Functional DNA molecules have been introduced into polymer-based nanocarrier systems to incorporate chemotherapy drugs for cancer therapy. Here is the first report of dual-responsive microgels composed of a core of Au nanorods and a shell of magnetic ionic liquid and DNA moieties in the cross-linking network simultaneously, as effective drug delivery vectors. TEM images indicated a magnetic polymer shell has an analogous “doughnut” shape which loosely surround the AuNRs core. When irradiated with a near-infrared-light (near-IR) laser, Au nanorods are the motors which convert the light to heat, leading to the release of the encapsulated payloads with high controllability. DNA acts not only as a cross-linker agent, but also as a gatekeeper to regulate the release of drugs. The internalization study and MTT assay confirm that these core–shell DNA microgels are excellent candidates which can enhance the cytotoxicity of cancer cells controlled by near-IR laser and shield the high toxicity of chemotherapeutic agents to improve the killing efficacy of chemotherapeutic agents efficiently in due course.

Neutrophil extracellular traps (NETs) induce tumor metastasis by riding tumor cells through interaction via the transmembrane protein CCDC25 on the surface of cancer cells. Recently, we applied positively charged polyamino acids to neutralize negatively charged NET-DNA and inhibit tumor metastasis as approved on several metastasis models. To elucidate the exact polymer structures on performance, herein, we precisely synthesized a series of cationic oligopeptides with defined numbers of arginine (R) and glycine (G). The oligopeptides R5, R7, R9, and R5G4 showed much better binding affinity and lower cytotoxicity than R3 and R3G6. From the pull-down assay, the oligoarginines, R5, R7, and R9, inhibited CCDC25 interaction with NET-DNA because of stronger competitive interaction than myeloperoxidase (MPO) and citrullinated histone H3 (H3Cit) in NETs. The subsequent inhibition of cell migration and tumor metastasis in triple-negative breast cancer (TNBC) mouse models demonstrated that R5, R7, and R9 efficiently prevented tumor cell metastasis to the liver and lung. This was corroborated by the distribution of oligoarginines in the liver and lung, which reduced NETs accumulation. Thus, in this study, we screened out oligoarginines with a precise number of units to inhibit TNBC metastasis to distant organs, which may benefit in the decreasing mortality rate because of tumor metastasis. With their low cytotoxicity, oligoarginines represent a major advancement for the clinical treatment of metastatic diseases.

Coronavirus disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused secondary pandemic, which still poses a serious threat to physical health and economic development. Herein, the target-driven functionalized DNA hydrogel capillary sensor based on cascade signal amplification and carbon coated cobalt manganese modified by prussian blue and platinum nanoparticles (MnCo@C-Pt-PB NPs) has been successfully developed for dual-mode detection of SARS-CoV-2. The cascade signal amplification triggered by target RNA causes the permeability of the DNA hydrogel loaded in the capillary to be destroyed, thereby releasing the embedded MnCo@C-Pt-PB NPs as signal molecules into 3,3′,5,5′-tetramethylbenzidine/hydrogen peroxide (TMB/H 2 O 2 ) solution under the driving of capillarity. The colorless TMB is then catalyzed to blue oxidation products (oxTMB) due to peroxidase-like activity of MnCo@C-Pt-PB NPs, and MnCo@C-Pt-PB NPs and oxTMB with photothermal properties synergistically increase the system temperature under near-infrared irradiation, which are recorded by portable devices to achieve dual-mode detection. Signals intensity are proportional to the logarithm of T-RNA concentration in a wide detection range (100 aM-100 pM), with a detection limit of 100 aM. Moreover, the reliability of the developed method in oropharyngeal swabs samples has also been validated. The signal conversion and amplification function of functionalized DNA hydrogel enhances the convenience, sensitivity and versatility of the developed method, which is promising to be applied in environmental safety, molecular diagnostic assays and disease prevention.

Synthetic DNA is essential for various areas. Array-based synthetic technologies have been developed, and owing to scalability their synthetic throughput can be dramatically increased with reduced time and cost. However, the traditional arrays are usually fabricated on solid and rigid substrates which can hardly be renewed, their scalability is still limited, which hinders further advance of the synthetic technology. Here, a renewable array based on transformable gallium (Ga) electrode for DNA synthesis and facilitate its downstream manipulation is reported. After the DNA synthesis based on electrochemically triggered phosphoramidite chemistry on the Ga electrode, the electrode can be simply renewed by converting its tip into a drop of liquid metal (LM) so that the remaining electrode can be used for a new synthetic cycle. It also found that by repeated deformations of the LM drop, producing increased oxidation layer on the Ga, the surface DNA can be encapsulated within the LM drop for protection and easy manipulation. It is demonstrated that programmable DNA synthesis and manipulation can be accomplished on a microfluidic chip, which can be used for highthroughput, low-cost DNA synthesis and manipulation for data storage.

Herein, four ruthenium(II) complexes, [Ru(L) 2 (PEIP)(CH 2 ) 4 Br)](ClO 4 ) 2 (L = phen, 1 ; bpy, 2 , PEIP = 2-(2-phenyl-ethenyl)-1 H -imidazole-[4,5 f ][1,10]phenanthroline) and [Ru(L) 2 (PEIP)(CH 2 ) 4 Erianin)](ClO 4 ) 2 (L = phen, 1E ; bpy, 2E ) have been synthesized and investigated as promising inducer to promote conformation inversion of AS1411 G4 DNA. All of these complexes can bind and stabilize AS1411 G4 DNA. Subsequent investigations reveal that in the presence of 1 and 1E, the described circular dichroism signal of AS1411 G4 DNA was reversed. Furthermore, it is discovered that in the presence of 1 and 1E, AS1411 G4 DNA can condense to a somewhat loose nano-network. Moreover, the analysis using molecular docking demonstrates that the conformation inversion of AS1411 G4 DNA is largely dependent on the binding sites of 1 and 1E. In conclusion, these complexes will be used for tumor diagnostics in the clinical context because of their capacity to stably bind AS1411 G4 DNA and dramatically boost its fluorescence emission. Besides, these complexes show promise as inducers of AS1411 G4 DNA conformational inversion.

Metastasis, the leading cause of mortality in cancer patients, presents challenges for conventional photodynamic therapy (PDT) due to its reliance on localized light and oxygen application to tumors. To overcome these limitations, a self-sustained organelle-mimicking nanoreactor is developed here with programmable DNA switches that enables bio-chem-photocatalytic cascade-driven starvation-photodynamic synergistic therapy against tumor metastasis. Emulating the compartmentalization and positional assembly strategies found in living cells, this nano-organelle reactor allows quantitative co-compartmentalization of multiple functional modules for the designed self-illuminating chemiexcited PDT system. Within the space-confined nanoreactor, biofuel glucose is converted to hydrogen peroxide (H 2 O 2 ) which enhances luminol-based chemiluminescence (CL), consequently driving the generation of photochemical singlet oxygen ( 1 O 2 ) via chemiluminescence resonance energy transfer. Meanwhile, hemoglobin functions as a synchronized oxygen supplier for both glucose oxidation and PDT, while also exhibiting peroxidase-like activity to produce hydroxyl radicals (·OH). Crucially, the nanoreactor keeps switching off in normal tissues, with on-demand activation in tumors through toehold-mediated strand displacement. These findings demonstrate that this nanoreactor, which is self-sufficient in light and oxygen and precise in striking tumors, presents a promising paradigm for managing highly metastatic cancers.

DNA methylation aberrations have a strong correlation with cancer in early detection, diagnosis, and prognosis, which make them possible candidate biomarkers. Electrochemical biosensors offer rapid protocols for detecting DNA methylation status with minimal pretreatment of samples. However, the inevitable presence of background current in the time domain, including electrochemical noise and variations, limits the detection performance of these biosensors, especially for low concentration analytes. Here, we propose an ultrasensitive frequency-domain electrochemical analysis strategy to effectively separate the weak signals from background current. To achieve this, we employed periodic magnetic field modulation of magnetic beads (MBs) on and off the electrode surface to generate a periodic electrochemical signal for subsequent frequency-domain analysis. By capturing labeled MBs with as low as 0.5 pg of DNA, we successfully demonstrated a highly sensitive electrochemical method for determination of genome-wide DNA methylation levels. We also validated the effectiveness of this methodology using DNA samples extracted from three types of hepatocellular carcinoma (HCC) cell lines. The results revealed varying genomic methylation levels among different HCC cell lines, indicating the potential application of this approach for early-stage cancer detection in terms of DNA methylation status.

Ochratoxin A (OTA) is a highly toxic mycotoxin with nepHrotoxicity, hepatotoxicity, and carcinogenicity. It poses a long-term threat to the health of humans and animals. Although screen printed electrodes (SPE) with aptamer functionalization provide promising detection capabilities, their applications are limited by inherent weak electrochemical signals, prompting the need for signal amplification strategies. In this study, tetrahedral DNA clusters (TDCs) were prepared as aptamer probes for OTA, and an aptamer-based electrochemical sensing method for OTA was established for signal amplification. Multiple single-stranded DNA (ssDNA) were designed to self-assemble into tetrahedral DNA nanostructure (TDN) through base complementarity, with the OTA aptamer attached to one vertex. Additional ssDNA strands were further utilized to extend TDNs at the three remaining vertices, forming a DNA tetrahedral cluster. The method had a linear range of 1 pg/mL to 100 ng/mL (LOD = 1 pg/mL) for detecting OTA, with excellent specificity and satisfactory recovery rate in feed samples

In vivo optical tumor molecular imaging encounters significant challenges in achieving adequate tumor specificity and sensitivity, largely attributed to off-tumor signal leakage and the relatively low expression levels of target molecules. Therefore, a double self-amplified programmable allosteric DNA nanomachine (named HPs-tFNA) is developed through two elaborately designed hairpin structures (HP1 and HP2) hybridized on tetrahedral framework DNA (tFNA), enabling rapid, specific, and sensitive tumor molecular imaging using the highly specific expression of apurinic/apyrimidinic endonuclease 1 (APE1) in the tumor cytoplasm as a stimulus-response target. In the presence of APE1, HP2 modifies two apurinic/apyrimidinic sites (AP sites), which can be specifically recognized and cleaved by APE1, releasing a significant number of cyclic sequences (cyclic-seq) and achieving initial APE1-assisted signal amplification. Subsequently, cyclic-seq hybridizes with HP1, inducing a conformational change that converts the stem-loop structure of HP1 to a linear form. This structural change facilitates the spatial separation of the fluorophore and quencher, thereby generating fluorescence signals. Furthermore, APE1 incises two AP sites within the HP1 loop region, resulting in the release of cyclic-seq. The released cyclic-seq can hybridize with additional HP1 to continuously amplify the fluorescence signal in a cyclic manner, thereby achieving the second round of signal amplification assisted by APE1. The experimental results of this study demonstrated that HPs-tFNA can achieve rapid in situ tumor molecular imaging and guide precise surgical excision in vivo, with superior spatial specificity. In particular, HPs-tFNA can effectively monitor drug resistance in neuroblastoma cells and stratify risk levels of neuroblastoma via plasma analysis.

Reactive oxygen species (ROS)-induced DNA oxidative damage is a significant manifestation of oxidative stress in the body and is closely associated with the onset and progression of various diseases. Although Pyripyropene A (PPPA) exhibits anti-tumor and anti-inflammatory activities, its antioxidant and protective effects against DNA oxidative injury remain unclear. In this study, using a hydrogen peroxide-induced oxidative injury model of L02 cells, it was found that PPPA could significantly reduce intracellular ROS and malondialdehyde (MDA) levels, enhance the activities of catalase (CAT) and reduced glutathione (GSH), and increase the 2,2-Diphenyl-1-picrylhydrazyl (DPPH) clearance rate, confirming its antioxidant effect. Comet assay showed a reduction in DNA breakage, and down-regulation of phosphorylated histone (γ-H2AX) and 8-hydroxydeoxyguanosine (8-oxodG), indicating that it effectively alleviates DNA oxidative injury. Meanwhile, the upregulated expression of poly ADP-ribose polymerase (PARP) suggests that PPPA may promote repair by activating the DNA damage response (DDR). This study systematically clarify for the first time that PPPA exerts a protective effect by synergistically antioxidizing, reducing DNA injury, and potentially activating repair pathways, providing a theoretical basis for its application in neoplasm and oxidative stress-related diseases.

DNA has demonstrated the abilities to differentiate single-wall carbon nanotubes (SWCNTs) with various chiralities and manipulate their analyte sensing properties. However, the fundamental mechanisms underlying these remarkable abilities remain unclear due to the lack of high-resolution determination of DNA structures on SWCNTs. Here, we combine atomic force microscopy and single-particle cryo–electron microscopy to determine DNA structures on five different types of single-chirality SWCNTs, achieving unprecedented subnanometer resolution. This resolution enables the direct observation of left-handed helical DNA structures with pitches ranging from 1.59 to 2.20 nm, depending on the DNA sequence and nanotube chirality. These findings provide structural insights into the mechanisms by which DNA differentiates the chirality of SWCNTs, and governs the sensitivity, dynamic response range, and analyte differentiability of SWCNT sensors. We propose a non–Watson-Crick hydrogen-bonding network model, which not only accounts for the observed ordered DNA structures but also facilitates the design of DNA sequences for targeted SWCNT purification and desired SWCNT sensor performance.

Self-assembled DNA nanostructures hold great promise in biosensing, drug delivery and nanomedicine. Nevertheless, challenges like instability and inefficiency in cellular uptake of DNA nanostructures under physiological conditions limit their practical use. To tackle these obstacles, this study proposes a novel approach that integrates the cationic polymer polyethyleneimine (PEI) with DNA self-assembly. The hypothesis is that the positively charged linear PEI can facilitate the self-assembly of DNA nanostructures, safeguard them against harsh conditions and impart them with the cellular penetration characteristic of PEI. As a demonstration, a DNA nanotube (PNT) was successfully synthesized through PEI mediation, and it exhibited significantly enhanced stability and cellular uptake efficiency compared to conventional Mg2+-assembled DNA nanotubes. The internalization mechanism was further found to be both clathrin-mediated and caveolin-mediated endocytosis, influenced by both PEI and DNA. To showcase the applicability of this hybrid nanostructure for biomedical settings, the KRAS siRNA-loaded PNT was efficiently delivered into lung adenocarcinoma cells, leading to excellent anticancer effects in vitro. These findings suggest that the PEI-mediated DNA assembly could become a valuable tool for future biomedical applications.
