Effects of curcumin on growth performance, intestinal morphology and microbiota, and liver transcriptome in Asian yellow pond turtle (Mauremys mutica)

Mingzuo Gao,Jingu Shi,Zike Yan,Hongyao Zhang,Kaijie Lin,Yutian Zhan,Yuhan Li,Shuyu Han,Jingzhen Liang

This study investigated the effects of dietary curcumin supplementation at different concentrations on the growth performance, intestinal morphology and microbiota, and liver transcriptome of Mauremys mutica . Healthy turtles were randomly assigned to five groups: a control group (C0, 0 mg/kg curcumin) and four treatment groups (C100, C200, C400, and C800) receiving curcumin at 100, 200, 400, and 800 mg/kg diet, respectively. Each group consisted of three replicates (30 turtles per replicate), and the trial lasted for 60 days. The results demonstrated that dietary supplementation with 100, 200, and 400 mg/kg curcumin significantly improved both weight gain rate (WGR) and specific growth rate (SGR), with the 200 mg/kg group showing the highest activities of α-amylase, trypsin, and lipase compared to the control group. Curcumin supplementation also significantly increased intestinal villus height, muscular thickness, and goblet cell counts per villus, indicating an improvement in intestinal morphology. Furthermore, curcumin modulated the gut microbiota composition by increasing the relative abundance of Prevotellaceae_unclassified while decreasing Desulfobacterota, Proteobacteria, Edwardsiella , Bacteroides , and Negativibacillus . Transcriptomic analysis of liver tissues revealed 215, 422, 265, and 243 unique DEGs in the 100, 200, 400, and 800 mg/kg groups, respectively. These DEGs were primarily enriched in immune-related pathways such as the PI3K-Akt, and TNF signaling pathways, as well as metabolism-related pathways including the PPAR signaling pathway, nucleotide metabolism, and various amino acid metabolism pathways. Collectively, these findings indicate that curcumin enhances growth performance and digestive capacity, improves intestinal morphology and digestive enzyme activity, modulates microbial community, and regulates hepatic gene expression related to immune and metabolic function in M. mutica . Based on regression analysis (R 2 > 0.5) of key indicators—including WGR, SGR, FCR, and the activities of trypsin and lipase, as well as malondialdehyde (MDA) content—and with no significant effect on survival, the optimal dietary inclusion level of curcumin for M. mutica was estimated to be between 199.74–266.73 mg/kg.

Engineered Cancer Cell Membrane-Functionalized Metal-Organic Frameworks for Irinotecan/Curcumin Codelivery in Colorectal Cancer: Enhanced Efficacy and Mitigated Toxicity

Shuiling Jin,Aifang Zhou,Qi Zhang,Di Zhao,Xintong Dong,Hong-Min Meng,Zhengquan Yu,Zhaohui Li

Purpose Irinotecan (CPT-11), a semisynthetic camptothecin derivative and topoisomerase I inhibitor, is globally used for treating cancers such as colorectal cancer. However, its clinical application is limited by severe adverse effects, including diarrhea and myelosuppression. Biomimetic nano-drug delivery is widely used and can improve chemotherapeutic activity and decrease side effects. Methods In this study, we developed a cancer cell membrane-functionalized metal-organic frameworks for irinotecan/curcumin (named MZCC) for colorectal cancer treatment. In this study, by combining irinotecan, curcumin, MOFs and functionalized cancer cell membrane, a pH-sensitive drug delivery system of MZCC was developed for colorectal cancer treatment with enhanced chemotherapeutic efficacy while reducing side effects. The characteristics and stability of MZCC were carefully examined, and its anti-cancer efficiency was studied in vitro and in vivo. In vivo experiments in a murine colorectal cancer mouse model given intratumorally MZCC injection every 4 days were used to assess antitumor efficacy and systemic toxicity. Results The MZCC system improved treatment antitumor efficacy by 2.3-fold compared with free CPT-11 through synergistic chemotherapy. MZCC-treated groups exhibited a significant reduction in inflammatory cytokine levels within colon tissues; inflammation levels decreased by 55% compared with free CPT-11, and colon length was restored to 79% of normal. Conclusion Our strategy effectively alleviates CPT-11-induced intestinal toxicity and mitigates adverse reactions. This approach may provide a novel method for enhancing chemotherapeutic efficacy while reducing gastrointestinal toxicity.

Covalent engineering of pectin-whey protein isolate conjugates enhances mucoadhesion and bioaccessibility of curcumin microparticles: Strategic impacts of crosslinking modalities

Haifeng Wang,Ting Fang,Ying Chen,Jiang Mou,Teng Li,Yuying Fu,Yun Zhai

This study systematically investigates the structure-function relationships between Maillard-glycated (PWM) and genipin-mediated crosslinked (PWG) pectin-whey protein isolate (WPI) complexes, with particular emphasis on their differential impacts on bioadhesive performance and digestion profiles for curcumin-loaded delivery systems. Both crosslinking strategies induced structural unfolding of WPI and endogenous pectin proteins, exposing amino-active sites that facilitated their reconstitution into stable complexes. PWG developed denser and more homogeneous hydrophobic microdomain networks compared to PWM. Curcumin was encapsulated via hydrogen bonding and hydrophobic interactions within these networks. Ex vivo intestinal adhesion assays demonstrated PWG loaded curcumin (PWG-Cur) achieved 28.69 % particulate retention in colonic environments, representing 7.0-fold and 1.75-fold enhancements over free curcumin and PWM loaded curcumin (PWM-Cur), respectively. In vitro digestion models confirmed PWG-Cur's superior bioaccessibility (22.46 %), reflecting a 37 % enhancement over PWM-Cur (16.35 %). These findings provide critical design principles for engineering mucoadhesive pectin-polyphenol delivery platforms capable of precisely modulating cargo release while enhancing bioaccessibility.

Curcumin encapsulation enhanced by soy protein isolate modified through deamidation and pH-shifting: Mechanistic insights using a novel protein-glutaminase

Yuelong Xu,Yu Zhang,Jingcheng Ma,Ting Wang,Chong Zhang

This study elucidates the mechanism of curcumin encapsulation in soy protein isolate (SPI) modified via enzymatic deamidation and pH-shifting. A novel Chryseobacterium sediminis -derived glutaminase (CS-PG) demonstrated superior thermal stability, catalytic efficiency, and specificity over commercial CP-PG. Controlled deamidation by both enzymes enhanced SPI functionality (solubility/emulsification/foaming) with minimal hydrolysis, where CS-PG induced hydrophobic self-assembly and CP-PG promoted hydrophilic domain formation. These distinct conformations dictated binding modes: CS-PG-SPI achieved 92.97 % higher encapsulation efficiency versus SPI through hydrophobic encapsulation, while attained an 85.00 % increase via surface-layer stabilization. pH-shifting further altered SPI conformation by inducing reversible unfolding and refolding, enabling formation of amorphous, stable protein–curcumin complexes. Dual modification significantly enhanced bioaccessibility versus free curcumin—1034 % for CS-SPI–Cur–pH and 1058 % for CP-SPI–Cur–pH. The work establishes enzymatic deamidation as a precision tool for tailoring SPI self-assembly and bioactive delivery in functional foods.

Construction of hyperbranched polyglycerol-based curcumin prodrug via phenol-yne click reaction for pH-responsive and bone-targeted osteoporosis therapy

Peng Luo,Jiaming Fu,Xiaowei Yang,Fengbo Mo,Yanlong Zhong,Qi Lai,Shaorong Huang,Xiaoyong Zhang,Bin Zhang,Yen Wei

Osteoporosis is one of the most common chronic metabolic diseases that mainly caused by an imbalance in bone homeostasis. Considered its severe concurrent disorders, osteoporosis has become a global health issue, however, the anti-osteoporosis medications available in clinical practice are confronted with challenges such as high costs, adverse side effects, and limited efficacy. Herein, we reported construction of pH-responsive and bone-targeting nanomedicine (termed HPGCA) via click conjugation of alkyne functionalized hyperbranched polyglycerol (HPG) with curcumin (Cur) and alendronate (ALN) for the first time. We demonstrated that HPGCA exhibit superior water dispersibility, small particle size, and pH-dependent drug release behavior. Results from biological assays indicated that HPGCA displayed desirable bone-targeting capability, strong anti-osteoporosis efficacy, good hemocompatibility, and negligible adverse effects. More importantly, HPGCA showed better anti-osteoporosis efficacy than both free Cur and ALN according results from micro-CT and TRAP staining. The synergistic osteoporosis therapeutic efficacy would be explained by targeting delivering HPGCA to bone tissues and in-situ releasing cargos under acidic environment. Therefore, considered the above aspects, we believe that the innovative nanomedicine is promising for osteoporosis therapy in clinic and will provide new insights for full realization anti-osteoporotic properties of natural compounds

Bhandari Shashikant V.,Kuthe Pranali,Patil Shital Manoj,Nagras Om,Sarkate Aniket Pardip

Introduction: Cancer is the major cause of death globally. Cancer can be treated with naturally occurring Curcumin nuclei. Curcumin has a wide range of biological actions, including anti-inflammatory and anti-cancer properties. Even though it is an effective medicinal entity, it has some limitations such as instability at physiological pH and a weak pharmacokinetic profile due to the β-diketone moiety present in it. To overcome this drawback, research was carried out on monoketone moieties in curcumin, popularly known as mono-carbonyl curcumin. Objective: The present review focuses on different synthetic schemes and Mono-carbonyl curcumin derivative's Structure-Activity Relationship (SAR) as a cytotoxic inhibitory anticancer agent. The various synthetic schemes published by researchers were compiled. Methods: Findings of different researchers working on mono-carbonyl curcumin as an anticancer have been reviewed, analyzed and the outcomes were summarized. Results: The combination of all of these approaches serves as a one-stop solution for mono-carbonyl curcumin synthesis. The important groups on different positions of mono-carbonyl curcumin were discovered by a SAR study focused on cytotoxicity, which could be useful in the designing of its derivatives. Conclusion: Based on our examination of the literature, we believe that this review will help researchers design and develop powerful mono-carbonyl curcumin derivatives that can be proven essential for anticancer activity.

Solid Lipid Nanoparticles Enhanced the Neuroprotective Role of Curcumin against Epilepsy through Activation of Bcl-2 Family and P38 MAPK Pathways

Ruiqi Huang,Yanjing Zhu,Lijuan Lin,Simin Song,Liming Cheng,Rongrong Zhu

Oxidative stress of neurons caused by a series of complex neuropathological processes will induce certain neurodegenerative disorders including epilepsy. Curcumin (Cur) is an effective natural antioxidant compound; however, the poor bioavailability obstructs its neural protective applications. In this study, Cur is encapsulated in solid lipid nanoparticles (SLNs) for better neuroprotective efficacy. In vitro study certified that Cur-SLNs functioned obviously better against neuronal apoptosis than Cur, by significantly decreasing the level of free radical and reversing mitochondrial function through the activation of the Bcl-2 family. In vivo experiments showed that SLNs transported Cur through the blood–brain barrier (BBB). The behavioral performance of epileptic mice was improved by Cur-SLNs, with more NeuN but less TUNEL positive cells observed in hippocampus. The in vivo mechanism was also explored. Cur-SLNs reduced neuronal apoptosis through Bcl2 family and P38 MAPK pathways. Overall, Cur-SLNs have better protective effects toward oxidative stress in neurons than free Cur both in vitro and in vivo, which suggests they may be a promising agent against neurodegenerative disorders including epilepsy.

Surface modification of MCM-41 by chain transfer free radical polymerization and their utilization for intracellular pH-responsive delivery of curcumin

Wenfeng Yan,Junmao Li,Wei Long,Yulin Feng,Mingzhen He,Weifeng Zhu,Meiying Liu,Hui Ouyang,Xiaoyong Zhang,Yen Wei

In this paper, a simple method that relied on direct chain transfer free radical polymerization was reported to synthesize MSNs based polymeric composites (named as MCM-41-SH-poly (PEGMA- co -VPA)) using poly (ethylene glycol) monomethyl ether (PEGMA) and 4-vinyl phenylboric acid (VPA) as monomers. The utilization of MCM-41-SH-poly (PEGMA- co -VPA) to load and delivery curcumin (CUR) was also evaluated in detail. The structure, properties, drug loading and release behaviors were evidenced by various characterization techniques. The results from biological studies have shown that CUR can be effectively loaded on MCM-41-SH-poly (PEGMA- co -VPA) with high drug loading rate through formation of borate bond between CUR and VPA. More importantly, resultant drug-loading complexes displayed excellent water dispersibility and CUR could be released from complexes with pH responsiveness. Taken together, we have developed a simple method for surface modification MCM-41 and resultant composites could display improved properties for intracellular controlled drug delivery applications.

Dual-crosslinked polysaccharide microspheres via glycated collagen and sodium alginate/chitosan for intestine targeted delivery of curcumin in ulcerative colitis treatment

Jing-jing Fu,Si-yi Mei,Yong-qiang Chen,Xiang-yu Fu,Yong-jie Long,Jun-xin Wu,Wen-qiang Cai,Yue-wen Chen

Polysaccharide-based hydrogel microspheres have demonstrated considerable potential for functional factors intestinal-targeted delivery and ulcerative colitis (UC) treatment. However, single-polysaccharide systems often struggle to achieve both stable loading and controlled release of functional factors. In this study, protein-polysaccharide composite microspheres were developed using a dual-crosslinking strategy. Initially, curcumin (Cur) was encapsulated within Andrias davidianus collagen (ADC) or its xylose glycated form (ADCXy) to form ADC/ADCXy-Cur complexes. These complexes were then crosslinked with sodium alginate (SA) and chelated with Ca 2+ ions to construct a primary hydrogel network (ADC/ADCXy-Cur-SA). Finally, the cationic polysaccharide chitosan (CS) with colon enzymolysis targeting, mucosal adhesion and pH responsive controlled release characteristics was incorporated with ADC/ADCXy-Cur-SA to form a double cross-linked multifunctional hydrogel microsphere (ADC/ADCXy-Cur-SA-CS). Compared with ADC-Cur-SA-CS, the ADCXy-Cur-SA-CS exhibited enhanced cellular antioxidant and anti-inflammatory activity, maintained high stability in simulated gastric conditions, and enabled controlled Cur release in colonic pH environment. In vivo experiments have shown that ADCXy-Cur-SA-CS alleviated UC based on the “one stone, four birds” strategy of antioxidant, anti-inflammatory, repairing the colon mucosal barrier, and regulating the gut microbiota. This study provides an important technical and theoretical foundation for the development of multifunctional polysaccharide-based hydrogel delivery systems.

Folic acid/3-carboxyphenylboronic acid dual-functionalized cellulose nanocrystals as a pH-responsive nanocarrier for improved curcumin delivery against hepatocellular carcinoma

Xi Wang,Yuying Wang,Ziqi Li,Bin Zou,Li Lin,Zhifeng Xue,Ruzhi Zhang,Weimin Wan

Hepatocellular carcinoma (HCC) remains a leading cause of cancer-related mortality worldwide due to late diagnosis, high metastasis, and poor therapeutic outcomes. Curcumin (Cur), a natural polyphenol with potent antitumor activity, is severely limited in clinical application by poor water solubility, low bioavailability, and insufficient tumor-targeting capability. Herein, a dual-targeted pH-responsive nanocarrier based on cellulose nanocrystals (CNCs) co-functionalized with folic acid (FA) and 3-carboxyphenylboronic acid (CA) was fabricated for enhanced Cur delivery to HCC cells. CNCs were aminated via silane coupling, followed by covalent co-grafting of FA and CA to obtain ACNCs@FA- CA. FA enabled targeting of folate receptors overexpressed on HCC cells, while CA played a dual role by reversibly binding Cur through boronate ester formation and recognizing sialic acid-rich tumor cell membranes. Cur was loaded with an efficiency of 13.90 ± 2.00% and exhibited pH-responsive release, reaching 81.61 ± 1.60% at pH 5.4 and 26.29 ± 1.47% at pH 7.2. In vitro studies demonstrated that Cur-loaded ACNCs@FA-CA significantly enhanced intracellular drug accumulation and cytotoxicity in HepG2 cells, with improved growth inhibition in a 3D hepatoma spheroid model. Collectively, this dual-targeted pH-responsive nanoplatform offers a promising strategy to improve the anti-HCC efficacy of Cur.

Self-Assembled Curcumin–Epigallocatechin Gallate Nanoparticles Enhance Antiviral Activity Against White Spot Syndrome Virus Through Immunomodulation, Antioxidant Effects, and Metabolic Regulation

Songlin Luo,Xudong Yang,Shunqiang Wei,Chushun Yi,Fansheng Zeng,Fali Zhang,Wei Zhang,Xiuli Chen,Yongzhen Zhao,Yinghui Wang,Aiguo Huang

White Spot Syndrome Virus (WSSV) remains a major factor limiting the sustainable development of crustacean aquaculture, creating an urgent need to develop comprehensive antiviral strategies with multiple protective functions. Natural bioactive compounds with antiviral, immunomodulatory, and antioxidant activities hold great potential for application; however, their poor stability and low bioavailability limit their practical use. In this study, self-assembled nanosystem (CE SA) composed of epigallocatechin gallate (EGCG) and curcumin (Cur) was constructed based on noncovalent interactions, and its protective effects against WSSV-infectedProcambarus clarkiiwere systematically evaluated. The results indicate that CE SA exhibits good stability, sustained-release properties, and biosafety. Compared with Cur, EGCG, and their physical mixture (CE), CE SA significantly improved the survival rate of infected shrimp and reduced viral copy numbers as well as the expression levels of key viral genes such asIE1,DNApol, andVP28. At the same time, CE SA significantly promoted the expression of relevant genes such asDorsal,Relish,SOD, andCAT, increased SOD and CAT enzyme activities and hematopoietic cell viability, and effectively alleviated damage to the hepatopancreas and intestinal tissues. Furthermore, metabolomic analysis indicated that CE SA regulates key metabolic processes such as amino acid metabolism, inflammation-related metabolism, nucleotide metabolism, and redox homeostasis, further elucidating its potential mechanism for enhancing the host’s antiviral capacity. In summary, by enhancing the stability and utilization efficiency of natural bioactive components, CE SA achieves superior anti-WSSV efficacy compared to individual components. Its protective effects are closely related to the inhibition of viral replication, enhancement of the immune response, improvement of antioxidant defenses, and regulation of host metabolic homeostasis. This study provides a promising, self-assembling nanostructure strategy for the green prevention and control of viral diseases in aquatic animals.

A novel edible, all-biomass Pickering emulsion coating based on Lys/Alg composite nanoparticles for curcumin delivery and active fruit preservation

Fengqiong Jiang,Chaoran Fan,Mingfei Dang,Fuxiang Wei,Lu Liang,Lihua Fu,Chuanhui Xu,Baofeng Lin

A stable food-grade amphiphilic Lys/Alg nanoparticle emulsifier was prepared by double crosslinking. • An Edible, all-biomass-derived multifunctional Pickering emulsion was successfully developed via simple emulsification. • The Lys/Alg served as effective Pickering stabilizer enhanced the stability and bioavailability of curcumin. • The developed emulsion exhibited high antioxidant properties (99.02%) and broad-spectrum antibacterial activity. • The shelf life of various fruits was effectively extended by the emulsion coating. The integration of nano-encapsulation with edible coatings holds great potential for active fruit preservation. However, conventional emulsions often rely on chemical surfactants, posing safety risks and limited functionality. Here, we developed an edible, all-biomass-derived Pickering emulsion (CLAPE) coating stabilized by amphiphilic lysozyme/alginate composite nanoparticles (Lys/Alg). The Lys/Alg were fabricated through electrostatic self-assembly followed by Ca 2+ cross-linking, forming a robust interfacial layer that effectively encapsulates curcumin (Cur). Molecular docking simulations revealed a strong binding affinity between Lys and Cur (binding energy: −87.2 kcal/mol), which significantly enhances the aqueous dispersibility and stability of Cur. The resulting Pickering emulsion enables sustained release of curcumin over 60 h, with a peak cumulative release of 94.0%, approximately twice that of the unencapsulated control. This release profile follows Fickian diffusion, extending the bioactive duration and increasing the total released amount, thereby enhancing curcumin's bioavailability. Moreover, CLAPE exhibits excellent thermal stability, maintaining high stability indices under varied accelerated storage conditions. It also exhibited high antioxidant (99.02%) and broad-spectrum antibacterial activity. Additionally, the application of CLAPE on strawberries, bananas and grapes has led to a doubling of their shelf life while maintaining the freshness and nutrient content of the fruits. The edible active preservative material, based entirely on biomass, not only forms a multifunctional protective coating that prevents spoilage but can also be easily washed off. This study presents an innovative strategy for the designs of active preservation materials utilizing all-natural plant-derived components.
