Synthesis and SERS activity of super-multibranched AuAg nanostructure via silver coating-induced aggregation of nanostars

Jian-Jun Li,Chen Wu,Jing Zhao,Guo-Jun Weng,Jian Zhu,Jun-Wu Zhao

The super-multibranched Au Ag bimetallic nanostructures are synthesized due to the aggregation of Au nanostars in the process of silver coating. The super-multibranched bimetallic nanostructures with different silver coating thickness are obtained by changing the concentration of silver nitrate and ascorbic acid . It has been found that the formation of these nanostructures is due to the stacking of several nanostars during the process of silver coating. By comparing the silver coating process of gold nanostars with different branch lengths, we found that the nanostars with longish branches are easy to aggregate and form the super-multibranched nanostructures in the process of silver coating. In the Au Ag bimetallic nanostructures, the silver layer is mainly covered on the surface of the cores and the thickness increases with the increasing of the AgNO 3 , which leads to the change of the surface-enhanced Raman scattering (SERS) activity. It has been found that the SERS activity is stronger when the silver layer is thin and the Au branches are still exposed to the outside of the Ag shell. The sample with the strongest SERS activity has been used to detect thiram with different concentrations. The Raman intensity increases linearly with the logarithmic concentration of thiram ranging from 10 −3 to 10 −7  M with a detection limit of 6.3 × 10 −7 M. These experimental results show that the super-multibranched bimetallic nanostructures have a broad application prospect in molecular detection and biologic sensing based on SERS.

Fabrication of a Flowerlike Ag Microsphere Film with Applications in Catalysis and as a SERS Substrate

Qijun Dai,Lu Li,Chungang Wang,Changli Lv,Zhongmin Su,Fang Chai

Graphical Thin films of Ag microflowers (MFs) were fabricated on substrates. SEM studies revealed that the Ag MFs are assembled from thin nanosheets composed of Ag nanoparticles. The potential of the film as a catalyst in the reduction of nitrophenols and as a substrate for surface-enhanced Raman scattering was investigated. Three-dimensional flowerlike Ag microstructures were assembled in situ on a substrate to fabricate Ag microflower (Ag MF) films. The morphology and size of the Ag MFs can be easily controlled by varying the reaction time. The mechanism of growth of Ag MFs was investigated by SEM. The shape evolution revealed that the Ag MFs are assembled from thin nanosheets composed of Ag nanoparticles (NPs), and the elemental composition of the NPs was evidenced by energy-dispersive X-ray spectroscopic analysis. The potential of the film as a catalyst in the reduction of nitrophenols was investigated. The reusability of the catalyst was estimated by recycling catalysis of 4-nitrophenol. Moreover, the Ag MF film could be used as a versatile substrate for surface-enhanced Raman scattering (SERS) for detection of Rhodamine 6G with a detection limit as low as 1 × 10 –7 m . The results demonstrated that the Ag MF film exhibits excellent SERS enhancement ability and catalytic activity.

SERS based protocol using flow glass-hemostix for detection of neuron-specific enolase in blood plasma

Dawei Li,Mingfeng Yang,Hanxia Li,Leilei Mao,Ying Wang,Baoliang Sun

An inexpensive and disposable lateral flow glass-hemostix (FGH) has been developed as an immunoassay, in which surface-enhanced Raman scattering (SERS) is utilized for sensing signal transduction. Gold nanocage@Raman reporter nanoparticles are developed as SERS tags, which is the key to the high sensitivity of the device. Compared with ELISA, the SERS-FGH exhibits superior performance in terms of sensitivity and limit of detection (LOD) in a blood plasma-containing sample matrix. In addition, the SERS-FGH has been successfully used for the detection of neuron-specific enolase (NSE), a traumatic brain injury (TBI) protein biomarker, in diluted blood plasma samples, achieving a LOD of 0.74 ng mL−1. Moreover, the SERS-FGH was successfully employed to measure the NSE level in clinical blood plasma samples taken from identified TBI patients. This work demonstrates that the SERS-FGH has great potential in assisting screening of TBI patients in the point-of-care setting.

In situ synthesis of gold nanoparticles (AuNPs) in butterfly wings for surface enhanced Raman spectroscopy (SERS)

Zhongde Mu,Xiangwei Zhao,Zhuoying Xie,Yuanjin Zhao,Qifeng Zhong,Ling Bo,Zhongze Gu

Recently, surface enhanced Raman spectroscopy (SERS) has been widely studied for chemical and biological sensing. SERS substrates combined with photonic architectures have attracted more and more attention. Photonic architectures in butterfly wings are too complicated to fabricate with either “top-down” or “bottom-up” approaches. Herein, we developed a simple, reproducible, inexpensive and green method to fabricate SERS substrates from different butterfly wings where the component chitosan/chitin was utilized as in situ reducer to synthesize gold nanoparticles in natural 3D photonic architectures. The SERS performances of three butterfly wings are compared. And the results show that a SERS substrate based on M. menelaus is better than the other three substrates, which could detect 10−9 M 4-ATP and has the lowest RSD and moderate SNR. In situ synthesis of AuNPs in butterfly wings with photonic architectures paves the way for fabricating multiple SERS substrates based on natural materials.

Hydrogen-Bond-Mediated in Situ Fabrication of AgNPs/Agar/PAN Electrospun Nanofibers as Reproducible SERS Substrates

Tong Yang,Hui Yang,Shu Jun Zhen,Cheng Zhi Huang

Reproducibility in surface enhanced Raman scattering (SERS) measurements is a challenge. This work developed a facile way to make highly dispersed uniform silver nanoparticles (AgNPs) loaded in the agar/polyacrylonitrile (PAN) nanofibers by the coupling the electrospinning technology from metal complex-containing polymer solution and in situ photoreductive technique. Agar, as hydrophilic component, was introduced into the electrospinning solution considering that its abundant hydroxyl group sites could greatly improve the contents of silver ions in the polymers because of the rich silver ion chelated with the hydroxyl group, whereas hydrophilic agar was integrated with hydrophobic PAN by −OH···N≡C– hydrogen bonds as a bridge. Meanwhile, the in situ photoreductive reaction was made under different light irradiations such as desk lamp, 365 nm UV-lamp, and 254 nm UV-lamp. High yield of stable AgNPs with highly uniform and dispersion are available in the agar/PAN nanofibers after the in situ photoreductive reaction, supplying the possibility of reproducible SERS signals. To identify that concept of proof, a facile approach for the determination of malachite green (MG) in three environmental practical samples was demonstrated by using the composite nanofibrous material irradiated by 365 nm UV-lamp, giving the minimum detection concentration of MG as low as 0.1 μmol/L with a good linear response ranging from 0.1–100 μmol/L (R2 = 0.9960).

Electrosprayed large-area membranes of Ag-nanocubes embedded in cellulose acetate microspheres as homogeneous SERS substrates

Yan Ke,Guowen Meng,Zhulin Huang,Ningning Zhou

For surface-enhanced Raman scattering (SERS)-based detection, it is desirable that the SERS substrates should not only have high SERS sensitivity, but also remarkable SERS-signal reproducibility and a good affinity for the target analytes. Herein, we report large-area membranes of plasmonic Ag-nanocubes (Ag-NCs) embedded in cellulose acetate (CA) microspheres (MSs) (denoted as Ag-NCs@CA-MSs), achieved by the electrospray technique, as highly sensitive and extremely homogeneous SERS substrates with good capture ability for analyte molecules in an aqueous solution. As a result, p-aminothiophenol (a probe molecule) and methyl parathion (a toxic pesticide) with concentrations down to 10−9 M and 10−7 M could be detected, respectively. Importantly, the membranes showed remarkable SERS-signal homogeneity over a large area, with a relative signal deviation down to 2.8% in a 500 × 500 μm2 area and 9.6% for the whole substrate (5 × 5 mm2). Moreover, Langmuir nonlinear fitting of the Raman intensity against the methyl parathion concentration was achieved, with a double-reciprocal plot of the Raman peak intensity versus the concentration showing a good linear relationship, making it possible for the quantitative SERS-based detection. Therefore, the Ag-NCs@CA-MS membranes showed potential for the quantitative SERS-based analysis of organic pollutants in the aqueous solution.

Simultaneous detection of clenbuterol and ractopamine based on multiplexed competitive surface enhanced Raman scattering (SERS) immunoassay

Meng Yu,Yongjun Hu,Jianzhi Liu

In recent years, the illegal abuse of β2-adrenergic agonists in livestock for growth promotion has been reported frequently. For conventional immunoassay methods, the development of quantitative and simultaneous detection is a great challenge. In this report, a highly sensitive multiplexed immunoassay based on competitive surface-enhanced Raman scattering (SERS) spectroscopy has been developed for the simultaneous detection of clenbuterol and ractopamine. The SERS nanoprobes based on gold nanoparticles (AuNPs) were attached by 4,4′-dipyridyl (DP), clenbuterol antibody, and 2,2′-dipyridyl (BP), ractopamine antibody respectively. The mode of detection of multiple antibodies was implemented by competitive binding between free clenbuterol, free ractopamine and clenbuterol–BSA, ractopamine–BSA immobilized on the substrates with their antibodies labeled on SERS nanoprobes. The results indicated that compared to conventional methods, the proposed method can detect clenbuterol and ractopamine simultaneously over a wide concentration range (1–1000 pg mL−1) with a lower limit of ca. 1.0 pg mL−1, which holds great potential for applications in biochemical analysis and food safety.

Silver nanotriangles-loaded filter paper for ultrasensitive SERS detection application benefited by interspacing of sharp edges

Chong Wang,Bingxin Liu,Xincun Dou

The strongly enhanced electromagnetic “hotspots” from the edges and tips of nanoplates are highly preferred to significantly promote the surface enhanced Raman scattering (SERS) activity. Here, Ag nanotriangles with a sharp edge of 20   nm were prepared by the reduction of AgNO 3 using N,N-Dimethylformamide (DMF) with the assistance of Polyvinyl pyrrolidone (PVP). By loading of Ag nanotriangles onto the filter paper, a flexible SERS substrate with good activity and reproducibility was obtained and the ultratrace detection of p -aminothiophenol (pATP) with a concentration as low as 10 −8   M was achieved. The sharp edges and tips boosted SERS activity was proved by the finite element method modeling of the plasmonic properties of Ag nanotriangles with different edge thicknesses (10 − 80   nm), Ag nanosheets with different tip acutances (squares and hexagons) together with Ag nanorods, nanocubes and nanospheres. The experimental and theoretical results confirmed that the interspacing created by filter paper plays an important role in enhancing the overall SERS activity. Furthermore, it is shown that the present silver nanotriangles-loaded filter paper SERS substrate can be employed to sensitive and selective detecting ultratrace military explosives picric acid (PA) of 10 −6   M.

Silver Nanoprism-Loaded Eggshell Membrane: A Facile Platform for In Situ SERS Monitoring of Catalytic Reactions

Yaling Li,Yong Ye,Yunde Fan,Ji Zhou,Li Jia,Bin Tang,Xungai Wang

We reported the fabrication of an in situ surface-enhanced Raman scattering (SERS) monitoring platform, comprised of a porous eggshell membrane (ESM) bioscaffold loaded with Ag nanoprism via an electrostatic self-assembly approach. The localized surface plasmon resonance (LSPR) property of silver nanoprism leads to the blue color of the treated ESMs. UV-vis diffuse reflectance spectroscopy, scanning electron microscope (SEM), X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS) measurements were employed to observe the microstructure and surface property of Ag nanoprisms on the ESMs. The silver nanoprism-loaded eggshell membrane (AgNP@ESM) exhibited strong catalytic activity for the reduction of 4-nitrophenol by sodium borohydride (NaBH4) and it can be easily recovered and reused for more than six cycles. Significantly, the composites also display excellent SERS efficiency, allowing the in situ SERS monitoring of molecular transformation in heterogeneous catalysis. The results indicate that the AgNP@ESM biocomposite can achieve both SERS and catalytic functionalities simultaneously in a single entity with high performance, which promotes the potential applications of ESM modified with functional materials. Keywords:eggshell membrane;silver nanoprism;catalysis;SERS

Facile synthesis of gold nanoflowers as SERS substrates and their morphological transformation induced by iodide ions

Zhen Shujun,Wu Tong,Huang Xin,Li Yuanfang,Huang Chengzhi

We report a new strategy to prepare gold nanoflowers (AuNFs) using a two-step seed-mediated method. The as-prepared AuNFs were employed as surface-enhance Raman scattering (SERS) substrates, showing strong signal enhancement. We further found that iodide ions (I – ) could selectively induce the morphological transformation of AuNFs to spheres, resulting in a blue-shift of the localized surface plasmon resonance (LSPR) bands, a color change of the AuNFs solution from blue to red, and decreased SERS activity. This behavior allows the AuNFs to be used in the determination of I – .

Ultrasensitive SERS detection of Bacillus thuringiensis special gene based on Au@Ag NRs and magnetic beads

Long Wu,Xiaoyan Xiao,Kun Chen,Wenmin Yin,Qin Li,Pan Wang,Zhicheng Lu,Jing Ma,Heyou Han

Abstrcact Highly sensitive and selective detection of specific DNA sequences is of great importance in clinical diagnosis, environmental and food monitoring, but it still remains challenges to develop a facile method for real sample detection in aqueous solution. Here, a simple and recyclable surface enhanced Raman scattering (SERS) sensor was constructed for Bacillus thuringiensis (Bt) special gene fragment detection by Fe 3 O 4 magnetic beads (MBs) and Au–Ag core-shell nanorods (Au@Ag NRs). A hairpin DNA with sulfhydryl and biotin was attached to Au@Ag NRs as indicator, and MBs with streptavidin (SA) were acted as the capture probe. On the basis of the biotin–SA specific interaction, target sequences were first hybridized with the hairpin DNA and exposed the biotin. Subsequently, the Au@Ag NRs were captured by the streptavidin modified MBs, which reduced the suspended NRs and led to the change of Raman intensity . Under the optimal conditions, the SERS intensity revealed a good linearity with Bt transgene fragment ranging from 0.1   pM to 1   nM with a detection limit of 0.14   pM (S/N=3). To demonstrate the specificity of the strategy, the single-base mismatch in DNA was discussed in the SERS assay. The results showed that the sensitivity and accuracy of the proposed method was acceptable in DNA detection, revealing a great potential in special gene detection.

Collapse-enhanced SERS effect of self-crosslinked plasmonic starch aerogels for sensitive detection of food contaminants

Jing-Xiang Wang,Yan-Dan Qin,Ri-Hui Su,Wei-Wei Zhu,Song-Song Zhao,Shu-Fang Zhang,Xue-Cai Tan,Ke-Jing Huang,Jun Yan

A flexible 3D plasmonic starch aerogel substrate was fabricated for susceptible detection of thiabendazole (TBZ) and doxycycline hydrochloride (DH). Based on the unique structure features of starch, an aerogel with high porosity and a large specific surface area was formed without any extra cross-linking agents. Silver nanoparticles (Ag NPs) uniformly distribute throughout starch aerogel and generate dense hot spots while aerogel structure collapses, which leads to significant SERS enhancement. The as-prepared aerogel substrate exhibits good SERS activity with a low detection limit of 1 × 10 −10  M for rhodamine 6G and 1 μg/L for 4-Aminothiophenol (4-ATP). In practical application for real sample, the TBZ residue on the surface of citrus was successfully detected by wipe sampling, with a good recovery rate of 96.2 %–98.8 % and a low limit of detection (LOD) of 0.031 mg/L range from 0.1 to 100 mg/L. The substrate also exhibited excellent performance in detecting DH in chicken, with recovery rates ranging from 96.9 % to 98.1 %. The LOD was 0.066 mg/L, and the detection range spanned from 0.1 to 100 mg/L. Results demonstrate that the prepared aerogel is an ideal material as a SERS substrate due to its unique properties such as strong adsorption capacity, dense hot spot, and ease of use. Therefore, the developed SERS platform-based flexible 3D plasmonic aerogel substrate paves a new avenue for highly sensitive SERS detection in the field of food safety.
