Piezo1 activation in endothelial cells aggravates microvascular ischemia–reperfusion injury in limbs by enhancing ferroptosis

Chen Fan-feng,Zhang Yin-he,Wu Zi-chang,Du Kaiyi,Chen Xinyuan,Lu Yang,Hu Qianqian,Du Anyu,Du Shenghu,Wang Jian,Shi Keqing,Chen Zimiao,He Zili,Zhou Kailiang,Xiao Jian

Acute limb ischemia–reperfusion injury (ALIRI) prominently involves microvascular dysfunction, with notable contributions from damage to microvascular endothelial cells (MECs). Previous research suggests that the mechanosensitive ion channel Piezo1 becomes active in response to mechanical stress conditions, including ischemia and trauma. However, its precise function within the ALIRI context remains elusive. Notably, the expression of Piezo1 was markedly elevated postreperfusion in mouse hind limb ischemia/reperfusion (I/R) models, implicating its crucial involvement in limb survival. Employing specific inhibitors of cell death pathways, the study delineated key molecular drivers of ferroptosis during limb damage. Here evaluations of limb vitality, western blot, quantitative PCR and immunofluorescence implicated that activation of Piezo1 by its agonist exacerbates I/R-induced microvascular perfusion deficits, tissue swelling, skeletal muscle damage and increased tissue infarction and MECs damage. Conversely, these detrimental impacts were mitigated through pharmacological blockade of Piezo1 or specific deletion of Piezo1 in MECs. Comprehensive untargeted metabolomic analysis revealed significant changes primarily in glycerophospholipid and arachidonic acid metabolism pathways. Further experiments demonstrated that RNA interference-mediated inhibition of cytosolic phospholipase A2 (cPLA2) and acyl-CoA synthetase long-chain family member 4 (ACSL4) negated the protective effects against ferroptosis and limb damage that were observed with Piezo1 deletion. Moreover, this study confirmed that protein kinase C phosphorylates ACSL4, which mediates Piezo1-induced ferroptosis and exacerbates limb damage, as shown through immunoprecipitation studies. In summary, Piezo1 contributes to the exacerbation of microvascular and skeletal muscle damage in ALIRI by facilitating the cPLA2-dependent release of arachidonic acid and promoting ACSL4-driven lipid peroxidation, thereby intensifying ferroptosis in MECs.

Chemosensory Pathways Involved in the Perception of Tomato-Derived Flavonoids and Alkaloids in Meloidogyne incognita

Qian Wang,Liping Peng,Youjing Wang,Jiajia Hu,Liangying Kong,Yajun Liu

Plant-parasitic nematodes rely on the perception of host-derived chemical cues released from plant roots to locate suitable hosts, representing a critical prerequisite for successful plant infection. In this study, we selected four representative tomato (Solanum lycopersicumL.) root-derived chemical signals and demonstrated their distinct effects on the host-seeking behavior of the southern root-knot nematode (Meloidogyne incognita). Among these compounds, the flavonoids quercetin and luteolin exhibited significant attractant effects, whereas the alkaloids dihydrocapsaicin and solasodine showed repellent effects. RNA interference (RNAi) experiments further indicated that multiple chemosensory neurons are involved in the recognition of plant-derived chemical cues. Silencing of key chemosensory genes significantly impaired the directed chemotactic response ofM. incognitatoward tomato roots and reduced its infectivity, demonstrating that the chemosensory system represents a crucial molecular basis regulating host localization and parasitic establishment. This study provides new insights into host recognition mechanisms in root-knot nematodes and offers a theoretical foundation for developing environmentally sustainable strategies for nematode management by targeting host perception processes.

Endoplasmic reticulum stress is involved in chronic chlorpyrifos-induced jejunal barrier injury in weaned mice

Junkang Zhao,Yue Zhang,Jinyu Li,Yuli Zhu,Xunuo Song,Yao Meng,Yuming Zhang,Qinghua Deng,Liyin Du

Chlorpyrifos (CPF), a widely used organophosphate insecticide, can cause intestinal toxicity under chronic exposure conditions, but the upstream mechanisms underlying intestinal epithelial barrier-associated injury remain unclear. This study investigated whether endoplasmic reticulum stress (ERS) contributes to CPF-induced tight junction (TJ) alterations and intestinal epithelial injury. We used an 8-week drinking-water exposure model in weaned ICR mice with 30, 60, and 120 mg/L CPF and a MODE-K cell model, combined with pharmacological ERS modulation using 4-phenylbutyric acid (4-PBA) and tunicamycin (TM). Quantitative single-cell immunofluorescence was performed at 12, 24, and 48 h to assess temporal changes in TJ proteins and apoptosis-related markers. CPF induced jejunal villus atrophy, reduced the villus-to-crypt ratio, downregulated ZO-1, Occludin, and Claudin-1, impaired TJ structural integrity, increased the total protein levels of PERK, IRE1, and ATF6, and enhanced apoptosis-related alterations, including increased Bax and total Caspase-3 levels and decreased Bcl-2 levels. 4-PBA partially restored TJ protein expression and attenuated apoptosis-related alterations, whereas TM further aggravated TJ protein loss and apoptosis-related changes. Time-course analysis showed that TJ protein reductions were detectable before marked apoptosis-related changes and persisted at later time points. Collectively, chronic CPF exposure induced jejunal epithelial barrier-associated injury in weaned mice, accompanied by coordinated increases in the total protein levels of UPR sensors, TJ protein reductions, and enhanced apoptosis-related alterations. The opposing effects of 4-PBA and TM support the involvement of ERS in CPF-associated epithelial injury. These findings suggest that ERS contributes to progressive intestinal epithelial injury induced by CPF.

In-situ formation of metal-polyphenol networks in MXene membranes for efficient molecular/ion sieving

Qiankun Wu,Wentian Zhang,Cunxian Lai,Houlong Yang,Fangang Meng,Shanshan Zhao

This study proposes a bio-inspired in-situ cross-linking strategy to address the critical issue of swelling and structural instability in MXene two-dimensional (2D) lamellar membranes. By anchoring gallic acid (GA) onto MXene nanosheets via hydrogen bonding and subsequently coordinating cross-linking with Al 3+ ions, a robust three-dimensional GA-Al network is constructed between the nanosheets. This approach effectively transforms the inherently swellable, loosely stacked MXene assembly into a coordination-crosslinked reinforced lamellar architecture, successfully yielding structurally stable MXene/GA-Al composite membranes with excellent separation performance. The GA-Al network significantly suppressed interlayer swelling (the dry-wet d-spacing difference is only 1.2 Å), enabling precise sub-nanometer channel tuning, mitigating interlayer defects, and improving surface hydrophilicity. The optimized membrane achieved over 99% rejection of Congo red (CR), a pure water permeability of 120.0 L m −2  h −1 bar −1 , and outstanding separation factors of 1105 and 1403 for CR/NaCl and CR/Na 2 SO 4 mixed solutions, respectively. These results outperform most documented 2D lamellar and nanofiltration membranes. This work offers a new design paradigm for developing high-performance, anti-swelling 2D lamellar separation membranes.

Mobility Control Mechanism of In Situ Viscosity-Enhancing Graphene Quantum Dots in Assisted CO2 Flooding

Fang Shi,Weibin Jin,Jingchun Wu,Bo Zhao,Chunlong Zhang,Lifeng Mao

To address gas channeling, low sweep efficiency, and water sensitivity in CO2flooding of shale reservoirs, amidine-functionalized graphene quantum dots (FN-GQDs) were synthesized via amidation of citric acid-derived GQDs. FTIR and UV-Vis confirmed successful grafting. Conductometric titration showed an optimal reaction time of 24 h with a grafting ratio of 58%, in good agreement with the 60% saturation predicted by molecular dynamics simulation. Upon CO2introduction, protonation of amidine groups induced a nonlinear viscosity increase from 0.298 to 2.0 mPa·s at 0.02 wt% via electrostatic attraction and hydrogen bonding, forming a dynamic crosslinking network. FN-GQDs maintained low oil-water interfacial tension of 0.12–0.25 mN/m at 80–120 °C and rapidly reversed rock wettability from strongly oil-wet to water-wet, reducing the contact angle from 141.7° to 38.9° within 80 min. The positively charged surface inhibited clay swelling, achieving 92% at 0.20 wt%. Core flooding and NMR T2spectra revealed that alternating CO2and FN-GQDs injection at a 2:1 gas–water ratio achieved a final oil recovery of 52.5%, significantly higher than pure CO2flooding. Through synergistic effects of interfacial tension reduction, wettability alteration, viscosity enhancement, and anti-swelling, FN-GQDs improve microscopic displacement efficiency and macroscopic sweep volume, showing great potential for CO2-enhanced oil recovery in shale reservoirs.

Overcoming the Thermo-Optic Limit in Silicon Photonics through Ferroelastic Domain Reconfiguration in Barium Titanate

Hongguang Sun,Biaolin Peng,Kun Peng,Yi Chen,Ying Qiu,Min Wang,Kanghua Li,Zhonghui Shen,Tianwen Dong,Liang Liao,Ming Luo,Shujun Zhang,Xi Xiao,Qiuyun Fu,ZhongLin Wang

A novel ferroelastic domain reconfiguration mechanism delivers giant thermo-optic response in sol–gel barium titanate (BTO) thin films. • 750 °C-annealed BTO achieves record thermo-optic coefficient of 9.69×10⁻⁴ °C⁻¹, multiple times higher than silicon. • Fully reversible a-c domain switching is realized within the practical 25 °C-100 °C working window for on-chip photonic devices. • Prototype Fabry-Pérot modulator reaches 40% modulation depth for photonic integrated circuits and wavelength selective switching. • The domain engineering strategy is potentially extendable to CMOS-compatible ferroelectric perovskites. Thermo-optic tuning in ferroelectric perovskites has emerged as a promising strategy for achieving broadband, energy-efficient optical modulation. Nevertheless, the physical origins governing the near-room-temperature thermo-optic response of ferroelectrics are still not clearly defined. In this work, we report a previously unexplored thermo-optic mechanism in sol-gel-processed (001)-oriented BaTiO₃ films. This mechanism is driven by thermally induced, reversible a-c ferroelastic domain switching, activated through the synergistic control of interfacial screening, domain, and defect engineering, resulting in a large change in refractive index in the 25°C-100 °C temperature range. Films crystallized at 750 °C exhibit high tetragonal phase purity, strong c-axis alignment, and an exceptional thermo-optic coefficient of 9.69×10 -4 °C⁻¹—over several times larger than that of crystalline silicon—together with reversible domain evolution upon thermal cycling. A Fabry-Pérot thermo-optic modulator incorporating this film delivers 40% intensity modulation in the C-band. These findings establish a ferroelastic-domain-driven route to thermo-optic enhancement in ferroelectric perovskites and lay the groundwork for scalable. Deep MDRB: An integrated platform enabling high-coverage in vivo metabolite annotation in natural medicines

Jingjing Shi,Junyu Zhang,Xuting Zhou,Dandan Zhang,Hui Qi,Simian Chen,Yunlong Hou,Zhenhua Jia,Caisheng Wu

Comprehensive identification of in vivo metabolites from natural medicines (NMs) remains challenging due to their chemical complexity and the limited reference spectra. Here, we presented the deep mass dataset relevance bridging (Deep MDRB) platform, an integrated platform powered by a diagnostic weighted cosine similarity (DWCS) algorithm that efficiently facilitates high-coverage annotation of in vivo metabolites. By integrating mass spectral databases with biosynthetic and metabolic reaction rules, Deep MDRB enables fully automated non-targeted analysis, including peak extraction, background correction, multimodal data alignment, multilevel similarity matching, and parent–metabolite network bridging. As a proof of concept, Deep MDRB was applied to in vitro liver microsome models of verapamil and berberine, as well as an in vivo metabolic study involving mixed reference standards (MRS) and Tongxinluo (TXL) capsule in rats. The platform identified 1565 parent compounds with a 94.4% identification rate for the TXL capsule and constructed a biotransformation network of 29 in vitro , 83 (MRS) and 2069 (TXL capsule) in vivo metabolite nodes, respectively, with a correlation rate exceeding 90%. Compared to existing software, Deep MDRB substantially improves annotation coverage and network integrity. Overall, the “knowledge-data” dual-driven framework of Deep MDRB provides a novel strategy for metabolite profiling in complex systems.

Exploring protein digestion in water buffalo milk vs. holstein milk: An in vitro comparative study

Xiaobing Wang,Yun Gao,Hanyu Chai,Kai Yang,Jinsu Liao,Chao Zhang,Jianhuan Wei,Fazheng Ren,Pengjie Wang,Yi Wang

Buffalo milk caseins coagulated faster into larger, rougher curds in gastric phase. • Buffalo milk showed slower and more sustained protein hydrolysis in gastric phase. • Fewer peptide species with lower sequence coverage were generated from buffalo milk. • Clot structure during gastric digestion was the key factor affecting hydrolysis. This study compared the digestion behavior and peptide profiles of water buffalo and Holstein skim milk using the standardized INFOGEST in vitro protocol. During the gastric phase, protein hydrolysis and amino acid release demonstrated that buffalo milk proteins underwent a slower but more sustained digestion process than those of Holstein milk. SDS–PAGE analysis further confirmed the slower rate of protein hydrolysis in buffalo milk. Particle size distribution and confocal microscopy revealed that buffalo milk coagulated more rapidly into larger and more wrinkled coagulates than Holstein milk, which likely reduced pepsin accessibility and contributed to the slower protein hydrolysis. LC–MS peptide profiling further showed that buffalo milk generated fewer peptide species with lower protein sequence coverage following gastric digestion. In the intestinal phase, proteins from both milks were rapidly degraded, and species differences in protein hydrolysis behavior and peptide profiles became less pronounced. The role of endoplasmic reticulum stress-mediated autophagy in cadmium-induced liver injury in rats

Chengxiang Guo,Hao Ling,Junbing Mao,Jiale Dong,Cai Zhang,Yue Wang,Shuai Wang,Shuai Guo,Jicang Wang

Cadmium (Cd) is a widespread toxicant with high bioaccumulation potential. This study explores the interplay of endoplasmic reticulum stress (ERS), autophagy, and apoptosis in Cd-induced hepatotoxicity, focusing on whether ERS-driven autophagy protects against liver injury. Male SD rats (4 weeks old, n = 24) were acclimatized for 7 days and randomized into four groups receiving CdCl₂ at doses of 0, 0.5, 1, or 2 mg/kg for 14 consecutive days. A second cohort (n = 36) was similarly allocated to six groups: control, Cd, 4-PBA, Cd + 4-PBA, CQ and Cd + CQ. All treatments were administered via daily intraperitoneal injection throughout the study. On day 14, blood and liver tissues were collected for analyses of liver function, hematological parameters, and histopathology. The expression of target factors was analyzed via qRT-PCR and Western blotting. Results indicate that Cd exposure causes liver injury and disordered hepatocyte morphology. ERS markers Grp78 and Caspase-12 and autophagy-related factors Beclin-1, Atg5, P62 and LC3 are upregulated at both mRNA and protein levels. The endoplasmic reticulum-phagy (ER-phagy) receptor FAM134B and the apoptosis effector cleaved Caspase3 are upregulated at the protein level. These findings indicate that Cd induces ERS, UPR activation, autophagy, reticulophagy, and apoptosis. The ERS inhibitor 4-PBA markedly attenuated Cd-induced hepatic injury. Compared with the Cd group, the Cd + 4-PBA group showed decreased serum ALT and AST levels, as well as reduced RBC, WBC, MCH, and MCV counts. At the molecular level, mRNA and protein expression of ER stress markers Grp78, Caspase-12, PERK, eIF2α, ATF4, IRE1α, JNK, and ATF6 were all downregulated. Protein levels of the ER-phagy receptor FAM134B and the apoptosis effector cleaved Caspase-3 were also decreased. The autophagy inhibitor CQ aggravated such injury. Compared with the Cd group, the Cd + CQ group showed increased serum ALT and AST levels, as well as elevated RBC, MCH, and MCV counts. At the protein level, expression of the autophagy-related factors P62 and LC3, and the apoptosis effector cleaved Caspase-3 was significantly increased. These results show that Cd-induced ERS activates autophagy and reticulophagy mainly via the PERK, IRE1α, and ATF6 pathways, and this adaptive response clears autophagic substrates to alleviate hepatocyte damage.

Significant role of intermittent PMS backwash in mitigating irreversible fouling in MnTiO3 catalytic ceramic membranes

Guangxu Zhang,Jin Wang,Wubin Wang,Yadong Hu,Mengmeng Dou,Chao Han,Xinyu Li,Kaibo Wang,Yaxuan Liu,Yuting Ma

MnTiO 3 catalytic ceramic membrane (MTPCM) is prepared via solid particle impregnation- drawing- sintering method. • MTPCM 3 exhibits high BSA rejection primarily through dynamic cake layer formation and pore constriction. • Intermittent PMS backwash reduces chemical consumption while mitigating irreversible fouling. • MTPCM exhibits stable filtration performance over multi-cycle operation with PMS backwash. Ultrafiltration membrane's irreversible fouling-induced pore blockage and gel layer frequently reduce membrane lifespan. Peroxymonosulfate (PMS) can generate free radicals under appropriate catalysts, offering a promising strategy for membrane fouling control. This study developed a MnTiO 3 -modified catalytic ceramic ultrafiltration membrane (MTPCM) via an impregnation-drawing-sintering process. MTPCM achieved a BSA rejection >92%, attributed to dynamic cake layer formation and fouling-induced pore constriction. Higher membrane pressure, greater pollutant concentration, and longer filtration duration enhanced BSA removal by accelerating filter cake layer formation. Under optimized PMS backwash conditions (1 bar, 30 min, pH 7), the cleaning efficiency reached ∼60% after five cycles, approximately 2.3 times higher than that of water backwash (∼26%). Furthermore, an intermittent backwash strategy (four water backwashes followed by one PMS backwash) was evaluated: the MTPCM maintained ∼50% of its initial flux after 15 h of operation (three cycles), demonstrating stable multi-cycle performance with reduced PMS consumption. Hermia membrane fouling model analysis suggests that PMS backwash plays an important role in reducing irreversible fouling and may contribute to maintaining membrane performance during cyclic operation.

Nanocrystal maturation in doxorubicin liposomes determines functional performance: real-time insights from in situ SAXS

Yiwen Li,Fulin Ye,Lei Qu,Jialin Duan,Liangliang Kong,Guangfeng Liu,Chunming Yang,Na Li,Xiaohui Wei

The clinical success of PEGylated liposomal doxorubicin (DOX) is fundamentally attributed to its unique internal nanocrystalline structure. However, a notable discrepancy exists between the rapid drug uptake predicted by loading kinetics and the extended incubation times employed in actual manufacturing. By integrating in situ time-resolved synchrotron small-angle X-ray scattering (SAXS), cryogenic transmission electron microscopy (cryo-TEM), and in vitro release kinetics analysis, this study systematically elucidates the multi-step structural evolution and formation mechanism of DOX-sulfate nanocrystals during remote loading via the ammonium sulfate gradient method, along with its correlation with the final functional performance of the formulation. Our results demonstrate that the rapid drug uptake and achievement of target encapsulation efficiency (EE%) do not necessarily indicate the formation of a mature, stable nanocrystalline structure. Instead, during a critical post-loading maturation stage, the intraliposomal nanocrystals undergo gradual lattice optimization into a thermodynamically stable structure. Liposomes containing “sub-mature” aggregates exhibit pronounced burst leakage, whereas those with fully matured nanocrystals ensure near-zero-order release kinetics and superior stability. These observations provide useful insights that may inform future process optimization and offer a potential basis for structural quality control of liposomal nanodrugs. In situ Photocrosslinked Fluorinated Polyacrylate Layer for Interfacial Modification in Carbon-Based Perovskite Solar Cells

Li Jialiang,Geng Mengqi,Jiang Le,Zhao Yongsheng,Chen Jianxin,Lin Yiting,Duan Zhaoxuan,Gou Hailing,Xu Tingting

Carbon-based perovskite solar cells (C-PSCs) have attracted considerable attention owing to their low cost and excellent commercial potential. However, insufficient interfacial contact and unfavorable energy-level alignment at the perovskite/carbon interface often lead to severe nonradiative recombination and inefficient hole extraction, thus limiting device performance. Herein, two photocurable polyacrylates, poly(isopropyl acrylate) (poly(IPAc)) and fluorinated poly(1,1,1,3,3,3-hexafluoroisopropyl acrylate) (poly(HFIPAc)), are in situ polymerized as interfacial layers to investigate the molecular structure effect on the perovskite/carbon interface. A comparative study reveals that the poly(HFIPAc) layer with the electron-withdrawing effect of fluorine substituents strengthens polymer/carbon interactions. Compared with the poly(IPAc) interlayer, the as-formed poly(HFIPAc) layer exhibits better ability to enhance film crystallinity, reduce defect density, and optimize surface electronic structure and improve energy-level alignment at the interface of the perovskite and the carbon electrode. Consequently, the optimized carbon-based perovskite solar cell incorporating the poly(HFIPAc) interfacial layer achieves the champion power conversion efficiency (PCE) of 18.44%, significantly higher than that of the poly(IPAc)-treated PSCs. Meanwhile, the unencapsulated poly(HFIPAc)-treated device retains 90.5% of its initial efficiency after 1000 h of storage under ambient conditions. This work provides valuable insights on how molecular structure of polymers regulates the perovskite/carbon interface. It also provides a sustainable route toward cost-effective and stable carbon-based perovskite solar cells, taking advantage of the room-temperature UV-induced photopolymerization method and the HTL-free carbon-electrode architecture.
