryl-containing tripeptide , is a key reason for PAT-mediated oxidative stress. Cystine/glutamate antiporter (system x c − )-mediated cystine uptake plays a critical role in maintaining redox balance via promoting GSH biosynthesis. In this study, we addressed if GSH reduction by PAT was associated with inhibition of system x c − -mediated GSH biosynthesis. Results showed that PAT significantly decreased activity of SLC7A11, a core subunit of system x c − , through activating AMPK-mediated formation of beclin1-SLC7A11 complex. Furthermore, PAT promoted ferroptosis induced by a known ferroptosis inducer RSL3 in normal renal cells, and exacerbated folic acid-induced nephrotoxicity in a mouse model of acute kidney injury . The findings of the present study provide new insights into PAT-induced kidney toxicity, and implicate that patients with ferroptosis-associated diseases maybe more susceptible to PAT.

Simultaneous electrochemical aptasensing of patulin and ochratoxin A in apple juice based on gold nanoparticles decorated black phosphorus nanomaterial

Zhao Haiyan,Qiao Xiujuan,Zhang Xuelian,Niu Chen,Yue Tianli,Sheng Qinglin

Simultaneous detection of patulin (PAT) and ochratoxin A (OTA) in food products is in great demand, which can prevent toxins from being exposed to human and animal bodies. However, simultaneous detection of multiple targets still faces a challenge. Herein, we developed a novel electrochemical aptasensor for the simultaneous detection of PAT and OTA in apple juice based on gold nanoparticles decorated black phosphorus (AuNPs-BP) nanomaterial. AuNPs-BP function?/work? as a sensing platform for loading much different electrochemical signal molecules functionalized aptamers. In this context, methylene blue functionalized PAT aptamers (Mb-PAT-aptamers) and ferrocene functionalized OTA aptamers (Fc-OTA-aptamers) have been introduced here to fabricate the aptasensor. Fc close to electrode surface showed a strong signal, whereas Mb was far away from electrode surface so exhibited a weak signal in the absence of OTA and PAT. Two kinds of electrochemical signal changes have been recorded dependent on target of OTA and PAT concentrations. So, simultaneous detection of OTA and PAT is achieved. Under the optimum conditions, using this developed biosensor, PAT and OTA can be quantified at a linearity range of 0.01 × 10 −7  μg·mL −1  ~ 0.10 μg·mL −1 . In addition, it also has good selectivity, stability and repeatability. For the practical application, it shows promising performance for the simultaneous detection of PAT and OTA in apple juice.

Isolation and identification of Bacillus mojavensis YL-RY0310 and its biocontrol potential against Penicillium expansum and patulin in apples

Yu Ding,Fengjuan Liu,Jin Yang,Yingying Fan,Lvjian Yu,Zenghao Li,Na Jiang,Jing An,Ziwei Jiao,Cheng Wang

Penicillium expansum is the main causal organism of apple rot and causes huge economic losses during apple storage. Patulin (PAT) produced by P. expansum , occurs widely in apples and their products because of its water-solubility and thermal stability . In the present study, we isolated bacterial strains with antagonistic activity against P. expansum to effectively control the growth of P. expansum and production of PAT. We isolated a bacterium named YL-RY0310, which inhibited the growth of P. expansum and production of PAT in apples by 77.4% and 48.4%, respectively. Strain YL-RY0310 was 96% homologous to Bacillus mojavensis using morphological and 16S rRNA gene sequencing analyses. In addition, DNA amplification analysis using Bacillus -specific primers showed that the genes responsible for synthesis of lipopeptides of surfactin, iturins and fengycin are expressed. Oxford cup agar diffusion test showed that its crude lipopeptides could effectively inhibit the growth of P. expansum . Furthermore, strain YL-RY0310 could significantly degrade PAT; the degradation efficiencies of the intracellular and extracellular metabolites of the strain were 62.6% and 56.9%, respectively. Therefore, B. mojavensis YL-RY0310 could be used as a biocontrol agent to inhibit the growth of P. expansum and control PAT contamination in apples. This study provides a theoretical basis for a novel biological control strategy and degradation of mycotoxin.

The determination of patulin from food samples using dual-dummy molecularly imprinted solid-phase extraction coupled with LC-MS/MS

Minjuan Zhao,Hua Shao,Yahui He,Hui Li,Mengmeng Yan,Zejun Jiang,Jing Wang,A.M. Abd El-Aty,Ahmet Hacımüftüoğlu,Feiyan Yan,Yanli Wang,Yongxin She

A molecularly imprinted polymer (MIP) with specific adsorption for patulin was successfully polymerized by precipitation polymerization using 2-oxindole (2-oxin) and 6-hydroxynicotinic acid (6-HNA) as dummy template molecules, methylacrylic acid (MAA) as a functional monomer , trimethylolpropane trimethacrylate (TRIM) as a crosslinker, 2,2-azobis-(2-methylpropionitrile) (AIBN) as a initiator, and methanol as a porogen solvent. The molecularly imprinted solid phase extraction (MI-SPE) column was prepared using the polymer as a sorbent and applied for the selective extraction of patulin from real samples. The results showed that the MI-SPE method had high selectivity and specific adsorption towards patulin with mean recoveries ranged between 81.3% and 106.3% and a relative standard deviation (RSD) < 4.5%. Additionally, the developed MI-SPE method coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) exhibited good linearity in the range of 1–100 ng mL − 1 with correlation coefficients ( R 2 ) >0.998. The limits of detection (LODs, S/N = 3) were 0.05–0.2 ng g − 1 , and the limits of quantification (LOQs, S/N = 10) were 0.2–0.5 ng g − 1 . The developed method showed a better purification and higher patulin recovery for real samples than the quick, easy, cheap, effective, rugged, safe “QuEChERS” method.

Immobilized short-chain dehydrogenase/reductase on Fe3O4 particles acts as a magnetically recoverable biocatalyst component in patulin bio-detoxification system

Mengyang Xing,Yong Chen,Wanqin Dai,Xiao He,Boqiang Li,Shiping Tian

Patulin is one of the most important mycotoxins that contaminates fruit-derived products and causes acute or chronic toxicity in humans. In the present study, a novel patulin-degrading enzyme preparation was developed by taking a short-chain dehydrogenase/reductase and covalently linking it to dopamine/polyethyleneimine co-deposited magnetic Fe 3 O 4 particles. Optimum immobilization provided 63% immobilization efficiency and 62% activity recovery. Moreover, the immobilization protocol substantially improved thermal and storage stabilities, proteolysis resistance, and reusability. Using reduced nicotinamide adenine dinucleotide phosphate as a cofactor, the immobilized enzyme exhibited a detoxification rate of 100% in phosphate-buffered saline and a detoxification rate of more than 80% in apple juice. The immobilized enzyme did not cause adverse effects on juice quality and could be magnetically separated quickly after detoxification to ensure convenient recycling. Moreover, it did not exhibit cytotoxicity against a human gastric mucosal epithelial cell line at a concentration of 100 mg/L. Consequently, the immobilized enzyme as a biocatalyst had the characteristics of high efficiency, stability, safety, and easy separation, establishing the first step in building a bio-detoxification system to control patulin contamination in juice and beverage products.

Surface molecularly imprinted polymer capped Mn-doped ZnS quantum dots as a phosphorescent nanosensor for detecting patulin in apple juice

Wengang Zhang,Yong Han,Xiumei Chen,Xueli Luo,Jianlong Wang,Tianli Yue,Zhonghong Li

A Mn-doped ZnS quantum dots (QDs) based nanosensor for selective phosphorescent determination of patulin (PAT) was synthesized with 6-hydroxynicotinic acid (6-HNA) as dummy template via a surface molecular imprinting sol–gel process. FTIR and XRD indicated the successful graft of molecularly imprinted polymer (MIP) onto crystal QDs. Binding tests revealed that the MIP-QDs presented higher selectivity, adsorption capacity and mass transfer rate than non-imprinted polymers, demonstrating a specific recognition for PAT among competitive mycotoxins and its analogues with the imprinting factor of 2.02. The MIP-QDs could recognize PAT in a linear range of 0.43–6.50   μmol   L −1 with a detection limit of 0.32   μmol   L −1 and a correlation coefficient (R 2 ) of 0.9945. Recoveries of 102.9–127.2% with relative standard deviations <4.95% were achieved in apple juice samples which were in good agreement with high-performance liquid chromatography (HPLC) (P   >   0.05). The results indicated a simple phosphorescent nanosensor for PAT detection in complex matrix.

A fluorometric aptasensor for patulin based on the use of magnetized graphene oxide and DNase I-assisted target recycling amplification

Ma Liang,Guo Ting,Pan Shuli,Zhang Yuhao

A fluorometric patulin (PAT) assay is presented that is based on the use of magnetic reduced graphene oxide (rGO) and DNase I. The fluorescence of the PAT aptamer labelled with 6-carboxyfluorescein (FAM) is quenched by magnetized reduced graphene oxide (rGO-Fe 3 O 4 ) due to fluorescence resonance energy transfer (FRET). However, in the presence of PAT, the labelled aptamer is stripped off from rGO-Fe 3 O 4 . The rGO-Fe 3 O 4 is then magnetically separated so that the fluorescence of free labelled PAT aptamer is restored. DNase I cannot hydrolyze the aptamer on rGO-Fe 3 O 4 , but it can cleave the free aptamer-PAT complex. This will release FAM and PAT which can undergo a number of additional cycles to trigger the cleavage of abundant aptamer. Recycling of DNase I-assisted target therefore leads to a strong amplification of fluorescence and consequently to an assay with low limit of detection. The detection limit for PAT is as low as 0.28 μg L −1 which is about 13 times lower than that without using DNase I. The method offers a new approach towards rapid, sensitive and selective detection based on an aptamer. Conceivably, it has a wide scope in that it may be applied to numerous other analytes if appropriate aptamers are available. Schematic of a fluorometric assay based on the use of magnetic graphene oxide and DNase I. It was applied to the determination of patulin. DNase I was introduced for recycling amplification. The detection limit is about 13 times lower than that without using DNase I. Figure a contains poor quality of text in image. Otherwise, please provide replacement figure file.Thank you. A novel insight into fluorescent sensor for patulin detection using thiol-terminated liposomes with encapsulated coumarin-6 as signal probe

Dongxia Nie,Dakai Guo,Qingwen Huang,Wenbo Guo,Jianhua Wang,Zhihui Zhao,Zheng Han

Nanocomposites of thiol-terminated liposomes encapsulating with coumarin-6 ( [email protected] ) were successfully prepared via a simple thin-film dispersion method to construct a novel fluorescent sensor for selectively detecting trace amounts of patulin. Thiol group ( SH) was designed not only as a specific recognition element to capture patulin, but also as a convenient linker for the further separation of patulin-derivatized and un-derivatized [email protected] by combination with magnetic NH 2 [email protected] 3 O 4 nanoparticles. Liposomes primarily provided an effective platform with a large surface area for facilitated accommodation of large amounts of recognition element ( SH group) and fluorophore (coumarin-6). In such an assay protocol, a reliable link was established between the changes in fluorescent intensity (ΔF) from the nanocomposites of [email protected] /NH 2 [email protected] 3 O 4 and the concentrations of patulin in the range of 0.05–20 ng mL −1 , with the correlation coefficient of 0.996. This approach also effectively eliminated the background interference from other mycotoxins and metal ions. Under the optimal conditions, the designed sensor displayed excellent performance for patulin analysis, with an extremely low detection limit (0.033 ng mL −1 ), high recovery (96.2–107.6 %) and excellent selectivity. Furthermore, the feasibility of its applications has also been demonstrated in the analysis of real juice samples, providing a novel tactics for rational design of a fluorescence assay, for point-of-care diagnostics of patulin, with great potential to be extended to other hazardous compounds by substitution of the recognition elements.

Three-birds-with-one-stone: MIP-decorated MOF/COF hybrid nanoplatform for multifunctional PEC sensing, adsorption, and photocatalytic degradation of patulin

Wenyan Yan,Shuyue Li,Baoqing Bai,Ying Zhang,Jinhua Zhang,Tao Bo,Qinghua Wang,Ying Gu,Yukun Yang,Shuo Wang

A MIP-modified MOF/COF hybrid nanoplatform for PEC sensing, adsorption, and photocatalytic degradation was developed. • The MOF/COF hybrid enhanced photocurrent through improved light absorption and charge separation. • The MIP layer provided high selectivity. • The platform enabled ultrasensitive PAT detection and efficient adsorption-photocatalytic degradation. Rational design of photoactive materials is essential for the sensitive detection and efficient removal of hazardous contaminants. Herein, a visible-light-responsive covalent organic framework/metal-organic framework (COF/MOF) hybrid with uniform COF layers grown in-situ on the MOF surface, followed by the introduction of a molecularly imprinted polymer (MIP) layer via a sol-gel process, was constructed for the PEC sensing, adsorption, and photocatalytic degradation of patulin. The hybrid architecture enhanced photoelectrochemical performance by facilitating charge separation and improving light harvesting. The resulting PEC sensor exhibited a wide linear range (0.1 nM-1 mM) and a low detection limit (0.055 nM), along with good reproducibility and stability in real samples. Moreover, the MIP/MOF/COF hybrid enabled efficient adsorption and photocatalytic degradation of patulin, driven by photogenerated holes and superoxide radicals. This work provides a promising strategy for the efficient and highly sensitive detection and removal of contaminants. Download: Download high-res image (236KB) Download: Download full-size image

Highly efficient and cost-effective removal of patulin from apple juice by surface engineering of diatomite with sulfur-functionalized graphene oxide

Manshun Liu,Jing Wang,Xin Wang,Wenxin Zhu,Xiaolin Yao,Lihong Su,Jing Sun,Tianli Yue,Jianlong Wang

Patulin (PAT) contamination of apple juice leads to a serious food safety issue. Developing an excellent adsorbent to efficiently remove PAT is more desirable. Herein, a cost-effective and efficient adsorbent (GO-SH/diatomite) with abundant active sites was successfully fabricated via surface engineering of diatomite with sulfur-functionalized graphene oxide (GO-SH) nanosheets, which exhibited excellent selective adsorption capacity toward PAT. The adsorption behavior, adsorption mechanism, stability and cytotoxicity were investigated by systematic studies. The adsorption results showed that its maximum adsorption capacity was 10.68 μg/mg. Moreover, attributed to the specific interaction between PAT and thiol group, more than 90% of PAT was removed from apple juice without any juice quality deterioration. Importantly, the risk of food safety issue of apple juice caused by residual GO-SH/diatomite was negligible due to the properties of easy removal and excellent biocompatibility, which guaranteed its potential application in apple juice industry for PAT removal.

Efficiency and mechanism of a magnetic immobilized PPL system for simultaneous removal of the hazardous mycotoxins patulin and ochratoxin A from fruit juice

Yuan Wang,Lingxiu Liu,Fan Chen,Shaoyu Fang,Xiaohai Yan,Wushuang Bai,Qinglin Sheng,Yahong Yuan,Kewei Feng,Tianli Yue

This study developed two novel magnetic nanocomposites with immobilized porcine pancreatic lipase (PPL) for simultaneously removing patulin (PAT) and ochratoxin A (OTA)—two hazardous environmental contaminants—from fruit juices. All detoxification experiments were conducted in real fruit juice matrices (apple juice for PAT, grape juice for OTA) under ambient temperature (25 °C) with realistic contamination ranges. The results demonstrated MNP@UiO-66-(SH)₂@PPL achieved removal rates of 77.03% for PAT in apple juice and 45.03% for OTA in grape juice, while MNP@UiO-66-NH₂@PPL showed removal rates of 85.74% for OTA and 48.44% for PAT. LC-MS analysis indicated that PAT undergoes ring-opening to ascladiol, while OTA is hydrolyzed to phenylalanine and OTα. Molecular docking revealed that PPL spontaneously binds to both toxins via hydrogen bonding and hydrophobic interactions. CCK-8 assays confirmed low cytotoxicity of the materials and their degradation products, with no adverse impact on juice quality. The composites also exhibited magnetic recovery rates > 93%, enabling facile separation and recyclability. This work provides a safe, efficient, and reusable strategy for multi-mycotoxin decontamination with demonstrated environmental relevance.

Continuous flow removal of patulin by cysteine and porcine pancreatic lipase-modified hierarchical mesoporous zirconium metal–organic framework aerogel for apple juice treatment

Xiaohai Yan,Xinru Dong,Qiannan Zhao,Gengan Du,Qi Guo,Yahong Yuan,Tianli Yue

Patulin (PAT) is a mycotoxin synthesized by fungi such as Penicillium expansum . It has become a key risk indicator in fruit juices and therefore needs to be removed. Here, we used an ex-situ method and a self-assembly strategy to prepare a green carboxymethyl cellulose@hierarchical mesoporous zirconium metal–organic framework-cysteine/porcine pancreatic lipase (CMC@HMMOF-Cys/PPL) aerogel for the safe and efficient removal of PAT from apple juice. The optimal CMC@HMMOF-Cys/PPL aerogel exhibited high adsorption capacity (38.41 μg/mg), excellent degradation capacity (28.68 μg/mg at 10 μg/mL PAT), good reusability and storage stability, and improved selectivity. Notably, it could be customized as a filler for flow reactors to achieve the safe and efficient removal of PAT from apple juice. More importantly, the CMC@HMMOF-Cys/PPL aerogel designed did not have a significant effect on the quality of apple juice. The present study provided a new method for removing PAT from apple juice and further expanded the real-world applications of MOFs.
