A self-assembled Ru nanozyme with H2O2-activated oxygenation for NIR-II photoacoustic imaging-guided photothermal/photodynamic therapy

Guang Liu,Zhilang Li,Zirong Lv,Qiuping Zheng,Cunji Gao,Jianniao Tian,Xing-Can Shen

The characteristic hypoxia of solid tumors seriously reduced the treatment efficacy of oxygen-dependent photodynamic therapy (PDT). Herein, an oxygen nanogenerator, comprising Ru-based nanomaterials, CS–DA@Ru nanoparticles (NPs), has been constructed to relieve tumor hypoxia along with photodynamic/photothermal therapy for efficient tumor eradication. CS–DA@Ru NPs possess targeting ability and act as catalase-mimic nanozymes to decompose endogenous hydrogen peroxide in the tumor to produce O2. The broad absorbance of CS–DA@Ru NPs in the second near-infrared (NIR-II) region enabled them to possess good photothermal properties and photoacoustic (PA) imaging ability. CS–DA@Ru NPs could generate enough hyperthermia and enormous amounts of 1O2 under NIR-II laser irradiation. Specifically, CS–DA@Ru NPs exhibit catalase-like activity as well as a significantly augmented antitumor efficacy through synergistic PTT/PDT guided by NIR-II PA imaging in both cells and in vivo experiments. This work will advance the development of noble metal nanozymes in enhancing the efficacy of O2-dependent antitumor modalities.

CeO2 In Situ Growth on Red Blood Cell Membranes: CQD Coating and Multipathway Alzheimer’s Disease Therapy under NIR

Mingyuan Chi,Jichun Liu,Lianxin Li,Yuewen Zhang,Meng Xie

Alzheimer’s disease (AD) has a complex etiology and diverse pathological processes. The therapeutic effect of single-target drugs is limited, so simultaneous intervention of multiple targets is gradually becoming a new research trend. Critical stages in AD progression involve amyloid-β (Aβ) self-aggregation, metal-ion-triggered fibril formation, and elevated reactive oxygen species (ROS). Herein, red blood cell membranes (RBC) are used as templates for the in situ growth of cerium oxide (CeO2) nanocrystals. Then, carbon quantum dots (CQDs) are encapsulated to form nanocomposites (CQD-Ce-RBC). This strategy is combined with photothermal therapy (PTT) for AD therapy. The application of RBC enhances the materials’ biocompatibility and improves immune evasion. RBC-grown CeO2, the first application in the field of AD, demonstrates outstanding antioxidant properties. CQD acts as a chelating agent for copper ions, which prevents the aggregation of Aβ. In addition, the thermal effect induced by near-infrared laser-induced CQD can break down Aβ fibers and improve the permeability of the blood–brain barrier. In vivo experiments on APP/PS1 mice demonstrate that CQD-Ce-RBC combined with PTT effectively clears cerebral amyloid deposits and significantly enhances learning and cognitive abilities, thereby retarding disease progression. This innovative multipathway approach under light-induced conditions holds promise for AD treatment.

Boosting often overlooked long wavelength emissions of rare-earth nanoparticles for NIR-II fluorescence imaging of orthotopic glioblastoma

Zheng Liu,Feng Ren,Hao Zhang,Qiang Yuan,Zhilin Jiang,Hanghang Liu,Qiao Sun,Zhen Li

Rare-earth nanoparticles (RE NPs) with narrow long wavelength emissions have been recently investigated for their potential application for fluorescence imaging in the second near-infrared window (NIR-II). Previously these RE NPs have a very limited application in the diagnosis and treatment of deep-seated tumors such as brain tumors, due to their weak fluorescence in the range of 1300–1700 nm. Herein, we report a significant enhancement of more than 10 times regular emission of NaNdF 4 nanoparticles at 1340 nm wavelength by coating them with an inert layer of NaLuF 4 , followed by sensitizing with a near-infrared dye (IR-808). We deliver these highly bright nanoparticles into the brain by using focused ultrasound to temporarily open the blood-brain barrier (BBB), and then detect the orthotopic glioblastoma by fluorescence imaging at 1340 nm. The images obtained from long wavelength fluorescence ( i.e. 1340 nm) exhibited better resolution and contrast compared to the short wavelength fluorescence ( i.e. 1060 nm). Our study not only provides insights for enhancing often overlooked emissions of rare-earth nanoparticles for NIR-II fluorescence imaging of deep-seated tumors, but also demonstrates great potential of focused ultrasound based technology in delivering nanotheranostic agents.

Site-specific mutated albumin for covalently targeting NIR-II dye enables noninvasive imaging of blood-testis barrier integrity

Junrong Lin,Zetao Dang,Yijing Du,Yanli Gao,Jia Li,Yuewei Zhang,Shoujun Zhu

Assessing the integrity of the blood-testis barrier (BTB) is critical, as it serves to protect developing sperm cells by creating a protective environment against toxins and pathogens. Here, we optimize a near-infrared (NIR) dye for in situ binding of endogenous albumin or constructing NIR-II fluorescent proteins (FPs) based on exogenous albumin, affording non-invasive in vivo imaging of BTB disruption with tunable time windows. To further enhance the targeting capability of FPs, we employ a site-directed mutagenesis (SDM) strategy to develop the mutant albumin with increased binding efficiency to CO-1080 at room temperature, which serves as a specialized exogenous albumin for accelerating the imaging window of BTB disruption. Given the common drawbacks of traditional modalities for assessing BTB integrity, non-invasive and real-time NIR-II imaging is an effective method for investigating the testicular microenvironment as well as understanding testicular physiology and pathology.

Metal–organic framework-based pH/NIR dual-responsive nanoparticles for combined photothermal/photodynamic therapy and chemotherapy of cancer

Lihua Huang,Ying Luo,Yiyang Cong,Jia Liu,Caiyan Xu,Zhaoxiang Zeng,Yihua Yin,Hao Hong,Wenjin Xu

Metal–organic frameworks (MOFs) are unique coordination polymers that have emerged as important candidates for drug delivery in recent years. Herein, we designed a drug delivery system for multi-modal combination therapy (IR-820/DOX@MIL-101-PDA-PEG-F3, simplified as ID@MIL-PPF3). In this study, pH-sensitive nanocarriers MIL-101(Fe) were prepared and loaded with the effective chemotherapy agent doxorubicin (DOX) and the phototherapeutic reagent new indocyanine green (IR-820) using a physical adsorption method. Dopamine was spontaneously polymerized in alkaline solutions to form a hybrid coating on MIL-101(Fe) (denoted as ID@MIL-101-PDA). The surface of ID@MIL-101-PDA was functionalized by the synthesized polymer NH2-PEG-OCH3/F3 to obtain ID@MIL-PPF3. Characterization results show that ID@MIL-PPF3 has a good drug loading rate (27.7%) and photothermal conversion efficiency (28.89%). In addition, Fe3+ released from MIL-101(Fe) can engage in a Fenton-like reaction to generate hydroxyl radicals (˙OH) to realize chemodynamic therapy (CDT). ID@MIL-PPF3 not only demonstrates the ability to intelligently release drugs, but also realizes chemotherapy combined with PDT/PTT/CDT therapy for tumor cells, which was proved by in vitro evaluation. In conclusion, ID@MIL-PPF3 provides a new idea and method for building a multi-modal combination therapy targeted drug delivery platform.

MoS2@AuNRs nanoparticles equipped with MnO2 nanosheets as an efficient NIR/pH/GSH triple-responsive drug delivery carrier

Menglu An,Weiying Wang,Jingguo Li,Shaokui Cao,Yingliang Liu,Jun Shi

MnO 2 nanosheets have attracted extensive attention in the field of photothermal therapy due to their excellent pH/glutathione (GSH) responsiveness. In this study, MoS 2 @gold nanorods (AuNRs)/polydopamine (PDA)@MnO 2 (400–500 nm) nanocomposites with controllable drug release property were prepared by covering MnO 2 nanosheets around near-infrared (NIR) responsive MoS 2 @AuNRs (100–200 nm) via in-situ redox reaction. The large surface area and strong negative charge of MnO 2 nanosheets ensured that MoS 2 @AuNRs/PDA@MnO 2 had a high loading efficiency of 92.44 % for doxorubicin hydrochloride (DOX). MoS 2 @AuNRs/PDA@MnO 2 revealed significant NIR/pH/GSH triple-responsive drug release properties owing to the synergistically enhanced NIR photothermal-response of MoS 2 @AuNRs (61.10 % of photothermal conversion efficiency) and the reaction between MnO 2 and GSH in weakly acidic media. In addition, in vitro cell viability results confirmed that MoS 2 @AuNRs/PDA@MnO 2 could efficiently kill tumor cells under the dual action of acidic media and NIR laser. This study develops a novel nanocarrier with NIR/pH/GSH triple-responsive drug delivery capability, which has broad applications in photothermal therapy and intelligent drug delivery.

NIR-Mediated Pyroelectric Catalysis for Sustained ROS/RNS Generation and Advanced Cancer Therapy In Vivo via Injectable Hydrogel

Yandai Lin,Yanxi Yang,Xueli Ren,Zhe Liu

Exogenous electrical stimulation has attracted considerable attention due to the advantages of microelectric induction and subsequent biological effects such as actin reorganization and reactive oxygen species (ROS) generation. Herein, an injectable hydrogel of BPR-ARG@Gel (BAG) with pyroelectric BPR nanoparticle loading and l-arginine (ARG) introduction was fabricated for advanced cancer therapy in vivo. Due to the photothermal effect, the holes and electrons in BPR nanoparticles were separated to produce an open-circuit voltage and consequently catalyze water H2O to generate toxic superoxide (•O2–) and hydroxyl radicals (•OH). These ROS substances further oxidize ARG to produce NO for synergistic tumor treatments. The mice experiments indicated that the employment of BAG hydrogel incorporation with a near-infrared laser downregulated the heat shock protein and recruited immune cells with 5-fold-enhanced expression of proinflammatory cytokines of interferon-γ. It was also noteworthy that the injectable hydrogel of BAG substantially induced the generation of reactive oxygen/nitrogen species (ROS/RNS) with reliable biosafety and strong tumor inhibition. Overall, these findings have provided potentially new inspirations and a feasible strategy to translate this multifunctional hydrogel toward tumor therapy in a pyroelectric stimulation manner.

NIR-light-induced plasmonic liquid metal/ionic liquid/MXene polyurethane films with excellent antifouling and self-healing capabilities

Peng Wang,Haohang Yuan,Baoluo He,Ruisheng Guo,Shujuan Liu,Qian Ye,Feng Zhou,Weimin Liu

The potential of organic coatings in antifouling applications has been well-documented. Beyond their exceptional antifouling effects, these coatings should also possess good mechanical strength and self-healing capabilities. Herein, a novel vinyl-based ionic liquid [VEMIM + ] [Cl – ] (IL) was in situ polymerized and then assembled onto the surface of liquid metal (GLM) nanodroplets to prepare GLM-IL. Subsequently, Ti 3 C 2 T x (MXene) was modified with GLM-IL nanodroplets to obtain GLM-IL/MXene composite, which acts as an efficient photon captor and photothermal converters and can be further composited with PU film (GLM-IL/MXene/PU). Notably, the composite film significantly increases by ∼117°C after exposure to 200 mW/cm 2 light irradiation. This increase is attributed to the high photothermal conversion efficiency of MXene and the excellent plasma effect of GLM-IL. Compared with pure PU, the GLM-IL/MXene/PU film shows a 50% improvement in tensile strength and above 85.8% healing efficiency with a local temperature increase. Additionally, the as-prepared GLM-IL/MXene/PU film reveals satisfactory antifouling properties, achieving a 99.7% reduction in bacterial presence and an 80.3% reduction in microalgae. This work introduces a novel coating with antifouling and self-healing properties, offering a wide range of applications in the fields of marine antifouling and biomedicine.

NIR-triggered and glucose-powered hollow mesoporous Mo-based single-atom nanozymes for cascade chemodynamic diabetic infection therapy

Jingwei Wang,Yin Yu,Lingzhi Chen,Jiaqi Yu,Xiaoying Jin,Runmin Zeng,Xiaomin Luo,Yanguang Cong,Guangxian Xu,Jianglin Zhang,Xueqin Huang,Jiang Pi

Diabetic infections/wounds remain to be a threatening challenge as it seriously leads to lower limb amputation with endless pains and subsequent high economic/psychosocial costs. The exceptional peroxidase-like activity of single-atom nanozymes (SAzymes) holds great promise for chemodynamic therapy (CDT) of diabetic infection, but is extremely restricted by the near-neutral pH and insufficient H 2 O 2 levels in physiological conditions. Herein, we innovated a hollow mesoporous molybdenum single-atom nanozyme (HMMo-zyme) featured with catalytic activity, photothermal performance and drug delivery properties for more effective antibacterial therapeutic in diabetic conditions. The glucose oxidase (GOx) was encapsulated into HMMo-zyme with phase-change material (PCM) to form HMMo/GOx@P system, which could be controllably disassembled by near-infrared ray (NIR) to trigger cascade CDT toward bacterial infections. The results revealed that the release of GOx accelerated by NIR could facilitate the continuous conversion of glucose (Glu) into gluconic acid, accompanied by a sharply decrease in pH to establish a low-pH environment that notably enhanced the catalytic activity of HMMo-zyme, which subsequently drives the conversion of generated H 2 O 2 into toxic hydroxyl radicals ( · OH) for amplified anti-bacterial treatment. As a proof of the concept, this NIR-assisted HMMo/GOx@P strategy could efficiently inhibit/kill bacteria and suppress tissue inflammations, thereby accelerating the wound healing processes both in in vitro and in vivo diabetic infection models. This study provides a novel strategy that may serve as a promising alternative for antibiotic therapeutics against diabetic infection, thus holding promise for more effective diabetic infection treatment manipulating Mo-based SAzymes.

Novel graphene oxide modified LDH@CEO antibacterial materials capable of NIR stimuli-responsive control release for food preservation

Xu Ma,Xinhe Zhao,Ning Zhang,XiaoYu Liu,JiYue Ding,Qingyang Xu,Ke Cui,Yi Wang,Qian Zhang,Hailin Cong

The development of antimicrobial packaging materials serves as an effective means to enhance food preservation and mitigate food waste. This study had developed a near-infrared (NIR)-controlled smart responsive system for the release of antibacterial cinnamon essential oils (CEO), which involves immobilizing CEO within the interlayer of layered double hydroxides (LDHs) and decorating it with graphene oxide (GO), designated as LDH@CEO-GO. LDH@CEO-GO was prepared by co-precipitation method. The release kinetics of CEO at various temperatures was investigated, revealing that the release dynamics conform to the Fickian diffusion mechanism. Under NIR light serves as an intelligent switch that can regulate temperature, thereby facilitating the release of CEO. Antibacterial tests demonstrated that LDH@CEO-GO exhibited good antibacterial properties, achieving inhibition rates of 77.5 % and 68.3 % against E. coli and S. aureus, respectively. NIR irradiation, the inhibition rates increased to 98.3 % and 97.2 %. Through practical exploration of strawberry preservation, it was found that under NIR light stimulation, LDH@CEO-GO can effectively enhance the preservation of strawberries.

Tunable Broadband NIR-II Emission via Cr4+-Er3+ Energy Transfer in CaMgGeO4:Cr4+,Er3+ Phosphors for Nondestructive Analysis

Hanyu Hu,Zeyu Lyu,Dashuai Sun,Shuai Wei,Jia Liu,Xin Wang,Luhui Zhou,Hongpeng You

Although there have been numerous reports on broadband near-infrared (NIR) emitting phosphors, their emissions are mainly concentrated in the range of 700–1000 nm (NIR-I). Herein, we successfully synthesized a broadband near-infrared phosphor CaMgGeO4:Cr4+(CMG:Cr4+) with an emission in the range of 1000–1600 nm (NIR-II). The introduction of Er3+ ions into CMG:Cr4+ resulted in a wider near-infrared emission phosphor CaMgGeO4:Cr4+,Er3+ (fwhm = 361 nm), compensating for the luminescence of 1500–1600 nm. More importantly, an energy transfer from Cr4+ to Er3+ ions has been discovered. Furthermore, a NIR pc-LED was fabricated by combining the CMG:Cr4+,Er3+ phosphor with a 590 nm chip. The changes in intensity and profile of the transmission spectra of light passing through different liquids reveal its potential application in organic compound recognition. This work opens a direction for the development of NIR-II phosphors.

Infectious microenvironment responsive bimetallic sulfide combined with NIR irradiation to enhance photodynamic gas synergistic therapy of maxillofacial trauma

Liao Yin-Han,Liao Hai-Qing,Wu Xing-Mou,Liang Xu-Zhi,Zhang Shi-Yu,Liu Yan,Mo Wei-Jiu,Li Sui-Hua,Jiang Ming-Yang,Gao Zhi-Wei,Wang Jia-Xi,Gao Ming,Li Hao,Li Cui-Ping

Maxillofacial trauma is an infection and oxidative stress-induced damage to maxillofacial area related cells. Increasing evidences have suggested that therapeutic strategies with controllable anti-infective and anti-inflammatory capabilities hold great potentials in the clinical treatment of maxillofacial trauma. Herein, we reported an infectious microenvironment responsive bimetallic sulfide (FeCuS x , FCS) combining with near infrared (NIR) irradiation for maxillofacial trauma therapy. In cellular levels, FCS exhibited the most efficient antioxidant and anti-inflammatory, cell migration, and angiogenesis behaviors through down-regulating the inflammatory factor (iNOS) expression level, up-regulating anti-inflammatory factors (Arg-1 and TGF-β) expression level, inducing macrophages M2 polarization (CD86 down-regulation and CD206 up-regulation), and promoting tissue repair factors (CD31) expression level. Besides, it presented the excellent antibacterial ability of Escherichia coli (99.74%) and MRSA (99.78%) for FCS + NIR. Significantly, it also confirmed that FCS + NIR presented the excellent functions of inhibiting wound infections, decreasing wound inflammation, as well as promoting trauma tissue repairing. Altogether, it had developed the strategy of FCS + NIR for the synergistic enhancement of infectious maxillofacial trauma healing, could also serve as a promising therapeutic strategy for the treatment of infectious diseases.
