Multifunctional curcumin-loaded soybean oil-based waterborne polyurethane films with the outstanding barrier, antioxidant and antibacterial properties for active fruit packaging

Qiongrui Jiang,Junxian Guo,Xiaoxian Wu,Xiyan Shen,Xiaoyun Li,Chengjun Fang,Zhuohong Yang,Zhuangzhuang Chu,Yang Hu

Bio-based waterborne polyurethane (WPU) films have attracted much attention to replacing plastic films in food packaging fields. Despite recent clear progress in constructing bio-based WPU films, it is still a challenge to fulfill suitable mechanical property, high water and gas-resistant behavior, outstanding UV-blocking performance, favourable antioxidant and antibacterial activities at the same time in a bio-based WPU film. Herein, curcumin (Cur)-loaded soybean oil-derived WPU multifunctional films are developed by an in-situ emulsion encapsulation method. The results indicate that Cur is homogeneously embedded in WPU film matrix, and interacts with WPU chain through hydrogen bonds to compactify film structure. And the incorporation of Cur can significantly enhance the overall performances of WPU films. The optimal WPU film displays favourable mechanical property with the tensile strength of 10.95 MPa and strain at break of 1511.02 %, high water barrier performance with water contact angle of 100.9° and water vapor permeability rate of 0.77 × 10 −10 g·m −1 ·s −1 ·Pa −1 , favorable gas barrier behavior with the oxygen and carbon dioxide permeabilities of 1.24 × 10 −10 g·m⁻¹ ·s⁻¹ ·Pa⁻¹ and 2.49 × 10 −10 g·m⁻¹ ·s⁻¹ ·Pa⁻¹ , and the nearly complete UV-blocking capability. Furthermore, the optimal WPU film demonstrates remarkable antioxidant activity with ABTS radical scavenging rate as high as 98.02 %, clear antibacterial ability with the inhibition rates of 89.35 % against E. coli and 98.33 % against S. aureus . Interestingly, the results of litchi preservation tests imply that Cur-loaded WPU films effectively delay litchis deterioration and extend litchis shelf life. This study provides a robust strategy for designing high-performance, bio-based active packaging films for fruit preservation application fields.

Self-Assembled Curcumin–Epigallocatechin Gallate Nanoparticles Enhance Antiviral Activity Against White Spot Syndrome Virus Through Immunomodulation, Antioxidant Effects, and Metabolic Regulation

Songlin Luo,Xudong Yang,Shunqiang Wei,Chushun Yi,Fansheng Zeng,Fali Zhang,Wei Zhang,Xiuli Chen,Yongzhen Zhao,Yinghui Wang,Aiguo Huang

White Spot Syndrome Virus (WSSV) remains a major factor limiting the sustainable development of crustacean aquaculture, creating an urgent need to develop comprehensive antiviral strategies with multiple protective functions. Natural bioactive compounds with antiviral, immunomodulatory, and antioxidant activities hold great potential for application; however, their poor stability and low bioavailability limit their practical use. In this study, self-assembled nanosystem (CE SA) composed of epigallocatechin gallate (EGCG) and curcumin (Cur) was constructed based on noncovalent interactions, and its protective effects against WSSV-infectedProcambarus clarkiiwere systematically evaluated. The results indicate that CE SA exhibits good stability, sustained-release properties, and biosafety. Compared with Cur, EGCG, and their physical mixture (CE), CE SA significantly improved the survival rate of infected shrimp and reduced viral copy numbers as well as the expression levels of key viral genes such asIE1,DNApol, andVP28. At the same time, CE SA significantly promoted the expression of relevant genes such asDorsal,Relish,SOD, andCAT, increased SOD and CAT enzyme activities and hematopoietic cell viability, and effectively alleviated damage to the hepatopancreas and intestinal tissues. Furthermore, metabolomic analysis indicated that CE SA regulates key metabolic processes such as amino acid metabolism, inflammation-related metabolism, nucleotide metabolism, and redox homeostasis, further elucidating its potential mechanism for enhancing the host’s antiviral capacity. In summary, by enhancing the stability and utilization efficiency of natural bioactive components, CE SA achieves superior anti-WSSV efficacy compared to individual components. Its protective effects are closely related to the inhibition of viral replication, enhancement of the immune response, improvement of antioxidant defenses, and regulation of host metabolic homeostasis. This study provides a promising, self-assembling nanostructure strategy for the green prevention and control of viral diseases in aquatic animals.

A novel edible, all-biomass Pickering emulsion coating based on Lys/Alg composite nanoparticles for curcumin delivery and active fruit preservation

Fengqiong Jiang,Chaoran Fan,Mingfei Dang,Fuxiang Wei,Lu Liang,Lihua Fu,Chuanhui Xu,Baofeng Lin

A stable food-grade amphiphilic Lys/Alg nanoparticle emulsifier was prepared by double crosslinking. • An Edible, all-biomass-derived multifunctional Pickering emulsion was successfully developed via simple emulsification. • The Lys/Alg served as effective Pickering stabilizer enhanced the stability and bioavailability of curcumin. • The developed emulsion exhibited high antioxidant properties (99.02%) and broad-spectrum antibacterial activity. • The shelf life of various fruits was effectively extended by the emulsion coating. The integration of nano-encapsulation with edible coatings holds great potential for active fruit preservation. However, conventional emulsions often rely on chemical surfactants, posing safety risks and limited functionality. Here, we developed an edible, all-biomass-derived Pickering emulsion (CLAPE) coating stabilized by amphiphilic lysozyme/alginate composite nanoparticles (Lys/Alg). The Lys/Alg were fabricated through electrostatic self-assembly followed by Ca 2+ cross-linking, forming a robust interfacial layer that effectively encapsulates curcumin (Cur). Molecular docking simulations revealed a strong binding affinity between Lys and Cur (binding energy: −87.2 kcal/mol), which significantly enhances the aqueous dispersibility and stability of Cur. The resulting Pickering emulsion enables sustained release of curcumin over 60 h, with a peak cumulative release of 94.0%, approximately twice that of the unencapsulated control. This release profile follows Fickian diffusion, extending the bioactive duration and increasing the total released amount, thereby enhancing curcumin's bioavailability. Moreover, CLAPE exhibits excellent thermal stability, maintaining high stability indices under varied accelerated storage conditions. It also exhibited high antioxidant (99.02%) and broad-spectrum antibacterial activity. Additionally, the application of CLAPE on strawberries, bananas and grapes has led to a doubling of their shelf life while maintaining the freshness and nutrient content of the fruits. The edible active preservative material, based entirely on biomass, not only forms a multifunctional protective coating that prevents spoilage but can also be easily washed off. This study presents an innovative strategy for the designs of active preservation materials utilizing all-natural plant-derived components. Yaozhi He,Siyuan Zou,Jiawei Wang,Wenbin Zhang,Sheng Lu,Mengxian Jia,Yumin Wu,Xiaowu Lin,Ziwei Fan,Qishun Liang,Yizhe Sheng,Qichuan Zhuge,Bi Chen,Minyu Zhu,Honglin Teng

Spinal cord injury (SCI) causes devastating neurological deficits, and cellular senescence critically contributes to the pathogenesis of various diseases. Notably, endothelial cells (ECs) senescence exerts a pivotal effect on the pathogenesis following SCI. In this study, we found that the number of senescent ECs increased by 18.87% ± 5.91%, and the disruption of the blood-spinal cord barrier (BSCB) was aggravated with an 18% increase in Evans Blue dye extravasation in diabetic mice with spinal cord injury (DM-SCI). To address this pathological process, a bioinspired nanotherapeutic platform utilizing milk-derived small extracellular vesicles with platelet membrane fusion (PM-sEVs) was developed for the targeted delivery of curcumin (Cur). In vitro , PM-sEVs-Cur effectively mitigated HG/IL-1β-induced HUVECs senescence by 54.19% ± 5.39% and increased expression of the tight junction protein ZO-1 by 4.33-fold. Mechanistically, Cur attenuated HUVECs senescence by activating the NRF2/HO-1 pathway. In vivo , platelet membrane modification enhanced the lesion targeting of sEVs. Treatment with PM-sEVs-Cur attenuated ECs senescence by 34.96% ± 6.59%, preserved BSCB integrity with a 17% reduction in Evans Blue dye extravasation, promoted axonal regeneration with a 6.36-fold increase in neurofilament expression, and improved motor function recovery with an increase of 2.4 ± 0.55 points in Basso Mouse Scale score in DM-SCI. This study highlights PM-sEVs-Cur as a promising therapeutic delivery platform for DM-SCI treatment.

Methoxy polyethylene glycol amine-modified curcumin derivative: Synthesis, photophysical properties, and photodynamic inactivation preservation on peeled Pacific white shrimp (Litopenaeus vannamei)

Ran Wei,Shuyi Zhao,Chuandong Fang,Long Jiao,Xiaoye Zhang,Xiang Chen,Soottawat Benjakul,Bin Zhang

To improve the water solubility and photodynamic inactivation (PDI) activity of curcumin (Cur), a novel methoxy polyethylene glycol amine (mPEG-NH 2 )-modified Cur derivative, mPEG-NH-Cur, was synthesized and characterized. The results showed that mPEG-NH-Cur had extremely high water solubility with a saturated solubility of 6.71 mg/mL, which was 15-fold that of Cur. Theoretical calculations revealed that the introduction of PEG markedly enhanced hydrogen bonding, electrostatic interactions, and van der Waals interactions between mPEG-NH-Cur and H 2 O while simultaneously reducing the energy gap to its excited state. These effects contributed to its enhanced stability and resistance to aggregation in aqueous solutions, thereby significantly improving its capacity for singlet oxygen ( 1 O 2 ) generation (achieving 100 % degradation of 1,3-diphenylisobenzofuran within 30 s). In the PDI preservation experiments, the peeled Pacific white shrimp ( Litopenaeus vannamei ) treated with mPEG-NH-Cur showed the greatest sensory acceptability after eight days of storage at 4 °C compared to the control group. Moreover, the inhibitory effects of mPEG-NH-Cur-mediated PDI treatment on the total volatile basic nitrogen content, pH, thiobarbituric acid value, and total viable count were significantly higher than those of the control group. Ultimately, the use of mPEG-NH-Cur increased the refrigerated shelf life of the peeled L . vannamei by 8 days. Further, the weak interaction analysis demonstrated that PEG effectively suppressed molecular aggregation through enhanced hydrogen bonding and steric hindrance over an extended duration (40,000 ps), thereby improving aqueous solubility and dispersion stability. Overall, the findings of this study provide novel insights into the potential application of Cur derivatives in PDI preservation.

Gengsheng Shi,Wenqin Yin,Jing Luo,Song Lin,Linluo Zhang,Yuan He,Hui Ding

This study developed transferrin (Tf) -functionalized magnetic nanoparticles (Tf-MNP-PEG-PLGA-Cur) to enhance curcumin (Cur)’s therapeutic potential for lung cancer. Tf-MNP-PEG-PLGA-Cur was synthesized by chemically coupling Tf (tumor-targeting ligand) to the surface of MNP-PEG-PLGA-Cur. Its size and morphology were measured, in vitro drug release was evaluated via dialysis, oral bioavailability and tissue distribution were studied in rats and mice. Cytotoxicity to normal cells was tested by CCK-8, A549 cell uptake was observed by confocal laser microscopy, and in vitro anti-lung cancer activity was studied using MTT assay. In vivo, its distribution in tumor-bearing nude mice was detected by near-infrared imaging, followed by measuring tumor growth and histopathological observation. Tf-MNP-PEG-PLGA-Cur nanoparticles had a small size (137.39 ± 3.72   nm, PDI 0.224 ± 0.010) with a biphasic release pattern, longer half-life (P<0.05), and 186% relative oral bioavailability. It promoted Cur accumulation in tissues, especially liver and spleen. Both Tf-MNP-PEG-PLGA-Cur and Cur showed good cytocompatibility. Tf-MNP-PEG-PLGA-Cur enhanced curcumin's anti-lung cancer activity in vitro and in vivo, indicating good tumor-targeting. H&E staining showed it protected nude mice's vital organs and inhibited tumor growth. Tf-MNP-PEG-PLGA-Cur is a promising polymeric sustained-release curcumin preparation, improving oral bioavailability and anti-lung cancer activity.

Effects of colloidal delivery systems for curcumin-Brassica rapa L. polysaccharide mixture encapsulation on physicochemical properties, stability, and gut microbiota modulation

Dongdong Guo,Wenlida Mu,Chang Liu,He Qian

Three colloidal systems (LP-CP, SC-CP, CYC-CP) were systematically compared for encapsulating curcumin- Brassica rapa L. polysaccharide mixture (CP). • SC-CP exhibited the highest curcumin loading capacity, superior curcumin thermal stability, and peak colonic accumulation of curcumin in vivo . • SC-CP significantly promoted the production of acetate, propionate, and butyrate during in vitro colonic fermentation, outperforming LP-CP and CYC-CP. • In a mouse model, SC-CP enriched beneficial genera, including Lachnospiraceae_NK4A136_group and Akkermansia and increased cecal butyrate levels. • The choice of colloidal carrier fundamentally governs the colonic delivery efficiency and gut microbiota remodeling capacity of the curcumin-polysaccharide mixture. The therapeutic potential of curcumin is severely constrained by its poor physicochemical stability and low oral bioavailability. Co-formulation with Brassica rapa L. polysaccharide offers a promising strategy to enhance its functionality; however, the impact of different colloidal delivery systems on the encapsulation performance and subsequent biological fate of this mixture remains unclear. In this study, we systematically compared three spray-dried delivery platforms, including liposomes (LP-CP), sodium caseinate nanoparticles (SC-CP), and β-cyclodextrin inclusion complexes (CYC-CP), for encapsulating CP. Our results demonstrated that the carrier system critically determined encapsulation performance, with SC-CP exhibiting superior curcumin loading capacity (7.24%), curcumin thermal stability (82.87% retention at 95 °C), and favorable hygroscopicity profiles. Notably, SC-CP facilitated enhanced colonic accumulation in vivo , achieving a peak accumulation of 53.45% at 8 h post-gavage, representing a 15-fold increase compared to curcumin from unencapsulated CP. By integrating in vitro fermentation models with in vivo animal experiments and employing 16S rRNA sequencing alongside short-chain fatty acid (SCFA) analysis, we systematically elucidated the carrier-specific modulatory effects on the gut microbiota. In the in vitro fermentation system, SC-CP significantly promoted the production of acetate, propionate, and butyrate, while enriching butyrate-producing genera such as Lachnospiraceae_NK4A136_group . In the in vivo animal model, SC-CP intervention resulted in a 1.6-fold increase in cecal butyrate levels and a marked increase in the abundance of beneficial genera, including Akkermansia , demonstrating superior modulation of microbial community structure and metabolic function. Collectively, these findings establish that the selection of a colloidal carrier not only dictates the physicochemical stability of the curcumin-polysaccharide mixture but also governs its spatiotemporal distribution within the gastrointestinal tract and its prebiotic capacity to remodel the gut microecology. This study provides a theoretical foundation for the rational selection of delivery systems to maximize the functional efficacy of bioactive ingredients in functional food applications. Solid Lipid Nanoparticles Enhanced the Neuroprotective Role of Curcumin against Epilepsy through Activation of Bcl-2 Family and P38 MAPK Pathways

Ruiqi Huang,Yanjing Zhu,Lijuan Lin,Simin Song,Liming Cheng,Rongrong Zhu

Oxidative stress of neurons caused by a series of complex neuropathological processes will induce certain neurodegenerative disorders including epilepsy. Curcumin (Cur) is an effective natural antioxidant compound; however, the poor bioavailability obstructs its neural protective applications. In this study, Cur is encapsulated in solid lipid nanoparticles (SLNs) for better neuroprotective efficacy. In vitro study certified that Cur-SLNs functioned obviously better against neuronal apoptosis than Cur, by significantly decreasing the level of free radical and reversing mitochondrial function through the activation of the Bcl-2 family. In vivo experiments showed that SLNs transported Cur through the blood–brain barrier (BBB). The behavioral performance of epileptic mice was improved by Cur-SLNs, with more NeuN but less TUNEL positive cells observed in hippocampus. The in vivo mechanism was also explored. Cur-SLNs reduced neuronal apoptosis through Bcl2 family and P38 MAPK pathways. Overall, Cur-SLNs have better protective effects toward oxidative stress in neurons than free Cur both in vitro and in vivo, which suggests they may be a promising agent against neurodegenerative disorders including epilepsy.

Antidepressant effects of curcumin and HU-211 coencapsulated solid lipid nanoparticles against corticosterone-induced cellular and animal models of major depression

Xiaolie He,Yanjing Zhu,Mei Wang,Guoxin Jing,Rongrong Zhu,Shilong Wang

Major depression is a complex neuropsychiatric disorder with few treatment approaches. The use of nontargeted antidepressants induced many side effects with their low efficacy. A more precise targeting strategy is to develop nanotechnology-based drug delivery systems; hence, we employed solid lipid nanoparticles (SLNs) to encapsulate HU-211 and curcumin (Cur). The antidepressant effects of the dual-drug nanoparticles (Cur/SLNs-HU-211) for major depression treatment were investigated in corticosterone-induced cellular and animal models of major depression. Cur/SLNs-HU-211 can effectively protect PC12 cells from corticosterone-induced apoptosis and can release more dopamine, which may be associated with the higher uptake of Cur/SLNs-HU-211 shown by cellular uptake behavior analysis. Additionally, Cur/SLNs-HU-211 significantly reduced the immobility time in forced swim test, enhanced fall latency in rotarod test, and improved the level of dopamine in mice blood. Cur/SLNs-HU-211 can deliver more Cur to the brain and thus produce a significant increase in neurotransmitters level in brain tissue, especially in the hippocampus and striatum. The results of Western blot and immunofluorescence revealed that Cur/SLNs-HU-211 can significantly enhance the expression of CB1, p-MEK1, and p-ERK1/2. Our study suggests that Cur/SLNs-HU-211 may have great potential for major depression treatment.

An injectable elastic hydrogel crosslinked with curcumin–gelatin nanoparticles as a multifunctional dressing for the rapid repair of bacterially infected wounds

Xiaohui Cai,Yutong He,Liu Cai,Jiamian Zhan,Qian Li,Saiqiong Zhong,Honghao Hou,Wenya Wang,Xiaozhong Qiu

Injectable self-healing hydrogel dressings with excellent elasticity and multifunctional repair effects have been in high demand in wound healing applications, while maintaining stable elasticity in injectable multifunctional hydrogel dressings is still a challenge. Based on carboxymethyl chitosan (CMCS), curcumin–gelatin nanoparticles (CG NPs), and sodium alginate oxide (OSA), we developed a double-crosslinking injectable elastic self-healing hydrogel without any chemical cross-linking agent as a multifunctional wound healing dressing. CG NPs were more stable than pure curcumin (Cur) nanoparticles and could regulate the cross-linking of injectable hydrogels for high elasticity and rapid self-healing. We found that the CG NPs endowed the injectable hydrogel with good anti-inflammatory, antibacterial, and reactive oxygen scavenging activities and could significantly shorten the wound healing time in infected full-thickness skin defect rats by promoting the polarization of M2-type macrophages, reducing oxidative damage, accelerating collagen deposition, enhancing granulation formation, and elevating angiogenesis. Taken together, the tunable elastic injectable hydrogel dressing exhibited a long-term service life with sustained repair function and can be taken as an optimal candidate for bacteria-infected wound healing.

Curcumin inhibits liquid–liquid phase separation of fused in sarcoma and attenuates the sequestration of pyruvate kinase to restore cellular metabolism

Wen-Pu Shi,Wen-Juan Lin,Wan-Yi Ge,Liang-Liang Chen,Tuo-Di Zhang,Wei-Hong Guo,Jian-Li Liu,Da-Chuan Yin

The abnormal accumulation of fused in sarcoma (FUS) is a pathological hallmark in a proportion of patients with frontotemporal dementia and amyotrophic lateral sclerosis. Therefore, the clearance of FUS aggregates is a possible therapeutic strategy for FUS-associated neurodegenerative diseases. This study reports that curcumin can strongly suppress FUS droplet formation and stress granule aggregation of FUS. Fluorescence spectra and isothermal titration calorimetry showed that curcumin can bind FUS through hydrophobic interactions, thereby reducing the β-sheet content of FUS. Aggregated FUS sequesters pyruvate kinase, leading to reduced ATP levels. However, results from a metabolomics study revealed that curcumin changed the metabolism pattern and differentially expressed metabolites were enriched in glycolysis. Curcumin attenuated FUS aggregation-mediated sequestration of pyruvate kinase and restored cellular metabolism, consequently increasing ATP levels. These results indicate that curcumin is a potent inhibitor of FUS liquid–liquid phase separation and provide novel insights into the effect of curcumin in ameliorating abnormal metabolism.

A fluorescent nanoprobe based on N/S co-doped carbon dots coupled with molecularly imprinted polymers for the detection of curcumin

Fang Fang,Shaohua Zhang,Chaofan Chen,Xueying Wang,Chuannan Luo,Qin Wei

Curcumin can be used to treat cancer diseases, however the side effects of excessive use are serious, and the dosage of curcumin should be accurately controlled. In this study, a fluorescence nanosensor using carbon quantum dots (N,S-CQD) embedded multifunctional molecularly imprinted polymer (N,S-CQD@MIP) composite was developed as a probe for the detection of curcumin. The N,S-CQD@MIP was synthesized by sol-gel method and found a maximum emission peak at 430 nm. The curcumin can be binded selectively by the specific interaction between recognition sites and template molecules. The corresponding sensing mechanism of N,S-CQD@MIP for the detection of curcumin is the result of fluorescence inner filter effect and the static quenching effect. The fluorescence nanosensor using N,S-CQD@MIP for the curcumin detection has a good linear relationship in the range of 0.339–136.0 μM, and the corresponding detection limit is as low as 1.357 nM. This method has been applied to detect the curcumin in real samples with the recoveries of 97.6–103.7%. These results indicate that N,S-CQD@MIP are potential sensing materials for curcumin detection.
