Comparison of non-covalent binding interactions of six caffeoylquinic acids with β-lactoglobulin: Spectroscopic analysis, molecular docking and embedding of curcumin

Jiayuan Liu,Gongshuai Song,Like Zhou,Danli Wang,Tinglan Yuan,Ling Li,Guanghua He,Gongnian Xiao,Jinyan Gong

In this study, we systematically compared the differences in the interactions of β-lactoglobulin (LG) with three mono-caffeoylquinic acids (MCQAs: 3-CQA, 4-CQA, and 5-CQA) and three dicaffeoylquinic acids (DCQAs: 3,4-DCQA, 3,5-DCQA, and 4,5-DCQA). Based on molecular docking techniques, it was concluded that the non-covalent binding of all six caffeoylquinic acids to LG was dominated by hydrophobic forces, among which the binding ability of DCQAs was stronger. The antioxidant assay revealed that caffeoylquinic acid significantly improved the antioxidant properties of LG, and the best antioxidant properties were achieved with 4,5-DCQA. The DPPH radical scavenging ability was increased by approximately 54 μmol Trolox/g-sample, and the ABTS radical scavenging ability increased by 29.26 μmol Trolox/g-sample. Six non-covalent complexes were used to embed curcumin , and the stability of the formed solution was further investigated and compared.

Enhancing solid-like characteristics of porcine plasma protein-carrageenan-based high internal phase emulsion: As solid fat alternative of loading curcumin

Jianglan Yuan,Ping Yan,Xiaocui Liu,Xu Kang,Yongguo Jin,Long Sheng,Jianxin Xia

This study explored the feasibility of a novel solid-like fat stabilized by porcine plasma protein (PPP) and carrageenan (CG) coupling emulsifier as curcumin delivery vehicle. The combination of 2.0% (w/v) PPP and 0.3% (w/v) CG successfully stabilized the oil of 80% (v/v) volume fraction at solid-like state, and the increasing of oil phase led to the increase in droplet size , storage modulus (G′) and apparent viscosity. Compared with free oil, oxidation of PPP-CG-stabilized solid-like fat could be significantly inhibited. The results from in vitro simulated digestion showed that the release rate of free fatty acids and bioaccessibility of curcumin in the solid-like fat were significantly higher than those in free oil, and curcumin was almost not released in simulated gastric fluid, but mainly released in simulated intestinal fluid. Therefore, PPP-CG-stabilized solid-like fat could be used as an effective nutrient delivery vehicle for food and pharmaceuticals, and the work provided some new insights into the structure design of functional food.

Production and Anti-Inflammatory Performance of PVA Hydrogels Loaded with Curcumin Encapsulated in Octenyl Succinic Anhydride Modified Schizophyllan as Wound Dressings

Lingyun Tu,Yifeng Fan,Yongfei Deng,Lu Hu,Huaiqing Sun,Bisheng Zheng,Dengjun Lu,Chaowan Guo,Lin Zhou

Amphiphilic polysaccharides can be used as wall materials and applied to encapsulate hydrophobic active chemicals; moreover, there is significant demand for novel medical high-molecular-weight materials with various functions. In order to prepare amphiphilic schizophyllan (SPG), octenyl succinic anhydride (OSA) was chosen to synthesize OSA-modified schizophyllan (OSSPG) using an esterified reaction. The modification of OSSPG was demonstrated through FT-IR and thermal analysis. Moreover, it was found that OSSPG has a better capacity for loading curcumin, and the loading amount was 20 μg/mg, which was 2.6 times higher than that of SPG. In addition, a hydrogel made up of PVA, borax, and C-OSSPG (OSSPG loaded with curcumin) was prepared by means of the one-pot method, based on the biological effects of curcumin and the immune-activating properties of SPG. The mechanical properties and biological activity of the hydrogel were investigated. The experimental results show that the dynamic cross-linking of PVA and borax provided the C-OSSPG/BP hydrogel dressing with exceptional self-healing properties, and it was discovered that the C-OSSPG content increased the hydrogel’s swelling and moisturizing properties. In fibroblast cell tests, the cells treated with hydrogel had survival rates of 80% or above. Furthermore, a hydrogel containing C-OSSPG could effectively promote cell migration. Due to the excellent anti-inflammatory properties of curcumin, the hydrogel also significantly reduces the generation of inflammatory factors, such as TNF-α and IL-6, and thus has a potential application as a wound dressing medicinal material. Keywords:schizophyllan;octenyl succinic anhydride;curcumin;PVA;hydrogel;wound-healing

In vitro corrosion resistance and dual antibacterial ability of curcumin loaded composite coatings on AZ31 alloy: Effect of amorphous calcium carbonate

Kui Xue,Yan-Jin Li,Tian-Hao Ma,Lan-Yue Cui,Cheng-bao Liu,Yu-hong Zou,Shuo-Qi Li,Fen Zhang,Rong-Chang Zeng

Rapid corrosion and bacterial infection are obstacles to put into use biodegradable magnesium (Mg) alloy as biomedical materials . In this research, an amorphous calcium carbonate (ACC)@curcumin (Cur) loaded poly-methyltrimethoxysilane (PMTMS) coating prepared by self-assembly method on micro-arc oxidation (MAO) coated Mg alloy has been proposed. Scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy and Fourier transform infrared spectroscopy are adopted to analyze the morphology and composition of the obtained coatings. The corrosion behaviour of the coatings is estimated by hydrogen evolution and electrochemical tests. The spread plate method without or with 808 nm near-infrared irradiation is applied to evaluate the antimicrobial and photothermal antimicrobial ability of the coatings. Cytotoxicity of the samples is tested by 3-(4,5)-dimethylthiahiazo(-z-y1)-2,5-di- phenytetrazoliumromide (MTT) and live/dead assay culturing with MC3T3-E1 cells. Results show that the MAO/ACC@Cur-PMTMS coating exhibited favourable corrosion resistance , dual antibacterial ability, and good biocompatibility. Cur was employed as an antibacterial agent and photosensitizer for photothermal therapy. The core of ACC significantly improved the loading of Cur and the deposition of hydroxyapatite corrosion products during degradation, which greatly promoted the long-term corrosion resistance and antibacterial activity of Mg alloys as biomedical materials.

A curcumin-crosslinked bilayer film of soy protein isolate and chitosan with enhanced antibacterial property for beef preservation and freshness monitoring

Yueyuan Yong,Shancan Wang,Laihao Li,Rui Li,Hafiz Nabeel Ahmad,Noshaba Munawar,Jie Zhu

In this study, antibacterial and antioxidant bilayer films were prepared by using curcumin (Cur) crosslinked soy rotein isolate (SPI) and chitosan (CS). Molecular docking simulations and multispectral analysis revealed that hydrogen bonding and hydrophobic interactions were the primary driving forces that promoted the self-assembly of the bilayer films. The tensile strength, the UV-blocking properties and the hydrophobicity was greatly improved of the bilayer antimicrobial films. Moreover, water vapor permeability, thermal shrinkage and opacity were all reduced significantly. In addition, the composite films with curcumin demonstrated effective antioxidant activity and a slow release characteristic. Morphology observation of the bacteria by AFM revealed that the antibacterial bilayer film had a significant damaging effect on the cell structures of S. aureus and E. coli due to the dual antibacterial effect of curcumin and chitosan. SPI + Cur-CS antimicrobial bilayer film effectively inhibited the growth of bacteria and extended the shelf life of beef. According to the findings, SPI + Cur-CS antimicrobial bilayer film can be used as an active package material for beef preservation and freshness monitoring.

Polyphosphazene Nanodrugs for Targeting Delivery and Inflammation Responsive Release of Curcumin to Treat Acute Lung Injury by Effectively Inhibiting Cytokine Storms

Xiaochen Su,Xunan Jing,Wanting Jiang,Meng Li,Kai Liu,Menghao Teng,Yayun Ma,Daquan Wang,Lingjie Meng,Yingang Zhang,Wenchen Ji

An excessive inflammatory response induced by cytokine storms is the primary reason for the deterioration of patients with acute lung injury (ALI). Though natural polyphenols such as curcumin (CUR) have anti-inflammation activity for ALI treatment, they often have limited efficacy due to their poor solubility in water and oxidising tendency. This study investigates a highly cross-linked polyphosphazene nano-drug (PHCH) developed by copolymerisation of CUR and acid-sensitive units (4-hydroxy-benzoic acid (4-hydroxy-benzylidene)-hydrazide, D-HBD) with hexachlorotripolyphosphonitrile (HCCP) for improved treatment of ALI. PHCH can prolong the blood circulation time and targeted delivery into lung inflammation sites by enhancing CUR's water dispersion and anti-oxidant properties. PHCH also demonstrates the inflammation-responsive release of CUR in an inflammation environment due to the acid-responsive degradation of hydrazine bonds and triphosphonitrile rings in PHCH. Therefore, PHCH has a substantial anti-inflammation activity for ALI treatment by synergistically improving CUR's water-solubility, bioavailability and biocompatibility. As expected, PHCH attenuates the cytokine storm syndrome and alleviates inflammation in the infected cells and tissues by down-regulating several critical inflammatory cytokines (TNF-α, IL-1β, and IL-8). PHCH also decreases the expression of p-p65 and C-Caspase-1, inhibiting NLRP3 inflammasomes and suppressing NF-κB signalling pathways. The administrated mice experiments confirmed that PHCH accumulation was enhanced in lung tissue and showed the efficient scavenging ability of reactive oxygen species (ROS), effectively blocking the cytokine storm and alleviating inflammatory damage in ALI. This smart polyphosphazene nano-drug with targeting delivery property and inflammation-responsive release of curcumin has excellent potential for the clinical treatment of various inflammatory diseases, including ALI.

Application of curcumin in mixed properties of quaternary N-alkyl tropine-based ionic liquid surfactant with nonionic, anionic and zwitterionic surfactants

Zhixia Wang,Hang Song

Curcumin has been found to be sensitive to the solution environment and has aroused great interest. Based on this, UV spectrophotometry with curcumin as a probe was employed to investigate the solution properties of surfactants. The reliability of UV spectrophotometry using curcumin as a probe was confirmed by comparing the outcomes with those obtained by surface tension method. The micellization behaviors of surfactant mixtures of tropine-based ionic liquid surfactant (TILS) with anionic surfactant sodium dodecyl sulfate (SDS), nonionic surfactant Triton X-100 (TX-100), and zwitterionic surfactant N-dodecyl-N,N-dimethylglycine (BS-12) were investigated, respectively. Based on the theory of regular solutions, the interaction parameters between the two surfactants and their corresponding thermodynamic parameters were analyzed. The negative β m and | β m | > |ln( CMC 1 / CMC 2 )| suggest intense attraction between two different surfactants. Thermodynamic parameters indicate that micelle formation is an exothermic and entropy-increasing process. Comparing the interaction parameters of the three systems, it is evident that TILS exhibits stronger synergistic effects in the formation of mixed micelles with SDS as compared to TX-100 and BS-12. These results demonstrate the feasibility of using curcumin for the study of surfactant compounding properties and broaden the application range of curcumin.

Fabrication of multifunctional materials based on chitosan/gelatin incorporating curcumin-clove oil emulsion for meat freshness monitoring and shelf-life extension

Runan Zhao,Haocheng Guo,Tianyi Yan,Jiaheng Li,Weidong Xu,Yong Deng,Jianwei Zhou,Xingqian Ye,Donghong Liu,Wenjun Wang

A new multifunctional film with active and intelligent effects was developed by incorporating curcumin-clove oil emulsion into natural materials. The basic properties, functional characteristics, and pH/NH 3 -sensitivity of films were investigated, and then these films were applied to extend shelf-life and monitor freshness of meat. Curcumin solution and emulsion illustrated significant color variations at different pH values. The incorporation of emulsion improved the UV–vis barrier and water resistance properties of films, which blocked most of UV-light and its water contact angle reached 100.03°. Meanwhile, the films had stronger mechanical strength and higher thermal stability, with elongation at break reaching 79.18 % and the maximum degradation temperature rising to 316 °C. Moreover, emulsion made films have a slow-release effect on clove oil, which not only enhanced the antioxidant property but also significantly improved their antibacterial activity. Additionally, the multifunctional films presented a significant color response to acidic/alkaline environments over a short time interval and could be easily identified by naked eyes. Finally, the films effectively extended the shelf-life of fresh meat by 3 days at 4 °C and visually monitored freshness through color changes in real-time. This knowledge provides insights and ideas for the development of novel food packaging with both active and intelligent functions.

Transcutaneous amorphous preparation co-delivering curcumin and modified aptamer as a synergistic approach for the amelioration of psoriasis-like skin inflammation

Nan Jin,Ruotong Chen,Yuling Li,Ziyi Zhang,Baomei Zhang,Jianmin Chen

The aim is to achieve the targeting function of Aptamer 21-2 (Apt) by modifying glycyrrhizic acid (GA) to obtain Apt-GA and by delivering it through mesoporous silica in μm-size (μmS). Moreover, it is planned to induce synergistic effects of the novel amorphous preparation of Apt-GA with curcumin (Cur) in the treatment of psoriasis. UV/vis spectrophotometric analysis and Fourier transform infrared spectroscopy were performed to demonstrate the linkage of Apt and GA. The replacement of GA with Apt-GA in GA-Cur-μmS did not make a difference in the amorphous state, but showed superior fluorescence of Cur in the inflamed dermis similar to psoriatic induced by imiquimod. Consequently, compared to GA-Cur-μmS and Apt, Apt-GA-Cur-μmS exhibited less pronounced psoriatic characteristics in vivo , including Psoriasis Area and Severity Index, histological image, immunohistochemistry and immuno-fluorescence labeling neutrophils. The enzyme-linked immunosorbent assay showed a similar phenomenon that less IL-17A was observed after treatment with Apt-GA-Cur-μmS than with GA-Cur-μmS or Apt. Our finding suggests that this novel Apt delivery system achieved the Apt targeting function in the dermis by taking advantage of GA and μmS, and exhibited synergistic anti-psoriatic effect by co-delivering curcumin .

Efficient Delivery of Curcumin by Alginate Oligosaccharide Coated Aminated Mesoporous Silica Nanoparticles and In Vitro Anticancer Activity against Colon Cancer Cells

Chennan Liu,Fangyuan Jiang,Zifeng Xing,Lihong Fan,Yuan Li,Shaoning Wang,Junhong Ling,Xiao-Kun Ouyang

We designed and synthesized aminated mesoporous silica (MSN-NH2), and functionally grafted alginate oligosaccharides (AOS) on its surface to get MSN-NH2-AOS nanoparticles as a delivery vehicle for the fat-soluble model drug curcumin (Cur). Dynamic light scattering, thermogravimetric analysis, and X-ray photoelectron spectroscopy were used to characterize the structure and performance of MSN-NH2-AOS. The nano-MSN-NH2-AOS preparation process was optimized, and the drug loading and encapsulation efficiencies of nano-MSN-NH2-AOS were investigated. The encapsulation efficiency of the MSN-NH2-Cur-AOS nanoparticles was up to 91.24 ± 1.23%. The pH-sensitive AOS coating made the total release rate of Cur only 28.9 ± 1.6% under neutral conditions and 67.5 ± 1% under acidic conditions. According to the results of in vitro anti-tumor studies conducted by MTT and cellular uptake assays, the MSN-NH2-Cur-AOS nanoparticles were more easily absorbed by colon cancer cells than free Cur, achieving a high tumor cell targeting efficiency. Moreover, when the concentration of Cur reached 50 μg/mL, MSN-NH2-Cur-AOS nanoparticles showed strong cytotoxicity against tumor cells, indicating that MSN-NH2-AOS might be a promising tool as a novel fat-soluble anticancer drug carrier.Keywords:pH-sensitive drug release;silica;brown algae oligosaccharides;curcumin;colon cancer cells

Impact of Emulsifier Structure and Concentration on Lipolysis Dynamics and Curcumin Bioaccessibility in the Nanoemulsions Stabilized by Polyglycerol Fatty Acid Esters

Zhang Shangwei,Xu Xiaofei,Yang Jiguo,Ren Jie

Polyglycerol fatty acid esters (PGFEs), a type of nonionic surfactants, have been widely used in food industry. However, the effects of the aliphatic chain lengths in PGFEs and the concentrations of PGFEs on digestive profiles in emulsion-based systems are poorly understood. The present study has investigated the physicochemical stability, lipolysis dynamics and curcumin bioaccessibility in the nanoemulsions stabilized by synthesized PGFEs with different aliphatic chains (C 10 -C 18 ) at various concentrations using an in vitro gastrointestinal tract (GIT) model. Shorter aliphatic chain or higher concentrations of PGFEs resulted in smaller droplets in the emulsions before and during digestion. PGFEs concentration had different impacts on lipolysis dynamics of nanoemulsions depending on the aliphatic chain lengths of PGFEs. Furthermore, long aliphatic chain of PGFEs contributed to a greater rate and extent of lipolysis, but a lower bioaccessibility of curcumin compared with medium ones, which was attributed to the formation of insoluble calcium soaps induced by calcium ions. These results are expected to facilitate the application of PGFEs for developing optimized nanoemulsions in encapsulating poorly water-soluble nutraceuticals in functional food industry.
