Carboxymethyl chitosan-pullulan edible films enriched with galangal essential oil: Characterization and application in mango preservation

Wei Zhou,Yunxia He,Fei Liu,Liangkun Liao,Xiaobing Huang,Ruyi Li,Ying Zou,Lei Zhou,Liqiang Zou,Yuhuan Liu,Roger Ruan,Jihua Li

This study aimed to develop an active edible film based on carboxymethyl chitosan (CMCS) and pullulan (Pul) incorporated with galangal essential oil (GEO) by the casting method. And their physical properties, structural and preservation effect on mangoes were characterized. The CMCS/Pul ratio was determined to be 2.5:2.5 after the optimization of physical properties, mechanical properties and barrier properties of the blend film. The results of FT-IR and XRD showed that hydroxyl groups of Pul interacted with the carboxyl groups of CMCS and the blend films had good compatibility. Good thermal stability of CMCS/Pul-GEO films was further proven by TGA curves. The CMCS/Pul-8 %GEO film showed effective preservations on mango fruits during 15 days of storage at 25 ± 1 °C, based on the characterization by fruits weight loss, firmness, titratable acidity, soluble solids. Consequently, CMCS/Pul-GEO blend films may be a promising eco-friendly packaging material for the industrial application of fruit preservation.

Structural and physiochemical characterization of novel hydrophobic packaging films based on pullulan derivatives for fruits preservation

Ben Niu,Ping Shao,Hangjun Chen,Peilong Sun

Pullulan acetate (PLAc), pullulan propionate (PLPr) and pullulan butylate (PLBu) with various degrees of substitution (DS) were synthesized from pullulan and different carboxylic anhydrides (acetic anhydride, propionic anhydride and butyric anhydride). The structure of PLAc, PLPr and PLBu were determined by Fourier Transform Infrared Spectrometer (FT-IR) and 1 H Nuclear Magnetic Resonance Spectroscopy ( 1 H-NMR) spectroscopy. Pullulan ester films were obtained by solvent casting. The water vapor permeability, oxygen permeability, surface hydrophobicity, color and mechanical properties were investigated. Compared with native pullulan, all pullulan esters film showed a lower TS. TS and EAB declined with the increase of DS. PLBu3 achieved the lowest water vapor permeability property at 0.792 × 10 −11  g/m s Pa and lowest oxygen permeability property of 0.625 × 10 −3  g/m 2 s. The onset of ripening was monitored in terms of reduction in flesh firmness loss and weight loss. Package fruits with pullulan ester films delayed the ripening and senescence and hence could be used as an effective edible coating for extending their shelf life.

Pullulan-Functionalized Fe3O4 Nanoparticles with Mesopore Silica-Loaded Tetraphenylporphyrin Tetrasulfonic Acid Hydrate for Targeting Photodynamic Therapy

Liu Xinmeng,Zhao Hongli,Gong Wenxiu,Zhang Zhengcheng,Lan Minbo,Zhao Xueling

In cancer diagnosis and treatment, it is significant to targetedly and efficiently deliver drugs to tumor cells. In our present work, a multifunctional drug delivery system (Fe3O4@mSiO2(TPPS)-PL), pullulan (PL) coated Fe3O4@mSiO2 core/shell nanoparticles with tetraphenylporphyrin tetrasulfonic acid hydrate (TPPS) as a model photodynamic drug, has been designed and prepared. The obtained Fe3O4@mSiO2(TPPS)-PL nanoparticles were discrete and uniformly sized with an average diameter of 180 ± 10 nm. The cellular uptake of the Fe3O4@mSiO2(TPPS)-PL nanoparticles against HepG2 cells and HeLa cells were evaluated by flow cytometry and confocal scanning laser microscopy (CLSM), respectively. The results indicated that the cellular uptake of the nanoparticles by HepG2 cells was more than that by HeLa cells due to that HepG2 cells express maximum asialoglycoprotein receptor (ASGPR), and the uptake process was time-dependent. Additionally, the phototoxicity of Fe3O4@mSiO2(TPPS)-PL nanoparticles against HepG2 cells was investigated by MTT assay, and the materials exhibited efficient phototoxicity. It was suggested that the multifunctional drug delivery system is capable of targeting specific receptors, which is promising for photodynamic therapy (PDT).

Comparison of direct synthesis of silver nanoparticles colloid using pullulan under conventional heating and microwave irradiation

Kun Zhang,Shiyun Ai,Jiazhuo Xie,Jing Xu

In this comparative study, synthesis of pullulan-stabilized silver nanoparticles (AgNPs) colloid using conventional heating (CH) and microwave irradiation (MI) methods was investigated. The resulting AgNPs were characterized by UV-Vis, X-ray diffraction, and transmission electron microscopy. The reaction time and temperature were optimized for CH, and MI are 110°C, 40 min and 90°C, 10 min, respectively. The microwave-assisted approach improved pullulan/AgNPs colloid formation, with higher efficiency, shorter time, and lower temperature. However, the CH method produced more stable AgNPs with larger mean sizes. The antibacterial activities of the samples from CH and MI processes were compared.

Preparation and characterization of chitosan/pullulan film loading carvacrol for targeted antibacterial packaging of chilled meat

Longquan Xiao,Shuai Kang,Molazi Lapu,Peng Jiang,Xinhui Wang,Dayu Liu,Jing Li,Mingxue Liu

In this research, the common microorganisms in chilled meat were used as the targeted antibacterial objects. Chitosan, pullulan, and carvacrol were chosen to prepare the edible antibacterial film. The effects of different concentrations of carvacrol on the microstructure, physical properties and antibacterial properties of the films were investigated. The results showed that the antibacterial activity of chitosan/pullulan film (CS/PU) was unsatisfactory, when carvacrol was added, the antibacterial activity of the chitosan/pullulan/carvacrol film (CS/PU/CAR) improved significantly ( p   <   0.05 ), and the water vapor permeability (WVP) of the CS/PU/CAR decreased significantly ( p   <   0.05 ). When the carvacrol concentration was higher than 1.25% (w/v), the tensile strength and percentage elongation at break of the CS/PU/CAR increased significantly ( p   <   0.05 ), and the CS/PU/CAR exhibited satisfying antibacterial activity against the common bacteria in chilled meat such as Pseudomonas fluorescens , Listeria monocytogenes , Escherichia coli , Pseudomonas putida , Enterobacter cloacae , and Staphylococcus aureus . Finally, the CS/PU/CAR film was applied to the preservation of chilled goat meat and extended the shelf life of goat meat to more than 15 days. These results suggested that the targeted CS/PU/CAR film can be used as biodegradable films for the active packaging of chilled meat.

Characterization and Application in Packaging Grease of Gelatin–Sodium Alginate Edible Films Cross-Linked by Pullulan

Shuo Li,Min Fan,Shanggui Deng,Ningping Tao

Gelatin–sodium alginate-based edible films cross-linked with pullulan were prepared using the solution casting method. FTIR spectroscopy demonstrated the existence of hydrogen bonding interactions between the components, and scanning electron microscopy observed the component of the films, revealing electrostatic interactions and thus explaining the differences in the properties of the blend films. The best mechanical properties and oxygen barrier occurred at a 1:1 percentage of pullulan to gelatin (GP11) with sodium alginate dosing for modification. Furthermore, GP11 demonstrated the best thermodynamic properties by DSC analysis, the highest UV barrier (94.13%) and the best oxidation resistance in DPPH tests. The results of storage experiments using modified edible films encapsulated in fresh fish liver oil showed that GP11 retarded grease oxidation by inhibiting the rise in peroxide and anisidine values, while inappropriate amounts of pullulan had a pro-oxidative effect on grease. The correlation between oil oxidation and material properties was investigated, and water solubility and apparent color characteristics were also assessed. Keywords:pullulan;gelatin;sodium alginate;oil oxidation;edible film;grease packaging

Ultrasound-assisted development and characterization of novel polyphenol-loaded pullulan/trehalose composite films for fruit preservation

Lixin Kang,Qiufang Liang,Arif Rashid,Abdul Qayum,Zhuzhong Chi,Xiaofeng Ren,Haile Ma

A novel food packaging film was developed by incorporating a tea polyphenols-loaded pullulan/trehalose (TP@Pul/Tre) into a composite film with ultrasound-assisted treatment of dual-frequency (20/35 kHz, 40 W/L) for 15 min to assess the physicochemical and mechanical properties of a composite film. The optimized ultrasound-assisted significantly increases elongation at break, tensile strength, and improves the composite film's UV/water/oxygen barrier properties. Structure analysis using attenuated total reflectance-Fourier transform infrared, X-ray diffraction and thermal stability revealed that these improvements were achieved through ultrasound-enhanced H-bonds, more ordered molecular arrangements, and good intermolecular compatibility. Besides, the ultrasound-assisted TP@Pul/Tre film has proven to have good antibacterial performance against Escherichia coli and Staphylococcus aureus, with approximately 100 % lethality at 4 h and 8 h, respectively. Moreover, the ultrasound-assisted TP@Pul/Tre film effectively delayed moisture loss, oxidative browning, decay, and deterioration in fresh-cut apples and pears, thereby extending their shelf life. Thus, ultrasound has proved to be an effective tool for improving the quality of food packaging films, with a wide range of applications.

A chitosan/fucoidan nanoparticle-loaded pullulan microneedle patch for differential drug release to promote wound healing

Ayesha Younas,Zhuolin Dong,Ziye Hou,Muhammad Asad,Mengru Li,Nan Zhang

Wound healing is a largely unmet medical issue in trauma, burn, and diabetes. In this study, a pullulan-based and nanoparticle-loaded smart microneedle patch is designed to release drugs differentially based on the needs of wound healing. Chitosan and fucoidan are first used to prepare moxifloxacin (MOX)-loaded nanoparticles (MOXNPs) with a diameter of 258.0 ± 10.86 nm, PDI 0.19 ± 0.06, and surface charge 45.1 ± 3.9 mV. MOXNPs, lidocaine (LH), and thrombin (TH) are then incorporated to a 30 % ( w /w) pullulan-based microneedle patch (TH + LH +  [email protected] ). TH + LH +  [email protected] possesses uniform and cone-shaped microneedles with a length of 725 μm, demonstrating good biocompatibility, sufficient strength for skin penetration, fast skin dissolution within 55 ± 5 min, rapid release of TH and LH within 1 h, and sustained release of MOX for 24 h. TH + LH +  [email protected] heals mice skin wounds completely within 7 days and restores collagen deposition with accelerated cell proliferation , granulation, and reduced pro-inflammatory cytokines. In conclusion, this study utilizes combined polysaccharides to develop a smart multifunctional microneedle platform that achieves rapid hemostasis/analgesia and sustained bactericidal action. The smart and combined therapy is a potential strategy for high-quality wound healing.

Development of pullulan-based nanocomposite films reinforced with starch nanocrystals for the preservation of fresh beef

Miaoqi Dai,Xiong Xiong,Anwei Cheng,Zhengtao Zhao,Qian Xiao

BACKGROUND Incorporation of polysaccharide-based nanofillers is an effective strategy to fabricate bio-nanocomposite films with preferable mechanical, barrier, and surface hydrophobicity properties compared to pure biopolymer films. The objective of this research is to investigate the influence of starch nanocrystals obtained from native (NSNC) and waxy rice starch (WSNC) on the physical–chemical properties of pullulan-based nanocomposite films and their preservation performance on fresh beef. RESULTS Continuous SNCs network structure was observed for pullulan-10% SNCs nanocomposite films, whereas the percolation network of SNCs was destroyed and became no longer continuous with increasing SNCs concentration up to 20% in pullulan films. Among the tested films, pullulan-10% SNCs films showed the highest TS values, lowest WVP and OTR values, due to the formation of percolating SNCs network in pullulan matrix. It is noteworthy that the WVP and OTR values of pullulan-10% WSNC films were significantly lower than that of pullulan-10% NSNC films, probably due to higher hydrophobicity and crystallinity of WSNC compared with NSNC. Beef pieces coated with pullulan-SNCs films had higher L * and a * values, lower TVB-N, TBARS, and TVC values during 7 days' storage at 4 °C compared with samples coated with pullulan films. CONCLUSION Pullulan-SNCs nanocomposite films, especially pullulan-WSNC films, could be potentially used as a coating material for fresh beef due to their desirable oxygen and water barrier properties. © 2022 Society of Chemical Industry.

Construction of Artemisia sphaerocephala Krasch. Polysaccharide based hydrogel complexed with pullulan and gelatin crosslinked by ferric ions

Xiaolin Yao,Xiaoxue Yao,Xiaoyu Chen,Jianxiong Yue,Dan Yang,Ning Liu,Katsuyoshi Nishinari

Artemisia sphaerocephala Krasch. polysaccharide (ASKP) was found to be crosslinked with ferric ions to form hydrogels in the previous study. In this work, it was demonstrated that ASKP-Fe 3+ hydrogel complexed with pullulan or gelatin contributed to a significantly enhanced gel strength at 1.5% ASKP, 60 mM Fe 2+ , pH 4.0, and the mixing ratio of 9: 1. The complexed hydrogels presented a dense semi-interpenetrating network along with the delay of gelation time and the increase of water retention. ASKP based complexes exhibited good compatibility, probably because pullulan and gelatin could be entangled with ASKP chain under hydrogen bonding and electrostatic interaction, respectively. The interaction between ASKP and pullulan or gelatin contributed to the formation of complexed hydrogels with dense network and significantly enhanced gel strength. It is inferred that ASKP would have great potential to be a new gelling material as well as for the ferric ions delivery.

Grafted modified chitosan/pullulan film loaded with essential oil/β-CD-MOFs for freshness preservation of peach

Linyun Mou,Ya Lu,Jiale Ma,Shuwei Shu,Jianlong Li,Ganpeng Li

In response to the rapid quality degradation of peaches due to post-harvest decay, this study developed a composite film for preservation. Two primary pathogens responsible for peach decay, Aspergillus flavus and Talaromyces funiculosus , were identified. Molecular docking attributed to the main binding position of citral and geraniol's interactions with type I polyketide synthase on their antifungal effects, as well as the caffeic acid with its tyrosinase on antioxidant efficacy. β -CD-MOFs were synthesized via vapor diffusion, demonstrating exceptional spatial loading via SEM. CEOs (citral + geraniol) encapsulated in β -CD-MOFs were incorporated into a chitosan/pullulan matrix, yielding a film with 260.27 % fracture tensile strain, significantly ( P  < 0.001) outperforming pure chitosan (21.87 %), possessing excellent mechanical properties, and the improvement in the water solubility of the film facilitates the release of active compounds. The utilization of a compositel film on peaches has shown significant efficacy in controlling decay index ( P  < 0.05), weight loss ( P  < 0.05), firmness ( P  < 0.05) and production of malondialdehyde ( P  < 0.05). Compared to untreated controls, the treated peaches exhibited an extended shelf life of 2 days, highlighting the superior preservation capabilities of the film. Hence, the film presents a promising solution for fruit preservation.

Development and characterization of BSA-pullulan gel matrices incorporating orange peel flavonoids: Structural interactions and antimicrobial functionality

Mahmoud Salah,Mabrouk Sobhy,Yajing Fang,Salwa M. El-Sayed,Ibrahim Khalifa,Ali Korin,Mohammed Mansour,Syed Asim shah Bacha,Sajid Maqsood,Yun Wang

This study investigates the molecular interactions and functionality of bovine serum albumin-pullulan (BSA-Pn) hydrogels integrated with citrus flavonoid extracts (Fs). Structural characterization via vibrational and fluorescence spectroscopies revealed noncovalent interactions, primarily H-bonding, between BSA, pullulan, and flavonoids, as confirmed by molecular docking modelling. Surface chemistry analysis revealed that increased C1s-peak intensity in BSA-Pn-Fs-9 composite, thereby indicating a structural reorganization. The hydrogel demonstrated potent an antimicrobial activity against Gram-positive ( Bacillus cereus and Staphylococcus aureus ) and Gram-negative ( Escherichia coli ) bacteria, with inhibition zones of 1.34–1.69 cm, and suppressed fungal growth ( Penicillium expansum and Aspergillus westerdijkiae ) by 40.72–43.24 %. Thermal stability of flavonoids within matrix was enhanced, as evidenced by thermal structure analysis. Antioxidant assays showed significant radical scavenging with values of 33.49–93.47 % versus free radicals scavenging, confirming preserved flavonoid bioactivity. These findings underscore the structure-function synergy of BSA-Pn-Fs composites, driven by polysaccharide-protein-flavonoid interactions, positioning them as promising candidates for bioactive and functional biomaterials/additives.
