In this work, a simple, safe, and low-cost two-step route for preparing spherical AlN powders was developed. Specifically, pure AlN spherical powder can be obtained by using water as the solvent, ammonium polyacrylate as the dispersant, carbon powder and alumina powder as the raw materials, spraying and granulating into a spherical precursor, and then sintering at high temperature under a nitrogen atmosphere. The results demonstrate that the average size and the associated distribution of the precursor can be adjusted by regulating the solid or dispersant content of the slurry, which is beneficial for governing the size of the AlN powder. The average particle size of the sintered spherical AlN powders is approximately 33.5 μm, presenting high sphericity (greater than 0.9) and excellent flowability. This approach developed for preparing spherical AlN powder is of great significance for promoting the widespread application of AlN powder in the field of high-performance fillers.

Pulmonary fibrosis is a chronic and irreversible progressive lung disease caused by various factors, such as age and environmental pollution. With countries stepping into an aging society and the seriousness of environmental pollution caused by global industrialization, the incidence of pulmonary fibrosis is annually increasing. However, no effective drug is available for pulmonary fibrosis treatment. C-phycocyanin (C-PC), extracted from blue-green algae, has good water solubility and antioxidation. This study elucidated that C-PC reinforces autophagy to block pulmonary fibrogenesis by inhibiting long noncoding RNA (lncRNA) biogenesis in vivo and in vitro. Cleavage under targets and release using nuclease (CUT & RUN)-PCR, co-immunoprecipitation (Co-IP), and nuclear–cytoplasmic separation experiments clarified that C-PC blocked the nuclear translocation of activating transcription factor 3 (ATF3) to prevent the binding between ATF3 and transcription factor Smad3, thereby hindering lncIAPF transcription. Human antigen R (HuR) truncation experiment and RNA binding protein immunoprecipitation (RIP) were then performed to identify the binding domain with lncIAPF in the 244–322 aa of HuR. lncIAPF exerted its profibrogenic function through the binding protein HuR, a negative regulator of autophagy. In summary, C-PC promoted autophagy via down-regulating the lncIAPF–HuR-mediated signal pathway to alleviate pulmonary fibrosis, showing its potential as a drug for treating pulmonary fibrosis. Exploring how C-PC interacts with biological molecules will help us understand the mechanism of this drug and provide valuable target genes to design new drugs.

Epimedium is thought to enhance the integrity of tendons and bones, ease joint discomfort and rigidity and enhance kidney function. Although glucocorticoids are commonly used in clinical practice, the mechanism by which the active compound Epimedin C (EC) alleviates glucocorticoid‐induced osteoporosis (GIOP) is not well understood. The therapeutic potential of EC in treating GIOP was evaluated using alizarin red S staining, calcein immersion and fluorescence imaging, and bone mineralization, bone mass accumulation and bone density in zebrafish larvae were determined. Using the Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, the key signalling pathways related to bone development were identified. A protein–protein interaction network (PPIN) was constructed to identify osteoclast characteristic genes and the findings were verified using real‐time quantitative PCR (RT‐qPCR). The bone tissue damage caused by prednisolone was reduced by EC. It also altered physiological processes, improved bone density, boosted mineralization and increased bone mass and activity. Subsequent empirical investigations showed that EC impacted the major signalling pathways involved in bone development, such as osteoclast differentiation, oestrogen, MAPK, insulin resistance, PPAR and AMPK signalling pathways. It also decreased the expression of genes typical of osteoclasts. The results of our study uncover a previously unknown function of EC in controlling bone formation and emphasize the potential of EC as a therapeutic target. The osteoprotective effect of EC indicates its potential as a cost‐effective strategy for treating GIOP.

Mitomycin C (MTC) was incorporated to a micelle system preparing from a polymer named deoxycholic acid chitosan-grafted poly(ethylene glycol) methyl ether (mPEG-CS-DA). mPEG-CS-DA was synthesized and characterized by 1H nuclear magnetic resonance (1H-NMR) and Fourier transform infrared spectroscopy. mPEG-CS-DA formed a core-shell micellar structure with a critical micelle concentration of 6.57 µg/mL. The mPEG-CS-DA micelles were spherical with a hydrodynamic diameter of about 231 nm. After poly(ethylene glycol)ylation of deoxycholic acid chitosan (CS-DA), the encapsulation efficiency and drug loading efficiency increased from 50.62% to 56.42% and from 20.51% to 24.13%, respectively. The mPEG-CS-DA micelles possessed a higher drug release rate than the CS-DA micelles. For pharmacokinetics, the area under the curve (AUC) of the mPEG-CS-DA micelles was 1.5 times higher than that of MTC injection, and these micelles can enhance the bioavailability of MTC. mPEG-CS-DA micelles reduced the distribution of MTC in almost all normal tissues and had the potential to improve the kidney toxicity caused by MTC injection.

Ultrafine C-doped ZnO/carbon nanocomposites with different photocatalytic activities have been prepared using TEMPO-oxidized cellulose as a template but also as the source of carbon. The result is an enhancement of the photocatalytic activity ascribed to different phenomena: a high mesoporosity beneficial to mass transport, a thin carbon layer onto ZnO increasing the charge transfer and hydrophobicity of ZnO, a narrowing of ZnO band gap and an increase of the zinc (V Zn ) and oxygen (V o ) vacancies effectively suppressing of the charge recombination. These are evidenced by photocatalytic test of photodegradation of methyl orange (MO) achieved to assess and compared the different photocatalysts. The highest rate constant value of photodegradation of MO is 0.0254 min −1 , three times higher than that of ZnO prepared without templates (0.0087 min −1 ). The present results introduce a new vision of the use of template with multiple roles in the preparation of inorganic materials and specially photocatalysts.

Ceramizable carbon fiber/boron-modified phenolic resin (C f /BPR) composites are cost-effective and suitable for fabricating large-scale components, making them vital for thermal protection systems in aerospace vehicles. However, their thermal resistance requires further improvement. This study investigates the modification of C f /BPR composites using ZrSi 2 , Al, and mica to enhance their thermal resistance. High-temperature static oxidation and oxyacetylene ablation tests are conducted to evaluate their high-temperature performance. The role of each filler on oxidation resistance is discussed. The results reveal that when ZrSi 2 is added at 33.75 phr, Al at 3.75 phr, and mica at 5 phr, the prepared composite exhibits a flexural strength of 56 MPa after 20 min of static oxidation at 1600 °C, with excellent protection of the carbon fibers. After oxidation at 1670 °C, the flexural strength remains at 45 MPa, despite some degree of carbon fiber ablation.

In this work, the (ZrHfNbTaCr)C high-entropy precursors were synthesized via a novel metal–organic-gel (MOG) strategy using pyromellitic acid as dual-functional carbon source and chelating agent, with transition metal chlorides (ZrCl 4 , HfCl 4 , NbCl 5 , TaCl 5 , CrCl 3 ) serving as metal source. The as-synthesized precursors were converted successfully into single-phase nanoparticles featuring regular morphology, average particle size of only about 40 nm and uniform elemental distribution by pyrolysis in vacuum at 1300 ℃. Through spark plasma sintering at 1950 ℃ for 1 h under 40 MPa pressure, the as-prepared nanoparticles were consolidated into dense ceramics achieving relative density of 99.3 %, while maintaining submicrometer-scale grain size of 0.48 ± 0.2 μm. The resultant ceramics exhibited enhanced mechanical properties, demonstrating the Vickers hardness of 22.3 ± 0.5 GPa coupled with the fracture toughness of 4.1 ± 0.1 MPa·m 1/2 . The process provide potential for the research and application of high-entropy carbide ceramics and composites.

The dichloromethyl group is a valuable functional motif in bioactive molecules and synthetic chemistry. Despite its utility, the dearomative dichloromethylation of arenes remains a significant challenge and has been largely unexplored. Herein, we develop a chromium η6-coordination-enabled strategy for dearomative 1,2-hydrodichloromethylation of arenes using inexpensive CH2Cl2 as the dichloromethylating source, providing efficient access to dichloromethylated 1,3-cyclohexadienes. Furthermore, by altering the workup to oxidative quenching with I2, this system is extended to formal arene C–H dichloromethylation, delivering dichloromethylated arenes directly. The method features a broad substrate scope, excellent functional group compatibility, and successful application in the late-stage modification of complex drug molecules.

Background: To investigate the biological functions of the transcription factor LRR binding FLII interacting protein 1 (LRRFIP1) in white adipocyte differentiation (WAD) and elucidate the underlying molecular regulatory mechanisms involved. Methods: Consensus clustering, differential gene expression screening, and intersection analysis were used to identify transcription factors involved in WAD. Adipogenic differentiation experiments were conducted using C3H10T1/2 cells, and a diet-induced obesity model in C57BL/6J mice was established to investigate the function of LRRFIP1 in WAD in vitro and in vivo. Molecular mechanisms were examined through quantitative real-time polymerase chain reaction, Western blotting, luciferase assays, and chromatin immunoprecipitation. Results: Bioinformatics analyses identified LRRFIP1 as a transcription factor associated with WAD. LRRFIP1 expression was downregulated in white adipose tissues from obese patients and in mature white adipocytes. Silencing LRRFIP1 significantly inhibited WAD in C3H10T1/2 cells and reduced differentiation biomarker expression; in contrast, overexpressing LRRFIP1 had the opposite effects. Mechanistically, LRRFIP1 bound to the E2F transcription factor 6 (E2F6) promoter to suppress E2F6 transcription, thereby downregulating a key differentiation regulator, CCAAT enhancer binding protein alpha (C/EBPα). Furthermore, in a diet-induced obesity model, LRRFIP1 could regulate the differentiation and maturation of inguinal white adipose tissue. Conclusion: Our findings reveal that LRRFIP1 plays a crucial inhibitory role in WAD by negatively regulating the E2F6/C/EBPα axis. This discovery not only enriches our understanding of the molecular networks governing WAD but also holds great promise for creating targeted therapies for obesity and associated metabolic conditions.

The solar-driven water splitting for the production of renewable green hydrogen fundamentally relies on the exploration of efficient photocatalysts. Nanostructured TiO2is widely recognized as a promising material for photocatalysis, yet it remains hindered by inadequate light harvesting and fast photogenerated carrier recombination. Herein, calcined C/TiO2xerogels with yolk–shell and core–shell nanostructures (denoted as YS-C/TiO2and CS-C/TiO2) were designed and fabricated via a typical sol–gel–calcination assisted approach. Thanks to the encapsulation of carbon nanospheres into TiO2, it effectively enhances light absorption, improves carrier separation, and lessens carrier recombination, making the well-designed YS-C/TiO2composite display a remarkable hydrogen evolution rate of 975 µmol g−1h−1under simulated solar light irradiation and without the use of any co-catalyst, which is approximately 21.7 times that of the commercial TiO2. The work provides an efficacious design concept in developing nanostructured TiO2-based photocatalysts and in boosting broad photocatalytic applications.

Summary cGAS-STING signaling is essential for innate immunity. Its misregulation promotes cancer or autoimmune and autoinflammatory diseases, and it is imperative to identify effective lead compounds that specifically downregulate the pathway. We report here that astin C, a cyclopeptide isolated from the medicinal plant Aster tataricus , inhibits cGAS-STING signaling and the innate inflammatory responses triggered by cytosolic DNAs. Moreover, mice treated with astin C are more susceptible to HSV-1 infection. Consistently, astin C markedly attenuates the autoinflammatory responses in Trex1 −/− BMDM cells and in Trex1 −/− mouse autoimmune disease model. Mechanistically, astin C specifically blocks the recruitment of IRF3 onto the STING signalosome. Collectively, this study characterizes a STING-specific small-molecular inhibitor that may be applied for potentially manipulating the STING-mediated clinical diseases.

A variety of disturbances such as starvation, organelle damage, heat stress, hypoxia and pathogen infection can influence the autophagic process. However, how the macroautophagy/autophagy machinery is regulated intrinsically by structural damage of the cell remains largely unknown. In this work, we utilized the C. elegans epidermis as the model to address this question. Our results showed that structural damage by mechanical wounding exerted proximal inhibitory effect and distant promotional effect on autophagy within the same epidermal cell. By disrupting individual mechanical supporting structures, we found that only damage of the basal extracellular matrix or the underlying muscle cells activated a distinct autophagic response in the epidermis. On the contrary, structural disruption of the epidermal cells at the apical side inhibited autophagy activation caused by different stress factors. Mechanistic studies showed that the basal promotional effect of structural damage on epidermal autophagy was mediated by a mechanotransduction pathway going through the basal hemidesmosome receptor and LET-363/MTOR, while the apical inhibitory effect was mostly carried out by activation of calcium signaling. Elevated autophagy in the epidermis played a detrimental rather than a beneficial role on cell survival against structural damage. The results obtained from these studies will not only help us better understand the pathogenesis of structural damage- and autophagy-related diseases, but also provide insight into more generic rules of autophagy regulation by the structural and mechanical properties of cells across species.Abbreviations : ATG: autophagy related; BLI-1: BLIstered cuticle 1; CeHDs: C. elegans hemidesmosomes; COL-19: COLlagen 19; DPY-7: DumPY 7; ECM: extracellular matrix; EPG-5: ectopic PGL granules 5; GFP: green fluorescent protein; GIT-1: GIT1 (mammalian G protein-coupled receptor kinase InTeractor 1) homolog; GTL-2: Gon-Two Like 2 (TRP subfamily); HIS-58, HIStone 58; IFB-1: Intermediate Filament, B 1; LET: LEThal; LGG-1: LC3, GABARAP and GATE-16 family 1; MTOR: mechanistic target of rapamycin; MTORC1: MTOR complex 1; MUP-4: MUscle Positioning 4; NLP-29: Neuropeptide-Like Protein 29; PAT: Paralyzed Arrest at Two-fold; PIX-1: PIX (PAK (p21-activated kinase) Interacting eXchange factor) homolog 1; RFP: red fluorescent protein; RNAi: RNA interference; SQST-1: SeQueSTosome related 1; UNC: UNCoordinated; UV: ultraviolet; VAB-10: variable ABnormal morphology 10; WT: wild type.
