Diabetic wound healing poses a significant challenge in the biomedical domain. Inadequate angiogenesis and fibroblast dysfunction are two primary characteristics of diabetic microenvironment. Fibroblast dysfunction is characterized by excessive accumulation of reactive oxygen species (ROS) and fragmented mitochondria. Herein, we developed a microenvironment responsive zinc–gallic acid nanosheets (ZnGA NSs) for diabetic wound healing, which slowly release Zn 2+ and gallic acid (GA) in response to low pH and excessive ROS. The released Zn 2+ enhanced cell proliferation, migration, and endothelial tube formation. And the released GA activated antioxidant enzyme system via the KEAP1/NRF2 pathway and promoted mitochondrial fusion by upregulating MFN1/2 and OPA1, which ultimately reprograms dysfunctional fibroblast. The application of ZnGA NSs effectively facilitated wound healing in a rat diabetic model with full-thickness cutaneous defects. More importantly, skin wounds treated with ZnGA NSs exhibited enhanced angiogenesis, alleviated inflammation, and improved tissue regeneration. These findings highlight the potential of ZnGA NSs as a promising therapeutic modality for diabetic wound treatment.

A novel core-shell structured alginate-based hydrogel bead modified by co-gelatinizing with starch and protocatechuic acid (PA), was designed to modulate physical properties of beads, release behavior and antioxidant stability of encapsulated bioactives. Core was fabricated by ionotropic gelation , and its formulation (ratio of sodium alginate/starch) was determined by particle size/starch distribution, texture and bioactive encapsulation capacity of core. Then, coating core with shell-forming solution co-gelatinized with different doses of PA, and subsequently cross-linked with Ca 2+ to obtain core-shell structured beads. Surface microstructure, mechanical characteristics, and swelling ratio of beads were affected by concentrations of PA. Besides, core-shell structure containing PA could enhance delivery and sustained release of encapsulated phenolic bioactives during in vitro digestion, and improve their antioxidant potential stability. Furthermore, interaction between PA and polysaccharide components was elucidated by FTIR and TGA . The present information was beneficial for the advancement of functional food materials and bioactive delivery systems.

Xinhui Chenpi (XHCP) is a well-known type of Chenpi (CP) widely used as both a Chinese herb and a food ingredient. While previous studies have explored how the quality of CP changes over time, there has been limited research specifically on XHCP. This study aims to assess the chemical components and quality of XHCP based on total flavonoid content (TF), antioxidant activity (AA), and color value (CV) at two stages: freshly harvested (XHCP-0Y) and after 3 years of storage (XHCP-3Y). Thirty-eight common volatile compounds were identified, and the content of 17 compounds among them, nine nonvolatile compounds, which included one alkaloid (synephrine), three phenolic acids (PA, protocatechuic acid, vanillic acid, and ferulic acid), and five flavonoids (narirutin, hesperidin, sinensetin, nobiletin, and tangeretin), were firstly detected by the newly developed gas chromatograph–mass spectrometer (GC–MS) and ultra-performance liquid chromatography (UPLC) methods. Compared to XHCP-0Y, the content of 17 volatile compounds and synephrine decreased in XHCP-3Y to varying degrees, while the content of PA, five flavonoids, TF, AA, and CV increased. The reduction of dryness caused by volatile compounds and the enhancement of efficacy related to PA, flavonoids, and AA suggested improved quality of XHCP after 3 years of storage. The methods developed in this study show promise for evaluating the quality of XHCP during the aging process.

This study aimed to investigate the changes in physicochemical properties, bioactivities and metabolites of fermented goji juice (FGJ) by Lacticaseibacillus rhamnosus at different fermentation stages. The results showed that Lacticaseibacillus rhamnosus fermentation significantly decreased the content of soluble protein, total phenolic, total flavonoid and total sugar. Meanwhile, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability and the inhibition rate of xanthine oxidase (XOD) activity were remarkably enhanced by Lacticaseibacillus rhamnosus fermentation. Flavor profiles analysis indicated that FGJ produced novel volatile compounds such as 4-methylpentanol and 2-butanol, which provide its distinct aroma. The non-targeted metabolomics analysis showed that the differential metabolites in the FGJ28 vs. FGJ0 group were mainly included 1,7-bis (3,4-dihydroxyphenyl) heptan-3-yl acetate, isoplumbagin, triacetylresveratrol, sulochrin, indole-3-acetaldehyde, etc. , which might have an effect on the promotion of the bioactivity of goji juice. These findings will contribute to understanding the biotransformation effect of Lacticaseibacillus rhamnosus fermentation on goji juice.

An exopolysaccharide-selenium nanoparticles (EPS-SeNPs) was successfully synthesized by conjugating with Weissella confusa EPS through the reduction of SeO 3 2− . The EPS-SeNPs composite was comprehensively characterized. These analyses confirmed that the EPS-SeNPs composite had an amorphous nature and a uniform size distribution of around 100 nm. The OH groups in EPS interacted with SeNPs, replacing intermolecular interactions in native EPS, which resulted in the stable dispersion of SeNPs within the EPS network. Furthermore, compared to native EPS, EPS-SeNPs with varying Se/EPS ratios demonstrated enhanced radical scavenging capabilities against ABTS, DPPH, superoxide anion radical (O 2 − ), H 2 O 2 , and hydroxyl group radicals (OH·). This suggests that the conjugation of SeNP improved the antioxidant properties of EPS. Furthermore, the investigation delved into the dynamics and mechanism of selenium liberation from EPS-SeNPs under simulated gastric (SGF) and intestinal fluids (SIF). The EPS-SeNPs experienced a decrease in particle size from 223.03 ± 1.67 nm to 98.40 ± 5.57 nm. The release kinetics of selenium in SIF followed a conventional Fickian diffusion pattern. Notably, EPS-SeNPs demonstrated significant Se release following SIF digestion while exhibiting minimal release after SGF digestion, indicating their potential use as a controlled-release selenium-enriched supplement for addressing selenium deficiency.

As the limited water solubility and stability severely restrict the food application of luteolin, a deep knowledge of luteolin is required to understand the reactivity in complex formation. In the present study, the formation mechanism between lactoferrin and luteolin via a pH-driven method was systematically explored to confirm the application in hydrophilic nutraceutical delivery. The physicochemical properties of lactoferrin-luteolin nanocomplexation were investigated through particle size, PDI, ζ-potential, encapsulation efficiency, and loading amount assays. Observation of complexation behaviors revealed the pH-driven method effectively incorporated luteolin into the cores of lactoferrin with pH-dependent solubility characteristics. The improved thermal stability resulted in a stable nanocomplex formation with a zeta potential near − 15 mV. Furthermore, multi-spectroscopy (FTIR and fluorescence spectroscopy) demonstrated that luteolin-lactoferrin nanocomplexation was a spontaneous process together with secondary structure alterations during acidification. Fascinating biological applications of luteolin inspire to do quantum chemical calculations in order to confirm its bioactivities. In-depth investigations of electronic structure (MEP), topology characteristic (ELF, and LOL), and frontier molecular orbital (HOMO and LUMO, energy gap) help confirm the nucleophilic and electrophilic regions of luteolin, which play a crucial role in recognition between luteolin and lactoferrin. Consistent with the observed behavior in vitro, molecular docking further elucidated that hydrophobic contact and hydrogen bonding dominated the location of luteolin into the hydrophobic and deprotonated pocket of lactoferrin, especially at pH 4. From the integrative nutriology perspective, the synergistically biological mechanism of lactoferrin-luteolin nanocomplexation possessed multi-target and multi-pathway properties in preventing oxidative stress-related diseases. In conclusion, experimental and theoretical studies demonstrate great potential in using the pH-driven method for manufacturing luteolin-lactoferrin nanocomplexation with better physical and biological characteristics for delivery of hydrophobic nutraceuticals in the food sector.

Multifunctional biodegradable packaging films with antibacterial and antioxidant properties are highly desirable for food preservation. In this study, multifunctional bioactive chitosan-based composite films were fabricated for loquat preservation after incorporation of licorice flavonoids/ZnO/attapulgite (LF/ZnO/APT) nanocomposites, which were prepared by anchoring ZnO nanoparticles on the surface of attapulgite followed by mechanochemical-assisted loading of licorice flavonoids. The structure and physicochemical properties of composite films were characterized using Fourier transform infrared spectroscopy, transmission electron microscopy, X-ray powder diffraction, etc., while the effects of the added amount of LF/ZnO/APT nanocomposites on the mechanical, antibacterial and antioxidant performances of composite films were investigated. The results indicated that introduction of LF/ZnO/APT nanocomposites obviously enhanced the mechanical properties and antibacterial ratios against Escherichia coli (97.04 ± 2.08%) and Staphylococcus aureus (98.68 ± 1.10%) of composite films. Moreover, the composite films also exhibited excellent scavenging capacities against 2,2-diphenyl-1-picrylhydrazyl and hydroxyl radicals. Therefore, the as-prepared composite films significantly extended the shelf life of loquats, and it is feasible to be served as promising active food packaging materials based on component synergistic effect of composite films.

Hydrogel materials have proven valuable in wound healing, but improving the safety of these hydrogels is still challenging. Therefore, designing multifunctional natural polymeric-based hydrogels with excellent mechanical properties to replace toxic or potentially risky, refractory chemical polymer-based hydrogels such as polyacrylamide and polyethylene glycol is of particular significance. Here, a green starch-based hydrogel (Starch@Ca/CGC hydrogel) with injectability, self-healing, and instant adhesion was constructed by coordination interaction, electrostatic interaction, and intramolecular and intermolecular hydrogen bonds. Therein, natural bioactive small molecules gallic acid (GA) and carvacrol (CA) were coordinated with metal ions by the ultrasonic-triggered self-assembly and ionic cross-linking codriven strategy to prepare Cu-gallic acid-carvacrol nanospheres (CGC NPs), which conferred the hydrogel with near-infrared light (NIR)-controlled CA release and photothermal synergistic sterilization properties, as well as antioxidant and anti-infection capabilities. More importantly, the multifunctional hydrogel platforms could completely cover an irregular wound shape to prevent secondary injury and significantly accelerate wound healing under NIR with more skin appendages like hair follicles and blood vessels appearing. Therefore, it is expected that this starch-based hydrogel could serve as a competitive multifunctional dressing in the biomedical field, including bacteria-derived wound infection and other tissue repair.

Background Silicosis, an interstitial lung disease caused by inhalation of silica particles, poses a significant health concern globally. Green tea polyphenol (TP) stands out as a promising therapeutic candidate, yet its specific protective effects and in-depth mechanisms against silicosis have not been thoroughly investigated. Purpose This study aimed to systematically assess the protective potential of TP against silicosis and to elucidate the underlying mechanisms of its action. Methods A combination of physiological, transcriptomic, molecular, and computational techniques was employed. HPLC was used to identify the components of TP, and its antioxidant properties were tested with DPPH and ABTS assays. The effects of TP on lung injury were assessed in silicosis mice using histopathology, qRT-PCR, and western blot. Transcriptomic analysis was applied to explore the differentially expressed genes and pathways in response to TP intervention. In vitro studies with mouse alveolar macrophages (MH-S) examined TP's effects on cell viability, proliferation, apoptosis, and inflammation responses. Integrated qRT-PCR, western blot, immunohistochemistry, and molecular docking were performed to confirm the molecular mechanism underlying the protective effects of TP against silicosis. Results TP effectively attenuated pulmonary inflammation and fibrosis in silicosis mice, as evidenced by significant reductions in inflammation and fibrotic markers. Moreover, TP's therapeutic benefits were linked to its cytoprotective effects on alveolar macrophages, notably its ability to protect MH-S cells from silica particle-induced apoptosis, inhibition of proliferation, and inflammatory response, underscoring its targeted protective effects at the cellular level. Mechanistically, TP exerted its anti-silicosis activity by targeting key pathways implicated in inflammatory responses, notably through the inhibition of the IL-17/NF-κB p65 signaling cascade. Molecular docking simulations corroborated these findings, demonstrating favorable binding affinities between TP's bioactive components (EGC, ECG, and EGCG) and crucial proteins (IL-17A, IL-17F, p65, TNF-α, IL-6, and IL-1β) involved in the IL-17/NF-κB p65 signaling pathway. This pathway inhibition led to a significant decrease in the production of pro-inflammatory cytokines, such as TNF-α, IL-6, and IL-1β, thus mitigated silicosis. Conclusion TP demonstrates efficacy in alleviating silica particle-induced lung injury by suppressing inflammation through the IL-17/NF-κB p65 signaling pathway, underscoring its potential as a valuable natural compound for silicosis management.

Currently, searching for safer and more effective approaches to promote skin wound healing and tissue regeneration is a significant research focus in the field of public health. Eliminating excessive reactive oxygen species (ROS) and using electrically active dressings to accelerate wound healing has gained significant attention. Herein, green tea-based carbon dots (GCDs) are synthesized from leaves of green tea and proven to possess strong antioxidant, antibacterial, and anti-inflammatory properties. Then, GCDs, polyvinyl alcohol (PVA), and poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS) are successfully constructed into a multifunctional conductive PVA-PEDOT:PSS/GCDs hydrogel (PPPCD) using a repeated freeze-thaw method for promoting skin wound healing. Highly electrical conductivity is imparted by introducing PEDOT:PSS, while antioxidant and antibacterial properties are conferred by GCDs. In vitro assays demonstrated the excellent biocompatibility of the hydrogel, as well as its ability to scavenge excessive ROS and promote cell migration. Furthermore, the hydrogel not only accelerates wound healing by promoting blood vessel formation and epidermal regeneration but also alleviating excessive inflammation in vivo. Overall, the developed hydrogel dressing demonstrates promising prospects for skin wound healing and offers a new perspective for tissue engineering.

This study optimized the extraction of flavonoids fromFagonia creticausing deep eutectic solvents (DESs), focusing on key factors such as the type of DES used, molar ratio, water content, solid/liquid ratio, extraction temperature, and time. Among six DESs tested, the betaine–acetic acid combination exhibited the highest extraction efficiency, attributed to its low viscosity (4.98 mPa·s). Optimal extraction conditions were determined to be a 1:4 molar ratio of betaine to acetic acid, a 25% water content, a solid/liquid ratio of 1:60 g/mL, an extraction temperature of 50 °C, and an extraction time of 30 min. Under these conditions, the flavonoid yield was maximized while preserving bioactivity. Antioxidant assays revealed that flavonoids extracted with DESs exhibited superior scavenging activity against DPPH and hydroxyl radical compared to ethanol-extracted flavonoids, highlighting DESs’ potential to enhance antioxidant properties. The recyclability of DESs was demonstrated using ultracapacitor porous activated carbon, achieving an 89.78% recovery efficiency. The reused DES maintained a high flavonoid extraction yield, retaining 92% efficiency after six cycles, emphasizing its sustainability and cost-effectiveness. This study establishes DES-based extraction as an environmentally friendly and efficient approach for isolating flavonoids with strong antioxidant properties, offering significant advantages in green chemistry and bioactive compound recovery.

Gelatin (Gel) hydrogels are widely utilized in various aspects of tissue engineering, such as wound repair, due to their abundance and biocompatibility. However, their low strength and limited functionality have constrained their development and scope of application. Tannic acid (TA), a naturally occurring polyphenol found in plants and fruits, has recently garnered interest as a crosslinking, anti-inflammatory, and antioxidant agent. In this study, we fabricated novel multifunctional gelatin methacrylate/alginate-tannin (GelMA/Alg-TA) hydrogels using chemical and physical crosslinking strategies with gelatin methacrylate (GelMA), alginate (Alg), and TA as the base materials. The GelMA/Alg-TA hydrogels maintained a stable three-dimensional porous structure with appropriate water content and exhibited excellent biocompatibility. Additionally, these hydrogels demonstrated significant antioxidant and antibacterial properties and substantially promoted wound healing in a mouse model of full-thickness skin defects by modulating inflammatory responses and enhancing granulation formation. Therefore, our study offers valuable insights into the design principles of novel multifunctional GelMA/Alg-TA hydrogels, highlighting their exceptional biocompatibility, antioxidant, and antibacterial properties. GelMA/Alg-TA hydrogels are promising candidates for wound healing applications.
