Curcumin–Cu(II) Ensemble-Based Fluorescence “Turn-On” Mode Sensing the Plant Defensive Hormone Salicylic Acid In Situ and In Vivo

Chong Chen,Lin-Lin Yang,A-Ling Tang,Pei-Yi Wang,Rong Dong,Zhi-Bing Wu,Zhong Li,Song Yang

Salicylic acid (SA), a crucial, plant-derived signal molecule, is capable of launching global transcriptional reprogramming to assist plants in obtaining the systemic acquired resistance (SAR) mechanism. Thus, the accurate detection of SA will not only significantly contribute to the understanding of the plant SAR but also contribute to crop protection and to the security of the agricultural production and food supply. However, detection of SA using fluorescent probes is a great challenge for scientists, because SA analogues can significantly interfere with the detection results. Herein, we first reported using a simple, natural curcumin–Cu2+ ensemble to selectively and sensitively monitor SA in situ and in vivo, directed by a fluorescence “turn-on” mode. A binary combination curcumin–Cu2+ was first fabricated with a fluorescence “turn-off” pattern caused by the paramagnetic nature of Cu2+. Subsequently, a fluorescence “turn-on” response was performed for detecting SA accompanied by the formation of the ternary complex curcumin–Cu2+–SA due to the high affinity of SA toward Cu2+, which reduced the fluorescent impact caused by the paramagnetism of Cu2+. Further study revealed that the rationally designed hybrid probe could monitor SA in living cell lines. We anticipate that this finding can inspire the discovery of a high-performance SA probe.

Fabrication of natural deep eutectic solvent based water-in-oil high internal phase emulsion: Improving loading capacity and stability of curcumin

Jinyu Miao,Xinyu Zuo,David Julian McClements,Liqiang Zou,Ruihong Liang,Lu Zhang,Wei Liu

Water-in-oil high internal phase emulsions (HIPE W/O ) are promising as fat substitutes and nutrient carriers in food systems. A method was developed to replace the water phase of HIPE W/O with natural deep eutectic solvents (NDES) to enhance the loading and stability of nutraceuticals. NDESs, comprising varying ratios of d -glucose, sucrose , and water, were used. Higher sucrose ratios resulted in larger crystals in NDES-based HIPE W/O . Glucose-to-sucrose molar ratios below 1 generated gel-like HIPEs with excellent thermal, centrifugal, freeze-thaw, and storage stability. Increased sucrose proportion in NDES promoted crystal formation, thus improving emulsion stability. NDES-HIPEs demonstrated high curcumin loading with high encapsulation efficiency. Crystal-containing NDES-HIPEs exhibited improved ultraviolet and gastrointestinal stability. This study presents a novel method for HIPE W/O preparation. Using NDES as the internal phase regulates crystal morphology, enhances emulsion stability, and protects nutrients. These unique emulsions hold potential as carriers for bioactive components in food, nutraceutical, and pharmaceutical applications.

Phase behavior of multi-stimuli-responsive ionic liquids-based micro-emulsion and its application in nano silica synthesis and curcumin encapsulation

Zhixia Wang

The enrichment and separation of trace compounds, as well as the preparation of nanomaterials, are all greatly facilitated by responsive micro-emulsion. A multi stimuli-responsive ionic liquids-based micro-emulsion system was established with alkyl-N-Tropine (trans) P-methoxycinnamate ([C n Tr][PMCA], TILS) as a surfactant. The factors affecting the phase behavior of micro-emulsion, such as the structure of the TILS, cosurfactant, oil, temperature, pH, and salinity, were investigated by multiple research methods. Five zones were identified inside the micro-emulsion phase region as O/W, B.C., liquid crystal (L.C.), L.C.+B.C., and W/O. The TILS-based micro-emulsion displayed high stability at measured temperatures. High salinity contributed to a higher water solubilization capacity of the micro-emulsion. For the potential applications of the micro-emulsion, with sizes ranging from 20 to 70 nm, nanoscale silica particles showed persistent stability. High levels of drug loading rate (about 8.6 mg·g −1 ) and encapsulation efficiency (about 99%) of the micro-emulsion loaded with curcumin were attained.

Remodeling the structure of soy protein fibrils to hydrogels for co-encapsulation of (−)-epigallocatechin gallate (EGCG) and curcumin: Role of EGCG

Fuyun Ji,Huihui Liu,Chuyan Wang,Na Guo,Yizhong Shen,Shuizhong Luo,Shaotong Jiang,Zhi Zheng

Curcumin (Cur) and (−)-Epigallocatechin gallate (EGCG) have synergistic bioactivity in several terms, but their polar differences and environmental sensitivity make co-utilization challenging. Soy protein fibrils (UF) have sparked broad attention as outstanding carriers for bioactive substances. In this study, EGCG was applied to remodel the structure of Cur-loaded UF complex (UFC) to form novel UF-based hydrogels co-encapsulated Cur and EGCG (UFCE). The fluorescence spectroscopy, ITC , and FTIR analysis demonstrated that EGCG absorbed and deposited on the surface of UFC through hydrophobic and electrostatic interactions. The binding of EGCG led to the formation of UFCE, which constructed a stable and uniform gel network during heat treatment. UFCE gels formed with varying EGCG/protein concentration ([E]/[F]) ratios showed different structural properties with different encapsulation effects. At 2.5% protein concentration, UFCE gels formed at [E]/[F] ratio of 0.08 (UFCE 0.08 ) exhibited better gelling properties. The encapsulation efficiency of Cur and EGCG in UFCE 0.08 gel was up to 97.71% and 91.02%, respectively. Moreover, UFCE 0.08 gel exhibited significantly enhanced stability of Cur (95.27%) and EGCG (70.27%) than that of free Cur (53.13%) and free EGCG (13.32%) after thermal treatment at 85 °C for 120 min. UFCE 0.08 gel was also effective in improving the UV light stability of Cur (88.70%) and EGCG (72.96%). Furthermore, the binding of EGCG had pronouncedly boosted the anticancer activity of Cur in the UFCE 0.08 gel against Caco-2 cells. This study provides new perspectives into the fibril-polyphenol hydrogel systems for the co-encapsulation of EGCG and Cur, which has substantial potential for health-promoting applications.

Fabrication of multi-functional and breathable superhydrophobic cotton fabric based on curcumin/ZIF-8 through mild thiol-ene click chemistry reaction

Hua-Bin Yuan,Jin Cheng,Desheng Sha,ManMan Zhao,Tieling Xing,Guoqiang Chen

Multifunctional superhydrophobic materials demonstrate remarkable performance and promising application prospects in various fields. However, there is still considerable interest in developing milder superhydrophobic modification techniques for materials. In this study, we aimed to improve the surface roughness of cotton fabric by utilizing an in-situ growth method to incorporate zeolitic imidazolate framework-8 (ZIF-8). The fabric underwent a subsequent treatment with curcumin at room temperature, which served as a bridge for connection and provided multifunction to form a core–shell structure (curcumin/ZIF-8). Eventually, by implementing a mild thiol-ene click chemistry reaction, specifically by grafting octadecanethiol onto the C = C bond within the curcumin structure, we successfully prepared superhydrophobic modified cotton fabric known as SHCZC. The contact angle of the SHCZC material reached 168.7°, accompanied by a sliding angle of just 1°. Furthermore, SHCZC exhibited exceptional stability in harsh environments, enduring exposure to strong acids, alkalis, organic solvents, high and low temperatures, repeated washing, and abrasion. The material also demonstrated excellent thermal stability and remarkable breathability. Additionally, SHCZC possessed self-cleaning, UV-blocking, anti-fouling, antibacterial, anti-icing, and oil/water separation characteristics. The presence of these characteristics expands the potential of SHCZC beyond the field of superhydrophobic materials, positioning it as a highly advanced and versatile multifunctional material.

“Dual sensitive supramolecular curcumin nanoparticles” in “advanced yeast particles” mediate macrophage reprogramming, ROS scavenging and inflammation resolution for ulcerative colitis treatment

Han Xiaoqin,Luo Ruifeng,Qi Shanshan,Wang Yanli,Dai Linxin,Nie Wenbiao,Lin Meisi,He Haoqi,Ye Naijing,Fu Chaomei,You Yu,Fu Shu,Gao Fei

Ulcerative colitis (UC) faces some barriers in oral therapy, such as how to safely deliver drugs to the colon and accumulate in the colon lesions. Hence, we report an advanced yeast particles system loaded with supramolecular nanoparticles with ROS scavenger (curcumin) to treat UC by reducing oxidative stress state and inflammatory response and accelerating the reprogramming of macrophages. In this study, the dual-sensitive materials are bonded on β-cyclodextrin (β-CD), the D-mannose (Man) is modified to adamantane (ADA), and then loaded with curcumin (CUR), to form a functional supramolecular nano-delivery system (Man-CUR NPs) through the host-guest interaction. To improve gastrointestinal stability and colonic accumulation of Man-CUR NPs, yeast cell wall microparticles (YPs) encapsulated Man-CUR NPs to form Man-CUR NYPs via electrostatic adsorption and vacuum extrusion technologies. As expected, the YPs showed the strong stability in complex gastrointestinal environment. In addition, the Man modified supramolecular nanoparticles demonstrated excellent targeting ability to macrophages in the in vitro cellular uptake study and the pH/ROS sensitive effect of Man-CUR NPs was confirmed by the pH/ROS-dual stimulation evaluation. They also enhanced lipopolysaccharide (LPS)-induced inflammatory model in macrophages through downregulation of pro-inflammatory factors, upregulation of anti-inflammatory factors, M2 macrophage polarization, and scavenging the excess ROS. Notably, in DSS-induced mice colitis model, Man-CUR NYPs can reduce the inflammatory responses by modulating TLR4/NF-κB signaling pathways, alleviate oxidative stress by Nrf2/HO-1 signaling pathway, promote macrophages reprogramming and improve the favorable recovery of the damaged colonic tissue. Taken together, this study not only provides strategy for “supramolecular curcumin nanoparticles with pH/ROS sensitive and multistage therapeutic effects” in “advanced yeast particles”, but also provided strong theoretical support multi-effect therapy for UC.

Curcumin encapsulation in self-assembled nanoparticles based on amphiphilic palmitic acid-grafted-quaternized chitosan with enhanced cytotoxic, antimicrobial and antioxidant properties

Yinzhuo Xie,Xiaochen Gong,Zheng Jin,Wei Xu,Kai Zhao

The practical application of curcumin (CUR) is greatly limited due to its instability, high hydrophobicity, low bioavailability, and inability to cross the mucosal barrier of gastrointestinal tract. To overcome these disadvantages, several delivery systems have been explored to formulate CUR for oral administration. Nanoparticles (NPs) can significantly enhance oral absorption, bioavailability and therapeutic efficacy of drug, however, NPs are limited by the gastrointestinal degradation, mucosal and epithelial barriers. A novel amphiphilic quaternary ammonium chitosan (N-2-HACC) based NP delivery carrier was prepared using palmitic acid (PA) to encapsulate CUR. Palmitoyl chitosan (PA-N-2-HACC) was characterized including FT-IR, 1 H NMR, TGA, and CAC. The particle size of PA-N-2-HACC NPs and PA-N-2-HACC NPs loaded with CUR ( [email protected] NPs) was 231.6 ± 9.24 nm and 264.5 ± 4.31 nm. The encapsulation efficiency and loading capacity of [email protected] NPs was 75.43 ± 1.25 % and 6.81 ± 0.16 %. [email protected] NPs exhibited sustained and controlled release. Compared with the CUR, minimum inhibitory concentration of the [email protected] NPs against Escherichia coli , Staphylococcus aureus and Candida albicans was reduced by 4.2 times, 1.6 times and 4.6 times, respectively. Moreover, the antioxidant activity of [email protected] NPs was dose-dependent and higher than that of free CUR. The PA-N-2-HACC NPs show little toxicity and are a promising delivery system for encapsulating hydrophobic drugs.

A curcumin-crosslinked bilayer film of soy protein isolate and chitosan with enhanced antibacterial property for beef preservation and freshness monitoring

Yueyuan Yong,Shancan Wang,Laihao Li,Rui Li,Hafiz Nabeel Ahmad,Noshaba Munawar,Jie Zhu

In this study, antibacterial and antioxidant bilayer films were prepared by using curcumin (Cur) crosslinked soy rotein isolate (SPI) and chitosan (CS). Molecular docking simulations and multispectral analysis revealed that hydrogen bonding and hydrophobic interactions were the primary driving forces that promoted the self-assembly of the bilayer films. The tensile strength, the UV-blocking properties and the hydrophobicity was greatly improved of the bilayer antimicrobial films. Moreover, water vapor permeability, thermal shrinkage and opacity were all reduced significantly. In addition, the composite films with curcumin demonstrated effective antioxidant activity and a slow release characteristic. Morphology observation of the bacteria by AFM revealed that the antibacterial bilayer film had a significant damaging effect on the cell structures of S. aureus and E. coli due to the dual antibacterial effect of curcumin and chitosan. SPI + Cur-CS antimicrobial bilayer film effectively inhibited the growth of bacteria and extended the shelf life of beef. According to the findings, SPI + Cur-CS antimicrobial bilayer film can be used as an active package material for beef preservation and freshness monitoring.

Polyphosphazene Nanodrugs for Targeting Delivery and Inflammation Responsive Release of Curcumin to Treat Acute Lung Injury by Effectively Inhibiting Cytokine Storms

Xiaochen Su,Xunan Jing,Wanting Jiang,Meng Li,Kai Liu,Menghao Teng,Yayun Ma,Daquan Wang,Lingjie Meng,Yingang Zhang,Wenchen Ji

An excessive inflammatory response induced by cytokine storms is the primary reason for the deterioration of patients with acute lung injury (ALI). Though natural polyphenols such as curcumin (CUR) have anti-inflammation activity for ALI treatment, they often have limited efficacy due to their poor solubility in water and oxidising tendency. This study investigates a highly cross-linked polyphosphazene nano-drug (PHCH) developed by copolymerisation of CUR and acid-sensitive units (4-hydroxy-benzoic acid (4-hydroxy-benzylidene)-hydrazide, D-HBD) with hexachlorotripolyphosphonitrile (HCCP) for improved treatment of ALI. PHCH can prolong the blood circulation time and targeted delivery into lung inflammation sites by enhancing CUR's water dispersion and anti-oxidant properties. PHCH also demonstrates the inflammation-responsive release of CUR in an inflammation environment due to the acid-responsive degradation of hydrazine bonds and triphosphonitrile rings in PHCH. Therefore, PHCH has a substantial anti-inflammation activity for ALI treatment by synergistically improving CUR's water-solubility, bioavailability and biocompatibility. As expected, PHCH attenuates the cytokine storm syndrome and alleviates inflammation in the infected cells and tissues by down-regulating several critical inflammatory cytokines (TNF-α, IL-1β, and IL-8). PHCH also decreases the expression of p-p65 and C-Caspase-1, inhibiting NLRP3 inflammasomes and suppressing NF-κB signalling pathways. The administrated mice experiments confirmed that PHCH accumulation was enhanced in lung tissue and showed the efficient scavenging ability of reactive oxygen species (ROS), effectively blocking the cytokine storm and alleviating inflammatory damage in ALI. This smart polyphosphazene nano-drug with targeting delivery property and inflammation-responsive release of curcumin has excellent potential for the clinical treatment of various inflammatory diseases, including ALI.

A fluorescent nanoprobe based on N/S co-doped carbon dots coupled with molecularly imprinted polymers for the detection of curcumin

Fang Fang,Shaohua Zhang,Chaofan Chen,Xueying Wang,Chuannan Luo,Qin Wei

Curcumin can be used to treat cancer diseases, however the side effects of excessive use are serious, and the dosage of curcumin should be accurately controlled. In this study, a fluorescence nanosensor using carbon quantum dots (N,S-CQD) embedded multifunctional molecularly imprinted polymer (N,S-CQD@MIP) composite was developed as a probe for the detection of curcumin. The N,S-CQD@MIP was synthesized by sol-gel method and found a maximum emission peak at 430 nm. The curcumin can be binded selectively by the specific interaction between recognition sites and template molecules. The corresponding sensing mechanism of N,S-CQD@MIP for the detection of curcumin is the result of fluorescence inner filter effect and the static quenching effect. The fluorescence nanosensor using N,S-CQD@MIP for the curcumin detection has a good linear relationship in the range of 0.339–136.0 μM, and the corresponding detection limit is as low as 1.357 nM. This method has been applied to detect the curcumin in real samples with the recoveries of 97.6–103.7%. These results indicate that N,S-CQD@MIP are potential sensing materials for curcumin detection.

Curcumin inhibits liquid–liquid phase separation of fused in sarcoma and attenuates the sequestration of pyruvate kinase to restore cellular metabolism

Wen-Pu Shi,Wen-Juan Lin,Wan-Yi Ge,Liang-Liang Chen,Tuo-Di Zhang,Wei-Hong Guo,Jian-Li Liu,Da-Chuan Yin

The abnormal accumulation of fused in sarcoma (FUS) is a pathological hallmark in a proportion of patients with frontotemporal dementia and amyotrophic lateral sclerosis. Therefore, the clearance of FUS aggregates is a possible therapeutic strategy for FUS-associated neurodegenerative diseases. This study reports that curcumin can strongly suppress FUS droplet formation and stress granule aggregation of FUS. Fluorescence spectra and isothermal titration calorimetry showed that curcumin can bind FUS through hydrophobic interactions, thereby reducing the β-sheet content of FUS. Aggregated FUS sequesters pyruvate kinase, leading to reduced ATP levels. However, results from a metabolomics study revealed that curcumin changed the metabolism pattern and differentially expressed metabolites were enriched in glycolysis. Curcumin attenuated FUS aggregation-mediated sequestration of pyruvate kinase and restored cellular metabolism, consequently increasing ATP levels. These results indicate that curcumin is a potent inhibitor of FUS liquid–liquid phase separation and provide novel insights into the effect of curcumin in ameliorating abnormal metabolism.

In vitro corrosion resistance and dual antibacterial ability of curcumin loaded composite coatings on AZ31 alloy: Effect of amorphous calcium carbonate

Kui Xue,Yan-Jin Li,Tian-Hao Ma,Lan-Yue Cui,Cheng-bao Liu,Yu-hong Zou,Shuo-Qi Li,Fen Zhang,Rong-Chang Zeng

Rapid corrosion and bacterial infection are obstacles to put into use biodegradable magnesium (Mg) alloy as biomedical materials . In this research, an amorphous calcium carbonate (ACC)@curcumin (Cur) loaded poly-methyltrimethoxysilane (PMTMS) coating prepared by self-assembly method on micro-arc oxidation (MAO) coated Mg alloy has been proposed. Scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy and Fourier transform infrared spectroscopy are adopted to analyze the morphology and composition of the obtained coatings. The corrosion behaviour of the coatings is estimated by hydrogen evolution and electrochemical tests. The spread plate method without or with 808 nm near-infrared irradiation is applied to evaluate the antimicrobial and photothermal antimicrobial ability of the coatings. Cytotoxicity of the samples is tested by 3-(4,5)-dimethylthiahiazo(-z-y1)-2,5-di- phenytetrazoliumromide (MTT) and live/dead assay culturing with MC3T3-E1 cells. Results show that the MAO/ACC@Cur-PMTMS coating exhibited favourable corrosion resistance , dual antibacterial ability, and good biocompatibility. Cur was employed as an antibacterial agent and photosensitizer for photothermal therapy. The core of ACC significantly improved the loading of Cur and the deposition of hydroxyapatite corrosion products during degradation, which greatly promoted the long-term corrosion resistance and antibacterial activity of Mg alloys as biomedical materials.
