Prussian blue and collagen loaded chitosan nanofibers with NIR-controlled NO release and photothermal activities for wound healing

Wenyu Wang,Dejun Ding,Kunpeng Zhou,Man Zhang,Weifen Zhang,Fang Yan,Ni Cheng

Nitric oxide (NO) has emerged as a potential wound therapeutic agent due to its pivotal role in the wound healing processes. Nevertheless, NO-based therapy for clinical applications is still restricted due to its gaseous state and short half-life. Here we exploited a wound dressing by incorporating sodium nitroprusside doped prussian blue nanoparticals and Type I collagen into the chitosan/poly(vinyl alcohol) nanofibers through the electrospinning method. This hybrid nanofibrous scaffold possess the excellent abilities of NIR controlled NO release, photothermal therapy, and imitation of extra-cellular matrix-like architecture. These synergistic effects could enhance their anti-bactericidal effects in vitro and furthermore accelerate wound healing in vivo when compared to control groups. Histological analysis demonstrated the scaffold could promote fibroblast growth and accelerate epithelialization. Moreover, no apparent histological toxicology and negligible damage to major organs were observed, which provided sufficient biosafety for in vivo application. These data indicate the fabricated hybrid nanofibrous scaffold could be used as an ideal candidate for accelerating wound healing and treating chronic wounds.

A Targeted Activatable NIR-IIb Nanoprobe for Highly Sensitive Detection of Ischemic Stroke in a Photothrombotic Stroke Model

Xiaohu Yang,Zan Wang,Haoying Huang,Sisi Ling,Rong Zhang,Yejun Zhang,Guangcun Chen,Chunyan Li,Qiangbin Wang

Ischemic stroke is a devastating disease resulting in high morbidity and mortality. To date, its early diagnosis still faces challenges. Herein, an efficient detection strategy is proposed, in which a targeted activatable NIR-IIb nanoprobe (V&C/PbS@Ag 2 Se) is constructed for in vivo highly sensitive detection of early ischemic stroke in a photothrombotic stroke model. At first, the fluorescence of V&C/PbS@Ag 2 Se displays an “off” state due to the competitive absorption of excitation irradiation between Cy7.5 fluorophores and PbS@Ag 2 Se quantum dots (QDs). Upon intravenous injection, the V&C/PbS@Ag 2 Se quickly accumulates in the lesion regions based on VCAM1 binding peptide target to the inflamed vascular endothelium of ischemic stroke. Later, the nanoprobes can be rapidly activated via Cy7.5 oxidation by peroxynitrite (ONOO − ), the prodromal biomarker of ischemic stroke, instantly illuminating the lesion regions. Such a targeted activatable strategy offers a favorable approach for in vivo early real-time assessment of ischemic stroke, which can be expanded to other diseases as a general mothed for in vivo precise diagnosis.

A novel AIE active NIR fluorophore based triphenylamine for sensing of Hg2+ and CN− and its multiple application

Xiaoye Wen,Li Yan,Zhefeng Fan

New strategies still need to be proposed that can be used to sense and remove toxic environmental pollutants in a sensing system. In this research, a novel NIR fluorescence sensor 1 was designed and prepared with aggregation induced emission (AIE) property. The fluorescence intensity of the sensor 1 in DMSO/H 2 O mixed solvent was changed along with the proportion of water. The sensor 1 can be successfully used for real-time detection and removal of Hg 2+ in 20% DMSO aqueous solution with high selectivity, quick response and so on. Furthermore it can be efficiently reused and recycled without any loss through Na 2 S. In addition, the sensor 1 displayed high sensitivity and selectivity to cyanide ions in 1% DMSO aqueous solution with the presence of other interference anions. The sensing mechanism for Hg 2+ and cyanide ion was evaluated by 1 H NMR spectra, Mass spectrometry. The sensor 1 exhibited low cytotoxicity for biological applications, which was used as an outstanding fluorescent transducer for detection of cyanide ion in living cells. Based on the visible fluorescence change for the sensor 1 to cyanide ion, the measurement was performed for food materials containing cyanide, such as potato, cassava powder and almond. This research provides perspective potential in solving the problem of other pollution and stimulating new thinking for designing of the novel fluorescence materials.

X-ray and NIR light dual-triggered mesoporous upconversion nanophosphor/Bi heterojunction radiosensitizer for highly efficient tumor ablation

Jie Liu,Jing Zhang,Fei Huang,Yong Deng,Bing Li,Ruizhuo Ouyang,Yuqing Miao,Yun Sun,Yuhao Li

ABSTRACT Developing a multi-functional radiosensitizer with high efficiency and low toxicity remains challenging. Herein, we report a mesoporous heterostructure radiosensitizer ( [email protected] ) containing Lu-based upconversion nanophosphor (UCNP) and Bi-based nanomaterial loaded with iron phthalocyanine for X-ray and NIR light dual-triggered tri-modal tumor therapy. NaLuF 4 :Yb,Tm, a Lu-based UCNP, offers radiosensitization and upconversion luminescence for optical bio-imaging. However, Bi has a higher X-ray mass attenuation coefficient than Lu . Thus, after stepwise fabrication, Na 0.2 Bi 0.8 O 0.35 F 1.91 :Yb (NBOF) was assembled with the UCNP to form a mesoporous heterostructure composite. This enhanced the radiosensitization effect and drug load to realize multi-modal tumor therapy. After coating it with folate-conjugated amphiphilic PEG (PFA), [email protected] realized tumor photothermal/photodynamic/radio-therapy. The structure of [email protected] was well characterized. Different properties triggered by X-ray and NIR light were evaluated. Finally, a highly efficient tumor ablation effect was demonstrated in vitro and in vivo . Consequently, this kind of nanocomposite provides a unique strategy for designing a theranostic platform for oncotherapy. Statement of Significance The synergy of enhanced radiotherapy and photothermal/photodynamic therapy is found to improve tumor therapeutic efficacy. On that basis, a heterostructure nanohybrid containing Lu-based UCNP and Bi-based mesoporous material is synthesized. The heterostructure nanohybrid can be loaded with FePc and decorated with folate-modified amphiphilic PEG to form a multi-functional theranostic nano-platform. The platform exhibits upconversion luminescence capacity, X-ray attenuation property, photothermal effect, and X-ray and NIR dual-light triggered ROS generation capability. These features can not only enable upconversion luminescence/CT bioimaging of living beings but also be applied to the photothermal/photodynamic/radio- synergistic tumor ablation. To sum up, the nanomaterial offers a novel method for the construction of a new theranostic platform.

NIR-II Responsive Hollow Magnetite Nanoclusters for Targeted Magnetic Resonance Imaging-Guided Photothermal/Chemo-Therapy and Chemodynamic Therapy

Xingyu Wang,Changwei Li,Junchao Qian,Xiaotong Lv,Hong Li,Jinglu Zou,Jiahui Zhang,Xiangfu Meng,Hongji Liu,Yong Qian,Wenchu Lin,Hui Wang

Phototherapy in the second near-IR (1000–1700 nm, NIR-II) window has achieved much progress because of its high efficiency and relatively minor side effects. In this paper, a new NIR-II responsive hollow magnetite nanocluster (HMNC) for targeted and imaging-guided cancer therapy is reported. The HMNC not only provides a hollow cavity for drug loading but also serves as a contrast agent for tumor-targeted magnetic resonance imaging. The acid-induced dissolution of the HMNCs can trigger a pH-responsive drug release for chemotherapy and catalyze the hydroxyl radical (·OH) formation from the decomposition of hydrogen peroxide for chemodynamic therapy. Moreover, the HMNCs can adsorb and convert NIR-II light into local heat (photothermal conversion efficacy: 36.3%), which can accelerate drug release and enhance the synergistic effect of chemo-photothermal therapy. The HMNCs show great potential as a versatile nanoplatform for targeted imaging-guided trimodal cancer therapy.

NaGdF4:Nd@NaGdF4 Core-Shell Down-Conversion Nanoparticles as NIR-II Fluorescent Probes for Targeted Imaging of Bacteria

Leilei Sun,Sisi Shi,Hongchao Geng,Yao Huang,Yan Qiao,Jie Song,Lan Yang,Craig A. Grimes,Xinxin Feng,Qingyun Cai

Bacterial infections are a serious ongoing threat to human health. As such, within the context of modern technology, there is an urgent need to develop fluorescent probes suitable for diagnosing bacterial infections in vivo. Herein, we synthesize NaGdF4:Nd@NaGdF4 core-shell down-conversion nanoparticles (DCNPs) demonstrating excellent emission intensity in the second near-infrared (NIR-II) window for use as fluorescent probes to enable high-quality imaging of pathogenic infections. The fluorescent probe is covalently linked with vancomycin (Van), which specifically binds to Gram-positive bacteria such as S. aureus. Both in vitro detection and in vivo imaging results show that the Van-modified NIR-II fluorescent probe can distinguish Gram-positive from Gram-negative bacteria with a high degree of selectivity. Biocompatibility is verified by MTT assay and histological analyses.

NIR-Triggered Multifunctional and Degradable Nanoplatform Based on an ROS-Sensitive Block Copolymer for Imaging-Guided Chemo-Phototherapy

Chao Wang,Baoxuan Huang,Guoliang Yang,Yingjie Ouyang,Jia Tian,Weian Zhang

Imaging-guided chemo-phototherapy based on multifunctional nanocarriers has emerged as a promising and high-efficient cancer treatment because of the inevitable limitations of single therapy. Herein, a near-infrared (NIR) light-activated degradable polymeric nanoplatform was fabricated for chemo-phototherapy. An NIR photosensitizer, IR780, and a chemotherapeutic drug, doxorubicin (DOX), were efficiently coloaded within a reactive oxygen species (ROS)-sensitive polymeric micelle based on an amphiphilic copolymer with degradable thioketal (TK) linkages. The obtained spherical nanoparticles (denoted as (IR780/DOX)@PTK) exhibited a notable photodynamic and photothermal effect upon NIR light exposure. Furthermore, due to the rapid cleavage of TK linkers induced by ROS generated from NIR-activated IR780, (IR780/DOX)@PTK also showed an NIR light-induced degradable feature, which can be used for light-triggered tumor-specific drug release and lead to ignorable systematic toxicity after biodegradation and drug delivery. Under the guidance of NIR fluorescence and photothermal dual modal imaging, (IR780/DOX)@PTK exhibited excellent tumor accumulation after intravenously injection into 4T1-tumor-bearing mice. As verified in both in vitro and in vivo study, (IR780/DOX)@PTK presented a significant tumor suppression effect by synergistic chemo-phototherapy.

Triple stimuli-responsive ZnO quantum dots-conjugated hollow mesoporous carbon nanoplatform for NIR-induced dual model antitumor therapy

Shuang Feng,Yuling Mao,Xiudan Wang,Meiting Zhou,Hongyan Lu,Qinfu Zhao,Siling Wang

Aiming at the inefficiency and toxicity in traditional antitumor therapy, a novel multifunctional nanoplatform was constructed based on hollow mesoporous carbon (HMC) to achieve triple stimuli response and dual model antitumor therapy via chemo-photothermal synergistic effect. HMC was used as an ideal nanovehicle with a high drug loading efficiency as well as a near-infrared (NIR) photothermal conversion agent for photothermal therapy. Acid-dissoluble, luminescent ZnO quantum dots (QDs) were used as the proper sealing agents for the mesopores of HMC, conjugated to HMC via disulfide linkage to prevent drug (doxorubicin, abbreviated as Dox) premature release from Dox/HMC-SS-ZnO. After cellular endocytosis, the Dox was released in a pH, GSH and NIR laser triple stimuli-responsive manner to realize accurate drug delivery. Moreover, the local hyperthermia effect induced by NIR irradiation could promote the drug release, enhance cell sensitivity to chemotherapeutic agents, and also directly kill cancer cells. As expected, Dox/HMC-SS-ZnO exhibited a high drug loading capacity of 43%, well response to triple stimuli and excellent photothermal conversion efficiency η of 29.7%. The therapeutic efficacy in 4T1 cells and multicellular tumor spheroids (MCTSs) demonstrated that Dox/HMC-SS-ZnO + NIR had satisfactory chemo-photothermal synergistic effect with a combination index (CI) of 0.532. The cell apoptosis rate of the combined treatment group was more than 95%. The biodistribution and pharmacodynamics studies showed its biosecurity to normal tissues and synergistic inhibition effect to tumor cells. These distinguished results indicated that the Dox/HMC-SS-ZnO nanoplatform is potential to realize efficient triple stimuli-responsive drug delivery and dual model chemo-photothermal synergistic antitumor therapy.

Cell Membrane-Camouflaged NIR II Fluorescent Ag2Te Quantum Dots-Based Nanobioprobes for Enhanced In Vivo Homotypic Tumor Imaging

Jing-Jing Zhang,Yi Lin,Hui Zhou,He He,Jiao-Jiao Ma,Meng-Yao Luo,Zhi-Ling Zhang,Dai-Wen Pang

The advantages of fluorescence bioimaging in the second near-infrared (NIR II, 1000–1700 nm) window are well known; however, current NIR II fluorescent probes for in vivo tumor imaging still have many shortcomings, such as low fluorescence efficiency, unstable performance under in vivo environments, and inefficient enrichment at tumor sites. In this study, Ag 2 Te quantum dots (QDs) that emit light at a wavelength of 1300 nm are assembled with poly(lactic- co -glycolic acid) and further encapsulated within cancer cell membranes to overcome the shortcomings mentioned above. The as-prepared ≈100 nm biomimetic nanobioprobes exhibit ultrabright (≈60 times greater than that of free Ag 2 Te QDs) and highly stable (≈97% maintenance after laser radiation for 1 h) fluorescence in the NIR II window. By combining the active homotypic tumor targeting capability derived from the source cell membrane with the passive enhanced permeation and retention effect, improved accumulation at tumor sites ((31 ± 2)% injection dose per gram of tumor) and a high tumor-to-normal tissue ratio (13.3 ± 0.7) are achieved. In summary, a new biomimetic NIR II fluorescent nanobioprobe with ultrabright and stable fluorescence, homotypic targeting and good biocompatibility for enhanced in vivo tumor imaging is developed in this study.

Green to red and NIR tunable luminescence in heavily Er3+-doped tellurite glasses via selective energy transfer mechanism

Weirong Wang,Guanliang Yu,Ruibin Wang,Chun Jiang

Here, we select simple Er 3+ -doped tellurite glass as model system to systematically explore the up-conversion, down-shifting mechanisms with different excitations (980 and 447 nm), respectively. We observe for the first time, to the best of our knowledge that tunable photo-luminescence occurs from green to red and NIR region, rather than merely from the long-accepted green to red region. Direct evidence of selective energy transfer mechanism is expounded in detail, and its potential applications are demonstrated. In addition, we provide evidence that the cross-relaxation process between dopant ions can enhance photo-luminescence in Er 3+ doped tellurite glasses with high dopant concentrations, whereas the crucial reason for emission decrease is the energy loss long-distance energy migration. These fundamental insights into the photophysical processes in heavily doped photonic glasses will broaden the applications of rare-earth-doped materials ranging from optical communications to medical imaging.

A poly (vinyl butyral)/graphene oxide composite with NIR light-induced shape memory effect and solid-state plasticity

Yongkang Bai,Jiwen Zhang,Didi Wen,Peiwei Gong,Xin Chen

A NIR light-induced shape memory composite with light-induced plasticity was prepared by incorporating graphene oxide (GO) into cross-linked poly (vinyl butyral) (PVB). The cross-linked reaction between PVB and aromatic diisocyanate not only endowed the composites with excellent shape memory properties, but also offered the performance of solid-state plasticity due to the formation of carbamate bonds. The photo-responsive performance was introduced to the system by doping GO, a conventional photothermal reagent, resulting in excellent NIR light-induced shape memory properties and light-induced plasticity. According to the stress relaxation tests, the solid-state plasticity of composites could be regulated by the cross-linked density, GO, catalyst dosage and temperature. Moreover, the composites can be repeatedly programmed to a new permanent shape via a light-induced plasticity process, which still exhibited excellent light-induced shape memory properties even after 5 cycles of reconfiguration. The results demonstrate the promising prospect of these composites as actuator elements for applications in self-deployment devices and soft robotic.

Supramolecular PEGylated Dendritic Systems as pH/Redox Dual-Responsive Theranostic Nanoplatforms for Platinum Drug Delivery and NIR Imaging

Yunkun Li,Yachao Li,Xiao Zhang,Xianghui Xu,Zhijun Zhang,Cheng Hu,Yiyan He,Zhongwei Gu

Recently, self-assembling small dendrimers into supramolecular dendritic systems offers an alternative strategy to develop multifunctional nanoplatforms for biomedical applications. We herein report a dual-responsive supramolecular PEGylated dendritic system for efficient platinum-based drug delivery and near-infrared (NIR) tracking. With a refined molecular/supramolecular engineering, supramolecular dendritic systems were stabilized by bioreducible disulfide bonds and endowed with NIR fluorescence probes, and PEGylated platinum derivatives coordinated onto the abundant peripheral groups of supramolecular dendritic templates to generate pH/redox dual-responsive theranostic supramolecular PEGylated dendritic systems (TSPDSs). TSPDSs markedly improved the pharmacokinetics and biodistribution of platinum-based drugs, owing to their stable nanostructures and PEGylated shells during the blood circulation. Tumor intracellular environment (low pH value and high glutathione concentration) could trigger the rapid disintegration of TSPDSs due to acid-labile coordination bonds and redox-cleavable disulfide linkages, and then platinum-based drugs were delivered into the nuclei to exert antitumor activity. In vivo antitumor treatments indicated TSPDSs not only provided high antitumor efficiency which was comparable to clinical cisplatin, but also reduced renal toxicity of platinum-based drugs. Moreover, NIR fluorescence of TSPDSs successfully visualized in vitro and in vivo fate of nanoplatforms and disclosed the intracellular platinum delivery and pharmacokinetics. These results confirm tailor-made supramolecular dendritic system with sophisticated nanostructure and excellent performance is a promising candidate as smart theranostic nanoplatforms.
