Highly spontaneous spin polarization engineering of single-atom artificial antioxidases towards efficient ROS elimination and tissue regeneration

Bihui Zhu,Zhenyang Zhao,Sujiao Cao,Yimin Sun,Liyun Wang,Songya Huang,Chong Cheng,Lang Ma,Li Qiu

The creation of atomic catalytic centers has emerged as a conducive path to design efficient nanobiocatalysts to serve as artificial antioxidases (AAOs) that can mimic the function of natural antioxidases to scavenge noxious reactive oxygen species (ROS) for protecting stem cells and promoting tissue regeneration. However, the fundamental mechanisms of diverse single-atom sites for ROS biocatalysis remain ambiguous. Herein, we show that highly spontaneous spin polarization mediates the hitherto unclear origin of H2O2-elimination activities in engineering ferromagnetic element (Fe, Co, Ni)-based AAOs with atomic centers. The experimental and theoretical results reveal that Fe-AAO exhibits the best catalase-like kinetics and turnover number, while Co-AAO shows the highest glutathione peroxidase-like activity and turnover number. Furthermore, our investigations prove that both Fe-AAO and Co-AAO can effectively secure the functions of stem cells in high ROS microenvironments and promote the repair of injured tendon tissue by scavenging H2O2 and other ROS. We believe that the proposed highly spontaneous spin polarization engineering of ferromagnetic element-based AAOs will provide essential guidance and practical opportunities for developing efficient AAOs for eliminating ROS, protecting stem cells, and accelerating tissue regeneration.

High-Performance Elastomer with Excellent Resistance to Low Temperature, Aging and Solvent

Xiwei Guo,Ronan Brown,Yue Dong,Haoyu Feng,Yang Xiang,Dongxu Wang,Qi Zhang,He Zhu,Shiping Zhu

Elastomers are essential for a variety of demanding applications, but their performance is significantly compromised under harsh conditions, such as extremely low temperatures and serious aging. Herein, a novel polyurethane elastomer (PFTA) is presented, having perfluoropolyether as the soft segment and catechol-containing molecules synthesized via Schiff base reaction as the chain extender, where the perfluoropolyether segments confer outstanding low-temperature and solvent resistance and the catechol groups provide exceptional anti-aging properties to the material. As a result, the obtained elastomer achieves an ultralow glass transition temperature of approximately −115 °C, demonstrating remarkable flexibility at extremely low temperatures, even in a liquid nitrogen environment, and possesses good aging and solvent resistance. These results highlight that the combination of catechol functionalization and perfluoropolyether segments provides a promising platform for the development of elastomers suitable for extreme environmental applications, such as aerospace and polar exploration.

Gelatin-based composite film integrated with nanocellulose and extract-metal complex derived from coffee leaf for sustainable and active food packaging

Fangfei Sun,Juan Zhao,Peng Shan,Kun Wang,Hui Li,Lincai Peng

Gelatin (GEL) is widely used in food packaging due to its excellent film-forming ability, non-toxicity, biodegradability, and biocompatibility. However, pure GEL films suffer from poor mechanical strength, high water sensitivity, and lack antibacterial properties, limiting their suitability for food packaging applications. In this study, we incorporated dialdehyde cellulose nanocrystal (DCNC) derived from coffee leaves as cross-linkable nanofillers and a coffee leaf extract-Fe 3+ complex (CFe) as a functional additive into GEL to develop active food packaging composite films. We investigated the effects of varying CFe content on the microstructure and performance of films. The addition of 1 wt% CFe enhanced the entanglement and intermolecular interactions among the film-forming components, resulting in a dense and continuous microstructure. This led to significant improvements in UV-light shielding, mechanical strength, water solubility, water vapor and oxygen barrier properties of the GEL-based composite films. Owing to the excellent photothermal conversion capability of CFe, the composite films exhibited strong antibacterial activity against S. aureus and E. coli , with an antibacterial efficiency exceeding 90%. Additionally, the inclusion of CFe imparted good antioxidant properties to the composite films. Packaging application tests further demonstrated that the functional films, when assisted by 808 nm near-infrared (NIR) laser irradiation, effectively slowed the quality deterioration of fresh pork, extending its shelf-life to 8 days under 4 °C storage. These findings suggest that the developed composite films hold significant promise for use in food packaging.

Gelatin/dextran active films incorporated with cinnamaldehyde and α-tocopherol for scallop (Patinopecten yessoensis) adductor muscle preservation

Fengjiao Fan,Chenlinrui Yue,Zhenni Zhai,Hailu Liao,Xiaoni Lian,Hongkai Xie

Scallops are rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid but perishable due to their microbial growth and lipid oxidation. In this study, gelatin/dextran films containing cinnamaldehyde and α-tocopherol (0% + 0%, 0.3% + 0.3%, 0.6% + 0.6%, 0.9% + 0.9%, and 1.2% + 1.2%, w/w) as active fillers were developed by solution casting method, and their preservation effects on scallop adductor muscle refrigerated at 4°C for 0, 3, 6, 9, and 12 days were evaluated. Inclusion of the two active fillers did not influence the thermal stability of the films but created heterogenous and discontinuous film microstructure and increased the film hydrophobicity. Increase in the concentrations of active fillers lowered the mechanical properties and water vapor permeability of the films but increased their crystallinity, thickness, water contact angle, opacity, antibacterial property, and antioxidant property. The longest release times for both cinnamaldehyde and α-tocopherol were found in 95% (v/v) ethanol solution. The gelatin/dextran films containing 1.2% (w/w) of active fillers (Gelatin [Ge]/Dextran [Dx]/1.2 film) improved the chemical stability of refrigerated scallop adductor muscle. The total viable count (TVC) of the unpackaged scallop adductor muscle exceeded the recommended limit of 7 lg CFU/g on day 6 (7.07 ± 0.50 lg CFU/g), whereas the TVC of the Ge/Dx/1.2 film-packaged scallop adductor muscle was still below the limit on day 9 (5.60 ± 0.50 lg CFU/g). Thus, the Ge/Dx/1.2 film can extend the shelf life of refrigerated scallop adductor muscle by at least 3 days. Overall, the developed gelatin/dextran active packaging films are promising for the preservation of aquatic food products.

Ga2Se3 nanosheets: Broad-spectrum antibacterial and antioxidant agents with low drug resistance

Henggang Wang,Shan Sun,Jin Yang,Haoyi Sun,Ke Cheng,Xinxin Wang,Si Li,Hengwei Lin

The efficient treatment of many infectious diseases is hindered by uncontrolled bacterial infections and inflammation induced by reactive oxygen species (ROS). Thus, there is an urgent need for the development of multifunctional therapeutic agents that possessing both antibacterial and antioxidant features. In this study, a novel antibacterial and antioxidant agent, Ga 2 Se 3 nanosheets (NSs), were firstly prepared by the liquid-phase exfoliation method. The Ga 2 Se 3 NSs exhibited broad-spectrum sterilization capabilities, even including multidrug-resistant strains. Noteworthily, Ga 2 Se 3 NSs showed a remarkable 800-fold enhancement in antibacterial efficacy compared to the bulk materials, while also demonstrating low drug resistance compared to the traditional antibiotics. Their superior antibacterial performance is primarily attributed to the disruption of basic bacterial metabolism induced by gallium and selenium components, as well as the dimensional engineering of Ga 2 Se 3 NSs. Cellular studies further confirmed the protective effects of pluronic F127 modified Ga 2 Se 3 NSs against oxidative stress damage, preserving cellular functions through ROS scavenging. This study highlights the superior antibacterial and antioxidant properties of Ga 2 Se 3 NSs, which offers an alternative opportunity for addressing the drug resistance issue in treatment of certain special diseases.

Gallic acid-grafted chitosan photothermal hydrogels functionalized with mineralized copper-sericin nanoparticles for MRSA-infected wound management

Lihua Li,Li Ren,Qiuhan Zhao,Ke Xu,Qiushuang Wu,Qianru Su,Xin Li,Xin Lü,Li Wang

The management of wounds infected with drug-resistant bacteria represents a significant challenge to public health globally. Nanotechnology-functionalized photothermal hydrogel with good thermal stability, biocompatibility and tissue adhesion exhibits great potential in treating these infected wounds. Herein, a novel photothermal hydrogel (mCS-Cu-Ser 1 ) was prepared through in situ mineralization in the hydrogel networks and ion cross-linking driven by copper ions (∼3 mM). Self-assembling polyphosphate sericin nanoparticles (Ser NPs) formed by an ultrasound-assisted anti-solvent method were as mineralization templates and gallic acid-grafted chitosan (mCS) was prepared as the sole matrix. Grafting of polyphenols and cross-linking of copper ions endowed mCS-Cu-Ser 1 with injectable, skin-adhesive and self-healing characteristics. Due to the nonradiative relaxation of Cu 2+ electron-hole pairs of copper phosphate on the surface of Ser NPs and the molecular thermo-vibrational effect of the mCS-Cu complex, mCS-Cu-Ser 1 rapidly warmed up to 50 °C within one minute under near-infrared (NIR) irradiation. Integrating such excellent photothermal properties with antimicrobial activity and intracellular reactive oxygen species scavenging from mCS, mCS-Cu-Ser 1  + NIR effectively accelerated methicillin-resistant Staphylococcus aureus (MRSA) infected wound healing. This work develops a novel dressing for the treatment of MRSA-infected wounds and provides some reference for the preparation of multifunctional acid-free chitosan hydrogels.

Fangfei Sun,Peng Shan,Bingzhen Liu,Yongshi Li,Kun Wang,Yongliang Zhuang,Delu Ning,Hui Li

In this study, dialdehyde carboxymethyl cellulose (DCMC, 10 wt% based on gelatin) and varying contents of coffee leaf extract (CLE, 1, 3, 5 and 7 wt% based on gelatin) were incorporated into gelatin (GEL) matrix to develop multifunctional food packaging films. DCMC acted as a physical reinforcing filler through crosslinking with GEL matrix by Schiff-base reaction, CLE served as an active filler to confer film functional properties. The micro-morphology, micro-structure, physicochemical and functional properties of the GEL/DCMC/CLE composite film were investigated. The results demonstrated that mechanical, barrier properties and thermal stability of films were significantly improved by incorporation of CLE. Compared with pure GEL film, the GEL/DCMC/5%CLE film exhibited excellent UV light blocking while kept enough transparency, the best mechanical property, water resistance, water vapor and oxygen barrier, as well as thermal stability. GEL/DCMC/5%CLE film also possessed strong antioxidant activity and some antibacterial activity against E. coli and S. aureus . Packaging application testing demonstrated that the resultant GEL/DCMC/5%CLE film effectively delayed the lipid oxidation of walnut oil and preserved the postharvest freshness of fresh walnut kernels under ambient conditions.

Ginkgo biloba leaf polysaccharide-stabled selenium nanozyme as an efficient glutathione peroxidase mimic for the preservation of bananas and cherry tomatoes

Jiahui Zou,Huimin Zhong,Cong Jiang,Guancheng Zhu,Xueer Lin,Yanyan Huang

To develop functional, sustainable and eco-friendly active packaging materials as alternatives to plastic films, we successfully prepared Ginkgo biloba leaf polysaccharide-stabilized selenium nanomaterials (Se-GBLP). Se-GBLP with glutathione peroxidase-like activity could efficiently remove harmful reactive oxygen species. As a functional additive, Se-GBLP was incorporated into degradable chitosan (CS) to fabricate CS/Se-GBLP films. The addition of Se-GBLP improved the mechanical properties, UV–visible light barrier performance, water vapor permeability, and antioxidant activity of the films. Preservation experiments demonstrated CS/Se-GBLP film could maintain quality and prolong the storage time of bananas and cherry tomatoes . It was the first time to use selenium-based nanozyme for fruit preservation. This work offered a cost-effective solution to reduce post-harvest losses, increasing sustainability and profitability. Future research should focus on more factors affecting freshness such as variety, maturity, harvest and storage conditions to improve preservation, as well as on the material's safety concern and environmental impact.

Extending the freshness of tilapia surimi with pulsed electric field modified pea protein isolate-EGCG complex

Zhong Han,Ze-Ling Chen,Xin-Dong Xu,Songyi Lin,Yan Wang,Lang-Hong Wang,De-Bao Niu,Xin-An Zeng,Meng-Wai Woo,Ruo-Yong Wang

The development of natural polyphenol-protein complexes for preserving surimi holds great promise. The effects of pea protein isolate (PPI) - (−)-Epigallocatechin-gallate (EGCG) non-covalent and covalent complexes modified by pulsed electric field (PEF) on the preservation of tilapia surimi were evaluated. Results showed that the PEF-treated PPI-EGCG non-covalent complex extended the shelf life of surimi by more than 6 days. This complex reduced the microbial activity and delayed the lipid and protein oxidation of surimi during chilled storage. Total viable count (TVC), thiobarbituric acid reactive substances (TBARS), and total volatile base nitrogen (TVB-N) of surimi with PEF-treated PPI-EGCG non-covalent complex on day 15 were 31.65 %, 68.67 %, and 54.46 % lower than untreated surimi, respectively. Moreover, the rate of colour deterioration and off-odour development were also delayed. The major products of lipid oxidation in surimi were reduced by 37.91 % for hexanal , 43.14 % for pentanal, 73.24 % for 1-octen-3-ol, and 39.32 % for 1-penten-3-ol on day 15. Structural analyses and computer simulations revealed that PEF was able to unfold the structure of PPI, thereby exposing more binding sites for EGCG. PEF-treated PPI-EGCG non-covalent complex allowed the slow dissociation of EGCG from PPI and maintained the antimicrobial and antioxidant activities of EGCG in a long storage time. Therefore, PEF-treated PPI-EGCG non-covalent complex was considered as a potential preservative in fish and fish products.

Extraction of phenolic compounds from Moringa oleifera Lam. leaves with ultrasonic-assisted deep eutectic solvents

Wang Zilin,Tian Yang,Yang Min,Yang Junyan,Wang Yifan,Tao Liang,Sheng Jun,Shi Chongying

Background: In this study, deep eutectic solvents (DESs) combined with ultrasound-assisted extraction (UAE) were used to extract bioactive compounds from the leaves of Moringa oleifera Lam.Methods: The FT-IR method was used to analyze the structural characteristics of the DESs, and the extraction efficiencies of the DESs for total phenolic content (TPC) and total flavonoid content (TFC) were evaluated. The stability of the extracts under high temperature and UV radiation was assessed, and their antioxidant activity was investigated after undergoing in vitro simulated digestion. Results: The results show that the seven DESs extracted more TPC and TFC than did the 70% ethanol (36.27 ± 1.58 mg GAE/g, 23.09 ± 1.47 mg RT/g), and the extraction process of UAE-DES was optimized by selecting choline chloride: citric acid as the DES solvent, which has the highest extraction of TPC (86.92 ± 1.34 mg GAE/g) and TFC (49.73 ± 0.85 mg RT/g). The stability results indicated that the DES phenolic extracts were less stable when exposed to high temperature and UV radiation, indicating that DES extracts have better bioactivity. Moreover, after in vitro simulated digestion, the DES extract shows a higher DPPH free radical scavenging capacity (12.79 ± 3.88 mmol Trolox/g of DES extracts, 6.99 ± 4.02 mmol Trolox/g of ethanol extracts) and ferric ion reducing antioxidant power (62.61 ± 1.71 mmol Trolox/g of DES extracts, 55.07 ± 1.66 mmol Trolox/g of ethanol extracts) than ethanol extracts. Conclusion: This study confirmed that DESs are a new and environmentally friendly solvent that can be used for the extraction of phenolic compounds.

Extraction, characterization, and bioactivity of soluble dietary fiber from Physalis alkekengi L. calyx

Wang Jiaming,Liang Bin,Li Zhentao,Wu Xinru,Wang Zifei,Yu Te,Gao Yang,Dai Yonggang,Wu Qiong

Physalis alkekengi L. is one of the most popular berries in the world, and its calyx is rich in nutrients. Still, the study of soluble dietary fiber (SDF) from Physalis alkekengi L. calyx (PC) has not been reported. This study aimed to extract Physalis alkekengi L. calyx soluble dietary fiber (PCSDF), and the optimal extraction process was investigated using response surface methodology. After ultrasonic pretreatment, the yield of PCSDF reached 8.74 ± 0.17% under the conditions of liquid-solid ratio of 30 mL/g, thermostable α-amylase concentration of 2000 U/mL, dispase concentration of 20,000 U/mL, amyloglucosidase concentration of 40,000 U/mL. For the first time, PCSDF was characterized by UV spectroscopy, infrared spectroscopy, scanning electron microscopy and thermogravimetric analysis, and the composition of the monosaccharides, namely mannose, glucuronic acid, rhamnose, galacturonic acid, glucose, galactose, xylose, arabinose and fucose, was identified by HPLC. For antioxidant activity, the half inhibitory concentration (IC 50 ) against DPPH, ABTS + , ·OH, and ·O 2 − radicals were 3.67, 2.96, 5.45, and 2.90 mg/mL, respectively, and there was a specific Fe 3+ reducing capacity. In terms of hypoglycemic feature, the IC 50 values for the inhibitory ability of α-glucosidase and α-amylase were 1.20 and 9.21 mg/mL, respectively. For lipid-lowering potential, the IC 50 value for pancreatic lipase inhibitory ability for lipid-lowering was 16.95 mg/mL. This paper fills the research gap of PCSDF and provides a basis for further evaluation of PCSDF using in vivo studies. Graphical Abstract
